Abstract:
The trial and verdict of Rakim Mayers, also known as ASAP Rocky, caused major media attention during the summer of 2019. The aim of this bachelor study was to examine how news journalism represented the case “ASAP Rocky” in Swedish tabloids. The research questions examined were: How does journalism portray the story of the victim and perpetrator in the case “ASAP Rocky” ? What are the differences and similarities in how victim and perpetrator are constructed? How does the story develop in Swedish tabloids during the case “ASAP Rocky”?    We used Fairclough’s critical discourse analysis (CDA) to identify reoccurring discourses in the description of the case “ASAP Rocky”. By examining the characters, victim and perpetrator in Swedish tabloids, we found that there was no idealistic role portrayed in the case. Both the victim and the perpetrator were described as insufficient. One discursive theme was that the victim is inadequate since ASAP Rocky´s status as a celebrity provides him an advantage. This discourse is identified also in texts describing ASAP Rocky as a perpetrator. The study showed that the different discourses concern moral values in various contradictions, such as good - evil, right - wrong and “we” versus “them”.    Our study shows that the discourses we found contributes to the creation of celebrities and the discourse that celebrities are unreachable. The conclusion of this study is that media contributes to the stereotype of celebrities.

Abstract:
This thesis deals with the problematice of measuring membrane potential and monitoring the propagation of electrical activity of cells. For this purpose, fluorescence membrane voltage sensors have been developed to detect changes in the membrane potential by changing their fluorescence intensity. The practical part is focused on the study of the properties of the ASAP1 fluorescence probe, which was transfected into the HEK293 cell line, which are kidney cells from the human embryo. Cell membrane potential was changed using the patch-clamp technique.

Abstract:
The Arizona Student Assessment Program (ASAP) is a major piece of legislation for Arizona, reducing norm-referenced standardized testing, providing performance-based assessments matching curriculum, requiring district articulation with state curriculum frameworks and assessments, collecting contextual information from districts, and producing complete profiles of schools, districts and the state. In its first year of implementation, the ASAP is appropriately examined through policy analysis rather than through an evaluation study. Six criteria for educational policy analysis developed by Mitchell (1986) were validated and used as interview questions with seven interviewees knowledgeable about the ASAP. Results of the interviews suggest the degree to which the ASAP is good educational policy and likely to make a difference in Arizona. Interviewees indicated that the ASAP is democratic, providing for both the needs of legitimate stakeholders and the general public interest. It recognizes and supports the organizational integrity of schools only if schools have begun to make some reform efforts of their own in the direction of the ASAP. The ASAP provides adequate means-end linkage for the first two years of implementation, including through school, district, and state profiles, but may need to provide additional help to districts during the first two years; furthermore, relief incentives may be needed, rather than sanctions or disincentives, to encourage continued implementation. The ASAP may not be integrated into overall state educational policy, primarily because there has been no unifying state policy until the ASAP. The ASAP may emerge as a force to reorient current and unify future policy. The ASAP will be expensive, but the interviewees felt the short and long-term benefits justify cost. The ASAP was the most politically feasible policy available to bring about the changes needed, but perhaps not the most palatable, especially to districts that have made no reform efforts of their own. Policy analysis using different criteria and evaluation studies are recommended.

