Abstract:
Additives with bioactive properties can improve chickens’ gut health. This study investigated the physiological status of the gut, including its morphological structure and microbiome activities in chickens fed diets supplemented with phytogenic ingredients with hops (Anta®Phyt) or a mixture of a free butyrate acidifier and gluconic acid (PreAcid). In this study, 1155 broilers were distributed to three dietary treatments with 5 replicate pens per treatment, 77 birds each. Anta®Phyt was added at 400/300/200/200 mg/kg diet whereas PreAcid was added at 3/2/1/1 g/kg starter/grower I/grower II/finisher diet respectively. Dietary treatments did not compromise body weight in different growth periods. In the birds fed PreAcid-supplemented diet, higher gut concentration of butyric acid was observed, particularly in the early stage of growth, while the profile of the short-chain fatty acids was maintained among the treatments. Neither additive significantly affected cecal bacterial enzyme activities. Feeding the birds with Anta®Phyt and PreAcid had beneficial effects on gut morphostructure indices, including intestinal wall thickness, crypt depth and the villus height to crypt depth ratio, in 35- and 42-day old birds. In conclusion, the feeding Anta®Phyt- or PreAcid-supplemented diet exerted beneficial effects on the indices determining the physiological status of the gut and maintained good performance of birds of different ages.

Abstract:
The mitochondrial permeability transition pore (MPTP) has resisted molecular identification. The original model of the MPTP that proposed the adenine nucleotide translocator (ANT) as the inner membrane pore-forming component was challenged when mitochondria from Ant1/2 double null mouse liver still had MPTP activity. Because mice express three Ant genes, we reinvestigated whether the ANTs comprise the MPTP. Liver mitochondria from Ant1, Ant2, and Ant4 deficient mice were highly refractory to Ca2+-induced MPTP formation, and when also given cyclosporine A (CsA), the MPTP was completely inhibited. Moreover, liver mitochondria from mice with quadruple deletion of Ant1, Ant2, Ant4, and Ppif (cyclophilin D, target of CsA) lacked Ca2+-induced MPTP formation. Inner-membrane patch clamping in mitochondria from Ant1, Ant2, and Ant4 triple null mouse embryonic fibroblasts showed a loss of MPTP activity. Our findings suggest a model for the MPTP consisting of two distinct molecular components: The ANTs and an unknown species requiring CypD.

Abstract:
The toxicity of conventional nitroaromatic explosives like 2,4,6-trinitrotoluene (TNT) is caused by their enzymatic free radical formation with the subsequent oxidative stress, the formation of alkylating nitroso and/or hydroxylamino metabolites, and oxyhemoglobin oxidation into methemoglobin. In order to get an insight into the mechanisms of toxicity of the novel explosives NTO (5-nitro-1,2,4-triazol-3-one) and ANTA (5-nitro-1,2,4-triazol-3-amine), we examined their reactions with the single-electron transferring flavoenzymes NADPH: cytochrome P-450 reductase and ferredoxin:NADP+ reductase, two-electron transferring flavoenzymes mammalian NAD(P)H:quinone oxidoreductase (DT-diaphorase), and Enterobacter cloacae NAD(P)H:nitroreductase, and their reactions with oxyhemoglobin. The reactivity of NTO and ANTA in the above reactions was markedly lower than that of TNT. The toxicity of NTO and ANTA in bovine leukemia virus-transformed lamb kidney fibroblasts (line FLK) was partly prevented by desferrioxamine and the antioxidant N,N′-diphenyl-p-phenylene diamine, and potentiated by 1,3-bis-(2-chloroethyl)-1-nitrosourea. This points to the involvement of oxidative stress in their cytotoxicity, presumably to the redox cycling of free radicals. The FLK cell line cytotoxicity and the methemoglobin formation in isolated human erythrocytes of NTO and ANTA were also markedly lower than those of TNT, and similar to those of nitrobenzene. Taken together, our data demonstrate that the low toxicity of nitrotriazole explosives may be attributed to their low electron-accepting properties.

