Abstract:

This paper presents the results of the work of the new field initiative launched by the British Museum at the Darband-i Rania pass in the Kurdistan Region of Iraq. The pass is located at the northeastern corner of Lake Dokan, where, though now subsumed into the lake, the Lower Zab flows from the Peshdar into the Rania Plain. It is a strategic location on a major route from Mesopotamia into Iran, and control of both the road and the river must always have been important. The aim of the work, which commenced in autumn of 2016, is to explore a cluster of sites that commanded the pass, with a particular focus on the first millennium b.c. Excavation is being carried out principally at two sites: Qalatga Darband, a large fortified site at the western end of the pass, and Usu Aska, a fort inside the pass itself. The occupations of these two sites are predominantly Parthian and Assyrian respectively. Smaller operations have also been carried out at Murad Rasu, a multi-period site situated on a headland across the waters on the southern shore of Lake Dokan. The results have included the discovery at Qalatga Darband of a monumental complex built of stone and roofed with terracotta roof tiles containing the smashed remains of Hellenistic statuary. Other features indicative of Hellenistic material culture are Mediterranean-type oil-presses and Corinthian column bases and capitals. At Usu Aska remains are being uncovered of an Assyrian fortification of massive proportions.

Abstract:

Successful bovine embryo transfer requires synchronization of luteolysis, estrus and ovulation. The objective of the present study was to evaluate the effect of a combination of a PRID, PGF2� and eCG, on estrus synchronization and pregnancy rate in recipient heifers. A PRID� (ASKA Pharmaceutical Co., Ltd., Tokyo, Japan) was inserted into the vagina at random days of the estrous cycle for 7 (n = 35) or 9 (n = 43) days. Two days before removal of the PRID, the heifers were injected with PGF2� IM (2 mL Resipron�-C containing 0.25 mg mL-1 cloprostenol; ASKA). About half of the heifers in each group received 250 IU eCG IM (Serotropin�; ASKA) at the time of PRID removal. Blood was collected several times from the start of treatment for 7 (n = 9) or 9 (n = 9) days and on the day of embryo transfer by jugular venipuncture; plasma was immediately separated and stored at -20�C until assayed for plasma concentrations of estradiol-17α (E2) and progesterone (P4). The E2 and P4 determinations were performed by enzyme immunoassay after extraction by diethyl ether. Pregnancy was determined by ultrasonography on Day 30 (Day 0 = estrus). The rates of successful standing estrus (no. in estrus/PRID inserted), embryo transfer (no. transferred/estrus), and pregnancy (no. pregnancy/transferred) were compared between groups. Data were analyzed by chi-square analysis or Fisher's PLSD test following ANOVA. Injection of eCG at the time of PRID removal had no significant effect on the rates of successful standing estrus, embryo transfer, or pregnancy (P > 0.05). The proportion of heifers treated for 9 days that exhibited standing estrus (93&percnt;, 40/43) was significantly higher than the proportion of heifers treated for 7 days that exhibited standing estrus (66&percnt;, 23/35, P < 0.01). Of the heifers that were treated for 9 days, the proportion of heifers exhibiting standing estrus within 2 days after the end of treatment was significantly higher (93&percnt;, 37/40) than for heifers that were treated for 7 days (65&percnt;, 15/23; P < 0.01). Pregnancy rates of heifers treated for 9 days (84&percnt;, 32/38) and 7 days (81&percnt;, 17/21) were not significantly different. The E2 : P4 ratio normally increases during follicle growth and CL regression. The plasma E2 : P4 ratio between the time of injection of PGF2α and the time of PRID removal was significantly higher for heifers that were treated for 9 days than it was for heifers that were treated for 7 days (P < 0.01). These results suggest that a combination of PRID treatment for 9 days and injection of PGF2α 2 days before PRID removal successfully synchronized estrus in recipient heifers and led to high pregnancy rates following embryo transfer.

Belum dimilikinya peralatan-peralatan dan sistem instalasi penunjang pekerjaan finishing ducco indoor, seperti ruang pengering/oven dan indoor spray booth merupakan kendala bagi UKM “nuriska intergraha” yang memiliki anggaran belanja kecil dan sangat terbatas. Upaya peningkatan produktivitas melalui pengembangan dan penerapan fasilitas ruangan kamar pengering / oven cat yang membantu proses produksi mebel pada UKM “nuriska intergraha”, khususnya untuk mempermudah proses pengeringan cat dan pelapisan dasar (epoxy) pada saat musim penghujan, menjadi fokus penelitian ini.

