Abstract:
Adeno-associated virus (AAV) capsid assembly occurs in the nucleus. Newly synthesized capsid proteins VP1, VP2 and VP3 contain several basic regions (BRs), which may act as nuclear localization signals (NLSs). Mutation of BR2 and BR3, located at the VP1 and VP2 N termini, marginally reduced nuclear uptake of VP1 or VP2, but not of VP3, when expressed in the context of the whole AAV type 2 (AAV2) genome. Combined mutation of BR1, BR2 and BR3 resulted in capsids with slightly reduced amounts of VP1. Expression of isolated VP1/2 N termini revealed an influence of BR3 on nuclear transport, whilst BR1 or BR2 had no effect. However, deletion of an N-terminal fragment in front of the BR elements strongly reduced nuclear uptake of VP1/2 N termini. Mutation of BR4, present in all three capsid proteins, led to their retention in the cytoplasm and to the formation of speckles, resulting in a lack of capsid formation and a significant reduction in VP levels. In a VP fragment comprising BR2, BR3 and BR4, the BR4 element was not necessary for nuclear localization. Mutation of BR5 in the C-terminal part of the VPs resulted in a speckled protein distribution in the nucleus, strongly reduced capsid assembly, and low VP1 and VP2 levels. Taken together, these results showed that BR2 and BR3 have a weak influence on nuclear transport of VP1 and VP2, whilst combined mutation of BR1, BR2 and BR3 influences the stoichiometry of VPs in assembled capsids. BR4 and BR5 play a crucial role in capsid assembly but have no NLS activity.

Abstract:
miR319 was the first plant miRNA discovered via forward genetic mutation screening. In this study, we found that miR319 family members had similar sequences but different expression patterns in Brassica campestris and Arabidopsis thaliana. RT-PCR analysis revealed that Bra-MIR319a and Bra-MIR319c had similar expression patterns and were widely expressed in plant development, whereas Bra-MIR319b could only be detected in stems. The overexpression of each Bra-MIR319 family member in Arabidopsis could inhibit cell division and function in leaf and petal morphogenesis. Bra-miR319a formed a new regulatory relationship after whole genome triplication, and Bra-MIR319a overexpressing in Arabidopsis led to the degradation of pollen content and affected the formation of intine, thereby causing pollen abortion. Our results suggest that Bra-MIR319 family members have functional similarity and difference in plant development.

Abstract:
The composition of a Brazilian green propolis ethanolic extract (Et-Bra) and its effect onTrypanosoma cruzitrypomastigotes and other pathogenic microorganisms have already been reported. Here, we further investigated Et-Bra targets inT. cruziand its effect on experimental infection of mice. The IC50/4 days for inhibition of amastigote proliferation was 8.5 ± 1.8 μg mL−1, with no damage to the host cells. In epimastigotes Et-Bra induced alterations in reservosomes, Golgi complex and mitochondrion. These effects were confirmed by flow cytometry analysis. In trypomastigotes, Et-Bra led to the loss of plasma membrane integrity. Thein vitrostudies indicate that Et-Bra interferes in the functionality of the plasma membrane in trypomastigotes and of reservosomes and mitochondrion in epimastigotes. Acutely infected mice were treated orally with Et-Bra and the parasitemia, mortality and GPT, GOT, CK and urea levels were monitored. The extract (25–300 mg kg−1body weight/day for 10 days) reduced the parasitemia, although not at significant levels; increased the survival of the animals and did not induce any hepatic, muscular lesion or renal toxicity. Since Et-Bra was not toxic to the animals, it could be assayed in combination with other drugs. Et-Bra could be a potential metacyclogenesis blocker, considering its effect on reservosomes, which are an important energy source during parasite differentiation.