Abstract:
Au cours de la division cellulaire, la mise en place d’un fuseau bipolaire correct est nécessaire au maintien de la stabilité génomique, en permettant une ségrégation égale des chromosomes dans les deux cellules filles. Les microtubules, éléments constitués d’un assemblage de tubuline sous une forme cylindrique et creuse, sont les constituants de base de ce fuseau mitotique. Lors de l’assemblage du fuseau, les centrosomes se séparent, et des changements surviennent au niveau de la dynamique des microtubules. Cependant, sans les propriétés fonctionnelles d’autres protéines venant réguler cette dynamique, les microtubules ne seraient pas capables d’accomplir seuls les tâches qui leurs sont attribuées. Les protéines responsables de cette régulation sont les Protéines associées aux Microtubules ou MAPs. Des variations de la stabilité et de la dynamique des microtubules sont susceptibles d’atteindre la formation et l’organisation du fuseau mitotique ainsi que l’attachement des chromosomes, et peuvent provoquer des problèmes d’ instabilité génétique et la cancérogenèse. Nous avons identifié au laboratoire une nouvelle protéine humaine associée aux microtubules que nous avons appelé ASAP pour ASter-Associated Protein. La protéine ASAP est associée au fuseau mitotique. Sa surexpression entraîne des mitoses aberrantes et abortives (fuseaux multipolaires et cellules multinucléées avec une amplification des centrosomes, fuseaux monopolaires avec non séparation/migration des centrosomes). Ces phénotypes sont fréquemment observés dans des cancers. Sa déplétion provoque des fuseaux anormaux, retarde la mitose en entraînant des défauts de congression et de ségrégation des chromosomes et aboutit à des cytokinèses abortives ou à la mort cellulaire. ASAP est donc une nouvelle MAP jouant un rôle crucial au cours du cycle cellulaire. Une étude plus ciblée sur son implication dans la régulation de la dynamique et de l’organisation des microtubules a ensuite été entreprise afin de mieux comprendre l’impact d’ASAP au niveau du réseau de microtubules. Nous avons ainsi pu mettre en évidence une implication étendue de la protéine, tant au niveau des différentes phases dynamiques de la polymérisation, qu’au niveau de l’assemblage et de l’organisation des microtubules. Nous avons également clairement mis en évidence la présence d’ASAP aux centrosomes, avec la possibilité pour cette protéine de participer à la nucléation des microtubules, un rôle qu’il reste encore cependant à démontrer in vivo.

Abstract:
In the beginning of this thesis is a short introduction into plasmid DNA which is in the form of a vector used in molecular biology. Plasmids can be used in the form of fluorescent probes to measure changes in membrane potential. Into their structure is added a dye called fluorophore. As an important representative of this thesis is a fluorescent probe ASAP1 which contains green fluorescent protein whose response to the membrane potential change is the decrease in the intensity of emitted light. The aim of this thesis was to make chemical transfection of this plasmid into the HEK293 cell line and carry out its characterization. In the work is also described the design of a method for the analysis of the time course of changes in fluorescence depending on the cell membrane depolarisation. In the end of this thesis is also desribed realized experiment including the discussion of aquired results.

In recent years – and especially since the beginning of the COVID-19 pandemic – online shopping has become a part of everyday life for many people. Yet, in contrast to buying at a traditional retail store, staying anonymous is at least difficult if not impossible when shopping online – in particular, when physical goods are to be delivered. From the customer perspective, reasons for seeking anonymity when shopping online can be manifold, for example some do not want anyone to know about their purchases, others do not want their data to be used by Big Data-enabled online retailers. From the point of view of online retailers, the prospect of anonymous online shopping should therefore not only be seen as a threat to their data-driven business models, but also as an opportunity to attract new customers. In this thesis we search and find support in the literature regarding the question whether there is indeed a demand for anonymous online shopping, and we discuss system architecture designs that were proposed by other authors for potentially realizing what we call All-Steps Anonymous Purchase Platforms (ASAPP). We propose a new architecture design that improves earlier work by realizing the concept of Separation of Data within a single platform: GhostBuy. We implement a working prototype of this platform that demonstrates not only the fundamental feasibility of the architecture but also that such a platform can be realized with a look-and-feel similar to that of common online shops. We also propose solutions for certain related aspects that are particularly important in the context of such a platform, as for example a guaranteed use of secure user passwords or application-level database encryption. We evaluate to what extent the proposed architecture and prototype preserve the customers’ anonymity/privacy, showing that the prototype provides it to the maximum possible extent that can be achieved based on the proposed architecture. We also show that the system provides 256-bit security against all but one considered cryptographic and mis-authentication attack vectors and discuss how this can also be achieved for the remaining attack vector. Closing our evaluation, we show how well the platform could presumably be deployed in the real world. Finally, limitations, possible improvements, and potential further future work are discussed and proposed.

