Abstract:
This thesis presents a proposal of newly designed concept for travel hydraulic circuit of vibratory single drum roller ASC 110. The maximum machine gradeability was chosen as the main parameter for hydraulic circuit proposal. After selecting the new hydraulic travel concept, suitable hydraulic components have been selected. Length and local losses for two different machine speeds were calculated for sufficient hydraulic circuit design. Hydraulic losses have also been calculated for use in different climatic conditions. A new machine cooling concept has also been proposed. Heating and cooling curves of hydraulic circuit were calculated. For the purpose of comparing gradeability and energy balance of the newly designed travel concept compared to the existing one currently in mass production, both machines were fitted with measuring points. On both machines were measured for example, pressure, flow and temperature in each hydraulic circuit branch and others. From the time dependencies of the above-mentioned parameters it was possible to determine the energy efficiency of the newly designed concept. To assess the maximum gradeability, the slope of the hill was measured. Comparing the two machines, it has been found that the two hydraulic circuit solution has significantly improved traction properties in demanding machine applications, especially when driving uphill and downhill. This concept also affects less fuel consumption and places less demand on the hydraulic oil cooler due to its energy efficiency.

Abstract:
This paper explores the leasing standard update ASC 842, and its potential impact on investors. In addition to analyzing the differences between the old and new standard, this paper explores if there will be any impacts on investors, by testing investor reactions to news releases of ASC 842, conducting interviews with accounting professionals and testing the sophisticated ownership in selected companies. The results from these tests conclude that while there might be a difference in the financial statements with the implementation of ASC 842, there will be no significant impacts on investors of companies with large lease obligations.

Abstract:
The Papanicolaou Smear is a screening test which detects premalignant lesions of the uterine cervix. By treating these lesions, cervical cancer can be evaded. In 1988, a cytological diagnosis which communicated a state of uncertainty in the atypicality of cervical cells was first created in the Bethesda Cytology Classification scheme. This diagnosis is now known as atypical squamous cells of undetermined significance (ASC-US) and still little is known about its natural history.
This paper analyzes the results of a longitudinal study incorporating repeated regular measurements of viral and cytological endpoints as well as lifestyle and behavioural aspects, to understand the natural history of an ASC-US Pap smear and identify determinants of ASC-US acquisition and clearance.
Overall, the median duration of ASC-US is short, and is dependent on the definition of clearance since most lesions regress to normal. The factors most predictive of ASC-US acquisition but not clearance relate to HPV infection.

Abstract:
Facial Aging is caused by changes compromising the SMAS, Platysma, Skin and Facial Adipose Tissue. Currently, Facial Rejuvenation treatments imply to correct, reposition and replenish the hypertrophic or lax tissues to obtain more natural results, but considering less traumatic techniques and with short-term recovery periods. Less invasive surgeries are possible through the assistance of modern techniques in conjunction with new and adequate technology. Materials and Methods: In 2010, we incorporated to our practice the use of a new Laser wavelength: 1210-nm. Its advantage as a surgical instrument resides in its great affinity/absorption by adipose tissue. Results: This is the first wavelenght that does not produce lysis, furthermore, it preserves the adipocytes and stimulates the adipose-derived stem Cells (ASC). This wavelength also stimulates skin retraction in two points in time: a) Immediate effect; as it thickens the dermis. b) Late effect (3 months); after stimulating collagen production. Fat Grafts are widely spread nowadays as face fillers, but the most important difference we obtain is stimulation of ASC on our injecting material. What we acquire is not only a increase in volume but an important improvement in the quality of the neighbouring tissue of our graft. The considerable skin retraction caused by the use of the 1210-nm wavelength, allows us to diminish the dissection of the facelift’s grafts on the cervical level. The low complication levels presented on our Lipolaser technique permits us to use this technology with safety in the face, cheeks and cervical area. Conclusions: The use of this new Laser wavelength (1210-nm) that preserves and stimulates Adipocytes and ASC is considered a safe, convenient and adequate tool for our facial rejuvenation treatments, as it offers us a better grafting/filler material and an important skin retraction with less trauma, making it possible to associate our short-scar facelift with less dissection. This thesis examines the impact of the new revenue recognition standard, Accounting Standard Codification 606 (ASC 606), on the preparation and presentation of the financial statements. This thesis also inspects the impact of this change on the financial markets. This study begins with a qualitative examination of the impacts pertaining to ASC 606 through expert interviews and close examination of previously published reports. This paper later analyzes the financials of 10 specific companies and the effects attributed to ASC 606. And finally, this paper analyzes the effect of this change on the financials markets by examining the stock prices of each firm in the days surrounding the release of the updated financials.

Em um cenário atual em que o mundo apresenta ritmos de crescimento populacionais acelerados e consequentes acréscimos de habitantes vivendo em zonas urbanas, deve-se considerar que as cidades apresentam desafios e questões como alto índice de congestionamentos de trânsito, poluição do ar, consumo de recursos naturais, insegurança, etc. Surge-se, neste contexto, o desenvolvimento das chamadas Smart Cities, que são cidades que apresentam iniciativas inteligentes para solucionar ou melhorar a qualidade de vida do cidadão, baseadas em dimensões que envolvem o meio ambiente, governança, economia, mobilidade, pessoas, infraestrutura e tecnologia. A iniciativa escolhida foi a cidade de Amsterdã, mais precisamente a organização ASC (Amsterdam Smart City), que hoje representa, globalmente, um dos principais exemplos de iniciativas de Smart Cities do mundo. O presente estudo busca responder à seguinte questão de pesquisa: Como é estruturada a governança da ASC (Amsterdam Smart City) e como podem ser descritas as experiências vividas em uma smart city? Para responder à questão relacionado à governança, foi utilizado o modelo de redução lógica de governança proposto por Lynn (2000). E como base das unidades de sentido da análise fenomenológica, foi utilizado o Framework Integrativo de Iniciativas de Smart Cities na Amsterdam Smart City proposto por Chourabi et al., (2012). Visando um melhor entendimento buscou-se utilizar uma abordagem qualitativa, tendo como método o estudo de caso. A coleta de dados obteve-se através de entrevistas semiestruturadas para duas pessoas com amplo conhecimento da organização. E, para a análise fenomenológica, uma entrevista semiestruturada teve como base, porém foi utilizada uma narrativa descritiva do próprio autor a partir de suas próprias experiências vividas na cidade. Os resultados demonstrados no presente estudo refletem a importância de um olhar mais abrangente para o desenvolvimento de cidades inteligentes que não só a tecnologia a ser aplicada. Uma governança inteligente é tão importante quanto a tecnologia, pois molda e viabiliza os projetos, cria parcerias essenciais, estrutura e operacionaliza as iniciativas e realiza um papel importante na gestão municipal de mudança de paradigmas e cultura direcionada para a inovação.
In the current scenario, in which the world faces accelerated population growth rates and consequent increases in the number of inhabitants living in urban areas, it must be considered that cities present challenges and issues such as high traffic congestion levels, air pollution, high resource consumption, insecurity, etc. In this context, the development of the so-called Smart Cities, which are cities that present intelligent initiatives to solve or improve citizens\' quality of life, are based on dimensions that involve the environment, governance, economy, mobility, community, infrastructure and technology. The initiative chosen was the city of Amsterdam, more precisely the organization ASC (Amsterdam Smart City), which today represents, globally, one of the main examples of Smart Cities initiatives in the world. The present study aims to answer the following research question: How is the governance of the Amsterdam Smart City structured and how can be described the smart city life experiences? Regarding the governance matter, the logic reduction model of governance proposed by Lynn (2000) was used. And as a basis for the units of meaning of the phenomenological analysis, the Integrative Framework of Smart Cities Initiatives in the Amsterdam Smart City proposed by Chourabi et al. (2012) was taken as ground basis. Targeting a better understanding, a qualitative approach was applied in a case study methodology. The data collection was obtained through semi-structured interviews with two people containing deep knowledge of the organization. And, for the phenomenological analysis, a semi-structured interview was used as basic step, however a descriptive narrative of the author himself was also used based on his own experiences lived in the city. The outcomes shown in the present study reflect the importance of a broader look for the development of smart cities that are not just technology based. Intelligent governance is as important as technology, as it shapes and enables projects, creates essential partnerships, structures and operationalizes initiatives, and plays an important role in municipal management of paradigm shift and innovation-driven culture.

