Abstract:
This dissertation describes the application and optimisation of an analytical technique, named thermally assisted hydrolysis and methylation - gas chromatography / mass spectrometry [THM.-GCMS]. THM.-GCMS is a modification of pyrolysis - gas chromatography / mass spectrometry [Py-GCMS] which involves an on-line derivatisation process known as thermally assisted hydrolysis and methylation. The method is based on the high temperature reaction of tetramethylammonium hydroxide with macromolecular materials containing functional groups susceptible to hydrolysis and methylation. During this research THM.-GCMS was used for the chemical characterisation of the most frequently-used binding media in 20th century art: oils, acrylics, alkyds and poly(vinylacetates). The major classes of binding media used in 20th century paints have been previously studied in conservation using a range of techniques, the most important being GCMS, Py-GCMS and Fourier transform infrared spectroscopy [FTIR]. However, this project was novel in that it demonstrated the possibility of chemically characterising a wide variety of both natural and synthetic binders found in modem works of art using a single technique, THM.-GCMS. Standard samples of oils, egg yolk, acrylics, poly(vinylacetates) and alkyds as well as samples containing these materials from nine painted works of art were successfully studied using THM.-GCMS. Among the art works studied, those by Marcel Duchamp and Pablo Picasso were investigated for the first time.

This project is focused on the study of synthetic materials used by Portuguese artists in the twentieth century. The aim of the study was the molecular characterization of vinyl and acrylic polymers used since the 1960s and the study of their photodegradation and evolution over time. In modern and contemporary art, water based synthetic emulsions have been widely used as painting binding media, especially acrylic and vinyl based emulsions. In some countries like Portugal where the first aqueous emulsions used as paint binders were the poly(vinyl acetate) – PVAc – ones, both as household and artist’s paints, the study of vinyl polymers is of particular importance. In Portugal these emulsions were used by outstanding artists like Joaquim Rodrigo and Ângelo de Sousa since the 1960s. In order to guarantee the preservation of the artworks for the future generations it is essential to understand how these paints will behave upon ageing. The molecular photodegradation of PVAc was studied and the presence of degradation products on reference and model samples, as well as on dated artworks was investigated. Paintings by Joaquim Rodrigo and Ângelo de Sousa, and also a hand painted catalogue for the Sabu vinyl artists’ paints, were selected as case studies. The characterization of micro-samples from the artworks provided useful information on the real evolution of the polymer binder over time. Accelerated photodegradtion studies were performed by the exposure of PVAc films on an ageing chamber equipped with a xenon-arc lamp. The behaviour of PVAc (homopolymer) was compared with a commercial emulsion(Vulcano V7) and paint reconstructions prepared in the laboratory. The results achieved by means of infrared microspectroscopy (FTIR) point out the considerably stable nature of these binding media as no molecular evidence was obtained concerning the formation of other carbonyl functions, the disappearance of the PVAc carbonyl or the formation of hydroperoxides. The polymer photodegradation process was further studied by means of size exclusion chromatography (SEC), following the molecular weight distribution and correspondent average molecular weights. It was concluded that no side-chain reactions are taking place and that main-chain scission is the foremost degradation mechanism, although not affecting the polymer performance significantly. The influence of pigments, such as titanium dioxide and iron oxide, on the polymer’s stability was also considered and it was concluded that they do not promote degradation, which might be explained due to pigment encapsulation in inert materials inhibiting its photocatalytic behaviour. The photodegradation quantum yield was determined as 7.4 x 10-8 at 313 nm for PVAc homopolymer; therefore, degradation is quantified and comparisons may be performed for different polymers. A parallel study, according to a similar experimental approach, was undertaken on the stability of poly(methyl methacrylate) – PMMA – used as acrylic sheet by the Portuguese artist Lourdes Castro.

