Full textLa bioinformatique fait maintenant partie intégrante de la recherche en génomique, car elle permet des analyses de bases de données larges et complexes. Conventionnellement, ces analyses impliquent l'utilisation de logiciels sophistiqués et sont généralement faites par des personnes expérimentées en bioinformatique qui utilisent des ressources informatiques adéquates.Les plateformes de séquençage haut débit d'ADN ont démocratisé le séquençage du génome, le rendant ainsi accessible aux biologistes. Pour de nombreux biologistes, ce sera leur première incursion dans les domaines de la bioinformatique et de la génomique. Par conséquent, ils ne sont probablement pas familiers avec la bioinformatique ou n'ont pas les ressources informatiques nécessaires afin d'analyser les résultats. Pour ces biologistes, l'utilisation des plateformes de séquençage haut débit permet l'obtention abordable de données génomiques, mais n'offre pas les outils pour les analyser facilement. Le développement de logiciels ciblant les chercheurs ayant une expertise en bioinformatique limitée ou avec peu de ressources permettrait de combler cet écart.Cette dissertation décrit un paradigme visant à réduire, voire même à fermer, l'écart entre l'analyse du génome et le biologiste. En utilisant ce paradigme, j'ai développé des outils informatiques pour trois tâches facilitant l'analyse génomique : [i] l'évaluation de l'assemblage du génome, [ii] l'affichage et l'analyse intégrée des données génomiques, et [iii] l'obtention de connaissances biologiques utilisant de l'information publique. Le premier outil que j'ai développé était cgb, un programme qui crée des navigateurs personnalisés « UCSC Genome ». Il permet aux biologistes d'utiliser ces navigateurs pour évaluer les séquences obtenues à partir de plateformes de séquençage haut débit. L'utilisation de cgb lors d'une étude génomique comparative de Clostridium difficile nous a permis d'identifier des marqueurs diagnostics d'ADN associés à la gravité de la maladie et de démontrer que son pan-génome est plus grand qu'estimé précédemment. Ensuite, j'ai développé contiGo, un outil d'usage général pour réviser les assemblages de séquences génomiques par l'intermédiaire d'un navigateur web. Cette application permet aux biologistes de contourner la nécessité d'installer un logiciel, de satisfaire les exigences de l'équipement informatique, et de télécharger des larges bases de données. Conjointement avec cgb, ce programme nous a permis d'évaluer la performance de la plateforme de séquençage haut débit Roche/454 Genome Sequencer FLX, à travers cinq installations de séquençage, ainsi qu'à générer une séquence génomique de grande qualité du champignon Ophiostoma novo-ulmi. Finalement, j'ai développé BL!P, un programme pour automatiser les recherches BLAST NCBI et pour explorer les résultats obtenus dans une interface dynamique. Ce programme a été inspiré par mon travail sur la caractérisation du génome d'une souche pathogène et multi résistante d'Escherichia fergusonii, et pour laquelle cgb et contiGo ont également été utilisés dans l'analyse des données. Ces applications ont été utilisées dans d'autres projets de génomique par des utilisateurs possédant un éventail de compétences et de ressources bioinformatiques. D'autres domaines scientifiques générant des multitudes de données pourraient bénéficier d'un paradigme similaire de développement de logiciel informatique.

A formação do biólogo comumente não apresenta espaços destinados a reflexão ética da atuação deste e isso pode gerar uma defasagem em relação a sua atuação. O presente trabalho propõe então entrevistas semiestruturadas com alunos de graduação em Ciências Biológicas em último ano para que se possa avaliar que questões éticas são identificadas por eles. Após a análise de suas falas, pode observar a identificação de questões problemáticas pelos estudantes, que versavam principalmente sobre a formação do biólogo e sua atuação. Pensando sobre a formação, as questões sobre uso animal (fins didáticos), espaços para reflexão ética, problemas de relações entre alunos-alunos e alunos-orientadores foram apontados como importantes. Com relação a atuação do profissional biólogo as questões trazidas foram sobre a valorização do biólogo, as possíveis atuações dele, e o foco sobre a pesquisa. Essas variadas questões problemáticas mostram que funcionamentos dos estudantes estão sendo negligenciados ou sendo afetados pela falta de reflexões éticas sobre a natureza da própria atuação do biólogo. Propiciar tais reflexões e encarar tais questões como de cunho ético pode ser o caminho para uma melhor inserção e valorização do biólogo na sociedade
The formation of the biologist usually does not presents spaces designated to the ethical reflection of the performance of this one and this can generate a gap related to its performance. The present work then proposes semi-structured interviews with undergraduate students in Biological Sciences in the last year so they can evaluate what ethical issues are identified by them. After analyzing their speeches, we can observe the identification of problematic issues by the students, which focused mainly on the formation of the biologist and his performance. Thinking about the formations, questions about animal use (didactic purposes), spaces for ethical reflection, and problems of relations between students and students and professors were pointed out as important issues. With regard to the professional biologists work, the questions raised were about the biologist's appreciation, his possible actions, and the focus on the research. These varied issues shows that student’s functioning are being neglected or being affected by the lack of ethical reflections about the nature of the biologist's own performance. To foster such reflections and to address such issues as ethical in nature may be the way to better integrate and value the biologist in society

