Full textOver the last decade, the discovery of microRNAs revealed a new mechanism of post-transcriptional regulation. MicroRNAs are involved in many biological processes such as development, differentiation, proliferation and cell death. Moreover, several evidences showed the pathogenic role of microRNAs in various diseases. A lot of studies used microarray technology to identify miRNAs involved in the pathogenesis, but also to obtain the expression pattern characteristic of pathology with diagnostic or prognostic assessment. These studies suggest that profiling of microRNAs may be used to understand the role they play in regulating pathophysiological processes. In this work we employed microarray technology to investigate the expression profile of microRNAs in two different experimental models: 1) an in vitro model, useful for understanding the molecular mechanisms underlying the immune response, 2) a in vivo model, suitable for studying the pathogenesis of non-alcoholic fatty liver disease, also known as NAFLD. Recently, has been explored the relationship between inflammation, innate immunity and microRNAs, which are described to be involved in regulating cellular response to microbial infection. Thus, we identified the specific expression profile of microRNAs in human dendritic cells, using an in vitro model of stimulation and activation by agonists of different Toll-like Receptors (TLRs): R848/Resiquimod, ligand of TLR7/8; LPS, ligand of TLR4; and poly(I: C), ligand of TLR3. Analysis of expression profiles identified groups of miRNAs expressed specifically in response to treatments with LPS, R848, or their combination with respect to control dendritic cells. This analysis will help to clarify their possible role in mechanisms of dendritic cells to discriminate pathogens. The non-alcoholic fatty liver disease or NAFLD is an emerging disease characterized by a wide spectrum of liver conditions from simple steatosis, steatohepatitis with or without fibrosis (NASH, non-alcoholic steatohepatitis). The etiopathogenesis of NAFLD is complex and still unclear. To identify possible molecular mechanisms involved in the development of NAFLD, we used an animal model, able to reproduce various aspects of human pathology. In this study we performed the analysis of microRNAs expression profiles in liver tissue of rats subjected to different diets. Our results showed that treatment with different ipercaloric regimens caused a significant increase in body weight and liver, and some metabolic parameters, compared to control animals, as well as different liver damage. The analysis of microRNAs showed the significant downregulation of three microRNAs (miR-122, miR-451 and miR-27a) and the up-regulation of other three microRNAs (miR-200A, miR-429 and miR-200B) in animals treated with ipercaloric diets respect to those with a standard diet. Among the potential targets of these microRNAs we found some molecules involved in the regulation of apoptosis and inflammation, but also intracellular signaling proteins, and lipid and glucose metabolism.

Resumo: O objetivo deste trabalho foi avaliar, in vitro, o grau de rugosidade radicular produzido pelos vários tipos de instrumentos utilizados durante o processo de instrumentação. Foram selecionados 110 dentes humanos extraídos. Estes dentes foram lavados em água corrente e acondicionados em recipientes com formol para serem manipulados todos ao mesmo tempo, por um único operador. Os dentes foram divididos aleatoriamente, em 11 grupos instrumentados com curetas Gracey; lima periodonta1; aparelho ultra-sônico; aparelho sônico; broca diamantada extra-fina, em caneta de alta e baixa-rotação; broca carbide multilaminada de 12 e 30 lâminas, em caneta de alta e baixa rotação; e grupo controle, sem instrumentação. Foram realizadas três leituras paralelas e três perpendiculares ao sentido de instrumentação, em cada amostra antes e depois da instrumentação, utilizando um aparelho rugosímetro. Os dados foram analisados estatisticamente pelos teste '1;" pareado e ANOV A. Os resultados mostraram que o uso de todos os instrumentos produziram aumento de rugosidade na superfície radicular. As pontas diamantada extra-finas utilizadas em alta-rotação produziram o menor aumento de rugosidade em relação ao grupo controle, seguido em ordem crescente de rugosidade pelos seguintes instrumentos: lima periodontal manual, cureta Gracey manual, broca de 30 lâminas em alta-rotação, ponta diamantada em baixa-rotação, broca de 12 lâminas em alta rotação, broca de 30 lâminas em baixa-rotação, broca de 12 lâminas em baixa-rotação, aparelho sônico e aparelho ultra-sônico. Foram também realizadas fotomicrografias das superfícies radiculares, após a instrumentação, em microscopia eletrônica de varredura, com aumento de 150 e 350 vezes, com fins ilustrativos
Abstract: The aim of this study was to evaluate the "in vitro" roughness degree generated by several instruments used for root planning. 110 extracted human teeth were selected. The teeth were washed and stored in formaline, to be manipulated all at the same time by only one person. Then, they were randomly divided into 11 groups and scaled with gracey hand curette, hand periodontal file, ultrasonic scaler, sonic sca1er, fine-grit-diamond bur with high and low-speed headpiece, 12 and 30-1amina bur with high and low-speed hand piece, and control group without instrumentation. Three parallel readings and 3 perpendicular readings were made on the instrumentation direction in the samples before and after instrumentation, with the use of a porfilometer. It was made a statistical data analysis through the doble "t" and ANOV A tests. The results showed that the fine-grit-diamond bur with high-speed hand piece had the lowest values of roughness increase in relation to the control group, followed by the groups: hand periodontal file; hand curette ; 30-lamina bur with high-speed hand piece; fine-grit-diamond bur with low-speed hand piece; 12-lamina bur with high-speed hand piece; 30-lamina bur with low-speed headpiece; 12-lamina bur with low-speed hand piece; sonic scaler and ultrasonic sca1er.
