Abstract:
The results of experimental studies of changes in the structure, microhardness, and wear resistance of the AMG6 aluminum alloy during equal channel angular pressing (ECAP) are presented in this work. The evolution of the fine structure and the formation of secondary phases in the AMG6 alloy during ECAP were studied. The dark-field image of the structure of the AMg6 alloy in the matrix reflex showed the splitting of the material into small disoriented fragments of about 0.5 μm in size with a small-angle disorientation boundary (about 2–5°). Optimal method and modes of ECAP of the AMG6 aluminum alloy were selected of the bases of experimental research, which make it possible to obtain a workpiece with enhanced tribological and mechanical characteristics. It was established that the most intensive grinding of the grain structure in the AMG6 alloy occurs at ECAP-12 at a channel angle intersection of 120°. It is shown that with a decrease in grain size, the microhardness of the alloy AMG6 after ECAP increases by 4 times, compared with the initial state.The results of the test samples for abrasive wear showed a decrease in mass loss after 12 passes of ECAP, which indicates an increase in the wear resistance of the alloy AMG6 by 13–14 %, compared with the initial state.

Abstract:
Экспериментально исследовано влияние наноструктурования на механические и нелинейные упругие характеристики поликристаллического алюминиевого сплава AMg6 и нанокомпозита n-AMg6/C60. В образцах алюминиевого сплава AMg6 и нанокомпозита n-AMg6/C60 измерены механические характеристики и, ультразвуковым методом, все независимые коэффициенты упругости второго и третьего порядков. Коэффициенты упругости третьего порядка были определены методом Терстона-Браггера по результатам экспериментальных измерений зависимости скорости сдвиговых и продольных объемных акустических волн в исследуемых образцах от величины одноосного сжатия. Спектральным акустическим методом исследованы нелинейные упругие свойства и определены нелинейные акустические параметры этих материалов. Образцы и исследования их механических характеристик выполнены в рамках госзадания Министерства образования и науки РФ на 2016 г. (проект N 3562). Ультразвуковые исследования были выполнены за счет гранта Российского научного фонда (проект N 14-22-00042).

The paper presents the results of tests to failure of aluminum alloys samples AMg6, AMg6M of various forms. Among the samples there were uniaxial samples, tubular samples in torsion, round membranes designed to hidrolipoclasia, thin cut plates, flat samples with holes and round solid samples with an annular undercut with different radius cuts. The impact of volume stress state, calculated by the method of mathematical simulation of each type samples loading processes, on maximum deformation is discussed.

Abstract:
The results were shown in influence of fast heating parameters on the structure and properties of cold-worked alloy AMg6 with original hot-forged structure. Based on the measured data, the change of mechanical properties of cold-worked alloy AMg6 during the process of short duration heating was evaluated. There was reviewed the role of the temperature and the time of heat on the processes of softening the samples of cold-worked alloy AMg6. The stability of mechanical characteristics of hammer-hardened alloy AMg6 under elevated test temperatures was evaluated. It is shown that the return processes in cold-deformed AMg6 alloy during heating in the temperature range studied receive the most intensive development in the first 5–10 minutes, reducing the hardening effect from cold deformation, determined by tensile strength, respectively: by 8–9% with 100 °C; 26–27% at 150 °C; 37–38% at 200 °C; 42–44% at 250 °C and 50% at 300 °C. A decrease in the yield strength during high-speed heating in the temperature range studied is much faster ,compared with the change in the tensile strength. Hour exposure at 200 °C reduces the hardening effect on the yield strength from 340 MPa to 258 MPa, while the tensile strength decreases from 430 MPa to 385 MPa.

AbstractThe ultrasonic study results for dependence of the elastic wave velocities and second-order elasticity coefficients of the polycrystalline aluminum alloy AMg6 and its nanocomposite n -AMg6/C_60 on hydrostatic pressure up to 1.6 GPa have been described. The ultrasonic research has been carried out using a highpressure ultrasonic piezometer based on the piston-cylinder device. The pressure derivatives of the secondorder elastic constants of these materials established in the present study have been compared with the results of the third-order elastic constants measurements of the test alloys using the Thurston–Brugger method. Involving available literature data, we determined the relationships between the pressure derivatives of the second-order elastic constants of the AMg6 alloy and the Mg-content and nanostructuring.

