Abstract:
Glioblastomas (GBMs) are one of the most malignant brain tumors in adults. This is partly due to the potential presence of so-called glioma stem-like cells (GSCs), which are characterized by the expression of stemness markers and a resistance to radio- and chemotherapy. Previous work from us showed that after combination treatment of GSCs with arsenic trioxide and gossypol, the protein BRCA1-associated ATM-activator 1 (BRAT1) was one of the most downregulated proteins. This protein is largely undescribed, but it has been shown to regulate DNA damage signaling through interaction with ATM, BRCA1, and DNA-PKcs in initial stages of DNA damage response. An unpublished analysis of The Cancer Genome Atlas and The Human Protein Atlas databases showed an increased expression of BRAT1 in GBMs compared to healthy tissues and that an increased expression is negatively correlated with patient survival. Due to these findings, our goal is to analyze the radio-sensitizing effect of BRAT1 on FCS-grown (i.e., differentiated) highly radio-resistant GBM cells and GSCs. Here, using stable knockdowns of BRAT1, we show that it is needed for effective DNA repair after irradiation using a γH2AX-foci assay, whereas it is dispensable for cellular proliferation. A cell death analysis using Annexin V/propidium iodide staining revealed a first hint that BRAT1 downregulation sensitizes GBM cells to irradiation. Moreover, through immunofluorescent staining, we showed that BRAT1 is needed for BRCA1 recruitment to DNA damage sites. Future experiments will aim at systematically analyzing the downstream effects of BRAT1 depletion and to determine further interactors. Thus, we hope to gain a deeper understanding of the mechanism of radio-resistance in GSCs, also in order to individually determine the effectiveness of radiotherapy.

Abstract:
ObjectiveTo investigate the pathogenicity of a novel homozygous BRAT1 variant in 2 siblings with nonprogressive cerebellar ataxia (NPCA) through functional studies on primary and immortalized patient cell lines.MethodsBRAT1 protein levels and ataxia-telangiectasia mutated (ATM) kinase activity in patient-derived and control cell lines were assessed by Western blotting. The impact of the novel BRAT1 variants on mitochondrial function was also assessed, by comparing patient and control cell lines for rates of oxygen consumption and for phosphorylation (S293) of the E1⍺ subunit of pyruvate dehydrogenase (PDH).ResultsTwo male siblings with NPCA, mild intellectual disability, and isolated cerebellar atrophy were found to be homozygous for a c.185T>A (p.Val62Glu) variant in BRAT1 by whole exome sequencing. Western blotting revealed markedly decreased BRAT1 protein levels in lymphocytes and/or fibroblast cells from both affected siblings compared to control cell lines. There were no differences between the patient and control cells in ATM kinase activation, following ionizing radiation. Mitochondrial studies were initially suggestive of a defect in regulation of PDH activity, but there was no evidence of increased phosphorylation of the E1⍺ subunit of the PDH complex. Measurement of oxygen consumption rates similarly failed to identify differences between patient and control cells.ConclusionsBiallelic pathogenic variants in BRAT1 can be associated with NPCA, a phenotype considerably milder than previously reported. Surprisingly, despite the molecular role currently proposed for BRAT1 in ATM regulation, this disorder is unlikely to result from defective ATM kinase or mitochondrial dysfunction.

Abstract:
Lethal neonatal rigidity and multifocal seizure syndrome (RMFSL) is a severe autosomal recessive epileptic encephalopathy characterized by rigidity, intractable multifocal seizures, microcephaly, apnea, and bradycardia immediately after birth. RMFSL is related to a mutation in breast cancer 1-associated ataxia telangiectasia mutated activation-1 protein (<i>BRAT1</i>). We report a case of a female infant born to non-consanguineous Korean parents who developed hypertonia, dysmorphic features, progressive encephalopathy with refractory seizures at birth, and worsening intermittent apnea, leading to intubation and death at 137 days of age. The initial repeated electroencephalographic findings were normal; however, a pattern of focal seizures emerged at 35 days of life. Rapid trio whole-exome sequencing revealed heterozygous mutations c.1313_1314delAG p.(Gln438Argfs*51) and c.1276C>T p. (Gln426*) in <i>BRAT1</i>. After genetic counseling for pregnancy planning, a preimplantation genetic diagnosis for targeted <i>BRAT1</i> mutations was successfully performed, and a healthy baby was born. To our knowledge, this is the first reported case of a Korean patient with compound heterozygous mutations in <i>BRAT1</i>. An early and accurate genetic diagnosis can help provide timely treatment to patients and indicate the need for reproductive counseling for parents for family planning.

