Abstract:
ABSTRACT The hyperthermophilic bacterium Thermotoga maritimaMSB8 possesses a reverse gyrase whose enzymatic properties are very similar to those of archaeal reverse gyrases. It catalyzes the positive supercoiling of the DNA in an Mg2+- and ATP-dependent process. Its optimal temperature of activity is around 90°C, and it is highly thermostable. We have cloned and DNA sequenced the corresponding gene (T. maritima topR). This is the first report describing the analysis of a gene encoding a reverse gyrase in bacteria. The T. maritima topR gene codes for a protein of 1,104 amino acids with a deduced molecular weight of 128,259, a value in agreement with that estimated from the denaturing gel electrophoresis of the purified enzyme. Like its archaeal homologs, theT. maritima reverse gyrase exhibits helicase and topoisomerase domains, and its sequence matches very well the consensus sequence for six reverse gyrases now available. Phylogenetic analysis shows that all reverse gyrases, including the T. maritimaenzyme, form a very homogeneous group, distinct from the type I 5′ topoisomerases of the TopA subfamily, for which we have previously isolated a representative gene in T. maritima(topA). The coexistence of these two distinct genes, coding for a reverse gyrase and an ω-like topoisomerase, respectively, together with the recent description of a gyrase in T. maritima (O. Guipaud, E. Marguet, K. M. Noll, C. Bouthier de la Tour, and P. Forterre, Proc. Natl. Acad. Sci. USA 94:10606–10611, 1977) addresses the question of the control of the supercoiling in this organism.

Abstract:
The main objective of this study was to analyse the relationship between players´ non-verbal body language and their performance in sport. The sample consists of a total of 477 actions from 40 players competing in the ATP Tour. All actions were taken from tie-breaks in official tournaments. The categories that were analyzed were previous performance, dominant non-verbal body language, submissive non-verbal body language, and performance later on. Results show how the players´ non verbal body language impacts on their performance later on. Thus, we can conclude that following dominant non-verbal body language, the probabilities of better performance in the next point are greater than following submissive non-verbal body language. Therefore, it is important to train these aspects as an extra tool to improve players´ performance.

Abstract:
To assess ball performance for research and development purposes requires greater understanding of the impact conditions a tennis ball experiences in professional tournament play. Ball tracking information taken from three consecutive years of an ATP 250 tour event played on hard court was analysed. The frequency of first serves, second serves, racket impacts and surface impacts was assessed per game and extrapolated to show how many impacts a single ball is subjected to. Where applicable the pre- and post-impact velocities and angles were found, and the distribution of each was analysed. In total, data from 65 matches comprising 1505 games were analysed. On average, each game contained 70.26 (±16.23) impacts, of which 9.23%, 3.16%, 37.78% and 49.83% were first serves, second serves, racket impacts and surface impacts, respectively. As a result, assuming all balls in play are used evenly, a single ball is expected to be subjected to 105 (±24) impacts over the course of the nine games that it is in play. The results of the investigation could be used to design a wear protocol capable of artificially wearing tennis balls in a way that is representative of professional play.

AbstractObjective:To examine the effectiveness of the Produce for Better Health Foundation grocery store tour programme as a nutrition education tool for changing consumers’ intention to consume various forms of fruits and vegetables (F&V).Design:Cross-sectional study.Setting:Ten grocery stores in a city in the Southeast USA.Participants:A total of 147 grocery shoppers in Alabama, who participated in a grocery store tour, completed a retrospective pre-/post-survey using the Theory of Planned Behaviour. Results:Results from independent-samples t tests indicated that mean values of attitude, subjective norm and perceived control were significantly increased after store tours (P < 0·01). Participants’ intentions to consume various types of F&V (fresh, dried, canned, juice and frozen), especially dried F&V, increased significantly after store tours. Results from structural equation modelling indicated that before store tours attitude (γ = 0·48, P < 0·01) was the most significant predictor of intention to consume F&V, followed by perceived behavioural control (γ = 0·24, P = 0·02) and subjective norm (γ = 0·21, P = 0·03). After store tours, attitude (γ = 0·51, P < 0·01) and perceived behavioural control (γ = 0·44, P < 0·01) were still strong predictors of intention to consume F&V, while subjective norm became an insignificant predictor of intention. Conclusions:Findings revealed positive changes in study participants’ attitude, subjective norm, perceived behavioural control and their intentions to consume various forms of F&V, which suggest potential benefits of providing grocery store tours as a tool to promote consumers’ F&V intake.