Abstract:
Electron microscopy and x-ray analysis and mechanical testing have been investigated the influence of severe plastic deformation on structure and mechanical properties of aluminum alloys. It is established that in the initial state in the alloy AMC has a high density of chaotically distributed dislocations with a density of 5-10*109 сm-2. It is shown that microdiffraction paintings in alloy AMC in the bulk of grains are observed uniformly distributed particles of the second phase. It is established that in the initial state in the alloy AMG6 there is a high density of chaotically distributed dislocations with a density of 2-6 *1010 сm-2. Determined that after ECAP the dislocation structure of alloys AMG6, AMC and changes: formed dislocation networks inside the fragments of the dislocation is practically not observed. Determined that after ECAP-12 increase the tensile strength and yield strength of alloys AMG6 and AMC.

Об’єкт дослідження – фізико-хімічні процеси, які відбуваються внасідок електроіскрового легування міддю та ультразвукової ударної обробки поверхні алюмінієвого сплаву АМг6. Мета роботи – синтез композиційних покриттів на поверхні сплаву АМг6 електроіскровим легуванням Сu з подальшою ультразвуковою ударною обробкою. Методи дослідження – мікродюрометричний, рентгеноструктурний та мікрорентгеноспектральний аналіз, просвічувальна електронна мікроскопія, а також випробування на корозійну стійкість. Практичне значення – одержані в роботі наукові результати представляють практичний інтерес для машинобудування та авіаційної галузі. Результати досліджень – запропоновано ефективний спосіб зміцнення поверхневого шару алюмінієвого сплаву АМг6 за допомогою комбінованоїобробки – ЕІЛ та УЗУО. Електроіскрове легування мідним електродом сприяє зміцненню поверхні за рахунок утворення в приповерхневому шарі товщиною до 25 мкм інтерметалідних фаз – Аl2Cu та Al6Mg4Cu. Ультразвукова ударна обробка обумовлює диспергування субзеренної/зеренної структури, підвищує твердість на 70-80% та сприяє інтенсифікації масопереносу легуючого елементу. Результатом комбінованої обробки є підвищення мікротвердості (в 3,5 рази) та корозійної стійкості поверхневих шарів сплаву АМг6 у порівнянні з вихідним станом.
The object of research is the physical and chemical processes that take place near the electrosurgical doping of copper and ultrasonic shock treatment of the surface of aluminium alloy AMg6. The purpose of the work is the synthesis of composite coatings on the surface of AMg alloy by the electrospray doping of Cu followed by ultrasonic shock treatment. Methods of research - microdyrometric, X-ray and micro-ray diffraction analysis, transdermal electron microscopy, as well as corrosion resistance tests. Practical significance - the scientific results obtained in the work are of practical interest to the mechanical engineering and aviation industry. The results of the research - an effective way to strengthen the surface layer of aluminium alloy AMg6 with the help of combined processing - EIL and UIT. Electrospray doping with copper electrode helps to strengthen the surface due to the formation in the near-surface layer up to 25 microns of intermetallic phases - Al2Cu and Al6Mg4Cu. Ultrasonic shock treatment causes the dispersion of the sub-grains / grain structure, increases the hardness by 70-80 % and promotes the intensification of the mass transfer of the alloying element. The result of the combined treatment is an increase in microhardness (3,5 times) and corrosion resistance of the surface layers of AMg6 alloy as compared to the initial state.

Abstract:
In the 21st century, cancer is becoming the curse of the ageing population in developed countries. No satisfactory therapies are available for many tumour types, and application of existing therapies is limited by severe side effects. Thus, there is a great need of new approaches in cancer treatment. DAB-Am16 is a dendritic polymer with a globular structure, consisting of poly(propylene imine) branches that emerge from a diaminobutane core. Its intrinsic tumoricidal activity in mouse models was published in 2005, but no information on the mechanism of action was available. This thesis presents novel findings on the pathways underlying the anti-cancer activity of this dendrimer in vitro and in vivo. Extensive chemical characterisation of DAB-Am16 confirmed its stability and purity. Severe time- and concentration-dependent cytotoxicity was observed for a panel of human tumour cell lines, while a small population of persisters was identified. Toxicity was accompanied by a delayed or abrogated cell cycle. There was an increased number of S phase cells, while the ability to synthesise DNA or to undergo mitosis was progressively lost with increasing DAB-Am16 concentration. The following cell death was found to be apoptotic and was biased in a cell cycle phase dependent manner. The order of apoptotic events upon DAB-Am16 exposure was determined. Finally, an in vivo experiment confirmed that DAB-Am16 has a pronounced effect against human pancreatic cancer xenografts in mice, while being well tolerated by the animals. Post mortem examination of tumour tissue revealed cell cycle blockage of tumour cells from DAB-Am16-treated mice. However, disposition for further proliferation was not diminished, and no significant difference in tumour vascularisation was observed.