W konkluzji autor doszedł do wniosku, że dla obojga poetów – choć w różny sposób i w różnym stopniu - postać Brata Alberta nie stanowi jednak autonomicznego tematu i problemu twórczości, ale jest raczej traktowana jako pretekst dla ich własnych przemyśleń i poszukiwań artystycznych.

Abstract:
Brat Albert Chmielowski stał się bohaterem kilku ważnych tekstów literackich, w tym jest bohaterem dwóch dramatów powstałych prawie w tym samym czasie – podczas II wojny światowej. Są to Adama Bunscha Gołębie Brata Alberta (1943), (wydanie polskie z roku 1947 Przyszedł na ziemię święty) oraz Karola Wojtyły pt. Brat naszego Boga (1944-1950). Zarówno Bunsch, jak i Wojtyła są zafascynowani postacią Brata Alberta i jego życiem. Bunsch pisze swój dramat jako żołnierz, artysta-malarz, literat, a Wojtyła, przyszły papież, jako ksiądz. W dramacie Bunscha mamy pokazaną drogę wyborów życiowych Adama Chmielowskiego, który szuka swego miejsca w życiu. Widzimy go najpierw jako żołnierza-powstańca z roku 1863, potem jako malarza, a w końcu jako zakonnika posługującego ubogim. Dramat Bunscha zbliżony jest do dramatów realistycznych, obyczajowych czy biograficznych. Autor nie wprowadził do niego treści teologicznych, tylko położył nacisk na kwestie etyczne. Natomiast w dramacie Karola Wojtyły mamy inne podejście do postaci Brata Alberta. Sztuka ta oparta jest na fundamencie personalizmu. Stawia on w centrum człowieka z jego tajemnicą istnienia w relacji do Boga opartej na miłości. Fabuła staje się pretekstem dla rozważań na temat Miłosierdzia, będącego czystym Pięknem. Wizualizacją Miłosierdzia w omawianej sztuce jest obraz Chmielowskiego Ecce homo. Dramat Wojtyły ma wymowę teologiczną. Jego teatr jest teatrem słowa, myśli, filozoficzno-wizyjnym. Tak więc pomimo znaczących różnic w koncepcji teatru i dramatu oba dramaty są świadectwem roli, jaką brat Albert odegrał we współczesnym świecie.

O osobach niepełnosprawnych mówi się i pisze w dzisiejszych czasach, choć wciąż zbyt mało porusza się problemów dotyczących tej grupy. Sytuacja niepełnosprawnego rodzeństwa dzieci została prawie zapomniana. Jednak to sprawne rodzeństwo niepełnosprawnego brata lub siostry od najwcześniejszych lat konfrontowane jest z ludzkimi cierpieniami, a często z niedostatkiem emocjonalnym rodziców. Często stają przed problemem podejmowania decyzji, które często determinuje fakt posiadania niepełnosprawnego brata lub siostry. Kontakt z niepełnosprawną siostrą lub bratem może w znaczący sposób doprowadzić dziecko sprawne do dojrzałości, wrażliwości, odpowiedzialności, tolerancji, altruizmu i zaangażowania społecznego. Wszystko zależy od odpowiedniego traktowania niepełnosprawnego i sprawnego dziecka przez rodziców. Jeśli patenty nie zaniedbują ani nie przeciążają sprawnego dziecka, można uniknąć większości konfliktów, a niepełnosprawna siostra lub brat odniosą korzyści z obecności ich sprawnego rodzeństwa.

