Abstract:
The aim of this thesis is to examine the use of the groupware system cAR/PE!, which is based on Mixed Reality technology. cAR/PE! was developed at the DaimlerChrylser Research Center in Ulm dedicated to distributed engineering group work and subsequently tested in two studies at various plants of DaimlerChrysler AG. Interviews and observations of engineer meetings in distributed project groups provided the basis for the conception of the two empirical studies. In the first study the first cAR/PE! prototype was tested. The study confirmed that less complex tasks could be solved equally well in both a conventional meeting room and in cAR/PE! Meetings. However, when solving a complex task, the conventional meeting was more favorable than cAR/PE!. These results were exploited for further developments of cAR/PE! resulting in cAR/PE!2. In the second study, the modifications were evaluated under two conditions to solve the complex design task amphibious car. Under the first condition, groups were still better at solving a complex task under the conventional meeting2 condition compared with cAR/PE!2, even the problem-solving processes were comparable and no longer structurally different than in the first study. The differences in the group results were caused by the software when using cAR/PE!2. Under the second condition, the influence of the meeting workspace was tested; this had a direct influence on the problem-solving processes of the groups, but the result was not significant. However, from this outcome it was deduced that only the relevant information for the tasks and the physical placement of the speaker leads to a successful conversation. On the theoretical level, the enhancement of the framework developed by Vertegaal et al. (1997) introducing the concept of task awareness and the appropriateness of the Model for the elements of distributed group work were confirmed. On the practical level, cAR/PE!2 was improved based on the results of both studies, then implemented and established as a permanent running groupware system between DaimlerChrysler´s plants in Sindelfingen and Bremen

Clyne, Courtney. "Emotion-Focussed Psychoeducational Group Therapy for Binge Eating Disorder in Women and Men." Thesis, University of Canterbury. Psychology, 2007. http://hdl.handle.net/10092/1384.

Full text
Abstract:
A plethora of research has linked negative affect with binge eating in people with binge eating disorder (BED). Cognitive behavioural therapy (CBT) and interpersonal therapy (IPT) have not traditionally addressed emotional regulation deficits. Failure to address emotional aspects of binge eating may explain why some individuals do not respond to CBT or IPT, and why many of those who do respond relapse shortly after finishing treatment. Dialectical behaviour therapy (DBT) specifically targets the inability to accurately recognise and regulate affect. Preliminary investigations have shown that DBT may be efficacious in treating BED. However, it can take up to 10 months, and a shorter intervention targeting affect regulation and recognition may produce similar effects. Four studies evaluating a ten session emotion-focussed group psychoeducational intervention for BED were conducted. In the first, 25 women diagnosed with subthreshold or full syndrome BED (using DSM-IV-TR criteria) were treated. The second study tested whether the specific components of the intervention, or the treatment as a whole, was required to produce positive outcomes in women. Study three, examined the efficacy of the treatment programme with three men diagnosed with subthreshold or full syndrome BED (also using DSM-IV-TR criteria). The fourth study compared the women's and men's response to treatment. Following the intervention with women, binge abstinence rates, comparable to those of CBT and IPT, and various other positive changes to eating and general pathology, were observed. These effects were well-maintained up to one-year later. Overall, it was concluded that the whole treatment programme was necessary to produce the optimum outcome for BED in women. A positive outcome was measured in the men, although the effects were not as dramatic as those found in the women. Suggestions for improvements, and suggestions for further research, are discussed. The results provide support for the Affect Regulation Model of BED in women and men.

Abstract:

Cooper, Samuel John. "Polar group exchange receptor-ligand engineering of protein kinase B's pleckstrin homology domain." Thesis, Imperial College London, 2010. http://hdl.handle.net/10044/1/5931.

