Abstract:
Penelitian ini bertujuan untuk meningkatkan efektivitas pelaksanaan suatu aktivitas perawatan sesuai dengan task card C05-Check Pesawat Airbus 320-200, sehingga dihasilkan aktivitas perawatan yang lebih efektive terhadap jumlah waktu yang dibutuhkan dalam mengerjakan sebuah tugas kerja dari mulai awal perawatan sampai selesai atau sering di sebut Turn Around Time (TAT). Permasalahan TAT pada aktivitas perawatan C05-Check Airbus 320-200 adalah waktu TAT yang cukup lama sehingga membutuhkan biaya operasional teknisi yang cukup besar, dan adanya kerugian karena waktu tunggu operasinal pesawat pada saat perawatan. TAT pada aktivitas perawatan C05-Check Pesawat Airbus 320-200 dengan jumplah 351 task card, selama ini masih memakai standart perusahaan yang di hitung dengan metoda giant chart yaitu selama 13 hari kerja. Permasalahan yang ada mendorong penulis melakukan analisis dan perhitungan TAT pada aktivitas perawatan C05-Check Pesawat Airbus 320-200 dengan metode Critical Path Method (CPM), sebuah metode yang digunakan untuk mengidentifikasi tugas-tugas yang ada dalam sebuah aktivitas pekerjaan dan membentuk desain jaringan CPM sesuai dengan urutan dan jalur kritis pekerjaan. Hasil perhitungan Turn Around Time dengan metode (CPM) pada aktivitas perawatan C05-Check Pesawat Airbus 320-200 dengan jumlah task card 315 adalah 10,46 hari kerja, dengan jalur kritis berada pada code aktivity A-B-D-D1-D2-D3-D4-D5-D6-D7-H-I-J, pekerjaan secara seri code aktivity A-B-H-I-J, Sedangkan pekerjaan code aktivity C-D-E-F-G. Desian jarring CPM ini menghasilkan waktu perawatan yang lebih sedikit jika di bandingkan TAT standart Perusahaan yaitu 13 hari kerja. Selisih waktu penyelesaian TAT dengan metode CPM dengan Standart perusahanan adalah 2,54 hari kerja, sehingga dapat di simpulkan bahwa TAT dengan perhitungan CPM lebih efektif pada aktivitas perawatan C05-Check Air Bus 320 - 200.

Clostridium perfringens 790-94 and 44071.C05 carrying a chromosomal and a plasmid cpe gene, respectively, were used to determine differences in heat resistance and growth characteristics between the genotypes. Heat inactivation experiments were conducted using an immersed coil apparatus. Spore germination, outgrowth, and lag phase, together named GOL time, as well as generation times were determined during constant temperatures in fluid thioglycollate (FTG) medium as well as in vacuum-packed, heat-treated minced turkey. GOL time and growth were also monitored during cooling scenarios from 65 to 10°C for 3, 4, 5, 6, and 7 h in vacuum-packed, heat-treated minced turkey. Spores of strain 790-94 were approximately 10-fold more heat resistant at 85°C than those of strain 44071.C05, and strain 790-94 also had a higher temperature growth range in FTG. The higher growth range for a chromosomal enterotoxin-producing CPE+ strain was confirmed using two other strains carrying a chromosomal (NCTC8239) and plasmid (945P) cpe gene. Moreover, strain 790-94 had shorter GOL times at 50°C in turkey and approximately half the generation time compared with strain 44071.C05 at temperatures ≥45°C in both FTG and turkey. Strain 790-94 increased with 0.3, 1.0, 1.7, and 2.0 logs, respectively, during cooling from 65 to 10°C in 4, 5, 6, and 7 h, which was signi cantly higher than for strain 44071.C05. A maximum acceptable cooling time of 5 h between 65 and 10°C is suggested.

Abstract:
The 1991 Local Government Code of the Philippines brought about a far-reaching transfer of authority and resources to its local governments. Despite bringing benefits to the nation as a whole, and to the individual local governments, devolution has also had some negative consequences. Although regional disparities were present in the country prior to devolution, their recent rise has coincided with the implementation of devolution. Regional disparities are analyzed in relation to general macroeconomic policies and trends, such as industrial policy and increased trade, and particular socioeconomic features of the Philippines, such as the significance of investments and migration. These policies and trends, coupled with faulty transfer systems and differing levels of efficiency and capacity, suggest that devolution is hampering the pursuit of regional equity. Hence it is concluded that, although many factors are resonsible for the spatial inequities, devolution in its present form plays a direct and interactive part in preserving and/or promoting regional disparities in the country.

Abstract:
Beyond their educational function, schools are frequently a focal point for community life. We argue that this latter role was compromised in New Zealand by a decade of neoliberal realignments within education policy. During the 1990s the abolition of school zones, the commodification of education, and the drive for efficiency in the allocation of educational resources undermined the place of school in community life. In this paper we explore the impacts of a school closure on an urban neighbourhood in Invercargill, New Zealand. We present and interpret narratives gathered during an interview-based study of the closure of Surrey Park Primary School. Our analysis highlights parents' perspectives on the role of the educational bureaucracy in the closure debate, the ambiguous role assigned to ‘community’ within the restructured system, the impact of the closure for low-income families, and the place of schools in contributing to the neighbourhood social cohesion. Our analysis concurs with international research suggesting that breaking links between schools and communities has potentially significant educational impacts on children.