AbstractA fear of interlocking twins is one factor that has led to a high Cesarean section (C/S) rate in breech (A)/vertex (B) (Bra/Vtxb) twins. We sought to estimate the frequency of occurrence of twin entanglement, and of interlocking Bra/Vtxb twins in vaginal deliveries. 541 twins and 48 195 deliveries were retrospectively studied for the period 1987–1995. The incidence of Bra/Vtxb deliveries was noted, and the number of deliveries marked by interlocking and collision of fetuses. The mode of delivery, reason for C/S, and sources of perinatal mortality were noted. Only 43 deliveries were Bra/Vtxb (7.9% of all twins) and of these only 14 (32.6%) were delivered vaginally. One case of interlocking (2.3% of all Bra/Vtxb pairs) and five cases of collision of twins (ie competition for entry into the pelvic inlet with obstruction) were noted. All cases mentioned were delivered by C/S. No perinatal mortality resulted from these cases. In recent years the trend has been for greater use of C/S and ultrasound in managing twin deliveries. In this study 67.4% of Bra/Vtxb twins were delivered by C/S. Although there are fewer vaginal deliveries of these twins and the rate for interlocking (2.3%; 95% CI: 0.06–12.3%) for the whole group has remained about the same, we found the rate in those twins allowed vaginal delivery was 6.7% (95% CI: 0.2–31.9%). The presentation at greatest risk for entanglement was found to be Bra/Vtxb.

Syftet med studien är att undersöka vad lärare i ett fritidshem anser är bra och dålig lek. Frågeställningar som ställs i studien är: Vad anser lärarna är bra och dålig lek? Vad grundar sig lärarnas resonemang i? Vad finns det för eventuella likheter eller skillnader mellan lärarnas resonemang? I studien används kvalitativ metod där empirin samlades in genom enskilda och semistrukturerade intervjuer. Lärarna är fem till antalet och arbetar på samma skola och i samma arbetslag. Tematisk analys används för att analysera empirin.Resultatet visar att lärarna i största utsträckning ansåg att lek präglad av delaktighet, inflytande och samspel var bra lek. Lek där eleverna utvecklas i något anseende ansågs också vara bra lek. Dålig lek ansåg majoriteten av lärarna vara lek med bristande demokrati. Brottningslek och lek där elever inte bidrog till något konstruktivt ansågs också vara dålig lek. Resultatet visar att lärarna resonerar på olika sätt gällande lek de ansåg var dålig och lek de avbröt.

Syftet med examensarbetet är att undersöka i vilka situationer barn själva uttrycker att de mår bra samt undersöka när föräldrar och pedagoger upplever att barn mår bra på förskolan.  Metoden som använts är en kvalitativ undersökning, intervjuer har gjorts med barn i femårsåldern och med föräldrar och pedagoger på två olika förskolor. Resultatet visar att barn upplever att de mår bra när de leker, denna uppfattning delas också av föräldrar och pedagoger i barnens närhet. För barnen är det viktigt med vänner och social samspel för sitt välbefinnande. Undersökningen visar också att pedagogerna i sin planering och med sitt sätt att tillrättalägga verksamheten ger möjlighet till KASAM och tillfälle till Flow. Undersökningen visar även att barn mår bra när de får vara med och bestämma och när de får utmaningar i sin utveckling och sitt lärande. I det hela visar resultatet att föräldrar och pedagogers uppfattning om barns välbefinnande stämmer bra överens med barns egen uppfattning om när de mår bra.

Denna studie har riktat sig till ett stort antal pedagoger som arbetar i barngrupp på förskola i Sverige. Undersökningens mål är att undersöka hur stor del av dagen barnen tillbringar på den inhägnade gården samt hur ofta det förekommer planerad lärarledd aktivitet utomhus respektive inomhus? Vidare så är undersökningens mål att få fram vad respondenterna anser vara det huvudsakliga syftet med utomhusvistelse på förskolan. Studien har genomförts i form av en metodkombination där vi använt oss av en kvantitativ enkät med inslag av kvalitativ metod. De kvalitativa inslagen har skett genom följdfrågor samt en öppen kvalitativ fråga. Enkäten har därefter skickats ut i tre olika diskussionsforum för förskolan på Facebook. Undersökningens resultat visade åsikter kring att utevistelsen var viktig men att en del föredrog annan typ av miljö för lärarledda aktiviteter än gården, då främst skogen. Resultatet visade även att det oftast är arbetslaget som utformat och bestämt hur den egna gården ska se ut.

