Full textThe antioxidants of natural origin have an important role on food science, because they may substitute the synthetic antioxidants. The phenolic compounds of vegetables play an important role as oxidation inhibitors. The pomegranate (Punica granatum, L.) is rich in phenolic compounds as anthocianins, flavonoids and phenolic acids. The antioxidant activity of extracts and acid phenolic fractions obtained from the pulp and seeds of brazilian pomegranate (from Pernambuco) was evaluated, by the co-oxidation of β-carotene and linoleic acid assay and the Rancimat method. The kinetic behavior of the antioxidant activity of the extracts and acid phenolic fractions by the co-oxidation of β-carotene and linoleic acid test was evaluated, too. It was noted that the aquous extracts from the pulp and seeds have significant antioxidant activity at slow concentration. It was possible by the high content of phenolic compounds present on those extracts. The kinetic behavior showed that the aquous extracts was the most efficient along the oxidative process, showing larger stability than the antioxidant BHT. The pulp\'s of bound acid phenolic fraction by soluble compounds was the most efficient among the acid phenolic fractions, and it has major amount of phenolic compounds. The pomegranate seed has the punicic acid on its fatty acid composition. The punicic acid is a I conjugated inolenic acid isomer.The punicic acid is present at 58% from the total fatty acids from the fruit, being its principal fatty acid.

Full textThe aims of this work were to evaluate the in vitro antioxidant capacity of listed sunflower cotyledon extracts, obtained by a sequential extraction with solvents of different polarities, and to evaluate the in vitro antioxidant potential of the sample extract with highest in vitro capacity. Ali the in vitro parameters (β-carotene/linoleic acid system, FRAP, OPPH, ORAC and Rancimat methods) indicated the aqueous extract (EAq) as the extract with highest capacity. In this extract, the chlorogenic acid (12.88%) was identified as the principal fraction of phenolic acids. In the in vitro antioxidant capacity evaluation, both determinations used (TBARS and fatty acids profile) gave indication that the EAq was capable to exerce a protective effect on the polyunsaturated fatty acids of the adipose, cerebral, hepatic and plasm tissues of Wistar male rats just-weaned.

Full textFree radicals are chemical species that react rapidly with various compounds and target cells, as they have a very short half life and are highly unstable. The formation of these compounds consists of a continuous, physiological action, which includes essential biological functions and occurs through the loss or addition of a single electron to a non-radical compound. These reactions may occur in biochemical processes of the immune system, or by chemical reactions, causing damage to the cells through the destruction of components such as proteins, lipids, sugars and nucleotides. It is known that compounds exist which are effective against these species, preventing damage caused by oxidative stress. The object of this work was to study heterocyclic compounds that have nitrogen in their structure (azoles), which appear in the literature as exemplary molecules of compounds with a wide spectrum of pharmacological applications. Of these compounds, derivatives of pyrazoles (26 compounds), thiazoles (7 compounds) and 1 isoxazole (3,5-dimethylisoxazole) were analyzed. These 34 compounds were evaluated by the ORAC (Oxygen Radicals Absorption Capacity) in order to determine and/or evaluate its antioxidant potential. The choice of ORAC method is based on the fact that the molecules studied have hydrophilic and lipophilic characteristics, as well as a method validated by the literature, which is available and widely used. The ORAC method evaluates the antioxidant capacity of the sample, measuring its ability to protect the fluorescence (FL) of the oxidation by the AAPH in the reaction medium. AAPH is a generator of free radicals which, at 37°C, removes hydrogen from the medium, promoting the reduction of fluorescence from fluorescein in λ measured by time. Six compounds present good to moderate antioxidant activity: 3,5-dimethyl-1H-pyrazole (2.382 µmol eq.Trolox/g); 3-phenyl-5-(4-fluorphenyl)-1-thiocarbamoyl-4,5-dihydro-1H-pyrazole (6.354 µmol eq.Trolox/g); 3-phenyl-5-(2-methoxyphenyl)-1-thiocarbamoyl-4,5-dihydro-1H-pyrazole (8.739 µmol eq.Trolox/g); 5-(2,4-diclorophenyl)-3-phenyl-1-thiocarbamoyl-4,5-dihydro-1H-pyrazole (6.022,226 µmol eq.Trolox/g); 2-[5-(4-methoxyphenyl)-3-phenyl-4,5-dihydro-1H-pyrazole-1-il]-4-phenylthiazole (3.135 µmol eq.Trolox/g); and finally, 2-[5-(3-nitrophenyl)-3-phenyl-4,5-dihydro-1H-pyrazole-1-il]- 4-phenylthiazole (2.700 µmol eq.Trolox/g). Experiments with the ORAC method for the azoles studied present reproducibility in the experimental execution, and have proven to be a viable alternative for studies of synthetic molecules with antioxidant potential.

Resumo: O estresse oxidativo e as inflamações estão associados a muitas doenças como diabetes, câncer, doenças cardiovasculares e neurodegenerativas, incluindo a aterosclerose, causando preocupação em todo o mundo. Como alternativa, as plantas podem desempenhar um papel chave no combate a essas desordens devido suas propriedades farmacológicas que são relacionadas a uma ampla gama de compostos com atividade antioxidante, entre eles, os compostos polifenólicos. Nesse contexto as pimentas podem ser uma alternativa para a obtenção desses compostos. Porém, a concentração desses metabólitos secundários é baixa e afetada por diversos fatores abióticos. Como alternativa para contornar esse problema foram utilizadas metodologias relacionadas com as culturas in vitro de células e de tecidos vegetais. O presente trabalho teve como objetivo padronizar a obtenção de explantes (Capítulo 1) a serem utilizados em uma cultura de calos otimizada através de uma abordagem sistêmica (Capítulo 2). No capítulo 1, por meio de um delineamento experimental fatorial completoadois níveis (24) determinou-se numericamente o efeito de quatro fatores [O tipo de solução para condicionamento osmótico (água e solução aquosa de KNO3 a 1%), meio germinativo (ágar e ágar + ácido giberélico a 1,88 mg.L-1), tempo (15 e 30 dias) e dois genótipos (Pitanga e Cambuci)] nas taxas de surgimento de cotilédones e na germinação. Abstract: The oxidative stress and inflammations are associated with many diseases such as diabetes, cancer, cardiovascular and neurodegenerative diseases including atherosclerosis causing concern worldwide. As an alternative, the plants can play a key role due to their pharmacological properties against these disorders; these attributes are most of the times associated with the presence of polyphenolic compounds. In this context, peppers can be an alternative to obtain these compounds. However, the presence of secondary metabolites is low and affected by several abiotic factors. As an alternative to circumvent this problem, methodologies related to in vitro cultures of plant cells and tissues were applied. The objective of the present work was to standardize the extraction of explants (Chapter 1) to be used in a callus culture optimized through a systemic approach (Chapter 2). In Chapter 1, through a two-level full factorial experimental design (24 ) was numerically determined the effect of four factors [The solution type for osmotic conditioning (water and 1% KNO3 aqueous solution), germination medium (agar and agar + gibberellic acid at 1.88 mg.L -1, GA3), post-sowing time (15 and 30 days) and two genotypes (Pitanga and Cambuci)] in cotyledon emergence and germination rates. The genotypes responses to the osmotic conditioning solution and to the germination medium were different. The optimal combination of treatments for in vitro germination and development of seedlings for Pitanga and Cambuci was water + agar and water + agar-GA3, respectively. Resumo: O excesso na geração de espécies reativas como os radicais livres pode resultar num desequilíbrio que, embora benéfico em situações como o combate a micro-organismos patógenos, está implicado com diversas patologias crônicas e com o envelhecimento. Assim é comum a indicação de compostos antioxidantes para a prevenção destas patologias e inclusive, algumas pesquisas com substâncias antioxidantes já resultaram no desenvolvimento de novos medicamentos. Desta forma, o estudo de fontes naturais que apresentam substâncias ativas sobre as diferentes espécies oxidantes geradas em nosso organismo é de grande importância, visto as implicações destas em várias patologias. Entre estas fontes, Agaricus blazei Murrill é um cogumelo comestível e medicinal, popularmente utilizado na prevenção do envelhecimento e no tratamento de diversas doenças, como câncer, artrite reumatóide, artrose, aterosclerose, diabetes e hipertensão. Tem sido relatado também, o uso de outros cogumelos como Agaricus bisporus (champignon), Lentinus edodes (shiitake) e Pleurotus ostreatus (shimeji ou hiratake), de forma similar ao Agaricus blazei. Além da função nutricional desse cogumelo é importante verificar possíveis atividades biológicas, o que poderia render novos produtos com alegações de propriedades funcionais ou terapêuticas, como as já existentes para o A. blazei. Assim, este projeto objetivou o estudo do perfil de atividade antioxidante (químico, enzimático e ex vivo) e análise de atividade antibacteriana de três cogumelos cultivados no Brasil em comparação com o Agaricus blazei Murrill. Abstract: The excessive generation of reactive oxygen species, like free radicals, can lead to an imbalance which, although benefical in such situations to combat pathogenic microorganisms, is involved with various chronic diseases and aging. Thus, it is common to indicate the use of antioxidants for prevention of these diseases, and some research about antioxidant products have already resulted in the development of new medicines. Thus, the study of natural sources that contain active compounds over the different oxidant species, generated in our bodies, is of great importance, due their implications in various pathologies. Among these sources, Agaricus blazei Murrill is an edible and medicinal mushroom, commonly used in the prevention of aging and the treatment of various diseases such as cancer, rheumatoid arthritis, osteoarthritis, atherosclerosis, diabetes and hypertension. It has also been reported, the use of other Agaricus bisporus (champignon), Lentinus edodes (shiitake) and Pleurotus ostreatus (shimeji or hiratake), similar to Agaricus blazei. Besides its nutritional properties, it is important to check possible biological activities, which could render functional property claims as those existing for the A. blazei. Thus, mushrooms such as this project aims to study the profile of antioxidant activity (chemical, enzymatic and ex vivo) of mushrooms cultivated in Brazil, comparing to Agaricus blazei Murrill. Antioxidant activities were evaluated through DPPH·, ABTS·+, O2 ·-, NO, H2O2 and HOCl and also by analysis of the oxidative system catalyzed by myeloperoxidase (MPO) and horseradish peroxidase (HRP). Full textThe antioxidant activity of two commercial extracts, rosemary and green tea, were compared with the activity of the synthetic BHA for total replacement of the synthetic in chicken burger. The spectrometric quantification was determined by UV - VIS methods: Folin - Ciocalteu , FRAP and DPPH . In the Folin-Ciocalteau method the BHA showed no difference with green tea (p > 0.05), in the FRAP method the BHA obtained better (p < 0.05) performance and for DPPH the green tea showed greater activity (p < 0.05). According to the analysis of antioxidant activity, dosages of natural extracts were determined, using as basis the limit dosage of BHA (0.01 % fat base) and applied in chicken burgers: T1: Control, T2: 0.002 % BHA, T3: 0.0038 % Green Tea, T4: 0.001 % Green Tea, T5: 0.048 % Rosemary and T6: 0.00186 % Rosemary. Chemical-physical analysis of chemical composition and pH were made, and also stability analysis: cooking loss and reducing the diameter, TBARs index, objective color (parameters L *, a * and b *) and sensory acceptance test. The results of TBARS showed a significant difference between samples and also samples interaction x storage time (p < 0.05), after 120 days the test with higher dosage of rosemary extract (T5) did not differ from synthetic (T2) (0.423 and 0.369 TBARS index, respectively), thus better performance than the other tests. The sensory evaluation results showed that the samples did not differ during the storage period (p > 0.05). According the experimental conditions can be concluded that the commercial rosemary extract can completely replace the antioxidant BHA.