Abstract:
Retinoic acid-inducible gene-I (RIG-I) is a putative RNA helicase and recently identified as a cytosolic RNA receptor in mammalian cells. The role of RIG-I in the regulation of vascular function under physiological and pathological conditions is unknown. Recent studies have shown that the inflammasome serves as a crucial initiator of cytokine-mediated inflammation mediating the pathogenesis of cardiovascular disease. The present study tested whether RIG-I activation triggers inflammasome formation and subsequent cytokine-mediated inflammation in the endothelium of mice coronary arteries. Using both genetic and pharmacological interventions of the RIG-I inflammasome, we first characterized whether specific activation of RIG-I via 3pRNA transfection induced the formation of an ASC-containing inflammasome in mouse vascular endothelial cells (MVECs). We confirmed that 3pRNA dose-dependently increased RIG-I protein levels and release of of type I IFNβ and IL-1b (a prototype cytokine from inflammasome activation), confirming RIG-I activation. We found that MVECs transfected with 3pRNA exhibited increased colocalization of RIG-I with apoptosis-associated speck-like protein (ASC) or caspase-1 and elevated active caspase-1 and IL-1β levels, indicating the formation and activation of the RIG-I inflammasome. This RIG-I inflammasome activation was accompanied by endothelial barrier dysfunction. In the presence of 3pRNA, ZO-1 and ZO-1 and VE-Cadherin expression diminished, and inhibiting caspase-1 and silencing inflammasome components attenuated this effect. To test the functional role of RIG-I and its affect on the permeability of mouse ECs, we performed a transwell permeability assay. Results confirmed that 3pRNA induced increased permeabilization of these mouse ECs, which was attenuated when inhibiting and silencing inflammasome components. These data indicate that increased expression and activity of RIG-I activate IL-1b producing inflammasomes in ECs, which may represent an early molecular mechanism mediating vascular inflammation and endothelial dysfunction independent of Nlrp3. Furthermore, we investigated the role of anti-aging gene Klotho in the regulation of RIG-I inflammasome activation. The Klotho protein has been shown to directly interact with RIG-I in senescent cells to block RIG-I multimerization and downstream production of pro-inflammatory cytokines. Administration of D-saccharic acid 1,4-lactone (saccharolactone) in vitro, a pharmacological inhibitor of Klotho activity, substantially increased inflammasome activation. In addition, mice injected with saccharolactone exhibited increased RIG-I inflammasome formation and activation within the coronary artery endothelium. These results suggest that decreased Klotho activity may activate RIG-I and thereby increase inflammasome activity. Therefore, the present study defines a novel role for RIG-I inflammasome activation in vascular dysfunction.

Les conditions actuelles de réchauffement de la planète ont mené aux pays du monde à s'engager dans la durabilité et l’efficacité énergétique et la réduction des émissions de gaz à effet de serre. En tant que troisième consommateur d'énergie, le bâtiment représente un élément clé envers l'efficacité énergétique et de la stabilisation de la température globale. Plusieurs solutions existent pour la réalisation de ces objectifs, et les travaux présentés tout au long de cette thèse concernent un composant solaire particulier à la construction externe du bâtiment, appelé cheminée solaire. Cette thèse de doctorat porte sur l'analyse expérimentale et numérique des dispositifs de stockage d'énergie, sous forme de matériaux à changement de phase (PCM), afin d'optimiser les performances de cette technologie solaire. Le but de cette étude est de caractériser l’impact des panneaux Rubitherm RT44 PCM sur une cheminée solaire en laboratoire et in situ afin de permettre une comparaison avec la version classique. De plus, un modèle numérique a été développé et testé dans le but d'obtenir un outil numérique capable de représenter le comportement d'une cheminée solaire. Enfin, une optimisation à deux objectifs du modèle numérique de cheminée solaire intégrée PCM a été réalisée afin de déterminer certains des paramètres optimaux de ce type de technologie afin d’obtenir le flux d’air sortant le plus élevé possible, tout en maintenant une température suffisamment élevée dans la cheminée atteindre la gamme de fusion des PCM
The current global warming conditions have led nations across the world to commit into energetic sustainability and greenhouse gas emission reduction. Being the third greatest energetic consumer, the building represents a major key towards energy efficiency and global temperature stabilization. Several solutions exist for the accomplishment of these goals, and the works presented throughout this dissertation concerns a particular external building solar-driven component known as solar chimney. This PhD thesis focuses on the experimental and numerical analysis of energy storage devices, in the form of Phase Changing Materials (PCMs), for the optimisation of the performance of this solar technology. The aim of this study is to characterize the impact of Rubitherm RT44 PCM panels on a solar chimney under laboratory and in-situ conditions to carry out a comparison against the classic version. Additionally, a numerical model was developed and tested in the interest of obtaining a numerical tool capable of representing the behaviour of a solar chimney. Finally a bi-objective optimization of the PCM integrated solar chimney numerical model was carried out in order to determine some of the optimal parameters of this type of technology to obtain the highest exiting air flow, all while maintaining a high enough temperature across the chimney to reach the fusion range of the PCMs

Prior to the implementation of FAS 142 in 2002, goodwill was amortized annually. Now, companies with goodwill on their books must test it annually for impairment. This paper examines the effectiveness of goodwill amortization and impairment of goodwill both before and after the adoption of FAS 142 by analyzing their effect on abnormal stock returns. My results regarding the effect of goodwill amortization on stock returns are inconsistent, suggesting that goodwill amortization is not useful in determining the value of a company. My results also suggest that the stock market anticipates goodwill impairments before they are announced.

Abstract:
Allogeneic bone marrow transplantation (BMT) is an effective curative therapy for the majority of haematological malignancies and severe immunodeficiencies. The therapeutic potential of this procedure arises from the graft-versus- leukaemia (GVL) effect, which eradicates recipient malignancy after BMT and is mediated by donor T and natural killer (NK) cells. Unfortunately, GVL effects are inextricably linked to graft-versus-host disease (GVHD), the major cause of transplant-related mortality. GVHD manifests in two forms; acute and chronic GVHD. During acute GVHD, the skin, gastrointestinal tract, and liver are preferentially damaged by the transplanted donor immune system. Chronic GVHD generally develops in the later phase after transplant characterised by end-organ fibrosis. In the early stages of GVHD, donor T cells react to recipient- derived alloantigens presented predominantly by recipient antigen-presenting cells (APCs). Additionally, the success of transplant outcome is also determined by the potential for graft failure due to responses against donor immunohaematopoietic cells by recipient T or NK cells. Thus, there remains a clear need for greater understanding of the factors which lead to GVHD, associated opportunistic infection and disease relapse. Ultimately the aim is to reduce GVHD and associated immune deficiency whilst promoting the immunological clearance of leukaemia.

Abstract:
Steps Toward A Universal Soil Classification Chapter one focusses on the segmented nature of many schemes around the world and the requirements for a unified system. The strengths, weaknesses and suitability of these world systems lead to the conclusion that there is no unified soil taxonomic system. Chapter two discusses the steps of creating a world taxonomic system. It suggests that many of the philosophical problems with Australian taxonomy can be solved. A new algorithm is formulated in which key extragrade points are nominated, turning them into the focal point of clusters. Data that is not extreme is free to resolve into typical fuzzy k means (FKM) clusters within the end points. Chapter three takes akromeson and applies it to data from the United States Department of Agriculture (USDA). This chapter discusses the implications of this new method of horizon partitioning. Chapter four takes the ideas of principal component analysis and taxonomic distances. Algorithms regarding next nearest neighbour analysis (NNA) at the Order level for both taxonomies enables a new test for taxonomic accuracy. Chapter five consists of work on the mathematical methods required for a global taxonomy. Data needs to be converted into the same taxonomic space. Two Orders almost identical in pedological description are selected for analysis. The mean taxonomic distances for each Order are calculated to gain an understanding on how each Order from each classification system compares to Orders of the opposing taxonomic system. NNA is then further developed to determine which tiers of which taxonomy are on a similar operational level. Chapter six suggests there are taxa within both ST and ASC that are so close in properties and attributes as to be considered duplicates. The tiers of ST are matched by NNA to the closest tier of the ASC. These levels are harmonized and an algorithm devised to remove duplicate soil taxa, then the taxonomies are combined and assessed. In Chapter seven, ST is assessed, compared and matched with the ASC. The combined data set is split into groups and the differentiating properties are found and a numerical taxonomic description scheme created. The role of a functional nomenclature is discussed. In chapter eight it is concluded that it is possible to harmonize soil information and to create a taxonomic scheme.

Introducción El sistema Bethesda se utiliza para clasificar los resultados de la prueba de Papanicolaou. Entre los resultados se tienen a las células ASC-US (células escamosas atípicas de significancia indeterminada) y ASC-H (células escamosas atípicas que no pueden descartar lesión intraepitelial). Se ha relacionado a estas células con neoplasia de cuello uterino; sin embargo, su asociación no ha sido muy estudiada en Perú. Objetivos Calcular la probabilidad de displasia severa, carcinoma in situ y cáncer de células escamosas, en mujeres con hallazgo citológico de ASC-H y ASC-US. Métodos Estudio observacional, transversal realizado en un hospital nivel IV en Lima-Perú, entre el 2012 y 2013, en el cual se evaluaron a 1060 pacientes con resultado de la prueba de Papanicolaou, Se realizó análisis bivariado y regresión logística binomial múltiple para evaluar la asociación. Se usó Chi cuadrado y se consideró el valor de p <0,05 como significativo. Resultados Se analizaron 1060 pruebas de Papanicolaou. La edad media fue 46,4 (DE: 13,2) años y 64,9% fueron menores de 50 años. La prevalencia de ASC-US fue 3,45% y la de ASC-H 0,32%. De 175 biopsias, el 22,9% presentó displasia moderada a severa o carcinoma escamoso. Se encontró asociación entre el hallazgo de ASC- H y la presencia de displasia moderada/severa (NIC 2/3), carcinoma in situ y carcinoma de células escamosas, en el estudio histopatológico (PR=2,39; IC(95%):1,49-3,81). Conclusión Dada la asociación encontrada, las mujeres con hallazgo de ASC-H, deben ser estudiadas de manera más profunda, con el fin de buscar displasia moderada/severa (NIC 2/3) o carcinoma de células escamosas.
Background The Bethesda system is used to classify Pap smear results. Some of the results are ASC-US (atypical squamous cells of undetermined significance) and ASC- H cells (atypical squamous cells cannot exclude intraepithelial lesion). It has been found an association between these results with cervical neoplasia; however they have not been well studied in Peru. Objectives To calculate the probability of severe dysplasia in situ or squamous cell cancer in women with cytology results of ASC-H and ASC-US. Methods An observational, cross-sectional study was carried out in a reference higher complexity health establishment in Lima, Peru (4th level), from 2012 to 2013, in which 1060 data of patients was analyzed. Bivariate and multiple binomial logistic regression analysis were performed. Chi square test was used and p-value < 0.05 was considered as significant. Results 1060 Pap smears were evaluated. Mean age was 46,4 (SD: 13,2) years and 64,9% were under 50 years old. ASC-US prevalence was 3,45% and ASC-H 0,32%. Out of 175 biopsies, 22,9% had moderate to severe dysplasia, carcinoma in situ or squamous carcinoma. There was an association between ASC-H finding and the presence of moderate/severe dysplasia (CIN 2/3), carcinoma in situ and squamous cell carcinoma (PR=2,39; CI(95%):1,49-3,81). Conclusion Given the association found, women with ASC-H findings should be further and thoroughly studied in order to look for moderate/severe dysplasia (CIN2/3) or squamous cell carcinoma.

