Abstract:
The white-clawed crayfish is a complex of species (Austropotamobius pallipes plus A. italicus, Crustacea) pivotal to riverine environments. Regrettably, it is endangered in many European countries due to a recent massive decline. We carried out a fine-scale survey on white-clawed crayfish distribution, morphology and genetics in an under-investigated region of central Italy (Lunigiana, Tuscany) to improve local knowledge and aid conservation management of the species. The torrential fauna of Lunigiana is scarcely known and habitat alterations due to the anthropic exploitation of the territory represent a potential threat to its persistence. We investigated crayfish occurrence in eight streams of the Magra River Basin. We performed nocturnal and diurnal transects, took morphometric measurements, collected samples for genetic analysis and checked for the occurrence of ectosymbionts. Crayfish were disclosed in two streams (Civasola, Verdesina). Morphometric and phylogenetic analysis (mtDNA COI gene Bayesian tree reconstruction) concurrently identified both populations as Austropotamobius italicus carinthiacus. In the Verdesina stream, crayfish were heavily infested by branchiobdellidans (Annelida). Verdesina crayfish were also significantly smaller than those found in the Civasola stream, where branchiobdellidans were absent. Hence, we hypothesized that such difference in size might be related to the high density of ectosymbionts. In the light of habitat features and of data herein provided, we propose that both Civasola and Verdesina streams should be considered as “natural ark sites” for A. italicus, with the Magra River Basin representing a regional stronghold for the conservation of the species.

Abstract:
In endangered crayfish conservation projects, it is paramount to map the distribution of the causative agent of crayfish plague, Aphanomyces astaci, in native populations. Considering the inapplicability of the destructive cuticular sampling protocol for monitoring endangered populations, we explored the use of non-invasive sampling techniques to detect this pathogen with molecular assays. In the present study, we exploited environmental DNA (testing increasing water volumes combined with different filter porosities) and cuticular swabs to collect A. astaci DNA. In addition, we evaluated the impact of the storage method on DNA preservation during field activities. After the first evaluations performed on both highly infected Austropotamobius pallipes and carrier Procambarus clarkii specimens in laboratory conditions, these sampling techniques were applied to wild populations of white-clawed crayfish. Our findings highlight better results with the filtration of 5 L of water with filters of 2.7 µm porosity for eDNA analysis and demonstrate that cuticular swabbing is equally effective as the World Organisation of Animal Health’s protocol. Storage in absolute ethanol proved to be the best solution to preserve swabs and filter samples for up to a week at room temperature. In conclusion, we suggest an integration of both sampling methods when monitoring A. astaci for conservation purposes.

The white-clawed crayfish A. pallipes has suffered in recent decades a strong decline throughout its entire distributional range, mainly due to the growing number of threats coming from anthropic influence, including habitat loss and degradation, overfishing, infectious diseases, and the introduction of non-indigenous crayfish species (NICS). The species is included in the red list of the IUCN (International Union for Conservation of Nature) as a species at risk of extinction. An important goal in conservation biology is to assess the genetic variability and thus the “genetic health” of populations and to identify any evolutionarily significant unit (ESU) within endangered species, before management decisions are taken. Within RARITY (http://www.life-rarity.eu), a LIFE project for the eradication of the invasive Louisiana red swamp (Procambarus clarkii) and for the preservation of the native white-clawed crayfish in Friuli Venezia Giulia (NE Italy), I was responsible for the genetic characterization of A. pallipes complex in this area, in order to define the taxonomic status, the genetic variability and the population structure and differentition. The analysis of two mitochondrial genes (COI and 16 rDNA) of about 500 individuals from 58 monitored sites showed that the FVG crayfish belong to the A. italicus species, with two different subspecies present: A.i. carsicus and A.i.meridionalis.
