Abstract:
Acute myeloid leukemias with t(8;21) or inv(16)/t(16;16) are commonly referred to as Core-Binding Factor AML and represent 5-8% of all AMLs . These genomic rearrangements are characterized by disruption of the CBF and CBF genes respectively. Both genes encode a subunit of core-binding factor complex, an important transcriptional regulator of hematopoiesis involved in myeloid differentiation. CBF AML are considered a favorable AML risk group based on high remission rate and survival probabilities. However, the presence of the cKIT D816V mutation has been reported to harbor an unfavorable prognostic implication in CBF-AML. The proto-oncogene cKIT encodes a receptor for SCF. It belongs to the type-III receptor of the tyrosine kinase subfamily, that is characterized by five extracellular immunoglobulin-like domains, a single transmembrane helix, a cytoplasmic juxtamembrane domain, and a kinase domain. The cKIT D816V mutation is found in 20–25% of patients with t(8;21) and in about 30% with inv(16)/t(16;16). cKIT D816V causes the indipendent-ligand activation of the tyrosine kinase receptor cKIT, and the activation of downstream pathways supporting cell proliferation and survival. CBF AML patients exhibiting D816V mutation in cKIT gene, are classified in the intermediate risk category, since they present an high incidence of relapse and a low overall survival. Moreover the replacement of the aspartic acid residue with valine at codon 816 causes resistence to Imatinib, the tyrosine kinase inhibitor usually used in the treatment of CBF AML to suppress the overexpressed cKIT. In this case, patients are treated with TKIs of second generation, such as Dasatinib or Nilotinib. So, the single point mutation D816V in cKIT gene, represents a critical prognostic factor for CBF AML patients and an important indicator to define the appropriate treatment. Several PCR-based methods are developed for the detection of D816V mutation, such as PCR followed by direct DNA sequencing, enzymatic digestion with HinfI enzyme (RFLP), or dHPLC. However, most of the PCR-based methods result expensive, time-consuming and labor-intensive. At the moment Sanger sequencing is still the gold-standard for D816V detection, even if it presents a low sensitivity (10–15%). The aim of this work is to improve the detection of cKIT D816V mutation in CBF AML patients by the development of rapid, user-friendly and cost-effective assay based on Allele-Specifc Loop mediated isothermal AMPlification (AS-LAMP) technology. The AS-LAMP assay is based on LAMP technology that amplifies target DNA at costant temperature thanks to the employment of the DNA polymerase with strand-displacement activity. AS-LAMP assay is able to detect the single point mutation thanks to the cooperation of two elements: a loop primer specific for the mutated cKIT sequence, and a blocker that binds only the wild type gene. While the blocker suppresses the aspecific amplification of wild type alleles, the LB primer recognizes the mutated codon and allows its amplification. The AS-LAMP assay includes also the amplification of the housekeeping ABL gene as internal control, that is informative to exclude false-negative results due to the failure of DNA extraction procedure. The target and the IC are detected in real-time in 500 nm and 530 nm channels respectively, thanks the use of two labeled probes specifically binding cKIT or ABL genes. The AS-LAMP assay was optimized on synthetic plasmid containing the cKIT sequence and was successfully validated on negative and positive clinical samples, confirming in all cases the results obtained with PCR-based methods. In conclusion the AS-LAMP assay is able to detect the D816V mutation with high sensitivity and specificity in only 40 minutes and it is easy-to-use. For these reasons the AS-LAMP assay represents a powerful tool for molecular diagnostics applications in clinical routine.

Abstract:
Gastrointestinal parasites Haemonchus contortus and Fasciola hepatica are major impediments to livestock production worldwide. Faecal egg counts remain the most commonly used and widely accepted diagnostic tool for these parasites; however, there is a need for improved, field-applicable diagnostics. In this study, loop-mediated isothermal amplification (LAMP) was optimised and evaluated for the detection of H. contortus (in sheep) and F. hepatica (in cattle) infection. LAMP assays were optimised to enable visual detection using calcein dye. DNA extraction techniques were developed that have the potential for on-farm application. Faeces suspended in water, heated, then centrifuged, with two cheap and stable chemicals, enabled detection of H. contortus at clinically relevant infection burdens. For F. hepatica, a faeces-water suspension was sieved to remove particulate matter, then physical disruption (bead-beating) was applied. LAMP was conducted under laboratory conditions and in the field; compared to FEC (the most commonly used diagnostic for the target parasites) and PCR. LAMP was conducted using three incubation methods: a commercially available thermocycler; a field-friendly low-cost portable styrofoam esky; and a dedicated field applicable LAMP incubator. The general trend was for LAMP to have high sensitivity but only moderate specificity when compared to FEC. However, the use of PCR (both pathogens) and a highly sensitive amended FEC (F. hepatica only) suggested that the apparent low specificity was the result of LAMP being able to detect low-level parasite infection when conventional FEC could not. A LAMP assay paired with a potentially field-applicable DNA extraction was able to adequately detect haemonchosis at ‘clinically relevant’ parasite burdens in a laboratory study. A field study for the detection of F. hepatica demonstrated the potential utility of LAMP on-farm. The studies conducted in this thesis demonstrate the potential of LAMP for parasitic disease diagnosis in agriculture.