Nel presente elaborato verrà descritto ed analizzato un caso reale di implementazione del flusso di acquisti di un’università privata di Milano nel sistema Enterprise Resource Planning (ERP) SAP. Il progetto ha l’obiettivo di gestire a sistema gli step approvativi di cui si compone il flusso di acquisto eliminando l’uso di supporti cartacei. L’utilizzo dei sistemi ERP porta numerosi vantaggi per le aziende sia per quanto riguarda la gestione interna, sia per la gestione dell’intera supply chain, in particolare in riferimento alla trasmissione e alla congruenza delle informazioni. Il progetto di dematerializzazione del flusso di acquisti in oggetto è stato seguito all’interno della società di consulenza Deloitte & Touche. La dematerializzazione permette una riduzione dell’impatto ambientale delle attività aziendali, ma soprattutto un maggior controllo su queste ultime e una migliore organizzazione interna. Sono stati calcolati gli impatti dal punto di vista ambientale e si è riscontrato un miglioramento molto lieve, ma rispetto alla situazione AS IS si evidenzia un miglioramento in termini di standardizzazione delle pratiche, gestione dell’archiviazione e gestione della reportistica.

These core modules are needed for invoicing tasks and most classical day-to-day scenarios: * IS-U BF Basic Functions * IS-U MD Master Data * IS-U DM Device Management * IS-U BI-IN Billing Invoicing * FI-CA Contract Accounts Especially for large customers in electricity and gas industry, IS-U EDM Energy Data Management is used for management of interval reading consumption and profiles. As utility industry evolves through market liberalization, unbundling processes are covered by IS-U IDE Intercompany Data Exchange (with PI Process Integration as and integration platform). Nos travaux se positionnement sur le supportinformatisé de ces processus de développement qui sont actuellement gérés par les systèmes PLM.L’objectif d’un support informatisé est d’accélérer le processus en automatisant la notification et ladiffusion des informations. Il permet également de garder trace des opérations et décisions effectuéeset d’accroître la standardisation des processus. Face à la rigidité des solutions actuellement proposéespour gérer les processus vis-à-vis des modifications survenant dans le processus, notre objectif est deproposer une approche permettant de modifier un processus en cours d’exécution sans devoir leredéfinir et le relancer dans son ensemble. Pour y parvenir, nous avons proposé, une approche quidécline une orientation services inspirée des architectures orientées services (SOA). Ces architecturespermettent de définir des applications modulaires, en utilisant des services faiblement couplés. Notreobjectif est de décliner une telle architecture exploitée essentiellement pour les systèmes logiciels et leWeb, au niveau métier de l’entreprise afin de modéliser et d’exécuter de manière flexible desprocessus de conception de produits par composition de services réutilisables. Nous proposons unedémarche d'identification des services du domaine métier des processus de conception produit et dudomaine fonctionnel du PLM. Ces services sont organisés dans deux catalogues de services métiers etfonctionnels. To cope with market dynamic and shortened time to market, industrial companies need toimplement an effective management of their design processes (DPs) and product information.Unfortunately, Product Lifecycle Management (PLM) systems which are dedicated to support designactivities are not efficient as it might be expected. Indeed, DPs are changing, emergent and nondeterministic, due to the business environment under which they are carried out. The aim of this workis to propose an alternative approach for flexible process support within PLM systems to facilitate thecoupling with the environment reality. The purpose of a support system is to accelerate the process byautomating the notification and dispatching of information and activities between actors. It also allowsto keep track of transactions and decisions made and to increase processes standardization. Our goal isto propose a solution which allows process change at run-time without having to redefine and restartthe whole of process activities. To achieve this, we proposed an approach based on service-orientedarchitectures (SOA). These architectures allow defining modular applications, using loosly coopledservices. They are mainly exploited for software systems and Web development. Our goal is to declinesuch architectures at the business level of a company in order to perform flexible DPs deploymentbased on services reuse and composition. We propose an identification approach for business levelservices (product design services) and functional PLM services. These services are organized in twocatalogs of business and functional services. Le laboratoire a caractérisé une nouvelle protéine associée aux microtubules (MAP) interphasique et au fuseau mitotique, appelée ASAP (ASter Associated Protein) ou MAP9. La dérégulation de son expression entraîne de sévères défauts mitotiques : fuseaux anormaux, retard de la progression mitotique avec des défauts de congression et de ségrégation des chromosomes aboutissant à une cytokinèse défectueuse, des cellules aneuploïdes et à la mort cellulaire. ASAP est donc nécessaire à l’assemblage du fuseau et au bon déroulement de la mitose. Les mécanismes contrôlant la mitose et l’assemblage du fuseau sont très finement régulés par phosphorylation assurées par plusieurs familles de kinases parmi lesquelles la kinase Aurora-A qui recrute et phosphoryle de nombreuses MAPs. Mon projet principal de recherche consistait à déterminer le lien fonctionnel entre ASAP et Aurora-A. J’ai montré qu’ASAP était phosphorylée par Aurora-A, caractérisé le site majeur de cette phosphorylation in vivo sur la sérine 625 et montré que cette phosphorylation était essentielle à l’assemblage du fuseau mitotique et à la progression correcte de la mitose. Par ailleurs, la déplétion d’Aurora-A induit la dégradation d’ASAP par le protéasome suggérant que cette interaction et/ou phosphorylation est nécessaire à la stabilisation d’ASAP. Ce travail a ainsi permis de caractériser le premier partenaire d’ASAP, de confirmer son rôle crucial au cours de la mitose et d’en déterminer les premiers mécanismes moléculaires. Enfin, la caractérisation de l’orthologue de souris et de son expression tissulaire m’a permis de valider le modèle murin comme modèle d’étude de différentes fonctions biologiques d’ASAP.