Abstract:
I have carried out this thesis in the Biophysical Chemistry of Macromolecules and Colloids research group of the University of Barcelona. In the recent years, the research group have made significant contributions to the study of the conformational, binding, diffusion and reactivity properties of weak polyelectrolytes and biopolymers. On the one hand, the group has contributed to the design and development of the Site Binding Rotational Isomeric State (SBRIS) model for weak polyelectrolytes. They have also successfully used the SBRIS model to reproduce the complex experimental titration curves of polyethylenimine. On the other hand, the research group has done significant contributions in the study of biopolymer diffusion and reactivity in crowded media. They tracked the diffusion of α-chymiotrypsin in solutions crowded by different- sized dextran macromolecules. They rationalized the experimental results comparing them with on-lattice Monte Carlo (MC) simulations, observing a qualitative agreement between both sets of data. The present thesis follows the efforts of the research group in the development of theoretical and computational models for weak polyelectrolytes and biopolymers. I investigate different theoretical aspects of the conformational, ionization, elastic and diffusive properties of weak polyelectrolytes and biopolymers using theoretical and computational simulation techniques. The thesis includes six original peer-reviewed publications, which are distributed in six different chapters. Moreover, a complete description of the fundamental concepts and methodologies relevant for the thesis is also given. On the one hand, I focus in the complex interplay of the ionization and conformational degrees of freedom of weak polyelectrolytes using the recently developed Site Binding Rotational Isomeric State (SBRIS) model. A new analytical technique, the Local Effective Interaction Parameters (LEIP) method, is presented to solve the SBRIS model including long range intramolecular electrostatic interactions. The model is validated against constant pH Monte Carlo (MC) simulations. The SBRIS model is used to study the elastic response of a model weak polyelectrolyte. A new force regime is found for which (i) the force-extension curves are significantly dependent on the pH and the ionic strength values (ii) the polyelectrolyte charge is modified by the stretching force. The effect of charge fluctuation in the conformational and stretching properties of weak polyelectrolytes is studied comparing the results obtained with SGCMC simulation with those obtained with MC simulation with the same charge but keeping it constant. The conformational and elastic properties of the polyelectrolyte are observed to be significantly affected by the presence of charge regulation. On the other hand, I study the diffusive and ionization properties of biopolymers in solution with a high concentration of macromolecules. These conditions, known as macromolecular crowding, are characteristic of biological media where up to the 40% of the volume is occupied by macromolecules. The diffusion of two globular proteins, α-chymiotrypsin and streptavidin, is analysed in different crowding conditions with Brownian Dynamics simulations. A new coarse grained model is proposed, named as Chain Entanglement Softened Potential (CESP), which is found to reproduce quantitatively the experimental data. I have investigated the effect of macromolecular crowding in the binding and conformational properties of two Intrinsically Disordered Proteins (IDPs), histatin-5 and β-amyloid 42, which are modelled using a bead and spring model. The crowders are modelled using the CESP model mimicking Bovin Serum Albumin (BSA). The investigation is motivated by the hypothesis that, in the same way that the conformational and ionization degrees of freedom are coupled in weak polyelectrolytes, the macromolecular crowding should also cause a charge regulation in IDPs whose flexible structure is similar to those of weak polyelectrolytes. Two possible mechanisms through by macromolecular crowding can alter the IDP ionization are proposed: (i) the increase in the effective ionic strength (same ions in a reduced volume) and (ii) the IDP compaction due to macromolecular crowding. The IDPs global charge is found to exhibit significant variations when neutral and charged crowders are added to the system, due to the increase in the effective ionic strength.
Els polímers són omnipresents a la nostra vida diària, presents en múltiples aplicacions industrials i involucrats en processos biològics essencials. Per exemple, l’ADN, les proteïnes i els sucres poden ser considerats polímers, normalment denominats biopolímers. Aquells polímers amb un gran nombre de grups carregats són coneguts com a polielectròlits i freqüentment classificats en termes de la seva ionització com a forts (completament ionizats) o febles (parcialment ionizats). En aquesta tesi, investigo diferents aspectes teòrics de les propietats conformacionals, elàstiques, difusives i d'ionització de polielectròlits febles i biopolímers. Per un costat, em centro en la complexa interació entre els graus de llibertat d'ionització i conformacionals dels polielectròlits febles utilitzant el model Site Binding Rotational Isomeric State (SBRIS), desenvolupat recentment. S’introdueix una nova tècnica analítica, el mètode Local Effective Interaction Parameters (LEIP), per resoldre el model SBRIS incloent les interaccions electroestàtiques intramoleculars de llarg abast. El model es valida comparant els resultats amb la solució obtinguda per simulacions Monte Carlo a pH constant. El model SBRIS s’utilitza per estudiar la resposta elàstica d’un polielectròlit model. Es troba un nou règim de força en el que (i) les corbes força-extensió són significativament dependents del valor del pH i la força iònica (ii) la càrrega del polielectròlit es modificada per la acció de la força d’estirament. S’observa que les propietats conformacionals i elàstiques del polielectròlit es veuen significativament i moderadament afectades per la presència de fluctuacions en la càrrega, respectivament. Per l’altre costat, avaluo les propietats difusives i d'ionització de biopolímers en solució amb una elevada concentració de macromolecules. Aquestes condicions, conegudes com crowding macromolecular, son característiques dels medis biologics on fins el 40% del volum es ocupat per macromolecules. S’analitza la difusió de dues proteïnes globulars, α-chimiotripsina i estreptavidina, en diferents condicions de crowding macromolecular amb simulacions de Dinàmica Browniana. Abstract:
Confucianism is the doctrine that ruled Eastern Asian countries for thousands of years. The doctrine focused on the order of the society and the dependence among individuals in the society. One critique for Confucianism is the under-evaluation of women's role and presence in the society. In the thesis, I will detect Confucianism-related gender stereotypes appearing on feature interviews in the entertainment section of online newspapers in Vietnam, one of former Confucian countries. The thesis also makes the effort to connect the global journalism theory to the solution for removing gender stereotypes from journalism. Three methods namely critical discourse analysis (CDA), conversation analysis (CA) and interviewing are employed in the thesis to address three research questions. Four Confucianism-inspired gender stereotypes are pointed out and the connection between Confucianism-related gender stereotyping and global journalism is basically sketched out in terms of journalistic style.

Abstract:
This study aims at tackling some of the problems often encountered in photostability testing and liquid formulation development. Three anti-cancer drugs will be employed as models; Dacarbazine (DBZ) has well established photostability issues, Axitinib (AXI) and Sunitinib (SUT) are two new drugs only commercially available in solid dosage forms. In ethanol, the photokinetics of these drugs were well described by the newly proposed Φ-order kinetic mathematical model. This has confirmed the photoreversible character of AXI and SUT’s and unimolecular photoreaction of DBZ’s photodegradations. Also, the Φ-order kinetics is proven to describe them better than the usually used classic thermal reaction orders. In aqueous solution, the drugs were found to undergo thermal and photochemical complex degradations, involving at least 3 photoproducts. A new photokinetic approach has been proposed in this work to solving and unravelling the attributes of such complex mechanisms. For the first time, the quantum yields (QY) of the three drugs were determined and found to increase with irradiation wavelength. SUT’s QY were comparable in ethanol and water (QY460 = 0.02), DBZ was found to be more photoefficient in water (QY330 = 0.04 and 0.1, respectively) and AXI in water (QY330 = 0.06 and 0.03). Φ-order kinetics’ potential for the development of reliable actinometers of the three drugs, without prior knowledge of unknown reaction parameters, has also been established. A general equation to describe the isotherm of a (Gn:Hm) guest-host multicomponent complex was proposed in this work to palliate the lack of a strategy for characterising nanosponge-drug complexes. It provides information on both stiochiometry and association constant of the complex. The results indicate that hydrophobic AXI forms a 1:0.8 complex, indicating the possibility of multiple association sites and/or different types of binding. The newly developed AXI/nanosponge liquid formulation has significantly increased solubility (5000-fold) and thermal stability. Furthermore, the photostability of DBZ and SUT were considerably improved by using a strategy based on light-absorption competitors. Their initial velocities reduced from 10 and 3 s-1 (respectively) to 1 and 0.13 s-1. The successful application of these methods to the model anti-cancer drugs has set out new approaches that might be found useful for future treatments of photodegradation data, development of drug-actinometers and liquid formulations of drugs.

