Abstract:
Abstract Hematopoietic system is maintained by the differentiation and proliferation of hematopoietic stem/progenitor cells (HSPCs) and their commitment to the mature blood cells should be tightly controlled by gene regulatory networks (GRNs) governed by transcription factors (TFs). To keep the homeostasis, GRNs should respond to the environmental changes, such as infection. However, the precise mechanism of such a system remains to be elucidated. TFs Bach1 and Bach2 belong to the basic region-leucine zipper family and recognize Maf-recognition elements containing AP-1 site (Oyake et al., 1996). We have previously shown that Bach1-/-Bach2-/-(DKO; double knockout) mice show erythropoiesis disorders with increased myelopoiesis from common myeloid progenitors (CMPs), which is an erythro-myeloid bifurcation point (ASH2015; Abstract ID# 81562) (Akashi et al., 2000). Since this phenotype is similar to that of LPS treated mice (O'Connell et al., 2008), we hypothesized that Bach factors work as sensors for infection. First, to evaluate the cell-intrinsic function of Bach factors, WT or DKO bone marrow cells were depleted of mature differentiated cells and transplanted to lethally irradiated WT mice. After 8 weeks, DKO donor cells showed greater myelopoiesis and lesser lymphogenesis compared to WT, suggesting Bach factors are necessary to suppress myelopoiesis to the appropriate level in regenerating hematopoiesis. To reveals the function of Bach factors in HSPCs from other aspect, LSKs (Lin-Sca1+c-kit+) were infected with retroviruses expressing Bach1-IRES-eGFP or Bach2-IRES-eGFP and transplanted to lethally irradiated WT mice. Cells derived from transgene induced LSKs were monitored by GFP fluorescence. After 2 weeks, Bach1 overexpressing LSKs did not show any difference in erythropoiesis and myelopoiesis. This might be explained by the high Bach1 expression levels in HSPCs according to the previous report (Lara-Astiaso et al., 2014). On the other hand, Bach2 overexpressing LSKs showed increased erythropoiesis and decreased myelopoiesis, suggesting that Bach2 regulates the erythro-myeloid lineage specification as expected by the observations of DKO mice. To assess the function of Bach factors under infection, we used M1 murine myeloid leukemia cells that differentiate to macrophage-like cells by LPS stimulation. LPS stimulation reduced expressions of Bach1, Bach2 and erythroid gene Gata1, and induced those of myeloid genes such as Cebpb and Csf1rin a dose-dependent manner. To determine if down-regulation of Bach factors is necessary for myeloid differentiation, Bach1 or Bach2 were transgenically overexpressed in M1 cells. Both of the M1 cells overexpressing Bach1 or Bach2 showed lower expression levels of myeloid marker CD11b compared to control under LPS stimulation. Thus, reductions of the expression of Bach factors in response to LPS were necessary for appropriate myeloid differentiation. To identify the direct target genes of Bach factors, Bach1 or Bach2 ChIP-seq data of M1 cells (Ebina-Shibuya et al., 2016) were merged with results of expression profile of WT and DKO CMPs. Several myeloid or inflammatory genes such as Cebpb, Fcgr1 and Tlr4 were identified as putative repressed target genes and several erythroid or lymphoid genes such as Klf1, Rag1 and Rag2 were identified as putative activated target genes. In addition, when Bach1 or Bach2 ChIP-seq data were merged by that of C/EBPb, which also possesses AP-1 site as its target motif, obtained from ENCODE database (ENCSR000AIB), we found that there were several co-localized regions of Bach and C/EBPb near the myeloid genes such as Cebpa, Il6 and Fcgr1. These observations suggest that Bach factors repress myeloid genes by competitively working with C/EBPb at same genomic regions. This is particularly interesting in the light of the latest findings showing the Bach2 function on AP-1 site in lymphoid cells (Sidwell et al., 2016). These results reveal a novel mechanism by which how the differentiation of erythro-myeloid bifurcation is controlled by responding to environmental changes. Bach factors regulate erythro-myeloid competitive differentiation by promoting and repressing erythroid and myeloid differentiation, respectively. We suggest that infection promote myelopoiesis at the expense of erythropoiesis by reducing the expression of Bach factors. Therefore, Bach factors may function as sensors for environmental changes. Abstract:
Members of the small Maf family (MafK, MafF, and MafG) are basic region leucine zipper (bZip) proteins that can function as transcriptional activators or repressors. The dimer compositions of their DNA binding forms determine whether the small Maf family proteins activate or repress transcription. Using a yeast two-hybrid screen with a GAL4-MafK fusion protein, we have identified two novel bZip transcription factors, Bach1 and Bach2, as heterodimerization partners of MafK. In addition to a Cap'n'collar-type bZip domain, these Bach proteins possess a BTB domain which is a protein interaction motif; Bach1 and Bach2 show significant similarity to each other in these regions but are otherwise divergent. Whereas expression of Bach1 appears ubiquitous, that of Bach2 is restricted to monocytes and neuronal cells. Bach proteins bind in vitro to NF-E2 binding sites, recognition elements for the hematopoietic transcription factor NF-E2, by forming heterodimers with MafK. Furthermore, a DNA binding complex that contained MafK as well as Bach2 or a protein related closely to Bach2 was found to be present in mouse brain cells. Bach1 and Bach2 function as transcription repressors in transfection assays using fibroblast cells, but they function as a transcriptional activator and repressor, respectively, in cultured erythroid cells. The results suggest that members of the Bach family play important roles in coordinating transcription activation and repression by MafK.