Postać Brata Alberta wpisała się w pejzaż intelektualny pierwszych dekad XX w., inspirując modernistycznych pisarzy do kreowania bohaterów na wzór tego Wielkiego Jałmużnika. Młodopolscy pisarze widzieli w nim wzorzec etyczny swojej epoki, w perspektywie jego duchowości rozważali jej dylematy światopoglądowe. Osoba Brata Alberta nabrała też szczególnego znaczenia po odzyskaniu przez Polskę niepodległości. Od drugiej dekady XX w. znacznie zintensyfikowana została publicystyczna aktywność mająca na celu upowszechnienie postawy i znaczenia Brata Alberta w polskim życiu społecznym czasu zaborów. W artykule prześledzony został właśnie sposób jego postrzegania oraz kategorie interpretacji życia i przemiany tego artysty w jałmużnika, które ukierunkowane zostały na ukazanie jego postaci jako integralnie związanej z przemianami polskiej świadomości po klęsce powstania styczniowego. Z analizy dość obszernego materiału źródłowego wynika, że Brat Albert – powstaniec przechodzący głęboką wewnętrzną przemianę – postrzegany był nie tylko jako dwudziestowieczne wcielenie św. Franciszka z Asyżu, jako wielki człowiek Kościoła, tworzący kanony etyczne dla swojej epoki, ale przede wszystkim jako unaoczniający w swojej postawie ewolucję polskiej świadomości kulturowej drugiej połowy XIX oraz początków XX w. W perspektywie rozważań o jego życiu scalała się polska biografia kulturowa tego czasu, odsłaniała się konstytuująca ją logika przenoszenia romantycznej „egzaltacji patriotycznej” na płaszczyznę marzeń o chrześcijańskiej doskonałości. Sylwetka Brata Alberta stała się w początkach XX w. istotnym elementem w polskim dyskursie tożsamościowym.

The character of Adam Chmielowski St. Brother Albert in the film Our God’s brother directed by Krzysztof Zanussi The film Our God’s Brother, directed by Krzysztof Zanussi in 1997, is the adaptation of Karol Wojtyła’s drama of the same title. Adam Chmielowski, who was later known as St. Brother Albert, is the main character in the film. He lived between 1845 and 1916 in the times of the industrial revolution, which led many people to misery. Brother Albert was an artist and painter by profession. Feeling that he was called by God, he decided to give up painting and he started helping the homeless. In his film, Krzysztof Zanussi shows Brother Albert’s path to holiness and the roots of his heroism. The article consists of five points. The first one focuses on the characteristics and structure of the film. The second one briefly shows the story of Adam Chmielowski’s life. In the third part the play by Karol Wojtyła entitled Our God’s Brother is described. The fourth part shows how Krzysztof Zanussi adapted the play into a film, and it also presents a film portrayal of St. Brother Albert. Finally, the conclusion shows the essence of the main character’s holiness. The film can be divided into two parts. In the first one Adam Chmielowski discerns his vocation. In the second one, after his decision to serve the homeless, his vocation develops and he becomes a friar. His service is based on his trust to the Creator and him following merciful Christ. It leads to the fullness of his humanity, freedom and happiness.

Genome evolution is partly driven by the mobility of transposable elements (TEs) which often leads to deleterious effects, but their activity can also facilitate genetic novelty and catalyze local adaptation. We explored how the intraspecific diversity of TE polymorphisms might contribute to the broad geographic success and adaptive capacity of the emerging oil crop Thlaspi arvense (field pennycress). We classified the TE inventory based on a high-quality genome assembly, estimated the age of retrotransposon TE families and comprehensively assessed their mobilization potential. A survey of 280 accessions from 12 regions across the Northern hemisphere allowed us to quantify over 90,000 TE insertion polymorphisms (TIPs). Their distribution mirrored the genetic differentiation as measured by single nucleotide polymorphisms (SNPs). The number and types of mobile TE families vary substantially across populations, but there are also shared patterns common to all accessions. Ty3/Athila elements are the main drivers of TE diversity in T. arvense populations, while a single Ty1/Alesia lineage might be particularly important for transcriptome divergence. The number of retrotransposon TIPs is associated with variation at genes related to epigenetic regulation, including an apparent knockout mutation in BROMODOMAIN AND ATPase DOMAIN-CONTAINING PROTEIN 1 (BRAT1), while DNA transposons are associated with variation at the HSP19 heat shock protein gene. We propose that the high rate of mobilization activity can be harnessed for targeted gene expression diversification, which may ultimately present a toolbox for the potential use of transposition in breeding and domestication of T. arvense.