Abstract:
Current diagnostic tools for malaria lack the sensitivity to identify individuals with low-density infections. Asymptomatic low-density infections are common in malaria endemic regions and these individuals provide an important reservoir of infection that enables transmission to mosquitoes. Failure to detect these individuals threatens the global health goal of malaria elimination. Loop-mediated isothermal amplification (LAMP) is a technique to amplify DNA and has the potential to diagnose these individuals. The LAMP assay was assessed in a field study in rural Vietnam. 5421 samples were collected and tested with a commercially available LAMP assay in Commune Health Care Centres in Binh Phuoc and Dak Nong Provinces. 101 positive LAMP cases (asymptomatic, smear, and RDT negative) were identified, with the proportion of positives ranging from 0.18% and 3.25% across five communes. In order for LAMP to be used as a screening tool, it must be cost effective and have a workflow suitable for minimally trained end users. To achieve this, an in-house LAMP assay was developed and compared to PCR. The assay was combined with instrument detection to simplify decision making for the end user and improve sensitivity. The in-house assay was as sensitive as the PCR assay and cost US$0.60 per reaction compared to US$3.57 for PCR and US$8.23 for the commercial LAMP. An integrated single cartridge, called T1, was assessed to further simplify this workflow of sample preparation, LAMP amplification and detection. Further development of the cartridge and the assay will be required for future deployment. The LAMP assay is suitable for detection of low density infections in asymptomatically infected individuals in field settings and has the potential for cost effective population based screening.

Windows Azure Platform is product of the Microsoft company aimed to host web applications. With this product, Microsoft is reacting on the new trend in IT -- cloud computing. This diploma thesis deals with all the aspects of developing and running PHP applications on Windows Azure Platform. Thesis is divided into two parts -- theoretical and practical. In the theoretical part, Windows Azure Platform is described in detail. The description consists of it's main parts, position in the world of cloud computing, price and main competitors. In the practical part, migration of school PHP application developed during 4IT445 course from LAMP platform to Windows Azure Platform is described. Except the migration itself, the benefits of new platform to application are described. In the thesis conclusion, development and running applications on both platforms is compared. The target of the thesis is to write first complete Czech text about running PHP applications on Windows Azure Platform and to help PHP developers to chose which way to go.

This work presents an efficient lighting system using high intensity light emitting diodes (LEDs). The goal is to develop a compact and low cost electronic circuit to drive and control the current of LEDs disposed in a single enclosure. The proposal aims energy saving, high luminous efficacy and high useful life when replacing traditional lighting systems, as dichroic, incandescent or compact fluorescent lamps by LEDs without any change in electrical system installation. Furthermore, an emergency lighting system integrated into the lamp is proposed, allowing the use of the same equipment in the daily activities, supplied by mains, and under a possible break in the electrical network. To supply the LEDs through the mains with universal input voltage and charge the battery, a Buck converter was employed. To supply the LEDs with a continuous current through the battery, a Boost converter was used. The converters operate at high frequency and control the current through the LEDs independently. The battery can be composed of three rechargeable Ni-MH batteries (1,2 V) or a Li-ion battery (3,6 V). The design attends the Brazilian standard and international standard for emergency lighting systems. To validate the proposed idea, the circuits were designed and implemented. The experimental results are shown with the intention to prove the satisfactory behavior of the lamps.
Este trabalho apresenta um sistema de iluminação eficiente empregando diodos emissores de luz (LEDs) de alta intensidade. O objetivo é desenvolver um circuito eletrônico, compacto e de baixo custo, para acionamento e controle da corrente dos LEDs contidos em um único invólucro. A proposta visa a racionalização no uso da energia elétrica através de uma elevada eficácia luminosa e longa vida útil na substituição dos sistemas de iluminação tradicionais, como lâmpadas dicróicas, incandescentes ou fluorescentes compactas, por LEDs sem qualquer alteração na instalação elétrica. Além disso, é proposto um sistema de iluminação de emergência integrado a esta lâmpada, permitindo a utilização de um único equipamento para iluminação convencional, alimentada pela rede elétrica, e sob uma possível falha no fornecimento de energia. Para alimentar os LEDs, através da rede elétrica, com tensão de entrada universal e ainda carregar a bateria, um conversor Buck é empregado. Para alimentar os LEDs em corrente contínua através da bateria, um conversor Boost é utilizado. Os conversores operam em alta freqüência e controlam a corrente nos LEDs de forma independente. A bateria pode ser composta por três pilhas recarregáveis de Ni-MH (1,2 V) ou por uma bateria de Li-Ion (3,6 V). O projeto atende às exigências da norma brasileira (NBR 10898) e da norma internacional (IEEE Std 446-1995) para sistemas de iluminação de emergência. Para analisar a idéia proposta, os circuitos são projetados e implementados. Os resultados experimentais são apresentados de forma a comprovar o comportamento satisfatório das lâmpadas.