La niche des cellules souches hématopoïétiques représente un compartiment complexe et radiosensible. Sa protection est nécessaire pour la restauration de l'hématopoïèse faisant suite à la myélosuppression due à l'exposition aux rayonnements ionisants. Nous avons dans un premier temps étudié l'effet des RI sur les progéniteurs endothéliaux et mésenchymateux de la niche par une étude de radiosensiblilité et une étude d'évaluation de la mort cellulaire. Nous avons proposé par la suite une stratégie innovante de thérapie génique basée sur la sécrétion locale et à court terme du morphogène Sonic hedgehog visant à favoriser la réparation de niche vasculaire et de stimuler les cellules souches hématopoïétiques et les cellules progénitrices résiduelles. Nous avons étudié la réponse hématopoïétique des singes irradiés à 8-Gy gamma après une seule injection intra-osseuse de cellules souches mésenchymateuses xénogéniques, multipotentes et d'origine adipocytaire transfectées avec un plasmide pIRES2-eGFP codant la protéine Shh. La durée de thrombocytopénie et celle de neutropénie ont été significativement réduites chez les animaux greffés et les clonogènes sont normalisés à partir du 42e jour. Les aires sous la courbe des numération des plaquettes et des neutrophiles entre 0 et 30 jours ont été significativement plus élevée chez les animaux traités que chez les témoins. La greffe d'explants de MatrigelTM colonisés ou non avec des ASC chez des souris immunodéprimées a démontré une activité pro-angiogénique notable des ASC transfectées avec le plasmide Shh . Le suivi à long terme (180 à 300 jours) a confirmé une reconstitution durable dans les quatre singes greffés. Globalement cette étude suggère que la greffe de cellules souches multipotentes Shh-peut représenter une nouvelle stratégie pour la prise en charge des dommages radio-induits de la niche
The hematopoietic stem cell niche represents a complex radiosensitive compartment whose protection is required for recovery from radiation-induced myelosuppression. We initially studied RI effects on endothelial and mesenchymal progenitors by an evaluating radiosensitivity and cell death. Then, we have proposed a new gene therapy strategy based on local and short term secretion of Sonic hedgehog morphogene to favour vascular niche repair and to stimulate residual hematopoietic stem and progenitor cells. We investigated the hematopoietic response of 8-Gy gamma irradiated monkeys to a single intra-osseous injection of xenogeneic multipotent mesenchymal stem cells transduced with a Shh pIRES2 plasmid. Thrombocytopenia and neutropenia duration were significantly reduced in grafted animals and clonogenics normalized from day 42. Areas under the curve of PLTs and ANCs between day 0 and day 30 were significantly higher in treated animals than in controls. Grafting MatrigelTM colonized or not with ASC in immunocompromized mice demonstrated a notable pro-angiogenic activity for Shh-ASC. Long term follow up (180-300 days) confirmed a durable recovery in the four grafted monkeys. Globally this study suggests that grafting Shh-multipotent stem cells may represent a new strategy to cure radiation-induced niche damage

Abstract:
Acute myocarditis is an acute inflammatory syndrome characterized by myocardial damage and dysfunction often due to a viral infection followed by a variable development over time. There are currently no specific treatments and standard treatments for heart failure are generally applied. The inflammasome is a recently identified macromolecular structure that occupies a central role in the amplification of the inflammatory response and promotion of cell death during acute and chronic infections. We hypothesized the formation of the inflammasome in acute myocarditis. To investigate, samples of patients were collected from the Cardiomyopathy Registry in Trieste, with 12 cases of biopsy-proven myocarditis and 11 cases of autopsy-proven myocarditis stained for major components of the inflammasome through immunofluorescence; 10 of the 12 (83.3%) biopsy cases and 8 of the 11 (72.7%) autopsy cases presented formation of the inflammasome in a variety of cells including resident cells (i.e. cardiomyocytes, endothelial cells, fibroblasts) and infiltrating cells (i.e. leukocytes) while varying in intensity and distribution. Control samples of 5 subjects not presenting with any acute cardiac events showed no formation of the inflammasome. While further studies should look to elucidate the correlation of inflammasome-formation and progression of disease, this finding paves the way for further insight into the pathophysiology of acute myocarditis.

Objetivou-se avaliar as altera??es na espermatog?nese provocadas pela Distrofia Muscular de Duchenne (DMD) e o efeito do tratamento com ?cido asc?rbico na preven??o dessas inj?rias. Neste trabalho foram utilizados 24 camundongos, sendo 12 linhagem C57BL/10 (n?o-distr?ficos) e 12 C57BL/10Mdx (distr?ficos) divididos em seis grupos com 4 animais cada (C30 = Controle de 30 dias; D30 = Distr?fico com 30 dias; C60 = Controle com 60 dias; D60 = Distr?fico com 60 dias; CS = Controle com 60 dias suplementados com ?cido asc?rbico e; DS60 = Distr?fico com 60 dias suplementados com ?cido asc?rbico. Os animais dos grupos C30 e D30 foram eutanasiados aos trinta dias de idade, enquanto que os animais dos demais grupos aos 60 dias de idade. A suplementa??o com ?cido asc?rbico foi ministrada na ?gua na dosagem de 0,005g/dia durante 30 dias. Ap?s eutanasiados, os test?culos (direito e esquerdo) foram coletados, imediatamente, pesados e seccionados transversalmente, fixados em solu??o de Karnovysky, inclu?dos em resina histol?gica (an?lises morfol?gicas e morfom?tricas), submetidos a an?lise ultraestrutural e submentidos t?cnica de imunohistoqu?mica para caspase-3. Houve aumento significativo no percentual de t?nica pr?pria em D30 em rela??o a C30 e D60. As an?lises ultraestruturais indicaram ind?cios de apoptose mitocondrial das c?lulas de Sertoli que podem reduzir a efici?ncia esperm?tica em CS60 e DS60. Verificou-se maior densidade de volume das c?lulas apopt?ticas postivas para Caspase - 3 em D30 versus C30 e DS60 em rela??o a CS60. . The objective was to evaluate spermatogenesis alterations caused by DMD and the effect of the treatment using ascorbic acid in preventing those injuries. Twenty four mice were used, 12 from the C57BL/10 lineage (non-dystrophic) and 12 from the C57BL/10Mdx (dystrophic). The sample was divided in six groups containing 4 animals each, as: C30 = 30 days control; D30 = Dystrophic with 30 days; C60 = 60 days control; D60 = Distrophic with 60 days; CS60 = 60 days control supplemented with ascorbic acid and DS60 = Dystrophic with 60 days supplemented with ascorbic acid. The ascorbic acid supplementation was given in the water, 0,005 mg/day. After euthanasia, the testicles (right and left) were collected, weighted and cross sectioned. The material was fixed in the Karnovsky solution for 24 hours, included in resin for histological studies (morphological and morphometric analyzes) submitted to ultrastructural analysis and immunohistochemistry for caspase-3. There was a significant increase in the tunica propria percentage in D30 compared to C30 and D60. The ultrastructural analysis showed mitochondrial apoptosis evidence of Sertoli cells that can reduce sperm efficiency in CS60 and DS60. A higher volume density of apoptotic cells postivas to Caspase-3 in C30 and D30 versus DS60 compared to CS60. There was severe hypertrophy of the Leydig cells between D30 and D60. However, with supplementation was observed reversal of this change in DS60. The ultrastructure of Leydig cells to early presence of lipid vesicles was observed in the group pre-pubertal dystrophic (D30). Thus, the DMD affect the organization of the seminiferous tubules and intertubule, however, the ascorbic acid supplementation used for the treatment of DMD has been just enough to reduce the hypertrophy of the Leydig cells.