Assisted reproduction technologies (ARTs) are generally considered safe; however, emerging evidence highlights the need to evaluate potential risks in adulthood to improve safety further. ART procedures like rederivation of embryos by vitrification differ from natural conditions, causing significant disparities between in vitro and in vivo embryos, affecting foetal physiology and postnatal life. This study aims to investigate whether hepatic transcriptome and metabolome changes observed postnatally are already present in foetal livers at the end of gestation. This study compared fresh and vitrified rabbit embryos, finding differences between foetuses obtained by the transfer of fresh and vitrified embryos at 24 days of gestation. Rederived embryos had reduced foetal and liver weights and crown-rump length. However, the offspring of vitrified embryos tended to be born with higher weight, showing compensatory growth in the final week of gestation (59.2 vs. 49.8 g). RNA-Seq analysis revealed 43 differentially expressed genes (DEGs) in the foetal liver of vitrified embryos compared to the fresh group. Notably, downregulated genes included BRAT1, CYP4A7, CYP2B4, RPL23, RPL22L1, PPILAL1, A1BG, IFGGC1, LRRC57, DIPP2, UGT2B14, IRGM1, NUTF2, MPST, and PPP1R1B, while upregulated genes included ACOT8, ERICH3, UBXN2A, METTL9, ALDH3A2, DERPC-like, NR5A2-like, AP-1, COG8, INHBE, and PLA2G4C. Overall, a functional annotation of these DEGs indicated an involvement in lipid metabolism and the stress and inflammatory process or immune response. Thus, our results suggest that vitrification and embryo transfer manipulation induce an adaptive response that can be observed in the liver during the last week of gestation.

Abstract The study, dedicated to the 100th anniversary of the birth of Vincent Blanár, offers a comparative Czech-Slovak view of the lexeme brother (Czech bratr – Slovak brat) in its paradigmatic relations. The analysis is based on a specific textual basis, the novel Bratrstvo (Brotherhood) by the Czech writer Alois Jirásek. The starting point of the comparison is the Czech original and two Slovak translations of this literary text, which shows a remarkably wide range of semantic concretizations of the polysemic word brother. In the Czech-Slovak comparison, various morphosyntactic, lexical-semantic, derivational and collocational differences appear. In a comparative view, the sphere of addressing is particularly interesting.

The reality show "Big Brother" represents a metaphor for the transformation of contemporary media toward its commercialization and the primate which is given to entertainment as opposed to classic informative and educational content, which, in Serbia, takes place in the context of post-socialist transformation. The hyper commercialized character of "Big Brother" is the product of a new guiding idea in the media sphere, which is to measure success only on the criteria of ratings. In all versions of "Big Brother" (the "ordinary" as well as VIP), the framework of the reality show is made up of two elements: the phenomenon of surveillance and the commercial principle of the programme. In this paper we wanted to take these two themes - surveillance and commercialization - and connect them through the concepts of the "panopticon"/"synopticon" and "ratings". In the analysis and discussion we will show that the newfangled criteria of "ratings" is directly dependent on the situation in which the viewers of the reality format find themselves, situations in which "many view and decide on the fate of the few". We will analyze the ways in which the very production of the reality show is geared toward influencing the ratings, through interventions and the direction done by the production team, but also the reception of the show in specialized forums. In the second case, a netography of the reception of VIP BB 2013 was undertaken, taking into account the discussions in the forum and the monitoring of YouTube videos. Based on this, we singled out the most viewed moments in the reality show, and considered the reasons why certain events and/or actors who participated in them garnered special attention among viewers, thus increasing the ratings of the show, but also multiplicating the commercial effect through the participation of viewers through different media platforms (websites, forums, online votes etc.)

