Abstract:
In this paper, we propose a bus-independent hardware (HW)-based approach to secure long protocol data units (PDUs) in Automotive Open System Architecture (AUTOSAR)-based automotive electronic control units (ECUs). Our approach is based on extending previous works that implemented two AUTOSAR communication (COM) application-specific instruction set processors (ASIPs). COM ASIP V1 introduced two instructions to handle the transmission and reception of PDUs no larger than 8 bytes and signals no larger than 32 bits individually through send signal and receive signal instructions. COM ASIP V2 introduced two extra instructions to handle long signals and PDUs of arbitrary lengths. We extended the instruction set architecture (ISA) of our previous ASIPs by introducing six new instructions, in COM ASIP V3, to hash PDUs that contain these signals to authenticate transmission and reception of such PDUs. The experimental results show that COM ASIP V3 can handle (i.e., transmit, receive, calculate hash, or verify hash) a 64-byte controller area network flexible data-rate (CAN FD) frame in 1.575 μs and a 254-byte FlexRay frame in 6.301 μs. These measurements indicate that the throughput of our new COM ASIP is much higher, 42× to 75×, than the throughput required by these communication buses.

Abstract:
Abstract The increase in complexity and size of modern ASIC designs in the HEP community and the use of advanced semiconductor fabrication processes raises the need for a shift toward a more abstract design methodology, that takes advantage of modularity and programmability to achieve a faster turnaround time both for design and verification. This contribution will present two complementary approaches, one using a RISC-V based System-on-Chip (SoC) and the other based on Application-Specific Instruction set Processors (ASIP). The SoC uses the PicoRV32 open-source RISC-V core and a rad-hard version of the AMBA APB bus to connect peripherals and is primarily geared towards control and monitoring applications. This solution is a demonstrator of what can become a more complete fully radiation-tolerant SoC platform with a standardized interconnect and an IP block library, to serve as the starting point for future ASIC designs. The ASIP based approach targets more the design of data path elements and the use in data processing applications. The presented approach makes use of a commercial ASIP Designer EDA tool to demonstrate an integrated workflow to define, benchmark and optimize an ASIP for a specific use case, starting from a general-purpose processor.

Abstract:
Agouti signaling protein (ASIP), the human (h) homolog of agouti, is an endogenous melanocortin peptide antagonist. To date, characterization of this protein has been performed with recombinant protein only and without the availability of an ASIP/agouti radioligand. In this report we describe the functional characteristics of a chemically synthesized truncated ASIP variant, ASIP-[90–132 (L89Y)], and the binding characteristics of its cognate radioligand,125I-ASIP-[90–132 (L89Y)]. Similar to full-length recombinant ASIP/agouti, ASIP-[90–132 (L89Y)] was a potent inhibitor of α-melanocyte-stimulating hormone cAMP generation at the cloned human melanocortin receptor (hMCR) subtypes hMC1R and hMC4R. It also displayed a lesser degree of inhibition at the hMC3R and hMC5R. However, ASIP-[90–132 (L89Y)] was found to be less potent than full-length recombinant ASIP and, surprisingly, only exhibited weak inhibitory activity at the hMC2R. In competition binding assays with the radioligand 125I-ASIP-[90–132 (L89Y)], ASIP-[90–132 (L89Y)] displayed a hierarchy of binding affinity that roughly paralleled its rank order of inhibitory potency at the various MCR subtypes, i.e., hMC1R ≈ hMC4R > hMC3R ≈ hMC5R > hMC2R. Structure-activity studies revealed that ASIP-[90–132 (L89Y)] possessed greater pharmacological potency than either the further truncated ASIP variants ASIP-(116–132) or cyclo(CRFFRSAC). Interestingly, the latter molecules were both weak agonists at the hMC1R. These studies further support the concept that ASIP/agouti inhibits melanocortin action by directly binding to target MCRs and provide additional insight into the structural requirements for maximal inhibitory potency.

