Resumo: Em termos quantitativos, a população universitária e a membresia das Assembleias de Deus são parecidas. Em 1991, os universitários eram 3.928.260 e os assembleianos 2.439.770. Em 2010, o número de universitários subiu para 12.679.010 e o de assembleianos para 12.314.410. Cresceu o numero de universitários e também o de assembleianos, inclusive de assembleianos universitários e de pastores. Quem são esses pastores assembleianos com nível superior e o que eles pensam? Foram enviados mais de mil emails para pessoas que integravam listagens de convenções, ministérios e igrejas, e também para amigos indicados por essas pessoas. Preenchidos e devolvidos, somaram 84 questionários. A primeira parte eram questões pessoais: residência, idade, sexo, estado civil, escolaridade, profissão e ministério, conversão. Além dessas questões, a pesquisa se dividiu em blocos: questões doutrinárias, institucionais, políticas e sociais. O caleidoscópio absolutamente multifacetado e plural mostra a cara dessa denominação que tem um nome único, Assembleias de Deus, mas essa pluralidade não está apenas no nome, mas também em sua natureza. Atualmente, são mais de 12 milhões de assembleianos (dados do Censo 2010), conquanto seja impossível quantificar o número de pastores/as. Desde a década de 1950, a Assembleia de Deus é a maior denominação pentecostal do país, embora diferentes entre si, distintas e, quase sempre, divergentes. Nasceram em 1911 já plurais, mas a terceira geração de pastores assembleianos leva isso ao extremo. Esse novo estamento assembleiano – pastores com curso universitário e/ou pós-graduação – é uma nova liderança: quais condutas, tendências doutrinárias e políticas é o que se pretende entender nesta pesquisa. Abstract: In quantitative terms, university student population and the membership of the Assemblies of God are alike in Brazil. There were 3,928,260 university students in 1991 and 2,439,770 members in the Assemblies of God. In 2010, the number of students had risen to 12,679,010 students and to12,314,410 for members of the Assemblies of God. Both the number of university students and Assembly of God members have increased, including university students who are members or pastors from the Assemblies of God. Who are these university graduate Assembly of God pastors and what do they think? Over a thousand emails were sent to people from listings of conventions, ministries and churches, and also to friends indicated by those people; and 84 questionnaires were filled and returned. The first part of the questionnaire dealt with personal information questions: residence, age, sex, marital status, education, occupation, ministry, and conversion. Besides that, the research was divided into blocks: doctrinal, institutional, political and social issues. The multifaceted and plural kaleidoscope shows the face of this denomination that has a unique name, Assemblies of God, but this plurality isn’t only in its name, but also in its nature. There are currently more than 12 million members in the Assemblies of God (2010 Census), and it is impossible to quantify the number of ministers both male and female. Since the 1950s the Assemblies of God has accounted for the largest Pentecostal denomination in the country; and its associated churches are diverse, different, and often divergent. They were born plural in 1911, but the third generation of the Assembly pastors has taken it to the extreme. This new Assembly of God estate (ou “stratum”) makes up a new leadership. This research intends to understand the conduct, doctrinal and political trends of the current Assembly of God leadership. Keywords: University Students. Assembly of God Pastors. Identity. Abstract:
Music has always been an indispensable tool in any church setting. In Nigeria, the early missionaries appropriated music in liturgical services in form of hymn singing, chant and anthem as part of worship in the church. With continual apt growth of Christianity in Nigeria, Assemblies of God church evolved from the Pentecostal movement, with emphasis on the activity of the Holy Spirit and on holy living. Hence, contents and styles of music were made to suit the pattern and beliefs of Assemblies of God such as spirit motivated songs, evangelistic songs, mission songs and salvation songs other than the conventional hymns singing and anthems. This paper therefore attempts to highlight the distinctive traits of Assemblies of God, Uyo District, and its reflection on the music ministration. Also, it showcases the basics for music ministers (choristers and music directors) to include: musical background, spiritual life, social and leadership skills as fundamental principles, beliefs and moral conduct patterned by the beliefs of the church for quality service to God and humanity. Qualitative design and typical forms of data drawn from interviews, participant observations and relevant documents were applied in the study for data collection. The significance of this paper is to provide justifiable guidelines to every music minister to attain the required moral and music standard widely accepted and appreciated by God and man. With the findings, it recommends periodical re-orientation towards musical knowledge and spiritual relevance as regard to what Assemblies of God believe for effective music ministration

