Full textThe main objective of this work is based on the search and the development of new antibacterial compounds able to interact with a bacterial wall. The proposed compounds are of polycationic nature, elaborated on calixarene structure or an aromatic platform and containing guanidinium functions in various number and position. Some of them were developed as water-soluble prodrugs of the quinolone: nalidixic acid. The chapter I talk about introduction and a bibliographical study on calixarenes and their applications in biology. The chapter II presents the results obtained for the synthesis, the characterization of the partially substituted analogues of the tétra-para (guanidinoéthyl) calix[4]arene. The objective of this part is to understand the role and the importance of the number and the positioning of the guanidines functions on the calixarene core. In chapter III, are presented the various steps leading to the synthesis, the characterization and the evaluation by HPLC ways of new water-soluble prodrugs. The work is centered on a set of ionic derivatives of calix[4]arenes carriers of nalidixic acid. The prodrug behavior was demonstrated for one of them. The chapter IV presents the syntheses and characterizations of star-like poly-ionic structures are described, based on a benzene core, including a variable number of para (guanidinoéthyl) phényléther subunits. The chapter V is dedicated to the results of the biological evaluations of all the structure synthetized. For some compounds, viability and cellular toxicity were evaluated on eukaryotic cells. We show that many of the new prepared derivatives, prodrugs or not, present very interesting anti-bacterial activities

Full textThe vast ecosystem of our oceans provides an important source of biologically active marine product. The biological activity of these marine natural products is often startling potent cytotoxic properties. Peloruside A, a polyoxygenated 16-membered macrolide, was recently isolated from the sponge Mycale Hentscheli. Like Taxol®, peloruside A has shown microtubule-stabilizing activity and arrests cells in the G2/M phase. Moreover, this agent presents a unique binding site on tubulin and is less susceptible than Taxol® to multidrug resistance. These properties make this compound a promising candidate for the development of new anticancer agents. Thus, within the Cancéropôle Grand Ouest (CGO), a project on the synthesis of peloruside and its analogues, but also on the synthesis of hybrid between peloruside and epothilones, has been initiated. After a bibliographic analysis, synthesis of a racemate fragment has been developed and has allowed access to several hybrids which differ by their side chain. The preparation of analogues was also considered, but could not succeed. Macrolides obtained were then tested on the platform in vitro of Rennes. An enantioselective synthesis based on the same retrosynthetic analysis was performed

Full textLa schizophrénie est une maladie complexe et multidéterminée caractérisée par plusieurs difficultés, incluant des déficits de cognition sociale. La cognition sociale réfère à l’ensemble des processus cognitifs qui permettent de comprendre les autres et regroupe différents domaines tels que la théorie de l’esprit, la reconnaissance d’émotions et les connaissances sociales. Les recherches récentes se sont intéressées à identifier la présence d’anomalies de la cognition sociale chez les apparentés de patients, ce qui appuierait l’hypothèse de la cognition sociale comme un marqueur biologique relié à la schizophrénie. À l’aide de quatre études, la présente thèse vise à mieux comprendre les difficultés de cognition sociale présentes chez les apparentés de patients atteints de schizophrénie afin de mieux cibler les aspects qui pourraient être des marqueurs d’une vulnérabilité à développer la pathologie. Premièrement, en combinant les résultats d’études antérieurs, la thèse démontre que la cognition sociale est affectée chez les apparentés de patients (Chapitre 2, Chapitre 4). Deuxièmement, la thèse démontre que le domaine de la cognition sociale qui nécessite la prise en compte de plusieurs éléments pour bien comprendre les autres (théorie de l’esprit) est le domaine le plus affecté chez les apparentés de patients (Chapitre 3). Finalement, la thèse démontre que différents réseaux cérébraux sont impliqués dans la théorie de l’esprit (Chapitre 5) et que parmi ces réseaux, les apparentés de patients présentent des anomalies d’activation cérébrale autant lors du traitement des informations contextuelles et du traitement des informations sociales (Chapitre 6). La thèse appuie ainsi l’idée que certains aspects de la cognition sociale pourraient être des marqueurs d’une vulnérabilité génétique à développer la schizophrénie.
Schizophrenia is a complex and multidetermined illness characterized by several deficits, including impairments in social cognition. Social cognition refers to the cognitive processes involved in the ability to understand others and includes domains such as theory of mind, emotion perception and social knowledge. Recent literature investigated the presence of social cognition difficulties in unaffected relatives of patients with schizophrenia, which would support the hypothesis that social cognition is a biological marker of schizophrenia. Using four studies, the present thesis aims to better understand social cognition difficulties observed in unaffected relatives of patients with schizophrenia in order to target which aspects might be markers of a vulnerability to develop the illness. First, by combining the previous results of the literature, the thesis demonstrates that social cognition is affected in unaffected relatives of patients (Chapter 2, Chapter 4). Second, the thesis also suggests that the domain of social cognition that requires integrating multiple elements to correctly understand others (theory of mind) is the most impaired in unaffected relatives of patients (Chapter 3). Finally, the thesis demonstrate that different sub-brain networks are implicated in theory of mind (Chapter 5), and that among these networks, unaffected relatives present abnormal cerebral activation during contextual processing as well as social information processing (Chapter 6). The thesis thus supports the idea that certain aspects of social cognition could be biological markers associated with a vulnerability to develop schizophrenia.

Several synthetic strategies aiming at the pharmacomodulation on mono- and polycyclic series have been studied: in 2-methyl-5-nitrothiazole series, the anti-Trichomonas pharmacomodulation on position 4 by SRN1 strategy did not improve the activity previously demonstrated in 2-methyl-5-nitroimidazole series, but led to derivatives displaying a selective in vitro antiproliferative activity toward the HepG2 cell line. In 4-arylsulfonylmethyl-2-methylthiazole series, the pharmacomodulation on position 5, by Suzuki-Miyaura cross-coupling reaction on the one hand, and by direct arylation and intramolecular Knoevenagel reaction on the other hand, led to mono- and polycyclic derivatives among which some displayed an encouraging in vitro antiplasmodial activity. In 5H-thiazolo[3,2-a]pyrimidin-5-one series, a double Suzuki-Miyaura cross-coupling reaction revealed that the phenyl group on position 6 contributes to the antiplasmodial effect of this series. Finally, the in vitro biological evaluation of the thieno[3,2-d]pyrimidin-4(3H)-one scaffold let to characterize the pharmacophore responsible for the significant antiplasmodial activity. Some preliminary encouraging results regarding a mechanistic antiplasmodial study show the specific inhibition of plasmodial kinases, as a potential mechanism of action of some of these compounds
