Abstract:
Probiotics can potentially prevent and treat diseases. We examined the inhibitory activity of bacteriocin-like inhibitory substances (BLISs) from potentially probiotic lactobacilli and streptococci on Candida albicans and non-Candida albicans clinical isolates from women with vulvovaginitis. Using agar well diffusion assays, BLISs inhibited both Candida albicans and non-Candida albicans isolates. The BLIS from L. pentosus isolates had the highest anti-Candida activity (33/45; 73.3%), followed by BLISs from isolates of L. paracasei subsp. paracasei (31/45; 68.9%), L. rhamnosus I (30/45; 66.7%), L. delbrueckii subsp. lactis I (30/45; 66.7%), and S. uberis II (30/45; 66.7%). Upon characterization according to the retained activity under variable physical and chemical conditions, the BLISs showed stability against heat, pH, and surfactants, but were protease-sensitive, which suggests a proteinaceous nature of the active substances. Using crystal violet assays, the BLISs reduced the Candida biofilm biomass significantly as compared to a control group that lacked BLISs. In vivo testing of the antagonistic activity was performed using the Galleria mellonella (G. mellonella) larvae model. BLISs significantly improved survival in G. mellonella larvae treated with Candida isolates on the first, second, and seventh days, as compared to larvae inoculated with Candida only (p < 0.01). The results show that BLISs can be used as biotherapeutic agents in vulvovaginal candidiasis.

ObjectiveTo assess the efficacy and safety of belimumab in paediatric versus adult patients with systemic lupus erythematosus (SLE).MethodsWe performed across-study comparisons of patients with active SLE who received belimumab or placebo, plus standard therapy, in PLUTO (paediatric phase II) and BLISS-52, BLISS-76, BLISS-NEA and EMBRACE (adult phase III). Analysed efficacy data included Week 52 SLE Responder Index (SRI)-4 response rate (EMBRACE: SRI with modified Systemic Lupus Erythematosus Disease Activity Index (SLEDAI) proteinuria scoring (SRI-S2K)); SRI-4 response rate (EMBRACE: SRI-S2K) according to baseline disease activity indicators (Safety of Estrogens in Lupus Erythematosus National Assessment-Systemic Lupus Erythematosus Disease Activity Index (SELENA-SLEDAI) score; anti-dsDNA/C3/C4 levels); Week 52 SRI-6 response rate; and time to first severe flare (SELENA-SLEDAI Flare Index) over 52 weeks. Safety data were compared for all aforementioned studies along with adult LBSL02 (phase II) and BLISS-SC (phase III).ResultsSRI-4 response rates were similar across the paediatric and adult studies; more belimumab-treated patients achieved SRI-4 responses versus placebo (PLUTO: 52.8% vs 43.6%; BLISS-52: 57.6% vs 43.6%; BLISS-76: 43.2% vs 33.8%; BLISS-NEA: 53.8% vs 40.1%; EMBRACE: 48.7% vs 41.6%). Across all studies, SRI-4 response rates were generally greater in patients with baseline SELENA-SLEDAI scores ≥10 than in patients with baseline SELENA-SLEDAI scores ≤9. A similar proportion of belimumab-treated patients achieved SRI-6 across all studies (PLUTO: 41.2%; BLISS-52: 46.2%; BLISS-76: 33.1%; BLISS-NEA: 43.9%; EMBRACE: 37.5%). Belimumab reduced the risk of severe flare versus placebo in all studies. The incidence of adverse events was similar across all studies.ConclusionsThese analyses demonstrate consistent efficacy and safety of belimumab plus standard therapy across paediatric and adult patients with SLE.Trial registration numbersPLUTO (NCT01649765); BLISS-52 (NCT00424476); BLISS-76 (NCT00410384); BLISS-NEA (NCT01345253); EMBRACE (NCT01632241); BLISS-SC (NCT01484496); and LBSL02 (NCT00071487).