Dabei werden als Grundlage des Konzepts die Standards der Workflow Management Coalition zur Beschreibung von Workflows und Workflow Management Systemen verwendet. Für die verteilte Abarbeitung von Prozessen wurde eine RMI-basierte Abwandlung des ASAP-Protokolls implementiert. Die Implementierung nutzt Enhydra Shark als Laufzeitumgebung und die Schnittstellen für die verteilte Abarbeitung sind für dieses System angepasst. Anhand von Beispielen und Testfällen wird die Funktionsweise des erstellten Konzepts und seiner Realisierung verdeutlicht.

La fabrication et le trafic illicite d’explosifs ont pour conséquence l’usage croissant d’engins explosifs lors d’attentats. L'identification d'explosifs est un enjeu majeur pour la lutte contre le terrorisme. La grande variété de formulations énergétiques rend cependant complexe leur caractérisation par une seule technique. Cette thèse s’est focalisé sur la spectrométrie de masse en cherchant à améliorer la détection de molécules explosives incluses dans des formulations énergétiques et en investiguant les processus mis en œuvre dans la technique de l’attachement d’anion permettant l’amélioration de la sensibilité de détection. Il apparaît que l’attachement de chlorure et de nitrate est la technique la plus appropriée à produire un signal intense. Une hypothèse a été avancée en se basant sur des expériences de CID et d’IRMPD. Le travail a aussi porté sur la caractérisation de la matrice environnant les explosifs pour créer une base de données. Ces analyses sont réalisées à l’aide des sources ASAP et DART, pour limiter les pertes de matière, avec en perspectives le travail sur des échantillons à l’état de traces. Ces méthodes ont montré leur capacité à produire des spectres de masse pour des polymères fluorés difficilement analysables. Afin d’obtenir une empreinte spectrale des polymères et de pouvoir comparer les spectres de ces polymères, une méthode d’analyse des défauts de masse de Kendrick a été développée et a permis d’identifier les polymères dans un mélange. The manufacture and the illicit traffic of explosives result in the increasing use of explosive devices during attacks. The identification of explosives is a major issue for the fight against terrorism. However, the wide variety of energetic formulations make their characterization complex by a single technique. This thesis focused on mass spectrometry by seeking to improve the detection of explosive molecules embedded in energetic formulations and by investigating the processes involved in the technique of anion attachment used to improve the sensitivity of detection. It appears that chloride and nitrate attachment is the most appropriate technique to produce an intense signal. A hypothesis has been advanced based on CID and IRMPD experiments. The work also focused on characterizing the matrix surrounding the explosives to create a database. These analyzes are performed using ASAP and DART sources, to limit material losses, with the prospect of working on samples in trace amounts. These methods have shown their ability to produce mass spectra for fluorinated polymers that are difficult to analyze. In order to obtain a spectral fingerprint for the polymers and to be able to compare the spectra of these polymers, a Kendrick mass defect analysis method was developed and allowed to identify the polymers in a mixture. An analysis was made on real samples of explosives to compare the chemical species present pre and post explosion

