Abstract:
Calcium (Ca2 +) homeostasis is essential in conducting various cellular processes including nerve transmission, muscular movement, and immune response. Changes in Ca2 + concentration in the cytoplasm are significant in bringing about various immune responses such as pathogen clearance and apoptosis. Various key players are involved in calcium homeostasis such as calcium binders, pumps, and channels. Sequence-based evolutionary information has recently been exploited to predict the biophysical behaviors of proteins, giving critical clues about their functionality. Ion channels are reportedly the first channels developed during evolution. Calcium homeostasis modulator protein 6 (CALHM6) is one such channel. Comprised of a single domain called Ca_hom_mod, CALHM6 is a stable protein interacting with various other proteins in calcium regulation. No previous attempt has been made to trace the exact evolutionary events in the domain of CALHM6, leaving plenty of room for exploring its evolution across a wide range of organisms. The current study aims to answer the questions by employing a computational-based strategy that used profile Hidden Markov Models (HMMs) to scan for the CALHM6 domain, integrated the data with a time-calibrated phylogenetic tree using BEAST and Mesquite, and visualized through iTOL. Around 4,000 domains were identified, and 14,000 domain gain, loss, and duplication events were observed at the end which also included various protein domains other than CALHM6. The data were analyzed concerning CALHM6 evolution as well as the domain gain, loss, and duplication of its interacting partners: Calpain, Vinculin, protein S100-A7, Thioredoxin, Peroxiredoxin, and Calmodulin-like protein 5. Duplication events of CALHM6 near higher eukaryotes showed its increasing complexity in structure and function. This in-silico phylogenetic approach applied to trace the evolution of CALHM6 was an effective approach to get a better understanding of the protein CALHM6.

Abstract:
Calcium homeostasis modulator (CALHM) family proteins are Ca2+-regulated adenosine triphosphate (ATP)–release channels involved in neural functions including neurotransmission in gustation. Here, we present the cryo–electron microscopy (EM) structures of killifish CALHM1, human CALHM2, and Caenorhabditis elegans CLHM-1 at resolutions of 2.66, 3.4, and 3.6 Å, respectively. The CALHM1 octamer structure reveals that the N-terminal helix forms the constriction site at the channel pore in the open state and modulates the ATP conductance. The CALHM2 undecamer and CLHM-1 nonamer structures show the different oligomeric stoichiometries among CALHM homologs. We further report the cryo-EM structures of the chimeric construct, revealing that the intersubunit interactions at the transmembrane domain (TMD) and the TMD–intracellular domain linker define the oligomeric stoichiometry. These findings advance our understanding of the ATP conduction and oligomerization mechanisms of CALHM channels.

Abstract:
BackgroundSteroid-sensitive nephrotic syndrome (SSNS), the most common form of nephrotic syndrome in childhood, is considered an autoimmune disease with an established classic HLA association. However, the precise etiology of the disease is unclear. In other autoimmune diseases, the identification of loci outside the classic HLA region by genome-wide association studies (GWAS) has provided critical insights into disease pathogenesis. Previously conducted GWAS of SSNS have not identified non-HLA loci achieving genome-wide significance.MethodsIn an attempt to identify additional loci associated with SSNS, we conducted a GWAS of a large cohort of European ancestry comprising 422 ethnically homogeneous pediatric patients and 5642 ethnically matched controls.ResultsThe GWAS found three loci that achieved genome-wide significance, which explain approximately 14% of the genetic risk for SSNS. It confirmed the previously reported association with the HLA-DR/DQ region (lead single-nucleotide polymorphism [SNP] rs9273542, P=1.59×10−43; odds ratio [OR], 3.39; 95% confidence interval [95% CI], 2.86 to 4.03) and identified two additional loci outside the HLA region on chromosomes 4q13.3 and 6q22.1. The latter contains the calcium homeostasis modulator family member 6 gene CALHM6 (previously called FAM26F). CALHM6 is implicated in immune response modulation; the lead SNP (rs2637678, P=1.27×10−17; OR, 0.51; 95% CI, 0.44 to 0.60) exhibits strong expression quantitative trait loci effects, the risk allele being associated with lower lymphocytic expression of CALHM6.ConclusionsBecause CALHM6 is implicated in regulating the immune response to infection, this may provide an explanation for the typical triggering of SSNS onset by infections. Our results suggest that a genetically conferred risk of immune dysregulation may be a key component in the pathogenesis of SSNS.