A hiperalgesia inflamatória é o processo pelo qual ocorre a sensibilização dos neurônios nociceptores aferentes primários por mediadores químicos inflamatórios, gerando assim uma diminuição do limiar nociceptivo e como consequência episódios de dor. Entre os principais mediadores envolvidos com a sensibilizacão das fibras nociceptivas periféricas está a prostaglandina E2 (PGE2), que é liberada como um produto final de uma cascata de mediadores inflamatórios. Dentro desta cascata de liberação hierárquica podemos destacar a interleucina -1? (IL)-1?, uma citocina importante na gênese da dor inflamatória, devido à sua capacidade de induzir a produção da enzima cicloxigenase-2 (COX-2), e consequentemente PGE2. O mecanismo de controle da produção da IL-1 ? envolvem dois passos intracelulares: a indução da expressão de uma forma protêica inativa (a pró-IL-1 ?) e a geração da forma biologicamente ativa (IL-1?) a partir da pró-IL-1 ?. Este último passo envolve a ação de uma cisteína-protease ativada em decorrência de um processo inflamatório, conhecida como Caspase-1, a qual cliva a pró-IL-1? em IL1?. Recentemente, nosso grupo demonstrou que a caspase-1 tem um papel importante na gênese da dor inflamatória, sendo crucial para a geração de IL-1? e consequentemente COX2/PGE2. Porém, não são conhecidos os mecanismos de ativação da caspase-1 na hiperalgesia inflamatória. Sabe-se que a ativação da Caspase-1 e clivagem da pro-IL-1? são dependentes de uma plataforma molecular intracelular denominada inflamassoma. Os principais inflamassomas ativadores de caspase-1 são formados pelas proteínas NLRP3, IPAF (NLRC4) e por sua molécula adaptadora ASC. O objetivo desse trabalho então foi avaliar a participação do inflamassoma na gênese da dor inflamatória. Nós identificamos que as moléculas IPAF e ASC, mas não o NLRP3, participa no desenvolvimento da hiperalgesia inflamatória mecânica e térmica induzida pela carragenina. Observou-se que estas moléculas são cruciais para a ativação da Caspase-1 e, consequentemente, para a produção da IL-1? ativa. Estes resultados evidenciam pela primeira vez um papel importante do inflamassoma no desenvolvimento da hiperalgesia inflamatória.
The inflammatory hyperalgesic is the process by which occurs the sensitization of nociceptors primary afferent neurons by inflammatory chemical mediators, that generating a decreased nociceptive threshold and result in episodes of pain. Among the main of nociceptive mediators involved with sensitization of peripheral nociceptive fibers are prostaglandin E2 (PGE2), which is released as a final product of a cascade of inflammatory mediators. Within this hierarchical cascade of release can highlight interleukin-1? (IL)-1?, a cytokine important in the genesis of inflammatory pain due to their ability to induce the production of the enzyme cyclooxygenase-2 (COX-2) and consequently PGE2. The control mechanism production of intracellular IL-1 ? involved two steps: induction of expression of a protein inactive form (pro-IL-1 ?) and the generation of the biologically active form from pro-IL-1 ? (IL-1?). This last step involves the action of a cysteine protease-activated due to an inflammatory process, known as Caspase-1, which cleaves pro-IL-1? to IL-1?. Recently our group has demonstrated that caspase-1 plays an important role in the genesis of inflammatory pain, crucial for the generation of IL-1? and consequently COX2/PGE2. However, there aren\'t known mechanisms of activation of caspase-1 in inflammatory hyperalgesic. It is known that the activation of Caspase-1 cleavage and pro-IL-1? are dependent on an intracellular molecular platform called inflammassome. The main inflammassome activators of caspase-1 proteins are formed by NLRP3, IPAF (NLRC4) and its adapter molecule ASC. The aim of this study was to evaluate the inflammassome participation in the genesis of inflammatory pain. We have identified molecules IPAF and ASC, but not NLRP3, is participate in the development of mechanical and thermal inflammatory hyperalgesic induced by carrageenan. It was observed that these molecules are crucial for the activation of Caspase-1 and thus for the production of active IL-1?. These results demonstrate for the first time an important role of the inflammassome in the development of inflammatory hyperalgesic.

This study used thematic analysis to investigate the blogs of parents of children diagnosed with an Autism Spectrum Condition, with a particular focus on themes that emerge when parents write about themselves, their child and their family relationships. The first stage of the study involved a broad analysis of the first six months of entries presented within eight blogs. Themes which emerged during this stage included ‘Coping’, ‘Searching for an explanation - Meaning making’ and ‘Sense of belonging vs not belonging’. The second stage of the study involved a deeper analysis of two of the eight blogs, used as case studies, which were selected due to differing on aspects of the themes found during the first stage of analysis. Themes which emerged from the second stage were a superordinate theme of ‘Searching for an explanation - Meaning making’, with subordinate themes of ‘Relationship with ASC’ and ‘Relationship with society’. There were only brief mentions of family relationships within the blogs, except the relationship with the child and it seemed that parents often made sense of their experiences through their relationship with ASC and their relationship with society. Many of the parents who blogged reported changes in their relationship with society due to feeling that others do not understand and a sense of not belonging. It seemed that blogging provided a sense of belonging and a way of being understood for these parents, as well as a source of information to help parents explore ASC and their relationship with ASC. The themes that emerged may provide important information for clinicians and the implications of the research findings are discussed. The limitations of the study are noted and there are suggestions for future research.

Salmonella enterica is an intracellular bacterial pathogen that injects effector proteins into host cells and induces rapid cell death. Mitochondria associated membranes (MAMs) are important contact sites between mitochondria and ER and selectively facilitate Ca²⁺ uptake from ER lumen into mitochondria. In the context of early Salmonella infection of THP-1 cells, a discontinuous sucrose gradient was used in combination with Stable Isotope Labeling by Amino acids in Cell culture (SILAC) to profile organelle proteomes. Protein profiles were generated for >800 mitochondria and ER proteins. I observed unique protein recruitments to MAMs during early Salmonella infection. The inflammasome adaptor protein, ASC is co-purified with MAM markers. SILAC immunoprecipitation (IP) experiments showed that ASC interacts directly with the VDAC/Stress-70 complex at MAMs, suggesting that ASC is specifically recruited to MAMs during Salmonella infection. SILAC IP experiments identified an interaction between Flightless-I and ASC. Flightless-I is an actin binding protein and also interacts with Salmonella flagellin (FliC). siRNA knockdown of Flightless-I and mitofusin 2 (a structural protein of MAMs) both leads to significant increase of IL-1β during Salmonella infection. Simultaneous knockdown of Flightless-I and mitofusin 2 does not have additive effect, suggesting that they are in the same pathway to dampen inflammasome activation. Actin isolation experiments showed that ASC is enriched at actin filaments during Salmonella infection. Therefore, I proposed a model that during early Salmonella infection, FliC interacts with Flightless-I that then interacts with and transports ASC to MAMs via actin filaments. This process specifically sequesters ASC at MAMs to dampen inflammasome activation in the cytosol. Experimental data from ΔfliCΔfljB Salmonella infection of THP-1 cells and inflammasome reconstitution in 293T cells supported this model. This study was the first characterization of MAM protein composition during bacterial infection. Furthermore, enrichment of ASC at MAMs was identified during early Salmonella infection, with Flightless-I interacting with and transporting ASC to MAMs via actin filaments at the presence of Salmonella flagellin. This novel pathway dampened the inflammasome activation during early Salmonella infection. This is the first report of the negative regulation of pyroptosis by Salmonella flagellin.

Este trabalho teve por objetivo avaliar os efeitos do estresse oxidativo identificado pela express?o da cyclooxygenase-2 (COX-2) na morfologia do tecido card?aco de camundongos mdx, bem como os efeitos do ?cido asc?rbico (AA) sobre estes par?metros. Foram utilizados 60 camundongos machos, sendo 30 C57BL/10 e 30 C57BL/10Mdx separados em seis grupos de acordo com a idade e tratamento (n=10/grupo): controle com 30 dias de idade (C30); distr?fico com 30 dias de idade (D30); controle com 60 dias de idade (C60); distr?fico com 60 dias de idade (D60); controle com 60 dias de idade suplementados com AA (CS60); e distr?fico com 60 dias de idade suplementados com AA (DS60). As amostras de tecido card?acos foram corados pelas t?cnicas de Tricr?mico de Masson, Picrosirius red sob luz polarizada e Imunohistoqu?mica da COX-2. Os resultados demonstraram aumento de fibras col?genas em D30 e diminui??o na ?rea ocupada por cardiomi?citos em D60. A express?o da enzima COX-2 foi maior nos grupos C30, D30 e D60 indicando, provavelmente, em C30 um processo de desenvolvimento celular e, em animais distr?ficos, aumento do estresse oxidativo que levaram ? fibrose. A suplementa??o com AA preveniu a perda muscular em DS60 em rela??o ao D60, bem como diminui??o da fibrose e da express?o de COX-2 em DS60, comprovando a a??o protetora do AA no cora??o de camundongos mdx.

Abstract:
Inflammation is the body’s innate immune system responding to harmful stimuli. The body reacts to this stimuli by forming and activating inflammasomes. Inflammasome protein activation plays major roles seen in metabolic and autoimmune disorders, showing the importance of comprehending the processes involved. The NLR protein family is involved in regulating innate immune responses. These types of proteins can sense pathogen-associated molecular patterns as well as damage-associated molecular patterns. Several miRNA families have been known to be regulators of the NLRP3 inflammasome, indicating that an improved understanding of how miRNAs work together to balance the inflammatory response is an area to be focused on. As well as increasing understanding of the miRNA networks and how those can be used to optimize the response of the inflammasome. The aim of this study was to determine the expression levels of the miR-34 family in relation to the NLRP3 inflammasome activation. Using THP-1 cells, mirRNA was isolated from cells taken at different time points after stimulation of the cells with LPS and ATP, followed by performing a two-step RT-qPCR. The Livak method with the RNU48 reference gene was used and indicated potential downregulation of the miR-34 family during NLRP3 inflammasome activation. Further studies should be carried out to confirm these findings.

