Full textThis work summarizes the results of the project “Molecular Endospectroscopy” at the Dresden University of Technology. The title expresses that tissue is characterized on the molecular level by coupling endoscopy and spectroscopy. Infrared (IR) and Raman spectroscopy offer advantages for these applications because they belong to the methods of molecular spectroscopy with the highest information content. Both methods probe molecular vibrations that provide a chemical fingerprint for the composition and structure of samples. The author was leader of a junior research group which developed vibrational spectroscopic methods for imaging of cells and tissues and applied them to clinical problems, in particular from the field of neuro-oncology. The current cumulative habilitation thesis is based on fourteen publications. The last one describes studies of a murine brain tumor model using a fiber-optic probe. In the first part various biophotonic methods are compared. Then biomedical applications of Fourier transform infrared (FTIR) and Raman imaging are reported. The papers are grouped into the chapters (i) Raman and FTIR imaging in neuro-oncology, (ii) FTIR microscopic imaging of tissue sections and (iii) Raman and FTIR microscopic imaging of single cells. It is discussed in the conclusions and outlook how molecular endospectroscopy as a new analytical tool can complement the standard diagnostic methods

Full textPollen represent one of the major causes for allergies in humans. Matrix-assisted laser desorption/ ionization time-of-flight is a suitable technique to characterize such complex biological samples. In pollen analyses, MALDI-TOF MS detects components that are extracted from the grain surface by the matrix solution and during laser desorption. Like any biological structure, pollen grains contain specific proteins, peptides, lipids and carbohydrates that produce characteristic signal patterns in the mass spectrum. The sample preparation applied in this work showed that the use of a conductive adhesive tape improves both the fixation of the pollen grains and the extraction of the analytes from them. The shown results prove that pollen of different plant species can be differentiated by comparing their respective mass spectral patterns. For the evaluation of such spectral patterns multivariate statistical methods, and in this work especially principal component analysis, are needed. PCA highlights differences in data sets and enables variance-weighting and improved classification. For efficient multivariate analysis, this method was integrated into a graphical user interface. In addition to the individual characterization of pollen samples, the mass spectrometric analysis of pollen grain mixtures posed a further challenge, since mixing and suppression effects can lead to discrimination of individual, possibly characteristic MS-peaks. For this purpose, the MALDI MS Imaging technique was applied. For the evaluation of such imaging data sets, further multivariate methods were applied, investigated, and evaluated. Thereby, in a model system individual pollen grains of different species could be identified and assigned their original position in the sample. The presented approach represents a new, reliable method for pollen determination, based on spectrometric-analytical basis rather than an individual visual assessment.

Full textIn this work, new concepts for the calibration free analysis by laser induced breakdown spectroscopy (LIBS) and for plasma diagnostics on laser induced plasma have been developed. In the course of this, already known techniques were analyzed for weaknesses and limitations. New techniques, based these findings, were presented. Using computer simulations of perfectly defined and entirely modifiable plasmas, the well known CF-LIBS (calibration free LIBS) technique was evaluated. The influence of all plasma parameters on the analysis result was investigated. It was shown that this technique, in its current form, is capable to deliver semi quantitative results only. An improvement of CF-LIBS was achieved by the use of spatially resolved measurements, overcoming the plasma heterogeneity. Combination of this approach with plasma simulation to find the parameters temperature and density, both important factors in calibration free analytics, much more stable results could be obtained. However, the well known technique for spatially resolved plasma investigation, the Abel Inversion, is limited to cylindrically symmetric plasmas only. Even weak disturbances of such symmetry can lead to major errors in the results. Using the inverse Radon Transformation, which is being introduced to plasma diagnostics on laser induced plasma by the present work, the first close to reality reconstructions and investigations of plasma with disturbed symmetry and even intrinsic asymmetry were conducted. This opens a new field in plasma diagnostics and helps to a better understanding of laser induced plasmas in general. It forms the foundation of further development of laser induced breakdown spectroscopy to become an absolute method.