Les chansons lient naturellement des mélodies à des paroles. Elles représentent l’une des formes les plus utilisées de l’expression musicale. De façon intéressante, les chansons nécessitent l’intégration simultanée d’informations verbales (les paroles) et d’informations musicales (la mélodie) pour former une seule trace mnésique. Le lobe temporal médian a été identifié comme région clé pour la mémoire épisodique et en particulier pour le processus de binding, qui consiste en la liaison de plusieurs éléments d’un souvenir. Néanmoins, les mécanismes par lesquels le cerveau réalise la liaison entre paroles et mélodies dans la mémoire des chansons restent peu connus. L’objectif de cette thèse est de montrer le rôle du lobe temporal médian dans le processus de binding de paroles et mélodies pour la création d’un souvenir unifié d’une chanson. Premièrement, nous avons étudié les effets d’une sclérose unilatérale de l’hippocampe associée à une épilepsie du lobe temporal sur le processus de mémorisation des chansons en IRM fonctionnelle (étude 1). Les patients avec une sclérose de l’hippocampe gauche avaient un déficit d’adaptation aux paroles et de la représentation intégrée des chansons. Puis, nous avons étudié l’organisation fonctionnelle de la mémoire des chansons dans une étude d’IRM fonctionnelle chez des volontaires sains (étude 2). Les résultats ont montré l’implication de l’hippocampe dans le processus de binding en mémoire des chansons ainsi que l’implication d’autres structures comme le gyrus frontal inférieur, les ganglions de la base et le cervelet. Enfin, dans une dernière étude (étude 3), nous avons examine la mémoire des chansons et plus particulièrement la mémoire de l’association (binding) des paroles au contexte musical chez des patients ayant bénéficié d’une résection unilatérale du lobe temporal médian pour traiter une épilepsie pharmacorésistante. La mémoire des paroles ainsi que celle du contexte mélodique des chansons a été testé dans une tâche explicite de reconnaissance. Un fort déficit en reconnaissance de paroles a été constaté chez les patients avec une lésion temporale gauche, et à un moindre degré chez les patients avec une lésion temporale droite. Ce déficit a été corrélé avec des déficits de la mémoire verbale. L'étude 3 suggère, en outre, que les structures du lobe temporal médian peuvent être cruciales pour le codage de la liaison détaillée entre les paroles et leur contexte mélodique, tandis que les effets implicites d'une représentation intégrée de la chanson peuvent être épargnés après des lésions du lobe temporal médian. Pour conclure, ces trois études ont apporté de nouvelles données sur le rôle du lobe temporal médian dans le processus de binding dans le domaine musical. Ce travail a également permis d’identifier un vaste réseau de régions corticales et sous-corticales impliqué dans l’encodage de nouvelles chansons avant de discuter les implications théoriques et cliniques de ces recherches
Songs naturally couple music with language, constituting one of the most broadly used forms of music expression. Interestingly, songs require the simultaneous and integrated process of verbal (lyrics) and musical (melody) information to form a single memory trace. The medial temporal lobe has been identified as the key region for the integration of features of an event in episodic memory, also called the binding function. Nevertheless, the mechanisms by which the brain binds lyrics and melodies in song memory remain poorly understood. The purpose of this thesis is to elucidate the role of the MTL on the binding of lyrics and melodies for the creation of a unified song memory trace. First, the effects of unilateral hippocampus sclerosis on song processing were investigated in an fMR-adaptation study (Study 1). Patients with left hippocampal sclerosis showed adaptation deficits in response to lyrics as well as to the integrated representation of songs. To further explore the relation of these results with the emergence of memory for songs, the functional architecture of song memory was examined in a subsequent memory fMRI study (Study 2). The results support the implication of the hippocampus in song binding. Furthermore, the role of other structures, including the Inferior Frontal Gyrus (IFG), the Basal Ganglia (BG) and cerebellum was highlighted and discussed. Finally, Study 2 was adapted to test binding in patients following a unilateral temporal lobe excision for the relief of intractable temporal lobe epilepsy behaviorally in Study 3. Memory for lyrics as well for their melodic context was tested in an explicit recognition task. A strong deficit in lyrics recognition was found in patients with a left temporal lesion, and to a smaller degree in patients with a right temporal lesion. This deficit was correlated with deficits in verbal memory. Evidence from Study 3 further suggests that MTL structures may be crucial for encoding the detailed binding of lyrics with their melodic context, whereas implicit effects of an integrated representation of the song may be spared after MTL lesions. Altogether the studies presented in this thesis provide novel evidence for the role of the medial temporal lobe structures in binding lyrics and melodies for song memory. The present thesis proposes a comprehensive network of cortical and subcortical regions cooperating to successfully encode new songs. Finally, theoretical and clinical implications of these findings are considered