Calcium homeostasis modulator 1 (CALHM1) is a voltage-gated ATP release channel that plays an important role in neural gustatory signaling and the pathogenesis of Alzheimer’s disease. Here, we present a cryo–electron microscopy structure of full-length Ca2+-free CALHM1 from Danio rerio at an overall resolution of 3.1 Å. Our structure reveals an octameric architecture with a wide pore diameter of ~20 Å, presumably representing the active conformation. The overall structure is substantially different from that of the isoform CALHM2, which forms both undecameric hemichannels and gap junctions. The N-terminal small helix folds back to the pore and forms an antiparallel interaction with transmembrane helix 1. Structural analysis revealed that the extracellular loop 1 region within the dimer interface may contribute to oligomeric assembly. A positive potential belt inside the pore was identified that may modulate ion permeation. Our structure offers insights into the assembly and gating mechanism of the CALHM1 channel.

Abstract:
Calcium homeostasis modulator protein-1 (CALHM1) and its Caenorhabditis elegans (ce) homolog, CLHM-1, belong to a new family of physiologically important ion channels that are regulated by voltage and extracellular Ca2+ (Ca2+o) but lack a canonical voltage-sensing domain. Consequently, the intrinsic voltage-dependent gating mechanisms for CALHM channels are unknown. Here, we performed voltage-clamp experiments on ceCLHM-1 chimeric, deletion, insertion, and point mutants to assess the role of the NH2 terminus (NT) in CALHM channel gating. Analyses of chimeric channels in which the ceCLHM-1 and human (h)CALHM1 NH2 termini were interchanged showed that the hCALHM1 NT destabilized channel-closed states, whereas the ceCLHM-1 NT had a stabilizing effect. In the absence of Ca2+o, deletion of up to eight amino acids from the ceCLHM-1 NT caused a hyperpolarizing shift in the conductance-voltage relationship with little effect on voltage-dependent slope. However, deletion of nine or more amino acids decreased voltage dependence and induced a residual conductance at hyperpolarized voltages. Insertion of amino acids into the NH2-terminal helix also decreased voltage dependence but did not prevent channel closure. Mutation of ceCLHM-1 valine 9 and glutamine 13 altered half-maximal activation and voltage dependence, respectively, in 0 Ca2+. In 2 mM Ca2+o, ceCLHM-1 NH2-terminal deletion and point mutant channels closed completely at hyperpolarized voltages with apparent affinity for Ca2+o indistinguishable from wild-type ceCLHM-1, although the ceCLHM-1 valine 9 mutant exhibited an altered conductance-voltage relationship and kinetics. We conclude that the NT plays critical roles modulating voltage dependence and stabilizing the closed states of CALHM channels.

Quartz crystal microbalances (QCMs) were utilized for the detection of the noble gas xenon (Xe) by combining them with different recognition layers such as permethylated calixarenes (tetramethyl-tert-butylcalix[4]arene (Cal4Me), hexamethyl-tert-butylcalix[6]arene (Cal6Me)), and polyurethanes, with covalently embedded Cal4OH (Poly4Cal), or Cal6OH (Poly6Cal). A third type of sensitive material is synthesized from polyacrylic acid (PAA) and polyvinyl alcohol (PVA) and utilized as a sensitive coating. The results demonstrate that the Cal4Me layer gives higher response towards Xe, while, by the use of a second uncoated QCM channel, the influence of ambient humidity could be nearly completely compensated by signal subtraction. Moreover, the Cal4Me sensor shows excellent reversibility and rapid response time, providing a potentially reliable way to determine Xe during anaesthesia.