Full textHyper-Raman scattering follows different symmetry selection rules than Raman scattering and, as a non-linear two-photon process, profits even more than Raman scattering from enhanced electromagnetic fields at the surface of plasmonic nanostructures. Surface-enhanced hyper-Raman scattering (SEHRS) could thus gain practical importance for spectroscopy. The combination of SEHRS and surface-enhanced Raman scattering (SERS) offers complementary structural information. Specifically, due to the localization of the enhancement to the close proximity of the nanostructures, this information can be utilized for the characterization of the interaction between molecules and metal surfaces. The aim of this work was to increase the understanding of the SEHRS effect and to assess its applicability to answer analytical questions. For that purpose, SEHRS experiments with excitation at 1064 nm and SERS experiments with excitation at the same wavelength, as well as with excitation at 532 nm - to detect SEHRS and SERS in the same spectral region - were conducted. As an example for non-resonant excitation, pH-dependent SEHRS and SERS spectra of para-mercaptobenzoic acid were examined. Based on these spectra, the interaction of different silver nanostructures with the molecules was characterized. beta-Carotene was used to study the influence of resonance enhancement by the excitation of a molecular electronic transition during SEHRS experiments. By the thiol-functionalization of carotene, a more intense interaction with the silver surface was achieved, which enables to obtain not only resonant SEHRS and SERS but also non-resonant SERS spectra of carotene. Hyperspectral SEHRS imaging in combination with microspectroscopy was demonstrated by analyzing the distribution of different dyes on structured plasmonic surfaces.

Full textRapid tests are widely used analytical methods for obtaining analytical information immediately on site. Surface enhanced Raman scattering (SERS) is a possible detection method for compounds in diluted samples. This thesis focuses mainly on three aspects: reproducibility of SERS signal intensity, interpretation of concentration curves and analysis of sample mixtures. The signal reproducibility was investigated using different deposition methods and measurement systems and the reproducibility of measurements was tested over longer periods of time. It was found that the most stable signal intensity was obtained using a combination of homogeneous deposition of a nanoparticle suspension on paper and a detection configuration that involves large diameters of both, the laser and the detection spot on the sample. It was shown with a laboratory setup, that comparatively stable measurements are possible with a relative standard deviation of approx. 20 % over a period of approx. 2.5 months. For the analysis and interpretation of the dependence of the SERS signal intensity on the concentration of the analyte, concentration series of different compounds were measured. The measurement data could be fitted with a Langmuir isotherm and explained with the Langmuir SERS model. For the measured thiol compounds an alternative option for quantification was found: the shift of certain SERS bands in the Raman spectrum as a function of analyte concentration. For the analysis of compound mixture in solution microfluidic paper-based analytical devices (µPADs) were used. It was observed that certain analytes which have a high affinity for the nanoparticles can be separated from the solution and thereby analyzed.

Full textUltrasonic levitation combines advantages of microfluidics like the required small sample volumes with a wall-less sample handling. While the coupling of analytical methods like optical spectroscopy as well as x-ray scattering are very well elaborated, an established mass spectrometric method to obtain molecular analytical information is still lacking. The herein presented work describes the fundamental processes for a contactless mass spectrometric analysis of levitated droplets. First, the influences of the specially designed levitator geometry on the levitation capabilities is described. During further experiments, the use of infrared lasers has proven useful as a combined desorption and ionization source for organic molecules from a mixture of water and glycerol as chromophore. Subsequently, sonic-spray ionization was used to gain a deeper understanding of the ionization processes occurring within the spray plume. Mass spectrometric mapping as well as laser-induced fluorescence were performed to investigate the ionization during an aerodynamic breakup of the micro droplets in the spray process. As a complementary sampling method, the ionization with a low- temperature plasma source is described. First, a time-resolved mass spectrometric investigation of the ionization process is shown. Sub- sequent to this fundamental study, the application of such a plasma source for the direct analysis of volatile compounds from within the droplets in the surrounding environment without interferences from the droplets bulk phase is described.

Full textSphingolipids are a family of bioactive signalling molecules. Besides their role in cellular structure, sphingolipids act as key regulators of many cellular functions including cell proliferation, differentiation, and apoptosis. The acid sphingomyelinase (ASM) catalyses the hydrolysis of sphingomyelin (SM) to ceramide, a metabolic hub of sphingolipid metabolism, and phosphoryl choline. Due to the lack of biochemical analytical tools, the molecular details of acid sphingomyelinase activity are still not fully discovered, and the development of pharmacological inhibitors is also hampered. The scope of this work is to develop new chemical probes for monitoring the activity of ASM both in vitro and in vivo.

However, an understanding of the reaction mechanisms can be considerably deepened with the help of time-resolved investigations. This work focuses on the use of time-resolved investigations, which rest largely on X-ray based analysis techniques, to identify the intermediate phases directly under realistic conditions. In the first part of this work, the reaction mechanisms of iron oxides and iron oxide hydroxides were investigated directly in a stabilizer-assisted synthesis. Time-resolved, simultaneous small-angle X-ray scattering and X-ray absorption spectroscopy experiments were performed on an iron oxide system for the first time using an acoustic levitator as sample holder after the optimisation of the instrumental set-ups. Further analytical methods were used to obtain detailed insights of the intermediate species in the three reaction mechanisms. Using X-ray photoelectron spectroscopy in combination with a microjet, species such as iron oxo oligomers could be detected as intermediates in the early stages of the hydrolysis process of Akaganeit nanoparticles. The first photoelectron spectrum for this species could be shown. In the last part of this work, time-resolved investigations were carried out on an aluminium oxo system. In situ-investigations were possible by combining wide-angle X-ray scattering experiments with the acoustic levitator. For the first time it was possible to detect the early stage crystallization of the Al13 cluster in a sulfate-containing solution as an aluminum sulfate cluster Al13SO4 directly in solution.