A planta selecionada para este estudo, Garcinia brasiliensis (família Clusiaceae), apresentou perfil químico com expressiva ocorrência de benzofenonas, conhecidas por seu potencial antimalárico, antioxidante, antibacteriano e antinflamatório. Os objetivos deste trabalho incluíram o estudo químico/biológico dos caules e folhas de Garcinia brasiliensis, com o isolamento e determinação estrutural de metabólitos secundários e avaliação de sua bioatividade. Apesar das plantas medicinais serem raramente utilizadas como antioxidantes na medicina popular, suas propriedades terapêuticas podem ser devidas a sua capacidade de sequestrar radicais livres de oxigênio, envolvidos em diversas patologias decorrentes do estresse oxidativo em sistemas biológicos. A ação antioxidante tem sido também apontada como forte indicativo de ação quimiopreventiva, associada à indução de enzimas destoxificantes do organismo como a quinona redutase, que pode evitar eventos iniciais da geração de tumores. O estudo fitoquímico dos extratos de folhas e galhos de G. brasiliensis resultou no isolamento ou identificação de diversas substâncias polifenólicas a partir de frações polares: os biflavonoides amentoflavona, podocarpusflavona, fukugetina, além de procianidina e seus trímeros e da benzofenona garcinol através do uso de técnicas cromatográficas e espetrométricas como CLAE-DAD, CLAE-EM, UV, IV e RMN. The phytochemical investigation of plant species contributes to conservation and sustainable use of São Paulo biodiversity. This project has addressed the phytochemical investigation of stems and leaves of Garcinia brasiliensis, with isolation, structural determination of secondary metabolites as well as evaluation of bioactivity. G. brasiliensis is a member of the Clusiaceae family with interesting secondary metabolites such as benzophenones which have potent antimalarial, antioxidant, antitumoral, antibacterial, antiinflamatory and chemoprevention capacity. Whereas medicinal plants are not used as antioxidants in popular medicine their therapeutic properties are derived from capacity to scavenge free radicals involved in oxidative stress. The antioxidant activity is indicative of chemoprevention associated with toxic enzymes such as quinone reductase that is associated with inhibition of tumor development. The phytochemical study of extracts from leaves and stems of G. brasiliensis led to the isolation or identification of several polyphenols from polar fractions: biflavonoids amenthoflavone, podocarpusflavone, fukugetin, in addition to procyanidin and its trimers and the benzophenone garcinol through chromatographic and spectrometric techniques as HPLC-DAD, HPLC MS, UV, IR and NMR. Low polarity extracts were analyzed by GC-FID and evidenced the presence of triterpenes and steroids, including stigmasterol, -amirinone, -amirin, taraxerol and lupeol, as major compounds. Metabolite target analysis was carried out using thin layer chromatographic tests with -carotene and anisaldehyde as revealing agents aiming the detection of antioxidant compounds, in addition to inhibitory activity against T. cruzi, human and plant pathogenic fungi, malaria, acetylcholinesterase, and HIV protease. The development of new drugs with antioxidant action has been an important subject of study, since the current drug therapy for the treatment of diseases such as Alzheimer's, Parkinson's, multiple sclerosis, Huntington's disease, epilepsy and some types of cancer can be performed because the antioxidant properties of many natural and / or synthetic compounds. Thus, the search for new antioxidants is extremely necessary and is currently the subject of interest in new research by the cosmetic, pharmaceutical and nutritional industry. In this context, this work will enable increased knowledge on Moringa oleifera by evaluating the antioxidant potential, sunscreen and anti-glycation of the extracts of this species, besides elucidating ethnobotanical reports of its use and enrich the scientific knowledge of this widely used species with medicinal plant. A planta selecionada para este estudo, Garcinia brasiliensis (família Clusiaceae), apresentou perfil químico com expressiva ocorrência de benzofenonas, conhecidas por seu potencial antimalárico, antioxidante, antibacteriano e antinflamatório. Os objetivos deste trabalho incluíram o estudo químico/biológico dos caules e folhas de Garcinia brasiliensis, com o isolamento e determinação estrutural de metabólitos secundários e avaliação de sua bioatividade. Apesar das plantas medicinais serem raramente utilizadas como antioxidantes na medicina popular, suas propriedades terapêuticas podem ser devidas a sua capacidade de sequestrar radicais livres de oxigênio, envolvidos em diversas patologias decorrentes do estresse oxidativo em sistemas biológicos. Abstract: The phytochemical investigation of plant species contributes to conservation and sustainable use of São Paulo biodiversity. This project has addressed the phytochemical investigation of stems and leaves of Garcinia brasiliensis, with isolation, structural determination of secondary metabolites as well as evaluation of bioactivity. G. brasiliensis is a member of the Clusiaceae family with interesting secondary metabolites such as benzophenones which have potent antimalarial, antioxidant, antitumoral, antibacterial, antiinflamatory and chemoprevention capacity. Whereas medicinal plants are not used as antioxidants in popular medicine their therapeutic properties are derived from capacity to scavenge free radicals involved in oxidative stress. The antioxidant activity is indicative of chemoprevention associated with toxic enzymes such as quinone reductase that is associated with inhibition of tumor development. The phytochemical study of extracts from leaves and stems of G. brasiliensis led to the isolation or identification of several polyphenols from polar fractions: biflavonoids amenthoflavone, podocarpusflavone, fukugetin, in addition to procyanidin and its trimers and the benzophenone garcinol through chromatographic and spectrometric techniques as HPLC-DAD, HPLC MS, UV, IR and NMR. Low polarity extracts were analyzed by GC-FID and evidenced the presence of triterpenes and steroids, including stigmasterol, -amirinone, -amirin, taraxerol and lupeol, as major compounds. Metabolite target analysis was carried out using thin layer chromatographic tests with -carotene and anisaldehyde as revealing agents aiming the detection of antioxidant compounds, in addition to inhibitory activity against T. cruzi, human and plant pathogenic fungi, malaria, acetylcholinesterase, and HIV protease. As mudanças climáticas globais podem alterar significativamente o metabolismo das células vegetais. Um cenário com alta concentração de CO2 atmosférico pode ser benéfico para plantas do tipo C3 devido ao estímulo à fixação de carbono. No entanto, esse aumento esperado na taxa de assimilação de carbono também pode aumentar a atividade e a expressão das enzimas envolvidas na cadeia de transporte de elétrons, resultando em maior acúmulo de espécies reativas de oxigênio. Nesse trabalho, foi estudada a resposta do sistema antioxidante em cloroplastos de Eucalyptus urophylla cultivados sob altas concentrações de CO2. As plantas de E. urophylla expostas a alta concentração de CO2 (980 ppm) mostraram um aumento no fechamento estomático e pequena, mas significativa, indução do estresse oxidativo em relação as plantas cultivadas em concentração atmosférica de CO2. Entretanto, essas respostas não foram observadas quando as plantas foram cultivadas em concentração de CO2 igual a 680 ppm, cujo tratamento induziu abertura estomática. Através de uma abordagem de proteômica de descoberta, foram identificadas e detectadas 19 proteoformas antioxidantes cloroplastidiais e 2 proteoformas diferencialmente reguladas. Dentre essas, destacam-se uma proteoforma de ascorbato peroxidase e uma de superóxido dismutase, as quais estão possivelmente envolvidas na neutralização de espécies reativas de oxigênio após cultivo em atmosfera enriquecida com CO2, e que podem ser utilizadas como marcadores bioquímicos de estresse abiótico ou como alvos em programas de engenharia genética.
Global climate change can significantly alter plant cell metabolism. A higher atmospheric CO2 scenario may be beneficial for C3 plants through the stimulation of carbon fixation. However, this predicted increase in the rate of carbon assimilation may also increase the activity and expression of enzymes involved in the electron transport chain, resulting in higher accumulation of reactive oxygen species. Here, we studied the responses of the chloroplast antioxidant system of Eucalyptus urophylla plants cultivated under high-CO2 conditions. E. urophylla plants exposed to a high concentration (980 ppm) of CO2 showed an increase in stomatal closure and a small, but significant, induction of oxidative stress in relation to plants grown at atmospheric CO2 concentration. However, these responses were not observed at a CO2 concentration equal to 680 ppm, which induced stomatal aperture. With the discovery proteomics approach used herein, we identified 19 chloroplast antioxidant proteoforms and pinpointed 2 differentially regulated antioxidants proteoforms. We highlight an isoform of ascorbate peroxidase and one of superoxide dismutase, which are possibly involved in the neutralization of reactive oxygen species upon cultivation in a high CO2 atmosphere and could be used as biochemical markers of abiotic stress or as targets in genetic engineering programs.

