Abstract:
This master thesis deals with the field of possibilities of using social networks to increase competitiveness. In first part there are theoretically explained terms such as social network, social media and then there is described possibility to use social networks in business. The thesis includes theoretical treatment procedure entry of the company on social networks and describes appropriate monitoring and analyzing tools. The practical part is focused on dance club Beton, there are made the critical analysis of current attitudes of the club forward to social network, competitive analysis and comparison between Beton's and competitor's social network activities. The last part is devoted to selection appropriate monitoring tool and designed a social media monitoring system.

Abstract:
This thesis deals with the issue of social networks in terms of business support and their use for commercial purposes. Based on long-term statistics from the most widely used social networks in the Czech Republic, this thesis describes the main principles of using these social networks as a means of increasing profit or turnover. Particular attention is paid to Facebook, the leading social network in the online world. This study concerns the combination of marketing and the IT world, which nowadays has a key role in the sale of products and services. This study should show the importance of the combination of these two worlds. The practical part of the study is the analysis of the communication of the RunCzech Running League (formerly the Prague International Marathon), including a detailed analysis of one their competitors on the market. For the analysis appropriate metrics were selected, which were measured using special tools. The conclusion includes a comprehensive evaluation and presents suggestions for more effective future communication of the RunCzech running leagues in the field of social networking.

Abstract:
The diploma thesis is focused on the social networking sites in terms of their use for increasing the competitiveness of companies. The first part deals with a theoretical introduction to social networks where the basic concepts are defined and then characterized by major networks globally. Each social network is analyzed both in terms of the structure of its users, and are listed as individual options, which provides for promotion within their system - they are mentioned both paid and unpaid opportunities. At the end of this section are described the general possibilities of using social networking for business - ways to measure the impact of these activities, risks and requirements of their operation within the company. The second part is a practical solution to use social networks to support the selected e-commerce business. The introduction provides basic parameters of the e-shop is defined by the competitive environment. The analysis carried out various solutions to various social networks. It is designed and implemented their own social network to the target audience the company. The practical part ends with a final report including recommendations for further progress in the use of social networks. The main contribution of the paper is to present a detailed analysis of selected enterprise communication on social networks, including putting all the relevant data from which the conclusion can be drawn in specific recommendations to further enhance competitiveness.

The thesis deals with free tools that can be used to analyze customers of SMB e-commerce enterprises on the social network Facebook in order to increase their competitiveness within the social network. The theoretical part focuses on defining key terms, distinguishes types of social media and describes present social networks situation in world and in the Czech Republic from a users perspective and in the Czech Republic also from an enterprises perspective. Furthermore, the theoretical part also deals with possibilities that social networks can offer to the SMB enterprises. And as well it identifies different types of analytical tools that can be used for purposes of monitoring customers or analysis of customers. In the beginning of the practical part there is introduced the company Dobrutka.EU s.r.o. (the e commerce B2C company). Afterwards is presented the analysis of the survey about how SMB companies that operate the best Czech e-shops monitor and analyze their activities and customers from the social network Facebook. Subsequently there is handled the selection of appropriate tools for SME companies, which are also analyzed in the following chapter. At the end of the practical part there is practical application of knowledge from the thesis to Dobrutka.EU s.r.o. through the design and pilot deployment of appropriate procedures and analytic tools, whose effectiveness is evaluated at the end of chapter. The main benefit is in act of finding and verification of free tools and effective method that allows for detailed analysis of company customers even for micro companies and thus also effectively achieve positive economic results even in emerging facebook presentations.

This master’s thesis deals with realization of fault recorder designed for a distribution transformer station. In theoretical part is created research about operation of distribution transformer stations and about analytic methods, which gives bases for fault detection in power network. The practical part consists of design and realization of fault recorder in LabVIEW interface using CompactDAQ measurement platform. Purpose of this recorder is monitoring and recording of voltage and current waveforms on secondary side of transformer station during the fault in high voltage network. Principle of fault detection is primarily based on level monitoring of voltage negative symmetrical component. Fault recorder has been successively optimized and the final version is presented in this thesis. At the end functionality of designed system is evaluated on simulated fault in a model of high voltage network.