Cardamonin, the chalcone class, is one of the natural components from the spicy herbaceous plant (Alpinia conchigera Griff) and has anticancer activities in many human cancer cell lines. There is, however, no information to show that cardamonin induces cell apoptosis and alters apoptosis associated gene expressions in mouse leukemia cells. Thus, we investigated the effects of cardamonin on the apoptotic cell death and associated gene expression in mouse leukemia WEHI-3 cells in vitro. Results indicated that cardamonin decreased total viable cell number via induced cell morphological changes and apoptotic cell death in WEHI-3 cells that were assay by contrast-phase microscopy and flow cytometry examinations, respectively. The flow cytometry assay indicated that cardamonin increased reactive oxygen species (ROS) and Ca[Formula: see text] production, decreased the levels of mitochondrial membrane potential ([Formula: see text] and increased caspase-3, -8 and -9 activities in WEHI-3 cells. Western blotting was performed to analyze expression of relevant pro- and anti-apoptotic proteins and results showed that cardamonin decreased anti-apoptotic protein of Bcl-2 but increased pro-apoptotic protein of Bax in WEHI-3 cells. Furthermore, cardamonin increased cytochrome c, AIF and Endo G release, increased GRP78, caspase-12 that were associated with ER stress and increased Fas, Fas-Ligand and FADD expression. Furthermore, cardamonin increased the gene expressions of DAP (death-associated protein), TMBIM4 transmembrane (BAX inhibitor motif containing 4), ATG5 (autophagy related 5) but decreased the gene expression of DDIT3 (DNA-damage inducible transcript 3), DDIT4 (DNA-damage-inducible transcript 4), BAG6 (BCL2-associated athanogene 6), BCL2L13 [BCL2-like 13 (apoptosis facilitator)] and BRAT1 (BRCA1-associated ATM activator 1) that are associated with apoptosis pathways. Based on those findings, we may suggest cardamonin induced apoptotic cell death through Fas and Fas-Ligand-, caspase- and mitochondria-dependently pathways and also affects the apoptotic gene expression in WEHI-3 cells in vitro.

Tri Hita Karana culture adopts a philosophy that includes Parhyangan, Pawongan and Palemahan. LPD growth adopts M. Porter's industrial growth strength model which includes: threat of new entrants, supply strength of suppliers, supply strength of buyers, threat of substitution products, and competition. Data collection uses questionnaire techniques and is analyzed with regression analysis and moderator regression analysis. The results of the data analysis show that leadership has a positive and significant effect on LPD growth, and THK culture strengthens the influence of leadership on LPD growth in Bali.

The various of brata that should be done are: no eating and drinking (Upavasa), not talking (Mauna Brata), and not sleeping (Jagra Brata). Each brata has it owns religious and ethical values. So far, Brata Śivalatri has been done because apart from spiritual cleansing, it also has a positive infact on worldly life, which can be implemented in real life, especially during the covid -19 outbreak. The implementation of Brata Śivalatri in society is being able to maintain the health; peace; and increase alertness and awareness. It happened because doing the Brata Śivalatri can control lust.

The DNA damage response (DDR) is crucial for genomic integrity. BRIT1 (breast cancer susceptibility gene C terminus-repeat inhibitor of human telomerase repeat transcriptase expression), a tumor suppressor and early DDR factor, is recruited to DNA double-strand breaks (DSBs) by phosphorylated H2A histone family, member X (γ-H2AX), where it promotes chromatin relaxation by recruiting the switch/sucrose nonfermentable (SWI–SNF) chromatin remodeler to facilitate DDR. However, regulation of BRIT1 recruitment is not fully understood. The baculovirus IAP repeat (BIR)-containing ubiquitin-conjugating enzyme (BRUCE) is an inhibitor of apoptosis protein (IAP). Here, we report a non-IAP function of BRUCE in the regulation of the BRIT1–SWI–SNF DSB-response pathway and genomic stability. We demonstrate that BRIT1 is K63 ubiquitinated in unstimulated cells and that deubiquitination of BRIT1 is a prerequisite for its recruitment to DSB sites by γ-H2AX. We show mechanistically that BRUCE acts as a scaffold, bridging the ubiquitin-specific peptidase 8 (USP8) and BRIT1 in a complex to coordinate USP8-catalyzed deubiquitination of BRIT1. Loss of BRUCE or USP8 impairs BRIT1 deubiquitination, BRIT1 binding with γ-H2AX, the formation of BRIT1 DNA damage foci, and chromatin relaxation. Moreover, BRUCE-depleted cells display reduced homologous recombination repair, and BRUCE-mutant mice exhibit repair defects and genomic instability. These findings identify BRUCE and USP8 as two hitherto uncharacterized critical DDR regulators and uncover a deubiquitination regulation of BRIT1 assembly at damaged chromatin for efficient DDR and genomic stability.