Abstract:
La perturbation des mécanismes de réponse au stress chez les organismes peut entraîner une dysfonction cellulaire et des maladies telles que le syndrome métabolique. L'équilibre énergétique est principalement régulé par le système nerveux central (SNC), qui intègre des signaux hormonaux, neuronaux et alimentaires provenant de divers tissus. Une dysfonction de ce système est liée à l'obésité et au syndrome métabolique, qui sont tous deux des précurseurs du diabète de type 2 (T2D). Notre laboratoire a découvert que la protéine kinase ID dépendante du calcium/calmoduline (CaMK1D), un gène associé au T2D, favorise la prise alimentaire médiée par la ghréline chez les souris. Cependant, le rôle de la signalisation de CaMK1D dans les neurones NPY/AgRP reste encore à éclaircir. Dans cette étude, nous avons réalisé un séquençage de l'ARN en utilisant la lignée cellulaire hypothalamique GT1-7. Nous avons ainsi découvert que CalHM6 est une cible potentielle en aval de la signalisation de CaMK1D. Les niveaux d'ARNm de CalHM6 sont ainsi significativement augmentés dans les cellules CaMK1D-/- et sont réduits lorsque CaMK1D est ré-exprimé. Cela a également été confirmé in vivo dans l'hypothalamus des souris CaMK1D-/-. CalHM6, probablement un canal calcique dépendant du voltage, a montré des niveaux intracellulaires de Ca2+ augmentés en réponse à la ghréline dans les cellules CaMK1D-/- par rapport aux cellules CaMK1D+/+ en utilisant la méthode jGCamps. En résumé, notre travail a montré que CalHM6 est une nouvelle cible de CaMK1D. Une expression élevée de CaMK1D, entraînant une faible expression de CalHM6, pourrait ainsi favoriser la prise alimentaire et l'obésité en modulant la signalisation dépendante du calcium dans les neurones NPY/AgRP
Disruption of stress response mechanisms in organisms can lead to cellular dysfunction and diseases like metabolic syndrome. Energy balance is mainly regulated by the central nervous system (CNS), which integrates hormonal, neuronal, and dietary signals from various tissues. Dysfunction in this system is linked to obesity and metabolic syndrome, both precursors to type 2 diabetes (T2D). Our laboratory discovered that calcium/calmodulin-dependent protein kinase ID (CaMK1D), a gene associated with T2D, promotes ghrelin-mediated food intake in mice. However, CaMK1D signaling in NPY/AgRP neurons still remains questions. In this work, we proformed RNA sequencing using the GT1-7 hypothalamic cell line. To this end, we found that CalHM6 is a downstream target of CaMK1D signaling. CalHM6 mRNA levels were significantly upregulated in CaMK1D-/- cells and downregulated when CaMK1D was re-expressed. This was confirmed in vivo in the hypothalamus of CaMK1D-/- mice. CalHM6, likely a voltage-gated calcium channel, showed increased intracellular Ca2+ levels in response to ghrelin in CaMK1D-/- cells compared to CaMK1D+/+ cells using jGCamps method. Altogether, our work showed CalHM6 is a novel target of CaMK1D. High CaMK1D, leading to low CalHM6 expression, may enhance food intake and obesity by modulating calcium-dependent signaling in NPY/AgRP neuron