Abstract:
VanS is an integral membrane protein, and a receptor histidine kinase representing on half of the VanSR two component system. In the presence of vancomycin, as well as certain other antibiotics, VanS undergoes autophosphorylation at a conserved histidine residue and then transfers this phosphoryl group to a conserved aspartate on VanR. The phosphorylated form of VanR binds to the promoter region of the vanHAX operon, inducing expression of these genes, which confer glycopeptide antibiotic resistance to the cell. Although the vancomycin resistance mechanism is mostly well-characterised, questions surrounding the structure and mode of activation of VanS remain unanswered. This thesis describes work towards the structural and ligand-binding characterisation of the extracellular \sensor" domain of VanS. Little is known of the structure of this domain, and while there exists evidence in the literature to support numerous theories surrounding the identity of the VanS ligand, no report has yet been made of a direct observation of a binding event involving any VanS protein and its ligand To identify the ligand to VanS, the catalytically active, full-length VanS proteins of Enterococcus faecium (VanSA) and Streptomyces coelicolor (VanSSC) were heterologously expressed, solubilised and purified and binding studies were carried out by solution-state NMR. We present evidence of interactions between vancomycin and both VanS proteins, indicating that direct binding of vancomycin may be the mechanism by which both proteins are activated. The VanSSC extracellular domain was isolated as a synthetic peptide, its structure characterised, and the binding interface between this domain and vancomycin was investigated. Evidence of binding at the N-terminal end of the extracellular domain, in agreement with the findings of Koteva et al. (2010), is presented here. An interaction between vancomycin and DPC detergent micelles was also observed and characterised, and the biological significance of such an interaction is discussed.

Attualmente non è ancora disponibile alcun vaccino ampiamente totalmente efficace per la prevenzione della meningite di gruppo B. Uno dei più promettenti candidati per lo sviluppo di un vaccino è la proteina che lega il fattore H (factor H-binding protein o fHbp), una lipoproteina di superficie espressa in tutti i ceppi di Neisseria meningitidis. La sua funzione è quella di legare il fattore H (fH), una molecola chiave della via alternativa della cascata del complemento. Il patogeno, legando sulla sua superficie il fH, riesce a bloccare l'attivazione del complemento e ad eludere le difese immunitarie dell'ospite. La fHbp è una componente di un nuovo vaccino per la protezione nei confronti della malattia meningococcica di gruppo B, che si trova attualmente nell'ultima fase di sperimentazione clinica. In base alla variabilità nella composizione amminoacidica, la fHbp è stata suddivisa in tre diverse varianti o due sottofamiglie e la sua architettura è modulare, costituita da 5 segmenti di residui variabili. Immunizzando topi con ciascuna delle 3 varianti della fHbp è stato prodotto un pannello di anticorpi monoclonali anti-fHbp. Gli epitopi riconosciuti da questi mAbs sono stati mappati tramite l'allineamento multiplo delle sequenze proteiche ed esperimenti di mutagenesi sito-specifica. Tuttavia questi approcci non hanno permesso di mappare l'epitopo di uno di questi mAb, denominato JAR 36. L'anticorpo JAR 36 è stato prodotto immunizzando un topo con la fHbp nella variante 3, inoltre esso cross-reagisce con la proteina della variante 2 e mostra attività battericida cooperativa con JAR 11 e JAR 13. E' stato effettuato uno screening di librerie di peptidi esposti sulla superficie di batteriofagi filamentosi allo scopo di identificare ì residui che costituiscono l'epitopo di JAR 36. Le ipotesi di mappatura sono state quindi verificate attraverso esperimenti di mutagenesi sito-specifica. Inoltre, utilizzando un approccio computazionale, sono stati individuati 17 residui che potrebbero essere importanti per il riconoscimento della proteina da parte dell'Ab. In particolare 8 di questi residui risiedono in una regione ritenuta essere fondamentale per il riconoscimento di JAR 36 e 4 di essi sono comuni alle varianti 2 e 3. Sia ì residui individuati usando la tecnologia del phage display che quelli predetti dall'analisi “in silico”, si trovano in prossimità del sito di legame del fH. Questi risultati possono sono in accordo con precedenti dati sperimentali in cui JAR 36 inibisce parzialmente il legame del fH. I dati ottenuti contribuiranno alla caratterizzazione del meccanismo molecolare dell'immuno-protezione contro MenB, per lo sviluppo di vaccini a base proteica sempre più sicuri ed efficaci per la prevenzione dalla malattia causata da questo patogeno.
No broadly protective vaccine is yet available for the prevention of group B meningococcal disease. One promising vaccine lead is the factor H binding protein (fHbp), a surface-exposed lipoprotein, which is present in all strains of Neisseria meningitidis. This protein binds human factor H (fH), contributing to the ability of these bacteria to avoid complement-mediated killing. The fHbp is a component of a serogroup B meningococcal vaccine currently in late-phase clinical development. Based on the sequence variability of the entire protein, fHbp has been divided into 3 variant groups or 2 sub-families and its architecture is modular, consisting of 5 variable segments, amino acid residues in position between 100 and 255 have been identified as important targets of variant-specific bactericidal antibodies. A panel of anti-fHbp mAbs has been produced from mice immunized with each of the 3 variants of fHbp and all their epitopes have been mapped, except that of the mAb designated JAR 36. This antibody was isolated from a mouse immunized with fHbp of the antigenic variant 3 group, but it cross-reacts with all fHbp sequences in the variant 2 group and elicits complement-mediated cooperative bactericidal activity with JAR 13 and JAR 11. We have screened phage displayed random peptide libraries to identify amino acid residues composing the JAR 36 epitope, putative mapping hypotheses have been then validated by constructing recombinant fHbps single-point variants through site-specific mutagenesis, and analyzing their reactivity with the mAb. Moreover, by using a computational tool, a total of seventeen residues have been identified as possibly constituting the epitope of JAR 36. Eight of these residues are located in a segment of fHbp previously recognized to play an important role in the binding of JAR 36 and four of them are common in variant groups 2 and 3. Both the amino acid identified by phage display and the “in silico”-predicted epitope are located in the proximity of the fH-binding site. This finding is according with previous experimental data, of JAR 36 partially inhibiting binding of fH to fHbp. The data reported will help in characterizing the molecular mechanism of immune protection against MenB, toward the development of safer and effective protein-based vaccines for the prevention of group B meningitis.