During the fourteenth century, with the rise of Dol po pa Shes rab rgyal mtshan (1292-1361), the gzhan stong philosophical tradition became a source of great controversy in Tibet. Dol po pa taught this new philosophical tradition for the first time to the wider Tibetan intellectual community. As Dol po pa's Jo nang teachings attracted an audience, many other philosophical giants of the day, such as Bu ston Rin chen grub (1290-1364), Red mda' ba Gzhon nu blo gros (1349-1412/13), and their students composed polemical works to refute Jo nang tradition. Lamp that Illuminates the Expanse of Reality was composed in the midst of this controversy to defend the Jo nang point of view. In it, its author, Gha rung pa Lha'i rgyal mtshan (1319-1402/03), attempts to prove that the Jo nang philosophical tradition is the definitive teaching and the quickest path to achieve the Buddhahood.

The aim of this thesis project was to migrate an existing excel-based application, used

to analyze energy use and carbon dioxide emission of companies, to a web-based application.

Special development questions were put around which software development process,

solution stack and user interface to be used according to the company needs. The spiral

lifecycle model has been chosen because it provides a clear view of the process and has the

concept of early prototypes. A solution stack based on Linux, Apache, PHP and MySQL has

been chosen because such approach has met the company requirements in terms of cost,

security, support, and maintenance. As a result, the developed web-based system overcomes

the problems normally found in excel-based applications, such as application deployment and

maintenance, and provides a more usable and richer user interface.

Esta pesquisa se propÃe a compreender as relaÃÃes participativas e de convivÃncia entre os moradores da comunidade SÃo Vicente de Paulo (Quadra) e a rÃdio poste Centro de ComunicaÃÃo Alternativa, localizada no bairro Aldeota, em Fortaleza (CearÃ). A partir da pesquisa participante (BrandÃo, 1984; 2006) como metodologia, foi possÃvel investigar a histÃria de criaÃÃo da rÃdio poste, refletir sobre as diversas formas de participaÃÃo (Bordenave, 1983) de dois grupos de moradores da Quadra, jovens catÃlicos e mulheres evangÃlicas, e compreender a relaÃÃo deles com a rÃdio dentro da convivÃncia com as paisagens sonoras (Schafer, 2011) da comunidade. Entre as consideraÃÃes finais, foi possÃvel entender que a participaÃÃo de jovens e mulheres religiosos na rÃdio deu-se atravÃs da mobilizaÃÃo dos prÃprios grupos religiosos. Por fim, o campo ainda proporcionou momentos interventivos, durante o aniversÃrio do programa âRecordar à Viverâ e participaÃÃo no projeto de reforma da rÃdio. Este percurso de pesquisa tambÃm me fez perceber que a intervenÃÃo esteve presente desde minhas primeiras aproximaÃÃes com a Quadra. PorÃm, antes, pesquisa e intervenÃÃo pareciam se distanciar. Agora, concluo que as metodologias participativas unem esses processos.
This research aims to understand the participatory coexistence and relations between the residents of SÃo Vicente de Paulo community (Quadra) and the radio post Centro de ComunicaÃÃo Alternativa, located in Aldeota district, in Fortaleza (CearÃ). From the participatory research (BrandÃo, 1984; 2006) as a methodology, it was possible to investigate the history of creation of the post radio, consider about the various forms of participation (Bordenave, 1983) of two groups of residents of Quadra, young Catholics and evangelical women, and understand their relationship within the radio in the living with soundscapes (Schafer, 2011) community. Among the final comments, it was possible to understand that the participation of young women and religious radio was given by mobilizing of their own religious groups. Finally, the course of this research also provided interventional times during the anniversary of the program "Remembering is living" and participation in the radio reform. This search path also made me realize that the intervention has been present since my first contact with Quadraâs community. However at first, research and intervention seemed to be distance. I concluded that the participatory methodologies unite these processes.