Abstract:
Research background: The rise of innovative (alternative) guided tours around the world and especially in Europe embody the idea of ‘glocalization’, since these tours and associated new business models (comprising both ‘free’ and paid offers) represent a global phenomenon that utilizes ‘localness’ as a major asset. Therefore, glocalization strategies of the tour providing companies are worth studying and so are the current transformations in their business models. The Bulgarian capital city of Sofia is an appropriate case study area, being recently denoted as an emerging tourist destination on the international scene, with a number of innovative city tours developed over the last years that form a particular aspect of Sofia’s urban experience. Purpose of the article: Building upon a previous study, the paper seeks to uncover glocalization strategies applied and current transformations observed in the innovative city tours of Sofia in terms of COVID-19 induced market change, product development, and delivery system adjustment. Methods: Subjected to analysis are five tour providers selected on the ground of their business models, tour content and market success. The study involves in-depth interviews with company managers and tour guides, personal observations, and information retrieved from social media and websites. Findings & Value added: The study results contribute to better understanding of how global trends are implemented in a local context and reveal the challenges triggered by the current CIVID-19 induced shift of scales from global to local in terms of markets, product development, and service organization.

Abstract:
Abstract The objective of the virtual Operation Main Street (OMS) program was to provide a live, interactive experience to educate audiences anywhere in the world about modern pork production. National Pork Board’s OMS began in 2004 to train pork producers to share their stories to improve the image of pig farming. Since its inception, they have trained >1,300 producers influencing >240,000 people However, there are locations that don’t have access to OMS speakers, and because presenters use slides, it doesn’t provide a real-time experience of being in the barn. Therefore, the virtual OMS was developed through a collaborative effort between the National Pork Board, Eidson & Partners, and South Dakota State University (SDSU). After a formal interview process, students participate in a 2-day OMS training. Students are trained to cover a set of defined speaking points, but discuss them in their own words. Each live tour is conducted in the SDSU Swine Unit, and is done entirely by the student on her/his own cellphone. Tours last approximately 10 minutes, with the audience asking questions at any time through a moderator at the venue. The first virtual tour was given on December 7, 2017, and since then there have been a total of 133 tours impacting 24,558 people. Audiences include Veterinary Colleges, high schools, the American Association of Critical Care Nurses, and the Association of Nutrition and Foodservice Professionals. In the last year, 51 tours were given to 1,678 people in 22 different states. Respondents to a Google survey offered at the end of every virtual tour stated the presentation and tour resulted in a >60% increase in a positive opinion of the pork industry. Virtual OMS is an impactful method to dispel myths about modern pig farming, and creates a personal connection between the faces of pork production and consumers.

Abstract:
This study constructs a new travel risks’ evaluation model for freelancers to evaluate and select tour groups by considering the interdependencies of the evaluation criteria used. First of all, the proposed model adopts the Nominal Group Technique (NGT) to identify suitable evaluation criteria for evaluating travel risks. Six evaluation criteria and 18 subcriteria are obtained. The six evaluation criteria are financial risk, transportation risk, social risk, hygiene risk, sightseeing spot risk, and general risk for freelancer tour groups. Secondly, the model uses the analytic network process (ANP) to determine the relative weight of the criteria. Finally, examples of group package tours (GPTs) are used to demonstrate the travel risk evaluation process for this model. The results show that the Tokyo GPT is the best group tour. The proposed model helps freelancers to effectively evaluate travel risks and decision-making, making it highly applicable to academia and tour groups.

Abstract:
Purpose:Difficulties in preserving physical capacities while on tennis tours necessitate targeted training prescription. This study analyzed training and match loads performed before and on tour for their relationship with posttour physical-capacity changes. A secondary aim was to determine whether the presence of a strength and conditioning (S&C) coach affected the type and volume of on-tour training load. Methods:The training and match loads of 30 high-performance junior tennis players were recorded over 8 wk: 4 wk before and 4 wk during an international tour. Fitness tests were conducted pretour and posttour, including double and single-leg (dominant and nondominant) countermovement jump, speed (5, 10, and 20 m), modified 5-0-5 agility, 10 × 20-m repeated-sprint ability, and multistage fitness tests. Tour training and match loads were categorized according to whether S&C support was present or absent. Results:Total and tennis training loads were significantly greater on tour than pretour (P ≤ .05, d > 0.8). Increases in on-tour, on-court training loads were moderately correlated with decrements in speed and aerobic power (r = .31-.52). Finally, S&C presence on tour significantly increased total, on-court, and off-court training load completed (P ≤ .05, d > 0.8).Conclusions:Training loads should be carefully prescribed to ensure that sufficient total and tennis loads are completed pretour. Specifically, speed and aerobic capacities may regress with increased training on tour. Finally, a practical observation was that on-tour S&C support resulted in increased S&C training load (around match loads), potentially countering the observed regression of physical capacities. Such a finding has the capacity to alter current physical-preparation structures in high-performance tennis environments with finite resources.