Factors present in the oocyte and surrounding follicular cells aid in the attainment of oocyte competence. Agouti-signalling protein (ASIP) is a known regulator of melanocyte function through binding to melanocortin receptors including MC1R and MC4R. Additionally, ASIP has been classified as an adipokine due to a link with insulin resistance and obesity in humans. In mice, expression is limited to hair follicles where ASIP regulates hair pigmentation. In cattle, however, ASIP mRNA has been detected in a variety of tissues, including adipose, skin, heart, testis, and the ovary. Despite ovarian expression, the role of ASIP in reproduction remains undetermined. Bovine ASIP is a secreted protein consisting of 133 amino acids. The aim of this experiment was to provide a detailed description of the ASIP expression profile within the bovine ovary and during early embryonic development. Reverse transcription PCR (RT-PCR) was conducted to analyse ASIP, MC1R, and MC4R mRNA expression. Samples examined included fetal ovaries from gestational day 90 to 250, adult ovary, fetal testis, adult testis, and 12 somatic tissues including adrenal, cerebral cortex, gut, heart, intestine, kidney, liver, lung, muscle, pituitary, stomach, and thymus. Amplification of ribosomal protein L19 (RPL19) served as a positive control for all samples. Expression of ASIP was detected in the fetal testis, 9 somatic tissues, and the fetal and adult ovary. In the fetal ovary, ASIP was detected as early as 90 days of gestation and continued throughout gestation. Expression of the ASIP receptors, MC1R and MC4R, were detected exclusively in the fetal ovary. To further characterise ASIP expression, quantitative real-time PCR (RT-qPCR) was utilised to examine samples including germinal vesicle and MII oocytes (pool of 10 oocytes), invitro-produced embryos ranging from the 2-cell to blastocyst stages (pool of 10 embryos), and cumulus and granulosa cells collected from a pool from 5 cumulus–oocyte complexes (COCs) and follicles, respectively. Theca cells from a single follicle were analysed. Samples with cycle threshold values below 35 were considered to express the gene of interest. Of the follicular cells examined, ASIP expression was present in theca, granulosa, and cumulus cells. ASIP expression was detected in both GV and MII oocytes. Early embryonic expression of ASIP was detected in the 2-cell embryo and continued to the blastocyst stage of development. In conclusion, ASIP is present in the bovine adult and fetal ovary, follicular cells including cumulus, granulosa, and theca cells, GV and MII oocytes, and invitro-produced embryos from the 2-cell to blastocyst stages. Future research will focus on identifying the function of ovarian and early embryonic ASIP in cattle.

Pemberian ASI pada Ibu Bekerja terhambat pada waktu menyusui karena intensitas pertemuan Ibu dan Bayi berkurang. Alternatif yang bisa ditempuh adalah pemberian ASIP. Tujuan penelitian adalah untuk mengetahui Hubungan Pendidikan dan Pengetahuan tentang ASIP dengan Pemberian ASIP pada Ibu Bekerja di Puskesmas Batangtoru Tahun 2021. Metode Penelitian bersifat Survey analitik desain cross sectional dilaksanakan di Puskesmas Batangtoru. Populasi seluruh Ibu bekerja yang memiliki bayi usia ≥ 6 bulan sampel sebanyak 36 orang teknik Purvosive sampling. Hasil Pengumpulan data dengan kuesioner dan uji statistic Fisher dan Continuity Correlation. Pada analisa bivariat didapatkan p value < α, ada hubungan pendidikan dengan pemberian ASIP ( P value = 0,004) dan ada hubungan pengetahuan dengan pemberian ASIP (P value = 0,002). Kesimpulan ada hubungan pendidikan dan pengetahuan ibu tentang asi eksklusif dengan pemberian ASI perah pada ibu bekerja. Disarankan kepada Puskesmas meningkatkan Promosi kesehatan tentang ASIP agar ibu mau melakukan ASIP.`

ASIP can fulfill data processing effectively and flexibly in specific application. Due to the instruction’s diversity and the flexibility of logical structure, it has increased test vectors and enhanced difficulty in the functional verification of ASIP [1].With the continuous spread of ASIP application, the scale of designing is enlarging and the functional verification has become the bottleneck of designing of ASIP. This paper presents a methodology able to accomplish the functional verification of ASIP. The “Bottom-up” methodology is composed of the component-level verification, the instruction-level verification and the FPGA-based prototype system verification. The paper proves the feasibility and efficiency through the experiments in functional verification of ASIP.

