Abstract:
Abstract Despite aggressive, multimodal therapies, the prognosis of patients with refractory or recurrent rhabdomyosarcoma (RMS) has not improved in four decades. RMS is thought to arise due to defective differentiation in skeletal muscle precursor cells. Differentiation-inducing therapy may improve outcomes for RMS patients with advanced disease. In RAS-mutant PAX fusion-negative RMS (FN-FMS), targeting ERK1/2 activation through MEK1/2 inhibition (MEKi) induces differentiation, slows tumor growth, and extends survival in preclinical studies. However, the duration of therapeutic response is short lived. Identifying additional targets for differentiation therapy is necessary. We propose that ASAP1, an Arf GTPase-activating protein (Arf GAP) highly expressed in FN-RMS and implicated in breast and colorectal cancer progression as well as osteogenic and retinal endothelium differentiation, contributes to FN-RMS differentiation. We find that knockdown (KD) of ASAP1 inhibits differentiation in myoblasts and FN-RMS cells, and its overexpression enhances differentiation. Moreover, myogenic differentiation-associated genes are not enriched upon loss of ASAP1. We discover that KD of ASAP1 homologs, ASAP2 and ASAP3, also blocks differentiation. However, loss of a paralogous Arf GAP, ARAP1, does not, indicating that ASAP regulates differentiation through a mechanism not explained by GAP activity alone. Interestingly, KD of Arf1 or Arf5, small GTPases inactivated by ASAP, also blocks differentiation of FN-RMS, suggesting a novel relationship between Arf and ASAP. Using RNAseq, qPCR, and immunoblotting techniques, we show that loss of ASAP blocks myogenic transcription factor expression. To determine the mechanism by which ASAP1 regulates myogenic transcription factor expression, we investigate the PI3K/AKT, MAPK, and Hippo pathways, which are known to be regulated by ASAP1. While the PI3K/AKT and MAPK pathways are unaffected, downstream components of the Hippo pathway are modulated by ASAP1 KD in FN-RMS cells treated with MEKi. YAP and TAZ are transcriptional coactivators that promote proliferation. Upon Hippo pathway activation, YAP/TAZ activity is blocked by phosphorylation at a nuclear exclusion site. Using cell fractionation and immunoblotting techniques, we find that induction of differentiation by MEKi increases TAZ phosphorylation, excluding it from the nucleus and blocking pro-proliferative transcription. Further, upon ASAP1 KD, TAZ phosphorylation is blocked, restoring nuclear localization, and inhibiting MEKi-induced differentiation. In conclusion, we discover that ASAP1 regulates MEKi-induced differentiation of FN-RMS cells by modulating TAZ localization and supports targeting the YAP pathway as a strategy for FN-RMS differentiation therapy. Our work also identifies ASAP1 as a potential effector of Arf1 activity, a novel interaction of these two proteins. The results described herein provide a deeper understanding of differentiation in FN-RMS and establish the groundwork for advancing differentiation therapy in FN-RMS. Citation Format: Katie E. Pirolis terjadi dalam tungku pembakaran dimana pada keadaan ini tempurung mengalami proses penguraian yang disebabkan oleh energi panas. Hasil pengujian yang dilakukan, dari 30 kg tempurung basah menghasilkan 13 kg arang dalam waktu pembakar 3 jam dan 5 kg tempurung kelapa tidak menggalami pembakaran.

Abstract Massachusetts’ 27 Aging Services Access Points (ASAPs) are nonprofit organizations responsible for guiding clients and families in their geographic service areas through a maze of financial and functional eligibility criteria to receive needed home and community-based services (HCBS). These services aim to sustain function and promote engagement for older adults and persons with disabilities, and to maintain individuals in the community rather than in residential settings. Payment for HCBS flows from numerous state and Federal funding programs, including but not limited to state Medicaid waivers and Older American Act programs. We conducted semi-structured interviews with 10 ASAP directors and gathered information from documentary sources to delineate the pathways to funded HCBS that these agencies open for clients. We identified variation in the 27 ASAPs’ contractual relationships with service providers. State data revealed variation in budgetary resources and area population characteristics. These nonprofit corporations appear to flexibly adapt to local conditions and needs. Understanding variation across ASAPs will support analysis of client data to examine whether ASAP processes result in different service outcomes for similar clients. Alternate payment models for health services (managed care, managed long-term services and supports, accountable care organizations) are broadening their reach, encouraging health systems to recognize the importance of functional supports and services. Some health systems are charging their own dedicated units with assigning HCBS to enrollees, avoiding the ASAPs. Understanding and modeling the allocation of services by the ASAPs will allow investigation of trends in services for persons with varying needs under various payment methods.

