Abstract:
Adeno-associated virus (AAV) capsid assembly occurs in the nucleus. Newly synthesized capsid proteins VP1, VP2 and VP3 contain several basic regions (BRs), which may act as nuclear localization signals (NLSs). Mutation of BR2 and BR3, located at the VP1 and VP2 N termini, marginally reduced nuclear uptake of VP1 or VP2, but not of VP3, when expressed in the context of the whole AAV type 2 (AAV2) genome. Combined mutation of BR1, BR2 and BR3 resulted in capsids with slightly reduced amounts of VP1. Expression of isolated VP1/2 N termini revealed an influence of BR3 on nuclear transport, whilst BR1 or BR2 had no effect. However, deletion of an N-terminal fragment in front of the BR elements strongly reduced nuclear uptake of VP1/2 N termini. Mutation of BR4, present in all three capsid proteins, led to their retention in the cytoplasm and to the formation of speckles, resulting in a lack of capsid formation and a significant reduction in VP levels. In a VP fragment comprising BR2, BR3 and BR4, the BR4 element was not necessary for nuclear localization. Mutation of BR5 in the C-terminal part of the VPs resulted in a speckled protein distribution in the nucleus, strongly reduced capsid assembly, and low VP1 and VP2 levels. Taken together, these results showed that BR2 and BR3 have a weak influence on nuclear transport of VP1 and VP2, whilst combined mutation of BR1, BR2 and BR3 influences the stoichiometry of VPs in assembled capsids. BR4 and BR5 play a crucial role in capsid assembly but have no NLS activity.

Abstract:
miR319 was the first plant miRNA discovered via forward genetic mutation screening. In this study, we found that miR319 family members had similar sequences but different expression patterns in Brassica campestris and Arabidopsis thaliana. RT-PCR analysis revealed that Bra-MIR319a and Bra-MIR319c had similar expression patterns and were widely expressed in plant development, whereas Bra-MIR319b could only be detected in stems. The overexpression of each Bra-MIR319 family member in Arabidopsis could inhibit cell division and function in leaf and petal morphogenesis. Bra-miR319a formed a new regulatory relationship after whole genome triplication, and Bra-MIR319a overexpressing in Arabidopsis led to the degradation of pollen content and affected the formation of intine, thereby causing pollen abortion. Our results suggest that Bra-MIR319 family members have functional similarity and difference in plant development.

Abstract:
The composition of a Brazilian green propolis ethanolic extract (Et-Bra) and its effect onTrypanosoma cruzitrypomastigotes and other pathogenic microorganisms have already been reported. Here, we further investigated Et-Bra targets inT. cruziand its effect on experimental infection of mice. The IC50/4 days for inhibition of amastigote proliferation was 8.5 ± 1.8 μg mL−1, with no damage to the host cells. In epimastigotes Et-Bra induced alterations in reservosomes, Golgi complex and mitochondrion. These effects were confirmed by flow cytometry analysis. In trypomastigotes, Et-Bra led to the loss of plasma membrane integrity. Thein vitrostudies indicate that Et-Bra interferes in the functionality of the plasma membrane in trypomastigotes and of reservosomes and mitochondrion in epimastigotes. Acutely infected mice were treated orally with Et-Bra and the parasitemia, mortality and GPT, GOT, CK and urea levels were monitored. The extract (25–300 mg kg−1body weight/day for 10 days) reduced the parasitemia, although not at significant levels; increased the survival of the animals and did not induce any hepatic, muscular lesion or renal toxicity. Since Et-Bra was not toxic to the animals, it could be assayed in combination with other drugs. Et-Bra could be a potential metacyclogenesis blocker, considering its effect on reservosomes, which are an important energy source during parasite differentiation.