Abstract: This study aims to determine the problems of pastoral care that occur in the Assemblies of God Church in North Sulawesi, Indonesia. Assemblies of God's efforts are reconstructing (providing a solution proposal) regarding pastoral care in an effort to develop the Assemblies of God congregation. To obtain research data, the authors used documents, both books and journals as a theoretical basis and conducted interviews with 14 church leaders as respondents/research subjects from regional organizational leaders, pastors and academics. The research was conducted using qualitative research methods by conducting interviews with 14 Assemblies of God leaders in North Sulawesi. Retrieval of research data using probability sampling with purposive sampling technique. The results showed that there were problems in Assemblies of God's pastoral care, especially with regard to pastoral care. The problem that occurs is the wrong paradigm regarding the concept of pastoral care in pastoral care caused by the old theoretical concept of pastoral care among church leaders, and the church's struggles in terms of independence. Therefore, in this study, two new theories were proposed in the form of proposed solutions, namely; 1) reconstruction of theological education (contextual pastoral care development), and 2) Striving for church independence. Abstract:
There are two main historical works on Assemblies of God in Britain. The first is Donald Gee's Wind and Flame (originally published under the title The Pentecostal Movement in 1941; later revised and enlarged for publication in 1967). Gee was intimately involved in much of AoG's development not only in the British Isles but also overseas, There are, however, three things which Donald Gee fails to do and which I decided to attempt in the history which follows. First, and very properly, Gee underestimates his own contribution to the shape of British pentecostalism. A natural modesty prevented Gee from seeing all the value of his own efforts. Second, Gee very rarely gives the source of any information he cites. There is a complete absence of footnotes, references, printed materials and the like in his book. We simply do not know what and whom he consulted when he wrote. And, third, Gee fails to make any mention of the immense social and technological changes which took place in his life time. He gives us the foreground without the background, and yet the background was important. It matters, for example, that ordinary commercial air travel opened up after the 1939-45 war or that telephones became common in the 1950s. The Pentecostal movement did not develop in a vacuum and sometimes successful events are explicable by reference to forgotten factors. For example, the success of the great Stephen Jeffreys crusades makes more sense when one knows that, at one stage, he moved from town to town, each within easy travelling distance of the others; this allowed those who had been attracted by one set of meetings to travel to the next. Or that these crusades took place when the national health service in Britain did not exist and people were more desperate in their search for healing. The second main work is Walter Hollenweger's The Pentecostals (SCM, 1972). This sets British pentecostalism in a world wide context and allows comparisons with Pentecostal churches in Latin America, Canada, Australia, New Zealand, the Continent and North America. Inevitably, therefore, Hollenweger's book paints on a broad canvas and omits many events within British Assemblies of God. At the end of this thesis a list is given of all the people I interviewed or consulted by phone. Not listed, however, because references are given at appropriate places in the text or notes, are the various documents which became available to me. These included letters, handbills, newspaper cuttings, minute books, diaries, reports submitted to the General Conference, accounts, short-lived magazines and, of course, all the volumes of Redemption Tidings. Undoubtedly Redemption Tidings proved to be the richest source of information. It was published continuously from 1924-85 and contained a whole variety of articles, crusade reports, letters, editorials, stenographically recorded sermons, advertisements and the like which, more than any other single source, recreate early pentecostalism. Redemption Tidings was published monthly 1924-33 and then fortnightly 1934-1956 and weekly 1956-1985. So far as the ordering of the following history is concerned, I have simply moved forward decade by decade and with little attempt to group subjects together thematically. This rather unimaginative approach has the virtue of being systematic and it was used by Adrian Hastings in his excellent A History of English Christianity: 1920-1985 (Collins, 1986). At the start of each major section, I have briefly outlined the economic and political events of the era. At the end of each major section, I have paused for sociological comment. These comments are not intended to be exhaustive. Rather, I have used some of the tools and concepts of sociology to illuminate the historical development previously described. Alternation between description and analytic comment is slightly clumsy, but seemed to be the only sensible way of handling the overall task. The events of Pentecostal history are simply not well enough known to take them for granted: they need to be described first. Any attempt to describe them while simultaneously analysing them would have proved confusing in the extreme. It is also necessary to point out that this history pays particular attention to Pentecostalism in Britain and only mentions missionary work overseas to the extent that this it is relevant to what was happening in Britain. In some respects this is unfortunate, but to do justice to the extraordinary work of men and women in various continents of the world would require a separate study of comparable length.

