Full textAmphiphilic block copolymers are molecules composed of hydrophilic and hydrophobic segments having the capacity to spontaneously self-assemble into a variety of supramolecular structures like micelles and vesicles. Here, we propose an original way to self-assemble amphiphilic block copolymers into a supported bilayer membrane for defined coating of nanoparticles. The heart of the method rests on a change of the amphiphilicity of the copolymer that can be turned off and on by varying the polarity of the solvent. In this condition, the assembly process can take advantage of specific molecular interactions in both organic solvent and water. The higher gene silencing activity of the copolymer-modified complexes over the complexes alone shows the potential of this new type of nanoconstructs for biological applications, especially for the delivery of therapeutic biomolecules

Full textThis project aims to study and develop a novel principle dedicated to the valorisation of recycled metals, especially palladium. It consists in the direct use of an organic phase arising from a solvent extraction process into organo-catalyzed cross-couplings performed in aqueous micellar phases. The palladium transposition from the organic phase into the aqueous phase is performed thaks to surfactants developed by the CBSA team (C. Pépin & F. Bonneté, IBMM), and which structure has to be optimized to answer to the specifications linked with the hyrometallurgical processes dedicated to palladium recovery from electronic wastes, performed by the LHYS team (D. Bourgeois, ICSM). Following a recent proof of concept involving these both teams from the ChemiSyst LabEx and a third team from the Strasbourg University (F. Bihel), the present project will consist in the rational description of the relationship between the surfactants, easily tunable, the physic-chemical properties resulting from their auto-assembly, and their aptitude to back-extract and stabilize the palladium in the aqueous medium. The fundamental knowledge thus acquired will enable and efficient valorization of the proposed systems

Full textA key challenge in RNAi-based gene therapy is the safe and effective siRNA delivery. Recently, our group has established amphiphilic dendrimers as robust and effective nonviral delivery vectors for siRNA, which combine the beneficial delivery features of both lipid and dendritic polymer vectors while overcoming their shortcomings.With the desire to understand the underlying mechanism of amphiphilic dendrimers for efficient delivery, I performed a structure/activity relationship (SAR) analysis of a series of dendrimers featuring hydrophobic tails of different lengths during my PhD thesis. We systematically investigated these dendrimers for their self-assembling characters and their capacities for both binding and delivery of siRNA. Our results demonstrate that an optimal balance between the hydrophobic alkyl chain length and the hydrophilic dendritic portion plays a crucial role in the self-assembly and the delivery activity towards siRNA.Furthermore, we developed a novel bola-amphiphilic dendrimer by combining bola-amphiphiles and our amphiphilic dendrimers and studied their self-assembly properties and the corresponding siRNA delivery efficiency. This peculiar bola-amphiphilic vector was able to respond to reactive oxygen species for specific delivery, opening a new perspective for the design of stimuli-trigged vectors for targeted siRNA delivery.Finally, I studied the “proton sponge effect” of the amphiphilic dendrimer vectors using the Langmuir monolayer film technique. Our results gave direct evidence of swelling of the amphiphilic dendrimers upon protonation, offering unambiguous experimental data to support the “proton sponge effect”

Full textAmphiphile molecules, endowed with a particular structure containing a hydrophilic head and a hydrophobic tail, have many important applications, e.g., fabrication of detergents, surface coating or surface functionalization, etc. Molecular aggregates of various forms, micelles, vehicle, membranes, etc. can be formed from amphiphile molecules. The complexity of these molecular aggregates involving a large number of atoms make the theoretical study of these system very challenging. Up to now, our understanding of the interaction between nanoparticles and aggregates of amphiphiles remains quite incomplete. Using a variety of molecular simulation methods and some theoretical approaches (Helfrich theory and perturbation theory), we have studied the following issues in the present thesis: 1. How the presence of nanoparticles, especially due to their highly curved surfaces, affects the aggregation of the amphiphiles? 2. How a lipid bilayer, a particular amphiphile aggregate, induces the self-assembly of hydrophobic nanoparticles.3. How the morphology transition of a membrane nanotube can be induced by nanoparticles?

Full textAmphiphilic polysaccharides are composed of a hydrophilic backbone grafted with hydrophobic moieties. They show specific rheological properties due to their capacity to self-associate in solution, but also interfacial properties due to their ability to adsorb at the water/oil interface. However, only few studies describe the chemical modification of complex heteropolysaccharides regarding their structure or their conformation, such as xanthan. Indeed, this polysaccharide can adopt in solution two distinct conformations depending on experimental conditions: an ordered rigid helix or a disordered flexible coil.The objective of this work is to determine the impact of the chain stiffness of hydrophobically modified xanthan on associative properties in solution, but also on stabilizing properties in emulsion.The conformation adopted by the polymer during the grafting procedure has a major impact on the rheological properties: xanthan modified under disordered conformation is an associative polysaccharide, contrary to xanthan modified under its ordered conformation.In addition, hydrophobically modified xanthan is able, under certain conditions, to form and stabilize O/W emulsion, without further addition of molecular surfactant, by partitioning between stabilization of the water/oil interface and viscosifying the continuous aqueous phase

