Abstract:
The increasing incidence of antimicrobial resistance (AMR) is a major global challenge. Routine techniques for molecular AMR marker detection are largely based on low-plex PCR and detect dozens to hundreds of AMR markers. To allow for comprehensive and sensitive profiling of AMR markers, we developed a capture-based next generation sequencing (NGS) workflow featuring a novel AMR marker panel based on the curated AMR database ARESdb. Our primary objective was to compare the sensitivity of target enrichment-based AMR marker detection to metagenomics sequencing. Therefore, we determined the limit of detection (LOD) in synovial fluid and urine samples across four key pathogens. We further demonstrated proof-of-concept for AMR marker profiling from septic samples using a selection of urine samples with confirmed monoinfection. The results showed that the capture-based workflow is more sensitive and requires lower sequencing depth compared with metagenomics sequencing, allowing for comprehensive AMR marker detection with an LOD of 1000 CFU/mL. Combining the ARESdb AMR panel with 16S rRNA gene sequencing allowed for the culture-free detection of bacterial taxa and AMR markers directly from septic patient samples at an average sensitivity of 99%. Summarizing, the newly developed ARESdb AMR panel may serve as a valuable tool for comprehensive and sensitive AMR marker detection.

Abstract:
This article proposes a novel network model to achieve better accurate residual binarized convolutional neural networks (CNNs), denoted as AresB-Net. Even though residual CNNs enhance the classification accuracy of binarized neural networks with increasing feature resolution, the degraded classification accuracy is still the primary concern compared with real-valued residual CNNs. AresB-Net consists of novel basic blocks to amortize the severe error from the binarization, suggesting a well-balanced pyramid structure without downsampling convolution. In each basic block, the shortcut is added to the convolution output and then concatenated, and then the expanded channels are shuffled for the next grouped convolution. In the downsampling when stride >1, our model adopts only the max-pooling layer for generating low-cost shortcut. This structure facilitates the feature reuse from the previous layers, thus alleviating the error from the binarized convolution and increasing the classification accuracy with reduced computational costs and small weight storage requirements. Despite low hardware costs from the binarized computations, the proposed model achieves remarkable classification accuracies on the CIFAR and ImageNet datasets.

Abstract:
Abstract Motivation The elucidation of dysfunctional cellular processes that can induce the onset of a disease is a challenging issue from both the experimental and computational perspectives. Here we introduce a novel computational method based on the coupling between fuzzy logic modeling and a global optimization algorithm, whose aims are to (1) predict the emergent dynamical behaviors of highly heterogeneous systems in unperturbed and perturbed conditions, regardless of the availability of quantitative parameters, and (2) determine a minimal set of system components whose perturbation can lead to a desired system response, therefore facilitating the design of a more appropriate experimental strategy. Results We applied this method to investigate what drives K-ras-induced cancer cells, displaying the typical Warburg effect, to death or survival upon progressive glucose depletion. The optimization analysis allowed to identify new combinations of stimuli that maximize pro-apoptotic processes. Namely, our results provide different evidences of an important protective role for protein kinase A in cancer cells under several cellular stress conditions mimicking tumor behavior. The predictive power of this method could facilitate the assessment of the response of other complex heterogeneous systems to drugs or mutations in fields as medicine and pharmacology, therefore paving the way for the development of novel therapeutic treatments. Information from cluster head to base station may have a lot of chances to get hacked. The most reliable way to manage energy consumption is clustering, and encryption will suit best for information security. In this paper, we explain clustering techniques and a new algorithm to encrypt the data in the network.