Der Artikel geht auf die Echtheit folgender J. C. Bach möglicherweise zuzuordnender Werke ein: Präludium und Fuge ES-Dur BWV Anh. 177, zwei Variationsreihen über Aria-Melodien in ES-Dur und a-moll und eine über eine Sarabande, sowie Choralbearbeitungen. Eine Zugehörigkeit zu den Sarabanden-Variationen J. C. Bachs wird im Artikel auch für BWV 990 diskutiert. Unter den Choralbearbeitungen wird besonders Ach Gott, vom Himmel sieh darein beleuchtet, die als BWV 741 bisher J. S. Bach zugeschrieben wurde, im vorliegenden Text aufgrund verschiedener Überlegungen jedoch J. C. Bach zugewiesen wird. Zudem werden besonders verschiedene Einflüsse von Werken J. C. Der Autor stellt sich der tradierten Auffassung entgegen, Bachs Vokalschaffen sei seinerzeit lediglich ausnahmsweise außerhalb seiner eigenen Wirkungsstätten erklungen und er lediglich als Tastenvirtuose von weitergehendem Ruf gewesen. Er skizziert die Genese dieses Bach-Bildes und zeichnet parallel die Entwicklung einer gegenläufigen Sicht nach, die sich auf verschiedene nach und nach zu Tage getretene Dokumente stützt. Um solche systematisiert und stichhaltig auswerten zu können, werden zunächst die Grundlagen der Auswahl und Auswertung solcher Quellen festgestellt. Vier Beispielbetrachtungen zu mitteldeutschen Kirchenmusikerkollegen Bachs (G. G. Wagner, J. T. Krebs d. Ä., J. A. Matthaei, G. F. Der Artikel versammelt vier Fallstudien mit dem Ziel, anhand von neuem Quellenmaterial einige offene Fragen stilkritischer, werkgeschichtlicher und biografischer Natur zu Johann Sebastian Bach einer Klärung entgegen zu führen. Die erste Studie widmet sich einem Beleg für Bachs frühe Beschäftigung mit der Gattung der italienischen Solokantate, den ein in der Staatsbibliothek zu Berlin verwahrtes Einzelblatt mit dem Beginn einer Komposition Antonio Biffis liefert, das von Bachs Hand stammt. Ebenfalls in der Berliner Staatsbibliothek fand sich der originale Textdruck zur Aufführung von BWV 194 bei der Orgelweihe in Störmthal, der im nächsten Abschnitt vorgestellt wird. Die besprochenen Quellen werden in Faksimiles wiedergegeben. Der dritte Teil widmet sich einer verschollenen Trauungskantate, die die Urbilder von BWV 195/1 und /5 enthalten haben soll. Über dieses Werk gibt ein Textdruck aus dem Schlossmuseum Sangerhausen Auskunft. Die letzte Studie behandelt die im Stadtarchiv Eilenburg vorhandene Akte zur Wiederbesetzung der Konrektorenstelle an der dortigen Stadtschule 1749, zu der die Superintendentur Delitzsch eine Parallelakte verwahrt und die eigenhändige Zeugnisse Bachs enthält. Die drei Fallstudien des Artikels stellen neue Erkenntnisse zur Biografie Johann Sebastian Bachs und der frühen Rezeption seines Schaffens vor. Schostakowitsch hat beide Bände des Wohltemperierten Klaviers bereits als 12jähriger gespielt, als er das Klavierspiel bei Ignati Gljasser, einem Schüler von Hans von Bülow, erlernte. So hat ein Petersburger Junge die deutsche Schule kennengelernt. Im Konservatorium studierte Schostakowitsch später das Klavierspiel bei dem hervorragenden Pädagogen Leonid Nikolajew, einem Schüler von Sergei Tanejew. Bei ihm herrschte der Kontrapunkt der strengen Schreibart vor. Nikolajew schenkte den Werken Bachs eine große Aufmerksamkeit.

Abstract:
Schostakowitsch hat beide Bände des Wohltemperierten Klaviers bereits als 12jähriger gespielt, als er das Klavierspiel bei Ignati Gljasser, einem Schüler von Hans von Bülow, erlernte. So hat ein Petersburger Junge die deutsche Schule kennengelernt. Im Konservatorium studierte Schostakowitsch später das Klavierspiel bei dem hervorragenden Pädagogen Leonid Nikolajew, einem Schüler von Sergei Tanejew. Bei ihm herrschte der Kontrapunkt der strengen Schreibart vor. Nikolajew schenkte den Werken Bachs eine große Aufmerksamkeit.

Abstract:
<p>The Bach Dang Bridge forms part of the Ha Long – Hai Phong Highway in Vietnam. It will help cut 50km from the journey between Hanoi and the Ha Long Bay UNESCO world heritage site. The focal cable-stayed section of the bridge will consist of two main spans of 240 m. The tallest of the three towers will reach a height of almost 100 m.</p><p>The construction technique used for the cable-stayed section is to cast 9.6 m-long and 28 m-wide concrete segments in situ, using an underslung form traveller (FT) which is launched to the next position once the segment is cast.</p><p>The FT has been designed by the Technical Centre of specialist contractor VSL. Its weight is minimised by using the stay cable as a support at the front of the traveller. The stay cable is connected to the FT by precasting the anchor block and securing it to the FT. This innovation was previously, successfully used by VSL on the Ironton-Russell project in the USA. Besides, the formwork system has been mechanised to be easily collapsed from one segment to the next.</p><p>This paper discusses the design of this innovative construction technique and how it was used on this major bridge project in Vietnam.</p>
