Abstract:
Molecule property prediction is a fundamental problem in many fields. To accurate and rapid prediction of molecules properties, molecule characterization and representation are key operations in the pretreatment stage. Generally, we represent molecule as graph based on different features such as element type, bond type, etc. The features we select could have the ability of enough representation and discrimination. However, in terms of bond embedding, one-hot coding is the most common processing method in the current research which means the features we used are discrete and could not distinguish different single or double bonds in a molecule. Here we add predicted bond energy feature as an extra chemical bond descriptor and compare three popular GNN models on two different datasets. The experiment shows that supplying additional unusual bond features—bond energy will improve the model performance significantly. The PDN model with bond energy has best performance among three model. Finally, we discuss the experimental result.

Abstract:
The problem of negative design of DNA languages is addressed, that is, properties and construction methods of large sets of words that prevent undesired bonds when used in DNA computations. We recall a few existing formalizations of the problem and then define the property of sim-bond-freedom, where sim is a similarity relation between words. We show that this property is decidable for context-free languages and polynomial-time decidable for regular languages. The maximality of this property also turns out to be decidable for regular languages and polynomial-time decidable for an important case of the Hamming similarity. Then we consider various construction methods for Hamming bond-free languages, including the recently introduced method of templates, and obtain a complete structural characterization of all maximal Hamming bond-free languages. This result is applicable to the θ-k-code property introduced by Jonoska and Mahalingam.

Abstract:
Effect of loading rate on bond property between corroded concrete and steel bar are studied. Simulation test and finite element are used to study the pull-out test of corroded reinforcement concrete members in present literature. Considering the corrosion of steel bar and concrete and their rate dependent characteristics, the bond property of corroded reinforcement and concrete under the different loading rate (10-4 s-1-10-2 s-1) are studied. It concluded that the bond property was improved when the rating rate form 10-4 s-1 to the static load; the bond property would decreased when the loading rate reached the fixed value (10-3s-1-10-2s-1), the members was failed under the smaller load.

Abstract:
This paper reports a Density Functional Theory (DFT) investigation of the electron density and optoelectronic properties of two-dimensional (2D) MX2 (M = Mo, W and X = S, Se, Te) subjected to biaxial strains. Upon strains ranging from −4% (compressive strain) to +4% (tensile strain), MX2 bilayers keep the same bandgap type but undergo a non-symmetrical evolution of bandgap energies and corresponding effective masses of charge carriers (m*). Despite a consistency regarding the electronic properties of Mo- and WX2 for a given X, the strain-induced bandgap shrinkage and m* lowering are strong enough to alter the strain-free sequence MTe2, MSe2, MS2, thus tailoring the photovoltaic properties, which are found to be direction dependent. Based on the quantum theory of atoms in molecules, the bond degree (BD) at the bond critical points was determined. Under strain, the X-X BD decreases linearly as X atomic number increases. However, the kinetic energy per electron G/ρ at the bond critical point is independent of strains with the lowest values for X = Te, which can be related to the highest polarizability evidenced from the dielectric properties. A cubic relationship between the absolute BD summation of M-X and X-X bonds and the static relative permittivity was observed. The dominant position of X-X bond participating in this cubic relationship in the absence of strain was substantially reinforced in the presence of strain, yielding the leading role of the X-X bond instead of the M-X one in the photovoltaic response of 2D MX2 material.

Abstract:
The goal of this study is to ascertain how partial leverage has an impact on bond ratings, between 2016- 2019, the influence of profitability on bond ratings, as well as the simultaneous effect of leverage and profitability on bond ratings in the property, real estate, and construction industries listed on the Indonesian stock exchange. Leverage is measured using debt-to-equity ratio proxies, and profitability is measured using return on assets as an independent variable, while bond ratings are measured using ratings from the rating agency PT. PEFINDO as a dependent variable. All companies in the property, real estate, and construction industries that were publicly traded on the Indonesian Stock Exchange between 2016 and 2019 are included in this study's population. The sample is comprised of firms that issued bonds between 2016 and 2019, and the sampling approach is non-probability sampling with purposive sampling, yielding a total sample of ten companies or forty financial statements. The data analysis approach employed is linear regression with a 5% significance threshold, utilizing the SPSS 26 and Eviews 10 software packages. According to the conclusions of this study, leverage has a limited impact on bond ratings, profitability has a limited impact on bond ratings, and both leverage and profitability have an impact on bond ratings. simultaneously.