Escherichia coli enteropatogÃnica (EPEC) à um importante patÃgeno associado Ãs doenÃas diarreicas. InfecÃÃes intestinais ocasionam comprometimento da barreira intestinal e um dos primeiros mecanismos de resposta à recuperaÃÃo à a migraÃÃo das cÃlulas intestinais. As principais proteÃnas que regulam esse processo sÃo as pequenas GTPases Rho, Rac1, RhoA e Cdc42A. A alanil-glutamina (Ala-Gln) estimula este processo migratÃrio, entretanto os mecanismos envolvidos nesta resposta ainda sÃo desconhecidos. Desse modo, investigou-se o efeito de uma cepa selvagem e padrÃo (E2348/69) de EPEC, e de uma cepa comensal E. coli HS na migraÃÃo celular intestinal, bem como a regulaÃÃo da transcriÃÃo e expressÃo gÃnica das GTPases Rho e o papel da suplementaÃÃo com Ala-Gln no processo de migraÃÃo na presenÃa ou ausÃncia da infecÃÃo. A infecÃÃo pelas cepas de EPEC e pela cepa comensal reduziram significativamente a migraÃÃo celular intestinal. Entretanto, houve uma maior reduÃÃo desse efeito nas cÃlulas infectadas pelas cepas de EPEC quando comparado Ãquelas infectadas pela cepa comensal de E. coli HS. Observou-se um alto percentual de cÃlulas necrÃticas, cerca de 30%, induzido pela cepa padrÃo de EPEC apenas nos tempo de 12 e 24 horas apÃs infecÃÃo. A adiÃÃo da Ala-Gln em cÃlulas nÃo infectadas estimulou significativamente e de modo dose dependente a migraÃÃo apÃs 24 horas. PorÃm, quando esse nutriente foi adicionado durante 12 e 24 horas na presenÃa da infecÃÃo, nÃo houve uma reversÃo do dano. Em relaÃÃo à expressÃo gÃnica das GTPases Rho, observou-se um aumento da transcriÃÃo de rac1 nas cÃlulas que haviam sido infectadas pelas cepas de EPEC e E. coli HS, bem como um aumento da transcriÃÃo de rhoA nas cÃlulas infectadas pela cepa padrÃo de EPEC apÃs 2 horas da infecÃÃo. Todavia, na anÃlise das proteÃnas por imunofluorescÃncia, RhoA e Cdc42 mostraram-se aumentadas nas cÃlulas infectadas pela EPEC padrÃo quando comparado ao controle. Enquanto que as cÃlulas infectadas com a cepa selvagem de EPEC observou-se um aumento de Rac1 e reduÃÃo de RhoA. Esses dados mostraram que a migraÃÃo das cÃlulas intestinais à reduzida principalmente pelas cepas patogÃnicas de EPEC, ao regular a transcriÃÃo e expressÃo gÃnicas das proteÃnas GTPases Rho. A suplementaÃÃo com Ala-Gln em cÃlulas intestinais promoveu a migraÃÃo celular apenas na ausÃncia da infecÃÃo.
Enteropathogenic Escherichia coli (EPEC) is an important pathogen associated with diarrheal diseases. Intestinal infections cause impairment of the intestinal barrier and one of the earliest response mechanisms to recover is migration of the intestinal cells to cover the injured area. The key proteins that regulate cell migration are small Rho GTPases, Rac1, Cdc42 and RhoA. The alanyl-glutamine (Ala-Gln) increases this migration process, however the mechanisms involved in this response are still unknown. Thus, we investigated the effect of a wild type strain and standard (E2348/69) of EPEC strain and a commensal E. coli HS on intestinal cell migration, as well as transcriptional regulation and gene expression of Rho GTPases and the role of supplemental Ala-Gln in the migration process in the presence or absence of infection. Infection by EPEC strains and commensal E. coli HS significantly reduced intestinal cell migration. However, this effect was more pronounced in cells infected by the strains of EPEC compared to those infected by the commensal strain of E. coli HS. We observed a high percentage of necrotic cells, about 30%, induced only by EPEC strain pattern 12 and 24 hours after infection. The addition of Ala-Gln in uninfected cells significantly stimulated in a dose dependent migration after 24 hours. However, when this nutrient was added over 12 and 24 hours in the presence of infection, there was no reversion of the damage. Regarding the gene expression of Rho GTPases, we observed an increase in transcription of rac1 in cells that had been infected by the strains of EPEC and E. coli HS as well as an increase in rhoA transcription in cells infected with EPEC strain pattern at 2 hours after infection. However, the analysis of proteins by immunofluorescence, RhoA and Cdc42 shown to be elevated in cells infected with EPEC pattern when compared to the control. Whereas cells infected with wild EPEC strain was observed an increase of Rac1 and reduction of RhoA. These data showed that cell migration is reduced mainly by the intestinal pathogenic strains of EPEC, in regulating gene transcription and expression of the protein Rho GTPases. Supplementation with Ala-Gln in intestinal cells only promoted cell migration in the absence of infection.

As proteínas ligadoras de ácidos graxos (Fatty Acid Binding Proteins, FABPs) de parasitos têm um papel importante no processo de infecção por estes organismos. Por este motivo, estas proteínas são antígenos candidatos para vacina contra a infecção por Schistosoma mansoni e Fasciola hepatica. No presente trabalho foram caracterizadas FABPs de F. hepatica e comparadas com a proteína Sm14 de S. mansoni, a FABP de parasito melhor caracterizada, mediante análise de sequências e estruturas modeladas. Também foram clonadas, expressas e purificadas as FABPs tipo 1 e tipo 3 de F. hepatica. Os resultados do presente estudo indicam que a FABP tipo 3 de F. hepatica é relacionada estrutural, imunológica e funcionalmente com a Sm14, um candidato vacinal amplamente estudado. Devido à importância da Sm14 como alvo para o desenvolvimento de vacina para a esquistossomose, as características apresentadas pela FhFABP3 de F. hepatica apontam esta proteína como um candidato importante também para o desenvolvimento de uma vacina contra a fasciolose
Parasites fatty acid binding proteins (FABPs) have an important role in infection process by these organisms. For this reason, these proteins are candidates as vaccine antigens against Schistosoma mansoni and Fasciola hepatica infection. In the present study, FABPs from F. hepatica were characterized and compared with Sm14 protein from S. mansoni, the best characterized parasite FABP, by sequence analysis and modeled structure. Type 1 and type 3 FABPs from F. hepatica were also cloned, expressed and purified. The results of this study indicated that type 3 FABP from F. hepatica is structural, immunological and functionally related with Sm14, a vaccine candidate widely studied. Due to the importance of Sm14 as a target for vaccine development for schistosomiasis, the characteristics presented by the FhFABP3 from F. hepatica suggest this protein as a candidate also important for the development of a vaccine against fasciolosis