Full text
Abstract:
A diverse range of cellular functions are influenced by the activity of protein kinase B (PKB, also known as Akt). PKB is involved in a number of physiological pathways, perturbation of which can lead to pathological conditions. PKB activation has been shown to involve binding of its pleckstrin homology (PH) domain to PtdIns(3,4)P2 and PtdIns(3,4,5)P3 generated on the inner face of the plasma membrane upon receipt of an external hormonal signal. To study PKB independently of other inositol-phospholipid binding proteins which recognise the same lipid signals, their mutual dependence on the same ligand must be broken. It is proposed that this can be achieved by mutating PKB‟s PtdInsPn binding PH domain to accept an unnatural ligand. By altering the inositol-phospholipid head-group to include an additional carboxylic acid substituent, its binding to the wild-type PH domains should be inhibited through steric compression. Disruption of a salt-bridge in the wall of the binding pocket by point mutation of glutamate 17 to alanine, E17A, introduces an additional positively charged pocket into the PH domain of PKB, which should bind with the tailored lipid, resulting in a polar group exchange receptor-ligand pair. To this end, a novel 2-O-acetic acid functionalised analogue of Ins(1,3,4,5)P4 was synthesised. The key steps were the protection/deprotection of myo-inositol‟s hydroxyls, enabling selective functionalisation of the 2-hydroxyl, and phosphorylation of the 1,3,4,5-O positions. Using an allyl ether as a masked form of acetic acid allowed a divergent synthetic strategy to be employed to access additional 2-O analogues. Both the wild-type and the E17A mutant of the PH domain were expressed in order to assess ligand binding, and determine whether an ion pair exchange receptor-ligand interaction occurred. Intrinsic tryptophan fluorescence spectrometry was initially used, but was unsuccessful due to denaturing problems arising from isolating the protein. Isothermal titration calorimetry suggested binding, but protein concentration could not be increased high enough for the binding energy to become significantly greater than the energy of mixing. The analogues were also tested as potential inositol poly-phosphate phosphatase catalytic (IPPc) domain inhibitors.

Abstract:
Chemical Looping Combustion (CLC) is an oxy-combustion like process where particles are used to supply oxygen to combustion. Further work is still needed for extrapolation and optimization of the CLC process, concerning properties of Group B particles and CFB technology. Hydrodynamic tests were made on a 18 m tall riser. Axial pressure profiles as well as radial flux profiles and radial momentum quantity profiles were obtained. Three types of Group B particles were used with Sauter mean diameters between 250 and 300 μm and densities between 2600 and 3300 kg/m²s. An important impact of particle sphericity on riser pressure drop has been revealed. At identical conditions, glass beads present about half the pressure drop generated by sand. In the developed region of the riser, the core-annulus regime has been found. A 1D model of the riser, based on experimental results and on the Euler-Euler gas-solid equations, has been developed. Moreover, a new cross section averaged drag force correlation is presented. A new boundary condition on the bottom of the riser has been investigated. The final 1D model is capable of predicted riser pressure drop from the operating conditions and it takes into account particle properties such as density, size and shape. A study on the adequacy of the use of the commercial CFD code Barracuda to simulate risers with Group B particles was made. It was shown that the code under estimates pressure drop along the riser for sand simulations

The use of behavioural indicators of suffering and welfare in captive animals has produced ambiguous results. In comparisons between groups, those in worse condition tend to exhibit increased overall rate of Behaviours Potentially Indicative of Stress (BPIS), but when comparing within groups, individuals differ in their stress coping strategies. This dissertation presents analyses to unravel the Behavioural Profile of a sample of 26 captive capuchin monkeys, of three different species (Sapajus libidinosus, S. flavius and S. xanthosternos), kept in different enclosure types. In total, 147,17 hours of data were collected. We explored four type of analysis: Activity Budgets, Diversity indexes, Markov chains and Sequence analyses, and Social Network Analyses, resulting in nine indexes of behavioural occurrence and organization. In chapter One we explore group differences. Results support predictions of minor sex and species differences and major differences in behavioural profile due to enclosure type: i. individuals in less enriched enclosures exhibited a more diverse BPIS repertoire and a decreased probability of a sequence with six Genus Normative Behaviour; ii. number of most probable behavioural transitions including at least one BPIS was higher in less enriched enclosures; iii. proeminence indexes indicate that BPIS function as dead ends of behavioural sequences, and proeminence of three BPIS (pacing, self-direct, active I) were higher in less enriched enclosures. Overall, these data are not supportive of BPIS as a repetitive pattern, with a mantra-like calming effect. Rather, the picture that emerges is more supportive of BPIS as activities that disrupt organization of behaviours, introducing ?noise? that compromises optimal activity budget. In chapter Two we explored individual differences in stress coping strategies. We classified individuals along six axes of exploratory behaviour. These were only weakly correlated indicating low correlation among behavioural indicators of syndromes. Nevertheless, the results are suggestive of two broad stress coping strategies, similar to the bold/proactive and shy/reactive pattern: more exploratory capuchin monkeys exhibited increased values of proeminence in Pacing, aberrant sexual display and Active 1 BPIS, while less active animals exhibited increased probability in significant sequences involving at least one BPIS, and increased prominence in own stereotypy. Capuchin monkeys are known for their cognitive capacities and behavioural flexibility, therefore, the search for a consistent set of behavioural indictors of welfare and individual differences requires further studies and larger data sets. With this work we aim contributing to design scientifically grounded and statistically correct protocols for collection of behavioural data that permits comparability of results and meta-analyses, from whatever theoretical perspective interpretation it may receive.