O excesso na geração de espécies reativas como os radicais livres pode resultar num desequilíbrio que, embora benéfico em situações como o combate a micro-organismos patógenos, está implicado com diversas patologias crônicas e com o envelhecimento. Assim é comum a indicação de compostos antioxidantes para a prevenção destas patologias e inclusive, algumas pesquisas com substâncias antioxidantes já resultaram no desenvolvimento de novos medicamentos. Desta forma, o estudo de fontes naturais que apresentam substâncias ativas sobre as diferentes espécies oxidantes geradas em nosso organismo é de grande importância, visto as implicações destas em várias patologias. Entre estas fontes, Agaricus blazei Murrill é um cogumelo comestível e medicinal, popularmente utilizado na prevenção do envelhecimento e no tratamento de diversas doenças, como câncer, artrite reumatóide, artrose, aterosclerose, diabetes e hipertensão. Tem sido relatado também, o uso de outros cogumelos como Agaricus bisporus (champignon), Lentinus edodes (shiitake) e Pleurotus ostreatus (shimeji ou hiratake), de forma similar ao Agaricus blazei. Além da função nutricional desse cogumelo é importante verificar possíveis atividades biológicas, o que poderia render novos produtos com alegações de propriedades funcionais ou terapêuticas, como as já existentes para o A. blazei. The excessive generation of reactive oxygen species, like free radicals, can lead to an imbalance which, although benefical in such situations to combat pathogenic microorganisms, is involved with various chronic diseases and aging. Thus, it is common to indicate the use of antioxidants for prevention of these diseases, and some research about antioxidant products have already resulted in the development of new medicines. Thus, the study of natural sources that contain active compounds over the different oxidant species, generated in our bodies, is of great importance, due their implications in various pathologies. Among these sources, Agaricus blazei Murrill is an edible and medicinal mushroom, commonly used in the prevention of aging and the treatment of various diseases such as cancer, rheumatoid arthritis, osteoarthritis, atherosclerosis, diabetes and hypertension. It has also been reported, the use of other Agaricus bisporus (champignon), Lentinus edodes (shiitake) and Pleurotus ostreatus (shimeji or hiratake), similar to Agaricus blazei. Besides its nutritional properties, it is important to check possible biological activities, which could render functional property claims as those existing for the A. blazei. A família Myrtaceae apresenta ampla distribuição, ocorrendo, preferencialmente, nas zonas tropicais e subtropicais, e é considerada uma das mais importantes famílias da flora brasileira em função da larga ocorrência de espécies comestíveis e/ou usadas na medicina tradicional. Eugenia jambolana Lam. é conhecida no Brasil como jambolão, e é de grande interesse pelas aplicações medicinais, especialmente de suas folhas e frutos, no tratamento da diabetes. O trabalho tem como objetivo a investigação fitoquímica das folhas e frutos de E. jambolana monitorado por testes para detecção de atividade potencialmente antioxidante, através dos métodos de descoramento do b-caroteno e seqüestro do radical DPPH. Folhas e frutos da planta em estudo foram coletados na Faculdade de Ciências e Letras - UNESP/CAr. Foi obtido o extrato etanólico das folhas que foi particionado com solventes de diferentes polaridades. Para a obtenção do extrato bruto dos frutos (casca e polpa) foi utilizado MeOH (1% HCl). O estudo fitoquímico das folhas através de técnicas cromatográficas usuais (cromatografia em coluna, CLAE e cromatografia em contra corrente de alta velocidade) resultou no isolamento de dois flavonóides que apresentaram atividade antioxidante em ensaio com solução de b-caroteno. Os flavonóides isolados foram identificados como 4'-Oramnopiranosídeo- 3',5,5',7-tetraidroxi-flavona (1) e miricetrina (2). Análises em CLAE-DAD das frações provenientes do extrato bruto das frutas evidenciaram a presença de três antocianinas majoritárias e uma minoritária, que foram detectadas por análises de CLAE-DAD e CLAE-MS, e em comparação com dados da literatura, proporcionou a identificação das mesmas como cianidina-3-xilosídeo, cianidina-3- glucosídeo, cianidina-3-rutinosídeo e cianidina-3-(6''-acetoil)-glicosídeo. The Myrtaceae family is widely distributed and occurs, mostly, in tropical and subtropical areas. It is one of the most important families of the Brazilian flora owing to the wide occurrence of edible species and/or species used in the traditional medicine. Eugenia jambolana Lam. is known in Brazil as jambolão and is of great interest for its folk medicinal applications, especially of their leaves and fruits, in the treatment of diabetes. Leaves and fruits of this species have been collected at UNESP (University Campus). The ethanol extract of the leaves was partioned with organic solvents and the crude extract of the fruits (skin and pulp) was obtained with acidified methanol (1% HCl). The phytochemical study of the leaves extract using chromatographic techniques (column chromatography, TLC, HPLC and HSCCC) resulted in the isolation of two flavonoids which presented strong antioxidant activity in the TLC assay using b-carotene solution as revealing agent. The isolated flavonoids have been identified as 4'-Oramnopyranoside- 3',5',5,7-tetrahydroxy-flavone (1) and myricetrin (2). Their strong antioxidant activity could be related with the presence of hydroxy groups in ring B which provides increased stability to the radical formed after donation of a hydrogen radical, and also the presence of the _,_-unsaturated carbonyl moiety on C ring.. HPLC-DAD analyses of the fractions obtained from the crude extract of the fruits evidenced the presence of three major anthocyanins and a minor one, which been identified by analyses of HPLC-DAD and HPLC-MS data, and comparison with literature. They were identified as cyanidin-3-xyloside, cyanidin-3-glucoside, cyanidin-3-rutinoside and cyanidin-3- (6''-acetoyl)-glucoside. Este trabalho objetivou determinar o efeito de diversos tratamentos de sanitização na qualidade físico-química, fitossanitária e o teor de antioxidantes (poliaminas, fenóis, flavonóides, ácido ascórbico, clorofila) além do potencial antioxidante de brócolis orgânicos e convencionais e qualidade enzimática de trinta e uma variedades de radicchio. Os brócolis orgânicos e convencionais foram adquiridos de produtores locais do município de Botucatu/SP. Os vegetais foram lavados, higienizados com água clorada e ozonizada por 5 e 10 minutos e armazenados em câmara fria a 6±1°C por sete dias. As coletas foram realizadas a cada três dias. O modo de cultivo aliado aos tratamentos de lavagem influenciou significativamente AT e os SS. Os tratamentos com água ozonizada por 5 e 10’ foram eficazes na diminuição da carga microbiana nos brócolis tratados. Não houve influencia do modo de cultivo nos teores de Cu, Zn e Mn. O uso do ozônio não alterou as qualidades físico-químicas dos brócolis, independente do modo de cultivo, o que pode ser promissor, como alternativa para uso como sanitizante. Houve influencia dos tratamentos de sanitização no período de avaliação para os antioxidantes, poliaminas e capacidade antioxidante nos brócolis orgânicos e convencionais. Os brócolis orgânicos apresentaram maiores teores de antioxidantes. Verificou-se que as variedades T e T 506 Chioggia e Chioggia 706 foram as que apresentaram maior atividade enzimática. A variedade Castellano Lucrezia apresentou maior atividade da polifenoloxidade. Para o teor de proteínas solúveis totais a 5070 F1 Chioggia foi a variedade que apresentou maior teor de proteínas solúveis totais
This study investigated the effect of various sanitization treatments on the physicochemical, plant and content of antioxidants (polyamines, phenols, flavonoids, ascorbic acid, chlorophyll) addition of the antioxidant potential of organic and conventional broccoli and quality of enzymatic thirty-one varieties of radicchio. The organic and conventional broccoli were purchased from local producers of Botucatu / SP. The vegetables were washed, sanitized with chlorinated water and ozonated for 5 and 10 minutes and stored in cold storage at 6 ± 1 ° C for seven days. Samples were collected every three days. The cultivation methods together with the washing treatments significantly affected TA and SS. The treatments with ozonated water for 5 and 10 'were effective at reducing the microbial load in broccoli treated. There was no influence of cultivation methods for Cu, Zn and Mn. The use of ozone did not change the physicochemical quality of broccoli, regardless of the method of cultivation, which can be promising alternative for use as a sanitizer. There was influence of sanitation treatments during evaluation for antioxidants, polyamines and antioxidant capacity in organic and conventional broccoli. The organic broccoli showed higher levels of antioxidants. It was found that the peroxidase activity was higher in the varieties T and T 506 and Chioggia Chioggia 706. In the variety Castellano Lucrezia was obtained the highest activity of polyphenyloxidase. For the total soluble protein content, the 5070 F1 Chioggia variety showed the highest content of total soluble proteins

Resumo: O estresse oxidativo das bactérias probióticas representa uma grande preocupação para a indústria de alimentos probióticos, pois a presença de oxigênio pode levar tais bactérias à morte em poucos dias fazendo com que o produto deixe de apresentar suas propriedades funcionais alegadas. Assim, estudos são cada vez mais necessários para prolongar a vida de prateleira dos produtos probióticos. Nesse sentido foi avaliado o potencial do extrato da casca de jabuticaba, obtido por extração com fluido supercrítico, como fonte natural de antioxidantes, que além de promover a redução do estresse oxidativo das bactérias probióticas, também pode trazer benefícios à saúde. O extrato de jabuticaba foi bastante eficiente na manutenção da contagem das culturas de Lactobacillus acidophilus e Bifidobacterium lactis no Petit Suisse ao longo de 28 dias de estocagem refrigerada. O Petit Suisse acrescido do extrato também apresentou menor valor de pH final e maior produção de ácidos orgânicos o que pode estar relacionado com a maior atividade antioxidante do extrato em relação aos demais antioxidantes estudados, ácido ascórbico, cisteína e glicose oxidase, o que favorece o metabolismo das culturas probióticas. O Petit Suisse com o extrato também apresentou boa aceitação por possíveis consumidores do produto, obtendo elevada intenção de compra
Abstract: The oxidative stress of the probiotic bacteria represents a great preoccupation for the probiotics food products, since the presence of oxygen may cause bacteria death in a few days making that the product don't present the functional properties cited. Thus, studies are each more necessaries to improve the shelf life of the probiotic products. Then it was evaluated the potential of the jaboticaba peel supercritical fluid extract, a natural source of antioxidants, that promotes the reduction of the oxidative stress of the probiotic bacteria, and also achieving health benefits. The jaboticaba peel was very efficient on the maintenance of the counts of the cultures of Lactobacillus acidophilus and Bifidobacterium lactis on Petit Suisse during 28 days of storage. The Petit Suisse added of the extract also presents lowest value of final pH and greatest production of organic acids which may be related with the greatest antioxidant activity of the extract in comparison with the others antioxidants studied, ascorbic acid, cistein and glucose oxidase, what favors the probiotic cultures metabolism. The Petit Suisse containing extract also present good acceptability between the potential consumers of the product, obtaining great purchase intent

