Abstract:
Honeycomb can be used as a source of antibacterial, this is due to the content of secondary metabolites in the form of flavonoids in the honeycomb which serves as a protective determinant of honey quality. This study aims to isolate, identify and test antibacterial isolates of Indigenous bacteria from Apis mellifera nests against gram-positive bacteria S. aureus and gram-negative bacteria E. coli. This research used descriptive method and the research data were obtained through laboratory experiments. The results of the isolation stage of indigenous bacteria obtained seven (7) isolates AM1, AM2, AM3, AM4, AM5, AM6, AM7 Indigenous bacteria from Apis mellifera bee hives. AM1 and AM4 isolates were similar to the Enterobacter genus, AM2 isolates to Corynebacterium genus, AM3 and AM6 isolates to Paracoccus genus, and AM5 and AM7 isolates to Azotobacter Sp. The seven isolates of Indigenous bacteria have potential as antibacterial and the diameter of the inhibition zone against E. coli bacteria is AM1 (8.88 mm), AM2 (8.65 mm), AM3 (8.03 mm), AM4 (6.41 mm) , AM5 (9.07 mm), AM6 (9.53 mm) and AM7 (9.44 mm) while against S. aureus AM1 (9.08 mm), AM2 (9.23 mm), AM3 (9.15 mm), AM4 (8.70 mm), AM5 (10.44 mm), AM6 (11.56 mm) and AM7 (9.0 mm).

Abstract A monoclonal human anti-soluble schistosomal egg Ag(SEA) antibody (E5) that stimulates anti-Id T cells and is idiotypically represented in pools of immunoaffinity-purified human anti-SEA antibodies from chronic, generally asymptomatic, intestinal (INT) patients (AM1 and AM5) was used to raise several monoclonal anti-Id: 1C2, 1C6, 4A8, 4F9, and 2A7. Cross-inhibition between these anti-Id identified distinct idiotopes on E5. Anti-SEA preparations from schistosomiasis patients (AM1, AM5, and others) were tested for their inhibition of the E5/monoclonal anti-Id reactions, in competitive ELISA. In either the E5/4A8 or E5/1C6 ELISA system, anti-SEA from INT (AM1 or AM5) or hepatointestinal (HI) (AM7) patients were able to inhibit these reactions. However, anti-SEA antibodies from acute (AM9) or hepatosplenic (HS) (AM3 or AM8) patients did not express Id that were inhibitory in these systems. These results suggest that a relatively high proportion of INT and HI anti-SEA antibodies express a dominant cross-reactive idiotope (CRI) recognized by 1C6/4A8. This CRI is also easily detected in plasmas from individual INT patients. Anti-Id 1C2 reacted strongly with an Id in AM1, AM5, or AM7, but one which also occurred, to a lesser extent, in AM3, AM8, and AM9. Monoclonal anti-Id 4F9 and 2A7 reacted weakly with idiotopes expressed by antibodies from all patients, regardless of the clinical form of their infection. These observations indicate that anti-SEA antibodies from INT and HI, but not acute or HS patients express dominant, CRI that are identified by 1C6, 4A8, or 1C2 and are also expressed on the INT-derived anti-SEA mAb E5.

Introdução: Instituída pelo Ministério da Saúde (MS), a Avaliação para Melhoria da Qualidade (AMQ) é um instrumento de avaliação e aperfeiçoamento das equipes da Estratégia de Saúde da Família (ESF). A Secretaria Municipal da Saúde de Curitiba conta, desde 2002, com o Plano Operativo Anual (POA) que é uma estratégia de gestão voltada para resultados, aprimoramento, planejamento e avaliação. Este instrumento é composto por 81 indicadores. Assim, o município do estudo conta com duas formas de avaliação de qualidade em todas as suas Unidades de Saúde da Família. O objetivo deste trabalho foi analisar as concordâncias e discordâncias entre o POA e o AMQ nos programas da saúde da criança, adolescente, mulher, diabéticos, hipertensos e saúde mental. Metodologia: Participaram do estudo 12 equipes de Saúde da Família. Para realizar a comparação foram levantados os indicadores que estavam presentes tanto no AMQ (instrumento cinco) quanto no POA-2010. Após as comparações foram realizadas porcentagens simples e análise das discordâncias. Resultados: Ao total, 28 indicadores estavam presentes tanto na AMQ como no POA. No geral houve concordância de respostas em 53,6%. Na avaliação da AMQ, 82% das respostas foram consideradas totalmente adequadas para o item avaliado, enquanto na avaliação realizada pelo POA este índice caiu para 46,4%. Conclusão: Observou-se discordância entre os itens comparados da AMQ e do POA. Alguns fatores podem ser responsáveis pela discordância entre eles: a utilização de dados recuperados automaticamente através das ações registradas pela equipe no prontuário eletrônico no POA e a obrigatoriedade da sua realização. Além disso, a realização facultativa da AMQ e a não parametrização das respostas (SIM x NÃO) podem contribuir para a divergência das respostas.

e13570 Background: The treatment of metastatic colorectal cancer has been improved by combining cytotoxic chemotherapy with bevacizumab. Newer anti-angiogenic agents are required to improve survival rates as response rates with the current therapies are modest. The preclinical development of such drugs is time-consuming and new methods are required to test the efficacy of lead drugs which represent the entire tumor micro-environment. Methods: Chemical screens were performed in zebrafish larvae to identify hits that affected intersegmental angiogenesis. Five lead drugs were then tested for cytotoxicity using the crystal violet assay. These drugs were tested using ex vivo colorectal tumor explants to determine their effect on secretion of IL-1β, TNF, IL-6 and VEGF. Human explants from 20 patients (Dukes A-D) were cultured for 72 hours and the levels of the above factors measured by ELISA. Comparisons were made between early (Dukes A&B) and late stages (Dukes C&D). Results: One thousand drugs were screened and 5 were found to reduce intersegmental angiogenesis in zebrafish larvae: AM1, AM2, AM3, AM4 and AM5. The drugs did not affect cell growth levels at 10 µM concentration. From the explant studies: AM1 decreased secretion of VEGF in late stages (p=0.005) and IL-6 in early stages (p=0.009). AM2 decreased VEGF in early (p=0.004) and late stages (p=0.02). AM3 decreased TNF secretion in late stages (p=0.02) and IL-6 secretion in early (p=0.009) and late stages (p<0.0001). AM4 decreased VEGF secretion in early stages (p=0.001), IL-6 in early stages (p<0.001) and late stages (p=0.03) and IL-1β in early stages (p=0.002). AM5 decreased IL-6 secretion in the early stages (p=0.03) and IL-1β in the early stages (p=0.001).Overall, IL-1β secretion was reduced by AM4, AM3 and AM5 (p<0.05). IL-6 secretion was reduced by AM1, AM3 and AM4 (p=0.001). TNF secretion was reduced by AM3 (p=0.02) and VEGF secretion was reduced by AM1, AM2 and AM4 (p<0.005). Conclusions: These studies show these drugs have stage-specific effects in colorectal tumor explants and may have the potential as new anti-angiogenic agents in colorectal cancer.

O objetivo do trabalho é analisar a validade de conteúdo dos instrumentos de autoavaliação que compõem a Avaliação para Melhoria da Qualidade da Estratégia Saúde da Família (AMQ). Trata-se de um estudo ecológico, retrospectivo, de análise de dados secundários, com utilização dos dados disponíveis de 46 dos 64 municípios que aderiram ao processo de AMQ no Espírito Santo até Dezembro de 2007, totalizando 241 equipes de saúde da família. Foram calculadas as médias de percentual de respostas "sim" que as equipes de saúde davam em cada padrão de qualidade do AMQ. Encontrou-se uma não linearidade entre a complexidade de implementação dos processos de trabalho sugeridos pelo AMQ a partir do elenco de padrões em cada estágio de qualidade e a autoavaliação das equipes de saúde da família dos municípios estudados. A identificação da não linearidade da relação entre os estágios de qualidade do AMQ e a complexidade de implantação dos processos de trabalho preconizados pelos padrões de qualidade evidencia a necessidade de se reorganizar os cadernos de autoavaliação, de forma a encontrar uma correlação mais consistente entre dificuldade de implantação dos processos de trabalho no cotidiano da ESF e estágios de qualidade.