This Diploma Thesis' main subject is Internet marketing. Its main objective is suggesting and realizing an online marketing campaign focused on using Facebook and Google AdWords in order to building brand awareness of a small business. This effort is followed by an analysis of its results. The contribution should be apparent to all the marketers of other small businesses, who find themselves in similar situation and try to solve it by using modern online marketing tools and channels. The thesis is divided into two main sections. The first covers necessary theoretical bases of online marketing, the next one puts the theoretical knowledge into practice of a real small company, Saloon Exclusive. Abstract:
Práce pojednává o možnostech získávání dat a jejich analýzy ze zachycené síťové komunikace. Jsou zhodnoceny možnosti aktuálně dostupných řešení jednotlivých nástrojů i celých prostředí pro síťovou forenzní analýzu. Provedením analýzy těchto nástrojů byly zjištěny nedostatky, pro které není možná integrace již hotových řešení pro záměry projektu SEC6NET, a dále byly stanoveny cíle, které navržené řešení musí splňovat. Na základě cílů a znalostí z předchozích prototypů řešení byla provedena dekompozice problému na jednotlivé funkčně související bloky, které byly implementovány jako nezávislé moduly schopny spolupráce. Správná funkcionalita je po každé změně v implementaci testována pomocí sad Unit testů, které pokrývají majoritní část kódu. Před zahájením samotného vývoje bylo nutné zhodnotit aktuální situaci v komerčních i open-source sférách řešení. Srovnání nástrojů používaných pro forenzní síťovou analýzu nám dalo jasnou představu, na kterou část trhu chce naše řešení směřovat a jaká funkčnost je v jednotlivých nástrojích nepříliš povedená. Následně byly stanoveny hlavní požadavky a směr, kterým by se měl vývoj ubírat. Na začátku vývoje rekonstrukčního frameworku stála fáze vytvoření návrhu architektury a dekompozice průběhu zpracování zachycené komunikace do ucelených částí jednotlivých modulů. Využití předchozích znalostí a zkušeností získaných vývojem rekonstrukčního nástroje Reconsuite nám pomohlo při formování fronty zpracování, kterou budou data při zpracování procházet. Následně byly navrženy základní komponenty provádějící práci se zachycenou komunikací v různých formátech PCAP souborů, rozdělení komunikace na konverzace, provedení defragmentace na úrovni IP a v případě komunikace TCP provedení reassemblingu daných toků. V rané části vývoje jsme se zaměřili na komunikaci zapouzdřenou v nízkoúrovňových protokolech Ethernet, IPv4/IPv6, TCP a UDP. Po definici rozhraní komponent bylo nutné provést další výzkum síťových protokolů a vytvoření algoritmů pro jejich zpracování ze zachycené komunikace, která se liší od standardní a není tedy možné ji zpracovávat dobře známými postupy z RFC či jader operačních systémů. Protože proces zpracování zachycených dat se na komunikaci přímo nepodílí, tak v případě, kdy dojde ke ztrátě či poškození při zachycení, nebo je komunikace směřována jinou cestou, atd., není možné data získat pomocí znovu zasílání, ale je nutné využít jiné mechanismy k označení či obnově takto chybějících dat - algoritmus provádějící IP defragmentaci a TCP reassembling. Po implementaci a otestování byl zjištěn problém se separací jednotlivých TCP toků (TCP sessions), který nebylo možné řešit původním návrhem. Po analýze tohoto problému byla změněna architektura procesní pipeline s výsledným zvýšením počtu rekonstruovaných dat v desítkách procent. V závěrečné fázi je popsána metodologie jakou bylo porvedeno testování výkonu implementovaného řešení a srovnání s již existujícími nástroji. Protože rekonstrukce aplikačních dat je příliš specifická záležitost, při srovnání výkonu byla měřena rychlost zpracování a potřebná paměť pouze při provádění separace toků, IPv4 defragmentace a TCP reassemblingu, tedy operace společné pro všechny rekonstrukční nástroje. Srovnání ukázalo, že Netfox.Framework předčí své konkurenty Wireshark i Network monitor v rychlosti zpracování, tak v úspoře paměti. Jako testovací data byl použit jak generovaný provoz, tak i vzorky reálné komunikace zachycené v laboratorním prostředí.

