Abstract:
The present paper covers reproductive performance in an artificial-insemination (AI) program of the Sheep CRC Information Nucleus with 24 699 lambs born at eight locations in southern Australia across five lambings between 2007 and 2011. Results from AI with frozen semen compared well with industry standards for natural mating. Conception rates averaged 72%, and 1.45 lambs were born per ewe pregnant for Merino ewes and 1.67 for crossbreds. Lamb deaths averaged 21% for Merino ewes and 15% for crossbreds and 19%, 22% and 20% for lambs from ewes that were mated to terminal, Merino and maternal sire types, respectively. Net reproductive rates were 82% for Merino ewes and 102% for crossbreds. From 3198 necropsies across 4 years, dystocia and starvation-mismothering accounted for 72% of lamb deaths within 5 days of lambing. Major risk factors for lamb mortality were birth type (single, twin or higher order), birthweight and dam breed. Losses were higher for twin and triplet lambs than for singles and there was greater mortality at relatively lighter and heavier birthweights. We conclude that reproductive rate in this AI program compared favourably with natural mating. Lamb birthweight for optimum survival was in the 4–8-kg range. Crossbred ewes had greater reproductive efficiency than did Merinos.

Abstract:
Suboptimal reproductive performance of maiden (primiparous) ewes remains a source of inefficiency for the Australian sheep industry. However, the extent and causes of the poorer reproductive performance of maiden ewes on Australian sheep farms are not well understood. Here, we show the reproductive performance of maiden ewes relative to their multiparous counterparts on the same farms across Australia using a cohort survey. The difference in marking rate for non-Merino maiden ewe lambs compared to multiparous ewes was 58% (74 vs. 132%; p < 0.001), and this was attributable to a 50% difference in reproductive rate (109 vs. 159%; p < 0.001) and 16% difference in lamb survival to marking (67 vs. 83%; p < 0.001). The difference in marking rate for maiden Merino two-tooth ewes lambing at approximately 2 years-of-age compared to mature multiparous ewes was 22% (80 vs. 102%; p < 0.001) and this was attributable to a 24% difference in reproductive rate (108 vs. 132%; p < 0.001) and 3% difference for lamb survival (75 vs. 78%; p < 0.05). Positive correlations for reproduction traits (reproductive rate, lamb survival and marking rate) between maidens and multiparous ewes were observed for maiden Merino two-tooth ewes (p < 0.001), but these correlations were weak or non-existent for non-Merino ewe lambs. Strategies to improve both reproductive rate and lamb survival can address the poorer and more variable reproductive performance of maiden ewes.

Estimated breeding values for the selection of more profitable sheep for the sheep meat and wool industries are currently based on pedigree and phenotypic records. With the advent of a medium-density DNA marker array, which genotypes ~50 000 ovine single nucleotide polymorphisms, a third source of information has become available. The aim of this paper was to determine whether this genomic information can be used to predict estimated breeding values for wool and meat traits. The effects of all single nucleotide polymorphism markers in a multi-breed sheep reference population of 7180 individuals with phenotypic records were estimated to derive prediction equations for genomic estimated breeding values (GEBV) for greasy fleece weight, fibre diameter, staple strength, breech wrinkle score, weight at ultrasound scanning, scanned eye muscle depth and scanned fat depth. Five hundred and forty industry sires with very accurate Australian sheep breeding values were used as a validation population and the accuracies of GEBV were assessed according to correlations between GEBV and Australian sheep breeding values . The accuracies of GEBV ranged from 0.15 to 0.79 for wool traits in Merino sheep and from –0.07 to 0.57 for meat traits in all breeds studied. Merino industry sires tended to have more accurate GEBV than terminal and maternal breeds because the reference population consisted mainly of Merino haplotypes. The lower accuracy for terminal and maternal breeds suggests that the density of genetic markers used was not high enough for accurate across-breed prediction of marker effects. Our results indicate that an increase in the size of the reference population will increase the accuracy of GEBV.