Abstract:
In order to realize the recycle of waste tires, rubber concrete can be utilized in road engineering. The bond property of interface between rubber concrete and matrix ordinary concrete is one of the key factors. This paper studied the interfacial bond property by splitting tensile strength test and shear strength test and considered several influence factors, such as rubber volume fraction, rubber particle size, interfacial roughness and curing time of matrix concrete. The results show that the shear strength decreases with the increase of rubber content, longer curing time of matrix concrete, and the larger size of rubber particle. The splitting tensile strength also deceases with the longer curing time of matrix concrete or smaller interfacial roughness. However, no regular relationship is found between the splitting tensile strength and the size of rubber particle or rubber volume content.

Abstract:
Polystyrene (PS) was synthesized by emulsion polymerization. 1,4-bis(chloromethoxy)butane (BCMB) was synthesized using 1,4-butanediol, formaldehyde and phosphorus trichloride as raw material. The linear poly(p-chloromethyl styrene) (PS-Meyl-Cl) was prepared high effectively using BCMB and anhydrous aluminium trichloride as the chloromethylation reagent and catalyst, respectively. In this method, carcinogenic compound (chloromethyl ether etc.) was avoided. The resultant polymers were characterized by gel permeation chromatography (GPC), Fourier transform infrared spectrometer (FTIR) and thermogravimetric analyzer. The results show that the molecular weight (Mw) of PS is big. BCMB has been prepared because of the strong absorption band observed at 1130cm-1 (due to C-O-C bond stretching vibration) and two new bands at 640 and 1315cm-1 (due to the bending vibration of C-Cl bond and stretching vibration of C-H bond in -CH2Cl, respectively). Chloromethyl groups were introduced into benzene ring of PS because of the appearance of the absorption peaks at 676 and 1419cm-1 (due to the bending vibration of C-Cl bond and stretching vibration of C-H bond in -CH2Cl, respectively) in the FTIR spectra. The thermal stability of PS-Meyl-Cl was decreased with increasing chloromethyl group. Moreover, there are two decomposing stages. The reaction condition is gentle and the operation is safe, simple and economical.

Abstract:
Within this work the effects of metal identity are investigated on the magnetic properties of a range of materials, in which different transition metals are shown to produce significantly different calculated magnetic coupling values when incorporated into the same structure. The model structures composed of transition metals mediated by hydrogen halide ligands of the form FHF-, ClHCl- and FHCl- enable the calculation of magnetic coupling values via hydrogen bonds (HBs), providing insight into a scarcely studied topic. The employment of the well known HF=35% functional, provides results anticipated to represent potential experimental trends. Interesting results are also reported which indicate a relationship between the binding energy of the atom and the level of coupling displayed. The quantum nature of the H atom, often difficult to localise, are also accounted for by use of a shooting algorithm in order to solve the one dimensional Schrödinger equation in relation to the hydrogen positions within the isotropic lattice. Hydrogen positions were fixed during geometry optimisations in order to obtain the E(x) potential energy curve required to allow this to be achieved. Both the in which proton motion is incorporated and J coupling values from the optimised ground state structures, in which the atomic positions including the H positions underwent a full geometry optimisation, are presented. The electronic effects of incorporating different group one and group two metals into the related Prussian Blue (PB), Prussian Yellow (PY) and Prussian White (PW) model lattices are also presented. More specifically band gaps within these complexes are calculated from projected density of state (DOS) plots of the electrons within the atomic orbitals from a converged calculation in which the B3LYP functional (HF=20%) is employed. This functional has previously been shown to provide band gap energies in good agreement with experimental values. The results obtained confirm that the value of ~3eV, associated with an absorption in the yellow region, lies within the PY band gap range, indicated by the HOMO and LUMO orbitals within the DOS plot. Band gaps calculated directly from SCF energy differences do not agree well with experimental values or trends for the various group one PB complexes. A number of related metal containing bromanilic acid (BA) and chloranilic acid (CA) complexes are synthesised, crystallised and their structures, as determined by X-ray single crystal diffraction, reported. The structural packing of the molecules is analysed by defining a number or recurring unit within the structures in order to obtain key similarities. The structures are also broken down in to lower dimensional units composed of the individual components within the asymmetric unit and built up into higher dimensional units such as clusters, chains and planes that all intersect with one another in order to produce the overall three dimensional structures. In most cases the point of intersection is on a key symmetry element such as an inversion centre. Particularly short M-M contact distances are observed in a number of the complexes. This proves interesting within the related isomorphous complexes in which the main effect of incorporating a larger metal cation into the same structure is in the ionic repulsions between the metals. This is particularly interesting in more extended structures in which the metals exist in chains or planes that run along a particular direction of the unit cell. The specific increase in cell parameter can in most cases be explained as a consequence of the repulsion between the shortest M-M contacts. The effect is also observed in structures in which the metals exist as isolated clusters, and also displays some interesting consequential effects on other interactions. A variable pressure neutron single crystal diffraction experiment on a short asymmetric HB is also reported, in which one lattice is compressed significantly more than the other two, an effect that is explained by the change in contact distances with pressure.