Abstract:
The main goal and contribution of this thesis are to create an artifact in the form of model that supports a recruitment of employees via social media networks and its implementation into practice. For this purpose has been used a depth analysis of current state of researched problematics leading to finding gaps that could be eventually solved while using mentioned model. The Author takes advantage of knowledge obtained from literature research and also from self-created research, particularly the analysis of content type investigated social media networks and specific data obtained manually and automatically; Comparative analysis of job advertisements; periodic questionnaire survey amongst HR staff. A significant part of the research has also been an investigation of already existing frames of social media networks recruitment. A depth analysis of current state determines that there is existence only of few models dealing with social media networks recruiting. Furthermore, these models lack candidate evaluation based on the social media networks behavior. Artifact as a model that supports a recruitment of employees via social media networks contains a suggestion of an automatic solution dealing with user´s data downloading and also a suggestion of subsequent analytical data processing and the creation of a predictive model for assessing the user´s behavior on the social media networks. Final evaluation of the effectiveness of the proposed model is done through the formal verification process and the case study. The case study verification has the suggested artifact been implemented in practice with a name Model PM that is using a recruiting application (Práce na míru) for an extraction of Facebook´s data. For user´s behavior predictor setting has been used a character test (MBTI). With the help of the cluster analysis and machine learning (Decision trees) has been created Stochastic predictive model that determines a character type of particular candidate (The accuracy of the prediction of the MBTI personality category is in the range between 68 % to 84 % in individual cases with confidence value between 43 % and 81 %). The case study verified a usefulness of model that supports a recruitment of employees via social media networks, and afterward, the mode has been implemented into the practice.

Transportation procurement is one of the most critical sourcing decisions to be made in many companies. This thesis addresses a real-life industrial problem of creating package bids for a company's transportation procurement auction. The purpose of offering package bids is to increase the carriers' capacity and to improve the reliability of services. In this thesis, we investigate the possibility of forming packages using the company's own distribution network. Effective distribution of packages requires balanced cycles. A balanced cycle is a cycle containing no more than 3 nodes with equal loads (or volume of package) on every link in the cycle. We develop mixed-integer programs to find the maximum amount of network volume that can be covered by well-balanced cycles. These models are deterministic models that provide a rough guide on the optimal way of package formation when loads are known in advance. Since demand is random in real life, we perform a stochastic analysis of the problem using various techniques including simulation, probabilistic analysis and stochastic programming. Results from the stochastic analysis show that the effectiveness of package distribution depends on how we allocate the volumes on the lanes to create balanced cycles. If we always assign a fixed proportion of the lanes' volumes to the cycles, then it is only possible to have well-balanced cycles when the average volumes on the lanes are very large, validating the advantage of joint bids between several companies. However, if we assign a different proportion of the lanes' volumes to the cycles each time demand changes, then it is possible to create cycles that are balanced most of the time. An approximated solution method is provided to obtain a set of balanced cycles that can be bid out.

Abstract:
The aim of this master's thesis is to design a language through which it would be possible to monitor a stream of network flows in order to detect network policy violations in the local network. An analysis of the languages used in the data stream management systems and an analysis of tasks submitted by the potential administrator were both carried out. The analysis specified resulted in the language design which represents pipelining consisting of filtering and aggregation. These operations can be clearly defined and managed within security rules. The result of this thesis also results in the Policer modul being integrated in the NEMEA system, which is able to apply the main commands of the proposed language. Finally, the module meets the requirements of the specified tasks and may be used for further development in the area of monitoring network policies.

New challenges are being raised in the networking field with the increasing number of connected devices. The growth of mobile data usage has to be considered as a requirement for the deployment of future 5G networks, especially regarding mobility scenarios. Software-Defined Networking (SDN) enables a greater degree of dynamism and simplification for the deployment of those 5G networks. SDN provides the separation of the control plane from the forwarding plane, allowing more control, adaptability and cost reduction. The growth of SDN integration in new mechanisms and network architectures led to the development of different controller solutions, with a wide variety of characteristics. Several SDN controllers exist, which originated from the different needs of operators and research teams. That resulted in the development of their own controller versions, which made comparison efforts more difficult. As such, this work provides a wider study of several open-source controllers, (namely, OpenDaylight (ODL), Open Network Operative System (ONOS), Ryu and POX), by evaluating not only their performance, but also their characteristics in a qualitative way. Taking performance as a critical issue among SDN controllers, several criteria were evaluated by benchmarking the controllers under different operational conditions, using the Cbench tool. Results are presented regarding both qualitative and quantitative comparisons between those SDN controllers under test.
Com o aumento do número de dispositivos ligados em rede, surgem novos desafios no ramo das redes. A necessidade de acompanhar o crescimento da utilização de dados móveis é um dos requisitos a ter em conta nas futuras redes 5G (5a Geração), sobretudo em cenários de mobilidade. As redes controladas por software (do inglês, Software-Defined Networking (SDN)) permitem a simplificação e dinamismo necessários à criação das referidas redes 5G. As SDNs promovem ainda a separação do plano de controlo do plano de dados, permitindo um maior controlo, adaptabilidade e redução de custos. O crescimento da tecnologia SDN levou ao desenvolvimento de diferentes controladores, com diferentes características. Existem vários controladores SDN, com origem em diferentes necessidades dos operadores e equipas de investigação. Este desenvolvimento individualizado tornou as comparações entre os controladores mais difíceis. Deste modo, o trabalho desenvolvido fornece um estudo mais abrangente de vários controladores open-source (OpenDaylight (ODL), Open Network Operative System (ONOS), Ryu and POX), avaliando não só a sua performance como as suas características de uma forma qualitativa. Considerando a performance crucial nos controladores SDN, foram considerados vários critérios na avaliação dos controladores sob diferentes circunstâncias, utilizando a ferramenta Cbench. Os resultados apresentados são relativos à comparação qualitativa e quantitativa dos controladores em teste.