<p><em>Background: Breastfeeding on Working Mothers is hampered at breastfeeding as the intensity of the mother and baby encounters decreases. The alternative that can be taken is the provision of ASIP. Objective: To know the Education Relationship and Knowledge of ASIP with ASIP Assessment on Working Mother in Desa Tanjung Aur Kelurahan Balai Gadang Work Area of Cold Water Health Center Year 2017. Method: Cross sectional design cross sectional study was conducted in Tanjung Aur Village Balai Gadang District Working Area of Puskesmas Cold Water 2017. Data collection dated 3-7 July 2017. Population of all working mothers with infants aged ≥ 2-11 months sampled as many as 36 people total sampling technique. Result: Data collection with questionnaire and chi-square statistic test. Univariate was found from 36 respondents, 25 people (69,4%) did not give ASIP to their babies, 21 people (58,3%) low education level, 29 people (80,6%) low knowledge level. In bivariate analysis obtained p value &lt;α, there is correlation of education with giving ASIP (P value = 0,002) and there is correlation of knowledge with giving ASIP (P value = 0,001). Conclusion: Mothers Working in breastfeeding in the village of Tanjung Aur Kelurahan Balai Gadang Working Area Cold Water Health Center partially did not provide ASIP to the baby. AbstractHere we report a heterozygous tandem duplication at the ASIP (agouti signaling protein) gene locus causing ubiquitous, ectopic ASIP expression in a female patient with extreme childhood obesity. The mutation places ASIP under control of the ubiquitously active itchy E3 ubiquitin protein ligase promoter, driving the generation of ASIP in patient-derived native and induced pluripotent stem cells for all germ layers and hypothalamic-like neurons. The patient’s phenotype of early-onset obesity, overgrowth, red hair and hyperinsulinemia is concordant with that of mutant mice ubiquitously expressing the homolog nonagouti. ASIP represses melanocyte-stimulating hormone-mediated activation as a melanocortin receptor antagonist, which might affect eating behavior, energy expenditure, adipocyte differentiation and pigmentation, as observed in the index patient. As the type of mutation escapes standard genetic screening algorithms, we rescreened the Leipzig Childhood Obesity cohort of 1,745 patients and identified four additional patients with the identical mutation, ectopic ASIP expression and a similar phenotype. Taken together, our data indicate that ubiquitous ectopic ASIP expression is likely a monogenic cause of human obesity.

Abstract:
The mammalian protein ASIP/PAR-3 interacts with atypical protein kinase C isotypes (aPKC) and shows overall sequence similarity to the invertebrate proteins C. elegans PAR-3 and Drosophila Bazooka, which are crucial for the establishment of polarity in various cells. The physical interaction between ASIP/PAR-3 and aPKC is also conserved in C. elegans PAR-3 and PKC-3 and in Drosophila Bazooka and DaPKC. In mammals, ASIP/PAR-3 colocalizes with aPKC and concentrates at the tight junctions of epithelial cells, but the biological meaning of ASIP/PAR-3 in tight junctions remains to be clarified. In the present study, we show that ASIP/PAR-3 staining distributes to the subapical domain of epithelial cell-cell junctions, including epithelial cells with less-developed tight junctions, in clear contrast with ZO-1, another tight-junction-associated protein, the staining of which is stronger in cells with well-developed tight junctions. Consistently, immunogold electron microscopy revealed that ASIP/PAR-3 concentrates at the apical edge of tight junctions, whereas ZO-1 distributes alongside tight junctions. To clarify the meaning of this characteristic localization of ASIP, we analyzed the effects of overexpressed ASIP/PAR-3 on tight junction formation in cultured epithelial MDCK cells. The induced overexpression of ASIP/PAR-3, but not its deletion mutant lacking the aPKC-binding sequence, promotes cell-cell contact-induced tight junction formation in MDCK cells when evaluated on the basis of transepithelial electrical resistance and occludin insolubilization. The significance of the aPKC-binding sequence in tight junction formation is also supported by the finding that the conserved PKC-phosphorylation site within this sequence,ASIP-Ser827, is phosphorylated at the most apical tip of cell-cell contacts during the initial phase of tight junction formation in MDCK cells. Together,our present data suggest that ASIP/PAR-3 regulates epithelial tight junction formation positively through interaction with aPKC.

Abstract:
Agouti signalling protein (ASIP) is a coat colour-related protein and also is a protein-related to lipid metabolism, which had first been found in agoutis. According to our previous study, ASIP is a candidate gene that affects the lipid metabolism in bovine adipocytes. However, its effect on milk lipid has not been reported yet. This study focused on the effect of the ASIP gene on the lipid metabolism of mammary epithelial cells in cattle. The ASIP gene was knocked out in bMECs by using CRISPR/Cas9 technology. The result of transcriptome sequencing showed that the differentially expressed genes associated with lipid metabolism were mainly enriched in the fatty acids metabolism pathways. Furthermore, the contents of intracellular triglycerides were significantly increased (p < 0.05), and cholesterol tended to rise (p > 0.05) in bMECs with the knockout of the ASIP gene. Fatty acid assays showed a significant alteration in medium and long-chain fatty acid content. Saturated and polyunsaturated fatty acids were significantly up-regulated (p < 0.05), and monounsaturated fatty acids were significantly decreased in the ASIP knockout bMECs (p < 0.05). The Q-PCR analysis showed that knockout of ASIP resulted in a significant reduction of gene expressions like PPARγ, FASN, SCD, and a significant up-regulation of genes like FABP4, ELOVL6, ACSL1, HACD4 prompted increased mid-to long-chain fatty acid synthesis. Overall, ASIP plays a pivotal role in regulating lipid metabolism in bMECs, which could further influence the component of lipid in milk.