Abstract:

The purpose of this research was to develop the components of a theory for retention of young people after their high school years by examining the factors that contribute to continued spiritual vitality in Assemblies of God (AG) post-high school young adults. Data was collected from a stratified sample of ninety-five young adults in the United States during their senior year of high school in 2011 and two years later in 2013. In line with research by the Fuller Youth Institute (FYI), continued spiritual vitality was operationalized by using the Religious Behavior Scale, the Religious Identity Scale, and the Risk Behavior Scale. The results identified nine elements from spiritual formation factors, social considerations, and high school youth group experiences that produced fourteen statistically significant correlations with higher levels of retention and spiritual vitality in the sample two years after leaving school. This research appears to suggest that it is the aggregated effect of intentional youth group experiences providing opportunity for the internalized guidance of the Holy Spirit, recognized as God's work, and not specific youth group programs or religious activities that have the potential to create a unique spiritual journey that would ensure spiritual vitality for the youth after they leave high school.

Abstract:
In this study, a thermosonic flip chip process has been developed to assemble gold bumped die to an Al2O3 ceramic substrate. The effects of bonding conditions, such as ultrasonic power, temperature, bonding force and bonding time, on the reliability of the interconnections were investigated. It was found that ultrasonic power and bonding force were the major factors influencing the reliability. High ultrasonic power and bonding force resulted in high reliability. The thermal cycling test results (from −55 °C to 125 °C) showed that the assemblies passed 1250 cycles with no change in resistance. The pressure cooker test results (15 psig pressure and 100% RH at temperature of 121 °C) showed that the assemblies passed 168 hours with no change in resistance. The shear force test results showed that the shear force at failure ranged from 10 to 40 grams per bump depending on bonding parameters. The interconnect cross-sections and the fracture surfaces on selected dies and substrates were observed with scanning electron microscope (SEM). It was found that the shear failure usually occurred between a gold pad and ceramic substrate, indicating a strong bonding interface between the gold bump and pad on the substrate. The performance of the assemblies fell into the reliable region as long as the bonding force, temperature, and ultrasonic power were high enough. The failure mechanisms consisted of separation between Au bump and Au pad, separation between Au pad and substrate and cracking inside Au pad or trace. The ultrasonic power drove relative movement between Au bumps and Au pads, causing friction and heat in the interface to result in micro-welding.

Abstract:
In this research we investigate the design of nesting forces for exactly constrained, robust mechanical assemblies. Exactly constrained assemblies have a number of important advantages including the ability to assemble over a wide range of conditions. Such designs often require nesting forces to keep the design properly seated. To date, little theory has been developed for the design of nesting forces. We show how the effects of tolerances on nesting forces, a key issue, can be analyzed and apply the analysis to a simple design problem. For the example problem, good agreement is achieved with results from Monte Carlo simulation.