Abstract: This study aims to create and define the Bliss Yoga Malang logo and standardize its application through applicative media. The design uses the Designing Brand Identity method with stages 1). Conducting Research, 2). Clarifying Strategy, 3). Designing Identity, 4). Creating Touchpoints, and 5). Managing Assets. Visual identity elements in the form of logos, supporting graphics, and GSM (Graphic Standard Manual) to facilitate the design of applicable promotional media. Observation results show that Bliss Yoga is a yoga class that was only established in early 2021 and does not yet have a logo. The process of designing the Bliss Yoga Malang logo begins with a mind mapping of Bliss Yoga Malang to form keywords that can represent Bliss Yoga Malang, namely women, calm, and flexible. The logotype is created using the Nunito font which belongs to the Sans Serif font category. Visual identity is manifested in three categories of application media to form a signature, namely 1) stationery set, 2) promotion media set, and 3) merchandise set to give an 'impression and perception' of Bliss Yoga through the administrative process and correspondence, attracting more enthusiasts and emotionally bringing Bliss Yoga's target consumers closer. Managing Assets is facilitated by the Graphic Standard Manual. This visual identity is expected to introduce Bliss Yoga in Malang with special customer segmentation in terms of gender, age, and appearance (hijab). Keywords: visual identity; logo; class yoga Abstrak: Penelitian ini bertujuan membuat dan menetapkan logo Bliss Yoga Malang beserta standarisasi penerapannya melalui media aplikatif. Perancangan menggunakan metode Designing Brand Identity dengan tahapan 1). Conducting Research, 2). Clarifying Strategy, 3). Designing Identity, 4). Creating Touchpoints, dan 5). Managing Asset. Elemen visual identity berupa logo, supporting graphic, dan GSM (Graphic Standard Manual) agar mempermudah perancangan media promosi aplikatif. Hasil observasi menunjukkan bahwa Bliss Yoga merupakan kelas yoga yang baru berdiri pada awal tahun 2021 dan belum memiliki logo. Proses perancangan logo Bliss Yoga Malang diawali dengan mind mapping mengenai Bliss Yoga Malang untuk membentuk kata kunci yang dapat merepresentasikan Bliss Yoga Malang, yaitu perempuan, tenang, dan fleksibel. Logotype dibuat menggunakan font Nunito yang termasuk kategori font Sans Serif. Visual identity diwujudkan dalam tiga kategori media aplikasi agar terbentuk sebuah ciri khas atau signature, yaitu 1) stationery set, 2) promotion media set, dan 3) merchandise set untuk memberi ‘kesan dan persepsi’ terhadap Bliss Yoga melalui proses administrasi dan surat-menyurat, menarik peminat lebih banyak serta mendekatkan target konsumen Bliss Yoga secara emosional. ObjectiveWe evaluated the discriminant capacity of the Lupus Low Disease Activity State (LLDAS) in post-hoc analysis of data from the BLISS-52 and BLISS-76 trials of belimumab in systemic lupus erythematosus (SLE).MethodsLLDAS attainment, discrimination between belimumab and placebo arms, and the effects in subgroups with high disease activity at recruitment were evaluated at week 52 using appropriate descriptive statistics, χ2 test and logistic regression.ResultsAt week 52, for belimumab 10 mg/kg, 17.0% and 19.3% of patients who achieved a Systemic Lupus Erythematosus Responder Index-4 also attained LLDAS in BLISS-52 and BLISS-76, respectively. Significantly more patients attained LLDAS on belimumab 10 mg/kg compared with placebo (12.5% vs 5.8%, OR 2.32, p=0.02 for BLISS-52; 14.4% vs 7.8%, OR 1.98, p=0.04 for BLISS-76). In a subgroup analysis, the difference in week 52 LLDAS attainment between belimumab 10 mg/kg and placebo was greater in patients who had higher disease activity at baseline, compared with the overall group.ConclusionsLLDAS was able to discriminate belimumab 10 mg/kg from placebo in the BLISS-52 and BLISS-76 trials. Our findings support the validity of LLDAS as an outcome measure in SLE clinical trials.

Abstract:
User-initiated security breaches are common and can be very costly to organizations. Information security training can be used as an effective tool to improve users’ secure behavior and thus alleviate security breaches. Via the lens of learning, working memory, and cognitive load theories, this research examines how to improve the effectiveness of security training through decreasing extraneous stimuli in the presentation of online security training. The authors conducted a realistic laboratory experiment to examine the influence of training with different levels of extraneous stimuli on secure behavior. They found that training presented with low levels of extraneous stimuli improved secure behavior more than training presented with high levels. The results question the effectiveness of elaborate training programs, and rather suggest that simple, direct training modules are most effective.