Full textThe emergence of "molecular biology" is a complexe process including the reconstruction of disciplinar boundaries and the transformation of research practices. In france, the creation of a new governmental agency - the dgrst - provided the main impulse. During the 60's, it supported the rare groups of biologists already studying the structure of nucelic acids or protein synthesis. Among them, the biochemists and geneticists working at the pasteur institute played the leading part, defining and implementing the dgrst's programs. This work describe the formation of this group, its contribution to the birth of regulatory genetics and the policy it set up. With the history of twenty french laboratories, it reveals the steps leading to the formation of new research communities. The comparison with the american and british contexts shows the part influnce of french traditions in biology. The thesis emphasizes the fact that the creation of molecular biology didn't proceed from simple knoweldge transfer but that it was rooted in the reshaping of scientific objects, based on judgements determined by laboratory cultures, experimental practices, collaboration networks and institutional constraints

Full textLandfill leachate often contains contaminants that can be harmful if they are discharged into the environment without proper treatment. At SÖRAB’s (Söderhalls renhållningsverk AB) waste treatment site in Löt there is an active landfill for non-hazardous waste that generates leachate containing high levels of nitrogen in the form of ammonium. If large amounts of nutrients leak out into the environment it can have a negative impact on biological processes and cause problems such as eutrophication and acidification. The waste treatment site is until 31 December 2014 under a trial period where the owner company is obliged to, amongst other things, characterise the chemical composition in the runoff from the treatment areas and leachate from the landfill. The characterisation is done in order to find the best treatment methods for the different streams of water. Due to the high levels of nitrogen in the leachate from the landfill at Löt waste disposal plant the current regulations that are set up for the plant have been difficult to follow. To avoid negative environmental impact on the receiving waters and surrounding lands SÖRAB has constructed a pilot treatment plant for continuous biological treatment (KBR) where the nutrient rich leachate will be treated to reduce the total emissions of nitrogen from the waste disposal plant. The KBR-plant uses nitrification and denitrification to remove nitrogen from the leachate. This project is aimed at optimising SÖRAB’s plant for continuous biological treatment with respect to removal of nitrogen. An important part of the project was to examine the effect of different parameters on the removal of nitrogen in the KBR-plant. The parameters examined and optimised were retention time in the treatment basins, water temperature, phosphorus demand, and the need of external carbon source. The results showed that for a operational flow of 420m3/d, complete nitrification and the denitrification could be achieved for temperatures above 14°C if the phosphate content was more than 1mg/L in the water. Furthermore, a need for an external carbon source in the denitrification basin was identified. It was found that the need of an external carbon source corresponded to a COD-addition of four times the content of nitrate in the incoming water. With these parameter settings almost 100% of the inorganic nitrogen in the water could be separated in the KBR-plant. The removal of total nitrogen was 66-85%. In the project a comparison was also done between the KBR treatment method and other available treatment methods for nitrogen removal from nutritious leachate. The comparison showed that the KBR treatment method is a very good alternative for removal of nitrogen with regard to treatment efficiency and costs. When both removal efficiency and costs were taken into account only traditional wastewater treatment plants could compete with the KBR treatment method used in this project.

