Abstract:
Twenty-seven strains representing 10 Mucor species were assayed for their ability to utilize various substrates as carbon sources. They were also examined for their exoenzyme activities by means of the API-ZYM microtest system. Cluster analysis of resulting data revealed a loose correlation between these physiological patterns and the species designation of the isolates. Though different strains of the same species often vary in their substrate utilization abilities, the value of this approach to provide complementary data for the characterization of Mucor isolates has been demonstrated.Key words: Mucor, substrate utilization, API-ZYM, numerical taxonomy.

Abstract:
The closely related species Actinobacillus actinomycetemcomitans, Haemophilus aphrophilus, andHaemophilus paraphrophilus are common findings in oral microbiota. The aims of this study were to evaluate the applicability of the Rapid NH and API ZYM kits and arbitrarily primed PCR (AP-PCR) in the identification and differentiation of the three species from each other. The material included 62 clinical isolates and three reference strains of A. actinomycetemcomitans representing the 5 serotypes and 18 AP-PCR genotypes. Haemophilus species included 12 clinical isolates and 11 reference strains of H. aphrophilus, H. paraphrophilus, and 5 other species. For the PCR amplification, the oligonucleotide 5′-CAGCACCCAC-3′ was used as a primer. Contrary to the consistent performance of API ZYM, the Rapid NH system was able to identify only 10 of 65 (15%) A. actinomycetemcomitans isolates, whereas allHaemophilus species were correctly identified. The API ZYM test differentiated A. actinomycetemcomitans from H. aphrophilus and H. paraphrophilus by negative β-galactosidase and α-glucosidase reactions and a positive esterase lipase reaction. However, the API ZYM test was unable to differentiateH. aphrophilus from H. paraphrophilus, it also could not differentiate A. actinomycetemcomitans serotypes from each other. Among the H. aphrophilus isolates three AP-PCR genotypes and among H. paraphrophilus isolates only one AP-PCR genotype, distinct from those of A. actinomycetemcomitans, were found. The Rapid NH test showed poor ability to identify clinical isolates of all A. actinomycetemcomitans serotypes. Moreover, AP-PCR genotyping proved to be a rapid method for the species differentiation of A. actinomycetemcomitans, H. aphrophilus, and H. paraphrophilus.

The API ZYM system was used to identify the bacterial fish pathogens Enterococcus seriolicida, β-hemolytic Streptococcus sp., Renibacterium salmoninarum, Aeromonas salmonicida, A. hydrophila, Pasteurella piscicida, Edwardsiella tarda, Vibrio anguillarum, Pseudomonas fluorescens, Flexibacter columnaris, and F. maritimus. As additional tests, Gram stain and the activities of cytochrome oxidase and catalase were examined. The results of APS ZYM and the additional tests were coded for easy identification. Several biotypes were demonstrated by Streptococcus sp., V. anguillarum, and A. hydrophila. Other bacteria showed uniform profiles. This system can distinguish each fish pathogen from all other bacterial species examined.