Abstract:
The article describes the main aspects of the development of photo-tourism in the Yamal-Nenets Autonomous District. It is shown that this type of tourism is new, so it does not appear in any of existing regulations. The authors have attempted to give a definition of this type of tourism; show its main consumers. Presented are the factors that affect the emergence and development of photo-tourism. As part of this work the classification of photo tours is suggested, which provides information on the basic criteria required for forming such tours. These criteria include: the age of tourists, comfort level, duration of the tour, the form of the tour, as well as the objects for making photos. The optimal number of participants in the photo tour is determined as 1-3 people for individual tours and 3-8 for group tours). The authors prove the possibility of the development of photo-tourism in the Yamal-Nenets Autonomous District. To do this, an analysis of the main indicators of tourism development in the region is conducted and identified are tourist resources necessary for the development of photo-tourism. The main problems hindering the development of photo-tourism in the Yamal-Nenets Autonomous District, and their solutions are also suggested. In conclusion, it is concluded that the photo-tourism, of course, is not a mass form of tourism, so tourist traffic statistics are not kept, since the number of arrivals with objectives of photo-tourism is insignificant. Currently, however, for most tourists mass tourism programs and direction are becoming unpopular, and in this regard popularization of unusual and unfamiliar forms of recreation, including photo-tourism, is increasing. Thanks to advances in technologies, promotion of social networks and advances in Internet photo art is now considered "fashionable". All this must surely affect the development of photo-tourism, in particular in the Yamal-Nenets Autonomous District.

Abstract:
Article is devoted to building the technological route maps on the events of the Crimean Conference of 1945. In this context, the blocks can serve as a framework of thematic tours on the history of espionage, diplomacy and is an introduction in the classic routes, "Magic South Coast of the Crimea", "Steppe. Mountains. Sea", "Three palaces", "To the rocks of Ai-Petri" etc. Particular attention is paid to those aspects of the Crimea Conference, which is almost not used in the current tour routes. This is the airport near the city ofSaki, who was the "Gate of the Crimea Conference", the rest house of Winston Churchill in Simferopol. Many classical objects, it is proposed to refocus on the events in February 1945. For example a "normal" natural history tour of the park Vorontsov Palace the author transforms in the "Local History" route. The latter focuses on the issues of the safety, the residence of British Prime Minister Winston Churchill (Vorontsov Palace) and oddities occurring in the park during the Crimean conference. Article provided with a detailed technical route guidance, which includes items such as: route, time, objects display, illuminated questions, route indication stops, guidelines, organizational instructions. In general terms the author describes the course of the conference and its important, local history items. The article will be useful not only for professional tour guides, tour leaders of organizations, but also designed for lovers who during their visits to the Crimea want to independently examine a number of lesser-known sites devoted to a major diplomatic summit of the anti-Hitler coalition in the Second World War.

Abstract:
Summary The increasing number of travelers to Bali has been accompanied by the increased risks of travel-related health problems. Travel agents and tour guides are in a key position to inform travelers about these matters; nonetheless, little evidence was found on their role in the promotion of travel health. This article aims to assess travel agent and tour guide perceptions on their possible involvement in promoting travel health measures in Bali. A cross-sectional study was conducted between July and October 2015, involving 500 respondents (250 travel agents and 250 tour guides), who were chosen systematically from the Bali-based members of the Association of the Indonesian Tours and Travel Agencies. Data were collected using questionnaires, and perception indicators were developed using concepts from social learning theory and health belief model. Both groups have good levels of knowledge of general travel health issues, with mean percentages (SD) of 67.1 (11.9) and 66.2 (10.0) for travel agents and tour guides, respectively. Tour guides were more likely than travel agents to deliver information on health risks [PR (95%CI) = 1.31 (1.10–1.56); p = 0.003]; and on safety hazards during travel [PR (95%CI) = 1.22 (1.01–1.46), p = 0.04]. Overall, the majority of respondents (>68%) have positive and very positive perceptions of possible involvement in travel health and safety programs, although tour guides have statistically better perceptions (z = −2.2, p = 0.03). Based on perception levels, previous experiences and levels of knowledge, travel agents and tour guides have great potential to be involved in travel health promotion programs in Bali.