Full textThis study aims to investigate the knowledge and interest in biodiversity and conservationamong sixth graders. The inquiry was made through a digital survey that was sent out to 44pupils in total from two different schools in the muncipality of Halmstad. One of theresponding classes participated in the project LIFE-Goodstream with a field excursion,inventory of aquatic organisms by heaving and discussion about the ecological benefits ofartificial wetlands during 2020. The survey consisted partly of questions with one or severalalternatives, partly of open questions with possibility to write own answers. Additionally, thestudents were asked to grade their attitudes towards biology as a school subject. Thequestions concerned both the students’ theoretical knowledge and their relation tobiodiversity. Participants in LIFE-Goodstream did also comment on their experiences of theexcursion. From these answers, conclusions about knowledge and interest could be drawn.The students generally found it difficult to define biodiversity and were unsure of themeaning of conservation. Nevertheless, the majority consider it as important and want toknow more about conservation biology. The study shows no significant correlation betweenparticipation in the field excursion and knowledge or interest. However, the students with more regular outdoor activities tend to appreciate nature more and be more interested infurther education. The study indicates a need for more education in biodiversity and more information aboutfeasible measures for conservation. The current curriculum should possibly involve morepractical moments and put the education of single concepts in a context, which could create adeeper understanding and a motivation to learn more about how they can act forsustainability.

Full textDet eksisterer mange kjente metoder for å etterligne menneskelig tenkegang eller utøve kunstig intelligens. Et interessant anvendelsesområde for slike metoder er i musikken, siden dette er en kunstform uten klare regler eller absolutte sannheter. Uten regler og sannheter er det heller ikke en korrekt måte å lage musikk på. Arbeidet beskrevet i denne rapporten går ut på å bygge et system for å tolke musikalsk input fra en musikkutøver. Til dette blir det forsøkt benyttet nevrale nettverk. Et nevralt nettverk kan utføre denne oppgaven ved å trenes opp til å gjenkjenne hvilke akkorder som blir spilt. Systemet blir testet ved å koble det sammen med programmet Happysound (Utvliket i prosjektoppgaven høst 05). Dette er et program som benytter fuzzy logikk for å genererer musikalske soloer. Ved å koble et nevralt nettverk til dette fåes et system som tar inn akkompagnement fra en medspiller og spiller av soloer som passer til dette. Resultatene som presenteres viser at nevrale nettverk er en god metode for musikalske formål men at det legges begrensninger av hvor mange forskjellige akkordrepresentasjoner ett enkelt nevralt nettverk klarer å kjenne igjen. Til slutt fremlegges en utvidelse for å overkomme disse begrensningene.

Genom inventering avKillingskogen undersöktes områdets potentiella naturvärden.Inventeringen visar att en mosaikartad skog har en högre potential för känsliga arter ochbiologisk mångfald. Som en del av studien så föreslås åtgärder för att bevara eller höjanaturvärdena i området samt metoder för att höja rekreationsvärdena.

Full textOECD (Organisation for Economics Co-operation and Development) is an international organization which works for economical growth and increased sustainability through establishments of standards. A well-established OECD standard is the Closed Bottle 301D which measures the biodegradability in organic components in an aerobic and aqueous environment. The method has been applied for some selected hydraulic fluids, as a result of the increased interest of switching to “green” hydraulic fluids to decrease the damage and cost which is caused by oil spill. The purpose is to evaluate the biodegradability of selected hydraulic fluids through the standard OECD, Closed Bottle 301D. The thesis should be able to give increased knowledge and insight in the method for future evaluations. One of the principles behind Closed Bottle 301D is to use closed BOD-bottles where the process of biodegradation is taking place. Moreover, is it required that the hydraulic fluids have biodegraded to a minimum of 60% in 28 days in order to be classified as biodegradable according to the standard. Certain conditions are required for the hydraulic fluids in order to give them the chance to biodegrade in a similar way as in nature. By inoculating the BOD-bottles with microorganisms from a sludge source and adding other necessary minerals, an environment where the hydraulic fluids are given the right conditions in order to biodegrade is being created. BOD (Biological Oxygen Demand) can be used to detect the amount of biodegradable substances and how fast the microorganisms utilizes oxygen in a given amount of water. A comparison between the utilized amount of oxygen (BOD) and the theoretical amount ThOD (Theoretical Oxygen Demand) is used to describe the hydraulic fluids ability to biodegrade. Through this experiment it turned out that no one of the selected hydraulic fluids achieved the required biodegrading value in order to be classified as biological biodegradable according to the OECD method. This doesn’t necessarily mean that the fluids cannot biodegrade, but that a longer period of time is required for that to happen. The experiment gave results which corresponded with the expected outcome regarding the biodegradation process of the fluids, which strengthens the OECD method to be a reliable method in analysing oil’s ability to biodegrade.