Abstract:
Syftet med vår studie har varit att undersöka förskollärares uppfattningar om det lärande som sker i barns fria lek, med fokus på förskollärarens egen roll och förhållningssätt. För att synliggöra förskollärarnas tankar om ämnet valde vi att använda oss av kvalitativa intervjuer. Intervjuerna har varit delvis struktur-erade vilket innebär att vi har haft möjlighet att påverka frågeställningarna beroende på våra informanters svar. I resultatet har det framkommit att samtliga av de fem förskollärare vi har intervjuat tycker att det är viktigt att barn lär sig socialt samspel i den fria leken. Samtliga pratar även om att barn utvecklar sitt språk när de leker med andra barn. Utöver det har förskollärarna olika fokus, där vissa fokuserar på ämneskunskaper så som matematik eller naturkunskap medan andra fokuserar på kompetenser så som fantasi och kreativitet. I diskussionen har vi kunnat visa på kopplingar mellan vilket lärande förskol-lärarna fokuserar på, vilken roll de tar i den fria leken, samt deras uttalanden om utbildning och kollegial reflektion. Vi ställde resultatet från vår studie mot tidigare forskning inom området och kunde visa att det i stor utsträckning överensstämde med andra författares studier.

Vår studie belyser kontrasten mellan förskollärarnas uppdrag utifrån den nya läroplanen för förskolan (2018) kring att introducera digitala verktygen i förskolan, samtidigt som WHO (2019) rekommenderar att minska barns skärmtid. Målet med vår studie var att undersöka hur förskollärare resonerar kring digitala verktyg i förskolan utifrån läroplanen och WHO, då vi ser att det finns en kunskapslucka om förskollärares resonemang kring digitala verktyg. Det är en kvalitativ studie som består av semistrukturerade enskilda intervjuer och kan fokusgruppsintervjuer. Vi har analyserat vårt empiriska material utifrån ett sociokulturellt perspektiv det har hjälpt oss i vår analys eftersom vi ville utgå från ett lärande när vi tittade på vår analys. Våra begrepp som vi har använt oss av har varit den närmaste utvecklingszon, artefakt, mediering och leken. Studiens resultat visar att förskollärarna ser på digitala verktyg i förskolan som något bra men att det också kan vara mindre bra om de inte används på rätt sätt. Förskollärarna beskriver att med rätt sätt menas att det finns ett lärande och ett syfte i användandet. I resultatet blev det även synligt att digitala verktyg uppfattas som något bra när barnen använder dessa på ett aktivt sätt tillsammans med andra barn och/eller vuxna. När barn använder digitala verktyg ensamma eller på ett passivt sätt så uppfattades detta som mindre bra. Genom denna studie har vi bidragit med en ökad förståelse för hur förskollärare resonerar kring användandet av digitala verktygen i förskolan i förhållande till barns lek och lärande.

Syftet med denna studie är att undersöka hur förskollärarna på två mångkulturella förskolor arbetar för att skapa trygghet i leken hos nyanlända barn. För att få fram ett resultat så genomfördes en kvalitativ studie där fyra förskollärare intervjuades enskilt kring ämnet, vilket sedan transkriberades och analyserades grundligt för att få fram ett resultat. Studien utgick ifrån ett interkulturellt perspektiv då det perspektivet berör mångkulturella grupper där en handling sker bland individer med olika kulturella bakgrunder. Resultatet visade på att förskollärarna använder sig av olika metoder som närvarande pedagog, ta del av barnens hemkultur, tecken, bilder och rörelser för att främja trygghet hos nyanlända barn i leken. Förskollärarens inflytande i barns gruppkonstellationer beskrivs då det skapar trygghet hos barn även där. En närvarande förskollärare som bidrar till att barnen leker i en varierad mångfald skapar på så sätt en trygghet i en interkulturell miljö.

Kultur, medier och estetik, examensarbete, Malmö Högskola.Syftet med detta arbete är att undersöka, jämföra och analysera fyra olika pedagogers synsätt på aktiviteterna lek och lärande. I undersökningen försöker jag även redogöra för hur pedagogernas inställning till lek eventuellt har förändrats eller utvecklats över deras tid i yrket. Ytterligare undersöker jag om och hur pedagogerna medvetet planerar för och använder leken i sitt praktiska arbete.Metoden jag använde är kvalitativa intervjuer och för att uppnå syftet i min undersökning försöker jag att besvara följande frågor: Hur definierar pedagogerna aktiviteterna lek och lärande? Hur förhåller sig dessa aktiviteter till varandra? Hur upplever pedagogerna att deras synsätt på leken har utvecklats eller förändrats över tiden i yrket? Hur arbetar pedagogerna praktiskt med att förena aktiviteterna lek och lärande?Litteraturavsnittet behandlar olika forskares och författares teorier kring begreppen lek och lärande. I min analys av resultatet jämför jag teorierna med resultatet av intervjuerna. Min slutsats är att de fyra intervjuade personerna menar att lek och lärande hör ihop och har tydliga samband. Synsättet på lek och lärande har förändrats något över tiden och lek används som en metod för lärande.