The teleost adrenomedullin (AM) family consists of three groups, AM1/AM4, AM2/AM3, and AM5. In the present study, we examined the effects of homologous AM1, AM2, and AM5 on drinking and renal function after peripheral or central administration in conscious freshwater eels. AM2 and AM5, but not AM1, exhibited dose-dependent (0.01–1 nmol/kg) dipsogenic and antidiuretic effects after intra-arterial bolus injection. The antidiuretic effect was significantly correlated with the degree of associated hypotension. To avoid the potential indirect osmoregulatory effects of AM-induced hypotension, infusion of AMs was also performed at nondepressor doses. Drinking was enhanced dose-dependently at 0.1–3 pmol·kg−1·min−1 of AM2 and AM5, matching the potency and efficacy of angiotensin II (ANG II), the most potent dipsogenic hormone known thus far. AM2 and AM5 infusion also induced mild antidiuresis, while AM1 caused antinatriuresis. Additionally, AMs were injected into the third and fourth ventricles of conscious eels to assess their site of dipsogenic action. However, none of the AMs at 0.05–0.5 nmol induced drinking, while ANG II was highly dipsogenic. AM2 and ANG II injected into the third ventricle increased arterial pressure while AM5 decreased it in a dose-dependent manner, and both AM2 and AM5 decreased blood pressure when injected into the fourth ventricle. These data suggest that circulating AM2 and AM5 act on a target site in the brain that lacks the blood-brain barrier. Collectively, the present study showed that AM2 and AM5 are potent osmoregulatory hormones in the eel, and their actions imply involvement in seawater adaptation of this euryhaline species.

This study aimed to isolate, molecular identify, and evaluate the antimicrobial activity of the three commercial types of household detergents; Persil (P), Arial (A), and Peros (Pe) in both forms of gel liquid and (Tate ) powder and antibiotics susceptibility test to assessed against isolates from used washing machines in Mosul city/ Iraq. A total of 46 bacterial isolates were isolated. Five isolates were designated as Kh-Am1, Kh-Am4 from the Drum part, Kh-Am3, Kh-Am6 from the Rubber door seal part and Kh-Am5 from Drain dictum; these isolates were selected for molecular identification by 16S rRNA. The 16S rRNA gene of Kh-Am1, Kh-Am3, Kh-Am4, Kh-Am5 and Kh-Am6. The disc agar diffusion method was adopted for each selected isolate using stock and diluted concentrations. After incubation at 37ₒC for 24 hours, inhibition of Aerea was recorded. The results showed that the powder and gel form detergent stock solution has more inhibitory efficacy for the assayed isolates than dilutions. Susceptibility to antibiotics assay revealed that almost all the selected isolates were resistant. Even though infectious strains were not found, opportunistic pathogens were detected. However, because many bacteria have been detected in wash water, washing items at home with it may result in microbial contamination. Ações de planejamento em saúde adaptadas ao contexto local são fundamentais no cotidiano da Estratégia Saúde da Família (ESF). Este trabalho visa avaliar as práticas de planejamento empreendidas pelas equipes de saúde da família do Espírito Santo, Brasil. Trata-se de um estudo ecológico com análise dos dados secundários de 241 equipes de saúde de 46 municípios deste estado que utilizam a Avaliação para Melhoria da Qualidade da Estratégia Saúde da Família (AMQ-ESF). Foram estabelecidos critérios de avaliação das respostas dos profissionais de saúde aos padrões de qualidade dos instrumentos do AMQ-ESF e elencados os indicadores que versam sobre planejamento em saúde. As respostas foram estratificadas por porte populacional do município, tempo de implantação e cobertura populacional da ESF. Encontraram-se no geral baixas médias de adequação dos processos de trabalho das equipes aos padrões de qualidade do AMQ, sendo que não houve influência das variáveis de estratificação nas práticas de planejamento local. Ações específicas neste campo são necessárias para a qualificação do trabalho da ESF.

ABSTRACT N-acetylanthranilate amidase (Amq), a 32.8-kDa monomeric amide hydrolase, is involved in quinaldine degradation by Arthrobacter nitroguajacolicus Rü61a. Sequence analysis and secondary structure predictions indicated that Amq is related to carboxylesterases and belongs to the α/β-hydrolase-fold superfamily of enzymes; inactivation of (His6-tagged) Amq by phenylmethanesulfonyl fluoride and diethyl pyrocarbonate and replacement of conserved residues suggested a catalytic triad consisting of S155, E235, and H266. Amq is most active towards aryl-acetylamides and aryl-acetylesters. Remarkably, its preference for ring-substituted analogues was different for amides and esters. Among the esters tested, phenylacetate was hydrolyzed with highest catalytic efficiency (k cat/Km = 208 mM−1 s−1), while among the aryl-acetylamides, o-carboxy- or o-nitro-substituted analogues were preferred over p-substituted or unsubstituted compounds. Hydrolysis by His6Amq of primary amides, lactams, N-acetylated amino acids, azocoll, tributyrin, and the acylanilide and urethane pesticides propachlor, propham, carbaryl, and isocarb was not observed; propanil was hydrolyzed with 1% N-acetylanthranilate amidase activity. The catalytic properties of the cysteine-deficient variant His6AmqC22A/C63A markedly differed from those of His6Amq. The replacements effected some changes in Km s of the enzyme and increased k cats for most aryl-acetylesters and some aryl-acetylamides by factors of about three to eight while decreasing k cat for the formyl analogue N-formylanthranilate by several orders of magnitude. Circular dichroism studies indicated that the cysteine-to-alanine replacements resulted in significant change of the overall fold, especially an increase in α-helicity of the cysteine-deficient protein. The conformational changes may also affect the active site and may account for the observed changes in kinetic properties.

A proposta Avaliação para Melhoria da Qualidade da Estratégia Saúde da Família (AMQ) foi desenvolvida para oferecer, aos gestores municipais, ferramentas de avaliação e gestão da qualidade da Estratégia Saúde da Família (ESF). No presente estudo, apresenta-se uma perspectiva de avaliação mediante a AMQ, focada na percepção dos profissionais de saúde. Foram realizadas entrevistas com os profissionais de nível superior das três categorias mais envolvidas com a ESF, utilizando-se os instrumentos da AMQ referentes à avaliação de equipes, as quais foram comparadas em relação à qualificação profissional e ao tempo de implantação. As médias dos escores das equipes foram comparadas pelo teste 't' de Student, assumindo-se um nível de significância de 5%. Registrou-se uma percepção positiva das equipes, exceto para as subdimensões 'participação comunitária e controle social' e 'saúde do adolescente'. Equipes qualificadas mediante residência na área de saúde da família mostraram melhor desempenho. Os autores recomendam a ampla utilização do instrumento como forma de divulgação e consolidação da proposta de avaliação e monitoramento da qualidade na ESF e reforçam o papel da residência como medida privilegiada para a qualificação dos profissionais de saúde na atenção primária