Full text
Abstract:
The term ‘hospitality’ has long been linked to accommodation providers and eateries, but the wider range of hospitality, and hospitable experiences, has received little attention from academics and practitioners alike. Although tourism and hospitality are undeniably linked, Lynch [1] notes that “in tourism, there is a curious neglect of welcome given its associations with the idea of universal hospitality” (p. 174). To this end, we argue that hospitality goes well beyond food and accommodation alone, and includes any host-tourist interactions, such as those between guides and tour participants. Participants aboard nature-based marine tours are expecting to learn [2]. However, the guide's ability to 'connect' with their audience and provide quality learning experiences is not part of an industry standard. As a result, the expertise and personality of guides varies, as do their duties and their levels of training. Learning is a critical component of nature-based tours, thus placing significant responsibility on the guide in the creation of a hospitable experience. For nature-based wildlife tour operators in New Zealand (and globally), in many cases guide training efforts are a choice left up to the operators. While individual operators' needs may vary, previous hospitality research suggests that beyond fulfilling visitor expectations, appropriate training not only benefits guests, but also increases workplace satisfaction [3]. This paper discusses the need for guide training in increasing the hospitable experience aboard nature-based wildlife tours. Through data captured in self-administered visitor surveys, we explored over 400 participants' perceptions of guide abilities related to a hospitable, or pleasant, experience [4]. Tour participants on swim-with wild dolphins tours were asked to rate the importance of items related to their on-tour experience. All items were rated on a five-point scale. Over 93% of the respondents thought that having a knowledgeable and helpful boat crew was important, and more specifically over 90% felt having a knowledgeable guide on board was important. Likewise, the majority of participants identified the importance of learning about various subjects, such as the natural environment (69%) and threats to marine life (66%) – a responsibility that falls to the guides and crew. Being able to understand the safety rules was of near comparable importance to the learning experience desires (86%), as was the cleanliness of the vessel (85%). Our findings demonstrated that in order to create a hospitable experience, marine tour guides must fulfil dual roles as crew and naturalists. The high values placed on educational and safety information emphasise the critical role of knowledgeable guides/crew aboard nature-based tours. While it is apparent that many guides take pride in their work, with many furthering their knowledge during their personal time, the findings from our study highlight the importance of the guide's role in the overall visitor experience. While some tour components, such as the safety messages, are matters of legal compliance (and clearly were of high importance to participants as well), other components such as fulfilling the educational desires of visitors should be carefully considered by operators. Our findings show that visitors place a high importance on tour components that are directly related to a guide's performance, personality and abilities. Options for operators include requiring a minimum qualification for guides (e.g., a bachelor's degree in a related field) and/or an investment in continuing and specialised education. In conclusion, guide training on the natural environment and conservation as well as vessel/marine safety is essential. More specifically, this means that operators stand to benefit from guide training that reaches beyond the factual information and includes interpretation techniques. This may seem like a large investment for operators; however, the findings from this research indicate that visitor satisfaction is dependent upon this investment. The role of guides aboard nature-based wildlife tours is central to the overall hospitable experience. Abstract:
In general, there exists numerous attractions installed in a theme park, and tourists in a theme park dynamically change their locations during a tour. Thus, a tourist may cope with the issues of selecting the attractions to visit while planning the tour route. This paper, based on the concept of location awareness, proposes a novel waiting time, called the personalized waiting time, to introduce a location-aware recommendation strategy. In addition, this paper presents an architecture of tourist service system using the proposed recommendation strategy to relieve the pressure on tourists and create the pleasant experience in their tours. The proposed location-based system consists of mobile app, ticket-reader, detecting/counting, and central subsystems, and the whole system was implemented in this study. We conducted numerous experiments and field testing results validated that the entire proposed system can correctly provide information, such as attraction introduction, recommended session time, estimated moving and waiting time, tour map, and the number of reservations. The system functions, including dynamical scheduling, attraction reservation, ticket verification, visitor detection, and visitor counting, also worked well.