Abstract:
Social insect workers can vary in terms of body size, space use, division of labor, and reproductive potential. Here, I begin to 'dissect' a social insect colony, using the bumble bee Bombus impatiens, to determine how this variation affects colony organization. I found that workers are spatially organized inside the nest and they remain at a specific distance from the colony center. Bees that feed larvae tend to remain in the center, whereas foragers are more often found on the periphery when not foraging. Smaller workers are more likely to feed larvae and incubate brood, and larger workers are more likely to fan or guard the nest. Still, workers perform multiple tasks throughout their life. The size of this task repertoire does not depend on body size or age. Furthermore, workers that remain further from the queen while inside the nest and avoid energy-expensive tasks during the ergonomic phase are more likely to reproduce by the end of the colony cycle. Inactive bees are not, however, defensive reserves. Although inactive bees increase their speed inside the nest when the nest was disturbed, they were not more likely to leave the nest (presumably to attack the simulated attacker) or switch to guarding behavior. This suggests that inactive bumble bees that remain farther from the queen may be storing fat reserves to later develop reproductive organs. Finally, I examined how within-group variation affects colony performance. I reduced variation in body size or temperature response thresholds by removing individuals from the colony with extreme phenotypes, and compared colony performance to colonies where random bees were removed. Colonies took longer to cool down the nest after bees were removed, but this effect was most striking when variation in temperature thresholds was reduced. Further, although larger bees are better at carrying items and are more likely to fan, the ability of colonies to perform undertaking behavior or thermoregulation was not affected when size variation was reduced. These studies provide evidence that (1) within-group variation affects colony organization and (2) variation among workers in their inherent tendencies to respond to stimuli positively affects colony performance.

Abstract:
Free-riding and coordination are two types of potential problems in group incentive contracts, specifically in incentive contracts based on budget target group. The social value orientation (SVO - Social Value Orientation), individual difference measure that classifies people in individualistic - maximizers of self-interest, competitors - maximizing the relative advantage, and prosocial - maximizing the outcome of the group -, is an important fator which has implications for the design of group incentive contracts. Upton (2009) revealed that based on the social value orientation of individuals, pro-social value orientation of individuals have higher performance and less likely to free-ride on the contributions of members of the group than individuals having individualistic value orientation and competitor. In addition, for companies that adopt this type of contract for the supply of incentive group, the selection of workers prefer the pro-social type would be best suited to ensure the group in labor productivity, and improving business performance. In this context, this thesis seeks to show that while the SVO is an important consideration for the contract design, other factors such as group identity is also an element to be incorporated in this process, since one of the major challenges to solve the problems of a social dilemma situation as the group incentive context, is to promote cooperative behavior in group production environment. Therefore, manipulations based on the activation of identity, the process by which individuals manifest their individual preferences rewards or group could have an influence on the performance of groups with different types of SVO. This study aimed to investigate the moderating role of identity activation in the effect of the SVO on the group of performance incentive contract based on budgetary target group. In one experiment, 135 students of undergraduate course of four higher education institutions of the Paraíba State conducted a repeated game of public good dilemma in the experimental conditions of personal identity and group identity. The results showed statistically significant main effect of identity in the group of performance, while the SVO had no significant effect. They have found evidence of moderation in SVO effect of group identity activation in groups of participants with SVO individualistic and competitive, thus confirming the hypothesis of transformation motives. However, no effect was observed in groups of participants with pro-social SVO, thereby disproving hypothesis amplification motives.