This essay deals with estimation methods while implementation of Type application software. After the introductory part, which stipulates objectives of the work and descriptions of how to achieve them, is a general description of basic features of the project. After this part, there are characterized basic attributes of estimates, various approaches to their formation and problems that can accompany estimation activity. This work also describes diversification of information systems according to their way of development on Individual application software and Type application software. Then it defines their mutual differences while implementation. Next chapter resolve the first objective associated with exploration of implementation methodologies used by companies operating on the Czech market in the area of IS / ICT. Part of this chapter is dedicated to the AcceleratedSAP methodology which describes the life cycle of projects. The following chapter deals with the performance of the second objective, which is a comprehensive overview of estimation methods. Some items from this list are later selected for more detailed evaluation, in account of some of them are marked under the third objective as suitable for projects dealing with implementation of Type application software. In the last section of this essay is solved the main given objective, which is to implement some of chosen estimation methods into AcceleratedSAP methodology. Later in this chapter is recorded a reflection on possibilities of using model based estimation methods in area of Type application SW. It is followed with a proposal for possible course of development of such a model. The work ends with a practical example of effort estimation through UCP method adapted for the Type application software environment. This is followed by conclusions evaluating the achievements of set objectives.

The purpose of this study was to evaluate the effectiveness of cooperative learning in a liberal arts mathematics course and to examine any gender-related differences in the effects of cooperative learning in terms of achievement, composition of the cooperative groups, mathematics anxiety, attitudes toward mathematics, attendance, and retention. The quasi-experimental design compared a control section using individualized learning methods with three treatment sections using cooperative learning methods based on the Learning Together model of Johnson and Johnson (1991). The compositions of the three treatment groups varied: heterogeneous ability/heterogeneous gender, heterogeneous ability/homogeneous gender, and self-selected. The Academic Skills Assessment Program (ASAP) was used as a pre-test to show that the groups were equal. The Revised Math Attitude Scale and the Mathematics Anxiety Rating Scale (MARS) were used as pre- and post-tests to measure changes in attitudes and anxiety. Achievement was measured by individual and group course grades. Group grades were determined by taking 67% of a student's individual exam score plus 33% of the group's average on the exam. Cooperative learning and composition of groups had no significant effect on achievement. The differences between individual and group grades were insignificant, and the group grading method benefited the grades of only five students. Attendance had a large effect on achievement, and the ASAP score and the Math Attitude Post-test were significant predictors of achievement.The MARS post-test was negatively correlated with individual course grade. While no significant gender-related differences surfaced, some trends appeared. The ASAP, MARS, and Math Attitude pre-test scores were equal, however, females achieved slightly higher course grades than males. In each of the four research groups the individual course grades were higher for females than for males. Also, females had a larger decrease in mathematics anxiety with a drop of 22 points compared to the males' drop of 10 points. Abstract:
Tuberculosis remains a major public health problem and one of the leading causes of death worldwide. Human genetic factors determine susceptibility to M. tuberculosis (M. tb) infection and predispose to clinical TB. Genome-wide association studies (GWAS) aim to discover human genes associated with susceptibility to TB. Recently, a GWAS conducted by our lab identified a new TB-associated gene ASAP1 that encodes an Arf GTPase-activating protein (GAP). ASAP1 is known to be involved in regulation of actin and membrane remodeling. My Ph.D. included three projects. In my first project, I used RNAi and CRISPR-Cas9 technologies to study the role of ASAP1 in dendritic cells and macrophages, cells that play critical roles during mycobacterial infection. I demonstrated that in these cells ASAP1 is essential for migration and phagocytosis of mycobacteria. I characterized proteins that ASAP1 interacts with during mycobacterial infection. Finally, I found that the ASAP1-mediated pathway regulates expression of a large number of the immune response genes. These findings emphasize the important role of ASAP1 in mycobacterial infection and explain its involvement in TB pathogenesis. In my second project, I was involved in a large