Abstract:
Studies of intact hearts suggest that cardiac myocytes may have the ability to reversibly suppress metabolic activity and energy demand in states of regional hypoperfusion. However, an ability to suppress respiration in response to hypoxia has never been demonstrated in isolated myocytes. To test this, isolated embryonic chick cardiac myocytes were exposed to progressive hypoxia while their rate of O2 uptake and concentrations of lactate, ATP, ADP, AMP, and phosphocreatine were measured. Compared with the value obtained at an oxygen tension (PO2) of 120 Torr, cellular O2 uptake decreased by 28 +/- 14% (SD) at PO2 = 50 Torr and by 64 +/- 25% at PO2 = 20 Torr (P < 0.05). This decrease was similar after 1 min or 2 h of hypoxia, was sustained for 16 h, and was completely reversible within 2 min after reoxygenation. The reduction in O2 uptake was associated with a decrease in the rate of ATP turnover, but no change in adenine nucleotide or phosphocreatine concentrations. In myocytes adherent to glass cover-slips, O2 uptake and contractile motion were decreased after 30-60 min at 50 and 20 Torr, compared with normoxic values. O2 uptake also was significantly decreased at 50 and 20 Torr in myocytes incubated with N,N,N',N'-tetramethyl-p-phenylenediamine, which suggests that the catalytic activity of cytochrome-c oxidase was partially inhibited during hypoxia. In summary, these results demonstrate that embryonic chick cardiac myocytes can suppress their rates of ATP demand, ATP utilization, and O2 uptake during moderate hypoxia through a mechanism that involves a reversible inhibition of cytochrome-c oxidase. This mechanism may represent a protective response to cellular hypoxia.

Abstract:
The response of endothelial energy metabolism to oxygen supply was studied in cultured coronary endothelial cells from the rat at defined PO2 levels between 0.1 and 100 Torr. In the presence of glucose (5 mM), endothelial respiration (4 nmol O2.min-1.mg protein-1) was independent of the exterior PO2 greater than 3 Torr; oxygen consumption was half maximal at 0.8 Torr. At 100 Torr, lactate production was 26 nmol.min-1.mg protein-1; the decrease of the PO2 to 0.1 Torr resulted in a 2.2-fold increase in lactate production. The contents of ATP, ADP, and AMP were 21, 4, and 2 nmol/mg protein, respectively; they remained constant for 2.5-h incubations at PO2 levels between 0.1 and 100 Torr. In the presence of palmitate (100 microM) plus glutamine (0.5 mM), oxygen consumption was 8 nmol.min-1.mg protein-1 at PO2 levels greater than 3 Torr, and the half-maximal rate was again observed at 0.8 Torr. Lactate production was negligible. At PO2 levels greater than 3 Torr, the cells remained well energized. Below 3 Torr, however, the adenine nucleotide contents rapidly declined. These results demonstrate that the oxygen demand of coronary endothelial cells is low compared with the beating myocardium. In the presence of glucose, aerobic glycolysis is pronounced and the Pasteur effect small. In severe hypoxia (PO2 less than 0.1 Torr) the energetic state remained stable. In the absence of glucose, the energetic state of coronary endothelial cells is sensitive to the exterior PO2 less than 3 Torr, declining concomitantly with the decrease in respiration.

Abstract:
Abstract Introduction Jet lag can significantly impact an athlete’s health and performance. However, the effect of ultra long-haul travel (> 12h flying time) westward across nine time zones on the sleep of female collegiate athletes is limited. We therefore studied the sleep behavior of NCAA Division I female volleyball players on an exhibition tour to China. Methods For this observational study, eleven females were recruited from a NCAA Division I Volleyball team. During the Tour, sleep behavior was monitored using wrist actigraphy (Actiwatch Spectrum, Phillips) before (5 days) outbound travel (PRE-FLIGHT), during air travel to (1) & from (2) China (FLIGHT-DAY 1 & 2) and during the athletes’ tour while in China (TOUR-DAY 1–8). Athletes were instructed to wear the actigraphs at all times, except during competition. Standard parameters were collected from the actigraph including sleep durations and sleep efficiency and expressed as mean ± sd. Results Sleep duration and sleep efficiency were similar for PRE-FLIGHT days (~8.3 hrs ± 1.5, 90.6 ± 3.2%). Sleep duration and efficiency were significantly lower on FLIGHT-DAY 1 & 2 compared to TOUR-DAY and PRE-FLIGHT, but were not significantly different from each other (~5.2 ± 2.4h, 80.5 ± 8.8%). Sleep duration during TOUR-DAY 1–7 followed a quadratic relationship, peaking on TOUR-DAY 1 and reaching a nadir on TOUR-DAY 5, before increasing again through TOUR-DAY 7 (p<0.05). Sleep duration on TOUR-DAY 8 (~5.9 ± 0.9 h) was significantly lower than PRE-FLIGHT and TOUR-DAY 1,2,6 & 7. Conclusion During travel female collegiate volleyball players showed sleep disturbance. Implementation of sleep interventions for jet lag are warranted for athletes traveling across multiple time zones. Support PAC-12 Student-Athlete Health and Wellbeing Initiative, Grant #1554240