AbstractA fear of interlocking twins is one factor that has led to a high Cesarean section (C/S) rate in breech (A)/vertex (B) (Bra/Vtxb) twins. We sought to estimate the frequency of occurrence of twin entanglement, and of interlocking Bra/Vtxb twins in vaginal deliveries. 541 twins and 48 195 deliveries were retrospectively studied for the period 1987–1995. The incidence of Bra/Vtxb deliveries was noted, and the number of deliveries marked by interlocking and collision of fetuses. The mode of delivery, reason for C/S, and sources of perinatal mortality were noted. Only 43 deliveries were Bra/Vtxb (7.9% of all twins) and of these only 14 (32.6%) were delivered vaginally. One case of interlocking (2.3% of all Bra/Vtxb pairs) and five cases of collision of twins (ie competition for entry into the pelvic inlet with obstruction) were noted. All cases mentioned were delivered by C/S. No perinatal mortality resulted from these cases. In recent years the trend has been for greater use of C/S and ultrasound in managing twin deliveries. In this study 67.4% of Bra/Vtxb twins were delivered by C/S. Although there are fewer vaginal deliveries of these twins and the rate for interlocking (2.3%; 95% CI: 0.06–12.3%) for the whole group has remained about the same, we found the rate in those twins allowed vaginal delivery was 6.7% (95% CI: 0.2–31.9%). The presentation at greatest risk for entanglement was found to be Bra/Vtxb.

Abstract:
SUMMARY This paper examines the impact of the development of Business Risk Auditing (BRA) on international audit standard setting and the implications regarding the potential for innovation in auditing practice. BRA was an audit methodology development promoted by the large international audit firms in the late 1990s as a significant innovation but, ultimately, it did not have the degree of impact on practice initially promised. Our analysis demonstrates that professional and regulatory priorities at the time interacted in ways that led to the most innovative elements of BRA not being advanced for inclusion in formal standards. Intriguingly this outcome was clear well before the critical attention given to auditing following Enron, which was the explanation offered for the fate of BRA in previous studies. We conclude that the capacity for innovative ideas to have a substantive influence on audit practice is highly dependent on interactions between a shifting mix of professional and regulatory interests and preferences. The BRA case illustrates how lasting innovation requires a willingness and commitment both to develop the scope of the audit and to accept variations in aspects of its execution. This will not happen if standardization per se is seen as an essential validation of successful innovation.

The garment manufacturing industry is a labor-intensive industry, with one of the slowest transitions to automation. Hence, it is essential to build a smart factory based on automated systems to improve productivity and allow responsive production in the market. In this study, the manufacturing processes for a smart sports bra were established and optimized using various automated machines. For this system, computer-based 3D virtual design software, a technical embroidery machine, an automatic cutting machine, an industrial robot arm with gripper, and an industrial pattern sewing machine were used. The design and materials of the sports bra were selected considering embroidery, cutting, robot gripping, and sewing processes. In addition, conductive thread and light-emitting diode (LED) sequences were used to implement smart functions to the sports bra. Transport of intermediate materials, work orders, and process conditions were optimized to improve the flexible connection of each process and the quality of the final product. This study suggests the concept of the automated manufacturing system that minimizes human intervention by connecting the processes needed to produce a smart sports bra using various automation equipment and programs already used in the industry.

Here, we show that BarH1 and BarH2, a pair of Bar homeobox genes, play essential roles in the formation and specification of the distal leg segments of Drosophila. In early third instar, juxtaposition of Bar-positive and Bar-negative tissues causes central folding that may separate future tarsal segments 2 from 3, while juxtaposition of tissues differentially expressing Bar homeobox genes at later stages gives rise to segmental boundaries of distal tarsi including the tarsus/pretarsus boundary. Tarsus/pretarsus boundary formation requires at least two different Bar functions, early antagonistic interactions with a pretarsus-specific homeobox gene, aristaless, and the subsequent induction of Fas II expression in pretarsus cells abutting tarsal segment 5. Bar homeobox genes are also required for specification of distal tarsi. Bar expression requires Distal-less but not dachshund, while early circular dachshund expression is delimited interiorly by BarH1 and BarH2.

The author discusses the role of NIRAKN in progressing the scholarship of Aboriginal early childhood education through its collaborative research program, raising the bar regarding Aboriginal knowledges in higher education through its network connections and aspects for holding ground in Aboriginal research capacity building, particularly in the area of qualitative research methodologies. These discussions serve to highlight that, as the NIRAKN model has been led by a strong vision, it has fed change and achieved its visions.