Abstract:
Adrenomedullin (AM), known as a multifunctional hormone in mammals, forms a unique family of five paralogous peptides in teleost fish. To examine their cardiovascular effects using homologous AMs in eels, we isolated cDNAs encoding four eel AMs, and named AM1 (ortholog of mammalian AM), AM2, AM3 (paralog of AM2 generated only in teleost lineage), and AM5 according to the known teleost AM sequences. Unlike pufferfish, not only AM1 but AM2/3 and AM5 were expressed ubiquitously in various eel tissues. Synthetic mature AM1, AM2, and AM5 exhibited vasodepressor effects after intra-arterial injections, and the effects were more potent at dorsal aorta than at ventral aorta. This indicates that AMs preferentially act on peripheral resistance vessels rather than on branchial arterioles. The potency was in the order of AM2 = AM5 ≫ AM1 in both freshwater (FW) and seawater (SW) eels, which is different from the result of mammals in which AM1 is as potent as, or more potent than, AM2 when injected peripherally. The minimum effective dose of AM2 and AM5 in eels was 1/10 that of AM1 in mammals. The hypotension reached 50% at 1.0 nmol/kg of AM2 and AM5, which is much greater than atrial natriuretic peptide (20%), another potent vasodepressor hormone. Even with such hypotension, AMs did not change heart rate in eels. In addition, AM1 increased blood pressure at ventral aorta and dorsal aorta immediately after an initial hypotension at 5.0 nmol/kg, but not with AM2 and AM5. These data strongly suggest that specific receptors for AM2 and AM5 exist in eels, which differ from the AM1 receptors identified in mammals.

Sibirioside A and angoroside C are two important phenylpropanoid glycosides of the traditional Chinese medicine Scrophulariae Radix. High performance liquid chromatography, coupled with an ion trap time-of-flight multistage mass spectrometry equipped with electrospray ionization source (HPLC-ESI-IT-TOF-MSn), was applied to the profile and we identified the metabolites of sibirioside A and angoroside C in vivo in rats. A total of four metabolites of sibirioside A were identified: SM1, SM2 and SM3 which were known as new compounds. A total of 25 metabolites were detected for angoroside C: AM4, AM5, AM6, AM7, AM16, AM17, AM20, AM21, AM22, AM23 and AM25 which were identified to be new compounds. The main metabolic reactions were hydrolysis, reduction, hydroxylation, methylation, sulfation, and gluconylation. The prototype of sibirioside A was widely distributed in tissues found in the heart, liver, spleen, lung, kidney, stomach and small intestine of rats, and mainly distributed in the stomach, small intestine, kidney and liver. But for angoroside C, nothing was found in the viscera except the stomach and small intestine. The metabolites of sibirioside A were mainly eliminated from feces, while it was urine for the metabolites of angoroside C. Furthermore, 19 metabolites were likely to have bioactivities based on the ‘PharmMapper’ analysis, which roughly matched the known pharmacological activities of Scrophulariae Radix (SR) and the prototypes. One of the main pharmacological activities of SR in traditional Chinese medicine is anti-diabetes, and the predicted results showed that SM1, SM2, SM3, AM2, AM4, AM5, AM6, AM9, AM10, AM11, AM12, AM13, AM15, AM18, AM19, AM24, and AM25 might be used to cure diabetes. These findings provide a reference for studying the metabolism, distribution and pharmacological actions of phenylpropanoid glycosides in vivo.

The “About Me” Questionnaire (AMQ) has been used to measure components of social identity, academic self-concept and self-worth in children and adolescents in the United Kingdom and abroad. Studies have reported simple reliability statistics but a comprehensive assessment of the scale’s psychometric properties has not been conducted. Confirmatory factor analysis, using a sample of 5,082 children aged 6 to 18 years from combined datasets of five cross-sectional research studies, was used to establish the psychometric soundness of the 29-item AMQ. Analysis revealed generally adequate reliability with the seven-factor structure confirmed in a replication sample. Results provide evidence of adequate psychometric properties, optimized with the omission of reverse-coded item and selected items, suggesting it is suitable for assessing social identity and academic self-concept of children and adolescents in applied settings. Tests for measurement invariance showed that the assessment of parallel constructs was strongly supported across males and females and partially supported across primary and secondary school–age groups.

Objetivou-se verificar a adequação de equipes de saúde da família aos padrões de qualidade do Ministério da Saúde e sua correlação com ações de atenção à saúde, segundo os profissionais de saúde. Foi utilizado o instrumento de Avaliação para Melhoria da Qualidade da Estratégia Saúde da Família (AMQ) em entrevistas conjuntas com médicos, enfermeiros e cirurgiões-dentistas. Foram definidos escores para cada uma das subdimensões avaliadas, segundo o número de conformidades aos padrões definidos no AMQ. Os escores mais baixos da dimensão de consolidação do modelo foram referentes à participação comunitária e controle social. Para a atenção à saúde, os escores mais baixos foram relativos à saúde do adolescente. As subdimensões "acolhimento e humanização" e "promoção da saúde" mostraram-se estatisticamente associados com a atenção à saúde da criança, do adulto e do idoso. A subdimensão "organização do trabalho" mostrou-se estatisticamente associada com a atenção à saúde da criança e do idoso. Existe, portanto, correlação positiva entre consolidação do modelo e melhoria da qualidade.

Atualmente a qualidade da atenção apresenta-se como um dos desafios ao Sistema Único de Saúde, devendo ser compreendida à luz dos princípios da integralidade, universalidade, equidade e participação social. Visando a melhoria da qualidade da atenção, o Ministério da Saúde desenvolveu a proposta “Avaliação para Melhoria da Qualidade da Estratégia Saúde da Família”, instrumento específico que busca o aprimoramento gerencial, técnico e científico. A Secretaria Municipal da Saúde de Curitiba aderiu de maneira voluntária e participativa a esta proposta, situando a avaliação como instrumento permanente para tomada de decisões e a qualidade como um atributo fundamental a ser alcançado no SUS-Curitiba, ambas apropriadas pelos profissionais envolvidos na Estratégia. Este estudo tem por objetivo avaliar a qualidade autorreferida da ESF no município de Curitiba por meio do AMQ, identificando o estágio de qualidade alcançado pelos atores da SF no município. Para tal utilizou-se um estudo transversal e descritivo. No terceiro momento avaliativo, Curitiba contava com 49 Unidades Básicas de Saúde com Estratégia de Saúde da Família respondentes (instrumento 3), 153 equipes de saúde respondentes (instrumentos 4 e 5), um gestor municipal (instrumento 1) e uma Coordenação Municipal da Estratégia Saúde da Família (instrumento 2). Os resultados dos estágios de qualidade encontrados foram: qualidade Elementar (93,98%), qualidade em Desenvolvimento (90,79%), qualidade Consolidada (89,71%), qualidade Boa (88,25%) e qualidade Avançada (78,23%). Na perspectiva de melhoria contínua da qualidade, observou-se um incremento processual, com maior percentual em elementos dos padrões do estágio E, que abordam as condições elementares de estrutura e infraestrutura (qualidade E). A grande positividade das avaliações apresentadas, superior a 65% em todas as avaliações, indica o alto padrão de qualidade encontrado no município em todos os estágios de qualidade da AMQ. A forma de resposta dicotômica da AMQ não possibilita ver a evolução do trabalho realizado, devendo ser revista.