Resumo: Cresce o número de consumidores que optam por hábitos mais saudáveis e melhor qualidade de vida, incluindo em sua dieta alimentos que exerçam funções essenciais para a promoção da saúde e prevenção de doenças. O Brasil possui uma das maiores biodiversidades do planeta, com fontes inexploráveis de alimentos, como frutas exóticas ricas em compostos bioativos com funções diversas e complementares. Neste sentido, o presente trabalho buscou investigar o potencial prebiótico e a presença de compostos antioxidante em frutas exóticas da Mata Atlântica como o Cambuí Roxo (Eugenia candolleana), Araçá Una (Psidium eugeniaefolia) e Araçá Morango (Psidium guianensis). A extração etanólica apresentou melhor performance em relação à analise dos oligossacarídeos nas frutas. A concentração de oligossacarídeos totais nestes extratos foi em media de 44; 14 e 46 mg/100g de fruta para Cambuí roxo, Araçá una e Araçá morango, respectivamente, sendo verificada através de cromatografia de íons acoplada a detector amperiométrico pulsado. Os oligossacarídeos que se destacaram na composição das frutas foram os fruto-oligossacarídeos GF2, a estaquiose e a maltotriose. Foi observado ao microscópio a formação de aglomerados e formações celulares do tipo rede nos cultivos indicando a formação de exopolissacarídeo por Lactobacilos acidophilus em meio enriquecido com os extratos da frutas analisadas. A produção máxima de EPS foi verificada em 24h de cultivo sendo de 45,39; 41,90 e 17,16 mg/100g de meio para o Cambuí roxo, Araçá una e Araçá morango, respectivamente. A concentração de fenólicos para as extrações alcoólicas foi em média de 685; 395 e 361 mg de ácido gálico/100g de fruta, respectivamente para o Cambuí roxo, Araçá una e Araçá morango. Os extratos das frutas exóticas da Mata Atlântica foram avaliados, ainda quanto à capacidade antioxidante in vitro através do método Oxygen Radical Absorbance Capacity (ORAC) apresentando teores médios de 3.212; 2.065 e 2.641µM Trolox equivalente/100g de fruta para Cambuí roxo, Araçá una e Araçá morango, nesta ordem. A importância da pesquisa de compostos bioquímicos em frutas exóticas tem como objetivo, além de populariza-las, promover uma maior variedade de alimentos, novos sabores, aliado a componentes funcionais que exercem benefícios, tais como atividade prebiótica no intestino humano e atividade antioxidante, principal causa de envelhecimento celular. Brazil has one of the greatest biodiversity on the planet with unusable food sources, such as exotic fruits rich in bioactive compounds with different and complementary functions. In this sense, the present study investigated the prebiotic potential and the presence of antioxidant compounds in exotic fruits of the Atlantic Forest as Cambuí roxo (Eugenia candolleana), Araçá una (Psidium eugeniaefolia) and Araçá morango (Psidium guianensis). The ethanol extract showed better performance in relation to the analysis of oligosaccharides in fruits. The concentration of total oligosaccharides in these extracts was an average of 44; 14:46 mg/100g of fruit to Cambuí roxo, Araçá una and Araçá roxo, respectively, and verified by ion chromatography coupled with pulsed amperiométrico detector. Oligosaccharides that stood out in the composition of fruit were the fructo-oligosaccharides GF2, stachyose and maltotriose. It was observed under the microscope the formation of agglomerates and mobile network type formations on crops indicating the formation of exopolysaccharide by Lactobacillus acidophilus amid enriched with extracts of fruit analyzed. The maximum production of EPS was observed in 24 hours of cultivation being 45.39; 41.90 and 17.16 mg/100g medium for purple Cambuí roxo, Araçá una and Araçá morango respectively. The concentration of phenolic for alcoholic extractions averaged 685; 395 and 361 mg of AG/100 g of fruit, respectively for purple Cambuí roxo, Araçá una and Araçá morango. The extracts of exotic fruits of the Atlantic Forest were evaluated, even as the antioxidant capacity in vitro through the Oxygen Radical Absorbance Capacity method (ORAC) having average levels of 3,212; 2065 and 2.641?M Trolox equivalent/100 g of fruit to purple Cambuí roxo, Araçá una and Araçá morango, in that order. The importance of research biochemicals in exotic fruits, aims, and popularized them, promote greater variety of foods, new flavors, combined with functional components that exercise benefits such as prebiotic activity in the human gut and antioxidant activity, leading cause of cellular aging. The results of this research show the richness of components that can be exploited these fruits

Full textTwenty eight Brangus males were used to determine the oxidative stability of muscle and liver in order to compare organic and inorganic selenium source and rosemary extract based on the analysis of selenium concentration in the liver and in the serum, on the analysis of glutathione peroxidase, superoxide dismutase and catalase activities in the liver and also based on TBARS analysis (substances related to 2-thiobarbituric acid) in the muscle. Treatments were: 1) Control . without selenium or rosemary extract; 2) Se IN . 2 mg selenium/kg dry matter, as sodium selenite; 3) Se . 2 mg selenium/kg dry matter, as selenium-methionine and 4)Rosemary . 1400 mg rosemary extract/kg dry matter. Weight gain was greater when inorganic selenium was used (P<0.05) in comparison with control, however there was no difference regarding the other treatments. Feed efficiency was higher than control for all treatments (P<0.05). Dry matter intake was not affected by treatments (P<0.05). Selenium concentration in the liver was greater for inorganic selenium (P<0.05) than for control, in spite of that rosemary and organic selenium did not differ between themselves (P<0.05). Selenium concentration in the serum was higher for organic and inorganic selenium treatments than for control during all the period (P<0.05). Fatty acid profile in the meat was not affected by the treatments (P<0.05). Glutathione peroxidase activity was greater for inorganic selenium treatment when compared to control (P<0.05). Superoxide dismutase activity was lower for the rosemary treatment (P<0.05). Neither catalase activity nor TBARS analysis differ among treatments (P<0.05).

Full textIn the present study, analytical methods (TLC and HPLC/UV) were used to evaluate the antioxidant activity of the flavonoids of sugar cane (Saccharum officinarum L.) regarding to its possible use as a functional food. HPLC/UV and HPLC microfractionation techniques allowed fractionation and isolation of the flavonoids, which were evaluated by TLC using specific reagents to detect the antioxidant compounds. The results allowed to identify the flavonoids of sugar cane with antioxidant activity, based on comparison of UV-DAD spectra and retention time: orientin-O-rhamnoside in the leaves and schaftoside, isoschaftoside, diosmetin-8-C-glycoside, orientin and 4\'-5\'-dimethyl-luteolin-8-C-glycoside in the juice. Theses results justify more detailed nutricional and pharmacologic studies to classify (or not) sugar cane as a functional food.

Full textO mercado mundial de polissacarídeos tem atraído grandes companhias industriais interessadas em conquistar novos e rentáveis campos de atuação. Polissacarídeos com propriedades tecnológicas e biológicas podem ser obtidos a partir de plantas, algas e de microrganismos. Dentre os polissacarídeos com propriedades biológicas, as glucanas tem se destacado por apresentarem atividade imunoestimulante e potencialidades no tratamento de doenças como câncer, hipercolesterolemia, diabetes, esclerose múltipla e doenças cardiovasculares. Recentes estudos demonstram a produção extracelular de β-glucanas por fungos filamentosos em cultivos submersos. A modificação na estrutura química das glucanas por carboximetilação é considerada uma importante rota para melhorar suas propriedades, podendo contribuir para o aumento da solubilidade da molécula, bem como atividades biológicas, especialmente aquelas associadas a mecanismos de ação antioxidante e antiproliferativa. Nesse sentido, o presente trabalho teve como objetivo a produção de β-1,6-D-glucana (lasiodiplodana) pelo fungo Lasiodiplodia theobromae MMPI em cultivo submerso, bem como a carboximetilação da molécula, caracterização e avaliação da citotoxicidade e atividade antioxidante. A carboximetilação da molécula foi confirmada através da verificação de sinais químicos específicos identificados por espectroscopia de FT-IR e RMN 13C e a molécula carboximetilada apresentou grau de substituição (DS) de 1,27. A análise térmica (TG/DTA) indicou que a amostra bruta e carboximetilada apresentaram quatro estágios de perda de massa. O primeiro estágio ocorreu em 125ºC (perda de água) e houve dois eventos consecutivos de perda de massa (200ºC-400ºC) atribuídos à degradação da molécula. O quarto estágio ocorreu entre 425ºC e 620ºC (decomposição final) com pico exotérmico em 510ºC. Análise por MEV indicou que a lasiodiplodana bruta apresenta estruturas granulares que se rompem após carboximetilação. Análise de DRX demonstrou que o polímero bruto e carboximetilado apresentam estrutura não cristalina. A carboximetilação contribuiu para melhorar a hidrossolubulidade da molécula (aumento de 60%) e para melhorar a atividade antioxidante avaliada pela capacidade de captura dos radicais ABTS, DPPH e poder redutor do íon férrico (FRAP). Não foi verificado efeito citotóxico da lasiodiplodana bruta e modificada sobre hemácias. Os resultados obtidos sugerem que a carboximetilação da lasiodiplodana pode contribuir para melhoria das propriedades biológicas e para o potencial de uso biotecnológico da molécula.
The world market of polysaccharides has attracted large industrial companies interested in gaining new and profitable fields. Polysaccharides with technological and biological properties can be obtained from plants, algae and microorganisms. Among the polysaccharides with biological properties, glucans have been highlighted by demonstrate immunostimulatory activity and potential for treating diseases such as cancer, hypercholesterolemia, diabetes, multiple sclerosis and cardiovascular diseases. Recent studies demonstrate the production of exocellular β-glucans by filamentous fungi in submerged cultivations. Modifications in the chemical structure of glucans by carboxymethylation is considered an important route to improve its properties, may contribute to the increased solubility of the molecule as well as biological activities, especially those associated with antioxidant and antiproliferative mechanisms of action. Therefore, the present work aimed the production of β-1,6-D glucan (lasiodiplodana) by the Lasiodiplodia theobromae MMPI fungus in submerged cultivation and carboxymethylation of the molecule, characterization and evaluation of cytotoxicity and antioxidant activity. The carboxymethylation of the molecule was confirmed by checking specific chemical signals identified by FT-IR and NMR and 13C spectroscopy. Carboxymethylated molecule presented degree of substitution (DS) of 1.27. Thermal analysis (TG / DTA) indicated that native and carboxymethylated samples had four stages of mass loss. The first stage was at 125 ºC (loss of water) and there were two consecutive events of weight loss (200 ºC - 400 ºC) attributed to the degradation of the molecule. The fourth stage occurred between 425 ºC and 620 ºC (final decomposition) with exothermic peak at 510 ºC. SEM analysis indicated that the raw lasiodiplodan presents granular structures which are broken after carboxymetylation. XRD analysis showed that native and carboxymethylated biopolymers have no crystalline structure. Carboxymethylation aided to improve water solubility the molecule (60% increase) and to improve antioxidant activity assessed by ability to capture the ABTS, DPPH radical scavenging and the ferric ion reducing power (FRAP). There was no cytotoxic effect of raw lasiodiplodana and modified on the erythrocytes. The results suggest that the carboxymethylation of lasiodiplodan can contribute to improved biological properties and the potential biotechnological use of the molecule.