Abstract:

Apoptosis signal–regulating kinases (ASK1, ASK2, and ASK3) are activators of the p38 and c-Jun N-terminal kinase (JNK) mitogen-activated protein kinase (MAPK) pathways. ASK1–3 form oligomeric complexes known as ASK signalosomes that initiate signaling cascades in response to diverse stress stimuli. Here, we demonstrated that oligomerization of ASK proteins is driven by previously uncharacterized sterile-alpha motif (SAM) domains that reside at the carboxy-terminus of each ASK protein. SAM domains from ASK1–3 exhibited distinct behaviors, with the SAM domain of ASK1 forming unstable oligomers, that of ASK2 remaining predominantly monomeric, and that of ASK3 forming a stable oligomer even at a low concentration. In contrast to their behavior in isolation, the ASK1 and ASK2 SAM domains preferentially formed a stable heterocomplex. The crystal structure of the ASK3 SAM domain, small-angle x-ray scattering, and mutagenesis suggested that ASK3 oligomers and ASK1-ASK2 complexes formed discrete, quasi-helical rings through interactions between the mid-loop of one molecule and the end helix of another molecule. Preferential ASK1-ASK2 binding was consistent with mass spectrometry showing that full-length ASK1 formed hetero-oligomeric complexes incorporating large amounts of ASK2. Accordingly, disrupting the association between SAM domains impaired ASK activity in the context of electrophilic stress induced by 4-hydroxy-2-nonenal (HNE). These findings provide a structural template for how ASK proteins assemble foci that drive inflammatory signaling and reinforce the notion that strategies to target ASK proteins should consider the concerted actions of multiple ASK family members.

Desain penelitian ini menggunakan cross sectional study ( studi potong lintang ) dengan tehnik penarikan sampel yaitu Purposive sampling terhadap populasi target sehingga didapatkan 57 responden.Dari uji statistik didapatkan hasil bahwa rata-rata responden yang diambil sebagai sampel mempunyai nilai persentasi yang hampir sama untuk kejadian terhadap derajat kegawatan penyakit diare, Hasil Uji Kai-kuadrat menunjukkan bahwa tidak ada hubungan status sosial ekonomi dengan sub varibel pendidikan, pengetahuan, pekerjaan dan pendapatan dengan derajat kegawatan penyakit diare di Puskesmas Aska Kabupaten Maros. Kesimpulan dari penelitian ini adalah Hasil dari uji univariat menggambarkan bahwa rata-rata klien yang datang untuk mendapat pertolangan terhadap anggota keluarga yang menderita penyakit diare memiliki nilai yang hampir sama dari setiap sub variabel sosial ekonomi dan hasil dari uji bivariat menunjukkan ; tidak ada hubungan antara tingkat pendidikan dengan derajat kegawatan penyakit diare ,tidak ada hubungan antara tingkat pengetahuan dengan derajat kegawatan penyakit diare,tidak ada hubungan antara tingkat pekerjaan dengan derajat kegawatan penyakit diare,tidak ada hubungan antara tingkat pendapatan dengan derajat kegawatan penyakit diare.Adapun saran yang dianjurkan agar pemerintah terkait kesehatan dapat lebih meningkatkan fungsi promotif untuk dapat lebih meningkatkan pengetahuan masyarakat tentang penyakit-penyakit endemic yang sering terjadi dimasyarakat, Untuk pemberi pelayanan kesehatan agar lebih memperhatikan kondisi-kondisi klien yang baru masuk dengan kondisi sakit parah atau dalam kondisi baik, sehingga penanganan akan lebih terarah,Untuk peneliti selanjutnya diharapkan agar dapat meneliti tentang sikap anggota kelurga terhadap penurunan angka kejadian kegawatan derajat dehidrasi pada penyakit diare.