Abstract:
How do authoritarian states define and communicate notions of appropriate work conduct and professional excellence? This article examines three channels of communication used by the Chinese state to signal professional expectations to the bar: the bar exam, the administrative rules governing lawyers, and the state-sanctioned National Outstanding Lawyer Award. We find that China’s state narrative about “the good lawyer” celebrates lawyers willing to work closely with the authorities and asks more stringent critics to separate private beliefs from public behavior. In contrast to assumptions often made in research on authoritarian law, this article highlights how lawyers can participate in politics without opposing the regime and how much work goes into curating an appealing state strand of legal professionalism rather than relying on coercion alone. We end with a call for future work on “varieties of legal professionalism” to better understand which state signals are most visible and persuasive to different segments of the Chinese bar, as well as the conditions under which alternate ideas about professionalism gain traction.

By using the modern mathematical tools such as singular theory, geometric topology, and group theory, a global scale analysis method for kinematic performance of mechanisms is proposed. Taking 6-bar with two rings mechanism as an example, a detailed analysis process is studied. The 6-bar ABLM designed by this theory is verified by virtual prototype simulation experiment. The global scale analysis of 4-bar linkage is also carried out by using this method, and the result is compared with the “Grashof criterion” to verify the correctness of this method. It provides a general theory and method for innovative design and global scale analysis of ABLM.

The “AirHopper” leg-wheel hybrid jumping robot we propose is expected to navigate uneven terrain difficult to traverse for ordinary vehicles carrying supplies for disaster-hit areas or isolated districts. To give it high mobility and enable jumping, the AirHopper consists of (1) 4 widespred spiderlike legs for stability, (2) active wheels on its feet driven by built-in motors and transmissions, (3) 2 decoupled pneumatic cylinders, and (4) a built-in piston that drives a 4-bar linkage leg. We propose a powerful leg with a wide movable range and a compact, lightweight wheel drive that minimizes adverse effects. Experiments verified the feasibility of these mechanisms. We also developed an “AirHopper-II” leg-wheel hybrid jumping robot using these 2 mechanisms. AirHopper-II jumped 850 mm vertically and landed softly by controlling in-cylinder pressure. In preliminary experiments, we tested wheel drive jumping to acquire basic data for the next model.

Purpose The aim of this paper is to evaluate using statistical methods how two soldering techniques – the convection reflow and vapour phase reflow with vacuum – influence reduction of voids in lead-free solder joints under Light Emitted Diodes (LEDs) and Ball Grid Arrays (BGAs). Design/methodology/approach Distribution of voids in solder joints under thermal and electrical pads of LEDs and in solder balls of BGAs assembled with convection reflow and vapour phase reflow with vacuum has been investigated in terms of coverage or void contents, void diameters and number of voids. For each soldering technology, 80 LEDs and 32 solder balls in BGAs were examined. Soldering processes were carried out in the industrial or semi-industrial environment. The OM340 solder paste of Innolot type was used for LED soldering. Voidings in solder joints were inspected with a 2D X-ray transmission system. OriginLab was used for statistical analysis. Findings Investigations supported by statistical analysis showed that the vapour phase reflow with vacuum decreases significantly void contents and number and diameters of voids in solder joints under LED and BGA packages when compared to convection reflow. Originality/value Voiding distribution data were collected on the basis of 2D X-ray images for test samples manufactured during the mass production processes. Statistical analysis enabled to appraise soldering technologies used in these processes in respect of void formation.

Abstract:
Abstract Background Patients with reported β-lactam allergies (BLA) are often given alternative perioperative antibiotic prophylaxis, increasing risk of surgical site infections (SSI), acute kidney injury (AKI), and Clostridioides difficile infection (CDI). The purpose of this study was to implement and evaluate a pharmacist-led BLA clarification interview in the preoperative setting. Methods This single-center, IRB-approved, quasi-experimental study compared surgical patients with a BLA between November 2017 and March 2018 (pre-intervention) vs. November 2018 and March2019. From November 2018 to March 2019, a pharmacist performed BLA clarification phone interviews for patients scheduled for a surgical procedure. Based on the allergy history and decision algorithm, first-line antibiotics, alternative antibiotics, or an allergy testing referral were recommended and documented in the EHR. The allergy label was updated as well. The primary outcome was the use of β-lactams preoperatively. Secondary outcomes included 30-day SSI and CDI, AKI, allergic reactions, allergy labels updated or removed, time to incision, and vancomycin doses administered. Results 87 patients were included in the study; 50 (57%) and 37 (43%) in the pre- and post-group, respectively. Most common surgeries: orthopedic 41 (47%), neurosurgery 17 (20%). In the post-group, all EHR BLA labels were updated after interview. 23 patients were referred for allergy testing, 12 (52%) completed BLA testing, and 7 BLA allergies were removed. 76% of pharmacy antibiotic recommendations were accepted (figure). Cefazolin use significantly increased from 28% to 65% post-intervention, P = 0.001; vancomycin use also increased from 19 (38%) to 22 (59%), P = 0.047. Time to incision decreased by a median of 8 minutes (P = 0.484). SSI occurred in 5 (10%) patients in the pre-group only, P = 0.051. All of these were associated with alternative antibiotics. Incidence of AKI and CDI were similar between the groups (P > 0.05). No allergic reactions occurred in either group. Conclusion Clarifying reported BLA in the perioperative setting significantly increased β-lactam preoperative use without negative clinical sequelae. Disclosures All authors: No reported disclosures.