Full textO Brasil, devido à sua grande produção agrícola, é um dos países que mais produz resíduos agroindustriais no mundo e observa-se um interesse crescente na busca de alternativas para a utilização da matéria orgânica gerada, que atualmente tem sido fonte de poluição do meio ambiente. A indústria vinícola tem gerado um grande volume de resíduos que podem constituir um material de baixo custo e boa rentabilidade em aplicações tecnológicas, além de conter substâncias bioativas de alto valor comercial. Neste estudo; foi avaliado o bagaço de três variedades de uva. Ácidos graxos e compostos fenólicos foram identificados por cromatografia líquida e gasosa, respectivamente. A capacidade antioxidante dos extratos fenólicos foi avaliada pelos métodos DPPH, ABTS+ e íon ferroso. O bagaço da uva Cabernet Sauvignon apresentou os valores mais elevados de compostos fenólicos totais (5.101,82 ± 119,03 mg -1 -1 EAG 100 g), flavonoides (2.983,91 ± 51,76 mg ECT 100 g ) e antocianinas monomérica totais -1 (2.092,93 ± 71,57 mg mal-3-gli.100 g ). Além disso, o bagaço de Cabernet Sauvignon apresentou os menores valores de EC50 nos testes de DPPH e ABTS, apresentando alta atividade antioxidante.
Brazil, due to its large agricultural production, is one of the largest producers of agro-industrial waste worldwide and there is a growing interest in finding alternatives to the use of organic material generated, which currently has been a source of environmental pollution. Wine industry has generated a large amount of waste that may be a low cost and good profitability in technological applications, besides containing bioactive compounds of high commercial value. Grape pomace of three varieties was evaluated in current study. Fatty acids and phenolic compounds were identified respectively by gas and liquid chromatography. Antioxidant capacity of the phenolic extracts was determined by three different methods, namely DPPH, ABTS+ and ferrous ion assays. Cabernet Sauvignon pomace revealed the higher rates of total phenolic -1 -1 compounds (5,101.82 ± 119.03 mg GAE 100 g ), flavonoids (2,983.91 ± 51.76 mg CTE 100g ) -1 and monomeric anthocyanin (2,092.93 ± 71.57 mg mal-3-glu.100 g), coupled to lowest EC50 rates in DPPH and ABTS assays, showing a high antioxidant activity.

Resumo: Este trabalho objetivou determinar o efeito de diversos tratamentos de sanitização na qualidade físico-química, fitossanitária e o teor de antioxidantes (poliaminas, fenóis, flavonóides, ácido ascórbico, clorofila) além do potencial antioxidante de brócolis orgânicos e convencionais e qualidade enzimática de trinta e uma variedades de radicchio. Os brócolis orgânicos e convencionais foram adquiridos de produtores locais do município de Botucatu/SP. Os vegetais foram lavados, higienizados com água clorada e ozonizada por 5 e 10 minutos e armazenados em câmara fria a 6±1°C por sete dias. As coletas foram realizadas a cada três dias. O modo de cultivo aliado aos tratamentos de lavagem influenciou significativamente AT e os SS. Os tratamentos com água ozonizada por 5 e 10' foram eficazes na diminuição da carga microbiana nos brócolis tratados. Não houve influencia do modo de cultivo nos teores de Cu, Zn e Mn. O uso do ozônio não alterou as qualidades físico-químicas dos brócolis, independente do modo de cultivo, o que pode ser promissor, como alternativa para uso como sanitizante. Houve influencia dos tratamentos de sanitização no período de avaliação para os antioxidantes, poliaminas e capacidade antioxidante nos brócolis orgânicos e convencionais. Os brócolis orgânicos apresentaram maiores teores de antioxidantes. Verificou-se que as variedades T e T 506 Chioggia e Chioggia "706" foram as que apresentaram maior atividade enzimática. A variedade Castellano Lucrezia apresentou maior atividade da polifenoloxidade. Para o teor de proteínas solúveis totais a 5070 F1 Chioggia foi a variedade que apresentou maior teor de proteínas solúveis totais
Abstract: This study investigated the effect of various sanitization treatments on the physicochemical, plant and content of antioxidants (polyamines, phenols, flavonoids, ascorbic acid, chlorophyll) addition of the antioxidant potential of organic and conventional broccoli and quality of enzymatic thirty-one varieties of radicchio. The organic and conventional broccoli were purchased from local producers of Botucatu / SP. The vegetables were washed, sanitized with chlorinated water and ozonated for 5 and 10 minutes and stored in cold storage at 6 ± 1 ° C for seven days. Samples were collected every three days. The cultivation methods together with the washing treatments significantly affected TA and SS. The treatments with ozonated water for 5 and 10 'were effective at reducing the microbial load in broccoli treated. There was no influence of cultivation methods for Cu, Zn and Mn. The use of ozone did not change the physicochemical quality of broccoli, regardless of the method of cultivation, which can be promising alternative for use as a sanitizer. There was influence of sanitation treatments during evaluation for antioxidants, polyamines and antioxidant capacity in organic and conventional broccoli. The organic broccoli showed higher levels of antioxidants. It was found that the peroxidase activity was higher in the varieties T and T 506 and Chioggia Chioggia "706". In the variety Castellano Lucrezia was obtained the highest activity of polyphenyloxidase. For the total soluble protein content, the 5070 F1 Chioggia variety showed the highest content of total soluble proteins

Resumo: A família Myrtaceae apresenta ampla distribuição, ocorrendo, preferencialmente, nas zonas tropicais e subtropicais, e é considerada uma das mais importantes famílias da flora brasileira em função da larga ocorrência de espécies comestíveis e/ou usadas na medicina tradicional. Eugenia jambolana Lam. é conhecida no Brasil como jambolão, e é de grande interesse pelas aplicações medicinais, especialmente de suas folhas e frutos, no tratamento da diabetes. O trabalho tem como objetivo a investigação fitoquímica das folhas e frutos de E. jambolana monitorado por testes para detecção de atividade potencialmente antioxidante, através dos métodos de descoramento do b-caroteno e seqüestro do radical DPPH. Folhas e frutos da planta em estudo foram coletados na Faculdade de Ciências e Letras - UNESP/CAr. Foi obtido o extrato etanólico das folhas que foi particionado com solventes de diferentes polaridades. Para a obtenção do extrato bruto dos frutos (casca e polpa) foi utilizado MeOH (1% HCl). O estudo fitoquímico das folhas através de técnicas cromatográficas usuais (cromatografia em coluna, CLAE e cromatografia em contra corrente de alta velocidade) resultou no isolamento de dois flavonóides que apresentaram atividade antioxidante em ensaio com solução de b-caroteno. Os flavonóides isolados foram identificados como 4'-Oramnopiranosídeo- 3',5,5',7-tetraidroxi-flavona (1) e miricetrina (2). Análises em CLAE-DAD das frações provenientes do extrato bruto das frutas evidenciaram a presença de três antocianinas majoritárias e uma minoritária, que foram detectadas por análises de CLAE-DAD e CLAE-MS, e em comparação com dados da literatura, proporcionou a identificação das mesmas como cianidina-3-xilosídeo, cianidina-3- glucosídeo, cianidina-3-rutinosídeo e cianidina-3-(6''-acetoil)-glicosídeo. Abstract: The Myrtaceae family is widely distributed and occurs, mostly, in tropical and subtropical areas. It is one of the most important families of the Brazilian flora owing to the wide occurrence of edible species and/or species used in the traditional medicine. Eugenia jambolana Lam. is known in Brazil as jambolão and is of great interest for its folk medicinal applications, especially of their leaves and fruits, in the treatment of diabetes. Leaves and fruits of this species have been collected at UNESP (University Campus). The ethanol extract of the leaves was partioned with organic solvents and the crude extract of the fruits (skin and pulp) was obtained with acidified methanol (1% HCl). The phytochemical study of the leaves extract using chromatographic techniques (column chromatography, TLC, HPLC and HSCCC) resulted in the isolation of two flavonoids which presented strong antioxidant activity in the TLC assay using b-carotene solution as revealing agent. The isolated flavonoids have been identified as 4'-Oramnopyranoside- 3',5',5,7-tetrahydroxy-flavone (1) and myricetrin (2). Their strong antioxidant activity could be related with the presence of hydroxy groups in ring B which provides increased stability to the radical formed after donation of a hydrogen radical, and also the presence of the _,_-unsaturated carbonyl moiety on C ring.. HPLC-DAD analyses of the fractions obtained from the crude extract of the fruits evidenced the presence of three major anthocyanins and a minor one, which been identified by analyses of HPLC-DAD and HPLC-MS data, and comparison with literature. They were identified as cyanidin-3-xyloside, cyanidin-3-glucoside, cyanidin-3-rutinoside and cyanidin-3- (6''-acetoyl)-glucoside. O trabalho tem como objetivo determinar propriedades funcionais de tecidos vegetal e microbiano visando fornecer subsídios para uma dieta saudável. O tecido vegetal escolhido para este estudo foi a cebola cultivada na região sul do Rio Grande do Sul e a microalga chlorella, que será avaliada como padrão de funcionalidade. As amostras de cebolas foram fornecidas e separadas em classes comerciais, conforme o Regulamento Técnico de Qualidade de Cebola, aprovado em 1995 pelo Ministério da Agricultura, do Abastecimento e da Reforma Agrária, pelos técnicos da EMATER - Rio Grande. A composição centesimal, pH e acidez (AOAC, 2000) e os compostos fenólicos da cebola que foram extraídos em três sistemas solventes sob condições padronizadas (etanólico, aquoso, acetato de etila) e quantificados colorimétricamente com reagente de Folin-Ciocalteau. Na microalga chlorella os fenóis foram extraídos em sistema solvente metanólico. O teor de lipídios foi determinado pelo método de Bligh & Dyer para ambas as matrizes, os açúcares redutores foram determinados conforme método do 3,5-DNS (MILLER) para as amostras de cebola e na microalga chlorella foi estimado por diferença. Nas cebolas foi estimada a atividade específica de catalase, peroxidase e polifenoloxidase. A estimativa do potencial funcional dos compostos fenólicos foi indicada pela capacidade de seqüestrar o radical livre (DPPH), inibição da ação de enzimas oxirredutases e inibição de produção de peróxidos e compostos oxidados em sistemas lipídicos. Nos extratos de cebola a média dos teores de fenóis em solvente metanol, água e acetato de etila foram respectivamente 2,3; 3 e 0,08 mg/g de cebola. Os valores para umidade variaram entre 88,3 e 88,6%, lipídios entre 0,16 e 0,19%, proteínas entre 0,86 e 1,04%, cinzas entre 0,73 e 0,76%, fibras entre 0,6 e 0,7%, açúcares redutores entre 3,73 e 5,2%, o pH entre 5,08 e 5,23, a acidez entre 0,29 e 0,38%. A microalga chlorella apresentou 56,9% de proteína, 4,1% de umidade, 6,3% de cinzas, 22,3% de açúcares redutores, 2,8% de fibra-bruta, 7,6% de lipídios, 0,2% de acidez e pH 5,7. A atividade específica da catalase nas diferentes classes de cebola variou entre 0,014 e 0,025 mg H2O2/min/mg proteínas, sendo os maiores valores encontrados nas classes 2 e 3. A polifenoloxidase não foi detectada nas diferentes classes enquanto que a atividade específica da peroxidase variou entre 0,003 e 0,031 abs/min/mg proteínas. Todos os extratos apresentaram atividade seqüestradora do radical livre DPPH, porém o extrato de cebola extraído com acetato de etila foi o que apresentou maior inibição específica (53%) em relação aos demais. As velocidades máximas da reação de escurecimento enzimático diminuíram em presença dos extratos de cebola e chlorella, o que caracteriza o efeito de inibição da oxidação do guaiacol catalisado pela peroxidase. O extrato mais promissor para evitar a oxidação ocasionada pela catálise metálica em sistema lipídico foi o extrato de cebola extraído com acetato de etila com uma inibição percentual de 53% acompanhado pela menor formação de peróxido (até três vezes menor) e de malonaldeído (1,5 vezes menor) ao longo do tempo do experimento.
The work has as objective to determine functional properties in vegetal and microbian tissues aiming at to supply subsidies a healthful diet. The tissue vegetal chosen for this study was the onion cultivated in the southern region of the Rio Grande do Sul and the microalgae chlorella, that it will be evaluated as functionality standard. The samples of onions had been supplied and separate in commercial classrooms, as the Regulation Technician of Quality of Onion, approved in 1995 for the Ministry of Agriculture, the Supplying and the Agrarian Reformation, for the technician of the EMATER – Rio Grande. The centesimal composition, pH and acidity (AOAC, 2000) and the phenolic composites of the onion that had been extracted in three solvent systems under conditions standardized (ethanolic, watery, acetate of etila) and quantified colorimetrically with reagent of Folin-Ciocalteau. In the microalgae chlorella the phenols had been extracted in methanolic solvent system. The content of lipids was determined by the method of Bligh & Dyer for both the matrices, the reducing sugars had been determined in agreement method of 3,5-DNS (MILLER) for the samples of onion and in the microalgae chlorella was esteem by difference. In the onions the specific activity of catalase was esteem, peroxidase and poliphenoloxidase. The estimate of the functional potential of phenolic composites was indicated by the capacity to scavenging the radical (DPPH), inhibition of the enzyme action oxirredutases and inhibition of production of peroxides and composites oxidated in lipídicos systems free. In onion extracts the average of phenol texts in solvent methanol, water and acetate of etila had been respectively 2,3; 3 and 0,08 mg/g of onion. The values for humidity had varied between 88,3 and 88,6%, reducing lipids between 0,16 and 0,19%, proteins between 0,86 and 1,04%, leached ashes between 0,73 and 0,76%, staple fibers between 0,6 and 0,7%, sugars between 3,73 and 5,2%, pH between 5,08 and 5,23, the acidity between 0,29 and 0,38%. The microalgae chlorella presented 56.9% of protein, 4.1% of humidity, 6.3% of leached ashes, 22.3% of reducing sugars, 2.8% of fiber-rude, 7.6% of lipids, 0.2% of acidity and pH 5,7. The specific activity of catalase in the different classrooms of onion varied between 0,014 and 0,025 mg H2O2/min/mg proteins, being the biggest values found in classrooms 2 and 3. Poliphenoloxidase w as not detected in the different classrooms while that the specific activity of peroxidase varied between 0,003 and 0,031 abs/min/mg proteins. All the extracts had presented scavenging activity of free radical DPPH, however the extract of onion extracted with acetate of etila was what it presented greater specific inhibition (53%) in relation to excessively. The maximum speeds of the reaction of enzymatic blackout had diminished in presence of extracts of onion and chlorella, what it characterizes the effect of inhibition of the oxidation of guaiacol catalyzed by peroxidase. The extract most promising to prevent the oxidation caused for the metallic catalysis in lipídico system was the extract of onion extracted with ethyl acetate with a percentile inhibition of 53% followed by the lesser peroxide formation (up to three times lesser) and of malonaldeído (1,5 lesser times) to the long one of the time of the experiment.