Background Physical literacy-focused afterschool activity programs (ASAPs) can be an effective strategy to improve children’s health-related parameters. We sought to compare physical activity, body composition, aerobic capacity, and fundamental movement skills between physical literacy-focused ASAP and a standard recreational ASAP. Method A pre–post (6 months) comparison study was conducted in 5- to 12-year-old children in a physical literacy-focused ASAP (physical literacy group, n = 14) and children attending a standard recreational ASAP (comparison group, n = 15). Physical activity guideline adherence was assessed using accelerometry, body composition was analyzed using bioelectrical impedance, aerobic capacity was estimated using the Progressive Aerobic Cardiovascular Endurance Run test, and fundamental movement skills were evaluated using the Test of Gross Motor Development–2. Results There were no significant differences between groups at baseline. After 6 months, the physical literacy group exhibited a significant improvement in their total raw score for the Test of Gross Motor Development–2 ( p = .016), which was likely due to improvements in object control skills ( p = .024). The comparison group significantly increased body mass index ( p = .001) and body fat ( p = .009) over time. No significant between-group differences were found; however, there was a trend for improved aerobic capacity in the physical literacy group ( d = 0.58). Conclusions Engagement in the physical literacy-focused ASAP contributed to an attenuated increase in adiposity and an improvement in object control skills.

ABSTRACT Mosquito-borne diseases, including dengue, malaria, and lymphatic filariasis, exact a devastating toll on global health and economics, killing or debilitating millions every year (54). Mosquito innate immune responses are at the forefront of concerted research efforts aimed at defining potential target genes that could be manipulated to engineer pathogen resistance in vector populations. We aimed to describe the pivotal role that circulating blood cells (called hemocytes) play in immunity by generating a total of 11,952 Aedes aegypti and 12,790 Armigeres subalbatus expressed sequence tag (EST) sequences from immune response-activated hemocyte libraries. These ESTs collapsed into 2,686 and 2,107 EST clusters, respectively. The clusters were used to adapt the web-based interface for annotating bacterial genomes called A Systematic Annotation Package for Community Analysis of Genomes (ASAP) for analysis of ESTs. Each cluster was categorically characterized and annotated in ASAP based on sequence similarity to five sequence databases. The sequence data and annotations can be viewed in ASAP at https://asap.ahabs.wisc.edu/annotation/php/ASAP1.htm . The data presented here represent the results of the first high-throughput in vivo analysis of the transcriptome of immunocytes from an invertebrate. Among the sequences are those for numerous immunity-related genes, many of which parallel those employed in vertebrate innate immunity, that have never been described for these mosquitoes.

CONTEXT AND OBJECTIVE: Preoperative fasting guidelines do not recommend H2 receptor antagonists or proton pump inhibitors. This study investigated prophylactic use of gastric protection and the incidence of dyspeptic symptoms in the immediate postoperative period.DESIGN AND SETTING: Non-randomized observational investigation in a post-anesthesia care unit.METHODS: American Society of Anesthesiologists risk classification ASAP1 and ASAP2 patients over 18 years of age were evaluated to identify dyspeptic symptoms during post-anesthesia care for up to 48 hours, after receiving or not receiving prophylactic gastric protection during anesthesia. History of dyspeptic symptoms and previous use of such medications were exclusion criteria. The odds ratio for incidence of dyspeptic symptoms with use of these medications was obtained.RESULTS: This investigation studied 188 patients: 71% women; 50.5% ASAP1 patients. Most patients received general anesthesia (68%). Gastric protection was widely used (n = 164; 87.2%), comprising omeprazole (n = 126; 76.8%) or ranitidine (n = 38; 23.2%). Only a few patients did not receive any prophylaxis (n = 24; 12.8%). During the observation, 24 patients (12.8%) reported some dyspeptic symptoms but without any relationship with prophylaxis (relative risk, RR = 0.56; 95% confidence interval, CI: 0.23-1.35; P = 0.17; number needed to treat, NNT = 11). Omeprazole, compared with ranitidine, did not reduce the chance of having symptoms (RR = 0.65; 95% CI: 0.27-1.60; P = 0.26; NNT = 19).CONCLUSION: This study suggests that prophylactic use of proton pump inhibitors or H2 receptor antagonists was routine for asymptomatic patients and was not associated with postoperative protection against dyspeptic symptoms.

