Full textReperfusion of the heart after an ischemic event leads to the opening of a nonspecific pore in the inner mitochondrial membrane, the mitochondrial permeability transition pore (mPTP). Inhibition of mPTP opening is an effective strategy to prevent cardiomyocyte death. For example, inhibition of mPTP opening via ischaemic preconditioning (PreC) and post-conditioning (PostC) decreased the myocardial infarct size after ischemia–reperfusion. Although the molecular composition of the mPTP remains unclear, the matrix protein cyclophilin-D (CypD) is the best defined regulatory component of mPTP. In this thesis, we demonstrated that Complex I of the respiratory mitochondrial chain also regulates mPTP in a CypD-dependent manner. We also proved that inhibition of Complex I by isoflurane prevents lethal reperfusion injury in an in vivo rat model of ischemia-reperfusion. Finally, we proved that cardioprotective inhibition of CypD modulates calcium homeostasis and fluxes between mitochondria and sarcoplasmic reticulum. In summary, our results suggest that mPTP is regulated by several interconnected factors like calcium, CypD, complex I and mitochondrial functions

Full textThis work is focused on the study of a profile of soil and soil solutions collected on an experimental plot covered with spruce. All these samples come from the watershed of the Strengbach (environment - OHGE Hydrogeochimique Observatory), were sampled at different depths (5, 10, 30 and 60 cm) and during the period between 2009 and 2013. Characterizations of soil extracts by infrared spectroscopy allowed to highlight changes in the organic functional groups with depth and that these changes have a significant impact on the behaviour of the cations (major and trace) in the soil. Ultrafiltration experiments helped to identify flows of colloidal and dissolved organic carbon as well as those of the major and trace-element present in soil solutions. The joint use of isotope tracers (87Sr / 86Sr and δ44 / 40 Ca) and chemical (Rare Earth Elements) have highlighted processes taking place at the water-soil-plant interface, as the uptake by root or soil alteration

Full textThe first contribution of this thesis is the analysis and discretization of multiphase flow in a microfluidic framework.We propose a new scheme wich is second order accurate, based on mixed finite volume / finite element method for the two phases Stokes system with surface tension.We then present the comparison of the accuracy of this new scheme with the MAC discretization in 2D and 3D axi.The second contribution is related to the study of numerical schemes in time for viscoelastic fluids.We present the current limitations in this area by studying the case of flows of wormlike micelles, polymers having the ability to reorganize themselves according to the shear rate.We show that a condition of stability related to the ratio of viscosity of the polymer and solvent in time - very restrictive - exists.A new scheme is then proposed to overcome this limitation and stability studies are conducted to demonstrate our results

Full textObjetivo: Avaliar o fluxo salivar e a fração de eliminação cintilográfica de pacientes tratados com bochecho de lauril-sulfato de sódio associado ao hidróxido de cálcio (HcT10) durante ou após o tratamento radioterápico na região cervicofacial. Casuística e método: Os pacientes foram divididos em dois grupos: O grupo I (G1) foi composto por pacientes pós-irradiados (n=20) e o grupo II (G2) por pacientes que seriam submetidos à radioterapia no início deste estudo (n=23). Os pacientes foram avaliados através de sialometria, tendo sido realizada uma coleta de saliva antes do início dos bochechos e 4 outras a cada 14 dias (C1-C5), e de cintilografia, sendo uma realizada antes do início dos bochechos e outra entre 3 e 4 meses depois. Dois pacientes do G1 e oito pacientes do G2 não fizeram as secundas cintilografias. Resultados: Houve, a partir de C4, aumento significativo na quantidade de saliva quando comparada à C1 (p<0,05) para o G1 e decréscimo da quantidade de saliva para o G2 mesmo com o uso do HcT10 (p<0,05). Os resultados da cintilografia demonstraram que houve aumento da função das glândulas parótidas do G1 (p<0,05), sem, entretanto haver diferenças para as glândulas submandibulares (p>0,05). Em relação ao G2 houve decréscimo da função de todas as glândulas salivares (p<0,05). Conclusão: O HcT10 mostrou-se benéfico para o aumento de saliva no G1, porém não foi capaz de manter a quantidade de saliva para o G2
Objective: To evaluate salivary flow and the scintigraphic elimination of patients treated with a rinse of sodium lauryl sulphate associated with calcium hydroxide (HcT10) during or after radiation therapy in the cervicofacial area. Methods: Patients were divided into two groups: Group I (G1) was composed with post-irradiated patients (n=20) and Group II (G2) was composed with patients who were undergoing radiotherapy at the start of this study (n=23). Patients were evaluated by sialometry - one saliva collection before the start of mouthwash and 4 other every 14 days (C1-C5), and scintigraphy, the first one performed before the start of the mouthwash and the last one between 3 and 4 months later Two patients in G1 and eight patients in G2 did not perform the secondary scintigraphy. Results: There was, from C4, a significant increase in the amount of saliva when compared to C1 (p<0.05) for G1 and a decreased amount of saliva into G2 even with the use of HcT10 (p<0.05). The results of scintigraphy demonstrated an augmentation in the parotid gland function in G1 (p<0.05), without, however there are differences in the submandibular glands (p>0.05). Regarding the G2 to a decrease in light of all the salivary glands (p<0.05). Conclusion: HcT10 proved to be beneficial to increase salivary flow in G1, but it was unable to keep the amount of saliva for the G2