ENGLISH ABSTRACT: Owners of directly held, large commercial properties mostly face problems regarding liquidity and sometimes higher-risk exposure due to large properties dominating their property portfolio. • Exit vehicles in the listed property sector on the JSE are gaining momentum but are, however, still facing the negative impact of overall bear market conditions. • An exit vehicle in the bond market can serve as an alternative, where a Limited Purpose Company/Special Purpose Vehicle (SPV) acquires a rental income stream from a portfolio of properties and issues bonds. The funds raised from bond investors, who focus on the credit rating of the income stream and not the properties, are then used to acquire the property assets. • Current market conditions, favour an asset-backed securitisation of property leases. Low interest rates and other factors in the current market, as discussed in this document, ensure lower costs of debt and easier access to capital when funds are raised on the bond market, relative to the equity markets, with highly geared structures not necessarily causing a negative impact on the credit rating (80% - 90% gearing possible with A - AA local credit rating). • Usually a promoter, a Merchant Bank and a few institutions/ pension funds are involved. Example: Institution X will be requested to take up an equity stake in the fund by selling some properties or stakes therein (Institution X buildings) and receiving payment in a combination of cash, junior bonds and equity. The benefits to Institution X are discussed on page 39.

In the first part of the article, a development for the concepts of provisional remedies and real or personal property bond and covering loss and damage, is proposed, under the Constitutional Court and our national doctrine view; as well as the scope of the first as a fundamental right, and second as a condition of execution. On the following part of the paper, the inclusion and the subsequent amendments to the article 159° of Tax Code is exposed, also its scopes and reasons. Finally, a possible modification of the article, that respect state´s raising goals and due process citizens’ right, is proposed.
El presente artículo desarrolla los conceptos de medida cautelar y contracautela, a la luz de lo resuelto por el Tribunal Constitucional y lo establecido por nuestra doctrina, así como los alcances del primero como derecho fundamental y del segundo como requisito de ejecución. De la misma forma, se analiza los alcances de la incorporación del artículo 159° del Código Tributario, mediante Decreto Legislativo N° 1121, y su modificatoria a través de la Ley N° 30230. Finalmente, se propone una posible modificación al mencionado artículo, de conformidad con los fines recaudatorios del Estado y el derecho a la tutela jurisdiccional efectiva de los administrados.

Nous decrivons les progres recents de la spectrometrie de masse dans l'etude structurale des proteines. Nos etudes associent la degradation chimique d'edman, les hydrolyses enzymatiques et les analyses par esims pour resoudre des problemes specifiques a la caracterisation de la structure primaire des proteines. Ainsi nous montrons qu'il est possible de preparer des sels d'ammonium quaternaires d'isothiocyanates par synthese chimique et de les utiliser dans les conditions de la degradation d'edman. La detection des produits resultants est realisee par esims afin d'ameliorer la sensibilite et d'assurer la caracterisation de certains residus. La connaissance de la structure primaire des proteines nous a conduit a l'etude de la localisation des ponts disulfures, responsables en partie du maintien de la structure tridimensionnelle des proteines. Une approche nouvelle utilisant la tris-(2-carboxyethyl) phosphine pour la reduction des ponts disulfures et le n-hydroxymethyl benzamide comme methode d'alkylation des cysteines libres en milieu acide est presentee. Apres avoir realise une etude comparative de peptides lies a l'aide d'un pont disulfure par sm avec divers modes d'ionisation, nous avons identifie ceux des puroindolines a et b de ble. Enfin, par esims, nous avons obtenu des renseignements sur la stabilite et la conformation des cytochromes c#2 et c#5#5#3 sauvages et mutes. Nous avons mis en evidence la presence de plusieurs conformeres de proteines par observation de la distribution des etats de charges et par la mesure des echanges isotopiques (h/d). Nous avons ainsi classe en fonction de leur stabilite, les mutants des deux cytochromes par rapport aux types sauvages. De plus, nous avons mis au point et realise l'etude des sites de deuteriation des hydrogenes amidiques du squelette polypeptidique. Celle-ci utilise les echanges isotopiques suivis d'une digestion rapide par la pepsine permettant d'obtenir des informations locales sur la penetration du solvant deuterie dans la proteine