Abstract:
Using performance from progeny born in 2007 and 2008 generated by the Information Nucleus program of the Cooperative Research Centre for Sheep Industry Innovation, preliminary estimates of heritability were obtained for a range of novel carcass and meat attributes of lamb relevant to consumers, including carcass characteristics, meat quality and nutritional value of lamb. Phenotypic and genetic correlations of live animal traits with carcass composition and meat quality traits were also estimated. The data were from progeny located at eight sites, sired by 183 rams from Merino, maternal and terminal meat breeds and were representative of the Merino, Border Leicester × Merino, Terminal × Merino and Terminal × Border Leicester-Merino production types of the Australian sheep industry. Data were available from 7176 lambs for weaning weight, 6771 lambs for ultrasound scanning and 4110 lambs for slaughter traits. For the novel meat quality traits, generally moderate to high heritability estimates were obtained for meat quality measures of shear force (0.27 aged 1 day, 0.38 aged 5 days), intramuscular fat (0.39), retail meat colour (range of 0.09 to 0.44) and myoglobin content (0.22). The nutritional value traits of omega-3 fatty acids and iron and zinc contents tended to have low to moderate heritabilities (0.11–0.37), although these were based on fewer records. Fresh meat colour traits were of low to moderate heritability (0.06–0.21) whereas measures of meat pH were of low heritability (~0.10). For the carcass traits, estimates of heritability were moderate to high for the various measures of carcass fat (0.18–0.50), muscle weight (0.22–0.35), meat yield (0.24–0.35), carcass muscle dimensions (0.25–0.34) and bone weight (0.27). Results indicate that for most lamb carcass and meat quality traits there is sufficient genetic variation for selection to alter successfully these characteristics. Additionally, most genetic correlations of live animal assessments of bodyweight, muscle and subcutaneous fat with the carcass and meat quality traits were favourable. Appropriate definition of breeding objectives and design of selection indexes should be able to account for the small unfavourable relationships that exist and achieve the desired outcomes from breeding programs.

Estimates of genetic parameters for carcass traits in Merino sheep are presented, including muscle pH and colour, which are indicators of meat quality. The inclusion of these in the parameter set for genetic evaluation will provide the basis for more accurate estimated breeding values and the design of better breeding programs for improving meat traits in straightbred Merino sheep and crossbred lamb production. Data from 1045, nineteen-month-old Merino rams, the progeny of 125 sires from 3 strains that are representative of the industry, were used. Heritability and genetic correlations were estimated by fitting an animal model using a REML procedure in univariate and multivariate analyses. The traits included fasted pre-slaughter liveweight; dressing %; carcass tissue depth at the GR site (FATGR); fat depth at the C site (FATC); eye muscle depth (EMD), width (EMW), and area (EMA); ultimate pH and colour (L*, a*, b*) of the eye muscle. Multivariate analyses were used to estimate genetic correlations among meat traits and between wool traits (clean wool weight and mean fibre diameter) and selected carcass and meat traits. Estimates of heritability were: liveweight 0.51 (±0.10), dressing % 0.39 (±0.10), FATGR 0.33 (±0.09), FATC 0.20 (±0.08), EMD 0.27 (±0.08), EMW 0.15 (±0.07), EMA 0.23 (±0.08), muscle pH 0.27 (±0.09), and 0.14 (±0.07), 0.02 (±0.06), and 0.04 (±0.06) for the meat colour measurements L*, a*, and b* respectively. There was a favourable genetic correlation between muscle pH and colour L* of –0.56 (±0.23). There were also significant genetic correlations between wool weight and FATGR (–0.34 ± 0.16) and between mean fibre diameter and muscle colour L* (–0.66 ± 0.27). The study indicates that there is genetic variation for carcass traits, including indicators of meat quality, with scope for selection to improve meat production and quality in Merino sheep.

Aims We examine the average performance of Border Leicester (BL) flocks that are part of LAMBPLAN, Australia’s sheep performance recording and genetic evaluation system. We also summarise outcomes from the New South Wales Department of Primary Industry’s research on BL improvement, and discuss the performance required for the BL to continue to be a pivotal part of the Australian prime lamb industry. Key results Modest gains have been recorded in the BL in growth since 2000 and faecal egg counts have reduced, suggesting improved worm resistance. The net reproductive rate has slightly improved, while inbreeding continues to increase. In work conducted by the New South Wales Department of Primary Industries, an improved BL flock, encompassing selection for multiple births and a broadening of the breed’s genetic base, had advantages over purebred BLs in many aspects of performance, including a 19.3% advantage in the net reproductive rate. Advantages in fertility were ~5%, litter size ~7% and lamb rearing ability ~5%. BL improved × Merino and (BL improved × BL) × Merino crossbred ewes also had advantages over traditional BL × Merino ewes in their joining performance when joined to Dorset rams. Conclusions Selection in the BL needs to emphasise traits in a manner consistent with their importance to commercial lamb production. Implications Embracing this selection and broadening the BL’s genetic base will assist the BL to maintain its place in the Australian lamb industry.