Abstract:
Abstract The excess production of monoclonal immunoglobulin is the hallmark of multiple myeloma (MM) diagnosis and is an essential element of the determination of disease response to therapy. The development of new monoclonal immunoglobulins that are distinct from the baseline diagnostic paraprotein during the course of MM therapy can therefore pose a challenge in assessing disease response or potential relapse. There are prior reports of the development of abnormal protein banding (APB) after autologous stem cell transplantation for MM comprised of transient non-myeloma related mono- or oligoclonal protein bands that have been correlated with higher rates of event-free and overall survival. We now report the development of atypical serum immunofixation patterns (ASIPs) for the first time outside of the setting of stem cell transplant during the course of MM induction therapy with the BiRD (Biaxin® [clarithromycin], Revlimid® [lenalidomide], Dexamethasone) regimen. 72 patients with newly diagnosed Stage II and III MM (Salmon-Durie Criteria) were treated in a phase 2 clinical study of BiRD. The BiRD treatment regimen was given in 28-day cycles as follows: Clarithromycin 500mg po BID for days 1 – 28, Lenalidomide 25mg po daily for days 1 – 21, and Dexamethasone 40mg po weekly on days 1, 8, 15, and 21. The median age of the patients was 63 (range 36–83), and the baseline monoclonal immunoglobulins detected on serum immunofixation were as follows: 61% IgG, 22% IgA, 17% light chain only. 24 patients (33%) developed one or more ASIPs with either a mono- or oligoclonal banding pattern during the course of BiRD therapy. The new protein bandsvaried widely in duration of appearance (range 26–315 days) as well as isotype with IgM-κ, IgM-λ, IgG-κ, IgG-λ, IgA-λ, free λ light chain, and free IgM heavy chain all observed in one or more instances. The extent of therapy prior to first ASIP development was variable as well, (range 27– 724 days, median of 178 days). Patients with ASIPs had significantly better response to BiRD vs. non-ASIP patients (p = .00002), with CR+sCR rate of 71% vs. 23%, and a VGPR or better rate of 96% vs. 61%. The extent of BiRD therapy prior to development of first ASIP did not correlate with response rate (p = .7284). Bone marrow examination by histology, karyotype, FISH, and PCR IgH / IgK clonality analysis confirmed the disease response and furthermore showed no evidence for newly emergent plasma cell or other B-cell malignancy to account for ASIP generation suggesting molecular remission in some of the patients. Peripheral blood analysis by flow cytometry also showed no evidence for a circulating B-cell neoplasm to account for the new immunoglobulin production. We propose that ASIP development during myeloma therapy with a lenalidomide-based regimen heralds a robust tumor reduction with a hitherto unprecedented rate of complete remission. The immune phenomena that contribute to ASIP generation are unclear at present, however it is neither due to a new clonal B-cell population or transformation of the original MM plasma cell clone.

The methods used are workshops and counseling. Infrastructure required was counseling room (Rumah Dataku dusun Cikoneng), phantom breast pocket books, videos milking the milk manually and using the pump, infokus, and speaker stuffs. The results of activities was increasing the knowledge of health workers in ASIP management during the COVID-19 pandemic. DNA mutations within genes associated with melanogenesis can affect melanin production, leading to dyschromias. Genes that are involved in synthesis of melatonin and may affect the color of skin are melanocortin 1 receptor (MC1R), agouti locus (ASIP), and tyrosinase-related protein-2 (TYRP2). In this study, SNP identification within ASIP, MC1R, and TYRP2 gene fragments in wild and farmed foxes (Vulpes vulpes) was performed. Nine mutations in the ASIP gene which allowed us to distinguish seven SNP profiles, fourteen mutations and five SNP profiles in the MC1R gene, and seven SNP profiles based on four polymorphic nucleotides in the TYRP2 gene were detected. Analyses of obtained profiles indicate that ASIP did not undergo mutations in the wild, and significant variability of SNP profiles was found for TYRP2, with specific haplotypes noted for farm foxes and American and European wild foxes.