Abstract:

The use of oxidizing agents is one of the most favorable approaches to kill bacteria in daily life. However, bacteria have been evolving to survive in the presence of different oxidizing agents. In this study, we aimed to obtain a comprehensive list of genes whose expression can make Escherichiacoli cells resistant to different oxidizing agents. For this purpose, we utilized the ASKA library and performed a genome-wide screening of ~4200 E. coli genes. Hydrogen peroxide (H2O2) and hypochlorite (HOCl) were tested as representative oxidizing agents in this study. To further validate our screening results, we used different E. coli strains as host cells to express or inactivate selected resistance genes individually. More than 100 genes obtained in this screening were not known to associate with oxidative stress responses before. Thus, this study is expected to facilitate both basic studies on oxidative stress and the development of antibacterial agents.

Abstract:

Recombinant protein production plays an essential role in both biological studies and pharmaceutical production. Escherichia coli is one of the most favorable hosts for this purpose. Although a number of strategies for optimizing protein production have been developed, the effect of gene overexpression on host cell growth has been much less studied. Here, we performed high-throughput tests on the E. coli a complete set of E. coli K-12 ORF archive (ASKA) collection to quantify the effects of overexpressing individual E. coli genes on its growth. The results indicated that overexpressing membrane-associated proteins or proteins with high abundances of branched-chain amino acids tended to impair cell growth, the latter of which could be remedied by amino acid supplementation. Through this study, we expect to provide an index for a fast pre-study estimate of host cell growth in order to choose proper rescuing approaches when working with different proteins.

ABSTRACT The efficient production of membrane proteins in bacteria remains a major challenge. In this work, we sought to identify overexpressed genes that enhance the yields of recombinant membrane proteins in Escherichia coli. We developed a genetic selection system for bacterial membrane protein production, consisting of membrane protein fusions with the enzyme β-lactamase and facile selection of high-production strains on ampicillin-containing media. This system was used to screen the ASKA library, an ordered library of plasmids encoding all the known E. coli open reading frames (ORFs), and several clones with the ability to accumulate enhanced amounts of recombinant membrane proteins were selected. Notably, coexpression of ybaB, a gene encoding a putative DNA-binding protein of unknown function, was found to enhance the accumulation of a variety of membrane-integrated human G protein-coupled receptors and other integral membrane proteins in E. coli by up to 10-fold. The results of this study highlight the power of genetic approaches for identifying factors that impact membrane protein biogenesis and for generating engineered microbial hosts for membrane protein production.

Abstract:

Repeat-breeding Holstein cows (RBCs), i.e. cows that have undergone three or more AIs, have reduced pregnancy rates. The present study aimed to determine whether application of a combination of a PRID� (ASKA Pharmaceutical Co., Inc., Tokyo, Japan) + prostaglandin F2α (PGF2α) for estrus synchronization and AI + embryo transfer (ET) would improve pregnancy rates in RBCs. All RBCs selected showed a normal interval between the previous insemination and estrus, and were free of clinical disorders. The RBCs (n = 107) received a PRID for 9 days with a capsule containing 10 mg of estradiol benzoate, 0.5 mg IM of cloprostenol (PGF2α, Resipron�-C; ASKA, Japan) at 2 days before PRID removal. The cows were inseminated with frozen–thawed semen from a bull at 0 to 12 h after onset of estrus. Seven days after AI, single in vitro-produced embryos were transferred to the same RBCs. Pregnancy was diagnosed by palpation per rectum at e60 days post-insemination. Exp. 1 estimated the response to estrus synchronization treatment (n = 107). The factors examined were lactation number (mean � SD: 2.3 � 1.5, range: 1–6), insemination number (5.2 � 2.2, range: 3–13), and the interval from calving to next AI (275.9 � 97.6 days, range: 93–650 days). Exp. 2 compared the pregnancy rate in control RBCs (n = 161) and PRID-treated RBCs (n = 100). The factors examined included lactation number (2.5 � 1.6, range: 1–12), insemination number (6.0 � 2.5, range: 3–19), the interval from calving to next AI (288.0 � 102.1 days, range: 102–797 days), and whether the ET was into the uterine horn ipsilateral or contralateral to the CL in a total of 261 RBCs. Data were analyzed using logistic regression. The results for Exp. 1: 103 cows (96.3%) were confirmed to be in estrus (2.0 � 0.3 days after PRID removal). Although the frequency of estrus was affected by the lactation number (P = 0.014) and the interval from calving to the first AI (P = 0.005), the AI number did not have an affect (P = 0.624). Exp. 2: PRID synchronization had no effect on pregnancy rate (control: 37.3%, 60/161; and PRID-treated: 40.0%, 40/100). For the RBCs, the pregnancy rate was not affected by lactation number (P = 0.093), AI number (P = 0.694), or the interval from calving to next AI (P = 0.762). No significant difference (P = 0.428) in pregnancy rate was observed in cows when the embryos were transferred into the uterine horn contralateral to the CL (43.5%, 20/46), compared to those embryos transferred into the uterus horn ipsilateral to the CL (37.2%, 80/215). In conclusion, our findings suggest that the pregnancy rate may be improved by considering lactation number and the interval from calving to the next AI of RBCs in estrus synchronization using a combination of PRID + PGF2α and AI + ET.