Full textAnti-CD20 monoclonal antibodies (mAbs) are an essential component of the treatment of patients with non-Hodgkin's lymphoma and chronic lymphocytic leukemia (CLL). They mediate their antitumor effects by activating the immune system or by direct apoptotic signaling in target cells. In a previous preclinical study, we showed that treatment with anti-CD20 mAbs, rituximab and GA101, resulted in upregulated expression of early growth factor 1 (EGR-1) (Dalle et al. 2011). EGR-1 is a calcium (Ca2+) regulated transcription factor and CD20 is hypothesized to regulate transmembrane Ca2+ flux. Therefore, we aimed to assess the role of EGR-1 and Ca2+ flux in the cytotoxic activity of anti-CD20 mAbs. We have shown that EGR-1 expression is rapidly upregulated in CD20+ cells following rituximab and GA101 exposure. Decreasing EGR-1 expression by shRNA abolishes the direct cytotoxic effect of GA101 both in vitro and in vivo, indicating that EGR-1 is required for CD20-mediated cell death. Additionally, the overexpression of EGR-1 enhances the cytotoxic activity of GA101 both in vitro and in vivo. Furthermore, our results indicate that anti-CD20 mAbs induce calcium influx. Blocking the Ca2+ flux with calcium channel blockers (CCB) abolishes EGR-1 induction and impaires the GA101 efficacy in vivo and ex vivo in CLL blood samples. More importantly, our data indicate that patients receiving CCBs and anti-CD20 therapy have worst progression free survival and overall survival. In conclusion we have identified EGR-1 as a potential biomarker to predict response to anti-CD20 therapy. We demonstrated that co-treatement with CCBs negatively impacts the outcome of patients receiving anti-CD20 mAbs

The chemical and isotopic compositions of the gaseous, liquid and solid phases show than: - for calcite dissolution, two systems could occur; an open system where the CO₂ is considered as an infinite pool, a closed system where the carbon dioxide in the soil is limited. - Two processes lead to calcite precipitation: outgassing and/or evaporation of the liquid phase. The isotopic composition (18O, 13C) of the secondary calcite indicates the process involves during precipitation. The dissolution and precipitation conditions defined, a isotopic mass balance gives the relative abundances of the different carbonated fractions in the soil. The 14C datations become then significant and show two successive pedogenetic phases

Full textA method for the simulation of compressible three-phase flows is proposed taking into account gravity and capillary effects. Governing equations are written in a fractional flow formulation in terms of a global pressure equation and two saturation equations. The global pressure satisfaying a "Total differential" (TD) condition was introduced by [Chavent and Jaffré, 1986] to simplify the mathematical formulation of three-phase flows. Thus, a constrained optimization procedure is used to determine the preliminary secondary variables of the fractional flow from the effective saturation ternary diagram. Another TD interpolation class approach recentkly developped by [Chavent, 2008] is also implemented. Compared to the first formulation, the new formulation is equivalent to the classical formulation with the computational efficiency of the original formulation. We discuss the construction using spectral composite finite element, in particular the boundary conditions which need to be satisfied for the three-phase data to honor given two-phase data on the boundary of the ternary diagram [di Chiara Roupert et al. 2010]. Two efficient numerical methods are then used to solve the global pressure equation and the two saturation equations for the water and oil phase. Discontinous finite elements are used to approximate the convective term of the two saturation equations while the mixed finite element method is chosen to solve the global pressure equation and the diffusive part of the saturation equations. Full textOs tratamentos utilizados para a prevenção da xerostomia não atingem a eficácia esperada pelos pacientes, podendo ainda produzir efeitos colaterais indesejáveis. O objetivo deste trabalho foi avaliar quantitativamente o índice de fluxo salivar total não estimulado de pacientes portadores de câncer de cabeça e pescoço durante e após tratamento radioterápico exclusivo tratados com a associação de laurildietileno- glicol-éter-sulfato de sódio e hidróxido de cálcio (HcT20). Este estudo foi realizado no Hospital Dr. Hélio Angotti envolvendo trinta e um pacientes (n=31) divididos em dois grupos: o grupo I composto por dezessete pacientes (n=17) submetidos à telecobaltoterapia, na dose total de 7.000 cGy e receberam o HcT20 durante o tratamento. O grupo II composto por quatorze pacientes (n=14) iniciaram o uso do HcT20 após a conclusão da radioterapia. Foram realizadas e mensuradas coletas semanais do índice de fluxo salivar total não estimulado para os pacientes dos dois grupos. Os valores obtidos foram analisados estatisticamente através dos testes de aderência à curva normal, teste de Bartlett, teste de Friedman e teste de Wilcoxon, visando avaliar a eficácia do HcT20 tanto na prevenção da queda acentuada do fluxo salivar durante a radioterapia quanto na hipofunção salivar no período pós-radioterapia. Os pacientes do grupo I, ao término da radioterapia, mantiveram o índice de fluxo salivar total não estimulado em relação aos valores basais; os do grupo II, que utilizaram a associação medicamentosa após radioterapia, apresentaram melhora acentuada no índice de fluxo salivar total não estimulado em relação ao basal; e o uso do HcT20 apresentou melhor resultado quando utilizado durante a radioterapia.
The treatments used to prevent the xerostomy do not reach the efficiency expected by the patients, and it can have undesirable effects. The goal of this report was to make a quantifiable evaluation of the total non stimulated salivary flow index in patients with head and neck cancers during and after the exclusive radiotherapy with an association of lauryl-diethylene-glycol-ether sodium sulfate and calcium hydroxide (HcT20). This study was held at the hospital Dr. Hélio Angotti involving thirty one patients (n=31) divided into two groups: Group one had seventeen patients (n=17) submitted to fractioned radiation treatment in the head and neck region at the total dose of 7.000 cGy and had HcT20 during the treatment. Group two with fourteen patients (n=14) started with the use of HcT20 after the end of radiotherapy. A weekly and measured collection of the index of the total non stimulated saliva flow was done on the patients of both groups. The values that were obtained were analyzed statistically through the Bartlett test, the Friedman test and the Wilcoxon test, to seek an evaluation of the effectiveness of the medication on the prevention of the intense drop of the salivary flow during the radiotherapy and do avoid the xerostomy during the postradiotherapeutic period. The patients from group one at the end of the radiotherapy kept the base index of the total non stimulated salivary flow, the ones from group two that used the medication association after the radiotherapy had a great improvement of the total non stimulated salivary flow and the usage of HcT20 had a better result on the index of salivary flow during the radiotherapy.