Abstract:
Malassezia spp. são fungos lipofílicos, reconhecidos há mais de cem anos como membros da microbiota cutânea normal humana, bem como agentes de doenças dermatológicas. Desde 1980, são relatados como causadores de infecções sistêmicas oportunistas. Estas leveduras são frequentemente encontradas em uma grande variedade de animais homeotérmicos, e sua ampla distribuição no meio ambiente agora está sendo explorada através de técnicas moleculares e bioquímicas. O enfoque deste estudo foi a espécie não lipido-dependente Malassezia pachydermatis, e a sua caracterização através do perfil enzimático pelo sistema Api-Zym®. Deste modo, utilizaram-se 30 amostras de M. pachydermatis isoladas de cães, com e sem sinais clínicos, identificadas pela técnica de Reação em Cadeia da Polimerase (PCR). As amostras foram cultivadas em meio de Dixon modificado, incubadas a 37°C durante 48 horas, para posterior quantificação através de espectrofotometria. Um inóculo de 65 μl foi colocado em cada orifício do kit comercial de verificação enzimática e, após, incubado a 37ºC por 4 horas. Os resultados foram obtidos através da leitura visual e interpretação das titulações por escalas pré-determinadas, também foram avaliadas as reações através de “escaneamento” eletrônico. Todos os testes foram realizados em triplicata. Os resultados demonstraram a atividade de enzimas específicas pertencentes ao grupo das peptídeo hidrolases (100%), fosfohidrolases (98,3%) e éster hidrolases (91,6%), enquanto que as enzimas α-galactosidase, β-galactosidase, β-glicuronidase, α-manosidase e α-fucosidase caracterizaram 10% de todos os isolados. Além disso, não se comprovou a viabilidade do sistema utilizado no estudo para a caracterização enzimática da levedura.
Malassezia spp. are lipophilic fungi, which have been recognised for over a century as members of the normal human cutaneous flora, and also as agents of certain skin diseases. In addition, since the 1980s, they have been reported as causing opportunistic systemic infections. These yeasts have frequently been found on a diverse range of other warm-blooded animals, and their wider distribution in nature is now being explored using molecular techniques. The focus of the present study was the species no lipid-dependent Malassezia pachydermatis, and its characterization through the enzymatic profile by the Api-Zym® system. Then 30 isolated samples of dogs had been used, with and without clinical signals, identified by the technique of Polymerase Chain Reaction (PCR). The samples were cultivated into Dixon’s modified medium, incubated at 37°C during 48 hours, for posterior quantification through spectrophotometry. An inocula of 65 μl was placed in each microwell of the commercial kit of enzymatic verification and, after, it was incubated at 37ºC for 4h. The results were obtained through the visual reading and interpretation of the titulations by predetermined scales, it were also evaluated the reactions through electronic scanning. All the tests were performed in triplicate. The results demonstrated the activity of specific enzymes pertaining to the group of the peptide hydrolases (100%), phosphohydrolases (98,3%) and ester hydrolases (91,6%), while the enzymes α-galactosidase, β-galactosidase, β-glucuronidase, α-mannosidase and α-fucosidase characterized 10% off all the isolates. Moreover, it was not proven the viability of the system used in the study for the enzymatic characterization of the yeast.

Abstract:
Pythium insidiosum, classificado no Reino Stramenipila e Classe Oomycetes, é o agente etiológico da pitiose, uma doença diagnosticada principalmente em equinos, caninos e humanos. A secreção de enzimas por micro-organismos é considerada um fator importante na invasão tecidual. O presente estudo teve como objetivo analisar a atividade enzimática de isolados de P. insidiosum obtidos de lesões equinas (n=13), coelhos infectados experimentalmente (n=2) e uma amostra padrão (ATCC). Para isso, utilizou-se o kit comercial API ZYM. Zoósporos foram cultivados em caldo RPMI 1640 por 24h a 37 oC. Após esse período, a presença de hifas foi observada, as quais foram separadas e 65μl do líquido restante foram inoculados em cada um dos 20 poços (um controle e 19 com substratos específicos para cada enzima) do kit. As galerias foram então incubadas por 4 h a 37 oC. Os resultados foram obtidos através da leitura visual das reações. As análises foram feitas em triplicata e submetidas à análise de variância utilizando um nível de significância de 5%. Houve uma variação intraespecífica na atividade enzimática entre os isolados, sendo que enzimas dos grupos fosfohidrolases e éster hidrolases foram as mais prevalentes. Esses dados demonstram a capacidade de P. insidiosum em secretar enzimas que degradam substratos presentes em animais e em plantas, bem como a variabilidade enzimática entre os isolados.
Pythium insidiosum is classified in the kingdom Stramenipila, class Oomycetes. It causes pythiosis, a disease mainly diagnosed in horses, dogs, and humans. This study aimed at analyzing the enzymatic activity of P. insidiosum isolates obtained from equine lesions (n=13), experimentally infected rabbits (n=2) and a standard strain (ATCC 58637). To address this issue, the API ZYM commercial kit was used. Zoospores were cultivated in RPMI 1640 broth for 24 h at 37oC. After this period, the presence of hyphae was observed and they were carefully separated from the liquid phase. Then, 65 μl of this liquid were inoculated in each of the 20 microwells (one control, 19 containing specific substrates for each enzyme) of the API ZYM kit. The strips were incubated for 4h at 37oC. Results were obtained by visual observation of the reactions. The tests were carried out in triplicate and submitted to an analysis of variance using a significance level of 5%. An intraspecific variation among the enzymatic activities was observed among the isolates, where the group of phosphohydrolases and ester hydrolases were conspicuous among most of the isolates. Data here obtained highlighted the capacity of P. insidiosum in secreting enzymes capable of degrading substrates present in animals and plants, besides the enzymatic variability among the isolates.