The results of financial statements analysis and corporate governance reporting of insurance companies ASKA-LIFE, TAS, KD Life, PZU Ukraine, UNIQA Life, MetLife were presented. It was substantiated that, within studying the life insurance companies’ reputation, along with main financial indicators, there is a need to analyze in details such indicators as insurance premiums and investment income for one insured from savings life insurance, average payments, current accounts payable, etc.It was proved that for reputation capital development, it is worth strengthening the role of corporate social responsibility, and to consider insurance companies’ assessment on the part of clients and employees who are brand advocates and affect the companies’ reputation formation.

Synovial sarcoma is a rare but aggressive soft-tissue sarcoma associated with translocation t(X;18). Metastasis occurs in approximately 50% of all patients, and curative outcomes are difficult to achieve in this group. Since the efficacies of current therapeutic approaches for metastatic synovial sarcoma remain limited, new therapeutic agents are urgently needed. Tilapia piscidin 4 (TP4), a marine antimicrobial peptide, is known to exhibit multiple biological functions, including anti-bacterial, wound-healing, immunomodulatory, and anticancer activities. In the present study, we assessed the anticancer activity of TP4 in human synovial sarcoma cells and determined the underlying mechanisms. We first demonstrated that TP4 can induce necrotic cell death in human synovial sarcoma AsKa-SS and SW982 cells lines. In addition, we saw that TP4 initiates reactive oxygen species (ROS) production and downregulates antioxidant proteins, such as uncoupling protein-2, superoxide dismutase (SOD)-1, and SOD-2. Moreover, TP4-induced mitochondrial hyperpolarization is followed by elevation of mitochondrial ROS. Calcium overload is also triggered by TP4, and cell death can be attenuated by a necrosis inhibitor, ROS scavenger or calcium chelator. In our experiments, TP4 displayed strong anticancer activity in human synovial sarcoma cells by disrupting oxidative status, promoting mitochondrial hyperpolarization and causing calcium overload.