Abstract:
The pre-symptomatic stage of neurodegenerative diseases such as Alzheimer’s disease (AD) and Parkinson’s disease (PD) occurs several decades before the clinical onset. Changes in the lysosomal network, i.e. the autophagosomal, endosomal and lysosomal vesicular system, are among the first alterations observed. There are currently no treatments to slow or cure neurodegenerative diseases, and there is a great need for discovery of treatment targets in cellular pathways where pathology pre-dates the neuronal death. It is also crucial to be able to diagnose neurodegenerative diseases earlier, both to enable early intervention treatment and aid in selecting clinical trial populations before the patient has widespread pathology. This thesis aims at investigating the potential of lysosomal network proteins as biomarkers and therapeutic targets in neurodegenerative disease. A targeted search for lysosomal network proteins was performed in cerebrospinal fluid (CSF) from AD patients, and seven proteins: early endosomal antigen 1 (EEA1), lysosomal-associated membrane proteins 1 and 2 (LAMP-1, LAMP-2), lysozyme, microtubule-associated protein 1 light chain 3 (LC3), Rab3 and Rab7, were elevated. The levels of EEA1, LAMP-1, LAMP-2, LC3, lysozyme and Rab3 were also measured in CSF from parkinsonian syndrome patients: PD, clinically diagnosed 4-repeat tauopathy, pathologically confirmed corticobasal degeneration (CBD) and pathologically confirmed progressive supranuclear palsy (PSP) patients. LAMP-1 and LAMP-2 were decreased in PD. LC3 and lysozyme levels were increased in 4-repeat tauopathy patients. EEA1 was decreased and lysozyme increased in PSP, and LAMP-1, LAMP-2, LC3 and lysozyme were increased in CBD. The lysosomal network proteins had different CSF protein profiles in all the parkinsonian syndromes, as well as in AD. It should be emphasized that only a select few of the lysosomal network proteins were observed to be changed, rather than a general change in lysosomal network proteins, which implicates the involvement of these seven proteins in specific pathological processes. The most interesting candidates, LAMP-2 and lysozyme, were selected for further study for their involvement in the pathology of AD. Lysozyme was found to co-localise with Aβ plaques in AD patients and overexpression prolonged survival and improved the activity in a Drosophila model of AD. Lysozyme was found to alter the aggregation pathway of Aβ1-42, to counteract the formation of toxic Aβ species and to protect from Aβ1-42 induced cell toxicity. Aβ1-42 in turn was found to increase the expression of lysozyme in both neuronal and glial cells. These data suggest that lysozyme levels rise in AD as a compensatory response which is protective against Aβ associated toxicity. LAMP-2 mRNA and protein were found increased in brain areas relevant for AD pathology and various cellular models showed complex involvement of LAMP-2 in Aβ related pathology, with extensive crosstalk between LAMP-2 and Aβ. Exposure to oligomeric Aβ1-42 caused an upregulation of LAMP-2 and in turn, overexpression of LAMP-2 caused a reduction in secreted levels of Aβ1-42, as well as changing the generation pattern of Aβ and affecting clearance and secretion of Aβ1-42. These data indicate that the increased levels of LAMP-2 in AD could be an attempt to regulate Aβ generation and secretion. In summary, this thesis reports that utilising lysosomal network proteins as biomarkers and novel therapeutic targets for neurodegenerative diseases holds great promise.

Abstract:
Virtualization is a technique used to abstract the operating system from the hardware. The primary gains of virtualization is increased server consolidation, leading to greater hardware utilization and infrastructure manageability. Another technology that can be used to achieve similar goals is containerization. Containerization is an operating-system level virtualization technique which allows applications to run in partial isolation on the same hardware. Containerized applications share the same Linux kernel but run in packaged containers which includes just enough binaries and libraries for the application to function. In recent years it has become more common to see hardware virtualization beneath the container host operating systems. An upcoming technology to further this development is VMware’s vSphere Integrated Containers which aims to integrate management of Linux Containers with the vSphere (a hardware virtualization platform by VMware) management interface. With these technologies as background we set out to measure the impact of hardware virtualization on Linux Container performance by running a suite of macro-benchmarks on a LAMP-application stack. We perform the macro-benchmarks on three different operating systems (CentOS, CoreOS and Photon OS) in order to see if the choice of container host affects the performance. Our results show a decrease in performance when comparing a hardware virtualized container host to a container hosts running directly on the hardware. However, the impact on containerized application performance can vary depending on the actual application, the choice of operating system and even the type of operation performed. It is therefore important to consider these three items before implementing container hosts as virtual machines.