The thesis investigates APT attacks, which are professional targeted attacks that are characterised by long-term duration and use of advanced techniques. The thesis summarises current knowledge about APT attacks and suggests seven symptoms that can be used to check, whether an organization is under an APT attack. Thesis suggests a system for detection of APT attacks based on interaction of those symptoms. This system is elaborated further for detection of attacks in computer networks, where it uses user behaviour modelling for anomaly detection. The detector uses k-nearest neighbors (k-NN) method. The APT attack recognition ability in network environment is verified by implementing and testing this detector.

The focus of this thesis is the fast network anomaly detection based on automata learning. It describes and compares several chosen automata learning algorithms including their adaptation for the learning of network characteristics. In this work, various network anomaly detection methods based on learned automata are proposed which can detect sequential as well as statistical anomalies in target communication. For this purpose, they utilize automata's mechanisms, their transformations, and statistical analysis. Proposed detection methods were implemented and evaluated using network traffic of the protocol IEC 60870-5-104 which is commonly used in industrial control systems.

This thesis is aimed to exploitation of fundamental analysis in automatic trading. Technical analysis uses historical prices and indicators derived from price for price prediction. On the opposite, fundamental analysis uses various information resources for price prediction. In this thesis, only quantitative data are used. These data sources are namely weather, Forex, Google Trends, WikiTrends, historical prices of futures and some fundamental data (birth rate, migration, \dots). Experiments show major improvement in results of the trading system; 8\% increase in success prediction accuracy thanks to involvement of fundamental analysis.

This dissertation thesis deals with application of probabilistic approach to assessment of earthing system safety in distribution networks, especially for cases with common earthing of high and low voltage side of distribution transformers HV/LV. In these cases, the increased potential during fault might be transferred from high voltage to low voltage network and thus the individuals from public can be exposed to increased risk. Thus, for these cases were in this thesis defined expectable touch scenarios together with the resulting risk imposed on individuals from the public. Based on the results it seems that adoption of probabilistic approach for these cases of earthing systems might be more suitable compared to the conventional deterministic worst case approach. In accordance to the aims of the thesis, a thorough analysis of currently adopted probabilistic approaches was carried out as well and it was pointed out to some new possible simplifications in the adopted probabilistic based methodologies. For example, it seems that appropriate modelling of human body resistance by the full lognormal distribution is not completely necessary and similar results can be obtained when only the resistance for 50 % of population together with c3 and c4 fibrillation curves are used. Much of the work was also devoted to the determination of possible uncertainty of calculated risk of evaluated earthing system, especially due to inappropriate modelling of earthing system. The appropriateness of different earthing system modelling methods together with other parameters on the value of calculated risk was investigated through conducting sensitivity analysis. Based on the analysis results it seems, that due to using more, or less simplified modelling method, a possible underrating in the resulting risk of about 40 % (about half an order/decade) is expectable. On the other hand, the change of parameters related directly to calculation of fibrillation probability seems to exhibit greater change in calculated risk by up to units of orders/decades.

This thesis deals with an echo state network and with acceleration of its learning by implementing the echo state network on a graphics processor. The theoretical part consists of the description of neural networks and some selected types of neural networks, on which is based the echo state network. After that, there are some other algorithms described used for time series analysis and last but not least, the tools that were used in the practical part of the thesis were briefly described. The practical part describes the creation of the accelerated version of the echo state network. After that, there is described the creation of input data sets of real financial indexes, on which the echo state network and the other algorithmns were then tested. By analyzing this accelerated version it was found that its learning speed did not reach the theoretical expectations. The accelerated version works slower, but with greater precision. By analyzing the results of the measurement of the other algorithmns it was found that the highest precision is achieved by solutions based on the neural network principle.

