Full textThese are exciting times in the world of computer animation. Individual artists can now produce animations which in the past required teams of artists and programmers to create, but the obstacles to an individual artist can still be daunting in terms of required manual input or technical skills. The goal of this thesis is to demonstrate a simple software tool (BRAT - Blender Rapid Animation Tool) which allows an artist to create cartoon-like 3D animations using the artist's 2D drawings, at the touch of a button. It could be useful for any artist producing 3D animations. The tool is a Python script addon to the Blender 3D modeling system. It is a high level program which links to Blender's Python API library of low level calls to automate the manual work normally required by 3D artists to create an animation. The output of BRAT is a 3D animation file showing a character traversing a rectangular matrix of buildings.
This thesis compares the manual way of creating such an animation with the BRAT automated process. The comparison explains the benefits of using BRAT. This thesis also discusses the design of BRAT and compares it to some other existing 3D software tools whcih also attempt to simplify the 3D animation production process. The conclusion discusses some future enhancements to make BRAT more useful.

Full textThe advent of new sequencing techniques has dramatically increased the diagnostic rate of intellectual disability (ID), and more than 2,000 genes are currently known to be involved. Despite these considerable progresses, interpreting the variants identified by sequencing methods remains challenging, and the natural history of newly described ID is often poorly understood. To better understand these disorders and their underlying mechanisms, we have studied four rare forms of ID with various inheritance patterns from both clinical and genetic perspectives. On one hand, we defined the clinical pictures associated with variations in BRAT1, CNOT3 and MTOR, and we investigated the existence of any phenotype-genotype correlations. On the other hand, we contributed to the design of a functional test to reclassify PQBP1 variants of uncertain significance

Full textManuell segmentering av hjärntumörer är en tidskrävande process, segmenteringarna är ofta varierade mellan experter, och automatisk segmentering skulle vara användbart för kliniskt bruk. Den här rapporten undersöker användningen av deep convolutional networks (ConvNets) för automatisk segmentering av gliom i MR-bilder. De implementerade nätverken utvärderas med hjälp av data från brain tumor segmentation challenge (BraTS). Studien finner att 3D-nätverk har generellt bättre resultat än 2D-nätverk, och att de bästa nätverken har förmågan att ge segmenteringar som liknar mänskliga segmenteringar. ConvNets utvärderas också som feature extractors, med linjära SVM som klassificerare. Den här metoden ger segmenteringar med hög känslighet, men är också till hög grad översegmenterade. Vikten av att ha mer träningsdata undersöks också genom att träna på två olika stora dataset, men metoden för att få fram de riktiga segmenteringarna har troligen också stor påverkan på resultatet. Nätverken slår inte de tidigare rekorden på BraTS, men flera viktiga men enkla förbättringsområden är identifierade som potentiellt skulle förbättra resultaten.

Full textThe objective of this thesis is to study three novels written by English-speaking authors of Pakistan or India, namely Bapsi Sidhwa, Bharati Mukherjee and Hanif Kureishi. One might be tempted to place the three writers of this study in the category of "literature of immigrants." They all write at a time of mass migration when the idea of "cultural shock" among Western peoples begins to be more evident. All three writers are affected by themes which appear only marginally in the debate evoked above, much of the emphasis being on the cultural and social difficulties of women in Indo-Pakistani society. As for Kureishi, the polarization mentioned above assumes a very different emphasis, involving the situation of an Asian born and brought up inside Western society. Within this overall assessment of the ideological and historical context common to all three writers, it will thus be important to examine the specific attitudes adopted by each writer in relation to his or her own personal experience. The main focus of this study will therefore be thematic, centering on these writers’ specific preoccupations and the way this is seen in their peculiar depiction of the tensions at stake

Bone morphogenetic protein induces signals through the stimulation of serine/threonine kinase receptors which phosphorylated BMP signaling mediators, Smad1, Smad5 and Smad4. The phosphorylated Smad1/5/4 complex is then translocated to the nucleus and regulates the BMP target genes. BMP receptor associated molecule 1 (BRAM1) is a molecule that associates with BMP receptor IA. However its role in BMP signaling is still unclear. Thus, the specific aims of this study are examined (1) if BRAM1 affects BMP signaling; (2) If so, are the Smad proteins classical BMP signaling mediators involved? In this study, we demonstrate that BRAM1 interacts with BMP receptor by co-immunoprecipitaion, Western blot and confocal microscopy. The results show that BRAM1 associates with wild type BMP receptor as well as the constitutively activated BMPR. The Mynd domain of BRAM1 is the main region for BRAM1-BMPR interaction. Confocal microscopy study showed that BRAM1 co-localized with BMPR and Smad5, but not with Smad1. Furthermore, BRAM1 prevents the nucleus translocation of Smad4 in response to BMP treatment. Thus the results suggest that BRAM1 may be involved in BMP signaling. This study will provide more information regarding the function of BRAM1.

Full textToy industry in the US is product driven and full of challenges. This case presents an overview of the California-based Micro Games of America (MGA) Entertainment, which is a consumer entertainment products company, engaged in innovative lines of proprietary and licensed products including toys and games, dolls, consumer electronics, home decor, stationery and sporting goods. It had more than 200 licences. In 2001, MGA launched a fashion doll called ‘Bratz’, and it sold 150 million Bratz dolls all over the world. Bratz line surpassed the legendary brand ‘Barbie’—Mattel, Inc.’s flagship brand—in a short span of time through many innovative marketing strategies and different product placement, roll-outs, tie-ins and other promotional tactics, despite facing many challenges, such as, fast-changing demographics, shorter product life cycle (PLC) and negative perceptions about brand. This case can be used to address two issues: first, structural change drivers and trends that shaped the toy industry in developed economies and, second, how to develop effective marketing strategies for product with shorter PLC in highly product-driven market?