Abstract:
Traveling around a city and making transit in certain areas is called a city tour. Furthermore, determining the optimal city tour route can be considered as a traveling salesman problem. There are many kinds of algorithms to solve this, one of which is the Genetic Algorithm (GA). In developing the City Tour application, a platform is needed to be taken to various places anywhere and anytime. Finally, we developed an application that runs on mobile devices. This application is built on the Android platform so that its use can be more efficient. Furthermore, it can be concluded that the GA applied to the Android-based City Tour Application is reliable to determine city tour routes; this is evidenced by comparing GA with the brute force method, where GA provides optimum results with less running time.

Abstract:
The object of this research is the plot of D. D. Shostakovich’s ballet “The Limpid Stream”. The subject is the interpretation of this plot in the versions of F. G Lopukhov (1935) and A. O. Ratmansky (2003), as well as peer review on these spectacles. The goal of this work consists in determination of the crucial for the concepts of ballet masters differences of libretto (as a literary foundation of the plot) in the three versions of the ballet, and comparison of perception of the plot in the year of its first staging and at the present. The considered problematic required application of historical approach – attraction of the materials and articles for the period of 1935-1936. The historiographical analysis allowed translating and examining one of the most recent peer reviews on the spectacle – the English-language reviews on the “Limpid Stream” of Ratmansky, presented on the London tour of Bolshoi Theatre in August 2019. The article also utilizes practical experience of author’s work with Ratmansky and participation in the aforementioned tours (performing the role of Zina). The main tool for solution of the set problem became the comparative analysis of the varieties of libretto (authors – Lopukhov and Piotrovsky) of the three versions of ballet “The Limpid Stream”. It is concluded that the first versions of ballet were popular among the public, but aroused negative or ambiguous feedback, which led to the removal of spectacle from the repertoire. The last version is regularly performed in the repertoire of Bolshoi Theatre, including on the tour, being well regarded by the public and sophisticated British critics.

Abstract:
The relationship between biochemical and physiological responses and tissue O2 during hypoxia was investigated in vivo in the dog brain by 31P nuclear magnetic resonance (NMR) spectroscopy. Our findings demonstrate how ATP synthesis in the brain can be maintained during hypoxia because of compensatory changes in NADH, ADP, and Pi. Eleven beagle dogs were anesthetized and mechanically ventilated, and a steady-state graded hypoxia was induced by decreasing the fraction of inspired O2 (FIO2) stepwise at 20-min intervals. Biochemical metabolites were measured using 31P-NMR and fluorescence spectroscopy. When sagittal sinus O2 partial pressure (PVO2) had decreased to 15 Torr, NADH increased by 30%, Pi increased by 50%, and phosphocreatine (PCr) decreased by 20%. In contrast, ATP remained constant. There was a 10% increase in ADP in dogs that maintained a steady temperature, but ADP decreased by as much as 30% in dogs in which body temperature decreased with the falling PVO2. PCr/Pi was logarithmically related to the phosphorylation potential during steady-state hypoxia. Compensation for the O2 lack is attributed to increases in ADP, Pi, and NADH as a result of the reciprocal relationship of the Michaelis-Menten equation. If the Michaelis-Menten constants (Km) of ADP, Pi, and O2 are the same as determined in vitro in mitochondria, the minimum brain cytosolic O2 capable of maintaining a steady-state ATP is near its Km (0.1 Torr) at a PVO2 of 7.5 Torr. At this critical O2 level, PCr/Pi is 0.9, intracellular pH is 6.75, phosphorylation potential is 38.5 mM-1, and the calculated maximum velocity of ATP formation by oxidative phosphorylation is 55% of normal.