piRNAs play pivotal roles in maintaining genome stability, regulating gene expression, and modulating development and immunity. However, there are few piRNA-associated studies on honey-bees, and the regulatory role of piRNAs in the development of bee guts is largely unknown. Here, the differential expression pattern of piRNAs during the developmental process of the European honey-bee (Apis mellifera) larval guts was analyzed, followed by investigation of the regulatory network and the potential function of differentially expressed piRNAs (DEpiRNAs) in regulating gut development. A total of 843 piRNAs were identified in the larval guts of A. mellifera; among these, 764 piRNAs were shared by 4- (Am4 group), 5- (Am5 group), and 6-day-old (Am6 group) larval guts, while 11, 67, and one, respectively, were unique. The first base of piRNAs in each group had a cytosine (C) bias. Additionally, 61 up-regulated and 17 down-regulated piRNAs were identified in the “Am4 vs. Am5” comparison group, further targeting 9, 983 genes, which were involved in 50 GO terms and 142 pathways, while two up-regulated and five down-regulated piRNAs were detected in the “Am5 vs. Am6” comparison group, further targeting 1, 936 genes, which were engaged in 41 functional terms and 101 pathways. piR-ame-742536 and piR-ame-856650 in the “Am4 vs. Am5” comparison group as well as piR-ame-592661 and piR-ame-31653 in the “Am5 vs. Am6” comparison group were found to link to the highest number of targets. Further analysis indicated that targets of DEpiRNAs in these two comparison groups putatively regulate seven development-associated signaling pathways, seven immune-associated pathways, and three energy metabolism pathways. Moreover, the expression trends of five randomly selected DEpiRNAs were verified based on stem-loop RT-PCR and RT-qPCR. These results were suggestive of the overall alteration of piRNAs during the larval developmental process and demonstrated that DEpiRNAs potentially modulate development-, immune-, and energy metabolism-associated pathways by regulating the expression of corresponding genes via target binding, further affecting the development of A. mellifera larval guts. Our data offer a novel insight into the development of bee larval guts and lay a basis for clarifying the underlying mechanisms.

Com o surgimento do Projeto Avaliação para a Melhoria da Qualidade da Estratégia Saúde da Família (AMQ . MS), inova-se na auto-avaliação municipal em saúde. O método escolhido foi o de estudo de caso, pois se tornou necessário examinar um conjunto de variáveis indispensáveis à compreensão de um fenômeno complexo. Foram selecionados municípios para utilização dos instrumentos/aplicativos do AMQ-MS, assim como para avaliação com instrumentos já validados em pesquisa avaliativa anterior de âmbito regional, coordenada pelo autor. Três dimensões de análise . político/institucional, organização da atenção e cuidado integral . estão sendo trabalhadas e articuladas na avaliação do processo de implantação da ESF, com forte contextualização dos efeitos. Uma das dimensões da análise dos resultados, no sentido amplo, é a verificação do desempenho do sistema . o cumprimento de metas e resultados. Atualmente, trabalha-se com 23 indicadores na análise do desempenho dos sistemas de saúde municipais. Com os procedimentos analíticos, reduziu-se a dimensionalidade desses bancos, resultando em uma síntese da robustez e da capacidade explicativa desses indicadores. Ao final, são ressaltados os aspectos exitosos e os potencializadores no desenvolvimento dessa experiência.

This study examined the degree to which time perspectives were associated with the processes and content of autobiographical memory. A sample of 230 young adults (118 women) completed the Zimbardo Time Perspective Inventory (ZTPI; Zimbardo and Boyd, 1999 ), the Big Five Inventory (BFI; John and Srivastava, 1999 ), and responded in writing to seven memory prompts as well as to items from the Autobiographical Memory Questionnaire (AMQ; Rubin et al., 2003 ). The Past Positive and Future subscales of the ZTPI predicted many aspects of memory even when controlling for BFI traits. There were gender differences in time perspectives and gender moderated a number of the correlations between time perspectives and autobiographical memory.

Abstract In an effort to analyze both IgM rheumatoid factor (RF) repertoire and regulation of RF production in humans, we developed a panel of four mouse monoclonal antibodies (mAb) defining distinct K light chain-associated idiotopes (id) of a human monoclonal IgM RF (Alt). These mAb (A75, AM1, AM2, AM3) had equivalent reactivities with the immunizing RF during classic inhibition of antigen-binding assays. These anti-id reagents were reacting to neither other tested monoclonal IgM RF nor normal polyclonal IgM. It was possible to distinguish the id defined by the mAb from the results of four sets of experiments: dissociation of Alt RF heavy (H) and light (L) chains showed that A75, AM1, and AM2 reacted to id located on the L chain, whereas AM3 defined a conformational RF id; recombination experiments of H and L chains showed that A75 and AM2 reacted well with both homologous (Alt H + Alt L) and heterologous (Alt L + unrelated H) recombinants, whereas AM1 reacted better with the homologous recombinant than with the heterologous one; the relative affinities of the mAb were drawn from their ability to shift already bound labeled Alt RF from solid phase IgG; and radiolabeling of two mAb (A75 and AM3) and experiments of inhibition of id binding with cold anti-id and cold anti-CK showed that A75 recognized a proximal id (close to the K constant region), whereas AM3 defined a more distal id, AM2 and AM1 being located between A75- and AM3-defined sites. Abstract Immunoaffinity-purified antibodies against Schistosoma mansoni soluble egg Ag (SEA) from infected patients' sera differ idiotypically according to the donor's clinical form of the disease. The Id differ both by their ability to stimulate proliferation of anti-Id T cells and their recognition by anti-Id-specific sera. Also, mice infected with S. mansoni develop anti-Id T and B cell responses against mouse anti-SEA antibodies. We now show that anti-SEA antibodies from serum pools of chronic, but asymptomatic patients (AM1 and AM5) stimulate proliferation of spleen cells from mice infected with S. mansoni. However, AM8, anti-SEA antibodies from hepatosplenic patients, did not stimulate these spleen cells. The murine responses directly parallel patient studies where AM1 and AM5 Id-stimulated human PBMC, but AM8 Id did not. In competitive ELISA, using AM1 or AM-5-specific rabbit antisera or human anti-SEA mAb E5-specific rabbit antiserum, sera from mice infected for 8 and 16 wk (but not from uninfected mice) compete with AM1, AM5, or E5. These sera do not compete in the AM8/anti-AM8 competitive ELISA. Sera from 8-wk-infected mice inhibit more against AM1, AM5, and E5 than do sera from later infections, and anti-SEA immunoaffinity-purified antibodies from 8-wk-infected mice stimulate spleen cells from infected mice more than anti-SEA antibodies from sera of mice late in infection. However, spleen cells from more chronically infected mice are more responsive to either the murine or human anti-SEA antibody preparations than cells from mice with earlier infections. Both the ELISA data and lymphocyte responses indicate that anti-SEA antibodies from mice infected with S. mansoni for 8 wk bear Id cross-reactive with those expressed on anti-SEA antibodies from humans with chronic, asymptomatic schistosomiasis, but not those from hepatosplenic patients.

Penelitian ini bertujuan untuk mengetahui pengaruh biomassa Azolla microphylla dan mendapatkan biomassa terbaik untuk meningkatkan kelimpahan zooplankton pada media pemeliharaan ikan Patin Siam (Pangasianodon hypophthalmus) menggunakan air gambut. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) 1 faktor dengan 4 taraf perlakuan dan 3 ulangan. Perlakuan dalam penelitian ini adalah A. microphylla dengan biomassa yang berbeda yaitu AM0 (kontrol), AM1 (20 g m-2), AM2 (40 g m-2) dan AM3 (60 g m-2). Hasil penelitian menunjukkan bahwa perlakuan terbaik adalah perlakuan AM3 (60 g m-2) yang menghasilkan kelimpahan zooplankton rata-rata 1503 ind L-1 dan mampu mendukung kehidupan Patin Siam (P. hypophthalmus) selama 28 hari pemeliharaan dengan menghasilkan pertumbuhan bobot mutlak (2.41 g ekor-1). Selama penelitian, 4 spesies zooplankton ditemukan dari kelas Protozoa dan Rotifera. Kelas Protozoa terdiri dari 1 spesies yaitu Pleodarina sp., sedangkan dari kelas Rotifera terdapat 3 spesies yaitu Brachionus bidentata, Monostyla sp. dan Philodina roseola.