Introduction It is well known that resident adipose stem/stromal cells (ASCs) are a heterogeneous population of multipotent cells characterized by (a) their ability to adhere to plastic; (b) immunophenotypic expression of certain cell surface markers, while lacking others; and (c) the capacity to differentiate into cells of mesodermal origin including osteocytes, chondrocytes and adipocytes. Adipose derived stromal cells offer great therapeutic potential in multiple medical fields, including, orthopedics, cardiology, oncology and degenerative diseases, to name a few. Combining different disciplines of medicine and engineering, organ and tissue repair can be achieved through tissue engineering and regenerative medicine. Adipose derived stromal cells (ASCs) can be utilized as biological vehicles for vector-based gene delivery systems, since they home to sites of inflammation and infection in vivo. In order to reach the long-term aim of clinical translation of cell-based therapy, preclinical safety and efficacy need to be shown in animal models. This has motivated the development of standardized isolation, characterization and differentiation operating procedures as well as an in vivo tracking system for ASCs and lentiviral vector transduction for a vector-based gene delivery system. Methodology Human ASCs were isolated from lipoaspirate, expanded in culture, immunophenotyped using flow cytometery and induced to differentiate into adipogenic, osteogenic and chondrogenic lineages. Tri-lineage differentiation was confirmed by microscopy. The ASCs were then transduced with green fluorescent protein (GFP)-expressing lentiviral vectors in vitro. The effect of the GFP lentiviral vector on ASCs was investigated by studying ASC immunophenotypic expression of surface markers as well as their capacity to differentiate into osteocytes, chondrocytes and adipocytes. Results The isolated and expanded cell population, from harvested lipoaspirate adhered to recommended ASC identity criteria. The heterogeneity of ASCs was confirmed by the presence of sub-populations. Transduction efficiency in ASC cultures of approximately 80% was observed after introducing a total of 300 μl of concentrated lentiviral vector suspension per 4.8 x 104 cells. No immunophenotypic differences were observed between GFP positive and GFP negative cultures. Flow cytometric analysis revealed a progressive increase in GFP expression following in vitro expansion of transduced ASCs. Both non-transduced and transduced cultures successfully differentiated into osteocytes, chondrocytes and adipocytes. Conclusion The isolated and expanded cell population conformed to the recommended characterization criteria. Heterogeneity was demonstrated with the identification of immunophenotypic sub-populations and semi-quantification of adipogenesis was performed. ASCs were efficiently transduced using the GFP lentiviral vectors produced in our facility. In addition, transduced ASCs maintained adherence to plastic, ASC immunophenotype and were able to differentiate successfully into cells of the three lineages of mesodermal origin. This optimized GFP-ASC transduction technique offers a feasible tracking system as well as a vector-based gene delivery system for future preclinical studies.

Bakgrund och problem: En av de konvergerade redovisningsstandarder som finns i IFRS och US GAAP är standarden om rörelseförvärv av bolag. Enligt regelverket får inte goodwillposten innehålla identifierbara materiella eller immateriella tillgångar och den ska endast utgöra en minoritet i förvärvsanalysen. Trots standardiseringar kvarstår skillnader mellan förvärv redovisas i Sverige och USA. Enligt tidigare forskning i Sverige utgör goodwill fortfarande en majoritet i köpeskillingen vilket går emot IFRS rekommendationer. En studie av bolag i USA görs för att jämföra med den svenska undersökningen. Syfte: Syftet med studien är att undersöka fördelningen av köpeskillingen vid rörelseförvärv i amerikanska bolag under år 2013. Detta för att se hur fördelningen av köpeskillingen skiljer sig mellan de amerikanska bolagen och tidigare studie av svenska bolag. Metod: Studien utgår från en kvantitativ metod där 80 stycken bolag undersöks. Urval har gjorts från de bolagen med störst börsvärde på Nasdaq. Datan om rörelseförvärv kommer från årsredovisningar från kalenderåret 2013. En kvalitativ forskningsstrategi används sedan med komparativ inriktning för att besvara syftet med studien. Författarna ämnar att tolka och hitta ett mönster för hur fördelningen av köpeskilling skiljer sig. Resultat: Resultatet av undersökningen visar att vissa skillnader finns i fördelningen av köpeskillingen i länderna men skillnaderna är inte markanta. Bolag i USA har en högre andel av immateriella tillgångar än bolag i Sverige. Detta kan förklaras med den hårdare kontrollen från SEC i USA. Med 57 % i immateriella tillgångar och 46 % i goodwill av den totala köpeskillingen kan slutsatsen dras att amerikanska bolag följer rekommendationerna men det är fortfarande en ganska hög andel i goodwill.

The increasing prevalence of Autism Spectrum Conditions (ASC) has led to a national response within the UK to implement ways of supporting children and families affected by these conditions. WIthin the field of education, current legislation has identified a need to consider how to incorporate parental views into policy and practice. Key transitions can be particularly stressful for children affected by ASC and their fam~s~owever, little research has explored the first major transition within the education system for children with ASC: starting school. This mixed methods sequential study explored parental experiences and perspectives of their child's transition from nursery to primary school. Based within one local authority, purposive sampling was used to select parents of children in their first year of formal school (reception year). In phase one, five parents shared their views through semi-structured interviews. The data were analysed using Interpretative Phenomenological Analysis (IPA), and three overarching themes were identified: 'being a parent of a child with ASC', 'relationships with professionals', and 'importance of early identification'. Phase two involved using the overarching theme 'relationships with professionals' to create a questionnaire consisting of a semantic differential scale. The questionnaire was distributed to more parents, and descriptive statistics were used to analyse responses. The quantitative results indicated that the majority of parents had overall positive transition experiences. A key implication of this study is for professionals to incorporate parental views into transition policies and practice for children with ASC. The questionnaire can be used in the future to evaluate practice, and measure parental satisfaction following the transition. Limitations of this study and recommendations for future studies are discussed.

A partir de 1 de janeiro de 2019, as novas normas contabilísticas relativas aos contratos de locação do IASB (IFRS 16) e do FASB (ASC 842) exigiram que praticamente todas as locações fossem reconhecidas no balanço, como ativo (direito de uso) e passivo (dever de pagamento), sendo previamente testados e classificados como operacionais ou financeiros. A Sistemas McDonald's Portugal, Lda., reporta com base em dois normativos: o local (SNC) e o americano (US GAAP). Como tal, foi obrigada a alterar a forma de contabilização dos seus contratos de locação neste último normativo. Este projeto teve como objetivo entender quais as principais alterações ao nível contabilístico e de processos, como consequência da adoção da norma ASC 842, bem como analisar o impacto da mesma nas demonstrações financeiras da empresa no primeiro semestre de 2019. A empresa teve a necessidade de ajustar os processos existentes para corresponder às exigências da nova norma. A aplicação desta norma teve um grande impacto quer no balanço, onde ocorreu um grande incremento do ativo e do passivo, quer na demonstração de resultados onde a empresa registou um gasto superior com as locações, ao contrário do que aconteceria se se regesse pelo normativo anterior. Também os rácios de Autonomia Financeira e Rentabilidade Operacional dos Ativos foram afetados negativamente.
Since January 1st 2019, the new accounting standards for lease agreements from IASB (IFRS 16) and FASB (ASC 842) required all leases to be recognized in the balance sheet as assets (right of use) and liabilities (payment obligation), being previously tested and classified as operational or financial. Sistemas McDonald's Portugal reports based on two accounting standards: local (SNC) and US (US GAAP). As such, it was required to change the way its lease agreements were accounted for in US book (US GAAP). This project aimed to understand the main accounting and process changes, as a result of the adoption of ASC 842, as well as to analyze its impact on the company's financial statements in the first half of 2019. The company had to adjust existing processes to meet the requirements of the new standard. This application had a great impact both on the balance sheet, where there was a large increase in assets and liabilities, and in the income statement where the company recorded a higher expense with leases than in the old standard. Also the Financial Autonomy and Operating Return on Assets ratios were negatively affected.

Prior research has shown that even the most subjective fair value estimates are value-relevant (Song et al. 2010, Kolev 2009, Goh et al. 2009) and that managers appear to use Level 3 valuations opportunistically (Valencia 2011, Fiechter and Meyer 2009). However, the association between “traditional” measures of aggressiveness in financial reporting and biased estimates of fair value has not been previously studied. I test whether aggressiveness, as measured by discretionary accruals, real activities manipulation, and meeting-or-beating analysts’ consensus estimates, is positively associated with realized and unrealized gains and losses on Level instruments. Overall, I find limited support that aggressive firms opportunistically use fair value measurements to overstate earnings. Inferences remain the same whether only the unrealized component of gains/losses are examined and whether firms are classified into “suspect” or “non-suspect” groups.