Abstract:
One of the best known economic theories of the early 21st c. is Pine & Gilmore’s of the experience economy (1999). This is nothing new for the tourism industry which has always been selling emotions, dreams and memories involving travel. Recently, however, it has become much more important to provide professionally (consciously and purposefully) prepared tourism products, strongly marked with emotion1. Efforts to create original experiences for tourists include not only various modifications of traditional tourism packages, but also a search for new recreation spaces and new forms of tourism. The aim of this article is to review new tourism-recreational areas (e.g. military areas, new churches, so-called ‘destination centres’, along with ordinary and extreme experience spaces), as well as new forms of travel and recreation (e.g. creative, event, sports, culinary or extreme tourism). The analysis includes those phenomena which above all are currently gaining in popularity as part of the tourist experience triad (WŁODARCZYK 2013).

Abstract:
The paper aims at introducing an advanced delivery tour planner to support operators in urban delivery operations through a combined approach which chooses delivery bays and delivery time windows while optimizing the delivery routes. After a literature review on tools for the management and the control of the delivery system implemented for optimizing the usage of on-street delivery bays, a prototypical tour delivery planner is described. The tool allows transport and logistics operators to book the delivery bays and to have real-time suggestions on the delivery tour to follow, through the minimization of the total delivery time. Currently, at development phase, the tool has been tested in a target zone, considering the road network and time/city delivering constraints and real-time data about vehicles location, traffic and delivery bay availability. The tool identifies the possible tours based on the delivery preferences, ranks the possible solutions according to the total route time based on information on the road network (i.e. travel time forecasts), performs a further optimization to reduce the total travel times and presents the user the best alternative along with the indications of which delivery bay to use in each delivery stop. The developed prototype is composed by two main parts: a web application that manages communication between the database and the road network simulation, and, an Android mobile App that supports transport and logistic operators in managing their delivering, pre trip and en route, showing and updating routing based on real-time information.

Abstract:
The shortest path problem is one of the optimization problems where the optimization value is a distance. In general, solving the problem of the shortest route search can be done using two methods, namely conventional methods and heuristic methods. The Ant Colony Optimization (ACO) is the one of the optimization algorithm based on heuristic method. ACO is adopted from the behavior of ant colonies which naturally able to find the shortest route on the way from the nest to the food sources. In this study, ACO is used to determine the shortest route from Bumi Senyiur Hotel (origin point) to East Kalimantan Governor's Office (destination point). The selection of the origin and destination points is based on a large number of possible major roads connecting the two points. The data source used is the base map of Samarinda City which is cropped on certain coordinates by using Google Earth app which covers the origin and destination points selected. The data pre-processing is performed on the base map image of the acquisition results to obtain its numerical data. ACO is implemented on the data to obtain the shortest path from the origin and destination point that has been determined. From the study results obtained that the number of ants that have been used has an effect on the increase of possible solutions to optimal. The number of tours effect on the number of pheromones that are left on each edge passed ant. With the global pheromone update on each tour then there is a possibility that the path that has passed the ant will run out of pheromone at the end of the tour. This causes the possibility of inconsistent results when using the number of ants smaller than the number of tours.

Purpose The purpose of this paper is to examine the factors that influence mainland Chinese film tourists’ behavioural intention in accepting an augmented reality (AR) feature film character-related tour APP (an IfilmAR-tour-APP) based on an extension of unified theory of acceptance and use of technology (UTAUT) model. Design/methodology/approach Systematic sampling was used to collect data; 360 valid questionnaires were collected from mainland Chinese film tourists visiting Macau. Partial least squares structural equation modelling (PLS-SEM) was applied to analyse the data. Findings The findings indicated that performance expectancy, effort expectancy, social influence, celebrity involvement and personal innovativeness have a significant impact on mainland Chinese film tourists’ behavioural intention in accepting the IfilmAR-tour-APP, while facilitating conditions do not significantly affect tourists’ behavioural intention. Furthermore, the study revealed that effort expectancy, celebrity involvement and personal innovativeness positively influence performance expectancy in the use of an IfilmAR-tour-APP. Practical implications The findings of this study provide implications for AR tour APP developers which will help them to enhance their designs, and for the governments of tourist destinations to formulate their promotion strategies in developing niche tourism. Originality/value This study extended the application of UTAUT to tourism technology by introducing two personality traits in the context of AR tour APPs for film-induced tourism. This research contributes to niche tourism research by linking niche tourism and tourism technology.