This thesis is focused on network behavior analysis (NBA) designed to detect network attacks. The goal of the thesis is to increase detection accuracy of obfuscated network attacks. Methods and techniques used to detect network attacks and network traffic classification were presented first. Intrusion detection systems (IDS) in terms of their functionality and possible attacks on them are described next. This work also describes principles of selected attacks against IDS. Further, obfuscation methods which can be used to overcome NBA are suggested. The tool for automatic exploitation, attack obfuscation and collection of this network communication was designed and implemented. This tool was used for execution of network attacks. A dataset for experiments was obtained from collected network communications. Finally, achieved results emphasized requirement of training NBA models by obfuscated malicious network traffic.

Priority based scheduling disciplines are widely used by existing computer operating systems. However, the mathematical analysis and modelling of these systems present great difficulties since priority schedulling is not compatible with exact product form solutions of queueing network models (QNM's). It is therefore, necessary to employ credible approximate techniques for solving QNM's with priority classes. The principle of maximum entropy (ME) is a method of inference for estimating a probability distribution given prior information in the form of expected values. This principle is applied, based on marginal utilisation, mean queue length and idle state probability constraints, to characterise new product-form approximations for general open and closed QNM's with priority (preemptive-resume, non-preemtive head-of-line) and non-priority (first-come-first-served, processor-sharing, last-come-first-served with, or without preemtion) servers. The ME solutions are interpreted in terms of a decomposition of the original network into individual stable GIG11 queueing stations with assumed renewal arrival processes. These solutions are implemented by making use of the generalised exponential (GE) distributional model to approximate the interarrival-time and service-time distributions in the network. As a consequence the ME queue length distribution of the stable GE/GEzl priority queue, subject to mean value constraints obtained via classical queueing theory on bulk queues, is used as a 'building block' together with corresponding universal approximate flow formulae for the analysis of general QNM's with priorities. The credibility of the ME method is demonstrated with illustrative numerical examples and favourable comparisons against exact, simulation and other approximate methods are made.

V této práci se zabýváme vylepšením systémů pro odhalení síťových průniků. Konkrétně se zaměřujeme na behaviorální analýzu, která využívá data extrahovaná z jednotlivých síťových spojení. Tyto informace využívá popsaný framework k obfuskaci cílených síťových útoků, které zneužívají zranitelností v sadě soudobých zranitelných služeb. Z Národní databáze zranitelností od NIST vybíráme zranitelné služby, přičemž se omezujeme jen na roky 2018 a 2019. Ve výsledku vytváříme nový dataset, který sestává z přímých a obfuskovaných útoků, provedených proti vybraným zranitelným službám, a také z jejich protějšků ve formě legitimního provozu. Nový dataset vyhodnocujeme za použití několika klasifikačních technik, a demonstrujeme, jak důležité je trénovat tyto klasifikátory na obfuskovaných útocích, aby se zabránilo jejich průniku bez povšimnutí. Nakonec provádíme křížové vyhodnocení datasetů pomocí nejmodernějšího datasetu ASNM-NPBO a našeho datasetu. Výsledky ukazují důležitost opětovného trénování klasifikátorů na nových zranitelnostech při zachování dobrých schopností detekovat útoky na staré zranitelnosti.

Com o aumento do número de plataformas de transmissão de vídeo, as operadoras têm sofrido uma maior sobrecarga nas suas redes. De forma a fornecer uma melhor gestão dessas mesmas redes, garantindo qualidade de serviço a todos os clientes, torna-se necessário dar prioridade ao tráfego correspondente a vídeo aplicando novos conveitos na área das telecomunicações, como é o caso de Software-Defined Networking. Esta dissertação procura, numa primeira fase, apresentar uma revisão de vários temas relacionados com a determinação de tráfego de vídeo, Software-Defined Networking e qualidade de serviço. Posteriormente, é apresentada uma solução de uma aplicação de monitorização, que tem como objetivo, a deteção de tráfego de vídeo, de forma a ajudar na priorização de tráfego e na otimização da rede. A solução é validada através de uma implementação, baseada na performance e na baixa latência do sistema, que procura responder o mais rápido possível com informação sobre um determinado fluxo de pacotes na rede. São ainda apresentados resultados relativos a esta implementação.
With the increase of live streaming platforms, service providers have been experiencing a overhead on their networks. In order to provide a better management of these networks, ensuring quality of service to all customers, it is necessary to prioritize video traffic using new concepts being introduced into the telecommunications field, such as Software-Defined Networking. Firstly, this dissertation aims to present a review of several topics related with video traffic determination, Software-Defned Networking and quality of service. Secondly, a monitoring application solution is presented, which aims to detect video traffic in order to help the prioritization of traffic and network optimization. The solution is validated through an implementation, based on the system’s performance and low latency, which tries to reply as quickly as possible with information about a certain flow of network packets. Results related with this implementation are also presented