Due to its significance in many cellular functions, fatty acid binding protein 7 (FABP7) has become a rising topic of interest for many scientists. Immunocytochemistry was used to map the distribution of FABP7 and test whether the amount of FABP7 immunoreactivity (FABP7-IR) differed in animals that were defeated in a fight, as compared to control animals that did not engage in any social interaction. The male green anole was used as the subject because its natural tendency to establish social classes within its species provides an ideal model to observe for variation in FABP7-IR. The results showed FABP7-IR in cells and fibers of the cortex, hypothalamus, thalamus, medial preoptic area, dorsoventricular ridge, amygdala, suprachiasmatic nucleus, nucleus accumbens, nucleus rotundus, habenular area, tectum, dorsal noradrenergic and lateral forebrain bundles, and lining the third and lateral ventricles. Qualitative observation suggested higher FABP7 levels in socially defeated males than controls in all areas.

MARK4 (MAP/Microtubule Affinity-Regulating Kinase 4) is a serine-threonine kinase that phosphorylates the Microtubule Associated Proteins (MAPs) taking part in the regulation of microtubule dynamics involved in cell cycle regulation and proliferation. MARK4 is ubiquitously expressed and two different isoforms are produced through alternative splicing: the constitutive MARK4S, consisting of 18 exons, and the alternative MARK4L encoded by a transcript that undergoes skipping of exon 16 and consequent frameshift of the reading frame. The differential expression of the two MARK4 isoforms in human tissues and, particularly in the Central Nervous System, point to their not fully overlapping roles. MARK4S is predominant in normal brain and differentiated neurons and has been associated to cell differentiation, while MARK4L is mainly expressed in neural progenitors and is up-regulated in hepatocarcinoma and glioma, suggesting a function of this isoform in cell proliferation. In glioma and glioblastoma-derived cancer stem cells (GBM CSCs), we pointed out an imbalance between the two MARK4 isoforms, directly proportional to cell de-differentiation and tumour grade. Such imbalance is triggered by decrease in MARK4S expression, associated with overexpression of MARK4L. The MARK4 expression profile observed in glioma matches that observed in mouse neural stem cells (NSCs). Moreover, MARK4L has been found expressed in the embryonic ventricular zone and adult sub-ventricular zone, both well known regions of neurogenesis, suggesting that the balance between the two MARK4 isoforms is critical for neural differentiation and proliferation. Therefore, in glioma a subverted MARK4L/MARK4S ratio may contribute to cellular de-differentiation and proliferation during gliomagenesis. Having ruled out mutations or copy number loss/gain as cause of deregulation of MARK4 expression in glioma, we hypothesised that alterations at the post-transcriptional level, possibly in alternative splicing, might be at the origin of the observed MARK4 isoforms imbalance. MARK4L total mRNA evaluation by real-time PCR in glioma tissue samples and GBM CSCs, failed to reveal differences in the overall expression of the kinase, even in the presence of significantly different expression levels of the two isoforms, supporting the hypothesis of an alteration in alternative splicing at the root of MARKL/MARK4S imbalance in glioma. Alternative splicing is the most important mechanism in generating proteomic diversity from a limited number of genes. Alternatively spliced isoforms expression is thinly controlled according to specific cells signalling and developmental stages by a complex interplay between spliceosome and splicing regulatory factors, which can be grouped into the two main classes of SR (serine-arginine rich) and hnRNP (heterogenous nuclear ribonucleoprotein) proteins. The intrinsic plasticity of alternative splicing in proteome modelling, make sthis process a tempting target for cancer cells to enhance the production of protein isoforms sustaining tumour growth and spread. Protein isoforms produced from exon skipping, like MARK4L, are commonly found enriched in cancer, including glioma, as a consequence of hnRNP protein overexpression. We thus performed bioinformatic analysis to identify putative binding sites for hnRNP proteins in MARK4 pre-mRNA. Among the predicted putative binding sites for hnRNPs, three binding motifs for PTB (polypyrimidine-tract binding protein) appeared of particular interest, given the role of PTB in regulating neural stem cells proliferation and differentiation and its established involvement in glioma, where it is aberrantly overexpressed and induces the expression of protein isoforms produced by exon skipping. We found two putative PTB binding sites in intron 15 (IVS15) and one in intron 16 (IVS16). One of the two identified regulatory sequences in IVS15 is embedded in a polypyrimidine rich context and may represent a high affinity PTB binding site that could favour MARK4 exon 16 skipping by competition with U2AF65 or, together with the other PTB binding sites, by polymerisation or intron looping. A functional role of these sites is also suggested by the high sequence conservation between human and mouse. Western blot analysis showed a significant overexpression of PTB in our astrocytoma and glioblastoma samples, correlating with MARK4L expression and thus with tumour grade and cell de-differentiation. Moreover, GBM CSCs, both undifferentiated and differentiated, showed high levels of PTB coherently with the nearly exclusive expression of MARK4L. However, while NSCs show undetectable levels of MARK4L upon differentiation, in differentiated GBM CSCs high expression levels of MARK4L are maintained possibly sustained by PTB overexpression. We then performed minigene splicing assays to identify the specific sequences involved in MARK4 alternative splicing. We focused our attention on the polypyrimidine tract in MARK4 VS15 and constructed a MARK4 splicing minigene by cloning, in a mammalian expression vector, the MARK4 genomic region comprising exons 15, 16 and 17 and IVS15 and IVS16. Sequential deletions of different IVS15 portions revealed that the last 87 nucleotides of intron 15 contain a functional intronic splicing silencer (ISS). However, mutagenesis of the PTB binding site contained in this region did not affect minigene splicing, suggesting that PTB may be involved in MARK4 splicing by binding to a non canonical ISS or by cooperating with other identified PTB binding sites. Moreover, electrophoretic mobility shift assays (EMSA) highlighted a specific shifted band probably due to the interaction of splicing factors with MARK4 pre-mRNA. Mass spectrometry experiments are in progress aiming at identifying the protein(s) bound to MARK4 IVS15, and seem to confirm the presence of PTB in the shifted band. The achieved data, suggest that PTB overexpression in glioma may favour MARK4L expression causing the observed imbalance between the two MARK4 isoforms, featuring alternative splicing as an oncogenic mechanism that through the fine tuned regulation of MARK4 isoforms may foster proliferation and de-differentiation in glioma.