Full textThe screening of one-bead-one-compound (OBOC) libraries is a well-established technique for the identification of protein-binding ligands. The demand for binders with high affinity and specificity towards various targets has surged in the biomedical and pharmaceutical field in recent years. The combinatoric peptide screening traditionally involves tedious steps such as affinity selection, bead picking, sequencing and characterization. In this thesis, a high-throughput “all-on-one chip” system is presented to avoid slow and technically complex bead picking steps. Beads of a combinatorial peptide library are immobilized on a conventional glass slide equipped with an electrically conductive tape. The beads are applied by using a precision sieve, which allows the spatially ordered immobilization of more than 30,000 beads on one slide. For the target screening, the immobilized library is subsequently incubated with a fluorophore-labeled target protein. In a fluorescence scan followed by matrix-assisted laser desorption/ionization (MALDI)-time of flight (TOF) mass spectrometry (MS), high-affinity binders are directly and unambiguously sequenced directly from the bead. The use of an optimized ladder sequencing approach improved the accuracy of the de-novo sequencing step to 100 %. This new technique was validated by employing a FLAG-based model system. In a first step, new peptide binders for the M2 anti-FLAG monoclonal antibody were identified. Finally, this system was utilized to screen for IgG-binding peptides. The screening of about 30.000 peptides on one chip led to the identification of peptide binders in the mid micromolar range. A magnetic enrichment technique was developed to increase the number of screened beads. By combining the magnetic enrichment strategy with the chip system, 1 million beads were screened and IgG-binders in the low micromolar range were identified.

Full textThe focus of the thesis was the characterization of a monoclonal antibody, which binds specifically to cisplatin-treated DNA and is already used for diagnostic purposes in cancer therapy. The structure and composition of the antibody, as well, as the antigen specificity were investigated by mass spectrometry. Using nano-ESI-MS it was possible to analyse the antibody in its native state. Tandem-MS experiments revealed not only specific antibody fragments but also gave information on the primary structure of the antibody. The Fc-bound glycans that are responsible for antibody structure and functionality, were cleaved enzymatically and studied with MALDI-MS. For a detailed determination of the primary structure, the antibody was proteolysed with different enzymes and analyzed via high resolution Tandem-MS. The data was evaluated by comparism with results from sequence analysis on DNA level and known antibody sequences from protein databases. Simultaneously many different posttranslational modifications were identified. Furthermore, the antibody was successfully labelled with a metal complex at its sugar structures without loss of its structure and function. For investigation of the antibody specificity antigens were generated by incubation of well-defined single and double stranded DNA oligomers with cisplatin. Antibody-antigen-complexes were synthesized and analyzed by mass spectrometry under native conditions. The stability of the complex with double stranded DNA was also investigated by MS/MS-experiments. Thus, it was also possible to determine the specific fragmentation behavior for the complex just as for the free antibody.

Full textAs a total analysis system (TAS) an instrument is called which carries out complete chemical analysis procedures independently. The introduction of such systems offers a more efficient workflow in analytical laboratories because the sample manipulation, purification and the actual automated analysis can be carried out in one single operation. Specialized and already existing micro total analysis systems require currently a small amount of sample in the $\mu$L range. Owing to contamination, agglomeration and thus cross-secion reduction of incorporated channels in micro fluidics total analysis systems it can lead to a complete system interruption. Hence, the implementation of acoustic levitation in these systems is interessting alternative in order to avoid such kind of problems by abandoning vessels and wall contacts completely. To ensure acoustic levitation in micro total analysis systems can be successfully applied, technical development of analytical methods and coupling techniques is required. In the present work, the coupling of levitation technology and mass spectrometry is the prioritized topic but, in addition, spectroscopic experiments based on total reflections within the levitated droplet are as well realized in order to gain process insights. The particularly good reflection at the freely levitated droplet's circumference is due to the fact that the phase boundary between air and liquid is renewed by molecular interactions constantly and has no production-related rough surface. The combination of automated droplet generation, spectroscopy as well as the developed method for ion generation from the sample volume and mass spectrometry forms the basis of a novel micro total analysis system for small sample quantities.