Den fria leken resulterar inte alltid i något konkret material så som många andra styrda aktiviteter på förskolan kan göra, vilket innebär att viss problematik kan uppstå. Bristande "bevis" på att lärande har skett kan bland vårdnadshavare skapa tvekan och undran över barnens dag på förskolan och om de verkligen har lärt sig något. Tidigare forskning har visat att lek och lärande hänger samman och skapar förutsättningar för barns utveckling men att den kan missuppfattas av allmänheten. Den fria leken beskrivs som en mycket viktig del av ett barns lärande där tidigare erfarenheter bearbetas och ny kunskap tillägnas. Syfte Syftet är att ta reda på förskollärares föreställningar kring den fria leken och redogöra för dess betydelse som pedagogiskt verktyg i förskolan. Frågeställningarna lyder: Vad innebär den fria leken för barns utveckling och lärande enligt förskollärare? Vad har förskolläraren för roll och betydelse i barns utveckling och lärande genom fri lek? Vad har den fria leken för pedagogiskt syfte? Metod Den här undersökningen är en intervjustudie och är baserad på en fenomenologiskt inspirerad ansats vilket innebär att andra personers livsvärdar och synsätt hamnar i fokus. Den metod jag har valt att använda mig av är intervjuer där 6 stycken förskollärare i olika åldrar på två avdelningar har deltagit. Resultat Mitt resultat visar på att den fria leken utgör en stor del av den pedagogiska verksamheten. Den fria leken ses som redskap för att lära känna barnen på en djupare nivå samtidigt som den fria leken skapar oändliga möjligheter för barns utveckling och lärande. Resultatet visar alltså på att det finns en pedagogisk tanke bakom det utrymme den fria leken ges och att pedagogens roll först och främst handlar om att vägleda, delta, stötta och inspirera barnen i deras lek och lärande

Abstract:
Vårt syfte med denna studie är att undersöka förskollärare och fritidspedagogers uppfattning om fri lek, hur de ser på den fria lekens betydelse för lärande och utveckling och sin egen roll i den. Läroplanerna för både förskola och fritidshem betonar lekens betydelse för barns utveckling och lärande, både fysiskt och psykiskt. Även den forskning vi har tagit del av har framhävt detta. Begreppet fri lek är dock något som inte definierats tydligt i läroplaner eller forskning, därför blev vi intresserade av hur pedagoger ser på detta begrepp och den aktivitet som ändå har en given plats i båda verksamheterna. Studien skaffar sin empiri genom kvalitativ metod där fem förskollärare samt fem fritidspedagoger har intervjuats. Resultaten visar i korta drag att det finns en enighet mellan förskollärare och fritidspedagoger när det gäller den fria lekens betydelse. En viss skillnad mellan förskollärare och fritidspedagoger kunde ses genom att de senare lyfter fram social kompetens som en av de viktigaste aspekterna av vad barnen lär sig genom leken, medan förskollärarna till större del ser leken som ett redskap för att på ett lustfyllt sätt möjliggöra all form av inlärning. Både förskollärare och fritidspedagoger ser sin roll som observerande och avvaktande i leken, och att om de går in i leken är det i första hand för att avstyra bråk eller för att förhindra att någon gör sig illa.