XML has become a widely used standard for data representation and exchange in applications. The growing usage of XML creates a need for ecient storage and recovery systems for XML data. Native XML DBs (NXDBs) are being developed to target this demand. NXDBs implement many characteristics that are common to traditional DBs, such as storage, indexing, query processing, transactions and replication. Most existing solutions solve the replication issue through traditional techniques. However, the exibility of XML data imposes new challenges, so new replication techniques ought to be developed. To improve the performance and availability of NXDBs, this thesis proposes RepliX, a mechanism for XML data replication that takes into account the main characteristics of this data type, making it possible to reduce the response time in query processing and improving the fault-tolerance property of such systems. Although there are several replication protocols, using the group communication abstraction for communication and fault detection has proven to be a good solution, since this abstraction provides ecient message exchanging techniques and conability guarantees. RepliX uses this strategy, organizing the sites into an update group and a read-only group in such a way that allows for the use of load balancing among the sites, and makes the system less susceptible to faults, since there is no single point of failure in each group. In order to evaluate RepliX, a new replication layer was implemented on top of an existing NXDB to introduce the characteristics of the proposed mechanism. Several experiments using this layer were conducted, and their results conrm the mechanism's eciency considering the dierent aspects of a replicated database, improving its performance considerably, as well as its availability.

Abstract:

The purpose of the study was to determine whether there were associations between hospital costs and hospital size, hospital location, hospital ownership type, and Group Purchasing Organization (GPO) participation. The 2008 American Hospital Association annual survey responses were used as the instrument for a retrospective secondary study. Analyzing results from the various tests employed in this study showed there was a statistically significant association between hospital costs and hospital size. There were statistically significant differences between the means when comparing cost with hospital location and ownership type. There was not an association between hospital costs and GPO participation, likely as a result of the unadjusted cost metric used. Further research using a time series approach would help determine the true association of GPO participation. Expanding the criteria to identify whether the hospitals participate in a regional alliance may also return different results relative to cost and GPO participation.

Abstract:

The purpose of this paper is to examine the role that multilingualism plays in the identity formation of German-speaking emigrants in South Africa. In the era of globalization society multilingualism has gained greater value, to such an extent that multilingualism can be seen as the norm, whereas monolingualism is the exception. The importance of multilingualism and its effect on an individual‟s social and language identity has been debated by leading German academics such as Esser, Dirim and Krumm. In the South African context multilingualism is part of daily life, therefore it is not questioned how this multicultural world influences the way South Africans see themselves and interact with the world around them. Here the question comes to mind in what way this language-cultural diversity effects the identity formation of emigrants, who speak a language that is not part of the 11 official languages of South Africa. Therefore it is important to determine in what way the language and the social environment influence the development of their individual and social identities as well as determine in what way they contribute to the active integration into the community. With the help of different theories as well as through an empirical study of German immigrants in South Africa this paper will focus on their social and language identity and what effect, if any, multilingualism has on their identity formation. The paper attempts to explain theoretically the connection between language and identity and then through empirical research demonstrate the influence of language on identity and integration.

Abstract:
Physical education classes are mandatory for all healthy students. Problem in physical education classes for pupils with health problems still remains. Most of these students are able not to atend physical education classes with doctors permission. Physical exercises are particularly important for poorer health students that are usually assigned to specific schools, a medical team of physical fitness. It implies an inferior physical development of students complaining impaired cardiovascular, respiratory, visual or other body systems. Specially adapted physical education not only enhances the body of these pupils and promote good physical development, it helps even in cases where medical treatment is helpless. Researchers demonstrated that exercise and fitness are important for various disorders and preventive measures. That not to affect the health of students, teachers must adequately select tasks, each track, as they are able to meet the requirements. Abstract:
This work aims to increase the predictivity of the two-fluid model in gas-solid fluidized beds of Geldart A particles. The first part deals with the integration of physical phenomena into the two-fluid model: shear-induced collisions, lubrication between particles and inter-particular forces. The second part deals with the practical resolution of the two-fluid model using numerical codes. It is shown that bed expansion for Geldart A particles have a strong dependance on the mesh size used to compute the flow, leading to a major overestimation. The filtered approach is used and points up the filtered drag as the key of the problem. Two subgrid drag models are proposed and tested on both academic and laboratory configurations

Shayesteh, Alireza. "High-resolution infrared emission spectroscopy of diatomic and triatomic metal hydrides." Thesis, University of Waterloo, 2006. http://hdl.handle.net/10012/1216.