Full textResumo: Estudos têm encontrado resultados promissores para o uso de antioxidantes na reversão da redução da adesão, ocasionada pela impregnação de radicais livres em dentina clareada. O objetivo do presente estudo foi avaliar o efeito de diferentes concentrações (15, 20 e 25%) de um gel de alfatocoferol na resistência à fratura, formação de camada híbrida e resistência de união de um adesivo em substrato clareado com peróxido de hidrogênio 38% (PH 38%). Sessenta incisivos bovinos foram distribuídos em 6 grupos (n=10): G1 - sem tratamento clareador (controle negativo); G2 - PH 38% (controle positivo); G3 - PH 38% + alfatocoferol gel 15%; G4 - PH 38% + alfatocoferol gel 20%; G5 - PH 38% + alfatocoferol gel 25%; G6 - PH 38% + ascorbato de sódio gel 10%. Os dentes foram preparados, submetidos a 3 aplicações de 15min de PH 38%. Os respectivos antioxidantes foram aplicados por 10 min e, em seguida, os dentes foram restaurados. O teste de resistência à fratura foi realizado 24h após o procedimento clareador em máquina de ensaio mecânico (EMIC). Abstract: The release and impregnation of free radicals in dentin damage the bond strength on bleaching teeth. The use of antioxidants has emerged as an interesting alternative to solve this problem. The aim of this study was evaluating the effect of different concentrations (15, 20 e 25%) of an alpha-tocopherol gel on fracture resistance, hybrid layer formation and bond strength of an adhesive system used in endodontic treated and bleached teeth (hydrogen peroxide 38% - HP 38%). Sixty bovine incisors were divided into six groups (n=10): G1 - negative control; G2 - 38% HP (positive control); G3 - 38% HP + 15% alpha-tocoferol; G4 - 38% HP + 20% alpha-tocoferol; G5 - 38% HP + 25% alpha-tocoferol; G6 - 38% HP + 10% sodium ascorbate. The specimens were prepared and submitted a bleaching using a gel with 38% HP (3x15 minutes). The antioxidant gels were applied for 10 minutes and the teeth was restored. The fracture resistance mechanical testing was realized 24 hours after. The analyze of the hybrid layer was done by scanning electron microscope (SEM). And the bond strength was analyzed by the microtensile bond strength test. Data were subjected to specific statistical analysis (Kruskal Wallis/Dunn - a=0,05). Resumo: Em pimentão a floração e a frutificação são contínuas, o que faz com que a planta tenha frutos e sementes em diferentes estádios de maturação. Dessa forma, identificar o estádio em que as sementes alcançam a maturidade fisiológica é fundamental para a determinação do momento ideal de colheita dos frutos. Diante do exposto, objetivou-se com este trabalho estudar o efeito do estádio de maturação e do repouso pós-colheita dos frutos na qualidade fisiológica, na resposta bioquímica e na composição química de sementes de pimentão. O trabalho foi dividido em dois capítulos, em ambos o delineamento experimental utilizado foi de blocos casualizados, com oito tratamentos, resultantes de um fatorial 4x2, com quatro repetições. O primeiro fator foi constituído de quatro estádios de maturação (35, 50, 65 e 80 dias após a antese (DAA)) e o segundo, sem e com o repouso dos frutos pós-colheita, por 7 dias. Foram avaliados dois genótipos que apresentam padrão de qualidade visual de sementes diferentes, ou seja, sem (1730) e com (190-2) o escurecimento do tegumento das sementes. Abstract: In sweet pepper the flowering and the fruiting are continuous, which causes that the plant has fruits and seeds in different stages of maturation. Thus, to identify the stage in which the seeds reach the physiological maturity is fundamental to determine the ideal moment to harvest the fruits. In view of this, the objective of this work was to study the effect of maturation stage and post-harvest rest of the fruits in physiological quality, biochemical response and chemical composition in pepper seeds. The work was divided in two chapters, in both the experimental design was randomized blocks, with eight treatments, resulting from a 4x2 factorial, with four replications. The first factor consisted of four maturation stages (35, 50, 65 and 80 days after anthesis (DAA)) and the second, without and with the post-harvest rest, for 7 days. Two genotypes with different seed quality standards were evaluated, that is, without (1730) and with (190- 2) darkening of seed integument. In the first chapter the characteristics evaluated in the seeds were the determination of the content of moisture, weight of a thousand seeds, germination, vigor (first germination test count and germination velocity index), superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) enzyme activity, lipid peroxidation (LIP) and hydrogen peroxide content (H2O2).In the second chapter were evaluated the dry mass of one thousand seeds, the macronutrient and protein contents in the seeds.The fruit harvest time indicated for seeds of genotype 1730 without and with the rest of the fruits is around 80 DAA and for the 190-2 genotype without the rest of the fruits it is at 72 DAA and with rest at 58 DAA. Full textThe ingestion copper chlorophyllin (Cu-Chl) in form of as food colorant or supplement has been associated with potential beneficiai effects and the prevention of chronic diseases. However, little is known about Cu-Chl absorption in vivo, its antioxidant activity and its impact on status nutritional. In view of these facts, three experiments were carried out on rats fed 1 and 3% Cu-Chl supplemented diets. In the first experiment, the absorption of the pigment into the bloodstream and distribution to the liver and kidneys was comproved. HPLC analysis of food-grade Cu-Chl evidenced two main components, copper-chlorin e6 and copper-chlorin e4. The Cu-chlorin e4 was effectively absorbed and found in serum, liver and kidneys, while Cu-chlorin e6 was not detected. This result indicates that Cu-chlorin e6 was either not absorbed and distributed to organs or was degraded during its passage through the gastrointestinal tract. Histological analyses of the intestine did not show any morphological alteration. In a second experiment the antioxidant potential of Cu-Chl in rats under oxidative stress induced by vitamin E deficiency and simultaneous Cu-Chl administration in diet was investigated. The presence of chlorophyllin protected brain tissue from increased TBARS leveis, however the same effect was not observed in liver. There was no significant differences in activities of antioxidant enzymes (CAT, SOD, GPX and GR) among control, vitamin E deficient and Cu-Chl-supplemented groups. Therefore, any antioxidant effect of Cu-Chl seems to be related to in situ protection against peroxidation but not to antioxidant enzyme modulation. Weight gains of the animais receiving Cu-Chl supplemented diets were lower than those of the control group. A third experiment was conducted to evaluate whether the reduced weight gain was caused by low food acceptance or by an antinutritional effect of Cu-Chl. This biological pair-fed test confirmed the negative effect of CuChl on the development of rats. The pigment ingestion also decreased the apparent protein digestibility indicating that N protein and possibly other nutrients had itsabsorption compromised. Literature evidenced formation of Cu-Chl complexes with different food components supporting the hypothesis that Cu-Chl might have interfered in the utilization of proteins and possibly other nutrients by the animal organism.