Assembly related issues along with component reliability are the main focus of this experimental research. This experimental study aims to evaluate the mechanical integrity of solder joints comprising of both lead-free and SnPb alloys. Lead-free and SnPb solder pastes were used to assemble the components. This will allow us to check the forward and the backward compatibility of the solder alloys. The test vehicle considered for this study contained a variety of components such as ultra chip scale package (UCSP192), package on package (PoP), plastic ball grid array (PBGA-676 & 1156), very thin chip array BGA (CVBGA432), thin small outline package (TSOP-40 & 48), dual row micro-lead frame (DRMLF), micro-lead frame (MLF-36 & 72), and chip resistors (0201, 0402, 0603). The scope of this paper is limited to the performance evaluation for area array packages only. Solder ball alloy for the area array packages included SAC 305, SAC405, SAC105, SnAg and SnPb. Three different PCB surface finishes, electroless nickel immersion gold (ENIG), SnPb hot air solder level (HASL), and immersion silver (ImAg) were used. Different solder ball alloys and surface finish combinations provided good comparison data for investigating the assembly performance. The PCB assemblies were subjected to mechanical shock test in the as-soldered condition and also after 200 and 500 thermal shock cycles at −55 to 125°C. For the mechanical shock test, the assemblies were subjected to 30 drops from a height of 3 ft, generating an average G force of 485N. After each drop the components were checked for the continuity of the total daisy chain. The number of drops for the first failure was used in analyzing the performance of the components for various combinations. Since each component had many independent daisy chains, the failure of the individual daisy chains was later used in determining the location of the failure and how it progressed. Two sets of test vehicles were assembled. One set comprised of components with lead-free solder balls of different composition (SAC305, SAC405, SAC105, SnAg) and the other set comprised of components with lead-free solder balls and SnPb solder balls (SAC305, SAC405, SnPb). This mix of alloy composition provided adequate data for comparison. It was critical to optimize the process in order to enable the melting of the mix of alloys. The area array package performance was evaluated when assembled with lead-free and SnPb solder paste. Some of the assemblies were cross-sectioned after the tests and the microstructure of the solder joint was analyzed to study the possible cause for assembly failure.

In the first two phases of this project, the accuracy and reliability of both conventional and more advanced nondestructive inspection techniques were evaluated by way of a round-robin program of blind inspections The results indicate that there is much variability in the accuracy and reliability of discontinuity detection and sizing depending on details of the NDT procedures adopted, which includes specification of equipment, techniques, calibration methods, and reporting requirements. In Phase III, optimized procedures for sleeve fillet welds and a limited number of branch groove welds were developed in the laboratory using fabricated assemblies containing Intentionally placed discontinuities. In response to a recommendation in Phase Ill, the current phase developed optimized procedures for a wide range of branch groove welds. The results of this phase indicate that the reliability and accuracy of nondestructive inspection techniques is not as high as obtained for sleeve fillet welds in the previous phase of the program In particular, the detection of sub-surface discontinuities by ultrasonic inspection is made more difficult by the added complexity of the branch weld geometries. As with sleeve fillet welds however, the probability of detecting weld toe cracks of a significant size can be quite good.