Combination of this optimal four-bar linkage with on of it's cognates and elimination of two redundant cognates will result in a Watt's six-bar mechanism, which generates straight and parallel motion. This mechanism can be utilized for legged machines. The advantage of this mechanism is that the leg remains straight during it's contact period and because of it's parallel motion, the legs can be as wide as desired to increase contact area and decrease the number of legs required to keep body's stability statically and dynamically. “Genetic algorithm” optimization method is used to find optimal lengths. It is especially useful for problems like the coupler curve equation which are completely nonlinear or extremely difficult to solve.

During Drosophila leg development, the distal-most compartment (pretarsus) and its immediate neighbour (tarsal segment 5) are specified by a pretarsus-specific homeobox gene, aristaless, and tarsal-segment-specific Bar homeobox genes, respectively; the pretarsus/tarsal-segment boundary is formed by antagonistic interactions between Bar and pretarsus-specific genes that include aristaless (Kojima, T., Sato, M. and Saigo, K. (2000) Development 127, 769–778). Here, we show that Drosophila Lim1, a homologue of vertebrate Lim1 encoding a LIM-homeodomain protein, is involved in pretarsus specification and boundary formation through its activation of aristaless. Ectopic expression of Lim1 caused aristaless misexpression, while aristaless expression was significantly reduced in Lim1-null mutant clones. Pretarsus Lim1 expression was negatively regulated by Bar and abolished in leg discs lacking aristaless activity, which was associated with strong Bar misexpression in the presumptive pretarsus. No Lim1 misexpression occurred upon aristaless misexpression. The concerted function of Lim1 and aristaless was required to maintain Fasciclin 2 expression in border cells and form a smooth pretarsus/tarsal-segment boundary. Lim1 was also required for femur, coxa and antennal development.

ABSTRACT The artificial light source is one of the most important factors in a controlled environment for vegetable production. It could provide new opportunities to improve growth and increase phytochemicals content in vegetables. Therefore, this study focused on the effects of artificial light sources on growth and phytochemicals in green oak lettuce (Lactuca sativa). The plants were grown under growth chamber with three artificial light sources, namely bar-LED (the 1:1:1 ratio of blue 460 nm : red 630 nm : red 660 nm), bulb-LED (the 2:1:1 ratio of blue 460 nm : red 630 nm : red 660 nm), and fluorescent lamp (FL) (the wavelength range 380-700 nm) for 4 weeks. The quality of bar-LED light was better than either bulb-LED or FL, when assessed by the parameters of photosynthetically active radiation (PAR), photosynthetic photon flux density (PPFD, 400-700 nm), PPFD-B (blue, 400-500 nm), PPFD-R (red, 600-700 nm), the yield photon flux density (YPFD), and the color of red. However, shoot and root (fresh and dry mass), leaf area, leaf number, and shoot/root ratio did not significantly differ between plants grown under bar-LED and bulb-LED, but they were significantly higher than plants grown under FL. Bulb-LED and bar-LED induced larger dry mass of the plants than FL. The dry mass per mole of artificial lighting was highest in plants grown under bar-LED. On the other hand, the fresh mass per mole of artificial lighting was highest in plants grown under bulb-LED. As regards power consumption, bar-LED provided the lowest consumption with 44.4% energy saving over the FL. Total phenolic content, 2,2’-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) and 2,2-diphenylpicrylhydrazyl (DPPH) activities were highest in plants grown under bar-LED. FL lighting gave the least contents of chlorophyll a and chlorophyll a + b. However, chlorophyll b and carotenoid contents did not significantly differ among the treatments. Thus, the results suggested that bar-LED has the potential to improve energy saving, and both growth and phytochemicals content of green oak lettuce grown in a controlled environment of vegetable production.