A cross-sectional telephone survey of post-weaning sheep management and mortality was conducted involving 1410 farmers from across Australia. The average reported post-weaning mortality was 4.6%. Mortality was greatest in Queensland and Western Australia, and least in New South Wales, Victoria, and South Australia. Weaner mortality was also greater in the pastoral zone than in the sheep–cereals or high-rainfall zones. Overall, 44% of farms had mortality identified as ‘high’, exceeding the suggested benchmark of ≤4% per annum. High mortality was reported on 50% and 32% of farms with predominantly Merino and crossbred weaners, respectively. There was no statistically significant association between high mortality and the main month of lambing for a flock. Larger sheep flocks, flocks with a smaller proportion of weaners, and farms of smaller area were associated with a greater likelihood of high weaner mortality. The odds of high mortality in weaner flocks that were routinely separated according to bodyweight or condition score was half that of flocks that were managed as one group. Overall, 84% of farmers regularly provided supplementary feed to weaner sheep, but the kind of supplement offered and the proportions of farms routinely supplementing differed between states and sheep production zones. Only high-protein supplementary feeding was associated with lower odds of high mortality. This survey confirms that poor post-weaning survival remains a widespread issue for the Australian sheep industry but identifies farm and management factors associated with reduced weaner mortality.

Genetic variation within and between Australian Merino subpopulations was estimated from a large breeding nucleus in which up to 8500 progeny from over 300 sires were recorded at eight sites across Australia. Subpopulations were defined as genetic groups using the Westell–Quaas model in which base animals with unknown pedigree were allocated to groups based on their flock of origin if there were sufficient ‘expressions’ for the flock, or to one of four broad sheep-type groups otherwise (Ultra/Superfine, Fine/Fine-medium, Medium/Strong, or unknown). Linear models including genetic groups and additive genetic breeding values as random effects were used to estimate variance components for 12 traits: yearling greasy and clean fleece weight (ygfw and ycfw), yearling mean and coefficient of variation of fibre diameter (yfd and ydcv), yearling staple length and staple strength (ysl and yss), yearling fibre curvature (ycuv), yearling body wrinkle (ybdwr), post-weaning weight (pwt), muscle (pemd) and fat depth (pfat), and post-weaning worm egg count (pwec). For the majority of traits, the genetic group variance ranged from approximately equal to two times larger than the additive genetic (within group) variance. The exceptions were pfat and ydcv where the genetic group to additive variance ratios were 0.58 and 0.22, respectively, and pwec and yss where there was no variation between genetic groups. Genetic group correlations between traits were generally the same sign as corresponding additive genetic correlations, but were stronger in magnitude (either more positive or more negative). These large differences between genetic groups have long been exploited by Merino ram breeders, to the extent that the animals in the present study represent a significantly admixed population of the founding groups. The relativities observed between genetic group and additive genetic variance components in this study can be used to refine the models used to estimate breeding values for the Australian Merino industry.

The hypothesis tested in this experiment was that Merino lambs with lower birthweights, due to poor ewe nutrition during pregnancy, would have more fat and less muscle than Merino lambs with higher birthweights. At two sites (Victoria and Western Australia) in each of 2 years, a wide range in the liveweight profiles of ewes was generated during pregnancy and lactation by varying the amount of supplements fed and feed on offer grazed. Progeny had fat and muscle depth at the C-site measured at various ages from 8 to 28 months of age. Across the four experiments, there were differences of 0.5 kg in birthweights and 8 kg in weaning weights between extreme treatments. The effects on the depth of fat and muscle were very small with fat depth increasing by 0.1–0.2 mm (5–7%) and muscle depth increasing by 0.2–0.7 mm (1–3%) when birthweights decreased by 1 kg. The effects of birthweight on fat depth are consistent with our hypothesis whereas the effects of birthweight on muscle depth are in contrast to our hypothesis. Nevertheless, the impacts of birthweight on the depth of fat and muscle measured at the C-site of progeny from Merino ewes, is unlikely to be of any commercial significance within the range of nutritional scenarios during pregnancy and lactation that are likely to be experienced within the Australian sheep industry.