Along with the society development, and the fluctuation in national politics and economics, human beings have been suffering from pressure due to their life styles, when emotions would be affected at the same time. Under this circumstance, people living with modern life start seeking for any possible merchandise or atmosphere to satisfy personal needs of sensual. Emotions and feelings are occurred under some particular conditions, while a variety of physiological and physical structures are involving. Early philosophers believe that feelings come from how human being reacts to environmental stimulation, which shows the importance of space atmosphere. In terms of appropriate atmosphere, it would influence someone’s mentality through physiological way, and then brings out the effect on curing and easing sentiments. Based on space atmosphere composition and influence factors theories, it’s been confirmed that lighting is one of the important elements. According to existing theories, this design covers the concept of integrating light, shadow, and space, in order to loosen stress and ease up tension by forming difference atmospheres in the space and representing different sensory experiences. This situational lighting idea was inspired by nature, and was integrated with innovative concepts, techniques, and characters of different components. Sponge, the most important portion in this design, plays the medium of changing light sources, and helps present unique atmospheres to create complete impression of space with steady incoming lights. During series of research and development, evaluations at different steps and examinations with stimulations both support execution effects of this design and verification of possible difficulties. Conclusion has been divided into space atmosphere establishment and material innovation. By space atmosphere establishment, this piece uses light and shadow to define the concept of space; changeable aura would be formed by multiple functions and artistic appearance at the same time. Moreover, making the light with shield shape allows users switch the way of using as different needs. Regarding material innovation, this design helps confirm that Sponge successfully acts the medium of transmitting the light and creates and variation in figures and situations; definitely it is an idea worth building further. On the other hand, the unique way to iron textures with several layers on the Sponge elaborates the identities of this material and leads future developments towards to a more diversified prospect. This design is expected to pioneer the newer thinking of situational lighting, and to proceed to the next step as the reference of future product development. In this article, we aimed to prepare PP / PP-g-silane / glass fiber composite materials, and to observe variations of the physical properties. The grafting of VTMS on PP in the presence of DCP was carried out in a melt process, and then by the mixing of which with PP and glass fiber. By the FT-IR spectroscopy, we will confirm the silane was essentially grafted onto PP. Through the degree of crosslinking measurement, we confirmed the coupling agent reacted with glass fiber. Microstructure analysis by scanning electron microscopy (FE-SEM) was used to observe the interface between polypropylene and glass fiber adhesion. By measuring tensile testing ,we made stress calculations based on single-arm pole. Form FT-IR and degree of crosslinking, we have confirmed the silane was essentially grafted and has the ability to crosslink. SEM of the fracture surfaces of the coupled composites shows a good adhesion at the fiber/PP interface. The tensile modulus values and tensile strength values of the composites coupled by the macromolecular coupling agent PP-g-Silane, are higher than those of the non-coupled materials form tensile test. Stress calculations show that when the poles are upon the 17 level wind, the roots are exposed to the stress of 11.2 MPa, and the deflection is 44.8 mm. Clamp on the light poles generate for the 1.7 MPa. The screw between ground and pole are generated for the 0.3 MPa. Openings are exposed to the stress of 15.9 MPa. From the above results, the PP-g-Silane as a coupling agent by adding PP / glass fiber composite materials, can significantly improve the mechanical properties. With a pole of 3000 mm high, and 150mm diameter, and 25mm thickness, the required stress did not exceed the critical value of linear elasticity, and can be fully recovered after the wind stop blowing.