The aim of the study was to examine effects of the Mindfulness-Acceptance-Commitment (MAC) programme on sport-specific dispositional mindfulness, sport anxiety, and experiential acceptance, compared to an inactive control group, in a population of Martial Arts (MA) athletes. Twenty-three MA athletes were subjected to the MAC intervention, and 22 MA athletes comprised the control group. To analyse potential differences in the outcome variables between the two groups, Bayesian repeated measures analysis of variance (RM-ANOVA) was used. Findings showed that the MAC-group had greater increases over time in the sport-specific mindfulness subscales awareness and acceptance and acceptance, compared to the control group. In contrast, no interaction effects were found for present moment attention (AMQ), experiential acceptance (BEAQ), and sport anxiety (SAS-2). Overall, the present study adds further information and understanding about the effectiveness and implementation of the MAC programme.

The aim of the present study was to compare tolerance to salinity and ion accumulation potential of Armeria maritima subsp. elongata. Three accessions (AM1 and AM2, both from Latvia, and AM3 from Sweden) from relatively dry sandy soil habitats in the Baltic Sea region were selected and compared using both in vitro cultivated shoot explants and long-term soil-cultivated plants at flowering stage. Growth of root non-forming explants treated with increasing concentrations of NaCl was significantly inhibited starting from 110 mmol L–1, and the rate of shoot formation was even more sensitive. Significant differences in morphology and responses to salinity were found between different accessions. For soil-grown plants, biomass accumulation in above-ground parts was relatively little affected by salinity in AM1 and AM2 in comparison to that in AM3. Differences in ion accumulation were evident between the accessions as well as in respect to cultivation system used. Maximum accumulation capacity for Na+ was up to 2.5 mol kg–1 both in shoot explant tissues and in old leaves of soil-grown plants treated with NaCl, but that for K+ reached 4.0 mol kg–1 in old leaves of soil-grown plants treated with KCl. Non-ionic component of osmotic value was relatively high in old leaves and significantly increased under NaCl treatment, especially for AM2 and AM3 plants at moderate salinity, but in AM1 only at high salinity. It can be concluded that a wide salinity tolerance exists within A. maritima accessions from dry sandy soil habitats, associated with the ability to accumulate surplus ions both in salt glands and old leaves.

Quinolinium compounds have been used as Cl-sensitive fluorescent indicators in cells and cell-free membrane fractions. To improve Cl sensitivity and for conjugation via nucleophilic reaction, the compounds 6-methoxy-N-(n-aminoalkyl)quinolinium bromide hydrochloride (AAQ) with alkyl chain lengths (n) of 2 (AEQ), 3 (APQ), and 4 (ABQ) were synthesized. AAQ was water soluble, fluorescent, and quenched by Cl. The Stern-Volmer constants (KCl) for quenching of protonated AEQ, APQ and ABQ by Cl were 354, 322, and 272 M-1, respectively, higher than KCl for 6-methoxy-N-(3-sulfopropyl)quinolinium (SPQ; 118 M-1). To eliminate pH-dependent fluorescence, 6-methoxy-N-(3-trimethylammoniumpropyl)quinolinium dibromide (TMAPQ) was synthesized (KCl, 310 M-1). To red shift fluorescence excitation and emission spectra, 6-phenyl-N-(3-trimethylammoniumpropyl) quinolinium dibromide (phenyl-TMAPQ) (emission 475 nm) and N-(3-trimethylammoniumpropyl)phenanthridinium dibromide (TMAPP) (excitation 380 nm) were synthesized. AEQ and ABQ were conjugated with neutral dextran activated by cyanogen bromide to give indicator-to-dextran mole ratios of 5 to 20. KCl values at pH 7.4 were 132 (AEQ-dextran) and 237 M-1 (ABQ-dextran). To construct a single molecule with Cl-sensitive and insensitive moieties, the bichromophores 6-methoxy-N-(n-dansylsulfonamidoalkyl)quinolinium with alkyl chains of two and four were synthesized. The new Cl-sensitive indicators were used for measurement of intracellular Cl activity and for the labeling of endocytic vesicles in 3T3 fibroblasts and T84 cells. Circular RNAs (circRNAs) are a class of novel non-coding RNAs (ncRNAs) that play essential roles in the development and growth of vertebrates through multiple manners. However, the mechanism by which circRNAs modulate the honey bee gut development is currently poorly understood. Utilizing the transcriptome data we obtained earlier, the highly expressed circRNAs in the Apis mellifera worker 4-, 5-, and 6-day-old larval guts were analyzed, which was followed by an in-depth investigation of the expression pattern of circRNAs during the process of larval guts development and the potential regulatory roles of differentially expressed circRNAs (DEcircRNAs). In total, 1728 expressed circRNAs were detected in the A. mellifera larval guts. Among the most highly expressed 10 circRNAs, seven (novel_circ_000069, novel_circ_000027, novel_circ_000438, etc.) were shared by the 4-, 5-, and 6-day-old larval guts. In addition, 21 (46) up-regulated and 22 (27) down-regulated circRNAs were, respectively, screened in the Am4 vs. Am5 (Am5 vs. Am6) comparison groups. Additionally, nine DEcircRNAs, such as novel_circ_000340, novel_circ_000758 and novel_circ_001116, were shared by these two comparison groups. These DEcircRNAs were predicted to be transcribed from 14 and 29 parental genes; these were respectively annotated to 15 and 22 GO terms such as biological regulation and catalytic activity as well as 16 and 21 KEGG pathways such as dorsoventral axis formation and apoptosis. Moreover, a complicated competing endogenous RNA (ceRNA) network was observed; novel_circ_000838 in the Am4 vs. Am5 comparison group potentially targeted ame-miR-6000a-3p, further targeting 518 mRNAs engaged in several developmental signaling pathways (e.g., TGF-beta, hedgehog, and wnt signaling pathway) and immune pathways (e.g., phagosome, lysosome, and MAPK signaling pathway). The results demonstrated that the novel_circ_000838-ame-miR-6000a-3p axis may plays a critical regulatory part in the larval gut development and immunity. Furthermore, back-splicing sites of six randomly selected DEcircRNAs were amplified and verified by PCR; an RT-qPCR assay of these six DEcircRNAs confirmed the reliability of the used high-throughput sequencing data. Our findings provide a novel insight into the honey bee gut development and pave a way for illustration of the circRNA-modulated developmental mechanisms underlying the A. mellifera worker larval guts.