Nos últimos anos houve uma explosão no interesse dos consumidores pelos alimentos denominados alimentos nutraceuticoss. Estudos epidemiológicos têm sugerido que esses alimentos tem um importante papel na prevenção de doenças crônicas. Acredita-se que os antioxidantes, principalmente os compostos fenólicos, carotenóides, tocoferóis, naturalmente encontrados em frutas, vegetais e grãos, são os responsáveis por promover o efeito quimioprotetor contra o estresse oxidativo. As frutas são um dos importantes componentes de uma dieta saudável. Seu consumo em quantidades adequadas pode reduzir o risco de doenças cardiovasculares e alguns tipos de câncer. Esse alimento é importante porque é uma fonte rica de micronutrientes, fibras e outros componentes com propriedades funcionais. Ademais, é um alimento de baixa densidade energética, isto é, com poucas calorias em relação ao volume da alimentação consumida, o que favorece a manutenção do peso corporal saudável. Dentro desse cenário, o Brasil ostenta uma grande fonte de espécies tropicais e exóticas ainda pouco exploradas, as quais podem ter um potencial agroindustrial, representando uma excelente perspectiva de oportunidade econômica. Considerando os benefícios do consumo de frutas e o sua importância mercadológica, este trabalho procura estudar a atividade antioxidante de diferentes extratos de frutas, tropicais e exóticas, para avaliar o potencial antioxidante destes extratos, visando aplicações nos setores farmacêuticos, alimentícios e de cosméticos. In recent years have seen an explosion in consumer interest in foods called nutraceutical foods. Epidemiological studies have suggested that these foods have an important role in preventing chronic diseases. It is believed that antioxidants, especially phenolic compounds, carotenoids, tocopherols naturally found in fruits, vegetables and grains, are responsible for promoting chemistryprotect effect against oxidative stress. Fruits are one of the important components of a healthy diet. Its consumption in appropriate amounts can reduce the risk of cardiovascular disease and some cancers. This is important because food is a rich source of micronutrients, fiber and other components with functional properties. Moreover, food is a low energy density, that is, low in calories compared to the volume of feed consumed, which contributes to the maintenance of healthy body weight. Full textOs objetivos deste estudo foram avaliar os efeitos da utilização do ascorbato de sódio e alfatocoferol na dentina de dentes tratados endodonticamente, através da avaliação da resistência das coroas dentais à fratura, mensuração da formação intradentinária da camada hibrida e sua repercussão sobre a resistência de união de um sistema adesivo. No primeiro estudo, foram utilizados cinquenta incisivos bovinos endodonticamente tratados, distribuídos em cinco grupos (n = 10). Sendo, G1 controle, G2: somente restaurados com resina composta, G3: clareado com peróxido de hidrogênio a 38% e restaurado imediatamente, G4: clareado e tratado com ascorbato de sódio a 10%, G5: clareado e tratado alfatocoferol. Os espécimes foram submetidos ao teste mecânico de resistência à fratura, sendo que G1 apresentou maior resistência à fratura que os demais grupos. Não foi observada diferença estatística entre os demais grupos (p > 0,05). No segundo estudo, foram utilizados quarenta incisivos bovinos tratados endodonticamente, divididos em quatro grupos (n = 10): G1 restaurado (controle), G2 clareado e restaurado imediatamente, G3 clareado, tratado com ascorbato de sódio a 10% e restaurado imedimediata, G4 clareado, tratado com alfatocoferol 10% e restaurado imediata. O protocolo de clareamento e a utilização dos agentes antioxidantes foram iguais ao do experimento 1. Somente a porção coronária seccionada, no sentido vestíbulo-lingual de cada espécime foi utilizada para análise em microscópio laser confocal com aumento de 1024X. This study evaluated the effects of sodium ascorbate and alphatocopherol on fracture resistance, mensuration of the hybrid layer formation and bond strength of an adhesive system used in endodontically treated teeth. The first study used fifty endodontically treated bovine incisors, divided into five groups (n = 10): G1: control; G2: restoration with composite resin; G3: bleaching with 38% hydrogen peroxide and immediate restoration; G4: bleaching, application of 10% sodium ascorbate and restoration; G5: bleaching, alphatocopherol application and restoration. The specimens were subjected to fracture resistance mechanical testing. The obtained data were analyzed by ANOVA and Tukey test,at significance level of 5%. G1 showed higher fracture resistance than the other groups. No statistical difference were found among other groups (p> 0.05).The second study used forty endodontically treated bovine incisors, divided into four groups (n = 10): G1: restoration; G2: bleaching and immediate restoration; G3: bleaching, use of 10% sodium ascorbate and immediate restoration; G4: bleaching, use of 10% alphatocorol and immediate restoration. The bleaching protocol and antioxidant application were similar to experiment 1. Full textO emprego de técnicas de criopreservação de gametas em felinos domésticos são consideradas importantes no aprimoramento de procedimentos na reprodução assistida. A vitrificação se baseia no uso de altas concentrações de crioprotetores com o intuito de inibir a formação de cristais de gelo. É possível que a criopreservação induza a produção de espécies reativas de oxigênio (EROs) ou altere o potencial antioxidante enzimático do oócito, sendo assim, aventa-se que a adição de antioxidantes aos meios de vitrificação poderia reduzir o estresse oxidativo causado pelos crioprotetores. No experimento I comparam-se quatro protocolos de vitrificação utilizando-se como meio base (MB) o TCM199 (Meio de cultura de tecidos 199) com glicose mais antibiótico, acrescido de crioprotetores: G1 (3M etilenoglicol–EG + 2M de dimetilsulfóxido-DMSO), G2 (3M EG + 3M 1,2 propanediol-PrOH), G3 (1,5M EG + 1M DMSO) e G4 (1,5M EG + 1,5M PrOH). Após descongelamento, este material foi avaliado quanto à alteração morfológica e viabilidade (coloração cFDA/trypan blue). Neste, não houve diferença estatística (p=0,2857) entre os oócitos descongelados dos quatro grupos estudados em relação à morfologia, mas em relação a viabilidade oocitária os grupos G1 e G2 obtiveram maior porcentagem de oócitos viáveis, porém não diferiram entre si (35,71% e 36,84%, respectivamente; p=0,018). No experimento II três concentrações de vitamina E (0,4, 0,6 e 0,8mM) foram acrescidas ao protocolo de MB com 3M EG + 3M PrOH, formando os grupos G 0,4mM, G 0,6mM, G 0,8mM e o G 0 (sem adição de vitamina E). The use of techniques of cryopreservation of gametes in domestic cats are considered important in enhancing procedures in assisted reproduction. Vitrification is based on the use of high concentrations of cryoprotectants in order to inhibit the formation of ice crystals. It is possible that cryopreservation induces the production of reactive oxygen species (ROS) or change the enzymatic antioxidant potential of the oocyte, so one might speculate that the addition of antioxidants to the means of vitrification could reduce the oxidative stress caused by cryoprotectants. After thawing, the material was assessed for viability and morphological changes (staining cFDA / trypan blue). In this , there was no statistical difference (p = 0.2857) among the four groups thawed oocytes studied with respect to morphology, but in relation to oocyte viability G1 and G2 groups had a higher percentage of viable oocytes , but did not differ ( 35.71% and 36.84 %, respectively , p = 0.018). In the second experiment three concentrations of vitamin E (0.4 , 0.6 and 0.8 mM) were added to the protocol MB with 3M EG + 3M PrOH , L forming the groups G 0.4 mM, G 0.6 mM, G 0.8 mM and G 0 (no added vitamin E). The choice of the base protocol was in accordance with the results obtained in experiment I. We observed improvement in percentage (p = 0.0032) of oocytes with normal morphology in groups G 0.6 mM (88.6 %) and G 0.8 mM (62.5 %). Full textO efeito oxidante provocado por radicais livres provenientes do metabolismo do O 2 , ocorre naturalmente no organismo. O estresse oxidativo, aumento dessa condição, está correlacionado ao aparecimento de doenças, e pode ser intensificado devido à exposição a fatores externos, como a radiação solar, causando assim danos c elulares significativos, que podem levar ao envelhecimento acelerado das células. Estudos com extratos vegetais tê m demons trado que os mesmos contêm compostos de interesse farmacológico para tratar e/ou prevenir os danos celulares causados pelo estresse oxidativo, resultando em uma ação fotoprotetora e antiglicante. Entre as espécies vegetais , a família Annonaceae possui representantes que apresentam compostos bioativos com diferentes atividades farmacológicas, tornando - as importantes para a análise de sua capacidade antioxidante e a relação com o potencial fotoprotetor e antiglicante. Partindo desse pressuposto, este estudo tem por objetivo avaliar o potencial antioxidante, fotoprotetor e antiglicante com ensaios in vitro , além de quantificar fenóis e flavonoides totais presentes nas amostras, e também realizar a caracterização por cromatografia de camada delgada para flavonoides, terpenoides e alcaloides dos extratos. Os resultados obtidos mostraram que A. crassiflora e A. cacans apresentaram os melhores valores na maioria dos testes de avaliação da capacidade antioxidante e entre os tipos de extratos avaliados, bem como para potencial fotoprotetor. The oxidative effect caused by free radicals derived from O 2 metabolism occurs naturally in the human body. The oxidative stress, the increase of this condition, is correlated with the disease's appearance and can be intensified by the exposure of extern factors, like solar radiation, thus causing significant cellu lar damage and also may cause faster aging of cells. Researches with plant extracts have shown that they may have compounds with pharmacological interesting to healing and/or preventing cellular damag es caused by oxidative stress, those may results a photo protective and antiglicant activity. Among the plant species, the Annonaceae family has representatives that contain bioactive compounds with different pharmacological activities, making them important to the study of antioxidative capacity and its relatio