Abstract The upper few meters of the ocean form a critical layer for air–sea interaction, but because of observational challenges this region is undersampled. However, the physical processes controlling momentum transfer, gas exchange, and heat transfer are all concentrated in the uppermost region of the ocean. To study this region, the Air–Sea Interaction Profiler (ASIP) was developed. This is an autonomous microstructure vertical profiling instrument that provides data from a maximum depth of 100 m to the ocean surface and allows measurements to be performed in an undisturbed environment. The core sensor package on ASIP includes shear probes, microstructure and CTD-quality temperature and conductivity sensors, a photosynthetically active radiation (PAR) sensor, and an oxygen optode providing a repeated high-resolution dataset immediately below the air–sea interface. Autonomous profiling is accomplished with thrusters that submerge the positively buoyant instrument. Once the desired depth is reached, ASIP ascends through the water column acquiring data. At the surface, ASIP acquires its position and transmits this over the Iridium satellite network. ASIP is then placed in a low-power mode for a specified period, whereupon it repeats the profile cycle. Two-way communication over the Iridium network allows mission parameters to be changed in real time. ASIP has been used to study several scientific questions, such as the impact of diurnal warming on atmospheric processes, turbulence scaling in the upper ocean, parameterizing air–sea gas exchange, salinity gradients in the ocean surface boundary layer (OSBL), and consequences for remote sensing.

Canine coat color is a readily observed phenotype of great interest to dog enthusiasts; it is also an excellent avenue to explore the mechanisms of genetics and inheritance. As such, multiple commercial testing laboratories include basic color alleles in their popular screening panels, allowing for the creation of genotyped datasets at a scale not before appreciated in canine genetic research. These vast datasets have revealed rare genotype anomalies that encourage further exploration of color and pattern inheritance. We previously reported the simultaneous presence of greater than two allele variants at the Agouti Signaling Protein (ASIP) locus in a commercial genotype cohort of 11,790 canids. Here we present additional data to characterize the occurrence of anomalous ASIP genotypes. We document the detection of combinations of three or four ASIP allele variants in 17 dog breeds and Dingoes, at within-breed frequencies of 1.32–63.34%. We analyze the potential impact on phenotype that these allele combinations present, and propose mechanisms that could account for the findings, including: gene recombination, duplication, and incorrect causal variant identification. These findings speak to the accuracy of industry-wide protocols for commercial ASIP genotyping and imply that ASIP should be analyzed via haplotype, rather than using only the existing allele hierarchy, in the future.

Abstract: The objective of this work was to identify single nucleotide polymorphisms (SNPs) in resequencing data from MC1R, ASIP, and TYRP1 genes derived from Crioula sheep (Ovis aris) with different coat colors. Polymorphisms in the ASIP (agouti-signaling protein), MC1R (melanocortin 1 receptor), and TRYP1 (tyrosinase-related protein 1) genes were analyzed in 115 sheep from Embrapa’s conservation nucleus of crioula sheep, in Brazil. A total of 7,914 bp were sequenced per animal, and 14 SNPs were identified. Two additional assays were performed to detect duplications and deletions in the ASIP gene. Ninety-five percent of the coat color variation was explained by epistatic interactions observed between specific alleles in the MC1R and ASIP genes. Evidence suggests an important role of TYRP1 variants for wool color, despite their low frequencies. The marker panel was efficient enough in predicting coat color in the studied animals and, therefore, can be used to implement a marker-assisted selection program in the conservation nucleus of sheep of the crioula breed.

Abstract. Unlike specific expression in the skin of wild mice, the agouti signaling protein (ASIP) is expressed widely in the tissue of cattle, including adipose and muscle tissue. Hence, it has been suggested that ASIP plays a role in bovine fat metabolism. An inserted L1-BT element was recently identified upstream of the ASIP locus which led to an ectopic expression of ASIP mRNA in cattle. In this study, we detected the indel of the L1-BT element at g. −14 643 nt and three SNPs in introns of the ASIP gene (g. −568A > G, g. −554A > T, and g. 4805A > T) in a Chinese Simmental steer population. The association analysis between variants of ASIP and economic traits showed that the homozygous genotype of L1-BT element insertion, AA genotype of g. −568A > G, and AT genotype of g. 4805A > T were significantly correlated with carcass and fat-related traits, such as live weight and back fat thickness. Moreover, three haplotypes (H1: AT; H2: AA; H3: GT) were identified by linkage disequilibrium analysis and formed six combined genotypes. Results indicated that Chinese Simmental steers with an H1H2 combined genotype had a higher measured value of fat-deposition-related traits (p<0.05), including thickness of back fat and percentage of carcass fat coverage, but a lower content of linoleic acid and α-linolenic acid (p<0.05). Individuals of an H3H3 combination had a lower marbling score, perirenal fat weight, and carcass weight (p<0.05). This suggests that these three SNPs and two combined haplotypes might be molecular markers for beef cattle breeding selection.