BRAM1, bone morphogenetic protein receptor (BMPR) associated molecule 1, is a cytoplasmic protein consisting of 198 amino acids. BRAM1 shares 99% homology with the MYND domain-containing carboxy terminus of a tumor suppressor protein, BS69, which interacts with adenovirus E1A and Epstein-Barr virus-encoded nuclear protein EBNA2 through the MYND domain. Interaction of BS69 with E1A or EBNA2 inhibits viral oncoprotein-mediated transactivation. On the other hand, BRAM1 is a cytoplasmic protein and interacts with BMPR. To date, the biological role of BRAM1 remains unclear. In this thesis work, we evaluate the physiological role of BRAM1 in zebrafish model system. zebrafish bram 1 cDNA was generated by bioinformatic search of zebrafish EST database, followed by 5’ and 3’ RACE analysis. We demonstrate that zebrafish BRAM1 associates with C-terminal region of zebrafish BMP receptor through MYND domain in vitro and in vivo. Interaction between zebrafish BRAM1 and zebrafish BMP receptor blocked BMP signaling pathway. In zebrafish, bram1 is a maternal factor and its expression profile is coordinated with bmp4 and bmp receptor in early developmental stage. bram1 was expressed throughout the embryos before blastula stage as examined by whole mount in-situ hybridization analysis, and was expressed ubiquitously in different tissues such as brain, heart, muscle, ovary, and especially ovary as examined by northern blot assay. Using antisense morpholinos to knock down BRAM1 resulted in defects in heart looping and curly tails. This phenotype of bram1 morphants was similar to phenotype of mutants involved in Bmp signaling. Injection of constitutively active bmp receptor mRNA resulted in defects in heart looping. Overexpression of BRAM1 reversed the heart position from no looping (N-looping) to rightward looping (D-looping). Further analysis of zebrafish left-right asymmetry genes incluidng cyclops, southpaw, lefty1, lefty2, and pitx2c demonstrate that BRAM1 affects the zebrafish left-right asymmetry. As zebarfish midline is critical for development of left-right asymmetry, we hypothesize that BRAM1 may have earlier effect in regulating midline. Indeed, we show that expression of flh and gsc in bram1 morphants is reduced. These findings indicate that BRAM1 may function as a negative regulator of Bmp signaling to participate in the midline development during early cell fate decision. As Bmp signaling involves interaction of BMP and BMPR, and its downstream Smad mediators, BRAM1 may affect the BMP signaling by inhibiting Smad signaling. As tested in BALB/c3T3 and COS7 cells, mammalian BRAM1 (mBRAM1) inhibit Smad signalling pathway. mBRAM1 interacted with Bmp receptor type IA through MYND domain in the absence or presence of Bmp2 treatment. mBRAM1 interacted with Smads proteins, especially Smad4 and Smad5. Such interactions blocked the nuclear translocation of Smad4 or Smad5 in Bmp2-mediated manner. Thus, mBRAM1 inhibits Smad signaling through preventing Smad4 and Smad5 to translocate into nucleus. In conclusion, these results suggest that BRAM1 may function as a negative regulator of BMP-mediated biological functions.

Latent membrane protein 1 (LMP1), a major oncoprotein encoded by Epstein-Barr virus (EBV), has been shown to be involved in cellular signaling pathways. In this thesis, we tried to identify new targets of LMP1 signaling that may help to explain the role of LMP1 in non-lymphocyte cell transformation. Using yeast two-hybrid screening method with the C-terminal sequence of LMP1 as the bait, we found that BMP receptor-associated molecule 1 (BRAM1) associated with LMP1 in vitro and in vivo. This finding was supported by cotransfection experiments and examined by immunoassays and confocal microscopy. The association involves mainly the C-terminal half, the MYND domain, of BRAM1 and the CTAR2 region of LMP 1. The latter is critical for LMP1-mediated NF-kB and JNK signaling pathways, suggesting the role of BRAM1 in LMP1 signaling. To test this hypothesis, we examined the effect of BRAM1 on the induction of NF-kB and JNK activation by LMP1. We demonstrate that BRAM1 interferes with LMP1-mediated NF-kB activation in BALB/c3T3 cells. Since LMP1-induced NF-kB activation through the NIK-IKK-IkB pathway, we thus examined the protein level of total and phosphorylation form of IkBa in the presence and absence of BRAM1. The results indicated that BRAM1 decreased the phosphorylation of IkBa and delayed the degradation of total IkBa in the presence of LMP1. Since the CTAR2 region interacts with the TNF-a receptor-associated death domain protein (TRADD), it is interesting to find that BRAM1 also interferes with NF-kB activation mediated by TNF-a. Since the CTAR2 region of LMP1 is critical for LMP1-mediated JNK activation, we examined the effect of BRAM1 on the JNK activation mediated by LMP1, TNF-a treatment and overexpression of LTbR. We found that BRAM1 plays a role either in LTbR-mediated or in TNF-a mediated JNK activation but not in LMP1-mediated JNK activation. To further examine the role of BRAM1 in LMP1 signaling, we observed that BRAM1 disrupts the resistance of LMP1-expressing cells to TNF-a-induced cytotoxicity. Thus, we propose that BRAM1, a molecule uniquely associated with LMP1, functions as a negative regulator of LMP1-mediated biological functions.

These considerations lead naturally to stabilised vector-Pade approximation of a vector-valued function (VPAStab), and an algorithm for the acceleration of convergence of a linearly generated sequence of vectors. A generalisation of this algorithm for the acceleration of convergence of a nonlinearly generated system is proposed here, and comparative numerical results are given.