Full textThe increasing exploitation of land and the changes in land use over the last few hundred years has led to a marked reduction in the biodiversity of the landscape. Species populations have diminished in number or even gone locally extinct due to habitat loss. Research has shown that military training areas have become a refuge for many endangered species, partly due to the fact that these areas have been spared from exploitation of the land and partly due to the fact that these species have thrived as a result of the disturbance of habitat caused by military activity. This study aims to clarify which types of disturbance have had a positive effect on the biodiversity of Torhamns udde in South East Blekinge through history. In this investigation biodiversity is analyzed at the landscape and species levels. Earlier results from a pollen analysis north of the study area show that the region has been affected by land use over a period of several thousand years. The military activity that has taken place in the area over the last hundred years, has contributed to the preservation of the unique natural environment that exists there today. An important conclusion from this study is that changes in biodiversity are strongly connected with changes in land use. The present composition in the flora and fauna may thus largely be explained by the historical land use. In order to positively influence the biodiversity of a military training area, it is important to have a palaeoecological approach, when conservation and management plans are being set up.

Full textThe time used by physicists is directed against life. More often than not, the time referred to by philosophers has been extraneous to life. Thus it became all the more necesary to go about in quest of initial temporality as all those images made up by time merely trengthened the primary role of reason and the fictions coming along with it. Temporality is a force of synthesis and organization resulting from the confrontation between the ideal character of the temporal structure and the existential character of facts. Temporality finds in chronobiology the very expression of this radical ability to achieve the synthesis of the different clocks which compose it. Living means changing and this sequence of events gives temporality both its richness and its mystery, for a living creature is never true to himself. Temporality is this intimate relation fron self to self which gives the organism its autonomy and its freedom. Both philosophers and scientists consider time as a regulating concept whose purpose is the thorough unification of what our reason knows. This nightmare has to vanish so as to make room for a genuine metaphysical grasp of temporality. If ever there exists a metaphysical illusion in the approach to the notion of time, the fault lies with the imaginary of ambiguity for which time cannot be described as a sheer invention of thought. True temporality is the expression of a biological pecularity, a form of transcendence corresponding to that palpitation which is the origin of life and freedom

Full textThis article deals with the woodlands as a biological cultural heritage and how it is taken care of in the protected woodlands in Skåne, in the south of Sweden, especially the deciduous trees which is dominated by beech. High biological values have emerged as a result of grazing and different kinds of forestry. The protected woodlands have been ripped of its cultural influences which changes the biological values on which the original reasons for protection was founded.

Full textModern methods for managing pine (Pinus sylvestris) create homogenized forests. This decreases nature’s potential for biodiversity and might threaten species in need of different types of milieu. The main purpose of this study was to investigate how important older pine trees are for biodiversity. In the Hall-Hangvar Reserve in the north-west part of Gotland, insects collected from traps showed that more species were found in old or dead trees compared to younger pine trees. A statistically significant difference was found for Coleoptera (beetles). The taxons of greatest interest for this study were Coleoptera and Hymenoptera (wasps). Certain families of Hymenoptera use ducts made by larvae from some families of Coleoptera.These larvae also serve as prey. Relevance concerning enviromental importance to species and diffrenences in inhabiting the three stages of pine trees was of importance.

Full textDuring the summer 2007 was a scrubber tested at Gruvön papper mill in Grums. The scrubber reduced NOx with 90 % in flue gas. NOx was transferred from the flue gas to a scrubber liquid as nitrate and nitrite. The scrubber liquid needs to be purified from nitrate and nitrite.
One possible solution is to clean the scrubber liquid in Gruvön biologic cleaning construction.
Microorganisms in the biologic cleaning construction need to assimilate nitrogen. There are environments free from oxygen in the cleaning construction. Microorganisms can reduce nitrate in environments free from oxygen.
At the implementation was a labmodel built of the two first steps from Gruvön papper mill. Wastewater was collected from Gruvön papper mill. The wastewater was dosed with salts of nitrate and nitrite and pumped into the labmodel.
The results showed that nitrate and nitrite can be reduced in content with help of the biological cleaning construction.