Abstract:
The trial and verdict of Rakim Mayers, also known as ASAP Rocky, caused major media attention during the summer of 2019. The aim of this bachelor study was to examine how news journalism represented the case “ASAP Rocky” in Swedish tabloids. The research questions examined were: How does journalism portray the story of the victim and perpetrator in the case “ASAP Rocky” ? What are the differences and similarities in how victim and perpetrator are constructed? How does the story develop in Swedish tabloids during the case “ASAP Rocky”?    We used Fairclough’s critical discourse analysis (CDA) to identify reoccurring discourses in the description of the case “ASAP Rocky”. By examining the characters, victim and perpetrator in Swedish tabloids, we found that there was no idealistic role portrayed in the case. Both the victim and the perpetrator were described as insufficient. One discursive theme was that the victim is inadequate since ASAP Rocky´s status as a celebrity provides him an advantage. This discourse is identified also in texts describing ASAP Rocky as a perpetrator. The study showed that the different discourses concern moral values in various contradictions, such as good - evil, right - wrong and “we” versus “them”.    Our study shows that the discourses we found contributes to the creation of celebrities and the discourse that celebrities are unreachable. The conclusion of this study is that media contributes to the stereotype of celebrities.

For this purpose, fluorescence membrane voltage sensors have been developed to detect changes in the membrane potential by changing their fluorescence intensity. The practical part is focused on the study of the properties of the ASAP1 fluorescence probe, which was transfected into the HEK293 cell line, which are kidney cells from the human embryo. Cell membrane potential was changed using the patch-clamp technique.

Abstract:
The Arizona Student Assessment Program (ASAP) is a major piece of legislation for Arizona, reducing norm-referenced standardized testing, providing performance-based assessments matching curriculum, requiring district articulation with state curriculum frameworks and assessments, collecting contextual information from districts, and producing complete profiles of schools, districts and the state. In its first year of implementation, the ASAP is appropriately examined through policy analysis rather than through an evaluation study. Six criteria for educational policy analysis developed by Mitchell (1986) were validated and used as interview questions with seven interviewees knowledgeable about the ASAP. Results of the interviews suggest the degree to which the ASAP is good educational policy and likely to make a difference in Arizona. Interviewees indicated that the ASAP is democratic, providing for both the needs of legitimate stakeholders and the general public interest. It recognizes and supports the organizational integrity of schools only if schools have begun to make some reform efforts of their own in the direction of the ASAP. The ASAP provides adequate means-end linkage for the first two years of implementation, including through school, district, and state profiles, but may need to provide additional help to districts during the first two years; furthermore, relief incentives may be needed, rather than sanctions or disincentives, to encourage continued implementation. The ASAP may not be integrated into overall state educational policy, primarily because there has been no unifying state policy until the ASAP. The ASAP may emerge as a force to reorient current and unify future policy. The ASAP will be expensive, but the interviewees felt the short and long-term benefits justify cost. The ASAP was the most politically feasible policy available to bring about the changes needed, but perhaps not the most palatable, especially to districts that have made no reform efforts of their own. Policy analysis using different criteria and evaluation studies are recommended.

Au cours de la division cellulaire, la mise en place d’un fuseau bipolaire correct est nécessaire au maintien de la stabilité génomique, en permettant une ségrégation égale des chromosomes dans les deux cellules filles. Les microtubules, éléments constitués d’un assemblage de tubuline sous une forme cylindrique et creuse, sont les constituants de base de ce fuseau mitotique. Lors de l’assemblage du fuseau, les centrosomes se séparent, et des changements surviennent au niveau de la dynamique des microtubules. Cependant, sans les propriétés fonctionnelles d’autres protéines venant réguler cette dynamique, les microtubules ne seraient pas capables d’accomplir seuls les tâches qui leurs sont attribuées. Les protéines responsables de cette régulation sont les Protéines associées aux Microtubules ou MAPs. Des variations de la stabilité et de la dynamique des microtubules sont susceptibles d’atteindre la formation et l’organisation du fuseau mitotique ainsi que l’attachement des chromosomes, et peuvent provoquer des problèmes d’ instabilité génétique et la cancérogenèse. Nous avons identifié au laboratoire une nouvelle protéine humaine associée aux microtubules que nous avons appelé ASAP pour ASter-Associated Protein. La protéine ASAP est associée au fuseau mitotique. Sa surexpression entraîne des mitoses aberrantes et abortives (fuseaux multipolaires et cellules multinucléées avec une amplification des centrosomes, fuseaux monopolaires avec non séparation/migration des centrosomes). Ces phénotypes sont fréquemment observés dans des cancers. Sa déplétion provoque des fuseaux anormaux, retarde la mitose en entraînant des défauts de congression et de ségrégation des chromosomes et aboutit à des cytokinèses abortives ou à la mort cellulaire. ASAP est donc une nouvelle MAP jouant un rôle crucial au cours du cycle cellulaire. Une étude plus ciblée sur son implication dans la régulation de la dynamique et de l’organisation des microtubules a ensuite été entreprise afin de mieux comprendre l’impact d’ASAP au niveau du réseau de microtubules. Nous avons ainsi pu mettre en évidence une implication étendue de la protéine, tant au niveau des différentes phases dynamiques de la polymérisation, qu’au niveau de l’assemblage et de l’organisation des microtubules. Nous avons également clairement mis en évidence la présence d’ASAP aux centrosomes, avec la possibilité pour cette protéine de participer à la nucléation des microtubules, un rôle qu’il reste encore cependant à démontrer in vivo.