La niche des cellules souches hématopoïétiques représente un compartiment complexe et radiosensible. Sa protection est nécessaire pour la restauration de l'hématopoïèse faisant suite à la myélosuppression due à l'exposition aux rayonnements ionisants. Nous avons dans un premier temps étudié l'effet des RI sur les progéniteurs endothéliaux et mésenchymateux de la niche par une étude de radiosensiblilité et une étude d'évaluation de la mort cellulaire. Nous avons proposé par la suite une stratégie innovante de thérapie génique basée sur la sécrétion locale et à court terme du morphogène Sonic hedgehog visant à favoriser la réparation de niche vasculaire et de stimuler les cellules souches hématopoïétiques et les cellules progénitrices résiduelles. Nous avons étudié la réponse hématopoïétique des singes irradiés à 8-Gy gamma après une seule injection intra-osseuse de cellules souches mésenchymateuses xénogéniques, multipotentes et d'origine adipocytaire transfectées avec un plasmide pIRES2-eGFP codant la protéine Shh. La durée de thrombocytopénie et celle de neutropénie ont été significativement réduites chez les animaux greffés et les clonogènes sont normalisés à partir du 42e jour. Les aires sous la courbe des numération des plaquettes et des neutrophiles entre 0 et 30 jours ont été significativement plus élevée chez les animaux traités que chez les témoins. La greffe d'explants de MatrigelTM colonisés ou non avec des ASC chez des souris immunodéprimées a démontré une activité pro-angiogénique notable des ASC transfectées avec le plasmide Shh . Le suivi à long terme (180 à 300 jours) a confirmé une reconstitution durable dans les quatre singes greffés. Globalement cette étude suggère que la greffe de cellules souches multipotentes Shh-peut représenter une nouvelle stratégie pour la prise en charge des dommages radio-induits de la niche.

This paper describes the investigation of the centesimal composition, chemical and antioxidant activity of Morinda citrifolia L. (Rubiaceae), native of Southeast Asia, popularly known as noni, which has emerged in Brazil as a plant with great healing potential. The empirical knowledge of the population has meant that the fruit is constantly present in the diet of Maranh?o. Early in the second half of 2010 seedlings of the species were planted in two experimental areas of the Federal Institute of Maranh?o (IFMA), Campus of Z? Doca-MA, broken down by planting time. After three months of the planting in the first area the fruiting process was initiated, and it was possible to perform bromatologic analyzes. For the centesimal composition was found in pulp, seed and mixtures of both, respectively: the ash content (0,82%?0,01; 0,60%?0,01 e 0,79%?0,03), moisture (90%?0,01; 28,34%?0,01; 80,64%?0,05), proteins (4,2%?0,01; 7,47%?0,06; 4,70%?0,02), total lipids (0,34%?0,04; 0,79%?0,03; 0,63%?0,07), carbohydrates (2,68%?0,01; 25,83%?0,02; 13,24%?0,01) and calorific value (30,58kcal/100g?0,02; 140,31 kcal/100g?0,03; 77,43 kcal/100g?0,03). It was found in the pulp and seed values soluble dietary fiber (0,51?0,04; 0,93%?0,03) and insoluble (1,44?0,05; 36,04%?0,38). For the chemical characterization it was found in the pulp, seed and mixtures of both: ? Brix (8,17?0,05;-), the total acidity (0,54?0,02; 0,28?0,01; 0,62?0,02%) and pH (3,95?0,07; 4,5?0,04; 4,1?0,05), the relationship TA / SS pulp was 14.97?0,07. The content of ascorbic acid was calculated by the method Tilmans, pulp and seed fractions (117,33?0,01; 24,33?0,03), expressed in mg/100 g sample. The antioxidant capacity analyzes were performed in three different parts: the pulp, peel and seed using alcoholic extracts (11,63?0,07; 10,03?0,04; 11,19?0,01) and aqueous extract (7,20?0,07; 6,98?0,07; 7,60?0,01) through the sequestration of free radical DPPH, expressed in mM of Trolox / g sample. It was further evaluated the antioxidant capacity of different extracts of the three parts of the fruit and alcoholic extract (98.78% in pulp, 84.76% in peel and 94.96% in the seed) showed much higher antioxidant capacity than the aqueous extract ( 8.08% in the pulp, 5.22% in peel and 22.66% in the seed) and the prominence given to the pulp, followed by seed and peel, respectively. It was demonstrated that the noni fruit grown in the municipality of Z? Doca presented a rich source of nutrition important fact that justifies its inclusion in the diet. It should be noted that the pulp showed a high content of vitamin C and antioxidant activity, and seed a significant amount of insoluble fiber, suggesting that this product can be inserted in the consumer market for being a nutritious food with great herbal medicine potential

Para caracteriza??o qu?mica verificou-se na polpa, semente e mistura de ambos: o ?Brix (8,17?0,05;-), a acidez total (0,54?0,02; 0,28?0,01; 0,62?0,02%) e o pH (3,95?0,07; 4,5?0,04; 4,1?0,05), a rela??o ATT/SS na polpa foi de 14,97?0,07. O Teor de ?cido asc?rbico foi calculado pelo m?todo Tilmans, nas fra??es polpa e semente (117,33?0,01; 24,33?0,03), expressos em mg/100g amostra. As an?lises da capacidade antioxidante foram realizadas em tr?s partes diferentes: polpa, casca e semente utilizando extrato alco?lico (11,63?0,07; 10,03?0,04; 11,19?0,01) e extrato aquoso (7,20?0,07; 6,98?0,07; 7,60?0,01), atrav?s do sequestro do radical livre DPPH, expressos em ?M de Trolox/g amostra. Foi ainda avaliada a capacidade antioxidante dos diferentes extratos, das tr?s partes do fruto e o extrato alco?lico (98,78% na polpa; 84,76% na casca e 94,96% na semente) apresentou capacidade antioxidante bem maior que o extrato aquoso (8,08% na polpa, 5,22% na casca e 22,66% na semente) sendo o destaque dado ? polpa, seguido da semente e da casca, respectivamente. Demonstrou-se que os frutos do noni cultivados no munic?pio de Z? Doca apresentaram uma rica fonte nutricional importante fato que justifica sua inser??o na dieta alimentar. Cabe destacar que a polpa apresentou um elevado teor de vitamina C, e atividade antioxidante, e a semente um relevante teor de fibras insol?veis, sugerindo que esse produto pode ser inserido no mercado consumidor por ser um alimento nutritivo e com grande potencial fitoter?pico.

Abstract:
The global epidemiology of Autistic Spectrum Condition (ASC) is not thought to vary as a function of ethnicity. However, evidence suggests that the identified prevalence of ASC may be inconsistent across ethnic groups in the UK. In the UK, educational psychologists (EPs) often play a key role in the ASC identification process. Given the believed value of accurate identification of ASC to a child’s education, and the importance of providing minority ethnic groups with an equitable service, the following study explores how EPs incorporate ethnic minority cultural factors (EMCF) within ASC assessments. A multiple embedded case analysis was conducted with three EPs. Each was highly proficient in ASC assessment and brought experiences from both different geographical areas, and from service delivery through varied providers; a local authority (LA), a social enterprise, and private practice. Participants’ responses in two semi-structured interviews were recorded and transcribed verbatim. A documentary analysis of the existing LA pathway for the assessment of ASC was completed. This was complemented with a quantitative demographic analysis of data relating to the regions in which each participant EP was working. Interview transcripts were analysed thematically, and findings are presented through thematic maps. Content analysis of the existing policies revealed considerable variation between LAs in how ASC is assessed in school age children. Integration of findings revealed six considerations made in ASC assessments with EMC children, potentially impacting upon their consultations with parents, and their direct work with the child. Further examination suggests that the EP’s work context can influence their considerations through four avenues. It appears that EPs’ understanding of EMCF within ASC assessments is influenced by professional experiences and opportunities within their local context. The study extends understanding how EPs consider EMCF in their assessments for ASC. Findings are discussed with regards to their implications for theory, practice and future research.