Projector replacement lamps used to be widely used only in schools and office meeting rooms. Now, projectors are no longer a gigantic and expensive machine. The technology advancement, that allows projectors being slimmer and smaller, makes a new application for this presentation tool to be installed at home as a home theater as well as a gaming device at a reasonable and affordable cost. This recent trend has created in a new segment for the projector market, together with a surging demand for its consumable market of aftermarket replacement lamps. With the expending install base of projectors, the aftermarket replacement lamps become a lucrative and eye-catching business for projector manufacturers such as Epson and Benq, compatible lamp manufacturers which will be described in this thesis hereafter and those Chinese copy-cat manufacturers also known as Shanzhai manufactures. The influx of different new comers to this aftermarket has resulted in an intensive competition that led to a relentless price war at the end. Moreover, the new light source of LED or solid-state lamp that has 5 times more life span as a substitute of the conventional lamp source of projector is becoming a threat that would possibly take a large chunk of the current existing aftermarket of projectors. This thesis is served to showcase the insights of the future outlook and trend of the aftermarket of projector replacement lamps. To make this thesis more comprehensive, there are some interviews with three experts from this industry included in this report. The interviewers generously shared their own views about the current situation, the forecast and some suggestions for Company A to formulate its marketing strategy. Lastly, there is a SWOT analysis to summarize those suggestions from the three experts, which can be utilized as a reference for Company A to stand out from the tough competition so as to sustain the continuous growth of Company A in the long run.

The invention of the light bulb not only enlarges our living space but earns us more time for various activities. With the advancement of technology, people start to realize that lamps are not just for lighting, they can be used to create cozy environments. Hence, some artists utilize lamps as the subject of artistic creations, and businessmen employ attractive lighting design to invite customers. Since lamps can be made of diverse materials and take countless shapes, lamp buyers have plenty to choose from. Lamp manufacturers who wish to survive and maintain sustainable development have to continuously improve the production quality and develop new products. This thesis aims at revealing the optical effects which the polypropylene boards might generate. Through the analyses of lamp formation and the tests of optics, I attempt to unveil new structures dissimilar to those found on the market and apply these innovations to lamp design. My findings suggest that “cutting”, “knitting”, and “unit re-formation” show great promise for new lamps. Therefore, I make models based on the three methods and assess the potential of each model for new lamp designs. The final products are categorized into the “light echo series”, “irradiation series”, and“petaloid series.“

In the new century, the market has highly competition, and is simultaneously affected in globalization, localization, and individualization. Traditionally black-boxed designing model can’t match what the consumers require, exactly and effectively, not mentioning the numerous chooses in the present. “Just for fun” may be the best choice. Therefore, the designers’ responsibilities are for the choices of appropriated use, suitable size, the favorable texture, the satisfactory style, and right message transformation in products. Let products fit in human and let users conduct products designing. Then they are really products that consumers really need. Take lamps for example. Study randomly one hundred and eighty people in Taichung metropolis by using Q & A evaluation scales to explore the proper texture designing. Apply visualization texture and psychological feeling of costumers, and add small group market, which is emphasized nowadays, designers know well about population’s sense image when developing product design. In Q & A investigating, the costumers fill the form based on the situation in using table lamps and the aspect of life style. The result helps to understand the life style of general costumers. By conducting SD and statistics methods, factors analysis, the evaluation of texture and the favor of the image analysis the relationship between costumers’ favor and the texture of table lamps, and get further information about the psychological feeling of specific population in the texture of table lamps. The results show that the characteristic of population variable does not exist in the texture image of table lamp materials. Costumers can be divided into five groups, stable group, practical group, guard group, active group, and selfhood group. According to the research, the difference in the life-style affects the image much. Because the sampling areas are too close to make obvious effect. Among nine kinds of table lamps, the texture of table lamps is great different between metal and agate, and the image in cloth is similar to that in plastics but plastics tables are more artificial. Glass table lamps are also similar, but fog glass was felt friendly than transparent glass. Table lamps made in vines and papers are close in modernization. Table lamps made in PP and metal are belonged to fashion texture. The evaluation way can be also divided into five aspects, including fineness, time, friendliness, lightness, and firmness. Hopefully, the research would be a reference in the relevant product designing when in manufacturing. From the research, designers would quickly and exactly match the costumers’ favor and need in material textures.

Photoreflectance (PR) spectra of a GaN thin film and an AlGaN/GaN heterostructure were measured by using a HeCd laser or a mercury lamp as a pump beam. The wavelengths (λ) of the HeCd laser and the mercury lamp are 325 nm and 253.7 nm, respectively. The energy of the HeCd laser is lower than band-gap energy of AlxGa1-xN (x > 0.2) so that electron-hole pairs cannot be generated in the AlGaN layer. Hence, the PR of the AlGaN was measured by using Argon ion laser (λ=300 nm) or quadrupled Nd:YAG (λ=266 nm) rather than HeCd laser in the previous works. In this work, the mercury lamp (λ=254 nm)was used as the pump beam. The problem with using the mercury lamp as the pump beam is because it is a diffused source so that it cannot be focused to a small spot. Nevertheless, defocused pump and probe beams were used in the PR measurement to improve signal to noise ratio. Hence, the diffused property of the mercury lamp is not a hindrance to the PR measurements.