En la actualidad, el tomate es la hortaliza de mayor consumo en el mundo, ya que su producción total al año alcanza los 130 millones de toneladas. La demanda ha ido aumentando significativamente por el creciente reconocimiento de su valor nutritivo, representado por el licopeno, entre otros. De hecho, la industria del tomate está generalmente concentrada en el área llamada la faja del tomate, situada en la zona semitropical, entre los 30 y 40° de latitud norte y los 30 y 40 grados de latitud sur.Sin embargo, nos parece difícil incrementar la producción del tomate aún más en esta zona, debido a la aguda competencia en torno a la distribución de recursos entre el tomate y otros productos agrícolas. En cambio, en la zona baja y tropical del Perú, situada entre los 0 y 30 grados, se ha reportado que la producción del pimiento páprika (que pertenece a la misma categoría botánica del tomate) ha prosperado exitosamente. Por otra parte, en la industria del tomate en el Perú, se han presentado diferentes fracasos consecutivos hasta el año 2000. No obstante, en el Proyecto ASKA (Proyecto Experimental de Tomate, realizado en la zona de Chavimochic, en Trujillo, con la participación de la Empresa Nacional KAGOME-Japón), llevado a cabo con la estrecha colaboración entre la industria del tomate y la Universidad Nacional Agraria La Molina, se pudo comprobar que el cultivo del tomate industrial era factible en la costa peruana. e23561 Background: While Synovial Sarcoma (SS) only accounts for about 10% of soft tissue sarcomas (STS), or ~10,000 cases/year, ~4,000 of these will be fatal. Little benefit has been seen with newer combination chemotherapies and immune checkpoint inhibitors. SS is a transcription factor-driven cancer characterized by an t(X;18) chromosomal translocation resulting in the SS18-SSX fusion gene. SS is also known to overexpress the anti-apoptotic protein BCL-2, which has been used as a diagnostic marker for SS. Venetoclax, the FDA approved BH3 mimetic which specifically inhibits BCL-2, has been used to treat BCL-2-driven hematological cancers. Yet, preclinical studies of venetoclax, currently in clinical use for BCL-2 related hematologic malignancies, have failed in SS, raising the possibility of other BCL-2 family members playing a role in resistance. Methods: Publicly available gene expression databases of SS were queried for expression of BCL-2 family member expression, including the endogenous MCL-1 inhibitor, NOXA. Six SS cell lines, including ASKA, HS-SY-II, Yamato, SW982, SYO.1, and an ex vivo line from a SS patient-derived xenograft (PDX) were tested in vitro with combinations of venetoclax and the MCL-1 inhibitor S63845, using cell viability assays. In vivo testing employed both a patient-derived xenograft (PDX) model of SS and a cell line derived (SYO.1) xenograft model. Results: Queries of published gene expression data for SS demonstrated that SS expresses low levels of the endogenous MCL-1 inhibitor, NOXA, nominating a mechanism for intrinsic venetoclax resistance in these tumors, and suggesting rational combination of venetoclax and the MCL-1 inhibitor, S63845. In vitro, SS all cell lines possessing the SS18-SSX fusion gene demonstrated synergistic sensitivity to combined S63845 and venetoclax, despite generally insensitivity to either drug as monotherapy. Importantly, in an SS PDX, we demonstrated S63845 and venetoclax synergize to induce tumor regression. In the SYO.1 xenograft model, combination therapy significantly reduced tumor growth compared to no treatment or single agent groups. Conclusions: These findings provide the preclinical rationale for translation of the rational combination of BCL-2 and MCL-1 inhibitors into clinical trials for SS.

Abstract:

Daily rhythms of behaviors and physiologies are generated by the circadian clock, which is composed of clock genes and the encoded proteins forming transcriptional/translational feedback loops (TTFLs). The circadian clock is a self-sustained oscillator and flexibly responds to various time cues to synchronize with environmental 24-h cycles. However, the key molecule that transmits cellular stress to the circadian clockwork is unknown. Here we identified apoptosis signal-regulating kinase (ASK), a member of the MAPKKK family, as an essential mediator determining the circadian period and phase of cultured cells in response to osmotic changes of the medium. The physiological impact of ASK signaling was demonstrated by a response of the clock to changes in intracellular redox states. Intriguingly, the TTFLs drive rhythmic expression of Ask genes, indicating ASK-mediated association of the TTFLs with intracellular redox. In behavioral analysis, Ask1, Ask2, and Ask3 triple-KO mice exhibited compromised light responses of the circadian period and phase in their activity rhythms. LC-MS/MS–based proteomic analysis identified a series of ASK-dependent and osmotic stress-responsive phosphorylations of proteins, among which CLOCK, a key component of the molecular clockwork, was phosphorylated at Thr843 or Ser845 in the carboxyl-terminal region. These findings reveal the ASK-dependent stress response as an underlying mechanism of circadian clock flexibility.

Plants possess multiple homologues of the SKP1 gene encoding an essential subunit of the SCF ubiquitin ligases, but only ASK1 (Arabidopsis SKP1-like 1) and ASK2 have been characterised genetically. In addition, little is known about the function of SKP1 homologues in monocots. Here we report on a winter wheat homologue of SKP1 named TSK1 (Triticum aestivum SKP1-like 1). Expression analyses revealed that it was expressed predominantly in young roots and floral buds. RNA in situ hybridisation showed that it was expressed in the shoot apical meristem (SAM) and anthers, especially the tapetum and microsporocytes at the time of meiosis. It was also expressed in almost the entire meristematic and elongation zones of the root. These observations indicated that TSK1 might function in dividing cells. The Arabidopsis ask1-1 mutant with overexpressed TSK1 driven by the CaMV 35S promoter exhibited partial fertility, suggesting that TSK1 could partially restore function in meiosis to the ask1-1 mutant. In addition, overexpression of TSK1 in wild type Arabidopsis resulted in changes in auxin responses and auxin-related phenotypes, consistent with a role of ASK1 in Arabidopsis auxin response. These results suggest possible functional conservation between TSK1 and ASK1.