De Acuerdo a la “Encuesta Nacional de Hábitos de Actividad Física y Deportes en la Población de 18 años y más” que se publicó el 31 de marzo de 2016 brinda información muy útil respecto a la práctica de Deporte en Chile en general, pero además revela datos que favorecen especialmente a la Segunda Región del País: ▪ La Región de Antofagasta es la que muestra los mejores índices de práctica deportiva (40%). Adicionalmente, es esta misma región la que presenta el más bajo porcentaje de personas sedentarias en el país. ▪ El porcentaje de población que practica deporte a Nivel Nacional aumentó en un 2.4% en relación con la misma encuesta realizada el año 2012. Asimismo, en igual período el sedentarismo disminuyó en un 2.5%. El Plan de Negocios que se presenta a continuación, consta de la creación de una Nueva UEN en un Hotel que hoy tiene infraestructura ociosa y que por otro lado tiene características diferenciadoras en relación a sus competidores más cercanos. Dada la importancia de los equipamientos, tanto desde el punto de vista social como físico del desarrollo de la ciudad, me parece importante abordar el tema deportivo y recreacional, ya que estos espacios destinados para el ocio y tiempo libre, se convierten en focos sociales importantes, otorgándoles un carácter de hito dentro de la ciudad, conformándose un espacio público característico diseñado para el encuentro ciudadano y expresiones culturales propias de la región. Se convierten en lugares para los ciudadanos consolidando su identidad y sentido de pertenecía a un lugar. La construcción de una infraestructura de las proporciones de un estadio, puede significar para una ciudad una activación y un posicionamiento que sólo es posible con una inversión de la envergadura que estas infraestructuras urbanas requieren. Con respecto a Calama, se pone en manifiesto que la inquietud en el ámbito deportivo no proviene sólo del sector público, sino que existe interés privado esfuerzo e inversión en el desarrollo de infraestructura deportiva. El estadio será construido con recursos correspondiente a la tercera etapa de los estadios bicentenarios y más el aporte de minera Radomiro Tomic. Remodelar el sector definiendo zonas de encuentro, recreacionales y deportivas deficientes en la ciudad. Recuperar la zona ferroviaria para generar áreas de esparcimiento agregándole un valor extra. La propuesta es habitar esta gran extensión de terreno con un potencial de articulador urbano y generador deactividades complementarias al centro cívico, mejorando la conectividad oriente poniente que se limita por la vía férrea. Concretar la integración y relación de dos zonas contrapuestas, por medio de un proyecto deportivo y recreacional que vincule y articule aquellos lugares, concretándose la unidad urbana. Lo principal es desarrollar el concepto de espacio público en función de cumplir un rol de nexo o conexión, que la gente adopte este espacio y le sirva problemática de unir ambos sectores y crear áreas libres de encuentro y participación ciudadana.

Nous partons d'une analyse de sa poetique implicite : elle se definit comme une triple demarche d'experimentation, dramaturgique, scenique, linguistique. Il travaglia (publiee en 1556)et la rhodiana (1553, sous le nom de ruzante) representent une tentative de subversion de la comedie erudite par adjonction de fragments comiques independants; il saltuzza (1551) est un point d'equilibre; les referents de la spagnolas (1549) sont a chercher dans le mariazo et la comedie alla bulesca; la potione (1552) et la fiorina (1553) affrontent la reecriture (de la mandragola pour l'une, de la comedie homonyme de ruzante our l'autre); les egloghe pastorali (1553) presentent des types theatraux elabores dans les autres comedies et annoncant la commedia dell'arte. Le moment de la representation est fondamental; le corps de l'acteur, l'objet theatral, le lieu scenique, les moments de pur spectacle (jeux de scene, chants, danse, acrobatie) y parlent d'une voix propre et definissent les axes de son experimentation scenique. Le plurilinguisme et une langue comique fondee sur l'accumulation sont herites a la fois des traditions des bouffons (art du contrafar et de l'invention langagiere d'un aretin. Les rapports entre les personnages et leur langue reposent avant tout sur des raisons internes au champ theatral. We start from an analysis of his poetics; it can be defined as a threefold process experimenting with dramatic art, stage functions, linguistics. Il travaglia and la rhodiana represent an attempt to overthrow the learned comedy by adding independant comic pieces. In il saltuzza, all the elements of his research are well-balanced. La spagnolas' origines are to be looked for in the mariazo and the comedy alla bulesca. La potione and la fiorina confront the rewriting (of la mandragola; of the homonimous comedy by ruzante). The egloghe pastorali set forth theatrical types already worked out in his other comedies and foreboding the commedia dell'arte. The performance is a fondamental moment; the actor's body, the stage articles, the stage geography, the moments of sheer entertainments (acting devices, singing, dancing, acrobatics feats) express themselves independantly and open up the ways for his stage experimentations. The multilingual deliberate choice and a comic language based on accumulation are inherited from both the jesters' tradition and the linguistic inventiveness of an aretino. The relationship between the caracters and their languages belong to the stage world first and foremost. Studying the letters brings out the contacts that calmo keeps with the popular culture (the grotesque realism, the attention paid to every day details, the"popular" view points) and with the venitian reality (ideological relation to a mythical venitianity which expresses itself when he pretends that the letters have been written by a fishermen's family; factual relation with the venier circle). Calmo's works open up the possibility of a venitian literature. The attention shown to the"repertoire" and the trade sets up a link with the comici dell'arte. The experimentation is an answer to the polemical situation turned against the comedy