Full text
Abstract:
Several hydrides of Group 2 and 12 elements were generated in the gas phase using an emission source that combines an electrical discharge with a high temperature furnace, and their high-resolution infrared emission spectra were recorded with a Fourier transform spectrometer. Two classes of molecules were studied: a) diatomic metal hydrides BeH, MgH, CaH, SrH, ZnH and CdH; b) linear triatomic metal hydrides BeH2, MgH2, ZnH2 and HgH2.

Infrared emission spectra of BeH, MgH, CaH, SrH, ZnH and CdH free radicals contained several vibration-rotation bands in their 2SIGMA+ ground electronic state. The new data were combined with all the previous ground state data from diode laser infrared spectra and pure rotation spectra available in the literature. Spectroscopic constants, i. e. , vibrational band origins, rotational, centrifugal distortion, and spin-rotation interaction constants, were determined for each observed vibrational level by least-squares fitting of all the data. In addition, the data from all isotopologues were fitted simultaneously using the empirical Dunham-type energy level expression for 2SIGMA+ states, and correction parameters due to the breakdown of the Born-Oppenheimer approximation were determined. The equilibrium internuclear distances (re) of 9BeH, 24MgH, 40CaH, 88SrH, 64ZnH and 114CdH were determined to be 1. 342424(2), 1. 729721(1), 2. 002360(1), 2. 146057(1), 1. 593478(2) and 1. 760098(3) angstroms, respectively, and the corresponding re distances for 9BeD, 24MgD, 40CaD, 88SrD, 64ZnD and 114CdD are 1. 341731(2), 1. 729157(1), 2. 001462(1), 2. 145073(1), 1. 593001(2) and 1. 759695(2) angstroms, respectively.

Gaseous BeH2, MgH2, ZnH2 and HgH2 molecules were discovered and unambiguously identified by their high-resolution infrared emission spectra. The ν3 antisymmetric stretching fundamental band and several hot bands in the ν3 region were rotationally analyzed, and spectroscopic constants were obtained for almost all naturally-occurring isotopologues. The rotational constants of the 000 ground states were used to determine the r0 internuclear distances. For BeH2, ZnH2, ZnD2, HgH2 and HgD2 molecules, the rotational constants of the 000, 100, 0110 and 001 levels were used to determine the equilibrium rotational constants (Be) and the associated equilibrium internuclear distances re. The re distances of ZnH2 and ZnD2 differed by about 0. 01%, and those of HgH2 and HgD2 differed by about 0. 005%. These discrepancies were larger than the statistical uncertainties by one order of magnitude, and were attributed to the breakdown of the Born-Oppenheimer approximation.

Children are increasingly more inserted in consumer relations, and in this context, the Tweens (children from 7 to 12 years) emerge as a promising public, for they are avid consumers, but vulnerable for being in a preliminary phase that may overvalue the benefits consumption. By listening to the voice of the children themselves, this study aimed to analyze how social relationships influence Tweens’ consumption of goods and brands. In order to achieve the general objective, we identified (1) how the context of consumption and interpersonal relationships influence the consumption of goods by Tweens, (2) how do relationships with peers are affected by the symbolic and functional aspects related to appropriation of goods and brands (3) how they deal with “not having”, in front of groups and in their individuality, and (4) how parents guide their consumption. For this we adopted a methodology of qualitative, exploratory characteristics, through in-depth interviews with the children and their family (mother or grandmother). The interviews with the children were initially developed through the e-collage technique, which served as guiding for the conduction the interviews. The collected data was analyzed by the use of content analysis technique. The results indicate that the Tweens are influenced by the market environment, parents and peers in different levels according to the categories of products. It was found that material goods facilitate interaction in groups while the charge for to have the "right" property can generate constraints affecting the status, self-esteem and self-concept. It also identified that the mothers caring for their children to have socially appropriate goods, with the concern that they do not have problems in their social relationships.