Abstract:

We have established an indoor artificial breeding system for the cynomolgus monkey in an effort to increase the number of MII oocytes that are required for enhanced reproductive efficiency. A conventional ovarian stimulus method requires FSH to be administered to monkeys intramuscularly once a day for 9 days. Recently, a novel implantable and programmable micro-infusion pump (iPRECIO™, Primetech Corp, Tokyo, Japan) has been introduced for small laboratory animals to infuse fluids continuously for long periods of time in vivo. We adapted this micro-infusion pump to administer FSH to cynomolgus monkeys. In this study, we optimized the controlled-release program of FSH for the appropriate ovarian stimulation. First, laparoscopic evaluation was performed to identify animals that had small, underdeveloped follicles and gonadotropin-releasing hormone (0.9 mg animal–1; Leuplin, Takeda, Osaka, Japan) was administered to all selected animals. Two weeks later, iPRECIO™ containing FSH (Gonapure, ASKA, Tokyo, Japan) was implanted subcutaneously and the continuous infusion was started at 15.0 IU kg–1 per day. Five days after implantation, follicular development was evaluated by laparoscopy and the infusion rate was adjusted based on follicular profile (high level: reduced to 12.5 IU kg–1 per day, n = 11; middle level: maintained at 15.0 IU kg–1 per day, n = 47; low level: increased to 20.0 IU kg–1 per day, n = 30). Four days later, hCG (400 IU kg–1, IM) was administered and follicular aspiration was performed 40 h later. In the control group (n = 6), FSH (25.0 IU kg–1 per day) was injected intramuscularly once a day for 9 days, followed by an hCG injection. Oocytes were collected and evaluated and MII oocytes were fertilized by intracytoplasmic sperm injection. Injected oocytes were cultured for 7 days in CRML-1066 medium supplemented with 20% bovine serum at 38°C, with 5% CO2 and 5% O2 in air and blastocyst development was evaluated. Data were analysed by a two-sided t-test. All animals treated with the controlled-release FSH using iPRECIO™ showed significantly higher MII maturation rates (mean: 59.4%, 22/37; P < 0.05) than those of the control group (MII rate: 46.3%, 19/41); however, there was no significant difference among the different FSH release programs. Blastocyst development rates of the test group were also significantly higher than those of the control group (test: 52.0%, control: 28.1%; P < 0.05); however, there was no significant difference among the different FSH programs. This controlled-release system did not require daily injections to the animal, which would be beneficial for decreasing stress. Further, the required dose of FSH using iPRECIO™ was much less than that of the conventional multiple-injection method. These results indicated that controlled release of FSH utilising an iPRECIO™ pump can be customized based on follicular profile and has financial and animal care advantages compared with the conventional method.

Abstract:

