Full textThe growth of new vessels is called angiogenesis. Special growth factors e.g. basic fibroblast growth factor (bFGF), vascular endothelial growth factor (VEGF) are potent stimulators of angiogenesis in vivo and in vitro. Angiogenin another angiogenic protein was isolated first by Fett et al. 1985 from human carcinoma cells. Part I: The effect of angiogenin to stimulate angiogenesis in ischemic hind legs of inbred male Lewis rats is described. In this new animal model ischemia was induced by the ligature of the femoral artery. To stimulate angiogenesis angiogenic proteins (bFGF, angiogenin) were locally applied and compared to untreated ischemic animals. Already 1.9 ng locally applied angiogenin significantly stimulated the growth of collaterals in ischemic rat hind legs which could be demonstrated already 7 days after the application of angiogenin. The best stimulation of angiogenesis was achieved by the local application of 100 ng angiogenin which was 2.5-fold more angiogenic than 250 ng bFGF in the ischemic rat legs. The angiogenic effect of angiogenin as well as of bFGF was dependent on the applied dose reaching an optimum concerning angiogenin between 10 and 100 ng and for bFGF at 250 ng. Part II: The patency of PTFE vascular grafts with different angiogenic luminal coatings was tested in an animal model. In White New Zealand rabbits the abdominal aorta was replaced by a 5 cm long PTFE vascular graft 2 mm in diameter. Angiogenin and bFGF luminal coated PTFE grafts were 24 hours patent. Angiogenin coated grafts were after 8 hours occluded by a small distal thrombus. bFGF coated grafts were occluded after 6 hours. Heparine coated grafts were totally occluded by a thrombus after only 2 hours and gelatine coated grafts were already occluded after 1 hour. Part III: Protein Kinase C (PKC) isolated from rat brain was able to phosphorylat angiogenin with an effiency reaching 49% using Ca2+, Phospholipids and Dioleins as stimulators of the reaction. Histone V S was phosphorylated to 50% and bFGF to 84% by the same PKC. The determined Michaelis-Menten- Factor was concerning angiogenin km = 50 µM, bFGF km = 3.3 µM and Histone V S km = 10 µM. The separation of phosphorylated angiogenin was possible with the method of Sephadex G25 molecular sieve centrifugation as well as with the Centricon 10 membran centrifugation. The presented results demonstrate that angiogenin is a potent angiogenic protein stimulating the formation of new collaterals which was quantitatively evaluated in a new standardized rat animal model. The stimulation of indirect revascularization by angiogenin may be of therapeutical value in the treatment of ischemic cardio-vascular diseases. The patency of PTFE vascular prostheses 2 mm in diameter can successfully be tested in an animal model in White New Zealand rabbits by replacing the abdominal aorta by a 5 cm long PTFE vascular graft. Testing variant angiogenic luminal coatings of these PTFE grafts showed that a combination of bFGF/angiogenin had the best patency. Phosphorylation of angiogenin with PKC in vitro was possible reaching an effiency of 49% by using the stimulators Ca2+, Phospholipids and Dioleins for the reaction. Resumo: O objetivo deste estudo foi analisar a associação entre a angiogênese e as características anatomopatológicas do carcinoma do colo uterino, através da densidade de microvasos determinada pela técnica de imunoistoquímica com anticorpos monoclonais anti-CD34, BNH9 e anti-CD31. Para tal, foi realizado um estudo observacional analítico do tipo transversal, que incluiu 62 pacientes com carcinoma invasivo do colo uterino nos estádios Ibl, Ib2 e TIa da FIGO, que foram submetidas a histerectomia radical com linfadenectomia pélvica no período de janeiro de 2000 a fevereiro de 2002. A mensuração da angiogênese foi feita através da densidade de microvasos que correspondeu à média de microvasos por campo contados em dez campos. A densidade de microvasos foi maior quando se utilizou o anticorpo monoc1onal anti-CD34 e BNH9 do que com anti-CD31. O coeficiente de kappa foi de 0,74 (IC 950/0=0,56-0,91) quando analisou-se a concordância entre CD34 e BNH9 e os coeficientes de kappa foram de 0,35 (IC 95%=0,12-0,59) e de 0,29 (IC 95%=0,05-0,53), respectivamente, quando analisou-se a concordância entre CD34 e CD31 e entre CD31 e BNH9. Maior densidade de microvasos determinada pelo anticorpo anti-CD34 associou-se com carcinoma do colo uterino do tipo histológico escamoso, enquanto que com o anti-CD31 associou-se com o carcinoma indiferenciado. Maior densidade de microvasos determinada pelo anticorpo BNH9 associou-se com linfonodos comprometidos no carcinoma escamoso do colo uterino. Quando se utilizou os anticorpos anti-CD34 e BNH9 observou-se tendência de associação com invasão linfática para o carcinoma do colo uterino. Com bases nestes resultados, podemos concluir que a densidade de microvasos é maior quando utilizou-se os anticorpos monoclonais anti-CD34 e BNH9 do que com anti-CD31. A concordância para determinar a densidade de microvasos é melhor entre os anticorpos monoclonais anti-CD34 e BNH9. A atividade angiogênica é mais alta nos carcinomas escamosos, nos carcinomas indiferenciados e nos casos de carcinoma escamoso com linfonodos comprometidos