study conducted by our laboratory that characterized transcriptional responses to M. tb infection in macrophages from a cohort of 144 healthy subjects. We used RNA-Seq to study transcriptomes of the infected and non-infected macrophages and identified differentially expressed genes. We also genotyped DNA polymorphisms of these subjects and studied the association between genetic variants and levels of gene expression, which allows us to identify expression quantitative trait loci (eQTLs), i.e., DNA polymorphism that affect gene expression. In particular, we identified an eQTL located in the TLR10-TLR1-TLR6 gene cluster. In non-infected macrophages, a group of polymorphisms in this region was associated in cis with the level of expression of TLR1, but not of the other two TLR genes. In M. tb-infected macrophages the same polymorphisms were associated in trans with levels of expression of 37 genes. This network includes essential immune response proteins, including multiple cytokines and chemokines. The discovery of this TLR1-driven network will help to better understand mechanisms of macrophage responses to mycobacterial infection. Our study also identified a DNA polymorphism located upstream of the ARHGAP27 gene, regulating its expression in infected and non-infected macrophages. In our GWAS this polymorphism was associated with TB risk, which implicated ARHGAP27 in TB pathogenesis. The ARHGAP27 protein is a Rho-GAP involved in the endocytic pathway. In my third project, I used CRISPR technology to establish the ARHGAP27-knockout macrophage cell model and characterized the function of ARHGAP27, showing that it is involved in cell migration and phagocytosis of mycobacteria. Taken together, my studies highlighted functional mechanisms implicating TB-associated GAP proteins ASAP1 and ARHGAP27 in mycobacterial infection and TB pathogenesis.

Abstract:
The use of lasers for diagnosis and treatment in medical and cosmetic applications is increasing worldwide. Not all of these modalities are superficial and many require laser light to penetrate some distance into the tissue or skin to reach the treatment site. Human skin is highly scattering for light in the visible and near infrared wavelength regions, with a consequent reduction of the fluence rate. Melanin, which occurs in the epidermis of the skin, acts as an absorber in these wavelength regions and further reduces the fluence rate of light that penetrates through the epidermis to a treatment site. In vivo fluence rate measurements are not viable, but validated and calibrated computer models may play a role in predicting the fluence rate reaching the treatment site. A layered planar computer model to predict laser fluence rate at some depth into skin was developed in a commercial raytracing environment (ASAP). The model describes the properties of various skin layers and accounts for both the absorption and scattering taking place in the skin. The model was validated with optical measurements on skin-simulating phantoms in both reflectance and transmission configurations. It was shown that a planar epidermal/dermal interface is adequate for simulation purposes. In the near infrared wavelength region (676 nm), melanin (consisting of eumelanin and pheomelanin) is the major absorber of light in the epidermis. The epidermal absorption coefficient is one of the required input parameters for the computer model. The range of absorption coefficients expected for typical South African skin phototypes (ranging from photo-sensitive light skin, phototype I on the Fitzpatrick scale, to the photo-insensitive darker skin phototype V) was not available. Non-invasive diffuse reflectance spectroscopy measurements were done on 30 volunteers to establish the expected range of absorption coefficients. In the analysis it became apparent that the contributions of the eumelanin and pheomelanin must be accounted for separately, specifically for the Asian volunteers. This is a new concept that was introduced in the diffuse reflectance probe analysis. These absorption coefficient measurements were the first to be done on the expected range of skin phototypes for the South African population. Other authors dealing with diffuse reflectance probe analysis only account for the dominant eumelanin. Both the epidermal absorption coefficient and thickness are important in the prediction of the fluence rate loss. The computer model was used to evaluate the effect of the epidermal absorption coefficient (a parameter dictated by an individual’s skin phototype) and the epidermal thickness on the fluence rate loss through the skin. The epidermal absorption is strongly wavelength dependent with the higher absorption at the shorter wavelengths. In the computer model a longer wavelength of 676 nm (typical for a photodynamic treatment (PDT) of cancer) was used. For the darker skin phototypes (V) only about 30% of the initial laser fluence rate reached a depth of 200 ìm into the skin (just into the dermis). For the PDT application, results from the computer model indicated that treatment times need to be increased by as much as 50% for very dark skin phototypes when compared to that of very light phototypes.