Abstract:
El sistema carcelario en América Latina atraviesa una crisis de calidad y efectividad. Esto debido al constante aumento de la población recluída, en donde un importante porcentaje de ésta se encuentra esperando condena, la violencia al interior de las cárceles y los altos índices de hacinamiento acompañados de una nula rehabilitación, creando así las conocidas “escuelas del delito”. Chile es uno de los países con mayores tasas de población presa por habitante de América Latina, y el diagnóstico de su sistema penitenciario es también desalentador. Caracterizado por un incremento constante de la población encarcelada, con niveles de hacinamiento importantes y una limitada capacidad de reinserción y rehabilitación de los internos. En los útimos años, se ha buscado revertir esta problemática mediante una alianza público-privada que permitiría mejorar la calidad de vida de la población penal y aumentar la efectividad de los programas de reinserción de los internos. Se trata del modelo de Infraestructura Penitenciaria Concesionada, el cual se encuentra en pleno proceso de implementación en el país, y tiende a convertirse en un referente para los demás países de la región. La infraestructura penitenciaria y su permanente déficit, además de ser un tema de gran contingencia, es una problemática en que la arquitectura debe incidir y jugar un rol fundamental, siendo un gran aporte en el proceso de rehabilitación de un interno. Dentro de este contexto, el Proyecto de Título aborda esta temática a partir del encargo por parte del Ministerio de Justicia, de realizar como ejercicio académico la cárcel de Calama, la cuál es parte de los programas de infraestructura penitenciaria concesionada. Es así como este Proyecto busca desarrollar una propuesta acorde a su contexto geográfico, que a su vez incentive a los reclusos a cambiar y mejorar sus conductas, y con esto, lograr una real rehabilitación y posterior reinserción en la sociedad. Este recinto de máxima serguridad, albergará a internos de máxima, alta y media seguridad, condenados e imputados, mujeres y hombres, por lo que la segregación de la población penal será un factor importante a considerar en el diseo del recinto.

El presente proyecto de título desarrolla el conjunto habitacional “Corredor de sombra”, el cual se encuentra en la ciudad de Calama. Se emplaza en av. Balmaceda colindante al casco histórico y al estadio municipal. Hitos propios de zonas desconectadas, poniente y oriente respectivamente. El conjunto está destinado para los trabajadores de la industria minera, relacionados al nuevo y más grande yacimiento a rajo abierto del país, División Ministro Hales. El proyecto es una búsqueda de la habitabilidad propia del clima desértico, dando origen al concepto guía, la construcción de la sombra. Como también responder a los problemas propios del entorno donde se emplaza, conectando esta zona de la ciudad en sentido oriente poniente –actualmente disociadas por la vía del tren y terrenos valdios-, bajo un lenguaje común que sea adecuado para las inclemencias del clima.

It has been alleged that a major deterioration was going to be inevitable in the U.S.-Hungarian relations under the Biden Admnistration. Due to political issues, some analysts are ready to expect a low ebb of amity between Washington and Budapest. The present study goes back to the foundations and revisits the nature of the uprgading of those relations in the past four years, the different layers of American policy-making as well as their objective outcomes. This paper argues that in spite of the real potential of conflict in political commmunication, the United States will remain the most important ally and strategic partner of Hungary outside the European Union with enhanced cooperation in many fields, including defense cooperation within NATO. Then the study goes on to revisit different scenarios of the future relationship between Hungary and the United States.

We study banks’ systemic risk-taking decisions in a dynamic general equilibrium model, highlighting the macroprudential role of bank capital requirements. Banks decide on their unobservable exposure to systemic shocks by balancing risk-shifting gains against the value of preserving their capital after such shocks. Capital requirements reduce systemic risk taking, but at the cost of reducing credit and output in calm times, generating welfare trade-offs. We find that systemic risk taking is maximal after long periods of calm and may worsen if capital requirements are countercyclically adjusted. Removing deposit insurance introduces market discipline but increases the bank capital necessary to support credit, implies lower (though far from zero) optimal capital requirements and has nuanced social welfare effects.