n with photoprotective and anti glicant potential. Then , this project is designed to measure the oxidative, photoprotective and antiglicant potential in in vitro assays, and also to quantify the total phenols, flavonoids and condensed tannins in the samples . Full textNo momento há apenas dois fármacos disponíveis para o tratamento da doença de Chagas, nifurtimox e benzonidazol, ambos de ações limitadas. Assim, a compreensão dos mecanismos utilizados por T. cruzi para diminuir os efeitos citotóxicos de fármacos e a busca de potenciais moléculas para quimioterapia, bem como os alvos para elas, são de extrema importância. O aumento na produção de certas enzimas do metabolismo oxidativo seria responsável pela resistência das cepas do parasito ao benzonidazol (BZ). Tais enzimas são: triparedoxina peroxidase (mTcTXNPx) que catalisa a redução de peróxidos, superóxido dismutase (mTcSOD) que realiza a dismutação do ânion superóxido e old yellow enzyme (cTcOYE) que está envolvida na redução de substâncias tripanossomicidas, como naftoquinonas e compostos nitroheterocíclicos. Estudos demonstram que a atividade antimalarial da cloroquina foi aumentada com a adição de ferroceno. Neste trabalho, foram avaliadas as diferenças de suscetibilidade entre seis cepas de T. cruzi (Y, Bolívia, SI1, SIGR3, SI8 e QMII) em relação a três estruturas derivadas de diaminas do ferroceno e a expressão das proteínas antioxidativas. A substância AAC09 mostrou maior citotoxicidade, com CI50 de 2,21 μM para a cepa menos resistente (cepa Y) e 15,20 μM (cepa QMII) para a mais resistente, enquanto que para BZ foi 27,28 μM (cepa SI1) e 105,28 μM (cepa SIGR3), respectivamente. Após tratamento, a expressão das enzimas mTcTXNP (25,5 kDa) e cTcOYE (43 kDa) não aumentaram em cepas mais resistentes aos derivados de diaminas do ferroceno e BZ. At the moment there are only two drugs available for the treatment of Chagas disease, nifurtimox and benznidazole, both of use restricted. Understanding the mechanisms used by T. cruzi to reduce de cytotoxic effects of drugs and finding potential molecules for chemotherapy, as well as targets for them, are extremely important. Resistance to benznidazole (BZ) in T. cruzi strains may be caused by increased production of enzymes that act on oxidative metabolism. Such enzymes are: tryparedoxin peroxidase (mTcTXNPx), which catalyzes the reduction of peroxides, superoxide dismutase (mTcSOD), which performs the dismutation of superoxide anion and old yellow enzyme (cTcOYE), which is related to the reduction of trypanocidal substances, such as naphthoquinones and nitroheterocyclic compounds. Studies showed that the antimalarial activity of chloroquine was increased with the addition of ferroceno. Herein the susceptibilities in six T. cruzi strains (Y, Bolivia, SI1, SIGR3, SI8 and QMII) were analyzed in relation to three structures of ferrocene diamine derivatives and the expression of antioxidant proteins. The AAC09 substance showed higher cytotoxicity, with IC50 of 2.21 μM for the less resistant strain (Y strain) and 15.20 μM (QMII strain) for more resistant, whereas to BZ was 27.28 μM (strain SI1) and 105.28 μM (SIGR3), respectively. After treatment, the expression of the enzymes mTcTXNP (25.5 kDa) and cTcOYE (43 kDa) did not increase in more resistant strains to ferrocene diamine derivatives and BZ. Resumo: Protium heptaphyllum March (Burseraceae), conhecido como almécega ou breu branco, é uma planta medicinal abundante na região Amazônica. Seu exudato resinoso in natura é um conceituado remédio popular com ação anti-inflamatória, antiulcerogênica, analgésica e cicatrizante. O gênero Protium compreende cerca de 135 espécies, estima-se que a maioria das espécies da família Burseraceae pertençam a este gênero, tendo como principal característica sua capacidade em exsudar resinas aromáticas voláteis, usadas como curativo de feridas, tratamento de dor de dente, antisséptico, analgésico e distúrbios gastrointestinais. O objetivo deste trabalho, foi avaliar o potencial gastroprotetor do óleo essencial comercial do Protium heptaphyllum March (Burseraceae) (BB), bem como os possíveis mecanismos de ação envolvidos, através de modelos de úlcera gástrica em ratos Unib:WH. A análise de cromatografia gasosa-espectrometria de massa (CG-EM) do BB indicou a presença de três monoterpenos majoritários: ?-pineno (40%), p-mentha-1.4(8)-dieno (12%) e ?-phellandreno (10%). Foram realizados diferentes modelos experimentais de úlcera gástrica que, com base em suas respectivas especificações, foram incluídos dois grupos controles, sendo um positivo (Lansoprazol 30 mg/kg, Carbenoxolona 200 mg/kg ou Cimetidina 100 mg/kg) e um negativo (Veículo - Tween 80® 12%, 10 mL/Kg). Após cada experimento, foi realizada eutanásia por deslocamento cervical, os estômagos removidos, abertos na região da maior curvatura e fotografados para quantificação da área de lesão ulcerativa (ALU) por meio do programa AVSoft®. O BB, na dose de 100 mg/Kg (dose mais efetiva), apresentou atividade antiulcerogênica contra lesões gástricas induzidas por etanol absoluto (96%) e por Drogas ant-inflamatórias não esteroidais (DAINEs) (97%). Para avaliar os mecanismos de ação envolvidos a atividade antissecretória, muco, prostaglandina, atividade antioxidante e envolvimento dos Grupamentos Sulfidrila (G-SH), foram analisados. Além disto, o modelo de úlcera gástrica induzida por ácido acético e análise por Western blotting da expressão de COX-2 e EGF foram utilizadas para avaliar a capacidade cicatrizante do BB. Foi verificado que este óleo essencial não interfere na secreção ácida na mucosa gástrica; porém, sua gastroproteção depende de compostos com G-SH. A atividade gastroprotetora do BB não ocorre devido ao aumento na produção de muco induzido pelos níveis gástricos de PGE2. Como o BB não atuou nos mecanismos citoprotetor e antissecretório, objetivou-se verificar o envolvimento do mecanismo antioxidante na atividade gastroprotetora. Existem evidências de que espécies reativas de oxigênio participem na etiologia da úlcera gástrica e muitos óleos essenciais foram recentemente qualificados como antioxidantes naturais. A capacidade do BB de reduzir ou prevenir estresse oxidativo foi avaliada in vivo, a partir de modelos de úlcera induzida por etanol absoluto. O BB apresentou atividade antioxidante modulando enzimas antioxidantes como Superóxido Dismutase (SOD), Glutationa Peroxidase (GSH-Px) e Glutationa Redutase (GSH-Rd), desta forma, diminuindo a peroxidação lipídica (LPO) e atividade da Mieloperoxidase (MPO). BB demonstrou também potente capacidade de cicatrização, com 95% de redução da ALU, aumentando a expressão de COX-2 (55%) e EGF (100%) na mucosa gástrica. Em adição, nenhum sinal de toxicidade foi observado durante os 14 dias de tratamento, considerando os parâmetros analisados. Portanto o BB é composto essencialmente por terpenos, não apresenta atividade antissecretora nem citoprotetora, apresenta atividade antioxidante e cicatrizante
Abstract: Protium heptaphyllum March (Burseraceae), known as "Almécega" or "Breu Branco", a medicinal plant is abundant in the Amazon region, its resinous exudates in nature, is a renowned folk remedy with anti-inflammatory, antiulcer, analgesic and healing. Protium genus comprises about 135 species, it is estimated that most species of Burseraceae, belong to this genus, the main feature, its ability to exude volatile aromatic resins, used for various purposes such as dressing for wounds, pain tooth, antiseptic, analgesic, treatment of stomach, among others. Based on the popular indications of this plant for the treatment of gastrointestinal disturbances, the present work aimed on evaluating the gastroprotective potential of the commercial essential oil of Protium heptaphyllum March (Burseraceae) (BB), as well as its possible mechanisms of action involved, through models of gastric ulcer in rats Unib: WH. Gas chromatography-mass spectrometry (GC-MS) analysis of BB indicated the presence of three majority monoterpenes as major compounds: ?-pinene (40%), p-mentha-1.4(8)-diene (12%) and, ?-phellandreno (10%). We performed different experimental models of gastric ulcer, based on your specifications included two control groups, a positive (Lansoprazole 30 mg/kg, Cimetidine 100 mg/kg or Carbenoxolone 200 mg/kg) and a negative (Vehicle - Tween 80, 12%, 10 mL/Kg). After each experiment, rats were killed by cervical dislocation, their stomachs removed and opened in the region of greater curvature and photographed for quantification of lesion area ulcerative through the AVSoft program. BB at a dose of 100 mg / kg (effective dose) showed antiulcer activity against gastric lesions induced by absolute ethanol (96%) and nonsteroidal ant-inflammatory drugs (NSAIDs) (97%). To evaluate the mechanisms involved in the BB aniulcerogenic activity, the antisecretory action, mucus, prostaglandins, antioxidant activity and involvement of sulfhydryl group (G-SH) were analyzed. The acetic acid-induced gastric ulcer model and western blotting assay COX-2 and EGF were also used to evaluate the BB healing capacity. This essential oil does not interfere with acid secretion in the gastric mucosa, but their gastroproteção depends on G-SH compounds. The gastroprotective activity of the BB is not due to increased production of gastric mucus induced by PGE2 levels. As the BB did not act in cytoprotective mechanisms and antissecretório, aimed to verify the involvement of the antioxidant mechanism in gastroprotective activity. There is evidence that reactive oxygen species involved in the etiology of gastric ulcer and many essential oils have recently been classified as natural antioxidants. The role of the essential oil in reducing or preventing the oxidative stress was evaluated in vivo, from absolute ethanol-induced gastric ulcer model. The BB showed antioxidant activity by modulating antioxidant enzymes like Superoxide Dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GSH-Rd), thereby decreasing lipid peroxidation (LPO) and activity Myeloperoxidase (MPO). The BB has demonstraded potent healing capacity, with 95% reduction of the ulcerative lesion area due to increased expression of COX-2 (55%) and EGF (100%) in the gastric mucosa. In addition, no signs of toxicity were observed during the 14 days of treatment, considering the analyzed parameters. So BB is composed mainly of terpenes, shows no activity antissecretora or cytoprotective, exhibits antioxidant activity and healing