We have shown that a simplified oestrus synchronization protocol using prostaglandin F2α (PG) and oestradiol benzoate (EB) resulted in a satisfactory conception rate, similar to that obtained in high-producing dairy cows with natural oestrus (Yamaguchi et al. 2018 Reprod. Fertil. Dev. 30, 147 abstr). However, we found no significant effect of PG+EB protocols on the first-service conception rates (CR) of dairy cows. This study investigated the effect of the first-service CR by using PG+EB protocols in the high-producing dairy cows that had not exhibited oestrus expression after calving. Data concerning the first-service CR were obtained for 481 Holstein cows from 4 commercial dairy herds in Hokkaido and Ibaraki, Japan, from 2014 to 2017. The average number of parity, average interval between calving and the first service, and average milk yield (at 305 days) of the cows were 2.2±1.3, 89.7±26.1 days, and 10314±1994kg, respectively. We carried out the PG+EB protocols on cows not showing evident standing heat from Days 50 to 150 postpartum regardless of the presence of a corpus luteum in the ovary. The statuses of ovaries of cows were confirmed using ultrasonography before starting the treatment. Prostaglandin F2α (25 mg; Lutalyse, Pfizer, New York, NY, USA) was injected into cows with a >20mm cutoff for functional corpus luteum diameter at a random stage of oestrus, and EB (1 mg; Ovahormon®, Aska Animal Health Co., Tokyo, Japan) was injected 24h later, and AI was carried out 24 to 48h after EB injection. Cows with a natural oestrus service from Days 50 to 150 postpartum were used as control. All the cows were inseminated after the onset of standing heat or removal of the tail chalk. Pregnancy diagnoses were performed using ultrasonography 30 to 45 days after AI. The CR and intervals of partum to conception were analysed using a chi-square test and ANOVA. The first-service CR (i.e. the number of pregnant cows/total number of cows in the treatment group) of the natural oestrus (n=330) and PG+EB (n=151) groups were 42.7 and 41.1%, respectively, and there were no significant differences between these 2 groups (P=0.7). The average interval between calving and the first service of the PG+EB groups was significantly longer than that of the natural oestrus groups (86.4±1.5 days v. 96.9±1.9 days; P<0.01). The average days open of the natural oestrus and PG+EB groups were 88.5±3.2 days and 98.1±2.3 days, respectively (P<0.05). These results confirmed that the first-service CR in the PG+EB and natural oestrus groups of high-producing dairy cows were similar. We suggest that conducting PG+EB protocols was effective in the high-producing dairy cows that had not exhibited oestrus expression after calving. Moreover, using PG+EB protocols could prevent the further increase in the number of days open.

Abstract:

The objective of this study was to evaluate the effect of inducing oestrus in superovulation-treated Japanese Black donor cows after egg collection using a controlled internal drug release (CIDR) device and prostaglandin F2α treatments. The test animals were 50 biparous or triparous Japanese Black cows that had been treated for superovulation during the period of 75-110 days post-calving and from each of which 10 or more ova or embryos had been recovered using a uterine reflux method. In the 25 cows of the control group that received the conventional treatment, a CIDR was not introduced into the vagina, the number of days from egg harvesting to standing oestrus was recorded, and AI was performed using frozen semen. At the time of AI, 50µg of gonadotrophin-releasing hormone (GnRH; Consultan, Aska Animal Health) was administered intramuscularly. The cows that did not conceive in the first AI attempt were again given the same AI and GnRH treatments in the next oestrus cycle. In the 25 cows of the treatment group, a CIDR (CIDR 1900, Zoetis) device was inserted in the vagina after egg harvesting and was kept there for 12 days. At the time of insertion and removal of the CIDR, 150µg of d-cloprostenol formulation (Dalmazin, Kyoritsu Seiyaku) was administered intramuscularly. The number of days from egg harvest to standing oestrus was recorded, and GnRH was administered at the time of AI. The cows that did not conceive in the first AI attempt were given the same AI and GnRH treatments in the next oestrus cycle. The control and treatment groups were compared for the number of days from egg harvesting to oestrus (t-test) and the conception rate after AI (chi-square test). The mean number of days from egg collection to standing oestrus in the control group was 38.6±11.9, and the conception rate was 60% (15/25). Among the animals that failed to conceive in the first AI attempt, the second standing oestrus occurred 60.7±13.1 days after egg collection, and 70% (7/10) of the cows conceived after the second AI. In the treatment group, on the other hand, the mean number of days from egg collection to standing oestrus was 15.8±3.9, and 72% (18/25) of the cows conceived after the first AI. Among the cows that failed to conceive, the second standing oestrus occurred 37.4±4.7 days after egg harvest, and 71% (5/7) of the cow conceived after the second AI. There was a significant difference between the two groups in the number of days from egg collection to the first standing oestrus (P<0.05). The results described above suggest that keeping a CIDR in the vagina for 12 days and intramuscularly administering prostaglandin F2α at the time of insertion and removal of the CIDR in superovulated cows are effective for early onset of standing oestrus after egg collection and thus can reduce the number of days to conception.