AbstractThe Egyptian ideal was to establish an everlasting mortuary cult in order to ensure for the deceased infinite commemoration in this world and eternal life in the otherworld. In order to achieve this bold aim, funerary institutions were endowed with land and income, the priests and personnel were bound strictly to observe their duties and principles, and future offspring were defined as legally constrained to take over the offices and obligations of their fathers. In most instances, however, families became incapable of affording the cult or were extinct after a few generations, personnel left the priesthood, or the funerary institutions were stripped of property and were discontinued. Over time, even royal mortuary cults were ended and royal monuments abandoned. The tombs and memorial monuments then remained uncared for and were left to an uncertain future. As time went by, some of those monuments evolved into local attractions for visitors, some were piously restored, some were usurped or reused, and some were dismantled in order to process the building materials for new edifices. The present contribution aims at tracing the Egyptians’ thoughts and experiences and at illustrating the manifold fates of Egyptian mortuary monuments.

In this project, fully generalised analytical expressions for longitudinal Young’s modulus are developed for zigzag Single-Walled Nanotubes (SWNTs), based on a hexagonal honeycomb motif, using an energy equivalent approach. Honeycomb geometry (bond lengths and angles), and bond-stretch and bond-angle force constants are considered as variable parameters and deformation is assumed due to simultaneous bond stretching and bond angle variation in response to an applied axial load. A parametric study is then performed to explore the structure-property relationships in auxetic (negative Poisson’s ratio) nanotube.

Abstract A process control tool has been developed for air plasma spraying of a NiAl (bond coat) and Al2O3 (top coat) coating systems. The process is employed at Volvo Aero Corporation for abrasive purposes, such as knife-edge applications on compressor parts. In-flight particle temperatures, velocities and diameters were measured by the DPV2000 system. Several samples were sprayed and the coating microstructures were evaluated using Image Analysis techniques on optical and scanning electron microscope images. Top and bond coat thickness, oxides, porosity, grit blast residues, delaminations, surface roughness (on top, bond and substrate) and tensile strength were evaluated. Statistical regression analysis was then used to establish relationships between process parameters (i.e. current and primary gas flow), particle in-flight characteristics (i.e. velocity and temperature), microstructure properties, and mechanical properties. The equations derived were finally used for development of a tool, which can be used by the operator for on-line monitoring and control of the coating characteristics based on information of the current particle inflight characteristics. The tool makes it possible to continuously adjust the process set points, ensuring a high reproducibility and stability of the process.

The life of a thermal barrier coating (TBC) system is governed by the microstructural evolution of the thermally grown oxide (TGO) layer between the ceramic top layer and the bond coat. While the TGO provides a barrier to the oxygen diffusion, its continuous growth imposes stresses on the TGO/bond coat and TGO/topcoat interfaces that will eventually lead to crack linkage and propagation followed by failure of the TBC system. In addition to TGO growth during thermal exposure, coating properties such as hardness, Young’s modulus and fracture toughness will also change. This study is undertaken to investigate the oxidation behaviour of an electron beam physical vapour deposition (EB-PVD) YSZ/PtAl coating system. Cyclic oxidation tests were carried out with each cycle consisting of 5 hr holding time at 1150°C followed by air cooling to room temperature. TGO evolution, coating property and maximum crack length as functions of total thermal exposure time were investigated in this study. Microstructural analyses of the coating were carried out using scanning electron microscopy (SEM) and energy dispersive x-ray spectroscopy (EDS). Both hardness and Young’s modulus of the bond coat were measured using micro-hardness tester. The results from this study showed a parabolic TGO growth rate as a function of thermal exposure time. While no distinct mathematical relationship was found between the TGO thickness and maximum crack length, a general trend of increasing TGO thickness and maximum crack length was found with respect to exposure time. The mechanical properties of the bond coat were also found to be influenced by the thermal exposure.

Abstract To investigate the properties of thermal sprayed glass-composite coatings, experimental measurements have been carried out. The coating demands such as thermal, electrical or mechanical resistance need to be optimised along with stress-minimizing procedures in order to reach high-quality surface coatings. The relevant properties aging, oxidation resistance and bond strength to the substrate are evaluated. Different material combinations were sprayed using the VPS, LPPS and HVOF processes, which combine several parameter sets and substrates. The results of the investigations show that coating properties such as conductivity, thermal expansion, porosity and surface roughness can be well directed influenced by controlling the mixture of the powder material, the pretreatment of the powder and particle size distribution.