INTRODUÇÃO: O antecedente de parto prematuro espontâneo em gestação anterior é considerado o principal e mais importante fator de risco clínico para prematuridade, principal causa de morbidade e mortalidade neonatal. Cerca de 25% das pacientes que tiveram parto prematuro apresentarão recorrência. A prevenção secundária consiste na pesquisa de marcadores de maior risco, com o intuito de instituir medidas terapêuticas apropriadas e de evitar tratamentos desnecessários. A hipótese do presente estudo é a de que existe correlação entre os resultados da avaliação do colo uterino e do teste para proteína-1 fosforilada ligada ao fator de crescimento insulina-símile (phIGFBP-1) e que a utilização de ambos em associação possa predizer a ocorrência de parto prematuro com maior sensibilidade. OBJETIVOS: Averiguar a utilidade da medida do comprimento do colo uterino e do teste para phIGFBP-1 na predição do parto prematuro antes de 37 e de 34 semanas, a existência de relação dos testes entre si, o melhor valor de corte da medida do colo em diferentes idades gestacionais e a melhor época de realização de cada um dos exames. MÉTODO: Foram compilados e submetidos a análise secundária os dados de 101 gestantes com antecedente de parto prematuro atendidas no Setor de Baixo Peso Fetal da Clínica Obstétrica do Hospital das Clínicas da Faculdade de Medicina da Universidade de São Paulo, entre 2003 e 2008. A medida do comprimento cervical e o teste para phIGFBP-1 foram realizados a cada três semanas, entre 24 e 34 semanas de gestação, e comparados com o desfecho de parto prematuro e nascimento com 34 semanas ou menos, e o melhor valor de corte do colo uterino foi estabelecido por meio de curva de características operacionais. RESULTADOS: Das 101 gestações estudadas, 25 (24,8%) terminaram em parto prematuro, das quais 12 (11,9%) ocorreram com 34 semanas ou menos. As idades gestacionais médias de avaliação foram de 24, 27, 30 e 33 semanas, e os valores de corte do colo uterino foram de 22, 21, 20 e 16 mm, respectivamente. A medida do comprimento do colo apresentou maior sensibilidade (cerca de 70%) e foi capaz de predizer o parto prematuro em todas as avaliações. O teste para phIGFBP-1 não foi útil com 24 semanas, porém foi capaz de detectar de forma independente o risco de prematuridade com 27, com 30 e com 33 semanas. Houve associação estatística dos exames entre si, de forma que o comprimento cervical médio foi menor em gestantes com teste positivo para phIGFBP-1. A associação dos exames elevou a sensibilidade e o valor preditivo negativo de forma significativa. CONCLUSÕES: A medida do comprimento do colo pela ultrassonografia transvaginal constitui bom marcador de risco para parto prematuro com 24 semanas, e o teste para phIGFBP-1 é útil após 27 semanas. A associação dos dois exames possui alta sensibilidade e alto valor preditivo negativo em gestantes de alto risco para prematuridade espontânea, e a realização do primeiro com 24 semanas e do segundo com 27 semanas constitui bom modelo preditivo para o parto prematuro
INTRODUCTION: The history of spontaneous preterm birth in a previous pregnancy is considered the main and most important clinical risk factor for preterm birth, the leading cause of neonatal morbidity and mortality. About 25% of these patients will deliver prematurely again. Secondary prevention consists in the search for markers of increased risk, in order to institute appropriate therapeutic actions and to avoid unnecessary treatments. The hypothesis of this study is that there is a correlation between the results of the evaluation of the cervix and the test for phosphorylated insulin-like growth factor binding protein-1 (phIGFBP-1) and that the use of both in combination can predict the occurrence of preterm delivery with higher sensitivity. OBJECTIVES: To investigate the usefulness of the measurement of the cervical length and phIGFBP-1 rapid test in the prediction of preterm birth before 37 and 34 weeks, the existence of a relationship between the tests themselves, the best cutoff value of cervical length measurement at different gestational ages and the best time to carry out each of the exams. METHODS: Data of 101 women with previous preterm birth assisted at the Obstetrical Clinic of the Hospital das Clínicas, Faculty of Medicine, University of São Paulo between 2003 and 2008 were collected and subjected to secondary analysis. The measurement of cervical length and the phIGFBP-1 test were performed every three weeks, between 24 and 34 weeks gestation, and compared with the outcome of premature birth before 37 and 34 weeks, and the best cutoff value of the cervix was determined by receiver operator characteristic curves. RESULTS: Of the 101 pregnancies studied, 25 (24.8%) ended in preterm birth, of which 12 (11.9%) occurred at 34 weeks or less. The mean gestational age in each evaluation was 24, 27, 30 and 33 weeks, and the cutoff of the cervix were 22, 21, 20 and 16 millimeters, respectively. The measurement of cervical length showed the highest sensitivity (approximately 70%) and was able to predict preterm birth in all evaluations. The phIGFBP-1 test was not useful at 24 weeks, but was able to independently detect the risk of prematurity at 27, 30 and 33 weeks. Statistical association between the exams was observed, so that the mean cervical length was lower in pregnant women testing positive for phIGFBP-1. The combination of both tests significantly increased the sensitivity and negative predictive value. CONCLUSIONS: The measurement of cervical length by transvaginal ultrasound is a good marker of risk for preterm delivery at 24 weeks, and the test for phIGFBP-1 is useful after 27 weeks. The association of the two tests is valuable and shows high sensitivity and high negative predictive value in women at high risk for spontaneous preterm birth, when the first is preformed with 24 weeks, and the second with 27 weeks