Cell polarity is fundamental to differentiation and function of most cells. Studies in mammalian epithelial cells have revealed that the establishment and maintenance of cell polarity depends upon cell adhesion, signaling networks, the cytoskeleton, and protein transport. Atypical protein kinase C (PKC) isotypes PKCζ and PKCλ have been implicated in signaling through lipid metabolites including phosphatidylinositol 3-phosphates, but their physiological role remains elusive. In the present study we report the identification of a protein, ASIP (atypical PKC isotype–specific interacting protein), that binds to aPKCs, and show that it colocalizes with PKCλ to the cell junctional complex in cultured epithelial MDCKII cells and rat intestinal epithelia. In addition, immunoelectron microscopy revealed that ASIP localizes to tight junctions in intestinal epithelial cells. Furthermore, ASIP shows significant sequence similarity to Caenorhabditis elegans PAR-3. PAR-3 protein is localized to the anterior periphery of the one-cell embryo, and is required for the establishment of cell polarity in early embryos. ASIP and PAR-3 share three PDZ domains, and can both bind to aPKCs. Taken together, our results suggest a role for a protein complex containing ASIP and aPKC in the establishment and/or maintenance of epithelial cell polarity. The evolutionary conservation of the protein complex and its asymmetric distribution in polarized cells from worm embryo to mammalian-differentiated cells may mean that the complex functions generally in the organization of cellular asymmetry.

Novel coat colour phenotypes often emerge during domestication, and there is strong evidence of genetic selection for the two main genes that control base coat colour in horses—ASIP and MC1R. These genes direct the type of pigment produced, red pheomelanin (MC1R) or black eumelanin (ASIP), as well as the relative concentration and the temporal–spatial distribution of melanin pigment deposits in the skin and hair coat. Here, we describe a genome-wide association study (GWAS) to identify novel genic regions involved in the determination of the shade of bay. In total, 126 horses from five different breeds were ranked according to the extent of the distribution of eumelanin: spanning variation in phenotype from black colour restricted only to the extremities to the presence of some black pigment across nearly all the body surface. We identified a single region associated with the shade of bay ranking spanning approximately 0.5 MB on ECA22, just upstream of the ASIP gene (p = 9.76 × 10−15). This candidate region encompasses the distal 5′ end of the ASIP transcript (as predicted from other species) as well as the RALY gene. Both loci are viable candidates based on the presence of similar alleles in other species. These results contribute to the growing understanding of coat colour genetics in the horse and to the mapping of genetic determinants of pigmentation on a molecular level. Given pleiotropic phenotypes in behaviour and obesity for ASIP alleles, especially those in the 5′ regulatory region, improved understanding of this new Shade allele may have implications for health management in the horse.

The brown hare (Lepus europaeus Pallas 1778) is a common palearctic and a popular game species therefore it has been an obvious subject for population genetic studies since the second part of the 20th century. Among the several mitochondrial DNA studies some have been carried out concerning nuclear genes as well. The agouti signaling protein gene (ASIP) is involved in regulating the synthesis of eumelanin and pheomelanin in melanocytes of mammals. Though many studies focused on it in relation with several mammalian species, minimal information is available on this topic concerning the brown hare. Here we present a short communication concerning the agouti signaling protein (ASIP) gene in four different country’s L. europaeus populations, namely Lithuania, Hungary, Serbia and Georgia. N=45 tissue samples have been investigated from overall 17 sampling sites of the different countries. There has not been found any polymorphism among the sequences. In an alignment with other Leporid species’ partial ASIP sequences downloaded from ENA we have found that based on a 178 base pairs long DNA sequence the haplotype of our samples contains three other Lepus species as well. This is concordant with the findings of a previous study focusing predominantly on the European rabbit (Orycto lagus cuniculus Linnaeus 1758) and the several mutations of its ASIP gene.

Interventions based on self-determination theory to help teachers support their students' autonomy have measured teachers' autonomy-supportive and controlling behaviors as unidimensional constructs. This study investigated whether a Web-Based Autonomy-Supportive Intervention Program (WB-ASIP) for physical education (PE) teachers would alter their students' perceptions of (a) the teachers' multidimensional autonomy-supportive and controlling behaviors, (b) their own psychological need satisfaction and frustration, and (c) perceived intrinsic motivation. We found that WB-ASIP significantly increased students' perceptions of all autonomy-supportive teacher behaviors and students' need satisfaction. At follow-up, we found that WB-ASIP led to a significant decrease in students' perception of teacher intimidation behaviors and students' autonomy frustration. These results suggest that a web-based intervention program to enhance PE teachers' autonomy-supportive behavior and minimize their controlling behavior was partially effective in positively changing their students' perceptions.