Abstract As a result of the need to extend the storage life of rocket motors, new emphasis has been placed upon the effect of cracks developed during storage and transport on the integrity of motor performance. In addressing this need, the authors have developed a three specimen test procedure for evaluating the stress intensity factor (SIF) distribution for cracks through the thickness of rubberlike materials near to and within simulated bond lines between the motor grain and its liner. A refined frozen stress method is employed for making measurements, and preliminary studies have shown that, for cracks in the bond line under both Mode I and Mixed Mode loads, the stress intensity factors are elevated primarily due to bond line residual stresses and may not be affected by modulus mismatch.

Abstract The paper analyzed microstructure and property of WC-17Co coatings sprayed by High Velocity Oxygen/Air Fuel Spray under three kinds of spray conditions, which are HVOF, HVO-AF and HVAF. Coatings bond well with the substrate. The average bonding strength exceeds 70Mpa. Coatings are dense and hard, and the average porosity is about 1%. Microhardness of coatings is between HV10000.2 and HV12000.2. Coatings are mainly composed of WC with little W2C and Co3W3C. With the increasing of Nitrogen, decarburization of WC was reduced.

Purpose – The rostrum of a paddlefish provides hydrodynamic stability during feeding process in addition to detect the food using receptors that are randomly distributed in the rostrum. The exterior tissue of the rostrum covers the cartilage that surrounds the bones forming interlocking star shaped bones. Design/methodology/approach – The aim of this work is to assess the mechanical behavior of four finite element models varying the type of formulation as follows: linear-reduced integration, linear-full integration, quadratic-reduced integration and quadratic-full integration. Also presented is the load transfer mechanisms of the bone structure of the rostrum. Findings – Conclusions are based on comparison among the four models. There is no significant difference between integration orders for similar type of elements. Quadratic-reduced integration formulation resulted in lower structural stiffness compared with linear formulation as seen by higher displacements and stresses than using linearly formulated elements. It is concluded that second-order elements with reduced integration and can model accurately stress concentrations and distributions without over stiffening their general response. Originality/value – The use of advanced computational mechanics techniques to analyze the complex geometry and components of the paddlefish rostrum provides a viable avenue to gain fundamental understanding of the proper finite element formulation needed to successfully obtain the system behavior and hot spot locations.

Original Objectives: 1. Purify and biochemically characterize RB60 orthologs in higher plant chloroplasts; 2. Clone the gene(s) encoding plant RB60 orthologs and determine their structure and expression; 3. Manipulate the expression of RB60; 4. Assay the effects of altered RB60 expression on thylakoid biogenesis and photosynthetic function in plants exposed to different light conditions. In addition, we also examined the gene structure and expression of RB60 orthologs in the non-vascular plant, Physcomitrella patens and cloned the poly(A)-binding protein orthologue (43 kDa RB47-like protein). This protein is believed to a partner that interacts with RB60 to bind to the psbA5' UTR. Thus, to obtain a comprehensive view of RB60 function requires analysis of its biochemical partners such as RB43. Background & Achievements: High levels of sunlight reduce photosynthesis in plants by damaging the photo system II reaction center (PSII) subunits, such as D1 (encoded by the chloroplast tpsbAgene). When the rate of D1 synthesis is less than the rate of photo damage, photo inhibition occurs and plant growth is decreased. Plants use light-activated translation and enhanced psbAmRNA stability to maintain D1 synthesis and replace the photo damaged 01. Despite the importance to photosynthetic capacity, these mechanisms are poorly understood in plants. One intriguing model derived from the algal chloroplast system, Chlamydomonas, implicates the role of three proteins (RB60, RB47, RB38) that bind to the psbAmRNA 5' untranslated leader (5' UTR) in the light to activate translation or enhance mRNA stability. RB60 is the key enzyme, protein D1sulfide isomerase (Pill), that regulates the psbA-RN :Binding proteins (RB's) by way of light-mediated redox potentials generated by the photosystems. However, proteins with these functions have not been described from higher plants. We provided compelling evidence for the existence of RB60, RB47 and RB38 orthologs in the vascular plant, Arabidopsis. Using gel mobility shift, Rnase protection and UV-crosslinking assays, we have shown that a dithiol redox mechanism which resembles a Pill (RB60) activity regulates the interaction of 43- and 30-kDa proteins with a thermolabile stem-loop in the 5' UTR of the psbAmRNA from Arabidopsis. We discovered, in Arabidopsis, the PD1 gene family consists of II members that differ in polypeptide length from 361 to 566 amino acids, presence of signal peptides, KDEL motifs, and the number and positions of thioredoxin domains. PD1's catalyze the reversible formation an disomerization of disulfide bonds necessary for the proper folding, assembly, activity, and secretion of numerous enzymes and structural proteins. PD1's have also evolved novel cellular redox functions, as single enzymes and as subunits of protein complexes in organelles. We provide evidence that at least one Pill is localized to the chloroplast. We have used PDI-specific polyclonal and monoclonal antisera to characterize the PD1 (55 kDa) in the chloroplast that is unevenly distributed between the stroma and pellet (containing membranes, DNA, polysomes, starch), being three-fold more abundant in the pellet phase. PD1-55 levels increase with light intensity and it assembles into a high molecular weight complex of ~230 kDa as determined on native blue gels. In vitro translation of all 11 different Pill's followed by microsomal membrane processing reactions were used to differentiate among PD1's localized in the endoplasmic reticulum or other organelles. These results will provide.1e insights into redox regulatory mechanisms involved in adaptation of the photosynthetic apparatus to light stress. Elucidating the genetic mechanisms and factors regulating chloroplast photosynthetic genes is important for developing strategies to improve photosynthetic efficiency, crop productivity and adaptation to high light environments.