As a part of the Modern Tools for Detection and Mitigation of Cyber Criminality on the New Generation Internet project, a Lawful Interception System was developed. This thesis describes additions to the system, which provide a capability to intercept application protocols (eg. an e-mail communication) directly in a network of an Internet service provider. This new functionality enables automatic detection and filtering of a related TCP transfer. It is also able to handle situations, in which the identity (an IP address) of a target user is not known yet, or when it is difficult to detect it (NAT is in progress, user is at an Internet café, behind the firewall, etc.). One of the most important requirements for the developed prototype is the ability of a fast packet proccessing with maximum throughput and minimal packet loss. Therefore, this thesis also consists of a performance profiling, an identification of critical points and their optimalization.

Análise de Redes Metabólicas foi aplicada à cepa de Saccharomyces cerevisiae CEN.PK113-7D, e a alguns mutantes interrompidos em genes que codificam para proteínas regulatórias envolvidas no fenômeno de repressão por glicose. Todas as cepas foram cultivadas em aerobiose, em meio mínimo contendo [1-13C]glicose como substrato limitante. As células eram recolhidas em situação de crescimento balanceado e submetidas à hidrólise, seguida de derivação e posterior injeção da amostra resultante num cromatógrafo gasoso acoplado a um espectrômetro de massa, para análise da marcação em alguns fragmentos de metabólitos intracelulares. Estes dados serviram como base para a identificação da atividade de algumas vias metabólicas no metabolismo central de S. cerevisiae. Além disto, utilizando-os juntamente com um modelo estequiométrico, foi possível obter uma estimativa para os fluxos no metabolismo central na cepa referência e nos mutantes estudados. Num primeiro momento, a metodologia foi validada para cultivos contínuos e descontínuos. Calculou-se um desvio padrão para a medida da marcação em cada fragmento de metabólito detectado pela metodologia empregada. Na cepa referência, observou-se que o ciclo de Krebs opera de forma cíclica em células que respiram e de forma não cíclica em células que apresentam metabolismo respiratório-fermentativo. Verificou-se que uma maior parte da glicose consumida é desviada para a via das pentoses fosfato no primeiro caso, em relação ao segundo. Foram encontradas evidências para a biossíntese de glicina através da enzima treonina aldolase e para a atividade da enzima málica. A ausência das proteínas Mig1 e Mig2 não altera os padrões de crescimento, produção de etanol e de marcação em metabólitos intracelulares de S. cerevisiae. Já a ausência de Hxk2, Reg1 ou Grr1 provoca alívio na repressão por glicose, observado pelo aumento das atividades respiratórias.
Metabolic Network Analysis was applied to the reference strain CEN.PK113-7D of Saccharomyces cerevisiae, as well as to some mutants disrupted in genes which code for regulatory proteins involved in the glucose repression cascade. All strains were cultivated under aerobic conditions, using minimal medium with [1-13C]glucose as the limiting substrate. Cells were harvested under balanced growth conditions and submitted to hydrolysis, derivatization and injection of the sample into a gas chromatograph coupled to a mass spectrometer for analysis of the labeling pattern in some fragments of intracellular metabolites. These data were used for identifying the activity of some pathways in the central metabolism of S. cerevisiae. Furthermore, using the data together with a stoichiometric model, it was possible to estimate the fluxes in the central metabolism of the reference strain and in the mutant strains. First, the methodology was validated for batch and continuous cultivations. Standard deviations were calculated for the measurement of the fractional labeling in each of the detected fragments. In the reference strain, it was observed that the Krebs cycle operates in a cyclic manner in respiratory cells, whereas it operates in a non cyclic manner under respiro-fermentative metabolism. It was also seen that a greater part of the glucose consumed by the cells enters the pentose phosphate pathway in the former than in the later case. Evidence for the activity of the threonine aldolase and the malic enzyme catalyzed reactions was also found. The absence of the Mig1 and Mig2 proteins does not alter the growth, ethanol formation and labeling pattern of intracellular metabolites in S. cerevisiae. In contrast, the absence of Hxk2, Reg1, or Grr1 provoques a relief in glucose repression, which was observed by an increased respiratory activity.