Abstract:
In today's ICs and SOCs, voltage level Shifter (VLS) circuits are commonly employed as interfaces for several voltage domains (SOCs). The major design considerations for high performance level shifters are low power dissipation and low latency. In this dissertation we propose two voltage level shifter designs. The first being, an energy efficient voltage CMOS voltage level shifter. The main purpose of this voltage level shifter is to convert the voltage level from one level to another. We verified our voltage level shifter in ASAP7 7nm Fin Fet technology. The proposed voltage level shifter is based on differential cascade voltage switch logic, which takes an input voltage in the range of 0.25V to 0.6V and provides an output of 0.7V. Our voltage level shifter improves propagation delay and power dissipation with 48% and 43%, respectively, with recently reported Wilson current mirror voltage level shifter with zero-Vth design. The proposed design technique comes up with significantly lower power consumption and drastically reduced propagation delay over a wide range of temperatures (-25 to 25 degree Celsius), as compared to existing technologies. The secondly proposed design is based on select signal (Vin) voltage switch logic, which takes an input voltage in the range of 0.3V to 0.6V and provides an output pulse of 1.2 to 0.6V peak to peak. We verified our voltage level shifter in ASAP7 7nm Fin-Fet technology. Our voltage level shifter improves propagation delay and power dissipation with 42.76% and 39.6% respectively with recently reported Wilson current mirror voltage level shifter with Zero-Vt design. The proposed design topology comes up with significantly lower power consumption and drastically reduced propagation delay.

In the midst of the highly competitive, globalizing digitalized era, the new transaction mechanism is shaping in digital economy and all the trends and properties existing in the omnipresent web-environment have resulted in great changes of organizational structure and the nature of transaction, and even in paradigm shift. Business turn toward making the best use of their superior position for competitive advantages, in order to promote their competitiveness and to increase beneficial advantages in specific industry. Many businesses maintained aspects of their organizations that reflected their core competencies within the larger marketplace, while the remaining non-core or relatively non-profitable manufacturing operations, were all moved as far as possible to outsourcing or subcontracting to utilize resources already available on the market, in order to diversify against risk in the industry and marketplace, reduce operation costs, secure competitive advantages and search for the most suitable profit base. Considering the characteristics of highly specialized corporation in the construction industry, it's crucial to select appropriate subcontractors to implement specific subprojects. In this research, the overall subcontracting supply chain of a construction project is considered as a global procurement system and an optimal combination of subcontractors can be obtained within this system. Combined the quick response mechanism of information technology, multiple attribute utility theory, with portfolio theory in financial management, an integrated XML (eXtensible Markup Language) of Accelerated Subcontracting And Procuring (ASAP) decision model was proposed. The ultimate goal of this study is not only to develop a web-based decision support system for general contractors to accurately decide an appropriate trade-off between risk and profit for different combinations of subcontractors, but to take the subcontracting and procuring process into re-engineering through omnipresent internet in the digital economy. The proposed ASAP architecture's initial exploration of using IT to speed up construction projects' subcontracting process, can help to improve the obvious limitations of traditional processes of selecting subcontractors, including the overly limited time for selection, high levels of uncertainty from information asymmetry, and difficulties in judging quality. Bringing together the perspectives of CAPP (Continuous Assessment of Project Performance) assessment model, it also uses historical case database to provide objective project performance standards to assess the total combined subcontracting risks, and applies XML document standardization technology to the production and management of bidding information in subcontracting, to create a web-based construction subcontracting risk assessment support system, for the standardization and economic optimization of the entire scheme of the highly interrelated subcontracting supply chain. Through this system and the development of examples, general contractors can not only work in an environment without excessive time pressure to choose the most appropriate subcontracting supply chain, but control relevant risks and obtain anticipated profits. This formulation of this subcontract schema may be worth consideration in the future from related industries and organizations in the creation of standard schemes. Finally, the relevant illustrative example studies of ASAP system show that this architecture is certainly usable, and that proposed ASAP model has completely transformed the traditional thinking way about selecting subcontractors. It contains positive significance in its use of computerization methods, and its beginnings in remaking the subcontracting procuring process. It will be possible to reduce the overall amount of time and capital required for procurement, and improve the ability to risk assessment. Most importantly, after being made into a widespread and workable process in the future, it will be effective in transforming traditional flaws of subcontractor selection, and greatly enhance the quality of subcontracting supply chain management and controlling the project performance.