Full textThis thesis deals with the study of shear thickening of an amphiphilic polymers solution by large amplitude oscillatory shear flow (LAOS). The associating polymers based on a weakly charged hydrophilic backbone bearing grafted hydrophobic moieties show a pronounced shear thickening. In order to obtain a deeper insight, the polymer solutions have been submitted to LAOS. In this way, frequency as well as strain or shear stress can be varied. Shear thickening is observed under LAOS, since after a linear regime, dynamic moduli increase significantly above a critical stress. It is shown that this phenomenon appears, for a given concentration (Cp), at a critical stress which increases when frequency (ω) decreases. With different Cp, an equivalence Cp/ω is obtain. The response of LAOS can be decomposed into elastic and viscous parts. This method is easy to determine nonlinear parameters

Full textThe objective of this work was to synthesize and characterize amphiphilic copolymers based on poly(ethylene glycol) (PEG) and poly(lactic acid) (PLA) intended for drug delivery applications. The polymers were chosen regarding to their biocompatibility and bioresorbability. Different architectures of amphiphilic copolymers were prepared, and their behavior in aqueous media, as well as their abilities to encapsulate drugs were studied. First, a graft copolymer was synthesized through copolymerization of a functional monomer, monopropargylated glycolide, with L-lactide to yield a functionalized polyester backbone. The latter was then grafted with different densities of hydrophilic branches of PEG. Then, a brush-like triblock copolymer was synthesized through ROP and ATRP. To this end, chain ends of a telechelic block of PLA were modified to yield a macroinitiator able to initiate oligo(ethylene glycol) methacrylate polymerization with variable substitution degrees. Self-assembly and drug loading studies revealed that architecture and hydrophobic/hydrophilic balance played a major role on the nature of the formed objects and on their encapsulation potential. Finally, to modulate and increase the efficacy of encapsulated drugs, functionalization strategies were realized. This is illustrated by the linking of a fluorescent model molecule on a triblock brush-like copolymer and, in a collaboration project, the linking of an immunostimulant peptide on an amphiphilic diblock system. Comparison with other formulations revealed that the conjugate allowed modulating and reinforcing the drug's efficacy

Full textThe objective of this thesis was to investigate the physical chemical properties of interpenetrating networks formed by self-association of responsive amphiphilic block copolymers. The first part of the work was to develop IPSAN hydrogels(InterPenetrated Self-Assembled Network) simply by mixing two triblock copolymers. Thus, the IPSAN corresponds to a combination of the two polymer networks. The first network is formed by a triblock copolymer based on poly(ethylene oxide) (tPEO) bearing UV-cross-linkable hydrophobic blocks. The second network consists of a pH-sensitive triblock polyelectrolyte based on poly(acrylic acid) (TH50). The influence of the concentration of the copolymers and of the pH on the structure and the mechanical properties of the IPSAN has been systematically studied. In the second part we investigated the influence ofthe formation of a transient network on the phase separation in mixtures of tPEO and linear polymers in aqueous solution. This study revealed an increase of the incompatibility between the two polymers induced by the association of the triblock copolymer. This system enabled the elaboration of photo-cross-linked macroporous hydrogels. Finally, the diffusion of linear polymers and solid particles in pH-sensitive networks was studied by fluorescence recovery after photobleaching (FRAP)and dynamic light scattering. The formation of a transient network restricts the movement of small linear tracers in the same way as a covalent hydrogel would. The control of the structure and dynamics of the network by the pH is a powerful tool to control the diffusion in this hydrogels

Full textRiboflavin (RF) is an essential vitamin for cell growth and development. It possesses interesting physicochemical properties and is internalized by the cells through specific transporters. The first aim of this study was to prepare amphiphile derivatives of RF (RFA) and study their auto-assembly. The second aim was to insert RFA into established drug delivery systems and test their tumour-targeting potential in vitro and in vivo. RFA were prepared by the molecule functionalization with lipid moieties in different positions. One of them, a phospholipid-like derivative (RfdiC14) was able to self-assembly in aqueous solutions into μm-sized 3D objects constituted from slightly curved multilayer lamellas. The bilayer architecture and dynamics were very different from ordinary phospholipids. In contrast, the insertion of small amount of RfdiC14 in a liposome did not influence membrane dynamics and physicochemical characteristics. RfdiC14-functionalised liposomes displayed high and specific uptake in vitro in A431, PC3 cells and HUVECs. The efficiency of RF targeting was also tested in vivo. For that purpose, liposome composition was optimized and a new RF amphiphile with a PEG spacer between RF and lipid was prepared. The tumour accumulation of the liposomes labelled with ICG was studied by photoacoustic imaging in A431 tumour model. The biodistribution of DiR labelled liposomes was accessed by combined μCT/FMT imaging in PC3 tumour model. The results show slight improvement of the tumour accumulation in A431 xenographts and the enhancement of vascular targeting in PC3 tumour model. The overall biodistribution of the RF-targeted liposomes was comparable to control