Village credit institutions or known as LPDs are microfinance institutions that have a very important position in traditional villages. Village credit institutions have the authority to implement their management based on the respective traditional village pararem. Given the important position of village credit institutions in improving the welfare of local communities, village credit institutions must be improved as a financial institution. Given the number of problematic village credit institutions in Tabanan Regency caused by many mismanagement of the leadership, this study was conducted to see the influence of Asta Brata's leadership on employee satisfaction at village credit institutions (LPD) in Tabanan Regency. This study used a sample of 93 people from a total population of 1190 people. Sampling using the Proportionate Stratified Random Sampling method. The data analysis technique uses path analysis. Collecting information from respondents using a questionnaire with a Likert scale. Data from respondents is still in ordinal scale units and the data processing requirements used are path analysis, so the data must be increased to intervals using the interval successive method.The results of this study are to answer the proposed hypothesis. The results showed that the leadership of Asta Brata with subvariables Indra Brata, Yama Brata, Surya Brata, Candra Brata, Bayu Brata, Kuwera Brata, Baruna Brata and Agni Brata had a positive and significant effect on employee job satisfaction at village credit institutions in Tabanan Regency. Asta Brata with the greatest influence on job satisfaction at village credit institutions (LPD) in Tabanan Regency is Indra Brata. If you want to increase job satisfaction in village credit institutions (LPD) in Tabanan Regency, you need to improve Indra Brata's leadership first and then the others.

Abstract:
Tulisan ini membahas tentang gaya kepemimpinan Asta Brata untuk diterapkan di era VUCA, dengan menggunakan metode penelitian kualitatif dengan tahap pengumpulan data, analisis dengan metode deskriptif-analitik, interpretasi, dan dilanjutkan tahap penulisan menjadi sebuah karya. Masalah yang di angkat dalam penelitian ini adalah bagaimana konsep Asta Brata dapat diterapkan sebagai pedoman kepemimpinan di era VUCA. Permasalahan tersebut dijawab dengan menjabarkan pengertian VUCA, pengertian Asta Brata, dan analisis Asta Brata sebagai pedoman kepemimpinan di era VUCA. Melalui kajian yang dilakukan, ditemukan bahwa terdapat sudut pandang lain dari VUCA yakni VUCA Prime yang bermakna positif. VUCA Prime terdiri atas (Vision, Undertsanding, Clarity, and Agile). Ditemukan juga Asta Brata merupakan konsep kepemimpinan ideal yang didasari peneladanan terhadap sikap unsur dalam alam yakni bumi, air, angin, bulan, matahari, samudra, gunung, dan api. Setelah dilakukan analisis ditemukan bahwa Asta Brata dapat diterapkan sebagai pedoman yang harus dimiliki pemimpin untuk mencapai VUCA Prime. Penerapannya adalah sebagai berikut: sikap air dan bumi dapat digunakan untuk mencapai Vision, sikap lautan dan bulan untuk mencapai Understanding, sikap angin dan api untuk mencapai Clarity, serta sikap gunung dan matahari dapat diterapkan untuk mencapai Agile. Berdasarkan temuan tersebut dapat diambil kesimpulan bahwa Asta Brata dapat dijadikan alternatif konsep kepemimpinan di era VUCA. This study examines Asta Brata's leadership style for the VUCA age. Utilizing qualitative research methodologies with data gathering phases, descriptive-analytic analysis, interpretation, and extending the writing step into work. The issue presented in this study is how to apply the Asta Brata idea as a leadership guide in the VUCA age. This issue is addressed by defining VUCA, Asta Brata, and analyzing Asta Brata as a guideline for leadership in the VUCA age. According to the research, another point of view from VUCA, VUCA Prime, has a positive significance. VUCA Prime is made from (Vision, Understood, Clarity, and Agile). It was also discovered that Asta Brata is an ideal leadership concept based on the attitudes of nature's elements, including earth, water, wind, moon, sun, ocean, mountains, and fire. Following an examination, Asta Brata may be used as a guideline for leaders to reach VUCA Prime. Water and earth attitudes can be used to gain Vision; ocean and moon attitudes can be used to gain Comprehension; wind and fire attitudes can produce Clarity; and mountain and sun attitudes can achieve Agile. Asta Brata, based on these findings, can be employed as an alternative leadership idea in the VUCA age.