Abstract:

Abstract:

Abstract:
Background and Objective: Antimycin A (AntA) is a potent Electron Transport System (ETS) inhibitor exerting its effect through inhibiting the transfer of the electrons by binding to the quinone reduction site of the cytochrome bc1 complex (Complex III), which is known to be impaired in Huntington’s Disease (HD). The current studies were undertaken to investigate the effect of complex III inhibition in the SH-SY5Y cell line to delineate the molecular and cellular processes, which may play a role in the pathogenesis of HD. Methods: We treated SH-SY5Y neuroblastoma cells with AntA in order to establish an in vitro mitochondrial dysfunction model for HD. Differential proteome analysis was performed by the nLCMS/ MS system. Protein expression was assessed by western blot analysis. Results: Thirty five differentially expressed proteins as compared to the vehicle-treated controls were detected. Functional pathway analysis indicated that proteins involved in ubiquitin-proteasomal pathway were up-regulated in AntA-treated SH-SY5Y neuroblastoma cells and the ubiquitinated protein accumulation was confirmed by immunoblotting. We found that Prothymosin α (ProT α) was downregulated. Furthermore, we demonstrated that nuclear factor erythroid 2-related factor 2 (Nrf2) protein expression was co-regulated with ProT α expression, hence knockdown of ProT α in SH-SY5Y cells decreased Nrf2 protein level. Conclusion: Our findings suggest that complex III impairment might downregulate ubiquitinproteasome function and NRF2/Keap1 antioxidant response. In addition, it is likely that downregulation of Nrf2 is due to the decreased expression of ProT α in AntA-treated SH-SY5Y cells. Our results could advance the understanding of mechanisms involved in neurodegenerative diseases.

In this study, the first step in anthranilate catabolism was characterized. A mutant unable to grow on anthranilate, ACN26, was selected. The sequence of a wild-type DNA fragment that restored growth revealed theantABC genes, encoding 54-, 19-, and 39-kDa proteins, respectively. The deduced AntABC sequences were homologous to those of class IB multicomponent aromatic ring-dihydroxylating enzymes, including the dioxygenase that initiates benzoate catabolism. Expression of antABC in Escherichia coli, a bacterium that normally does not degrade anthranilate, enabled the conversion of anthranilate to catechol. Unlike benzoate dioxygenase (BenABC), anthranilate dioxygenase (AntABC) catalyzed catechol formation without requiring a dehydrogenase. In Acinetobacter mutants, benC substituted for antC during growth on anthranilate, suggesting relatively broad substrate specificity of the BenC reductase, which transfers electrons from NADH to the terminal oxygenase. In contrast, the benAB genes did not substitute for antAB. An antA point mutation in ACN26 prevented anthranilate degradation, and this mutation was independent of a mucK mutation in the same strain that prevented exogenous muconate degradation. Anthranilate induced expression of antA, although no associated transcriptional regulators were identified. Disruption of three open reading frames in the immediate vicinity ofantABC did not prevent the use of anthranilate as a sole carbon source. TheantABC genes were mapped on the ADP1 chromosome and were not linked to the two known supraoperonic gene clusters involved in aromatic compound degradation.

Abstract:

Abstract:
Objective of the study was to conduct phylogenetic analysis of Yersinia pestis strains of the main subspecies, belonging to antique biovar, phylogenetic branch 0.ANT, isolated in XX – early XXI centuries in the foci of Tien-Shan and Pamir-Alay to identify the regularities of spatial-temporal circulation of plague agent in the territory of the foci. Materials and methods. We have carried out whole genome SNP-analysis of Y. pestis strains of antique biovar, isolated in natural foci of Kyrgyz Republic in 1928–2016. Phylogenetic investigation is based on 1646 identified core SNPs in 51 included in the analysis strains of different phylogenetic lines. Phylogenetic tree was constructed using Maximum Likelihood algorithm, PHYML software package, and HKY85 model. Results and discussion. All 29 studied Y. pestis strains isolated between 1928 and 2016 in the foci of Tien-Shan and Pamir-Alay fall under the phylogenetic branches 0.ANT3 and 0.ANT5 of antique biovar of the main subspecies. Strains of 0.ANT3 branch were collected predominantly in Aksay and Alay foci, while strains of 0. ANT5 – in Upper-Naryn and Sarydzhas foci of Tien-Shan. Strains of phylogenetic lines 0.ANT1 and 0.ANT2 were not found in the foci of Kyrgyz Republic. According to the results of whole genome SNP-analysis, Y. pestis strains isolated in the XXI century belong to phylogenetic branch 0.ANT5. This branch also comprises a strain obtained from a patient (lethal case) in Sarydzhas plague focus in 2013. Genetic homogeneity of the modern strains of 0.ANT5 branch and their wide dissemination testify to the extension of the areal of this population and activation of Tien-Shan foci, caused by climate warming. For the first time ever, genomes of Y. pestis 0.ANT5 branch, isolated in XXI century, including clinical strain dated 2013, have been sequenced. We have obtained the whole-genome sequence of Y. pestis strain, 0.ANT3 branch, which caused pneumonic plague outbreak in Tien-Shan in 1928.