Designers apply different design techniques to create more beautiful and innovative products, to achieve higher market preference. Through the observation of contemporary products, many of them exhibit both typical and novel features to increese their uniqueness and value in the market. Based on the procedure of design, it is found that most of the designers regard the prototype of a product as a basis for reference and then deviate from it to make a product more unique and novel. For example: the light bulb of the ice lamp designed by Harri Koskinen was enclosed in the ice to present a vision of both warm and cold. The model of the Miss Sissi table light designed by Philip Starck in 1990 is a typical lamp which was made unique by using plastic materials to show both typical and unique feelings. As per Raymond Loewy’s claim, the preferred product is usually the best combination of typical and novel features. This concept implies a contradiction. Designs with contradiction were found to gain higher overall preference in many former studies. Ludden (2008) also mentioned in her study that products can improve the meaning and interaction, and even create surprise feeling if they can evoke different experience or sensations. As a result, these products can easily attract people’s attention and leave a deep impression on them. This study focuses on the discussion of the relationship between contradiction and surprise, and provides guidelines to the designers for contradictory design. Three conclusions as the followings: (1)In general, contradiction can trigger surprise based on the research results and correlations of trend observations. (2)This study applied the k-means cluster classification method to observe the distribution of the sample in the Nine Chequers. The surprising and preferred samples all fall into the contradiction and uniqueness categories. Participants with different education backgrounds have significantly different perceptions toward contradiction and preference. (3)The stunned and disliked samples are mainly caused by the lack of understanding of what the designers wanted to express. This even resulted in a resentment towards the products which may have further reduced the level of preference. Indeed, the designers must be careful about whether the design technique can be easily understood and avoid excessively applying design language and elements.

Due to the popularity of astrology and fortune-telling TV programs in recent years, buying fortune products has gradually become a habit of many Taiwanese consumers. Fortune products are mainly created based on the preference and perception of experts or celebrities. They are made to satisfy the mental needs of consumers but without consideration of user perceptions and practical needs. This study investigated lamp designs to induce elements and images of lamp designs that can meet user needs for fortune and product usefulness. The aim of this study was to find a consensus over fortune products between fortune experts and general consumers and provide a reference for developers of this kind of products. In the first stage, fortune experts were invited to evaluate 30 lamp design samples based on the five elements theory. 15 of them met the conditions and were introduced into a web-based two-dimensional analytical tool. Later, a survey was conducted to explore consumer perceptions of fortune products and evaluate the image words carried by the products. The images and weight of each image word obtained from the survey became a basis for subsequent lamp design. Three lamp products were designed. These designs and the pre-test samples were evaluated using importance-performance analysis (IPA) and grey relation analysis (GRA). The results indicated that the lamp designs were good among all the samples. In conclusion, application of the five elements theory to lamp design could not only retain the meanings of fortune products but also satisfy consumers’ expectation for perceptual images of fortune products. These single and independent women are so-called “underdogs” or “Heidis” . In recent years, researcher and the friends of researcher are gradually to the year of economically independent women who are so-called “underdogs”. Many of researcher’s friends decide to rent outside or buy houses on their own. For the influence of these friends, researcher started to be interested on the home life style and the design of housing products of the single and unmarried female.There are lots of bachelor women putting more focus on their living qualities. Recently, more and more real estate commercial are focusing on bachelor women, which shows that single women put more consideration on their independent living environment. Those economical independent bachelor women care much about life quality. They have good taste and really put emphasis on choosing every material that fits their life style. However, currently there is not so many products that are suitable or designed for them. Since they are living alone, they think much on good atmosphere of the environment. Moreover, because of their heavy workload, they also need a relax feel after work especially at home. Therefore, among all the housing products, essential products are commonly attractive to them, especially fragrant lamps. The warm bulb/candle light creates romantic atmosphere with fragrance. They can be relaxed by the fragrance of aromatherapy.This study will focus on the market of volatile oil decorative lighting to collect data and analyze it. The single women who living alone were interviewed to understand their life style and their demand for volatile oil decorative lighting. The results of interview will be used to design volatile oil decorative lighting which suit for the single women who living alone. The designed volatile oil decorative lighting will be sent to the interviewees to test it, try to learn more about the views of this products from single women and their recommendations, in order to let the design of volatile oil decorative lighting could be closer to single women’s home living. Furthermore, volatile oil decorative lighting can have more design innovations and breakthroughs in the market.