Although the European roe deer (Capreolus capreolus) population of North-West Germany has a remarkable number of melanistic specimens between 10% and 25%, the underlying genetic mutation-causing melanism is still unknown. We used a gene targeting approach focusing on MC1R and ASIP as important genes of coat coloration. Overall, 1384 bp of MC1R and 2039 bp of ASIP were sequenced in 24 specimens and several SNPs were detected. But only the ASIP-SNP c.33G>T completely segregated both phenotypes leading to the amino acid substitution p.Leu11Phe. The SNP was further evaluated in additional 471 samples. Generally, all black specimens (n = 33) were homozygous TT, whereas chestnut individuals were either homozygote GG (n = 436) or heterozygote GT (n = 26). Latar Belakang : Keberhasilan ASI Ekslusif pada ibu bekerja dipengaruhi oleh keberhasilan manajemen pemberian ASI perah (ASIP). Umumnya ibu yang gagal dalam manajemen ASIP akan segera memberikan susu formula karena merasa ASI ibu jumlahnya kurang, bahkan ada beberapa orang tua yang sengaja mengenalkan susu formula dari awal sebelum mulai bekerja. Bayi yang sudah mendapatkan susu formulacenderung berkurang frekuensi menyusuinya dan berdampak pada menurunnya produksi ASI. Tujuan: Penelitian ini bertujuan untuk mengetahui hubunganmanajemen ASIP (asi perah) terhadap keberhasilan asi eksklusif (studi kuantitatif pada ibu yang bekerja di instansi pemerintahan kota yogyakarta) Metode: Penelitian ini menggunakan jenis penelitian kuantitatif. Rancangan atau desain penelitian menggunakan pendekatan cross sectional.Sampel pada penelitian ini berjumlah 44 ibu yang memenuhi kriteria inklusi penelitian.Instrumen penelitian menggunakan kuesioner yang sudah dilakukan uji validitas dan reliabilitas. Metode analisis yang digunakan adalah uji statistik menggunakan analisis bivariat (Chi Square). Hasil : Karakteristik responden berdasarkan umur ibu paling banyak pada kategori umur > 35 tahun yaitu sebanyak 36 responden (75%), paritas ibu paling banyak berada pada kategori ibu dengan multipara sebanyak 30 responden (62,5%) dan cara persalinan paling banyak adalah persalinan normal yaitu sebesar 32 responden (66,6%). Ibu bekerja yang menerapkan manajemen ASIP yang paling banyak adalah pada kategori tidak tepat yaitu sebanyak 28 responden (58,3%) dan mayoritas responden tidak berhasil memberikan ASI ekslusif yaitu sebanyak 31 responden (64,6%).Hasil uji chi squaremenunjukkannilai p-value : 0,000 (

Animals exhibit a wide variety of genetically determined coat colors and pigmentation patterns that serve important roles in adaptation and communication. Although the genetics of the main coat colors in dogs have been studied extensively, there are types of coat pigmentation that have not been explained yet. Recently, an association between the variants in the ASIP gene Ventral (VP) and Hair Cycle (HCP) promoters with different coat colors in dogs has been established. Here, we used the new findings as a basis to investigate the genetics of the red sesame coat color in Shiba Inu dogs. Our study revealed that red sesame dogs carry a specific heterozygous ASIP promoter diplotype, VP2-HCP1/VP2-HCP3, where VP2-HCP1 is responsible for the red coat with a dark overlay, and VP2-HCP3 for a tan point-like pattern. This finding explains the inheritance of this coat color pattern and can be used by breeders to produce dogs with this rare phenotype. A comparison of sesame dogs (VP2-HCP1/VP2-HCP3) to a dog homozygous for the VP2-HCP1 promoter haplotype suggests that the incomplete dominance between the ASIP alleles may be involved in the sesame coat formation. These results are in good agreement with the new model explaining how different levels of ASIP gene expression affect the regulation of pigment synthesis in melanocytes.

A structured diary was used to collect information about the informal peer interactions of 536 children in their final year at nine primary schools. Measures of self-concept, social avoidance, and popularity were also taken. Initially, seven different scores were derived from the information contained in the diary to produce the Adolescent Social Interaction Profile (ASIP). Previous research with the ASIP subsequently identified three factors labelled scale, scope, and enjoyment of peer interactions, that were separate from a fourth generic self-concept factor derived from self-report questionnaires. Inspection of the factor scores for 19 children with full data sets who received the fewest peer nominations showed considerable heterogeneity, with only 1 child scoring low on more than two of the four factors. More expedient composite scores were then derived that reflected the factor loadings for the two complex ASIP scores labelled scale and scope. The utility of these scores for identifying and monitoring specific aspects of peer interaction difficulties is discussed.