The educative project How to live well with the animals has been applied to educational institutions in the city of São Paulo SP Brazil and has two modules: responsible pets ownership and the other synanthropic fauna. The current study was written to analyze the dynamics of the educational process, evaluating this research subjects in the role of re-passing the information about synanthropic animals. The course offered by the Municipal Center of Zoonosis Control (CCZ), in São Paulo (SP), is offered to multiplier teachers. This course was accompanied and the taught topics to the multipliers were evaluated by applying a self-filling-questionnaire, before and after the course. The teaching action on this taught topics, was accompanied in the school where two multipliers worked. The taught topics were evaluated in the same way again, but considering the teachers, who had received information from the two multipliers, and the children, who studied in the school, and these childrens parents or tutors. The self-filling-questionnaire was composed of free open answers in order to evaluate the taught topics. The topics were statistically studied by McNemar and Kappa concordance test. Another questionnaire with part-closed questions was used to study the social insertion which was studied by the Social Homogeneous Group (SHG) methodology. The correspondence analysis ANACOR-was used to study relations among the SHG and the taught points. The study indicated that PVBB project increases multipliers information about synanthropic animal, but the multiplier could not act on re-passing the information and technical taught to the teachers in the school. The participation of the multipliers in the course was not sufficient for preparing them to act in this way. According to the possibilities the teachers could re-pass part of the topics with their students. There was no sigificative impact on the students action of re-passing the information to their parents or tutors, but it was interesting to observe that the re-passing actions was different according to the homogeneous social group, and the possibility of dialogue between the child and the familiars. The use of SHG methodology could evaluate the social insertion and different association tendencies between the SHG and the answers written by the parents and tutors.

ABSTRACT: This is a pre - trial basis study with repeated measures that allows to analyze the impact of the activity of the group singing / choir, as a recovery strategy, the subjective welfare of the people . Five scales were used, as " Life satisfaction " , " Spane " and " Flourishing " helped analyze the Subjective well -being and "Need for Recovery Scale" and " Self- Perception of Individual Performance " helped analyze variables related to fatigue and labor activity . A sample was collected from 69 subjects in groups of four singing groups/ independent choruses, wherein one of the groups had religious/spiritual effect. The results revealed a temporal stability (3 weeks) in five scales, and the correlation varied between 0.60 and 0.81. There was also a significant statistically decrease in the level of positive affect, and a statistically significant increase in the level of positive affect after participation in the activity of group singing / choir. The religious/ spiritual influence tends to be a moderator of affective responses after participation in the activity role , although not statistically significant. It was found in the total sample, a high level of negative affect ( Spane state version ) and lower levels in the scale " Satisfaction with life " can be predictors of a high need for recovery. For individuals who work, it was found that high levels on the scale " Flourishing " constitute a predictor for elevated levels in the " Self- Perception of Individual Performance " scale.

Abstract:
Microfinance has generally been acclaimed as one way to reduce poverty through the provision of financial services targeting the previously “unbankable” poor. Persons with Disabilities (PWDs) are amongst society’s most excluded groups financially and the absence of PWDs within mainstream Microfinance urges Disabled People’s Organisations (DPOs) to directly engage in Microfinance. The practices by such alternative actors are suggested to be generally rejected because they risk being inefficient and failing. The objective of this study is to analyse examples of how DPOs in Nepal practice Microfinance and whether they generally follow recommended best practices. The correlation with what is perceived as best practices provide evidence to suggest whether the DPOs’ practices should be generally rejected or motivated as plausible and justified accordingly. To accomplish this, a well-justified analytical framework of recommended best practices for DPOs engaged in Microfinance is created, and field research is undertaken in Nepal, April-May 2014. It can be concluded that recommended best practices are followed at a general level; however, the extent varies within and between the analysed practices. The results provide sufficient evidence to suggest that the practices are plausible and justified accordingly, and should not be generally rejected.