The purpose of this study was to determine the macroscopic and microscopic characteristics of endophytic bacteria that live in the root and leaf tissue of several wet habitat trees, such as: kayu marsegu (Anthocephalus cadamba), ketapang hutan (Terminalia copelandii) and kayu burung (Elaeocarpus ganitrus). This study shows that the results of microscopic identification of 20 samples of bacterial isolates, there are only 11 samples of endophytic bacteria that live in the root and leaf tissue of tree samples. Furthermore, based on microscopic identification with the Gram Staining Method, it was found that there were 17 samples of endophytic bacterial isolates categorized into gram-negative (-), namely: DM1, DM3, DM4, AM1, AM2, AM3, AM4, DKH2, DKH3, DKH4, AKH1, AKH2, AKB1, AKB2, AKB3, DKB3, and DKB4. While 3 other bacterial isolate samples were classified into gram-positive (+), namely: AKH3, AKH4, and AKB4. Based on observations with a microscope with 1000 times the magnification size, all bacteria isolates were categorized as a rounded bacterial group, and therefore they are classified as genera Monococcus, Staphylococcus, and Streptococcus. The study also found that all three genera of bacteria have the ability to live in root and leaf tissue, even though the tree habitat is in a long-wet condition. Keywords: endophytic bacteria, isolation, morphology, wet habitat trees ABSTRAK Tujuan dari penelitian ini adalah untuk menentukan karakteristik makroskopis dan mikroskopis dari bakteri endofit yang hidup di akar dan jaringan daun dari beberapa pohon habitat basah, seperti: kayu marsegu (Anthocephalus cadamba), ketapang hutan (Terminalia copelandii) dan kayu burung (Elaeocarpus ganitrus). Penelitian ini menunjukkan bahwa dari hasil identifikasi mikroskopis terhadap 20 sampel isolat bakteri, hanya ada 11 sampel bakteri endofit yang hidup di akar dan jaringan daun sampel pohon. Selanjutnya, berdasarkan identifikasi mikroskopis dengan Metode Pewarnaan Gram, ditemukan bahwa ada 17 sampel isolat bakteri endofit yang dikategorikan menjadi gram‑negatif (-), yaitu: DM1, DM3, DM4, AM1, AM2, AM3, AM4, DKH2, DKH3, DKH4, AKH1, AKH2, AKB1, AKB2, AKB3, DKB3, dan DKB4. Sedangkan 3 sampel isolat bakteri lainnya digolongkan kedalam gram-positif (+), yaitu: AKH3, AKH4, dan AKB4. Berdasarkan pengamatan dengan mikroskop ukuran pembesaran 1000 kali, semua isolat bakteri dikategorikan sebagai kelompok bakteri bulat, dan oleh karena itu mereka diklasifikasikan sebagai genus Monococcus, Staphylococcus, dan Streptococcus. This study aims to determine the distribution of types of love based on Stenberg's Triangle Love Theory in primary adult of Ba'alawi Arab descent couples in Jakarta who experienced an arranged marriage. This study uses a quantitative descriptive method with descriptive statistical analysis. The measuring instrument used is the Triangular of Love Scale (TLS) to measure the type of love and the Arranged Marriage Questionnaire (AMQ) to see attitudes in selecting a partner for marriage. Respondents in this study were 154 people, with the sample characteristics being early adults aged 21-40 years, experiencing arranged marriages, married with a marriage age range of 0-10 years, and the last education was at least high school, and lived in Jakarta. The results of this study indicate that the distribution of the type of love that respondents have is more dominant in non-love (53.9%), consummate love (22.1%), and empty love (11%). In this study, it is understandable that respondents prefer to marry in a way that tends not in an arranged marriage (56.5%).

<em>This work investigated the impact of verbal praise on teachers’ motivation. The study was conducted with 612 teachers in six public primary school in Owerri West Local Government Area of Imo State, Nigeria. Teachers from the schools were assigned into treatment and control groups respectively. On the day of their experiment a Personal Orientation Questionnaire (POQ) was given to teachers as pretest. Teachers were asked to take up four topics in primary science which lasted for four weeks. At the end of each lesson, teachers in the experimental group were encouraged verbally by the head of their schools and the research assistants but those in control group were not encouraged. At the end, Actualization Motivation Questionnaire, (AMQ) was given to the teachers as post-test. The data collected were analyzed with mean scores, standard deviation and ANCOVA. It was found that verbal praise promoted teachers actualization and motivation more than those non-verbal praise. Again,verbal praise enhanced female teachers performance higher than the male teachers. Recommendations were made based on the findings that school heads and government should accompany teachers’ school performance with public verbal praise.</em>

The receptors for the calcitonin gene-related peptide (CGRP)/adrenomedullin (AM) family peptides were characterized in the mefugu Takifugu obscurus, a euryhaline fugu species very close to Takifugu rubripes, which has as many as five adrenomedullin genes (AM1–5). CGRP and AM share a G protein-coupled core receptor called calcitonin receptor-like receptor (CLR), and the specificity of the CLR is determined by the interaction with receptor activity-modifying proteins (RAMPs). Through database mining, three CLRs (CLR1–3) and five RAMPs (RAMP1–5) were identified, and all of them were cloned by RT-PCR and characterized by functional expression in COS7 cells in every possible combination of CLR-RAMP. The following combinations generated cAMP in response to physiological concentrations of CGRP, AM1 (an ortholog of mammalian AM), AM2, and AM5: CLR1-RAMP1/4 (CGRP), CLR1-RAMP2/3/5 (AM1), CLR2-RAMP2 (AM1), CLR1-RAMP3 (AM2), and CLR1-RAMP3 (AM5). Their expressions were found by Northern blot analysis to be tissue specific and salinity dependent. For example, CLR1-RAMP5 and CLR1-RAMP2 are expressed specifically in the gill and kidney, respectively, suggesting their involvement in osmoregulation. Furthermore, relatively high levels of CLRs and RAMPs were found in the spleen and ovary, suggesting roles in the immune and female reproductive systems. Immunohistochemistry revealed that AM receptors of the following types are expressed in the locations, indicated in brackets, of the mefugu gill and kidney: CLR1-RAMP5 (interlamellar vessels), CLR2-RAMP2 (pillar cells), and CLR1-RAMP2 (apical side of renal proximal tubule cells).

A avaliação da qualidade em Saúde parte de parâmetros que vão direcionar as ações ofertadas aos usuários além de subsidiar a construção de novos instrumentos avaliativos. São objetivos deste estudo: verificar o estágio de desenvolvimento de uma Unidade de Saúde da Família (USF) da cidade do Recife, segundo os padrões de qualidade preconizados pelo Ministério da Saúde. A metodologia constou de: estudo descritivo de corte transversal com abordagem quantitativa. Foram aplicados 27 questionários semi-estruturados aos profissionais, residentes e alunos de graduação da USF. As respostas foram confrontadas com os Padrões de Qualidade do AMQ (Avaliação para Melhoria da Qualidade). Os resultados demonstraram que a USF apresenta-se 100% adequada ao nível ‘elementar’ e 12,5% ao nível ‘consolidado’ para Infra-estrutura e Equipamentos; para Insumos, Imuno-biológicos e Medicamentos apresenta variação de 50% a 100% de adequação. Para Organização do Trabalho 76% adequado ao ‘elementar’ e ‘avançado’. O acolhimento, a utilização do cronograma e a participação dos Agentes Comunitários no planejamento das ações foram os aspectos positivos apontados. Conclui-se que uma avaliação participativa com enfoque na qualidade, refletindo sobre o estágio de desenvolvimento dos serviços oferecidos pela Estratégia Saúde da Família, envolvendo os estudantes contribui para o aprimoramento dos serviços e do processo ensino-aprendizado.

ResumoObjetivo: verificar a qualidade da atenção à saúde da criança pelas equipes da Estratégia Saúde da Família (ESF) em Capital do Nordeste do Brasil. Método: trata-se de um estudo transversal, descritivo, com abordagem quantitativa, realizado durante o ano de 2010, por meio de inquérito com os profissionais médicos e enfermeiros de 66 equipes da ESF de Maceió-AL, estratificadas por tempo de funcionamento em anos. Utilizou-se parte do instrumento de auto-avaliação denominado Avaliação para Melhoria da Qualidade da Estratégia Saúde da Família (AMQ) do Ministério da Saúde, especificamente as questões referentes às ações e atividades realizadas no campo de atenção à “saúde da criança”, dentro da dimensão “atenção à saúde”. Resultados: os padrões elementares de qualidade foram encontrados em 84,1% das ações realizadas. Os padrões considerados avançados apresentaram o menor percentual de conformidade entre todas as equipes (47,7%). Quanto ao tempo de implantação da ESF, as equipes com menos de quatro anos tenderam a relatar melhores padrões de qualidade. Conclusões: os achados sugerem que as ações de saúde da criança estão sendo realizadas em melhor conformidade nas equipes com menos tempo de implantação, e que a grande maioria das equipes ainda estão atendendo a padrões considerados menos avançados em termos de qualidade.