Cellulose is an insoluble dietary fiber and is the most abundant polysaccharides in nature. It has been proved that cellulose can improve human health, such as promoting gastrointestinal peristalsis, reducing weight, preventing constipation and colon cancer. It has been reported that macrominerals binding capacity of fiber is related with particle size and water holding capacity. Volume mean diameter of cellulose was significantly reduced by media-milling, resulted in both the reducing particle size and molecular weight. For example, the molecular weight was reduced from 646.29 kDa to 117.1 kDa after 90 min media-milling. Via sediment method, the suspended cellulose yield a zeta potential of -16.6 mV, a negative charge. After media-milling, the absolute value of zeta potential was increased, indicates that nano/submicron cellulose dispersion is more stable, Isoelectric point(IEP) of cellulose raw material is pH 3.03, and dropped to 2.74 after 90 min media-milled, shows that after media-milling, cellulose particles are more negative-charged. Media-milling also improves WHC of cellulosic particles. By centrifugation method, WHC increased 7.9 fold after 30 min media-milling, but decreased after 30 min. Viscosity of cellulose dispersion increased first from 1.71 cp to 25.46 cp and then decrease after 30 min. WHC and viscosity were found correlated. Media-milling can also enhance Mg, Ca, Na, K binding capacity with a relationship with WHC.

The binding properties of two new cyclophane receptors in aqueous media were explored. Replacement of two benzene rings of the host P with furans or thiophenes was expected to enhance cation-[pi] interactions, but significant improvements in the binding of positively-charged guests were not observed. Ab initio calculations provided a rationalization for the experimental findings and a better understanding of the cation-[pi] interaction. The binding in water of various guanidinium compounds to host P was also investigated. These molecules represented a new type of cationic guest for the receptor. Arginine was not measureably bound by P, but several alkylated guanidiniums were well bound, especially considering their high water solubility. Hexamethylguanidinium, a nonplanar molecule with [...] symmetry, was a particularly interesting guest which displayed enantioselective binding to P. In addition, some progess was achieved towards the synthesis of receptors that have either an amide or a carboxylate group attached to the rim of the cavity. The appended group is positioned such that it can form hydrogen bonds or electrostatic interactions with a bound guanidiniumn guest. Finally, exploratory work was carried out towards the synthesis of a cyclophane host that possesses a disulfide near the binding cavity. This receptor was designed to mimic the acetylcholine binding site of the nicotinic acetylcholine receptor. Although various disulfide containing macrocycles were synthesized, they were not sufficiently water soluble to permit studies of their binding properties.

Retouching material is the paint to fill the lacuna of color on art works during conservation process. The concept and technique of retouching from the western countries are more developed and comprehensive, the over viewing of the developing history and the material evolvment since hundreds centuries reveals the foundation of conservation wasn’t simply built in oe day. The binder of retouching material holds the key to determine the physical, chemical and photochemical properties, which also closely bonded with the handling ability and environmental condition. Owing to the fact of higher stability and easy to be modifying, the study of retouching binder leads toward the direction of synthetic resin. Based by the western conservation concept and technique, the conservation in Taiwan is striding forward to the international level. However, if the material that has been tested by museum condition can be directly transfer to the island climate of Taiwan is still a question of uncertain. Thus, this study choose three of the most recommended retouching material (1)watercolor coated with 5% acrylic resin Paraloid® B72, (2)acrylic resin Paraloid® B72 and (3)low molecule weight resin Laropal® A81 to proceed accelerate aging process with the simulation of Taiwan’s exterior condition, and measure the alter of color, gloss, adhesion and solubility to evaluate the stability and reversibility of the materials. The results shows that both the water color coated 5% Paraloid® B72 and Laropal® A81 have lower resistance against moisture, thus are not recommended for the use of exterior area. The stability and reversibility of Paraloid® B72 after aged is durable, and when the physical damages such as hollow and flaking occurred, the consolidation is achievable simply with pressure and heat. But the material is more suitable for hatching technigue rather than paint large area at once.

My Ph.D. work was aimed at developing new methodologies based on combination of traditional spectroscopic techniques with multivariate analysis tools to analyze binding media used in traditional easel European paintings. My dissertation explored the possibility of fully characterizing traditional neat binding media and complex systems such as oil-paint models, applying Principal Component Analysis (PCA) on combined first derivative Fourier Transform Near Infrared (FT-NIR) and micro-Raman spectra. Furthermore, I also carried out a preliminary study to evaluate potentialities and limits of the 2D-Fluorescence spectroscopy, applied in combination with PCA to study and characterize neat binding media. This work was performed within the framework of the national project “SICH PRIN 2010-11 - Sustainability in Cultural Heritage from diagnosis to the development of innovative systems for consolidation, cleaning and protection” that was funded by the Italian Ministry of University and Research.