Full textThe objective of this thesis is to significantly reduce the computational cost associated with numerical simulations governed by partial differential equations. For this purpose, we consider reduced-order models (ROMs), which typically consist of a training stage, in which high-fidelity solutions are collected to define a low-dimensional trial subspace, and a prediction stage, where this data-driven trial subspace is then exploited to achieve fast or real-time simulations. The first contribution of this thesis concerns the modeling of gas flows in both hydrodynamic and rarefied regimes. In this work, we develop a new reduced-order approximation of the Boltzmann-BGK equation, based on Proper Orthogonal Decomposition (POD) in the training stage and on the Galerkin method in the prediction stage. We investigate the simulation of unsteady flows containing shock waves, boundary layers and vortices in 1D and 2D. The results demonstrate the stability, accuracy and significant computational speedup factor delivered by the ROM with respect to the high-fidelity model. The second topic of this thesis deals with the optimal transport problem and its applications to model order reduction. In particular, we propose to use the optimal transport theory in order to analyze and enrich the training database containing the high-fidelity solution snapshots. Reproduction and prediction of unsteady flows, governed by the 1D Boltzmann-BGK equation, show the improvement of the accuracy and reliability of the ROM resulting from these two applications. Finally, the last contribution of this thesis concerns the development of a domain decomposition method based on the Discontinuous Galerkin method. In this approach, the ROM approximates the solution where a significant dimensionality reduction can be achieved while the high-fidelity model is employed elsewhere. The Discontinuous Galerkin method for the ROM offers a simple way to recover the global solution by linking local solutions through numerical fluxes at cell interfaces. The proposed method is evaluated for parametric problems governed by the quasi-1D and 2D Euler equations. The results demonstrate the accuracy of the proposed method and the significant reduction of the computational cost with respect to the high-fidelity model

Full textApoptosis is a key cellular process for tissue homeostasis. Apoptotic cell death is under control of Bcl-2 family proteins which regulate outer mitochondrial membrane permeability. However, beyond their role in apoptosis, Bcl-2 family proteins are also involved in other cellular processes such as cell cycle or metabolism. In our laboratory we are interested in non-apoptotic functions of Bcl-2 family proteins in embryonic development. Using zebrafish model we have shown that Bcl-2 proteins control different processes during early development thanks to their ability to regulate calcium homeostasis. Indeed, we have shown that the anti-apoptotic protein Nrz participates in actin cytoskeleton remodeling during epiboly by regulating cytosolic calcium concentration via an interaction with the IP3 receptor (IP3R). We have also demonstrated that Nrz decreases calcium release from the endoplasmic reticulum by inhibiting IP3 fixation on its receptor. We have furthermore identified a new pro-apoptotic member of Bcl-2 family, Bcl-wav which is expressed only in fish and frogs. This protein regulates mitochondrial calcium homeostasis by interacting with VDAC. We have moreover shown that this activity is essential for convergence and extension movements during early zebrafish development