Abstract:
The mycelium biofloc bioaugmented by Cordyceps strain C058 effectively purifies water, which may be related to the synthesis of extracellular polymer substances. To verify this conjecture, we analyzed the changes in extracellular polymer substances content in the mycelium biofloc under various hydraulic retention times (36 h, 18 h, and 11 h). The microstructure and microflora composition were analyzed using a scanning electron microscope and high-throughput sequencing. The ordinary biofloc without bioaugmentation was taken as a control. The results showed that under the above hydraulic retention time, the extracellular polymer substances contents of the mycelium biofloc were 51.20, 55.89, and 33.84 mg/g, respectively, higher than that of the ordinary biofloc (14.58, 15.72, and 18.19 mg/g). The protein content or the polysaccharide content also followed the same trend. Meanwhile, the sedimentation performance of the mycelium biofloc was better than that of the ordinary biofloc, attributed to the content of the extracellular polymer substances. It is worth noting that C058 is the main biofloc content, which promotes the synthesis of extracellular polymer substances in the mycelium biofloc. Other functional microorganisms in the mycelium biofloc were Janthinobacterium, Phormidium, Leptolyngbya, Hymenobacter, and Spirotrichea, which also promote the synthesis of extracellular polymer substances.

Abstract Introduction Variability in endometrial cancer (EC) risk and outcomes exist, with Black women disproportionately negatively affected related to White women. The biologic etiologies of these observations are poorly understood. Our objective was to describe the somatic mutations of the tumor genome and compare them to RNA expression in self-identified Black and White women diagnosed with EC. Methods Women with newly diagnosed EC and undergoing hysterectomy were recruited prospectively. Fresh tumors obtained at the time of surgery were flash-frozen, underwent DNA and RNA extraction and were submitted for whole exome sequencing (WES) analysis (DNA) or bulk RNA sequencing analysis. Differentially expressed genes between racial groups and tumor grade were calculated using Deseq2. Genomic ancestry was determined using ADMIXTURE. Chi-square and Mann Whitney U tests were used for analyses, with a threshold for significance of p<0.05. Results A total of 49 patients with EC were included. Thirty (61.2%) women were diagnosed with low-grade EC and 19 (38.8%) with high grade EC. Twenty (40.8%) women self-identified as Black, 27 (55.1%) as White and 2 (4.1%) as Asian/other. Admixture analysis showed that 36 (73.4%) had >14% African ancestry. The remaining 13 (26.5%) patients had admixed European and Native American ancestry, with <1% African ancestry. In the RNA-seq, we found significant DEGs by tumor grade; with 2132 DEG in High grade compared to Low grade tumors with a false discovery rate of <0.05 and a fold change (FC) of ≤-1.5 or ≥1.5. Top 10 DEG included MT4, TAF15, SLFN5 and PLEKHS1 (upregulated) and MCRIP1, MAZ, MYOM3, PRPH, BCL7C, AC016700.5 (downregulated). In the WES analysis, 8 Black women had mutations in GON4L compared to 1 in the White woman (44.4% vs 3.7%, padj=0.04); and 7 Black women had a mutation in PRAMEF14 compared to 1 White woman (38.8% vs 3.7%, padj= 0.06). Conclusion GON4L, which is a transcription factor associated with decreased apoptosis and promotion of tumor growth through CD24 regulation, was significantly more mutated in the EC genome of Black women relative to White women. Somatic mutation and transcriptional changes are seen between tumor grade and self-identified race and require further exploration for its relationship to aggressive disease. Citation Format: Alex P. Abstract:
Influenza is a yearly threat to global public health. Rapid changes in influenza surface proteins resulting from antigenic drift and shift events make it difficult to readily identify antibodies with broadly neutralizing activity against different influenza subtypes with high frequency, specifically antibodies targeting the receptor binding domain (RBD) on influenza HA protein. We developed an optimized computational design method that is able to optimize an antibody for recognition of large panels of antigens. To demonstrate the utility of this multistate design method, we used it to redesign an antiinfluenza antibody against a large panel of more than 500 seasonal HA antigens of the H1 subtype. As a proof of concept, we tested this method on a variety of known antiinfluenza antibodies and identified those that could be improved computationally. We generated redesigned variants of antibody C05 to the HA RBD and experimentally characterized variants that exhibited improved breadth and affinity against our panel. C05 mutants exhibited improved affinity for three of the subtypes used in design by stabilizing the CDRH3 loop and creating favorable electrostatic interactions with the antigen. These mutants possess increased breadth and affinity of binding while maintaining high-affinity binding to existing targets, surpassing a major limitation up to this point.