Abstract:
The LaRonde Zone 5 (LZ5) mine is part of the Doyon-Bousquet-LaRonde mining camp and is located in the southern part of the Abitibi greenstone belt in northwestern Quebec. The LZ5 deposit consists of three stacked mineralized corridors: Zone 4, Zone 4.1, and Zone 5. Zones 4 and 4.1 are discontinuous satellite mineralized corridors, whereas Zone 5 represents the main mineralized body. The mineralized zones of the LZ5 deposit and adjacent Ellison property (Ellison A and B zones) are hosted in the strongly-deformed, 2699-2695 Ma transitional to calcalkaline, intermediate to felsic, volcanic and volcaniclastic rocks of the Bousquet Formation upper member, which is part of the Blake River Group (2704-2695 Ma). Zones 4, 4.1, and 5 at the LZ5 mine are hosted in intermediate volcanic and volcaniclastic rocks of the Westwood andesitic to rhyodacitic unit (unit 5.1a), which forms the base of the upper member of the Bousquet Formation. The Ellison Zone A is hosted higher up in the stratigraphic sequence within a newly described intermediate volcanic unit. The Ellison Zone B is hosted in felsic volcanic and volcaniclastic rocks of the Westwood feldsparphyric rhyolite dome (subunit 5.3a-(b)). Mineralization in all three zones of the LZ5 deposit consists of discordant networks of millimeter- to centimeter-thick pyrite ±chalcopyrite ±sphalerite ±pyrrhotite veins and veinlets (10-20 % of the volume of the rock) and, to a lesser extent, very finely disseminated pyrite and boudinaged veins (less than or equal to 5 vol. % each) in strongly altered host rocks. Gold commonly occurs as microscopic inclusions in granoblastic pyrite and at the triple junction between recrystallized grains. The veins, stockworks, and disseminations were intensely folded and transposed in the steeply south-dipping, east-west trending S2 foliation. The vein network is at least partly discordant to the stratigraphy. A distal alteration halo envelops the LZ5 mineralized corridors and consists of a sericite-carbonate-chlorite- feldspar ±biotite assemblage. A proximal sericite-carbonate-chlorite-pyrite-quartz- feldspar-biotite ±epidote alteration assemblage is present within the LZ5 mineralized zones. A local proximal alteration assemblage of sericite-quartz-pyrite is also locally developed within Zone 4 and Zone 5 of the LZ5 deposit. Mass gains in Fe2O3 (t) and K2O, and mass losses in CaO, MgO, Na2O, and locally SiO2, are characteristic of the LZ5 alteration zones. The Ellison zone A and B are similar to LZ5 in terms of style of mineralization, but thin (10-20 cm) veins or bands of semi-massive to massive, finely recrystallized disseminated pyrite (0.1-1 mm) are distinctive. Chalcopyrite and sphalerite are also slightly more abundant in the mineralized corridors of the Ellison property and are usually associated with elevated gold grades. The zones are also slightly richer than at LZ5 in terms of gold and silver content, but narrower and less continuous in general. The Ellison Zone A is characterized by gains in Fe2O3 (t) and K2O and losses in CaO, MgO, Na2O, and SiO2. Gains in Fe2O3 (t) and local gains in K2O, MgO, and MnO, and losses in CO2, Na2O, P2O5, and SiO2, characterize the felsic host rocks of the Zone B corridor. The style of mineralization at LZ5 (pyrite ±chalcopyrite veins and veinlets, ±disseminated pyrite with low base metal content), its setting (i.e. in rocks of intermediate composition at the base of the upper member of the Bousquet Formation), and the geometry of its ore zones (stacked lenses of sulfide veins and veinlets, without massive sulfide lenses) differ from the other major deposits of the Doyon-Bousquet-LaRonde mining camp. Despite these differences, this study indicates that the LZ5 and Ellison mineralized corridors are of synvolcanic hydrothermal origin and have most likely been formed by convective circulation of seawater below the seafloor. An influx of magmatic fluids from the Mooshla synvolcanic intrusive complex or its parent magma chamber could explain the Au enrichment at LZ5, as has been suggested for other deposits of the camp. Evidence for a pre-deformation synvolcanic mineralization at LZ5 includes ductile deformation and recrystallization of the sulfides, the stacked nature of its ore zones, subconcordant alteration halos that envelop the mineralized corridors, evidence that the mineralized system was already active when the LZ5 lenses were deposited and control on mineralization by primary volcanic features such as the permeability and porosity of the volcanic rocks.