Most studies employing bioelectrical impedance analysis (BIA) for estimating appendicular skeletal muscle mass using descriptive BIA models rely on statistical rather than biophysical principles. The aim of the present study was to evaluate the feasibility of estimating arm and leg muscle volume (MV) based on multiple bioimpedance measurements and using a recently proposed mathematical model and to compare this technique to conventional segmental BIA at high and low frequencies. MV of the arm and leg, respectively, was determined in 15 young, healthy, active men [age 22 ± 2 (SD) yr, total body fat 15.6 ± 5.1%] by magnetic resonance imaging (MRI) and BIA using a conventional and new bioimpedance model. MRI-determined MV for leg and arm was 6,268 ± 1,099 and 1,173 ± 172 cm3, respectively. Estimated MV by the new BIA model [leg: 6,294 ± 1,155 cm3 (50 kHz), 6,278 ± 1,103 cm3 (500 kHz); arm: 1,216 ± 172 cm3 (50 kHz), 1,155 ± 157 cm3 (500 kHz)] was not statistically different from MRI-determined MV (leg: P= 0.958; arm: P= 0.188). The new BIA model was superior to conventional BIA and performed best at 500 kHz for estimating leg MV as indicated by the lower relative total error [new: 3.6% (500 kHz), 5.2% (50 kHz); conventional: 7.6% (500 kHz) and 8.3% (50 kHz)]. In contrast, the new BIA model, both at 50 and 500 kHz, did not improve the accuracy for estimating arm MV [new: 10.8% (500 kHz), 10.6% (50 kHz); conventional: 11.8% (500 kHz), 11.4% (50 kHz)]. It was concluded that modeling of multiple BIA measurements has advantages for the determination of lower limb muscle volume in healthy, active adult men.

The global prevalence of antimicrobial resistance and Extended-Spectrum and AmpC Beta- Lactamases is continuously widespread among Escherichia coli during recent years, especially in Viet Nam. In Viet Nam, there have been researches on ESBL and AmpC-carrying E. coli inhabiting animal and human. However, studies of antimicrobial resistance in E. coli residing in pets, especially dogs are unavailable. The aim of the study was to investigate the antimicrobial sensitivity testing (AST), the resistance to 3rd cephalosporin and penicillin, also to assess the molecular detection of ESBL and Amp-C-beta -lactamase in E. coli isolates inhabiting the digestive tract of dogs at kennels Dak Lak. By using double disk synergy test (DDST), and ceftazidime-imipenem antagonism test (CIAT) to detect phenotypic characteristic of E. coli strains producing extended-spectrum beta- lactamases (ESBLs) and plasmid-mediated Amp-C-beta -lactamase, and by using multiplex polymerase chain reaction (multiplex PCR) to confirm the presence of ESBL genes (class A): blaCTX-M(1;2;8;9;25), bla TEM, bla SHV , bla OXA and genes encoding AmpC-type beta lactamase (class C): bla MOX-1;2 , bla CMY- (1;2-7;8-11) , blaLAT-(1;4) ,bla DHA-(1;2), bla ACC, bla FOX-(1-5B) ,bla MIR-1 ,bla ACT-1. From of three hundred twelve bacteial strains isolated from sixty-four rectal swabs two hundred sixty-nine E. Coli, isolates accounting for 86%, were identified and isolated, forty-four (16%) and twelve (4%) E. coli isolates encoding with ESBL and Amp-C-beta -lactamases. From molecular diagnosis with regard to phenotype, production of ESBL was shown in thirty-nine (15%) E. coli isolates and Amp-C enzymes in eight (3%) E. coli isolates. The high percentage of E. coli exhibiting antibiotic resistance revealed the accelerated overuse of antibiotics. Result of this study will contribute to the monitoring of epidemiologic resistance.