The study is intended for the development of a cognitive analytical structure for the ASAP(After School Alternative Program) item bank, providing an explanation on the sources of item difficulty and cognitive load, as the reference to the automatic report system on student performance. In this research, items on "Numbers and Quantities" (to the 2nd and 3rd grades) are used for coding, exploring how well it works for explaining the variety of item difficulty and describing the cognitive characteristics of different levels. The average correct rate can help on discussing the performance of the students.There are 121 problems on "Numbers and Quantities". In this study we used six cognitive components to predict the Rasch difficulty of the items: information load, quantitative relation transformation, sorts of operators, sorts of unit transforming, fractions(decimals) and distractions. It reveals that these six components can predict 66% of variety of item difficulty. Dividing it into 2 levels, we find items in different levels show apparent deviation on information load, sorts of operators, fractions( decimals), and distractions. Average correct rate in the ASAP program and the normal distribution reveals that ASAP students need to be proficient in 2 information and 1 opeartor.

Task difficulty is a significant issue that the test developers are concerned. Purposes of many large-scale assessments are to select examinees for certain purposes. To control or balance item difficulty of a whole assessment helps to improve the validity and discrimination. Test developers construct tests from the cognitive perspective; make the test constructing process more scientific and efficient, and the scores obtained represent the proficient level more precisely. An analysis framework of cognitive components related to English listening comprehension (LC) test is proposed in the research. The data used in this study is derived from the norm of the After School Alternative Program English test (ASAP-ENGLC) item pool. 150 items are analyzed; item difficulty is concurrently calibrated under Three-Parameter-Logistic Item Response Theory from the response of 1459 participants from the fifth to seventh grade. Components may affect item difficulty are proposed and discussed from the cognitive perspectives. All items are coded with cognitive components, e.g. phonetic/ non- phonetic discrimination item type, number of plausible distractors, and content familiarity etc.. Multiple regression analysis is conducted to calibrate the correlation between the components and item difficulty. The result of this research indicates that phonetic/ non- phonetic discrimination item type, number of plausible distractors, necessity of inference, heterogeneity of sentence patterns in options and lexical overlap in key are the best predictors in the predicting model. The total explained variance of this cognitive predicting model is 53.5%. The research result is in expectation to make the task constructing procedure more organized, to make the task difficulty under controlled within the desired extent, so that the intended ability can be measure effectively. Besides, the purpose of ASAP English test is to assist in remedial instruction for low achievement students; the cognitive components and research results proposed in the present research will provide reference for developing teaching materials of English listening comprehension remedial instruction and constructing listening comprehension test items.