The demand in the world market for high quality fine and superfine wool is increasing, responding to greater wool textile industry demands and price incentives. There are few research studies focused on hair follicle population in fine and superfine Merino sheep in Uruguay. This study evaluates the association between wool follicle population and the most relevant economical wool traits. This work was carried out on Ultrafine Australian Merino (UAM) and Dohne Merino (DM) animals (lambs and rams). Phenotypic correlations between the secondary/primary follicles (S/P) and other wool traits (fiber diameter, staple length, clean fleece weight, scoured yield, coefficient of variation of fiber diameter and comfort factor parameter, were performed using histological skin cuts. Two skin samples were extracted from each lamb at two and nine months of age. These were processed at the skin histology laboratory to obtain a histological preparation for further biopsy, being able to differentiate the structures obtained to be evaluated. The results showed in the UAM group, S/P ratio in nine-month-old lambs was greater than two-month-old lambs (31.08 vs 23.12 P< 0.05). For DM group, there were no differences in the S/P ratio between age groups. Phenotypic correlations for wool characteristics obtained were of low to medium or null magnitude in both genotypes.

Ewes at two sites were fed to be either condition score 2 or 3 by Day 90 of pregnancy and then grazed on various levels of feed-on-offer (FOO) from Day 90 of pregnancy until weaning, to investigate whether nutrition of Merino ewes during pregnancy and lactation affected the muscle, fat and immunity to worms of their progeny. Eye muscle and fat depth at the C-site, and faecal worm egg counts (FWECs) of the progeny were measured between 7 - 27 months of age. Ewe condition score at day 90 of pregnancy did not impact largely on the eye muscle depth, fat depth or FWEC of the progeny. Increasing FOO available to ewes during the last 60 days of pregnancy and throughout lactation increased the eye muscle depth of progeny at one site and decreased the fat depth of progeny at the other site (P<0.05), but did not affect the majority of FWECs of the progeny at either site. The FWECs of the progeny were low, indicating a relatively low larval challenge, which limits the likelihood of differences in immunity to worms between the progeny being expressed. This Masters demonstrated that levels of nutrition available to Merino ewes typical of commercial grazing conditions had only small effects on the eye muscle and fat depth at the C-site, and on the faecal worm egg counts of their progeny

Abstract:
Muscular dystrophies (MDs) are neuromuscular disorders characterised by chronic, usually progressive, skeletal muscle weakness. Individuals often lose walking ability and can suffer terminal cardiorespiratory complications. Determining the genetics of a disease helps provide diagnosis, prognosis, genetic counselling, and the basis for rational therapeutic design. A naturally occurring sheep model of autosomal recessive congenital MD was identified in WA in the 1950’s and preserved as a research colony. The pathological features and distribution of this MD is novel. A sheep model of MD is incredibly valuable; sheep have similar skeletal muscle mass to humans, representing a significant improvement over smaller mammals in which to trial therapies. Successfully characterising the causative gene(s) would enable a possible target for new therapies and may open new lines of investigation into better understanding and treatment of MD in humans. This project utilised a two-pronged approach to investigate the genetics of this ovine MD. First, bioinformatics analysis of SNP genotyping for multiple individuals in the flock by a 50,000 SNP array in combination with the latest sheep genome reference build released by the International Sheep Genome Consortium, enabling homozygosity mapping, genetic linkage and association mapping. Second, molecular biological approaches further explored the identified prime candidate gene by cDNA sequencing. This research project identified ROCK2 as the prime candidate gene most likely harbouring a mutation causing the muscular dystrophy in this internationally unique ovine model. It also demonstrated for the first time in sheep the existence of ROCK2m, an isoform of ROCK2 preferentially expressed in skeletal muscle. This work has set the stage for further investigations into ROCK2m and the ovine MD which will hopefully pinpoint the causative disease mutation.

Abstract:
Succeeding phases of British economic growth prompted strikingly different imperatives for expansion, for natural resource exploitation, and for the social organization of extra-European production. In the eighteenth century, sugar, African slaves, and shipping in the Atlantic world provided one major dynamic of empire. But in the nineteenth century, antipodean settlement and trade, especially that resulting from expanding settler pastoral frontiers, was responsible for some of the most dramatic social and environmental transformations. Plantations occupied relatively little space in the new social geography of world production. By contrast, commercial pastoralism, which took root most energetically in the temperate and semi-arid regions of the newly conquered world, was land-hungry but relatively light in its demands for labour. The Spanish Empire based in Mexico can be considered a forerunner. By the 1580s, within fifty years of their introduction, there were an estimated 4.5 million merino sheep in the Mexican highlands. The livestock economy, incorporating cattle as well as sheep, spread northwards through Mexico to what became California by the eighteenth century. Settler intrusions followed in the vast landmasses of southern Latin America, southern Africa, Australia, and New Zealand. Australia was one of the last-invaded of these territories, and, in respect of the issues that we are exploring, was in some senses distinctive. Unlike Canada and South Africa, there was no long, slow period of trade and interaction with the indigenous population; like the Caribbean, the Aboriginal people were quickly displaced by disease and conquest. The relative scale of the pastoral economy was greater than in any other British colony. Supply of meat and dairy products to rapidly growing ports and urban centres was one priority for livestock farmers. Cattle ranching remained a major feature of livestock production in Australia. Bullock-carts, not dissimilar to South African ox-wagons, were essential for Australian transport up to the 1870s. But for well over a century, from the 1820s to the 1950s and beyond, sheep flooded the southern lands. Although mutton became a significant export from New Zealand and South America, wool was probably the major product of these pastoral hinterlands—and a key focus of production in Australia and South Africa. The growth in antipodean sheep numbers was staggering.