Abstract: The aim of this study was to analyse the association between angiogenesis and the pathoanatomic features of cervical cancer through microvessel density, determined by an immunohistochemical technique using anti-CD34, BNH9 and anti-CD31 monoclonal antibodies. For this purpose, a cross-sectional analytical observational study was conducted, including 62 patients with Stage Ibl, 1h2 and na cervical cancer as described by FIGO. These patients underwent radical hysterectomy with pelvic lymph node ressection 1Tom January 2000 to February 2002. Angiogenesis was measured through microvessel density which corresponded to the mean number of microvessels per field in 10 fields. Microvessel density was higher when using anti-CD34 and BNH9 monoclonal antibodies compared to using anti-CD31 monoclonal antibody (p<0,01). The kappa coefficient was 0.74 (CI 950/0=0.56-0.91) when analysing agreement between CD34 and BNH9. Kappa coefficients were 0.35 (CI 95%=0.12-0.59) and 0.29 (CI 95%=0.05-0.53), respectively, when analysing agreement between CD34 and CD31 and beween CD31 and BNH9. Higher microvessel density detennined by anti-CD34 antibody was associated with squamous cell cervical carcinoma, whereas it was associated with undifferentiated carcinoma when determined by anti-CD31 antibody. Higher microvessel density detennined by BNH9 antibody was associated with lymph node involvement in squamous cell cervical carcinoma. When anti-CD34 and BNH9 antibodies were used, a trend towards lymphatic invasion was observed in cervical carcinoma. Based on these results, we can conclude that microvessel density is higher when using anti-CD34 and BNH9 monoclonal antibodies compared to using anti-CD31. The agreeement between anti-CD34 and BNH9 monoclonal antibodies is better for determining microvessel density. There is a higher leveI of angiogenic activity in squamous cell carcinomas, undifferentiated carcinomas and squamous cell carcinomas with lymph node involvement

Resumo: Para investigar se carcinomas salivares com e sem diferenciação mioepitelial poderiam apresentar diferenças em relação ao nível de angiogênese, nós comparamos a vascularização tumoral entre os carcinomas adenóide cístico (31 casos) e epitelial mioepitelial (14 casos) versos carcinoma mucoepidermóide (37 casos). A expressão da peroxiredoxina I foi também estudada para verificar a relação potencial entre o metabolismo celular e a densidade microvascular. A densidade microvascular para o CD34 e CD105 foi significantemente menor em carcinomas com diferenciação mioepitelial. Porém, nenhuma relação foi encontrada entre a angiogênese e a quantidade de células mioepiteliaís. A forte expressão de peroxiredoxina I foi encontrada em 73.7% de carcinomas mucoepidermóides enquanto que 85.1% dos carcinomas com diferenciação mioepitelial apresentaram fraca expressão. Em conclusão, carcinomas com diferenciação mioepitelial, independente da quantidade de células mioepiteliaís, estão associados com significante baixa densidade microvascular. As razões para essa baixa atividade angiogênica ainda deve ser esclarecida, mas pode ser relacionada com as características metabólicas das células cancerosas.
Abstract: To investigate whether salivary carcinomas with and without myoepithelial differentiation could present differences regarding degree of angiogenesis, we compared tumor vascularization between adenoid cystic (31 cases) and epithelial- myoepithelial carcinomas (14) versus mucoepidermoid (37) carcinoma. The expression of peroxiredoxin I was also studied to verify the potential relationship between cellular metabolism and microvascular density. Microvascular density for CD34 and CD105 were significantly lower in carcinomas with myoepithelial differentiation. However, no correlation was found between degree of angiogenesis and amounts of myoepithelial cells. High-grade peroxiredoxin I expression was found in 73.7% of mucoepidermoid carcinomas whereas 85.1 of carcinomas with myoepithelial differentiation presented low-grade expression. In conclusion, carcinomas with myoepithelial differentiation, regardless of the amounts of myoepithelial cells, are associated to a significantly lower vascular density. The reasons for this lower angiogenic activity remain to be determined, but could be related to metabolic characteristics of the cancer cells.