Abstract:
En général, les écrivains francophones du Maghreb ont une liaison très forte avec l’eau étant donné que leurs pays d’origines sont riverains de la mer Méditerranée et de l’océan Atlantique. L'écrivain que je vous présente, Driss Chraïbi, est originaire de El Jadida, une petite ville située au bord de l’Atlantique à l’embouchure de l’Oum-er-Bia, le plus grand fleuve du Maroc. Pour lui, la présence de l’eau est importante pas seulement dans sa vie mais aussi dans son œuvre. Un de ses romans, La Mère du printemps, a comme sous-titre le nom du fleuve auprès duquel il est né. Chraïbi, comme signale François Mauriac dans Le romancier et ses personnages, souligne l’importance des lieux vécus même dans ses romans de fiction, et l’eau apparaît toujours comme un élément remarquable. D’un autre côté, l’eau sert à relier les différentes civilisations, la Méditerranée a toujours servi de moyen d’union entre les habitants d’une ou de l’autre rive ; elle représente aussi le radeau de sauvetage des immigrants qui cherchent une nouvelle opportunité, comme est le cas des personnages des Boucs. Une perspective postcoloniale (Bessière, Moura, Ashcroft, Griffiths, Tiffin) peut nous dévoiler toutes les représentations de l’eau dans l’imaginaire de cet écrivain du Maghreb et c’est l’approche que je propose pour cette communication.DOI: http://dx.doi.org/10.4995/XXVColloqueAFUE.2016.2995

Abstract:
L’histiocytose langerhansienne est une maladie rare causée par la prolifération et l’infiltration d’un ou plusieurs organes, par des cellules dendritiques de type Langerhans. Elle s’exprime par des manifestations cliniques extrêmement polymorphes et peut toucher l’os, la peau, l’hypophyse, les poumons, le système nerveux central, et plus rarement le foie et le système digestif. Elle a été initialement décrite chez les enfants. L’histiocytose langerhansienne de l’adulte présente une entité particulière tant par ses manifestations cliniques que par sa prise en charge. Le cas présenté est celui d’un patient âgé de 53ans, en bon état de santé apparent et non fumeur, qui s’est présenté à la consultation pour des lésions nécrotiques et douloureuses des muqueuses gingivales mandibulaires et maxillaires, associées à des mobilités dentaires sévères. L’examen exobuccal ne révélait aucune asymétrie faciale ni adénopathies. L’examen endobuccal confirmait la présence de lésions nécrotiques gingivales et une parodontite sévère au niveau mandibulaire antérieur et maxillaire postérieur. Le secteur maxillaire antérieur présentait un parodonte sain. L’examen radiologique panoramique et Cone Beam CT , révélaient des lyses osseuses moyennes à terminales s’étendant de la 44 à la 38 et au niveau des molaires maxillaires droites et gauches. Les dents antérieures ne présentaient quant à elles pas de lyse. Par ailleurs des images lacunaires à l’emporte pièce siégeaient au niveau du secteur mandibulaire édenté. Ces manifestations évoquaient un lymphome ou une manifestation orale d’une infection virale type VIH. L’examen biologique révélait une légère hyperleucocytose et une augmentation de la vitesse de sédimentation à la première heure. L’examen anatomopathologique des lésions muqueuses, a rapporté la présence d’éléments histiocytaires se regroupant en nodules, concluant en une histiocytose langerhasienne de l’adulte. Le bilan d’extension ne révélait aucune atteinte associée, concluant en une histiocytose localisée. L’étude moléculaire a montré la présence d’une mutation V600E du gène BRAF (Facchetti et al, European journal of pathology, 471(4); 2017). Ce dernier est situé sur le chromosome 7, et il est impliqué dans l’envoi des signaux qui déterminent la croissance cellulaire. L’évolution et le pronostic de cette maladie sont étroitement liés à l’âge et aux organes atteints. Les régressions spontanées ont été rapportées, et peuvent être induites par un curetage ou par une simple biopsie. (Goncalves et al, Biomedical research notes,9(19); 2016) Pour notre cas, les extractions des dents mobiles avec curetage des lésions ont permis une cicatrisation complète. La thérapeutique médicamenteuse peut comprendre une corticothérapie locale ou systémique, des bisphosphonates, voire même une radiothérapie. Par ailleurs la confirmation de la mutation BRAF permet d’oser un traitement spécifique par inhibition de l’enzyme produite par ce gène. Abstract:
The paper presents the design of a lower leg orthotic device based on dimensional synthesis of multi-loop six-bar linkages. The wearable device is comprised of a 2R serial chain, termed the backbone, sized according to the wearer’s limb anthropometric dimensions. The paper is a result of our current efforts in proposing a systematic process for the development of 3D printed customized assistive devices for patients with reduced limb mobility, based on anthropometric data and physiological task. To design the wearable device, the physiological task of the limb is obtained using an optical motion capture system and its dimensions are set such that it matched the lower leg kinematics as closely as possible. As a next step a six-bar linkage is synthesized and ensured that its motion is as close as possible to the physiological task. Next, the 2R backbone is replaced by the wearer’s limb to provide the skeletal structure for the multi-loop wearable device. During the final stage of the process the 2R backbone is relocated to parallel the human’s limb on one side, providing support and stability. The designed device can be secured to the thigh of the user to guide the lower leg without causing any discomfort and to ensure a natural physiological gait trajectory. This results in orthotic device for assisting people with lower leg injuries with compact size and better wearability.