Abstract:
This report covers that part of a Phase II effort that was completed by Magnasonics, Inc. prior to its liquidation by an adverse court ruling. The Phase I program investigated several configurations of Electromagnetic Acoustic Transducers (EMAT5) to arrive at an optimum approach to the problem of detecting and sizing stress corrosion cracks (SCC) in operating pipelines. Phase II was designed to optimize the most promising configuration by applying it to as many pipe samples as possible. Three pipe samples that contained several colonies of 5CC were made available to Magnasonics and a computerized data collection apparatus was assembled to collect ultrasonic data on the cracked areas. In agreement with the Phase I findings, high order Lamb wave modes were found to reflect strongly from the stress corrosion cracks and thus provide a sensitive detection method. In order to develop sizing capacitates, the crack depths in the colonies of SCC were measured with an eddy current technique and by a new surface acoustic wave method. Good qualitative correlation between the severity of cracking and the amplitude of the reflected ultrasonic energy was observed but real quantitative comparisons must wait on destructive measurement of the actual crack depths. Since only 30% of the funding has been used to date, it is hoped that additional measurements can be made on even more samples in the future.

Abstract:
The main recommendations of the panel report can be summarised under five key headings:  Building blocks: The ultimate aim should be to build rich, detailed and testable narratives situated within a European context, and addressing phenomena from the longue durée to the short-term over international to local scales. Chronological control is essential to this and effective dating strategies are required to enable generation-level analysis. The ‘serendipity factor’ of archaeological work must be enhanced by recognising and getting the most out of information-rich sites as they appear. o There is a pressing need to revisit the archives of excavated sites to extract more information from existing resources, notably through dating programmes targeted at regional sequences – the Western Isles Atlantic roundhouse sequence is an obvious target. o Many areas still lack anything beyond the baldest of settlement sequences, with little understanding of the relations between key site types. There is a need to get at least basic sequences from many more areas, either from sustained regional programmes or targeted sampling exercises. o Much of the methodologically innovative work and new insights have come from long-running research excavations. Such large-scale research projects are an important element in developing new approaches to the Iron Age.  Daily life and practice: There remains great potential to improve the understanding of people’s lives in the Iron Age through fresh approaches to, and integration of, existing and newly-excavated data. o House use. Rigorous analysis and innovative approaches, including experimental archaeology, should be employed to get the most out of the understanding of daily life through the strengths of the Scottish record, such as deposits within buildings, organic preservation and waterlogging. o Material culture. Artefact studies have the potential to be far more integral to understandings of Iron Age societies, both from the rich assemblages of the Atlantic area and less-rich lowland finds. Key areas of concern are basic studies of material groups (including the function of everyday items such as stone and bone tools, and the nature of craft processes – iron, copper alloy, bone/antler and shale offer particularly good evidence). Other key topics are: the role of ‘art’ and other forms of decoration and comparative approaches to assemblages to obtain synthetic views of the uses of material culture. o Field to feast. Subsistence practices are a core area of research essential to understanding past society, but different strands of evidence need to be more fully integrated, with a ‘field to feast’ approach, from production to consumption. The working of agricultural systems is poorly understood, from agricultural processes to cooking practices and cuisine: integrated work between different specialisms would assist greatly. There is a need for conceptual as well as practical perspectives – e.g. how were wild resources conceived? o Ritual practice. There has been valuable work in identifying depositional practices, such as deposition of animals or querns, which are thought to relate to house-based ritual practices, but there is great potential for further pattern-spotting, synthesis and interpretation. Iron Age Scotland: ScARF Panel Report v  Landscapes and regions:  Concepts of ‘region’ or ‘province’, and how they changed over time, need to be critically explored, because they are contentious, poorly defined and highly variable. What did Iron Age people see as their geographical horizons, and how did this change?  Attempts to understand the Iron Age landscape require improved, integrated survey methodologies, as existing approaches are inevitably partial.  Aspects of the landscape’s physical form and cover should be investigated more fully, in terms of vegetation (known only in outline over most of the country) and sea level change in key areas such as the firths of Moray and Forth.  