Abstract:
The findings from chronic complex diseases modelled in animals are difficult to extrapolate to humans. In metabolic (dysfunction) associated fatty liver disease (MAFLD), dietary models are commonly used to study the mechanisms for disease progression. The current dogma is that diets rich in fat, simple carbohydrates, and cholesterol can lead to systemic alterations in metabolism that leads to the accumulation of lipids in the adipose and liver tissues. This leads to lipotoxicity and a vicious cycle of inflammation and liver injury which can drive disease progression. Hence, there has been a considerable interest to model and comprehend the role of metabolism and the immune response in this disease. However, our knowledge of the underlying mechanisms in MAFLD is still limited. A major caveat is that pre-clinical models do not represent the full spectrum of human diseases. This has been a major source of failure in clinical trials. Understanding the impact of dietary challenges in animal models and where they resemble or diverge from human disease can help to resolve the current dilemmas that have hampered progress in the field. In this thesis, I used different dietary models in mice and characterised liver pathology, the liver immune profile, and the landscape of liver gene expression. Initially, we used multiple dietary models containing simple (sucrose) or complex carbohydrates, with/without cholesterol (2%), and with/without an added bile acid (cholic acid): A) normal chow (NC); B) high sucrose (HS); C) high sucrose and high cholesterol (HS_Chol2%); D) high sucrose, high cholesterol (2%) and cholic acid (HS_Chol2%_CA); E) high cholesterol (2%) and cholic acid (Chol2%_CA); F) cholic acid (CA). From a liver pathology perspective, diets containing cholesterol (Diets C to E) induced a dramatic change in liver pathology. Consistently, immune profiling of the liver of mice fed these diets induced infiltration of a broad range of immune cells including myeloid and lymphoid cells (diets C to E). Of note, the combination of cholesterol and cholate (diets D and E) had synergistic effects and dramatically enhanced liver immune cell infiltration. Subsequently, we performed RNA sequencing on the liver of mice fed these 6 diets. In agreement, we detected the highest differentially expressed genes in diets containing cholesterol and cholate (diets D and E). We conclude that this combination disturbs liver homeostatic functions the greatest. To gain a systems perspective of perturbations in liver homeostatic function, we undertook a systems approach and applied weighted gene co-expression network analysis (WGCNA) on liver transcriptomes. We noticed several gene expression modules (networks) that were associated with diets. Most of these modules were enriched for metabolic pathways and immune responses. Of interest, there was a negative correlation between immune and metabolic-related modules. This was reminiscent of immunometabolism and a co-variance in gene regulatory networks between metabolic and immune modules. The up-regulation of immune responses and down-regulation of metabolic networks within the liver were prominent in mice exposed to cholesterol and cholate. A published report and our unpublished data (not the subject of this thesis) indicated that diets containing cholesterol and cholate induce a heightened immune response with anti-tumorigenic properties. Thus, from a phenotypic perspective, this immune response and the outcomes are divergent from human fatty liver that increases the risk of liver cancer. One caveat in our dietary models was that they contained supra-physiological levels of cholesterol (2%). I detected a suppression in the expression of genes in cholesterol biosynthesis pathway in all diets that contained cholesterol (Diet C) or cholic acid (Diets D to F). This was in contrast with a study on clinical fatty liver disease (in humans) which reported up-regulation of genes in cholesterol biosynthesis pathways. I hypothesised that reduction in cholesterol biosynthesis could be related to a higher immune response. Hence, I omitted cholic acid and reduced the amount of cholesterol to 0.2% and investigated the liver pathology in mice exposed to a HS_Chol0.2% diet. This diet also induced minimal liver pathology similar to the HS diet. Despite a reduction in the cholesterol content, I detected a suppression in the expression of genes in cholesterol biosynthesis in mice exposed to the 0.2% cholesterol in the diet. This indicates a diverged response in cholesterol metabolism between mice and human liver. This diet did not induce pathological features resembling human MAFLD, however it resembled some characteristics of metabolic syndrome such as adiposity with systemic glucose intolerance. One of the models that is often used to simulate MAFLD is the MCD (methionine choline deficient) diet. This diet induced pathological features resembling human MAFLD, however, our analysis of liver transcriptome data on mice fed with MCD diet indicated down-regulation in metabolic pathways and cholesterol biosynthesis. Indeed, the behaviour of these modules in mice on the MCD diet resembles those in mice exposed to high levels of cholesterol, which are divergent from human fatty liver disease. Overall, my results have shown that mice dietary models do not fully resemble clinical fatty liver either phenotypically or at the gene expression level. A possible strategy to overcome these limitations is to use multiple models in which each model could represent a specific aspect of the disease. At the molecular level, undertaking a module-based approach to understand the link between the behaviour of preserved modules (e.g., between mice and humans) to phenotypic outcome is an alternative strategy. One unmet need in human fatty liver is dissociating liver tissue inflammation from protective immune responses (immunosurveillance), which I would like to delve into in my future endeavours.