Abstract:
Abstract Motivation Acute myeloid leukemia (AML) is one of the most common hematological malignancies, characterized by high relapse and mortality rates. The inherent intra-tumor heterogeneity in AML is thought to play an important role in disease recurrence and resistance to chemotherapy. Although experimental protocols for cell proliferation studies are well established and widespread, they are not easily applicable to in vivo contexts, and the analysis of related time-series data is often complex to achieve. To overcome these limitations, model-driven approaches can be exploited to investigate different aspects of cell population dynamics. Results In this work, we present ProCell, a novel modeling and simulation framework to investigate cell proliferation dynamics that, differently from other approaches, takes into account the inherent stochasticity of cell division events. We apply ProCell to compare different models of cell proliferation in AML, notably leveraging experimental data derived from human xenografts in mice. ProCell is coupled with Fuzzy Self-Tuning Particle Swarm Optimization, a swarm-intelligence settings-free algorithm used to automatically infer the models parameterizations. Our results provide new insights on the intricate organization of AML cells with highly heterogeneous proliferative potential, highlighting the important role played by quiescent cells and proliferating cells characterized by different rates of division in the progression and evolution of the disease, thus hinting at the necessity to further characterize tumor cell subpopulations. The residual effect of a few herbicides viz: Atrataf (atrazine), 2,4-D and Aresin (monolinuron) sprayed in the previous season on the pigment concentration and primary productivity of the crops (viz: Zea mays and Cajanus cajan) grown in rotation, as well as their associated weeds has been studied. The results indicate that the residues of all the three herbicides were non toxic to Zea mays and toxic to Cajanus cajan as an increase in chlorophyll content and primary productivity occurred in the former, whereas the contrary is true for the latter. Weeds from residual plots have shown a considerable decrease in the chlorophyll content and primary productivity. The most susceptible weed to all the three herbicides was Acalypha malabarica, while Echinochloa colonum is susceptible to the residues of Atrazine but is tolerant to the residues of the other two herbicides. Abstract:
The topic of degrees of being in Aristotle is almost universally ignored. A very few scholars do discuss the topic or make use of it in passing. This situation mightbe explained by a scholarly consensus that (a) Aristotle did have a doctrine ofdegrees of being, but (b) this doctrine is too uninteresting to be worth much discussion. Conversation with a number of scholars from several countries has convinced me, however, that a rather different consensus lies behind the current silence. It turnsout that many experts in the subject deny that Aristotle believed in degrees of being. No one, to my knowledge, has defended this denial in print. But the reason forsilence is not that the topic is dull, but that it is scandalous. Both defenders andopponents of the view that Aristotle had a doctrine of degrees of being tend, in conversation, to find the topic embarrassing. Our contemporary metaphysical prejudices areso opposed to degrees of being that people find themselves unable to make anysense of such a doctrine. As a result, one group of scholars is embarrassed on Aristotle's behalf at the suggestion that he might have held such a senseless doctrine.Another group, less sure of where the philosophical truth lies, finds itself in theinterpretative embarrassment of being unable to explain and motivate the doctrine. So bothgroups avoid the subject.

Grandparenting parenting pattern is childcare done by grandparents. Childcare should be the responsibility of both parents, but because there are several factors, namely: both parents are busy working, the economic state of the family and there is no parental trust in others in the care of the child except from a close family, so the responsibility is transferred to grandparents. In RT/04 RW/02, there are 7 families who transfer temporary parenting roles to grandparents. The types of parenting patterns applied are: democratic, authoritarian and permissive parenting patterns. From this type of parenting pattern, it will affect the personality of the child. There are 5 personality types: sanguin, phlegmatic, melancholic, cholelic and arsetif types. In childcare each grandparent has its own parenting pattern but the more instrumental in parenting is grandmotherhood. Parenting patterns are influenced by educational background, knowledge, activities, socioeconomic circumstances and so on. But the supporting factor in the implementation of parenting patterns well depends not only on the type of parenting pattern applied by grandparents, but also on the characteristics in the family and community environment. The forms of foster care patterns that are applied are very closely related to the personality of the child after becoming an adult. Abstract:
ABSTRACT Whole-genome sequencing (WGS) is now routinely performed in clinical microbiology laboratories to assess isolate relatedness. With appropriately developed analytics, the same data can be used for prediction of antimicrobial susceptibility. We assessed WGS data for identification using open-source tools and antibiotic susceptibility testing (AST) prediction using ARESdb compared to matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) identification and broth microdilution phenotypic susceptibility testing on clinical isolates from a multicenter clinical trial of the FDA-cleared Unyvero lower respiratory tract infection (LRTI) application (Curetis). For the trial, more than 2,000 patient samples were collected from intensive care units across nine hospitals and tested for LRTI. The isolate subset used in this study included 620 clinical isolates originating from 455 LRTI culture-positive patient samples. Isolates were sequenced using the Illumina Nextera XT protocol and FASTQ files with raw reads uploaded to the ARESdb cloud platform (ares-genetics.cloud; released for research use in 2020). The platform combines Ares Genetics’ proprietary database ARESdb with state-of-the-art bioinformatics tools and curated public data. For identification, WGS showed 99 and 93% concordance with MALDI-TOF MS at the genus and species levels, respectively. WGS-predicted susceptibility showed 89% categorical agreement with phenotypic susceptibility across a total of 129 species-compound pairs analyzed, with categorical agreement exceeding 90% in 78 species-compound pairs and reaching 100% in 32. Results of this study add to the growing body of literature showing that, with improvement of analytics, WGS data could be used to predict antimicrobial susceptibility.

Abstract:
The succession of extraordinary traumatic events, such as the poles defrosting, the fires in Australia and Amazon, the COVID-19 pandemic, the financial global crises revival, the Black Lives Matter movement, seems to call for science (to solve problems) and schooling (to make time to the unemployed) for help. Science and schooling are some of the big successes of post-modern culture and states. Given these successes, questions remain. Why do they not deliver hope? We are left to ponder (a) why these extreme events takes us by surprise; (b) why those who have been afforded the best education ever areso anxious, so frozen; (c) why is it that our best policies, science, and minds continue to fail us when it comes to solving the problems associated with the environment, this pandemic, capitalism, and racism; and (d) given the promise of science, why are we so in need of solutions?