In the beginning of this thesis is a short introduction into plasmid DNA which is in the form of a vector used in molecular biology. Plasmids can be used in the form of fluorescent probes to measure changes in membrane potential. Into their structure is added a dye called fluorophore. As an important representative of this thesis is a fluorescent probe ASAP1 which contains green fluorescent protein whose response to the membrane potential change is the decrease in the intensity of emitted light. The aim of this thesis was to make chemical transfection of this plasmid into the HEK293 cell line and carry out its characterization. In the work is also described the design of a method for the analysis of the time course of changes in fluorescence depending on the cell membrane depolarisation. In the end of this thesis is also desribed realized experiment including the discussion of aquired results.

AbstractThere is a wide variety of global thematic Land Use Cover (LUC) datasets characterizing agricultural covers. Most of them focus on cropland areas, providing information on their extent or the percentage of cropland cover on the ground. In some cases, the focus is more specific and they provide information on cropland irrigation practices. In other cases, specific maps charting the extension of different crops are also available. In this chapter, we review 8 different datasets with a spatial resolution of at least 1 km. There are many other datasets characterizing agricultural covers at coarser resolutions, such as the Historic Croplands Dataset, GMRCA or GIAM. Their coarse resolution hampers their potential application in practice, which is why they are not described in detail in this chapter. Nor do we analyse FROM-GC, a dataset mapping the extent of global cropland at 30 m, because it is not currently accessible. GFSAD30 has the highest resolution of all the datasets reviewed (30 m). It also provides some of the most up-to-date information (2015). However, it only charts the extent of cropland. As part of an associated project, GFSAD1KCD and GFSAD1KCM characterize cropland areas in 9 and 7 categories respectively at 1 km for 2010. They provide information on the irrigation status of the crops. GFSAD1KCD and GFSAD1KCM were obtained from data fusion. This method is commonly used in the production of many of the cropland datasets reviewed: IIASA-IFPRI cropland map, Global Synergy Cropland Map, Unified Cropland Layer (UCL) and ASAP Land Cover Masks. The IIASA-IFPRI (2005) and ASAP maps provide information on the proportion of cropland at a spatial resolution of 1 km. ASAP also includes a map on rangeland covers, and as such is the only dataset described in this chapter that maps a cover other than croplands. The Global Synergy Cropland Map (2010) and the Unified Cropland Layer (2014) also map cropland proportions, although they have been produced at higher spatial resolutions: 500 and 250 m respectively. The Global Cropland Extent product maps the extent of cropland at 250 m based on imagery from 2000-2008. Although thematically limited, this dataset is less affected by time variability, as it is based on imagery taken over a long period (8 years). Finally, GRIPC maps the extent of three types of cropland area (irrigated, rainfed and paddy crops) at 500 m for 2005.

Este artigo apresenta ASAPe, uma arquitetura para sensoriamento preditivo em computação vestível. Técnicas e estratégias de implementação da nossa proposta são discutidas e avaliadas nesta proposta. Os resultados alcançados até o presente momento indicam que a ASAPe pode ser muitoútil tanto em contextos de coleta de dados em tempo real como também em quantidade massiva de dados. A principal contribuição é fornecer um novo paradigma para trabalhar componentes e predição em Computação Vestível.