The correct classification of individuals in pre-established groups has become of great importance in breeding. The multivariate statistical techniques commonly used in this type of problem are the discriminant functions of Fisher and Anderson, which are used to allocate an initially unknown individual in one of the g populations or pre-defined groups. In recent decades a new computing paradigm, artificial neural networks, has come along to solve various problems of Statistics, such as grouping of similar individuals, time series forecasting and also of particular interest, the problem of classification. The objective of this research was to conduct a simulation study in order to compare the discriminant functions of Fisher and Anderson and neural networks. We evaluated the number of incorrect classifications of individuals known to belong to different populations with different levels of dissimilarity measured by the Mahalanobis distance. Simulations were conducted using the software Genes (Cruz, 2006). Although Artificial Neural Networks presented a rate of incorrect classification of individuals rejected for being considered ambiguous as to its discriminatory characteristics, it proved to be a promising technique, since it presented a lower number of incorrect classifications of individuals when compared to the discriminant functions.
A correta classificação de indivíduos em grupos pré-estabelecidos tem se tornado de grande importância no melhoramento genético. As técnicas de estatística multivariada usualmente utilizadas nesse tipo de problema são as funções discriminantes de Fisher e as funções discriminantes de Anderson, que são usadas para alocar um indivíduo inicialmente desconhecido em uma das g populações ou grupos pré-definidos. Nas últimas décadas vêm surgindo um novo paradigma de computação, as redes neurais artificiais, que podem ser utilizadas para resolver diversos problemas da Estatística, como agrupamento de indivíduos similares, previsão de séries temporais e em especial, os problemas de classificação. O objetivo dessa pesquisa foi realizar um estudo comparativo entre as funções discriminantes de Fisher e de Anderson e as redes neurais artificiais quanto ao número de classificações erradas de indivíduos sabidamente pertencentes a diferentes populações, com distintos níveis de dissimilaridade. Essa dissimilaridade, medida pela distância de Mahalanobis, foi um conceito de fundamental importância na utilização das técnicas de discriminação, pois quantificou o quanto as populações eram divergentes. Quanto maior o valor observado para essa medida, menos similares foram as populações em análise. A obtenção dos dados foi feita através de simulação utilizando o programa computacional Genes (CRUZ, 2006). As redes neurais artificiais apresentaram uma taxa de indivíduos rejeitados por serem considerados ambíguos quanto às suas características discriminatórias. No entanto, mostraram-se uma técnica promissora no que diz respeito a problemas de classificação, uma vez que apresentaram um número de classificações erradas de indivíduos menor que aqueles dados pelas funções discriminantes.

The next generation wireless system (NGWS) has been conceived as a ubiquitous wireless environment. It integrates existing heterogeneous access networks, as well as future networks, and will offer high speed data, real-time applications (e.g. Voice over IP, videoconference ) and real-time multimedia (e.g. real-time audio and video) support with a certain Quality of Service (QoS) level to mobile users. It is required that the mobile nodes have the capability of selecting services that are offered by each provider and determining the best path through the various networks. Efficient radio resource management (RRM) is one of the key issues required to support global roaming of the mobile users among different network architectures of the NGWS and a precise call admission control (CAC) scheme satisfies the requirements of high network utilization, cost reduction, minimum handover latency and high-level QoS of all the connections. This thesis is going to describe an adaptive class-based CAC algorithm, which is expected to prioritize the arriving channel resource requests, based on user¿s classification and channel allocation policy. The proposed CAC algorithm couples with Fuzzy Logic (FL) and Pre-emptive Resume (PR) theories to manage and improve the performance of the integrated wireless network system. The novel algorithm is assessed using a mathematical analytic method to measure the performance by evaluating the handover dropping probability and the system utilization.