Landscapes beyond settlement merit further work, e.g. the use of the landscape for deposition of objects or people, and what this tells us of contemporary perceptions and beliefs.  Concepts of inherited landscapes (how Iron Age communities saw and used this longlived land) and socal resilience to issues such as climate change should be explored more fully.  Reconstructing Iron Age societies. The changing structure of society over space and time in this period remains poorly understood. Researchers should interrogate the data for better and more explicitly-expressed understandings of social structures and relations between people.  The wider context: Researchers need to engage with the big questions of change on a European level (and beyond). Relationships with neighbouring areas (e.g. England, Ireland) and analogies from other areas (e.g. Scandinavia and the Low Countries) can help inform Scottish studies. Key big topics are: o The nature and effect of the introduction of iron. o The social processes lying behind evidence for movement and contact. o Parallels and differences in social processes and developments. o The changing nature of houses and households over this period, including the role of ‘substantial houses’, from crannogs to brochs, the development and role of complex architecture, and the shift away from roundhouses. o The chronology, nature and meaning of hillforts and other enclosed settlements. o Relationships with the Roman world

Abstract:
Rugose wood is a complex disease of grapevines, which occurs in all growing areas. The disease is spread in the field by vector transmission (mealybugs). At least five elongated-phloem- limited viruses are implicated in the various rugose wood disorders. The most fully characterized of these are Grapevine virus A (GV A) and GVB, members of a newly established genus, the vitivirus. GVC, a putative vitivirus, is much less well characterized than GV A or GVB. The information regarding the role of GVC in the etiology and epidemiology of rugose wood is fragmentary and no sequence data for GVC are available. The proposed research is aimed to study the etiology and epidemiology of rugose wood disease, and to construct genetically engineered virus-resistant grapevines. The objectives of our proposed research were to construct transgenic plants with coat protein gene sequences designed to induce post-transcriptional gene silencing (pTGS); to study the epidemiology and etiology of rugose wood disease by cloning and sequencing of GVC; and surveying of rugose wood- associated viruses in Californian and Israeli vineyards. In an attempt to experimentally define the role of the various genes of GV A, we utilized the infectious clone, inserted mutations in every ORF, and studied the effect on viral replication, gene expression, symptoms and viral movement. We explored the production of viral RNAs in a GV A-infected Nicotiana benthamiana herbaceous host, and characterized one nested set of three 5'-terminal sgRNAs of 5.1, 5.5 and 6.0 kb, and another, of three 3'-terminal sgRNAs of 2.2, 1.8 and 1.0 kb that could serve for expression of ORFs 2-3, respectively. Several GV A constructs have been assembled into pCAMBIA 230 I, a binary vector which is used for Angrobacterium mediated transformation: GV A CP gene; two copies of the GV A CP gene arranged in the same antisense orientation; two copies of the GV A CP gene in which the downstream copy is in an antigens orientation; GV A replicase gene; GV A replicase gene plus the 3' UTR sequence; and the full genome of GV A. Experiments for transformation of N. benthamiana and grapevine cell suspension with these constructs have been initiated. Transgenic N. benthamiana plants that contained the CP gene, the replicase gene and the entire genome of GV A were obtained. For grapevine transformation, we have developed efficient protocols for transformation and successfully grapevine plantlets that contained the CP gene and the replicase genes of GV A were obtained. These plants are still under examination for expression of the trans genes. The construction of transgenic plants with GV A sequences will provide, in the long run, a means to control one of the most prevalent viruses associated with grapevines. Our many attempts to produce a cDNA library from the genome of GVC failed. For surveying of rugose wood associated viruses in California vineyards, samples were collected from different grape growing areas and tested by RT-PCR for GV A, GVB and GVD. The results indicated that some of the samples were infected with multiple viruses, but overall, we found higher incidence of GVB and GV A infection in California vineyards and new introduction varieties, respectively. In this research we also conducted studies to increase our understanding of virus - induced rootstock decline and its importance in vineyard productivity. Our results provided supporting evidence that the rootstock response to virus infection depends on the rootstock genotype and the virus type. In general, rootstocks are differ widely in virus susceptibility. Our data indicated that a virus type or its combination with other viruses was responsible in virus-induced rootstock decline. As the results showed, the growth of the rootstocks were severely affected when the combination of more than one virus was present.