Abstract:
This study aimed to investigate the effect of young adult literature (YAL) on the morals of students in high school. This research is an adapted form of quasi-experimental cum survey design. A sample of one hundred and forty (140) students was randomly selected through the simple random sampling (SRS) technique from the target population. The Attitude to Morals Questionnaire (AMQ) instrument was utilized for data collection. The reliability and validity of the research instrument was assessed using Cronbach Alpha with result of 0.91. Descriptive analysis and two independent sample t-tests were applied to the data and all test were computed at the 0.05 levels of significance. The findings of this study revealed that young adult literature significantly impact on the morals of the high school students. It was also discovered that honesty and respect for elders varies significantly in both the experimental and control groups. More so, the result showed that tolerance of male does not vary significantly with that of the female students in the experimental group. Schools should equip their libraries with sufficient novels on young adult literatures, the teachers should dwell more on the plot, theme and characters in the novels. Students should be guided and emphasis should be made on the need to read young adult literatures, and more time should be allotted on the school timetable for reading of young adult literature.

Wheat (Triticum aestivum L.) is a vital cereal crop for food security in Pakistan. In Zn-deficient soils, its productivity and quality suffer, affecting grain yield, Zn bioavailability, and nutrition, which can lead to malnutrition. Field experiments were conducted using factorial randomized block design at the Agricultural Research Institute (ARI) Tarnab, Peshawar, Pakistan to evaluate the impact of wheat genotypes (G1-TRB-72-311 synthetic hexaploid, G2-TRB-89-348 advanced line, and G3-Pirsabak-19-approved variety), Zn application methods (AM1: no Zn application, AM2: seed priming with 0.5% Zn, AM3: soil application of 10 kg ha−1 Zn, and AM4: foliar application of 0.5% Zn), and the experiment also explored the use of ZSB (BF1: with bacteria, BF0: without bacteria) to cope with Zn deficiency. The study revealed significant impacts on wheat’s Zn content, uptake, and nutrient efficiency, arising from genotypes variance, Zn application approaches, and ZSB. TRB-72-311 synthetic hexaploid genotype with 0.5% foliar Zn and ZSB excelled, enhancing grain (17.8%) and straw Zn (23.1%), increasing total Zn uptake (55.0%), reducing grain phytic acid (11.7%), and boosting Zn-related efficiencies in wheat. These results prompt further discussion regarding the potential implications for agricultural practices. In conclusion, utilizing the TRB-72-311 genotype with 0.5% foliar Zn application and ZSB enhances wheat’s Zn content, uptake, grain quality, and addresses malnutrition.

BackgroundDischarge against medical advice (AMA) occurs infrequently but is associated with poor outcomes. There are limited descriptions of discharges AMA in national cohorts of patients with acute myocardial infarction (AMI). This study aims to evaluate discharge AMA in AMI and how it affects readmissions.MethodsWe conducted a cohort study of patients with AMI in USA in the Nationwide Readmission Database who were admitted between the years 2010 and 2014. Descriptive statistics were presented for variables according to discharge home or AMA. The primary end point was all-cause 30-day unplanned readmissions and their causes.Results2663 019 patients were admitted with AMI of which 10.3% (n=162 070) of 1569 325 patients had an unplanned readmission within 30 days. The crude rate of discharge AMA remained stable between 2010 and 2014 at 1.5%. Discharge AMA was an independent predictor of unplanned all-cause readmissions (OR 2.27 95% CI 2.14 to 2.40); patients who discharged AMA had >twofold increased crude rate of readmission for AMI (30.4% vs 13.4%) and higher crude rate of admissions for neuropsychiatric reasons (3.2% vs 1.3%). After adjustment, discharge AMA was associated with increased odds of readmissions for AMI (OR 3.65 95% CI 3.31 to 4.03, p<0.001). We estimate that there are 1420 excess cases of AMI among patients who discharged AMA.ConclusionsDischarge AMA occurs in 1.5% of the population with AMI and these patients are at higher risk of early readmissions for re-infarction. Abstract:
A simple reversed-phase HPLC method for measuring hepatic levels of acetaminophen- (APAP-) protein adduct following an overdose of APAP was developed. An aliquot of liver homogenate in phosphate-buffered saline pH 7.4 (PBS) was placed on a Nanosep centrifugal device, which was centrifuged to obtain a protein residue. This residue was incubated with a solution ofp-aminobenzoic acid (PABA), the internal standard, and bacterial protease in PBS, transferred to a Nanosep centrifugal device, and centrifuged. A 100 μL portion of the filtrate was analyzed on a YMC-Pack ODS-AMQ C18 column, using 100 mM potassium dihydrogen phosphate-methanol-acetic acid (100 : 0.6 : 0.1) as the mobile phase, a flow rate of 1 mL/min, and photometric detection at 254 nm. PABA and APAP-cystein-S-yl (APAP-Cys) eluted at~14.7 min and 22.7 min, respectively. Method linearity, based on on-column concentrations of APAP-Cys, was observed over the range 0.078–40 μg. Recoveries of APAP-Cys from spiked blank liver homogenates ranged from~83% to 91%. Limits of detection and of quantification of APAP-Cys, based on column concentrations, were 0.06 μg and 0.14 μg, respectively. RSD values for interday and intraday analyses of a blank liver homogenate spiked with APAP-Cyst at three levels were, in all cases, ≤1.0% and <1.5%, respectively. The proposed method was found appropriate for comparing the antidotal properties of N-acetylcysteine and taurine in a rat model of APAP poisoning.

Abstract:
Malaria is a devastating parasitic disease which caused around 216 million cases and 445,000 deaths worldwide in 2016. This might be attributed to a wide spread of drug resistant parasites. The plant Aloe megalacantha is indigenous to Ethiopia where the sap of the leaves is traditionally used for the treatment of malaria. This study was aimed at evaluating the antimalarial effect of leaf latex and isolates obtained from Aloe megalacantha against chloroquine sensitive Plasmodium berghei ANKA strain in Swiss albino mice. Peters’ 4-day suppressive test method was used to test the antimalarial activity of both leaves latex and isolates. Three isolates were obtained using thin layer chromatography and were coded as AM1, AM2, and AM3 in ascending order of their retention factor. After treatment of Plasmodium berghei infected mice with leaf latex of Aloe megalacantha for four days at 100, 200, and 400 mg/kg, it shows 30.3%, 43.4%, and 56.4% suppression of the parasite growth, respectively. 32.3%, 51.3%, and 67.4% chemosuppression after treatment with AM1, 39.8%, 50.6%, and 64.2% chemosuppression after treatment with AM2, and 52.6%, 69.4%, and 79.6% chemosuppression after treatment with AM3 were observed at doses of 100, 200, and 400 mg/kg/day, respectively. The observed parasite suppression of leaves latex and isolates was statistically significant (P<0.05) as compared to negative control. Moreover, both the leaves latex and isolates were also observed to prevent Plasmodium berghei induced body weight loss and hypothermia and increased the survival time of Plasmodium berghei infected mice as compared to the negative control. Hence, the present study supports the traditional claim of the plant for the treatment of malaria.

Chemical composition of the essential oils derived from Atractylodes macrocephala (AMA), Astragalus membranaceus (AME) and AMA-AME herb pair was investigated using gas chromatography-mass spectrometry (GC-MS). Antioxidant activities were evaluated by 1,1-diphenyl-2-picyrlhydrazyl (DPPH) radical-scavenging and Trolox equivalent antioxidant capacity (TEAC) assays. Forty-five, ten and forty-three components were identified in AMA, AME and AMA-AME essential oils, respectively. AMA-AME essential oil exhibited a significantly higher radical scavenging capacity than the theoretical sum of those of the respective herb essential oils ( P < 0.05). Principal component analysis showed that twenty-three components contributed to the scavenging activities against DPPH and ABTS•+ radicals. Moreover, the concentrations of these major components exhibited various increases to some extent when compared with the theoretical sum of the respective herb essential oils. These findings suggest that combination of two or more herbs might be used as a promising source of natural antioxidants in the pharmaceutical and food industries.