In this research is the use of high-CRI cool white LED light source to design surgical light lamps, use of two different types of secondary optics design, TIR (Total Internal Reflection) lens type and reflective cover type surgical lamps, improve the efficiency of light use, to achieve the best results within the specification. in the design can be divided into two parts, The first part is the calculation and design of LED TIR (Total Internal Reflection) lens type and reflective cover type a single module; The second part is the use of FRED optical simulation software to arrange, modular design of the overall model, make it can reach the illumination value of more than 130,000 Lux, and average CRI of more than 90, and between color temperature of 3000K ~ 5000K, and IEC (International Electrotechnical Commission) the norms of medicallam.

Patent Act exactly encourages the creation of products and protects the authority of industrial innovation. Recently, with an increase in patent analysis approaches to the study of patent creation, we have seen an obvious change in research patterns. Moreover, more Taiwanese companies have realized the importance of developing complete and steady patent products, and believed that they can support higher profits, firm business reputation, and long-term development. These companies would spend a lot of time finding what patents they need, which products they have to create, and how many resources they should use to produce products. However, little research has focused on patent analysis and patent making. In fact, the production and analysis of patent map can provide the appropriate patent portfolio, and increase the success probability of the product. The aim of this paper is to explore the strong relationship between a useful patent creation and patent-map analysis. This study includes patent search, patent analysis, and patent creation. Around 500 patent instructions concerning patent creation and analysis strategies were collected, and every page of them was reviewed. In the example of LED (Light Emitting Diode) lamps, our analyses using the proposed methodology indicate that the development of LED lamp has gradually gone into a maturity stage. Therefore, the results of this study not only provide the direction of new patent creation via the searching of patent vacancy, but also can save the resources during the research and development stage of product. This dissertation explores the design of LED street lighting fixtures, with a case study on the collaborative design of LED modules commissioned by TYC Brother Industrial Co., Ltd. TYC Brother Industrial Co., Ltd. was established in 1986, concentrating on manufacturing and selling vehicle lights and auto-parts, aiming at the automotive lights AM, and is one of the largest manufacturers of automobile, truck, bus and motorcycle lights. They supply the global OEM and AM with light fixtures of various functions, and has in recent years become actively involved in the development of LED street lighting equipments. In this study, data and references are collected to analyze and compare existing LED street lighting products. Analysis shows that in the public domain, or in larger regions, the population structure would be more complex and diverse, thus resulting in a relatively simple LED street lighting fixture design with neutral features; if the geographical placement of the street lamp is in a medium-sized regional community environment, the design generally exhibits more cultural characteristics; and if the street lighting fixture is placed in the private sector, the design tends to be manifold, reflecting the personal taste of the owner. This case study focuses on the design of LED street lighting fixtures that belong to the public domain.

To save energy, many local governments replace mercury street lights with energy-saving ones. Subsidized by central government, Taoyuan City Government took ESCO (Energy Saving Company) approach to execute such replacement. Nevertheless, New Taipei City Government, without subsidies from central government, took the lead in adopting PFI model. ESCO refers to an energy service company whose purpose is to provide the energy users with a whole spectrum of services, from energy diagnosis, retrofitting plan evaluation, retrofitting design, engineering, supervision and management, to financial planning for fundraising and guaranteed retrofitting investment return. PFI, a private financing proposal referred to by scholars, is a way of private sector’s participation in public constructions through government’s procurement of public service (energy) provided by a private company during its operation. So far in Taiwan, only New Taipei City government has the PFI experience, and therefore, few cases can be studied. Most researches of government procurement behavior focus on optimal procurement model, that is, the design to award the contract shall be the most advantageous tender, the lowest tender pass grading or the lowest tender, and seldom will the agency theory in the filed of public administration be discussed at the same time. Starting from the fundamental question – agency, the problems arising from self-interest, such as concealment of information and concealment behavior can be solved. For example, how to prevent adverse selection and moral hazard resulting from target conflict and information asymmetry? Which model fits municipal government most-ESCO or PFI? This research focuses on the principal-agency theory as well as incomplete contract. For case studies, interview outlines are designed specifically for Taoyuan City Government and New Taipei City Government, which took different approaches toward tender strategies, and in-depth interviews are conducted with interested parties to reach findings, and policy advises will be made accordingly. In summary, in consideration of interface integration, the PFI model shall be adopted for tender of installment of energy-saving street lights to municipality’s advantage if the project reaches certain economic scale. As for key performance indicators for service, the computer system shall be built eliminating subject evaluation factors and use objective evaluation standard as much as possible.