Background Brucellaspecies are Gram-negative, facultative intracellular bacterial pathogens that cause brucellosis, the most prevalent zoonosis worldwide. Brucellosis is characterized by increased abortion, weak offspring, and decreased milk production in animals. Humans are infected with Brucellaby consuming contaminated milk products or via inhalation of aerosolized bacteria from occupational hazards. Chronic human infections can result in complications such as liver damage, orchitis, endocarditis, and arthritis. Brucellaspp. have the ability to infect both professional and non-professional phagocytes. Because of this, Brucellaencounter varied environments both throughout the body and within a cell and must adapt accordingly. To date, few virulence factors have been identified in B. melitensisand even less is known about how these virulence factors are regulated. Subsequently, little is known about how Brucellaadapt to its rapidly changing environments, and how it alternates between acute and chronic virulence. Our studies suggest that decreased concentrations of cyclic dimericguanosinemonophosphate (c-di-GMP) lead to an acute virulent state and increased concentrations of c-di-GMP lead to persistent, chronic state of B. melitensisin a mouse model of infection. We hypothesize that B. melitensisuses c-di-GMP to transition from the chronic state of an infected host to the acute, virulent stage of infection in the placenta where the bacteria prepare to infect a new host. Studies on environmental pathogens such as Vibrio choleraeand Pseudomonas aeruginosasupport a mechanism where changes in c-di-GMP levels cause the bacterium to alternate between virulent and chronic states. Little work exists on understanding the role of c-di-GMP in dangerous intracellular pathogens, like Brucellathat is a frequent pathogen in Israeli domestic animals and U.S. elk and bison. Brucellamust carefully regulate virulence factors during infection of a host to ensure proper expression at appropriate times in response to host cues. Recently, the novel secondary signaling molecule c-di-GMP has been identified as a major component of bacterial regulation and we have identified c-di-GMP as an important signaling factor in B. melitensishost adaptation. C. Major conclusions, solutions, achievements 1. The B. melitensis1453 deletion mutant has increased c-di-GMP, while the 1520 deletion mutant has decreased c-di-GMP. 2. Both mutants grow similarly in in vitro cultures; however, the 1453 mutant has a microcolony phenotype both in vitro and in vivo 3. The 1453 mutant has increased crystal violet staining suggesting biofilm formation. 4. Scanning electron microscopy revealed an abnormal coccus appearance with in increased cell area. 5. Proteomic analysis revealed the 1453 mutant possessed increased production of proteins involved in cell wall processes, cell division, and the Type IV secretion system, and a decrease in proteins involved in amino acid transport/metabolism, carbohydrate metabolism, fatty acid production, and iron acquisition suggesting less preparedness for intracellular survival. 6. RNAseq analysis of bone marrow derived macrophages infected with the mutants revealed the host immune response is greatly reduced with the 1453 mutant infection. These findings support that microlocalization of proteins involved in c-di-GMP homeostasis serve a second messenger to B. melitensisregulating functions of the bacteria during infection of the host.