Ibu pekerja harus memerah asi selama jam kerja bila ingin memberikan asi secara eksklusif. Penyimpanan ASIP harus memperhatikan level suhu dan durasi waktu penyimpanan agar tetap aman dikonsumsi bayi. Tujuan penelitian ini adalah mengetahui perbedaan keasaman ASIP berdasarkan lama penyimpanan. Populasi dalam penelitian ini adalah ibu pekerja yang memberikan ASI lewat ASI Perah. Sampel dalam penelitian ini adalah ASI dari Ibu Pekerja. Metode penelitian ini adalah metode kuantitatif secara deskriptif. Manfaat dari hasil penelitian ini yaitu diharapkan para ibu tidak ragu memberikan ASI eksklusif pada bayinya, para ibu pekerja bisa menyimpan ASIP nya di cooler bag atau lemari pendingin sehingga ibu pekerja masih tetap bisa memberikan ASI eksklusif selama 6 bulan dan dilanjutkan hingga bayi berusia 2 tahun.

We have synthesized cubic and linear polysiloxanes containing polyoxyethylene branches (ASiP-Cu) using tetraethoxysilane, polyoxyethylene glycol, and copper chloride as precursors; the products are stable to self-condensation. The effect of copper chloride content on the chemical structure of ASiP-Cu has been established. A special study was aimed at defining the modifying effect of ASiP-Cu on the sorption characteristics of membranes based on microporous, optically transparent block copolymers (OBCs). These OBCs were produced using 2,4-toluene diisocyanate and block copolymers of ethylene and propylene oxides. The study demonstrated significantly increased sorption capacity of the modified polymers. On the basis of the modified microporous block copolymers and 1-(2-pyridylazo)-2-naphthol (PAN) analytical reagent, an analytical test system has been developed. Additionally, the modified OBCs have the benefit of high diffusion permeability for molecules of organic dyes and metal ions. It has been shown that the volume of voids and structural features of their internal cavities contribute to the complex formation reaction involving PAN and copper chloride.

Considering the variability of bay coat colour, we aimed to investigate the association of different shades of bay with ASIP and MC1R genotype combinations and we studied the discrimination between the bay and black coat colour. We phenotypically characterized coat colour using a spectrophotometer. The measurements were based upon international standards as defined by the CIE L*a*b* colour system and we phenotyped five different body parts (neck, armpit area, shoulder, belly, croup) of 43 bay and 14 black horses kept under standardized conditions. From the five measuring points a stepwise discriminant analysis revealed that chromacity and luminescence of armpit area and luminescence of the neck were the most important traits to differentiate between black and bay horses, whereas it was shown that the red colour spectrum of neck, luminescence of the neck, and luminescence of the armpit area grouped bay horses according to their ASIP and MC1R genotype combinations. Within the group of bay horses the analyses identified a single ASIP/MC1R genotype combination (A/a E/E) where colour variables differed significantly from the three remaining genotype groups. A/a E/E horses were characterized in all body parts except in the armpit region by significant darker shades (lower luminescence, less chromacity). Regarding classifications of coat colour, we found in the significant cluster of A/a E/E horses the coat colour categories seal brown and dark (mahogany) bay. Overall, we were able to show that the characterization of equine coat colour based upon international standards as defined by the CIE L*a*b* colour system represents a valuable tool for a precise description of colour variation and association analyses.

Reconfigurable instruction set processors provide the possibility of tailor the instruction set of a CPU to a particular application. While this customization process could be performed during runtime in order to adapt the CPU to the currently executed workload, this use case has been hardly investigated. In this paper, we study the feasibility of moving the customization process to runtime and evaluate the relation of the expected speedups and the associated overheads. To this end, we present a tool flow that is tailored to the requirements of this just-in-time ASIP specialization scenario. We evaluate our methods by targeting our previously introduced Woolcano reconfigurable ASIP architecture for a set of applications from the SPEC2006, SPEC2000, MiBench, and SciMark2 benchmark suites. Our results show that just-in-time ASIP specialization is promising for embedded computing applications, where average speedups of 5x can be achieved by spending 50 minutes for custom instruction identification and hardware generation. These overheads will be compensated if the applications execute for more than 2 hours. For the scientific computing benchmarks, the achievable speedup is only 1.2x, which requires significant execution times in the order of days to amortize the overheads.