Introdução: Tumores adrenocorticais (TAC) são raros, correspondendo somente a 0,2% de todas as neoplasias pediátricas, sendo que a maioria dos casos são diagnosticados no Brasil e estão associados com a mutação TP53 p.R337H. A cirurgia é o único tratamento efetivo conhecido para os TAC, sendo os tumores em estadios avançados frequentemente fatais. A família das Aurora-quinases é formada por três membros (Aurora-A, -B e -C) os quais atuam em diversas fases do ciclo celular, como alinhamento dos cromossomos, formação do fuso mitótico e citocinese. Diferentes trabalhos mostraram a expressão alterada de membros desta família em vários tipos de tumores e a inibição da atividade destas proteínas tem sido considerada uma potencial abordagem para o tratamento do câncer. Objetivo: A partir da análise da expressão dos genes Aurora-A e Aurora-B em amostras de TAC pediátrico, foram investigados os efeitos do AMG 900, um pan-inibidor de aurora quinases, na proliferação, apoptose, síntese hormonal e perfil transcricional da linhagem H295A. Além disso, foram avaliados os efeitos do AMG 900 combinado com diferentes quimioterápicos. Metodologia: Os níveis de expressão dos genes Aurora-A e Aurora-B foram analisados em 60 crianças com TAC através das técnicas de RT-qPCR e imuno-histoquímica. A proliferação celular foi avaliada por coloração com Giemsa e a apoptose foi realizada por citometria de fluxo. A análise de combinação de drogas foi feita com base no método de Chou-Talalay e o ensaio de microarray foi realizado utilizando a plataforma da Agilent. Resultados: A expressão dos genes Aurora-A e Aurora-B foi associada com estadios avançados da doença e a expressão do Aurora-A foi associada com a presença da mutação TP53 p.R337H. O tratamento com o AMG900 causou a inibição da proliferação, aumento da apoptose e sensibilizou as células para os inibidores de topoisomerase II (doxorrubicina e etoposídeo). Adicionalmente, o AMG 900 levou à redução da síntese de hormônios bem como modulou a expressão de genes envolvidos com esta atividade. A inibição das aurora-quinases alterou a expressão de genes associados com a regulação da fase G1 do ciclo celular e afetou a expressão de genes da via de sinalização Notch. Conclusão: A inibição das aurora-quinases pelo AMG 900 pode ser uma alternativa para o tratamento dos tumores adrenocorticais.
Introduction: Pediatric adrenocortical tumors (ACT) are rare malignancies representing only 0.2 % of all pediatric cancers. Most cases are diagnosed in Brazil and are associated with TP53 p.R337H mutation. Surgery is the only effective treatment known for the ACT however this approach has a small impact on survival in advanced disease. The Aurora kinase family is comprised of three members (Aurora-A, -B and -C) which act at different phases of the cell cycle, such as chromosomes alignment, mitotic spindle formation and cytokinesis. Several studies have demonstrated altered expression of members of this family in various types of tumors and the functional inhibition of the aurora kinases have been considered as a potential approach to cancer treatment. Aim: On the basis of analysis of Aurora-A and Aurora-B gene expression in the samples from pediatric ACT, we investigated the effects of AMG 900, a pan-aurora kinase inhibitor, on proliferation, apoptosis rate, hormone synthesis and transcriptional profile of H295A cell line. Furthermore, we evaluated the effects of AMG 900 combined with different chemotherapeutic agents. Methods: The mRNA expression levels of Aurora-A and Aurora-B genes were analyzed in 60 children with ACT by RT-qPCR and immunohistochemistry. Cell proliferation was assessed by Giemsa staining and apoptosis was performed by flow cytometry. Drug combination analysis was made on the basis of Chou- Talalay method. Microarray experiments were carried out using the Agilent human microarray. Results: Aurora-A and Aurora-B overexpression was associated with advanced disease. Patients carrying the TP53 p.R337H mutation presented significantly higher expression values of Aurora-A. Treatment with AMG900 caused inhibition of proliferation, increased apoptosis and sensitized the cells to topoisomerase II inhibitors (doxorubicin and etoposide). Additionally, the AMG 900 led to decreased synthesis of hormones and modulated the expression of genes involved in this activity. Finally, Aurora kinases inhibition altered the expression of genes associated with G1 cell cycle phase regulation and affected the Notch signaling pathway target genes. Conclusion: These data suggest that Aurora kinase inhibition by AMG900 may be a new therapeutic approach to adrenocortical carcinoma treatment.