Objetivos: Evaluar el riesgo de patología subyacente en las pacientes ASC-US HPV positivas de nuestra área de influencia analizadas por el método COBAS 4800. Se estudió el riesgo de CIN2+ en el momento del diagnóstico de las mujeres ASC-US HPV positivas según el genotipo del virus del papiloma: HPV16, HPV18 ( ambos incluyendo coinfecciones) u otros HPV de alto riesgo (HR-HPV). El objetivo es detectar aquellas mujeres con citologías ASC-US HPV positivo con un mayor riesgo de progresión y por lo tanto tributarias de un seguimiento más exhaustivo. Métodos: Se trata de un estudio retrospectivo de 183 pacientes ASC-US HPV positivas en la citología de cribaje por el método COBAS 4800, sin antecedentes de patología cervical, desde el 1 de junio de 2014 al 31 de julio de 2015. El método COBAS 4800 detecta un pool de 12 genotipos de HR-HPV y separadamente el HPV16 y HPV18.Se realizó colposcopia a todas ellas, y biopsia cervical en caso de que ésta fuera patológica o no satisfactoria. Se analizó el riesgo absoluto y relativo de patología subyacente CIN2+ en el momento del diagnóstico según el genotipo fuera HPV16, HPV18 u otros HR-HPV. Se analizaron éstos riesgos globalmente y posteriormente separados en grupos de edad, por rangos de 10 años, mayor o menor de 30 años y mayor o menor de 40 años. Resultados: Globalmente, el riesgo absoluto de patología subyacente CIN2+ de las pacientes ASC-US HPV16 fue del 9,09%, el de las HPV18+ del 20% y el de las pacientes ASC-US HPVno16no18 del 6.3%. El RR de CIN2+ en el momento del diagnóstico de las pacientes ASCUS-HPV fue de 1,26 para las pacientes HPV16+ y de 0.6 para el grupo de pacientes ASC-US HPVno16no18. En el caso de las pacientes HPV18 fue del 3,05. Analizando el RR de CIN2+ por grupos de edad, en el grupo de ASC-US HPV 18+ éste fue entre un 1,6 y un 7.8, pero sin ser significativo, en cambio en el rango de mujeres menores de 30 años el RR fue de 5,8 y significativo estadísticamente. Conclusiones: Parece existir un mayor riesgo de patología subyacente CIN2+ en las pacientes ASC-US HPV 16 y HPV 18 positivas (con o sin coinfecciones), que en aquéllas con HPVno16no18. Se necesitan estudios con un tamaño muestral más amplio para valorar si estos resultados son significativos y así poder usar el genotipado del HPV como método de tamizaje de las pacientes ASC-US HPV positivas.
Objective: To assess the risk of underlying pathology among ASC-US HPV-positive patients in our catchment area using the COBAS 4800 test. The risk of CIN2+ was analysed at the time of diagnosis of ASC-US HPV-positive women according to papillomavirus genotype: HPV16, HPV18 (both including co-infection) or other high-risk HPV (HR-HPV). The aim is to detect ASC-US HPV-positive women at a higher risk of progression of the disease and therefore requiring a more thorough monitoring. Methodology: A retrospective study of 183 ASC-US HPV-positive patients diagnosed via cytology screening COBAS 4800 with no history of cervical pathology, carried out from June 1, 2014 to July 31, 2015. The COBAS 4800 test detects a pool 12 HR-HPV genotypes and, separately, HPV16 and HPV18. All patients underwent a colposcopy, as well as a cervical biopsy if the former were pathological or unsatisfactory. The absolute and relative risk of underlying pathology CIN2 + was analysed at the time of diagnosis depending on whether the genotype was HPV16, HPV18 or other HR-HPV. These risks were first analysed globally and then by age groups according to 10-year ranges, above or below 30 years-old, and above or below 40 years-old. Results: Overall, the absolute risk of underlying pathology CIN2+ was 9.09% in ASC-US HPV-positive patients, 20% in HPV18+ patients, and 6.3% in ASC-US HPVno16no18 patients. The RR of CIN2+ at the time of diagnosis of ASCUS-HPV patients was 1.26 for HPV16+ patients, 0.6 for ASC-US HPVno16no18 patients, and 3.05 for HPV18 patients. When analysing the RR of CIN2+ by age group, it was between 1.6 and 7.8 in the ASC-US HPV 18+ group, but without being statistically significant, while RR was 5.8 and statistically significant in the range of women under 30 years. Conclusion: There seems to be a higher risk of underlying pathology CIN2+ in ASC-US HPV 16 and HPV 18 positive patients (with or without co-infection) than in those with HPVno16no18. Studies with a larger sample size would be needed to assess whether these results are meaningfull so that the genotyping of HPV could be used as a screening method for ASC-US HPV positive patients.

The human papillomavirus (HPV) is recognized as a major risk factor related to cervical cancer. The identification of HPV types of high risk can aid in the prevention of malignant lesions of the cervix. This study aims to determine and genotype HPV in women with a cytological result of ASC-US and LSIL. This is a cross-sectional study with an analytical component in women attended the Fundação Alfredo da Matta - FUAM. The patients were selected from the file examination of the Laboratory of Cytology of FUAM from January 2009 to July 2011 and requested to attend the DST clinic through active search in order to make a new collection of material for revaluation cytological, molecular detection and genotyping of HPV. Molecular detection was performed by Nested-PCR targeting the L1 region of the viral capsid. PCR products were analyzed on an agarose gel. Out of 100 patients selected, 70% (70/100) participated in the study, 34 of them had the cytological result of ASC-US and LSIL of 36. After reevaluation cytological 8 (11.4%) patients had cytology within normal limits, 33 (47.2%) inflammatory cytology, 22 (31.4%) ASC-US, 6 (8.6%) LSIL, 1 (1.4%) HSIL. HPV was identified in 28.6% (20/70) of the samples. Of the 20 patients HPV DNA-positive 1 had cytology within normal limits, 6 inflammatory cytology, 10 ASC-US and 2 LSIL and 1 HSIL. After the genotyping were identified the following HPV types 6, 16, 58, 61, 70, 83, 84 and 85. The most prevalent HPV 25% was HPV 58. The presence of HPV high-risk oncogenic stresses the importance of actions aimed at preventing the transmission of this virus and tracking of diseases in the city of Manaus.
O Papillomavírus humano (HPV) é reconhecido como o principal fator de risco relacionado do câncer cervical. A identificação de tipos de HPV de alto risco pode auxiliar na prevenção de lesões malignas do colo do útero. Este estudo tem como objetivo determinar e genotipar o HPV em mulheres com resultado citológico de Lesão Intraepitelial de Baixo grau (LSIL) e Células Escamosas Atípicas Significado Indeterminado (ASC-US). Trata-se de um estudo de corte transversal com componente analítico em mulheres atendidas na Fundação Alfredo da Matta - FUAM. As pacientes foram selecionadas a partir do arquivo de exames do Laboratório de Citologia da FUAM no período de janeiro de 2009 a julho de 2011 e solicitadas a comparecer ao ambulatório de DST por meio de busca ativa, a fim de fazer uma nova coleta de material para reavaliação citológica, detecção molecular e genotipagem do HPV. A detecção molecular foi realizada pela técnica de Nested-PCR tendo como alvo a região L1 do capsídeo viral. Os produtos da PCR foram analisados em gel de agarose. Do total de 100 pacientes selecionadas, 70% (70/100) participaram do estudo, sendo que 34 delas tinham resultado citológico de ASC-US e 36 de LSIL. Após reavaliação citológica 8(11,4%) pacientes apresentaram citologia dentro dos limites da normalidade, 33(47,2%) citologia inflamatória, 22(31,4) ASC-US, 6(8,6%) LSIL, 1(1,4%) HSIL. O HPV foi identificado em 28,6% (20/70) das amostras examinadas. Das 20 pacientes HPV-DNA positivas 1 caso apresentou citologia dentro do limite da normalidade, 6 citologia inflamatória, 10 ASC-US, 2 LSIL e 1 caso apresentou HSIL. Após a genotipagem foram identificados os seguintes tipos de HPV: 6, 16, 58, 61, 70, 83, 84 e 85. O HPV mais prevalente com 5 casos em 20 positivos foi o HPV 58. The majority of children with ASC are educated within a mainstream secondary setting. The challenges within the new environment may lead to some children experiencing a breakdown in provision resulting in temporary or permanent exclusions. A gap in the literature highlights a need for research which seeks to understand the views and experiences of year 7 children with ASCs about theirtransition to mainstream secondary school. The research involved four year 7 children with ASC, from across three settings, who had recently transitioned from a mainstream primary school to a mainstream secondary school (without resource based provision). The views of their parents and teachers were also sought. A multiple-embedded case study design was employed involving four cases from across three school settings. This involved utilising a transition Q-sort and a semi-structured interview with four children and semi-structured interviews with six parents and three teachers. The data was analysed using content analysis and thematic analysis (Braun & Clarke, 2006). The findings highlight a need for individualised transition planning for the child with ASC; consideration of co-occurring difficulties and the importance of including the views of the child and parents in transition planning. The findings are discussed in relation to psychological theories and existing literature. The implications for future research, local authority policy, school staff, parents and educational psychologists are considered.

A number of studies have reported that many children with Autism Spectrum Condition (ASC) experience unusual responses to sensory stimuli, however it is argued that there is a lack of conceptual understanding regarding this phenomenon and an underdeveloped evidence base regarding appropriate support for these responses within an educational context. Despite this, practitioners from a range of professions are called upon to offer consultation, advice and intervention. Therefore, the present study sought to qualitatively explore the experiences of these professionals with regards to the sensory needs of children with ASC within an educational setting. An exploratory single-embedded case study design was adopted. Two Specialist Teachers, two Educational Psychologists (specialists in ASC) and two Occupational Therapists were recruited opportunistically and individual semi-structured interviews were conducted with each participant, resulting in six interviews. Each interview was audio recorded and transcribed verbatim. Interview data were analysed using thematic analysis and the findings presented as three thematic maps according to research question. Six organising themes were identified for Research Question 1: conceptualisations, assessment, defining unusual sensory responses, pragmatism, impact at school and professionals’ roles. Three organising themes arose from the data for Research Question 2: interventions, differences between special and mainstream, and efficacy. The data from the interviews yielded two organising themes for Research Question 3: barriers and facilitators. The study extends conceptual understanding by presenting a proposed Interactive Factors Framework (IFF) for unusual sensory responses in children with ASC. A number of recommendations for supporting children with ASC and unusual sensory responses within educational settings are also proposed. Implications for professionals supporting the sensory needs of children with ASC are discussed with reference to the pragmatic issues involved in translating a developing evidence base into practice.

Existing research has started to explore the experiences of the people around children and adults diagnosed with Asperger's Syndrome (AS) and High Functioning Autism (HFA). However, there are very few studies looking at the experiences of fathers, and there are less studies in relation to adults rather than children. This study aims to take an in depth look at the experiences of these fathers in order to start building our knowledge of the experiences of being the father of someone with a diagnosis of AS or HFA. Four fathers of sons and three fathers of daughters with a diagnosis of AS or HFA were interviewed using a semi-structured interview. The experiences of the fathers were explored qualitatively, using Interpretative Phenomenological Analysis. Analysis of the transcripts brought out four superordinate themes for the seven fathers. The first theme: 'The changing role but constant responsibility of being a "Father"' described the fathers' perceptions of their roles and responsibilities towards their child as they have grown up and into the future. The second theme: 'The importance of knowledge and understanding' described the importance the fathers placed on developing their understanding of their child and the need for other people to understand them and their child. The third theme: 'The intense emotional impact' came from the fathers' descriptions of the positive and negative emotional reactions to the challenges their children and families have faced. The final theme: 'The importance of good support' includes the fathers' good and bad experiences of support services and support from family and friends. The results of the analysis are discussed in relation to the existing literature and in terms of what they mean for the clinical practice of people working with fathers of children and adults with AS or HFA. The strengths and limitations of the study are also discussed.