L’organisation de la réhabilitation maxillo-faciale implanto-portée en oncologie tête et cou reste sujet à débat. Il n’existe pas de consensus quant au moment de la réhabilitation implantaire (pendant ou à distance de l’exérèse) ou quant aux prérequis anatomiques qui garantissent son succès. Le but de cette étude est de mettre en place un protocole de prise en charge implanto-portée de patients ayant été traités en oncologie tête et cou. Il s’agit d’une étude rétrospective entre 2013 et 2017, analysant la réhabilitation implantoportée de neuf patients traités pour un cancer tête et cou par chirurgie et radiothérapie (groupe 1 : 5 patients) ou par chirurgie seule (groupe 2 : 4 patients). Les critères étudiés étaient les données personnelles des patients, leurs antécédents oncologiques, les bilans oraux pré-opératoires effectués, les thérapeutiques utilisées ainsi que le suivi. Tous les patients ont été traités pour des carcinomes épidermödes majoritairement situés au niveau du plancher buccal. Ils présentaient tous une limitation de la mobilité linguale, une absence de vestibule et des tissus crestaux mobiles. Les projets prothétiques prévoyaient de réaliser pour sept patients une prothèse amovible complète mandibulaire stabilisée sur implants et pour deux patients des couronnes unitaires sur implant. Les patients ayant recu un lambeau libre de péroné latéral ont bénéficié d’une greffe d’apposition associée à un lambeau de FAMM. Les deux greffes ont permises la pose des implants. Sur neuf patients, cinq ont bénéficié d’une libération linguale, six ont recu une vestibuloplastie et huit ont eu une greffe épithélio-conjonctive péri-implantaire. Les implants ont été posés sous anesthésie générale avec un guide chirurgical pointeur. Il s’agissait d’implants Tissu Level RN SLA Roxolid (Straumann, Basel, Suisse). Pour les patients du groupe 2, la pose des implants a eu lieu en même temps que la vestibuloplastie. Lors de cette étude quatre patients étaient réhabilités par une prothèse sur implants. Le groupe 2 n’a pas présenté de complications. Deux patients du groupe 1 ont nécessité plusieurs libérations linguales et deux implants ne se sont pas ostéointégrés dans ce groupe. Lors de chirurgies en deux temps, des greffes épithélio-conjonctives crestales ont dû être réalisées pendant la vestibuloplastie puis pendant la pose implantaire. Deux greffes osseuses ont été réalisées chez deux patients ayant été irradiés bien que cela soit contre indiqué dans la littérature (Raoul 2009). Une greffe osseuse d’apposition peut donc être une solution viable chez ces patients afin de limiter le bras de levier prothétique. Cette étude a permis de mettre en exergue : l’importance de prévoir en amont la réhabilitation implantoportée lors de la reconstruction pour une optimisation du positionnement du greffon osseux et l’intérêt d’effectuer dans le même temps vestibuloplastie, pose des implants et aménagement péri-implantaire pour la pérennité implantaire. Cette étude rétrospective a permis de proposer un protocole de prise en charge standardisé de la réhabilitation implanto-portée de patients traités pour un cancer tête et cou.

Abstract:
This paper uses path synthesis techniques to design four-bar linkage modules to constrain the movement of a 3R chain. The result is a 10-bar linkage. The goal is to develop a design procedure for a robotic system that guides the human leg during the walking gait cycle. A 3R chain is designed to match the dimensions of a human leg and the two four-bar linkages are synthesized using 9 point path synthesis to constrain the trajectory of the ankle and the toe. Precision points are derived from a basis spline equation. A numerical example is given using data collected from a motion capture system.