Motivation is one of the main factors in learning and it can affect different behavioral aspects of students in academic environment. Academic motivation can be considered as the only factor with direct effect on academic achievement. The present meta-analysis study is an attempt to determine academic motivation and academic achievement based on Vallerand’s scale and Hermans’ questionnaire in students in Iranian universities of medical sciences. The study was carried out as a meta-analysis work on studies published from 2001 to 2018. The relevant studies were searched using keywords “academic motivation” and “academic achievement” in SID, Medline (PubMed), and ScienceDirect. Heterogeneity in the studies was examined using I2 index and data analysis was done in Comprehensive Meta-Analysis software. Out of 16 articles that entered the meta-analysis process; seven articles were based on Vallerand’s scale (AMS) and nine articles were based on Hermans’ questionnaire (AMQ). Mean and standard deviation score of academic motivation based on Vallerand’s scale was 109.9±16.4 and mean and standard deviation score of academic achievement based on Hermans’ questionnaire was 96.83±6.38. The effects of sample size and year of publication were measured based on meta-regression. In terms of sample size, the mean score increased with an increase in sample size in the both Vallerand and Hermans studies. In terms of year of publication, the mean score increased with increase in the year of publication in Vallerand’s studies and decreased in Hermans’ studies (P<0.05). The students of Iranian universities of medical sciences were at good level in terms of academic motivation and academic achievement.

The proboscis extension response (PER) assay revealed the responsiveness of three subspecies of the honeybee Apis mellifera [A. m. jemenitica (AMJ), A. m. carnica (AMC), and A. m. ligustica (AML)] to water and different concentrations (0.00001, 0.0001, 0.001, 0.01, 0.1, 0.5, 1.0, and 1.5 M) of three sugars (fructose, glucose, and sucrose) during the summer and fall seasons. The tested bee subspecies showed significantly different PERs to sugar types across the seasons. The water responsiveness of AMJ, a native bee subspecies, was significantly lower than that of AMC and AML, which showed an equally higher water response in both seasons. During the summer season, AMJ and AMC were equally responsive to each sugar type at all tested concentrations. AML was relatively less responsive to glucose at 0.001, 0.001, 0.01, 0.1, 0.5, and 1.0 M than to fructose and sucrose during the summer season. During the fall season, AMJ was equally responsive to glucose and sucrose at all tested concentrations but showed a significantly different response between fructose and sucrose at 0.001, 0.01, 0.1, 0.5, and 1.0 M concentrations. The PER of AMJ to fructose was lower than that of glucose and sucrose. AMC was equally responsive to all tested sugars at all concentrations, and AML showed a differential response between glucose and sucrose at different concentrations during the fall season. The inter-specific species comparisons revealed that all tested subspecies were equally responsive to fructose at all tested concentrations, and AMJ was more responsive to glucose and sucrose than AMC and AML during both seasons. AMC and AML showed no differences in PER to glucose and sucrose in either season. The AMJ, AMC, and AML nectar and pollen foragers showed no significant differences in PER to glucose and sucrose. The AMC nectar foragers were highly responsive to sucrose than pollen foragers at higher sucrose concentrations (0.1, 0.5, 1.0, and 1.5 M). The AML (nectar forager vs. pollen forgers) showed identical PER to sucrose and glucose but a higher response of nectar foragers to high glucose concentrations (0.5, 1.0, and 1.5 M) than pollen foragers. For water responsiveness, AMJ nectar and pollen foragers showed similar PER to water, whereas AMC and AML pollen foragers were significantly more responsive to water than nectar foragers.

Young adult literature (YAL) is an important and necessary part of moral upbringing in teenagers and young adult lives. The present study seeks to investigate the effect of young adult literature on the morals of students in high school. The study utilized the quasi-experimental cum survey design and a sample of one hundred and forty (140) students of two schools were drawn from the eleven senior secondary schools in Bwari Area Council of Abuja, Nigeria. The intact class was utilized in each of the two schools, and the instruments for data collection was an Attitude to Morals Questionnaire (AMQ). The reliability and validity of the research instrument was assessed using Cronbach Alpha test with result of 0.91. The research questions and hypotheses were tested using mean score and two independent sample t-tests statistical methodology and all tests were computed at the 0.05 levels of significance with Statistical Package for Social Science (SPSS) version 26. The findings of this study revealed that young adult literature significantly impact on the morals of the high school students. It was also discovered that honesty and respect for elders varies significantly in both the experimental and control groups. More so, the result showed that tolerance of male does not vary significantly with that of the female students in the experimental group. Schools should equip their libraries with sufficient novels on young adult literatures, the teachers should dwell more on the plot, theme and characters in the novels. Students should be guided, and emphasis should be made on the need to read young adult literatures, and more time should be allotted on the school timetable for reading of young adult literature.

Com base nisso, o objetivo deste trabalho foi avaliar a produção de mudas de abobrinha irrigadas com águas salinas e cultivadas em diferentes tipos de ambientes e substratos. O delineamento experimental foi o inteiramente casualizado, utilizando esquema de parcelas subsubdivididas, sendo a parcela os três ambientes de cultivo (AM1= pleno sol; AM2= telado vermelho com 50% de sombreamento; AM3= telado preto com 50% de sombreamento), a subparcela os três substratos (SB1= biocarvão + solo; SB2= casca de arroz carbonizado + solo, SB3= esterco bovino + solo) e a subsubparcela as duas águas de irrigação (AI1= 0,8 e AI2= 2,5 dS m-1), com quatro repetições de vinte e cinco sementes. As variáveis analisadas foram: porcentagem de emergência (PE), índice de velocidade de emergência (IVE), tempo médio de emergência (TME), velocidade média de emergência (VME), diâmetro do caule (DC), altura de plântula (AP), comprimento da raiz (CR), massa seca da parte aérea (MSPA), massa seca da raiz (MSR) e massa seca total (MST). O ambiente AM3 apresentou melhores condições para as variáveis: PE, IVE e VME. Já o ambiente AM2 apresentou melhores condições para a variável TME com a utilização dos substratos SB2 e SB3 independente da salinidade da água de irrigação. O substrato SB3 apresentou melhores condições de crescimento para as variáveis: AP, DC, MSPA e MST. O substrato SB2 proporcionou melhores condições para a variável MSR.

Background & Aims: The superior mesenteric artery syndrome (SMAS) is an uncommon syndrome characterized by the compression of the third part of the duodenum between the superior mesenteric artery (SMA) and abdominal aorta with resultant proximal duodenal dilatation. The radiological diagnosis of the SMAS is based on reduced angle and distance between the SMA and aorta in presence of proximal duodenal dilatation. A reduction in these is closely associated with depletion of the mesenteric fat between the vessels. Our primary aim is to establish the relationship, if any, of body mass index (BMI) with the angle and distance between the SMA and abdominal aorta in general population. Materials and Methods: This study was carried out in 200 patients who had undergone contrast enhanced computed tomography for various other complaints. Various parameters such as aortomesenteric distance (AMD) and aortomesenteric angle (AMA) along with the body mass indices were calculated. Pearson correlation coefficients were calculated to establish the relationship between BMI, AMD and AMA. Results: Pearson’s correlation coefficient for BMI and AMD was 0.868, indicating strong positive correlation and 0.577 for BMI and AMA, indicating moderate positive correlation. Furthermore, AMD and AMA also showed positive correlation with Pearson’s correlation coefficient of 0.568. Conclusion: There is significant positive correlation of BMI with AMD and AMA in general population suggesting people with low BMI are at an increased risk of developing SMAS.