Abstract:
In September 2006 an outbreak of 'Bluetongue like' disease struck the cattle herds in Israel. Over 100 dairy and beef cattle herds were affected. Epizootic hemorrhagic disease virus (EHDV) (an Orbivirusclosely related to bluetongue virus (BTV)), was isolated from samples collected from several herds during the outbreaks. Following are the aims of the study and summary of the results: which up until now were published in 6 articles in peer-reviewed journals. Three more articles are still under preparation: 1. To identify the origin of the virus: The virus identified was fully sequenced and compared with the sequences available in the GenBank. It appeared that while gene segment L2 was clustered with EHDV-7 isolated in Australia, most of the other segments were clustered with EHDV-6 isolates from South-Africa and Bahrain. This may suggest that the strain which affected Israel on 2006 may have been related to similar outbreaks which occurred in north-Africa at the same year and could also be a result of reassortment with an Australian strain (Wilson et al. article in preparation). Analysis of the serological results from Israel demonstrated that cows and calves were similarly positive as opposed to BTV for which seropositivity in cows was significantly higher than in calves. This finding also supports the hypothesis that the 2006 EHD outbreak in Israel was an incursive event and the virus was not present in Israel before this outbreak (Kedmi et al. Veterinary Journal, 2011) 2. To identify the vectors of this virus: In the US, Culicoides sonorensis was found as an efficient vector of EHDV as the virus was transmitted by midges fed on infected white tailed deer (WTD; Odocoileusvirginianus) to susceptible WTD (Ruder et al. Parasites and Vectors, 2012). We also examined the effect of temperature on replication of EHDV-7 in C. sonorensis and demonstrated that the time to detection of potentially competent midges decreased with increasing temperature (Ruder et al. in preparation). Although multiple attempts were made, we failed to evaluate wild-caught Culicoidesinsignisas a potential vector for EHDV-7; however, our finding that C. sonorensis is a competent vector is far more significant because this species is widespread in the U.S. As for Israeli Culicoides spp. the main species caught near farms affected during the outbreaks were C. imicolaand C. oxystoma. The vector competence studies performed in Israel were in a smaller scale than in the US due to lack of a laboratory colony of these species and due to lack of facilities to infect animals with vector borne diseases. However, we found both species to be susceptible for infection by EHDV. For C. oxystoma, 1/3 of the Culicoidesinfected were positive 11 days post feeding. 3. To identify the host and environmental factors influencing the level of exposure to EHDV, its spread and its associated morbidity: Analysis of the cattle morbidity in Israel showed that the disease resulted in an average loss of over 200 kg milk per cow in herds affected during September 2006 and 1.42% excess mortality in heavily infected herds (Kedmi et al. Journal of Dairy Science, 2010). Outbreak investigation showed that winds played a significant role in virus spread during the 2006 outbreak (Kedmi et al. Preventive Veterinary Medicine, 2010). Further studies showed that both sheep (Kedmi et al. Veterinary Microbiology, 2011) and wild ruminants did not play a significant role in virus spread in Israel (Kedmi et al. article in preparation). Clinical studies in WTD showed that this species is highly susceptibile to EHDV-7 infection and disease (Ruder et al. Journal of Wildlife Diseases, 2012). Experimental infection of Holstein cattle (cows and calves) yielded subclinical viremia (Ruder et al. in preparation). The findings of this study, which resulted in 6 articles, published in peer reviewed journals and 4 more articles which are in preparation, contributed to the dairy industry in Israel by defining the main factors associated with disease spread and assessment of disease impact. In the US, we demonstrated that sufficient conditions exist for potential virus establishment if EHDV-7 were introduced. The significant knowledge gained through this study will enable better decision making regarding prevention and control measures for EHDV and similar viruses, such as BTV.