Abstract:
Soybean is one of the most economically important crops and has experienced adverse physiological and biochemical effects when subjected to drought stress and heat, resulting in lost productivity. Thus, the objective of this work was to evaluate the physiological, metabolic and growth responses of well-watered and drought-treated soybean cultivars under high temperature. The experimental design was set up in randomized blocks, in a factorial scheme with three soybean cultivars (7739 M, Anta 82 and Desafio) and two water levels (100% and 40% field capacity). The experiment was conducted in a controlled growth chamber with a gradual rise in temperature at 41°C for 5 hours daily. Morpho-physiological and metabolic analyses were performed 12 days after the treatments imposition. The parameters of water and osmotic potentials, relative water content, photosynthetic rate, stomatal conductance, transpiratory rate, electron flux for the carboxylation and oxygenation of RuBisCO were decreased for all cultivars under water deficit and high temperature. The results showed that the photorespiration and the rate of electrolyte leakage were increased as well. These results showed that these physiological behaviors are standard for soybean plants under water deficit, regardless of cultivars. The cultivars 7739 M and Desafio showed lower performance than the cultivar Anta 82 for the parameters of total electron flow and effective quantum yield of PS II. The 7739 M and Anta 82 were the only cultivars to show increased non-photochemical quenching dissipation and total soluble sugar content, respectively, under stress conditions. Desafio cultivar demonstrated greater physiological and growth traits stability, which could potentially indicate double tolerance to these stresses.

Abstract:
ABSTRACT The carbazole-degradative plasmid pCAR1 of Pseudomonas resinovorans strain CA10 has two gene clusters, carAaAaBaBbCAcAdDFE and antABC, which are involved in the conversions of carbazole to anthranilate and anthranilate to catechol, respectively. We proved that the antABC gene cluster, encoding two-component anthranilate 1,2-dioxygenase, constitutes a single transcriptional unit through Northern hybridization and reverse transcription-PCR (RT-PCR) analyses. The transcription start point of antA was mapped at 53 bp upstream point of its translation start point, and the −10 and −35 boxes were homologous to conserved σ70 recognition sequence. Hence the promoter of the ant operon was designated P ant . 5′ Deletion analyses using luciferase as a reporter showed that the region up to at least 70 bp from the transcription start point of antA was necessary for the activation of P ant . Luciferase expression from P ant was induced by anthranilate itself, but not by catechol. Two probable AraC/XylS-type regulatory genes found on pCAR1, open reading frame 22 (ORF22) and ORF23, are tandemly located 3.2 kb upstream of the antA gene. We revealed that the product of ORF23, designated AntR, is indispensable for the stimulation of P ant in Pseudomonas putida cells. Northern hybridization and RT-PCR analyses revealed that another copy of P ant , which is thought to be translocated about 2.1 kb upstream of the carAa gene as a consequence of the transposition of ISPre1, actually drives transcription of the carAa gene in the presence of anthranilate, indicating that both ant and car operons are simultaneously regulated by AntR.

Abstract:
Doing Western-type studies of Africa will always be problematic until we are able to wrest control of the paradigm from narrow Western objectives in order to satisfy the fundamental needs of Africa and not of the West. The late Cheikh Anta Diop, the greatest African scholar of the twentieth century, put forth the idea that Western scholars as a whole would find themselves unable to extricate their research from the intellectual interests of the West in order to serve the social and cultural aims of knowledge for Africa. Whether Cheikh Anta Diop was correct or not remains to be seen. However, from what we have already witnessed the most anti-African elements, whether in terms of the research priorities, projects, careers and programmatic orientations, structures, and objectives in the field, continue to have the most awesome hold on studies of Africa. The travesty is that it has lasted for so long.

Abstract:
The megalithic tomb of Currais do Galhordas (Castelo de Vide, Alto Alentejo, Portugal): Architecture, chronology and chemical analysis of the organic content of ceramic vasesThe organic content of three ceramic vases uncovered in the megalithic tomb of Currais do Galhordas (Central Eastern Portugal) – a monument built in the second half of the 4th millennium cal BC and reused in the 3th and in the 2nd millennia cal BC – was analysed by gas-chromatography with mass detection. The vases are allegedly connected with the latest use of the monument, during the Bronze Age (2nd millennium cal BC). Two of them presented traces of grapes or red fruits, together with fish; the third vase exhibited animal fat remains, possibly milk, associated with plant oils. Our results are compatible with other data from Iberian archaeological sites of identical period, in some cases located relatively close to the megalithic tomb of Currais do Galhordas.