Abstract:
In last decades, researchers have demonstrated that lipids are not only structural compounds of cellular membranes. These compounds have several functions in cell metabolism, specifically, phospholipids are cleaved by phospholipases, and their metabolites are secondary messengers in inflammation pathways. Moreover calcium-independent phospholipase A2 (iPLA2) might be linked to schizophrenia. Synthesis of new inhibitors of this enzyme has been the main aim of this work.
In our group we have designed different analogues of the only inhibitor of this enzyme (BEL). We introduced several modifications and functional groups, like trifluoroketones, in BEL's structure. Unfortunately, any of these compounds could not inhibit the activity of the enzyme. Some compounds that have an ariloxigroup in alpha position of ketone increase activity of iPLA2 and not produce the same effect in cPLA2.
Then, we have synthesized analogues of the natural substrate of phospholipases with several modifications; thioether group in sn-1 position, phosphorilcholine group in sn-3 position and different functional groups (amide, carbamate, sulfonamide.) in sn-2 position.
Some of these compounds show inhibition of iPLA2, moreover, they also inhibit cPLA2.
To know more information about catalytic mechanism of this phospholipase and design new inhibitors we have cloned and expressed two fragments of catalytic site of the enzyme. However these proteins are not soluble and its levels of expression were very small, and we could not do structural studies like X-ray crystallization.
Furthermore, we have designed and synthesized analogues of ceramide. Biological assays of these compounds in several enzymes of sphingolipid metabolism proved that this family opens a new research line in sphingolipid field.

Abstract:
This doctoral thesis deals with the socio-institutional trajectories of a group of youth with low-level qualifications or no qualifications at all, who benefit from a reinsertion policy measure designed to help them find job. The purpose of this research is to study how this target-group of youth – characterized by multiple forms of disqualifications – is getting the proposed support. First and foremost, it should be noted that those who have resorted to public assistance have been long wrongly considered a homogeneous social stratum, whereas indeed, there are several ways to deliver social support, depending on individuals, their different life experiences or even the specific phases they are go through by the time they ask for help. In line with this assertion, following the model of medical diagnosis, I undertook an“anamnesis” towards understanding, more or less, the etiological history of those male and female as well, finding themselves “ à posteriori” playing the institutionalized part of“pathological” trainees. From that point of view, I had to study different spaces that are namely : the family, the school and the institutional environment as well as their impacts on the socio-professional trajectories of the individuals concerned. In other words, the objective was to check to what extent, based on social supports received (nature, intensity, quality of resources ...), that youth may find the springs and dynamics allowing them to trace their own path.It is in this context that I integrated two associations active in social work towards facilitating the process of inserting young people having difficulties to enter the world of employment. Considered as communicational spaces, the “Mission Locale” and particularly “l’Atelier” were my privileged places of ethnographic observation, where I strived to single out the games of actors, power games, objectives and the multiple interactions entwined in these territories. A specific interest was put on personal experiences, perceptions, conflictual and /or cooperative interactions between social workers and beneficiaries. Jogging, basket (shoes), cap position (back / front) or street language, this field survey also allowed me to study the esthetics, postures and body expressions of young people by looking at the significance they take as individuals and / or collective action, particularly within social associations. In a broader prospect, I have tried to see how and to what extent the agents / actors of the social institutions take into account the specificities of this public and manage to create points of balance between the request of the beneficiaries and the institutional command, between imperatives as mediate and immediate structural constraints

Abstract:

Abstract:
Group 3 Innate Lymphoid cells (ILC3) actively maintain mucosal homeostasis through the production of Interleukin-22 (IL-22). ILC3 encompass 2 major populations, LTi (« Lymphoid Tissue inducer »), characterized by the expression of the chemokine receptor CCR6, and ILC3 that express the transcription factor T-bet, which include a population expressing the surface marker NKp46, a receptor originally used to identify group 1 ILC (ILC1). ILC1 plays a major role in the defense against intracellular pathogens and anti-tumoral responses. Three major ILC1 populations have been identified: the cytotoxic lymphocytes « Natural Killer » (NK or ILC1b), which largely rely for on the transcription factor Eomes their generation and express the integrin CD49b; hepatic and intestinal ILC1 that depends on the T-bet transcription factor and express CD49a (ILC1a); and a population that expresses CD49a and CD49b (ILC1ab) and populates the salivary gland and the uterus, which is independent of the transcription factor Nfil3. My work aimed to understand the biology of NKp46 expressing ILC, as well as factor involved in their development, maturation and function. The major part of my work focuses on NKp46+ ILC3. First, we demonstrate a major role for the chemokine receptor CXCR6 in their localisation in the lamina propria villi (Satoh-Takayama et al. 2014). Second, I showed that NKp46+ ILC3 could lose NKp46 expression (Verrier et al. 2016). Induced by TGFβ, this loss of expression was associated with higher IL-22 production and by the acquisition of markers identifying LTi (CCR6, MHC-II), demonstrating NKp46+ ILC3 plasticity. Finally, in collaboration with Rachel Golub’s group, we confirmed a putative role for Notch-signaling in this plasticity (Chea et al. 2016). In this manuscript, I will discuss the development and the heterogeneity of ILC3, ILC1a, ILC1b and ILC1ab. All the results I generated support a dynamic vision of ILC biology, which reflects how they adapt in response to environmental cues. By characterizing the different actors involved in this dynamic process, my work could be used to design therapies aiming at controlling the equilibrium between these different populations in diverse pathologies such as cancer, viral infection, or intestinal diseases