Children on the autism spectrum commonly display a reading profile characterised by strengths in decoding alongside weaknesses in reading comprehension (Nation et al., 2006). Reciprocal Teaching (RT; Palinscar & Brown, 1984) is an evidence-based instructional approach for supporting reading comprehension skills based on cooperative learning principles and endorsed by National Reading Panel (NRP; NICHD, 2000) research; however there is little evidence around the use of RT with children with Autism Spectrum Condition (ASC). Using an action research methodology, I sought to develop my knowledge as a practitioner by exploring how I could make adjustments within the context of delivering a group-based RT intervention to enhance its application for two children (aged 8-9) with diagnoses of Asperger’s Syndrome. A key feature of the research was eliciting the views of participating children and using these to inform the ongoing planning and delivery of the intervention. Qualitative data including feedback from participants, session records and a bespoke assessment of RT strategy-use (alongside my own reflective records) contributed to two cycles of action research in which my learning informed my subsequent actions. Within smaller micro-cycles of action and reflection, I made four adjustments to RT involving visual aids to activate children’s prior knowledge and support them to ask questions about text and summarise non-fiction passages. I discuss my findings with reference to theoretical models of comprehension and ASC and generate my own living theory of practice. The study addresses a gap in the literature and has direct implications for educational professionals and for the practice of Educational Psychologists (EPs) who frequently support children with ASC but often do not feel skilled in supporting reading comprehension (Greenway, 2002). Throughout my inquiry, I highlight questions for further research and future practice.

Le dysfonctionnement de l'exocytose des granules cytotoxiques est responsable d'une susceptibilité accrue aux pathogènes intracellulaires qui s'accompagne de l'activation continue et anarchique des lymphocytes cytotoxiques et des macrophages. Ce phénomène conduit à la lymphohistiocytose hémophagocytique (HLH), un syndrome auto-inflammatoire fatal en absence d'intervention thérapeutique. Les mutations des gènes codant pour la perforine (PRF-1) ou pour certaines des protéines impliquées dans la biogénèse ou le transport vésiculaire des granules cytotoxiques sont causales des formes familiales ou primaires de la HLH (FHL). La HLH fait également partie des complications secondaires aux infections à herpesviridae et à certains désordres immunologiques importants tels que l'arthrite juvénile idiopathique (SoJIA). Au moment d'entreprendre les travaux présentés dans ce manuscrit, le premier cas de HLH induite par une mutation menant à l'activation constitutive de la composante NLRC4 de l'inflammasome était décrit. L'inflammasome est une structure multimérique composée d'un récepteur cytosolique, de la protéine échafaud ASC et de la Caspase-1. Son activation mène à la maturation des pro-formes de l'IL-1β et l'IL-18 ainsi qu'à leur sécrétion. L'activation constitutive de NLRC4 étant suffisante au déclenchement de la HLH, nous avons tenté de comprendre si cette structure y était essentielle dans le cadre des défauts génétiques de cytotoxicité. Nous avons donc invalidé la protéine ASC ou Caspase-1 dans le modèle murin de HLH déficient pour la perforine (PRF1 -/-). Nous avons également testé l'hypothèse qu'un déficit de cytotoxicité pouvait expliquer le développement de la HLH chez les patients souffrant de SoJIA. Nos résultats montrent que l'inflammasome est nécessaire à la production d'IL-18 lors de la HLH mais qu'il n'est pas essentiel au développement de la maladie dans le cadre des FHL. Par ailleurs, nous montrons que la cytotoxicité des cellules NK semble normale chez les patients atteints de SoJIA, ce qui suggère que les mécanismes immunologiques à l'origine de la HLH dans les FHL et dans les maladies autoinflammatoires comme la SoJIA sont distincts. Dans la seconde partie de ce manuscrit, nous avons étudié sur le rôle de la sérine/thréonine/tyrosine kinase Styk1 dans la régulation des lymphocytes cytotoxiques NK. Ces derniers sont responsables du contrôle immunitaire précoce des pathogènes intracellulaires et contribuent à l'immunosurveillance des cellules tumorales. Suite à leur activation, ils relâchent de très grandes quantités d'IFN-y et de TNF-α, faisant ainsi le lien entre l'immunité innée et adaptative. La reconnaissance des cellules cibles par les lymphocytes NK est gouvernée par l'expression d'un éventail de récepteurs qui transduisent des signaux, activateurs ou inhibiteurs, et dont la balance se traduit par l'activation ou la tolérance. Ces récepteurs sont codés au sein de deux complexes génétiques très denses, le complexe de cytotoxicité naturelle (NCR) et le complexe des récepteurs des leucocytes (LRC). Au moment de commencer ces travaux, nous avions révélé que l'expression de la kinase Styk1 fait partie de la signature transcriptionnelle des lymphocytes NK. Sa fonction dans le système immunitaire demeure toutefois inconnue. Néanmoins, la localisation génétique favorable de Styk1, près du NCR, ainsi que son implication dans la voie PI3K-AKT, en faisaient un candidat potentiel de régulation des lymphocytes NK. Afin de connaître le rôle de Styk1 dans le développement et les fonctions effectrices des lymphocytes NK, nous avons donc généré une souris pour laquelle Styk1 est invalidé. Nos résultats confirment que Styk1 est exprimée de façon spécifique par les cellules NK. Nous avons également détecté une diminution de l'activité constitutive de la voie AKT/mTOR dans les cellules NK, mais le développement, l'homéostasie et la fonction des cellules NK sont cependant normaux dans les souris déficientes en Styk1
Upon recognition of infected or target cells, CD8+ T and Natural Killer (NK) lymphocytes initiate a polarized degranulation of vesicle storing cytotoxic molecules (perforin: PRF1 and granzyme B). By altering the target cell’s cellular membrane integrity, perforine allows granzyme B to translocate to its cytosol. Genetic anomalies may affect normal cytotoxic functions and severely hamper the control of intracellular pathogens. In this context, the pathogenic signal remains uninterrupted and both cytotoxic lymphocytes and macrophages are continuously stimulated. This auto-inflammatory pathological condition is named hemophagocytic lymphohistiocytosis (HLH) and is fatal without therapeutic intervention. HLH can also occur secondary to infection with viruses from the herpesviridae family, or be concomitant to important immune alterations such as systemic onset juvenile idiopathic arthritis (SoJIA), with no clear etiological cause identified. In 2014, a case of HLH mediated by the constitutive activation of the NLRC4 inflammasome receptor was first described. The inflammasome is a multimeric structure involving a cytosolic receptor, a scaffold protein – ASC – and Caspase-1. In the immune system, the inflammasome is expressed in macrophages and dendritic cells and senses pathogenic (PAMP) and danger (DAMP) associated signals. Once activated, inflammasome’s protein Caspase-1 catalyzes the maturation of pro-IL-1b and pro-IL-18 and leads to their secretion. Since NLRC4 constitutive activation appears to be sufficient for triggering HLH, we aimed to understand if the inflammasome structure was essential to the development of the syndrome. In order to address this question, we invalidated the inflammasome through the abrogation of ASC or Caspase-1 in PRF1 -/- HLH mouse model. We also tested the hypothesis that an altered cytotoxic function could explain the high prevalence of HLH in the proinflammatory context of SoJIA. The results we present here show that the inflammasome is responsible for the elevated levels of IL-18 in the serum of HLH patients. However, the inflammasome is facultative for its development. We also demonstrate that in patients suffering from SoJIA, NK cells show normal cytotoxicity, suggesting that immunological mechanisms involved in FHL and secondary HLH are distinct. In the second part of this manuscript, we aim at understanding the role of Styk1 serine/threonine/tyrosine kinase in NK cells’ regulation. NK cells are in charge of eliminating stressed, virally infected or transformed cells. Upon activation, they secrete large amounts of IFN-γ and TNF-α, thus bridging innate and adaptive immunity. Capabilities for recognition of target cells is endowed by the expression of numerous stochastically expressed activating and inhibitory receptors. The balance between activating and inhibitory signal allows for self-tolerance or activation upon engagement of abnormal cells. Activating and inhibitory receptor are germline encoded in two dense, large complexes, the Natural Killer Complex (NKC) and the Leukocyte Receptor Complex (LRC). At the moment of starting this work, we had recently identified that Styk1 was a signature transcript of NK cells. However, its function in NK cells and more generally in the immune system remains unknown. Nevertheless, its genetic localisation near the NKC and its potential implication in the PI3K-AKT pathway prompt that it may play a role in NK cell development and/or functions. In order to evaluate the role of Styk1 in NK cells’ regulation, we generated a mouse model in which its expression is abrogated. Our data confirms that amongst all immune subsets, Styk1 is specifically expressed by NK cells. Styk1 was also able to discriminate NK cells from other ILCs. In this study, we show that Styk1 invalidation lead to a decrease of activity in the AKT/mTOR pathway. However, NK cells development, homeostasis and function were surprisingly normal in Styk1 -/- mice