Full textEtt generellt problem i dagens svenska gymnasieskola är att elever ofta använder vardagliga förklaringar istället för vetenskapliga när de ska förklara naturvetenskapliga teorier och begrepp. När elever ska förklara orsaken till att biologisk evolution sker, uppkommer olika alternativa idéer, idéer som inte är förenliga med vetenskapliga förklaringar. Denna studie beskriver vilka alternativa idéer som elever ger när de skriftligt besvarar en uppgift innehållande ett evolutionsbiologiskt fenomen. De förklaringar eleverna ger på uppgiften diskuteras och utvecklas i intervjuer, för att öka kunskapen och förståelsen för vilka uppfattningar gymnasieelever har om biologisk evolution. Den vanligaste alternativa idén som framkommer när den skriftliga uppgiften löses är att evolution drivs för att ett behov finns(giraffen behöver utvecklas för att klara sig). Andra förekommande alternativa idéer är inlärning (giraffen sträcker på sig och därför blir halsen längre) och anpassning (giraffen anpassar sig till en ny omgivning). Fem begrepp är centrala för att förstå helheten i evolutionsteorin; variation, överlevnad, ärftlighet, reproduktion och anpassning. Variation visar sig vara ett nyckelbegrepp för elevens förmåga att frångå en alternativ idé om evolution och uppnå en högre grad av vetenskaplighet i sin förklaring.

Full textSweden and its municipal wastewater treatments plants, has as other Baltic countries, problem meeting the requirement on nitrogen compounds discharged via municipal wastewater. Raising the temperature of the wastewater, in the biological nitrogen removing process, is a possible alternative in order to achiev a higher reduction efficiency during the cold season when the wastewater has a bad cleaning outcome because of the low temperature. Hammarby Sjöstadsverk has, together with Oskarshamns municipality, developed a project named ITEST,-( Increased Technology and Efficiency in Sewage Treatment), where the project is based on studying the temperature influence on the biological nitrogen reduction process. The aim with this thesis, has been to study how the temperature, but also other important factors, influence the biological nitrogen reduction process, and then mainly the nitrification process. The main objective has been to run tests in a pilot plant and to do a literature review about the temperature influence on the nitrogen reduction process and the influence on the environment. The performance of the project was at Sjöstadsverket where two testing line had been set up. The testing plant concisted of one line with adjusting temperature and another line acted as a reference line. Measurements have been performed from both lines, where the outgoing concentrations of ammonia, nitrate and totalnitrogen has been studied. From the studies a discussion of the temperature influence of the denitrification and the nitrification process has been fulfilled. The result of the literature study part of thesis thesis shows that the nitrogen reduction rate is dependent on the temperature of the wastewater and it thought to be possible to get a better treatment in the biological nitrogen removal step if the temperature can be higher. The test in the pilot plant didn´t show a clear result. The nitrogen reduction rate showeds from both lines quite similar results despite the differences of both lines temperature. The difference of temperature in the two lines was probably too small to show a clear result.

Full textA biological dryer was developed to provide a discharge system for residues and to create a new design to enhance usability. The current version of the product, called a biological dryer, contains no discharge system. This means that the discharge operation today is performed by the user, with the assistance of a separate product which uses air to collect the residues and put it in a bag. The work was conducted at the University of Skövde in cooperation with the firm Pegil Innovations AB, which is conducted in Skultorp. The project began with the analysis of the existing dryer’s func-tions. Information was collected about prospective users’ work methods, existing solutions to similar discharge systems and studies that specializes in design. An idea generation was conducted to identify possible solutions to the discharge system and the design of the dryer. The concepts that arose were measured using a weighted concept selection matrix and the design alternatives that arose were evaluated with a task analysis. The final conceptual solution uses the already installed vacuum pump as a means of transport for the residues. The system was tested with a vacuum pump that creates 100 mBar which was loaned by the company. The results show that the sys-tem works for the purpose. The design and the discharge system were modelled in Creo Parametric 2.0 to show the final results.

The work aims to expose, gather and deepen matters pertaining to mathematics and biology with an emphasis on education for high school students. There was a research on the motivational and knowledge area with students that study on the second and third years in the state high school Teacher Liceu Domingos Brasileiro to analyze the reaction of students when they observed the contents of matrices and exponential and logarithmic functions in a signification through biology. Also work shows an intersection between the worlds of both science-based examples and applications. It was found that both relate content efficacious, especially when compared to traditional said instruction.
O trabalho tem o intuito de expor, reunir e aprofundar assuntos pertinentes a matemática e a biologia com ênfase no ensino para os alunos do ensino médio. Foi realizado uma pesquisa no campo motivacional e de conteúdos com alunos do 2˚ e 3˚ anos do Colégio do estado Liceu Professor Domingos Brasileiro a fim de analisar a reação dos alunos quando observaram os conteúdos de matrizes e funções exponenciais e logarítmicas em uma significação através da biologia. Além disso, o trabalho nos mostra uma interseção entre as esferas das duas ciências baseada em exemplos e aplicações. Verificou-se que relacionar ambos os conteúdos se mostra eficaz, principalmente se comparado ao ensino dito tradicional.