Special needs children integration into comprehensive schools affects a school, teachers‘ skills, curriculum, education management, children‘s assessment. Therefore it is essential to reveal comprehensive school teachers‘ opinion on special needs children as well as on existing competencies of special work, to analyse special needs‘ disorder groups and their degrees, to compare urban and rural school help for children with special needs. The research revealed that comprehensive school teacher treats special needs children the same as other ones. The classification of special needs individuals‘ disorder groups and their degrees helps to orient in the variety of disorders. Teachers are competent to work with pupils having specific cognitive, complex, emotional, behavior and development disorders, because they are experienced. The least information that teachers have is about sight, hearing, movement and position, chronic somatic and neurology disorders. Teachers are lacking various special knowledge and teaching methods. In urban schools there is a special pedagogue‘s , social pedagogue‘s, logopedical help provided for special children, but only a few schools provide psychologist‘s and teaching assisstant‘s help. There is no help provided for special needs children in most of rural schools. They are educated by teachers only. Just in a few rural schools there is a partial help provided by specialists for children in need.

Abstract:
Banco Espírito Santo Group, with a significant position in the banking sector, collapsed in 2014. The collapse had its apex in the fraud of the financial statements. When unveiled, the accumulated losses amounted to 2,750 million euros in the accounts of BES Group (CPI, 2015) already. The purpose of this study is to understand if there were fraud red flags in financial statements before the bank's collapse that could have allowed well informed investors' decisions. The financial reporting for the period between 2009 and 2013 were analyzed on the basis of a set of red flags as identified in previous studies, in order to understand if there were sufficient indicators that would have allowed both auditors and investors to incorporate the information in their analysis. Out of a total of 17 red flags analysed, 6 indicate possible fraud and all of them are related to the structure and management of the Banco Espírito Santo Group, such as: complexity of the Group, variable portion of executives' remuneration dependent on the Group's profits, dependency on BES - the "heart" of the entire group- to raising funds to complete the capital increases made in both 2011 and 2013. GBES's winding-up process is not yet complete, much information is still confidential and this limited this research. It should be noted that the "fraud" phenomenon is not sufficiently studied in Portugal.

Abstract:
Cooperation is pervasive in nature but paradoxically also provides opportunity to cheaters. My dissertation involves the study of both cooperation and conflict in two species of Megalopta bees. Megalopta is a Neotropical genus of halictid bees whose biology is characterized by complex life cycles that can range from solitary to eusocial. These bees nest in dead wood and forage under dim light conditions. Megalopta’s nests are inhabited by an extensive array of organisms and each nest therefore constitutes a miniature ecosystem providing opportunities for cooperation and conflict, both within and between species. I first delineate the social structure of M. genalis and M. ecuadoria nests in several Panamanian populations and integrate the factors that play a role in the behavioral decisions of females when joining a social group or not. Within a kin-selection framework, I discuss how genetic relatedness plays a role in the formation of social nests. Second, I investigate the conflict between host bees and a congener social parasite, and I elucidate reproductive structures that are relevant for understanding the evolution of parasitism. Finally, I describe a cleaning mutualism between Megalopta bees and their mite associates. Bee-mite associations encompass a broad spectrum of interspecific interactions. Some bee-mites are thought to perform cleaning services for their hosts in exchange for suitable environments for reproduction and dispersal. Field observations and experimental manipulation reveal a significant correlation between the presence of mites and the absence of fungi inside the brood cells, as well as between the absence of mites and increased bee mortality. This study therefore provides evidence of the sanitary effect of mites in nests of Megalopta bees. This bee-mite association constitutes one of the few examples of terrestrial cleaning mutualisms.