Abstract:
La diversité fongique des meules de plusieurs espèces de Macrotermitinae a été analysée au niveau taxinomique, fonctionnel et génétique à l'aide d'une approche polyphasique associant plusieurs techniques complémentaires. L'objectif étant d'évaluer la spécificité des taxons fongiques associés aux meules ainsi que les relations qu'ils entretiennent avec les termites champignonnistes. Une grande variété de phylotypes cultivables appartenant majoritairement au phylum des Ascomycètes a été obtenue par isolement et séquençage des ITS fongiques, et peu de séquences se sont révélées être spécifiques à un genre de Macrotermitinae particulier. Les profils physiologiques obtenus ont mis en évidence la nature saprophytique de la majorité des phylotypes et confirmé l'absence de taxons spécifiques. Par PCR-DGGE de l'ADN total de meules, 100% des phylotypes ITS et 28S fongiques identifiés étaient affiliés au genre Termitomyces. La technique Suicide Polymerase Endonuclease Restriction (SuPER) a été adaptée à la mycoflore des meules pour limiter l'impact de l'ADN majoritaire du Termitomyces symbiotique. Celle-ci a permis la détection de plusieurs autres populations fongiques. Les analyses phylogénétiques ont montré d'une part la spécificité des Xylaria associés aux meules de Macrotermitinae bien qu'aucune co-évolution n'ait été observée avec les termites hôtes et d'autre part leur affiliation dans un sous-genre spécifique. Une analyse préliminaire des facteurs d'inhibition a également révélé l'implication des termites dans la régulation des communautés fongiques des meules. Dans leur ensemble, nos résultats illustrent clairement l'influence des Macrotermitinae sur les communautés fongiques telluriques pendant leurs différentes activités.

Zusammenfassung Mobile Health hat bereits eine über 10-jährige Forschungshistorie hinter sich. Kommend aus dem Bereich des Quantified-Self zur Dokumentation eigener Gesundheitsparameter, sollte insbesondere die Forschungsfrage der Compliance den Schulterschluss zur medizinischen Anwendung schaffen. Phänomene wie die „App-Attrition“, also die Abnutzung der App-Nutzung, erfragen jedoch nach Anreizmodellen für eine dauerhafte Nutzung, wie sie gerade bei Chronikern notwendig sind. Daten – das „neue Blut“ – scheinen das Potenzial zu haben Mehrwerte für den Patienten wie auch für die Leistungserbringer generieren zu können. Die Autoren zeigen, wie mHealth mit Aspekten der Datenverarbeitung zur Verbesserung der medizinischen Versorgung zusammenspielen kann. Das führende Konzept bilden die sog. Digitalen Biomarker – validierbare Algorithmen, welche die durch Sensoren und Apps erfassten Daten verarbeiten und quantifizierte Aussagen zum Gesundheitszustand treffen können. Zwei Fallbeispiele zeigen, wie ein solcher Systemansatz im Bereich der Epilepsie und der Parkinsontherapie unterstützen kann.

Der Bereich Sachrechnen wird immer wieder als Problembereich des Mathematikunterrichts beschrieben. Zum Aufbau und zur Förderung der Sachrechenkompetenz von Schülerinnen und Schülern bedarf es stetiger und vielfältiger Aktivitäten. Im folgenden Beitrag wird aufgezeigt wie die Förderung der Sachrechenkompetenz durch das Erfinden von Sachaufgaben realisiert werden kann. Mit der Tablet-App Book Creator erstellen die Schülerinnen und Schüler digitale Bücher, in denen die erfundenen Sachaufgaben in verschiedenen Repräsentationsebenen abgebildet werden. Zunächst werden die allgemeinen Funktionen der Tablet App und deren Einsatzmöglichkeiten im Unterricht thematisiert. Zudem werden weitere Möglichkeiten des Einsatzes der App zu verschiedenen Inhalten im Mathematikunterricht der Primarstufe vorgestellt.

In diesem Beitrag werden zentrale didaktische Aspekte zum Sachrechenunterricht der Grundschule skizziert. Dabei wird das Sachrechnen als integriertes Modellieren und Problemlösen aufgefasst. Der auf diesem Verständnis fußende Lösungsprozess lässt sich mit Stop-Motion-Filmen schon im Anfangsunterricht sehr gut begleiten. Mögliche Realisierungen dieses Zusammenspiels werden anhand der App „Stop Motion Studio“ aufgezeigt.

Ausgehend von theoretischen Überlegungen werden eine digital unterstützte Lernumgebung zu Rechendreiecken sowie die Pilotierung dieser Lernumgebung und ausgewählte Ergebnisse dargestellt.
