Abstract:
This research problematises the contemporary phenomenon of alternative arts education after art’s ‘Educational Turn’, encompassed by evidence of a critical discourse between 2006 and 2016. The thesis addresses the questions: what are the alternatives to models of the alternative art school having emerged through the Educational Turn? And, how might dialogic engagement with organisations outside of the Turn propose something other for the future of alternative arts education? Contemporary art’s capacity to instrumentalise education, through its reimagining by artists and the co-option of ‘the alternative’ by arts institutions, must be countered by considering organisational models that sit outside of the Educational Turn. The field is contextualised by a ‘crisis in education’ in the UK, contributing to an abundant manifestation of ‘alternative’ art schools. An often-overlooked plurality exists to ‘the alternative’ that, in its co-option by contemporary art, is rendered homogenised. Existing discourse considers artistic, self-organised and curatorial practices, framed by institutional and infrastructural critique, but neglects to step outside of the Turn to imagine other models for alternative arts education. ‘Knowledge mobility’, ‘the dialogic’ and ‘(trans)formation’ form a framework for the thesis, functioning according to a methodology of critique and proposition. The research derives ‘knowledge mobility’ to critique the Turn’s instrumentalisation of education, by examining existing discourse and practice that problematise the paradoxes of the Turn and frame knowledge as a form of social organisation. The research aligns ‘the dialogic’ from Mikhail Bakhtin and Paulo Freire, with Julia Kristeva and Roland Barthes’ ‘intertextuality’ and Maurice Blanchot’s ‘infinite conversation’. The function of ‘the dialogic’ is twofold: as a structural metaphor and conversational research practice. Four dialogues with organisations operating outside of the remit of the Turn consider the productive and transformative capacities of models not framed as alternative art schools. These are with: Leeds Creative Timebank, IF Project, THECUBE and Syllabus programme. Negotiating critical and applied interpretations of ‘knowledge mobility’, findings from these are reconciled with the research through a process of ‘(trans)formation’, resulting in the proposition of speculative principles to contribute to the field of alternative arts education. The research has been produced as part of the UK Arts and Humanities Research Council’s (AHRC) Creative Exchange knowledge exchange hub, providing the context for stepping outside of the domain of contemporary art. The value of this approach for the field of alternative arts education is in its capacity to have drawn together thinking from each organisation. This research makes its contribution to the field of alternative arts education by working dialogically with organisations where the practice of knowledge is central, establishing a connection between organisations outside of the Turn, which would otherwise be excluded from its discourse, with contemporary art. The research formulates and puts into practice methods of critique, conversation and proposition: producing a critical vocabulary, lens and through deriving speculative propositions towards a possible future for alternative arts education.

This work aimed to evaluate control strategies of hairy fleabane (Conyza bonariensis) and also study the arthropods associated with this weed. For this, the work was divided into three chapters. The Chapter I aimed to evaluate the selectivity of herbicides with potential use to control hairy fleabane on forming species winter pastures, which are: oats, black oats, ryegrass and tetraploid ryegrass. Experiments were conducted under greenhouse conditions and evaluated the phytotoxicity of plants and production of shoot dry matter (SDM). The treatments were: 2,4-D amine (806 and 1612 g a.i. ha-1), metsulfuron-methyl (2.4 and 4.8 g a.i. ha-1); clorimuronethyl (20 and 40 g a.i. ha-1); imazetaphyr (350 and 700 g a.i. ha-1); diclosulam (16.8 and 33.6 g a.i. ha-1); saflufenacil (35 and 70 g a.i. ha-1) and flumioxazin (50 and 100 g a.i. ha-1), and the control. According to the results obtained, the selectivity of herbicides is species cultivated dependent and it is important to know the botanical composition of pasture before applying any herbicide. In Chapter II aimed to evaluate the combination effect of the herbicides glyphosate and saflufenacil at different doses on the control of hairy fleabane plants. The treatments consisted of doses that followed the logarithmic scale, and used the herbicides glyphosate and saflufenacil both applied alone and in mixture. Were evaluated the visual control and the SDM production of hairy fleabane plants. The results indicate that there is a synergistic relationship between the herbicide glyphosate and saflufenacil. Also, the association of saflufenacil and glyphosate is effective in controlling glyphosate-resistant hairy fleabane. The addition of the herbicide glyphosate with saflufenacil, besides improve control, prevents the occurrence of regrowth of hairy fleabane plants. In Chapter III, hairy fleabane plants were collected fortnightly in Boa Vista and Sao Vicente Incra South in order to evaluate the arthropods associated with this weed. The results show that hairy fleabane takes an important role as an alternative host of important pests of the soybean, such bugs and caterpillars, and too natural enemies.
Este trabalho teve como objetivo avaliar estratégias de controle de buva (Conyza bonariensis) e também estudar a artropodofauna associada a esta planta daninha. Para isso, o trabalho foi dividido em três capítulos. O capítulo I teve como objetivo avaliar a seletividade de herbicidas de uso potencial para o controle de buva sobre espécies formadoras de pastagens de inverno, sendo elas: aveia branca, aveia comum, azevém comum e azevem tetraplóide. Para isso, foram conduzidos experimentos em casa de vegetação e avaliada a fitointoxicação de plantas e a produção de massa de matéria seca da parte aérea (MMSPA). Os tratamentos avaliados foram: 2,4-D amina (806 e 1612 g i.a. ha-1); metsulfuron-methyl (2,4 e 4,8 g i.a. ha-1); clorimuron-ethyl (20 e 40 g i.a. ha-1); imazetaphyr (350 e 700 g i.a. ha-1); diclosulam (16,8 e 33,6 g i.a. ha-1); saflufenacil (35 e 70 g i.a. ha-1) e flumioxazin (50 e 100 g i.a. ha-1), além da testemunha. De acordo com os resultados obtidos, a seletividade dos herbicidas testados é dependente da espécie que está sendo cultivada, sendo importante se conhecer a composição botânica da pastagem antes de se aplicar qualquer herbicida. No capítulo II, objetivou-se avaliar o efeito da associação dos herbicidas glifosato e saflufenacil, em diferentes doses sobre o controle de plantas de buva. Os tratamentos foram compostos por doses que seguiram a escala logarítmica, sendo utilizados os herbicidas glifosato e saflufenacil tanto em aplicação isolada quanto em mistura. Foram avaliados o controle visual (%) e a produção de MMSPA das plantas de buva. Os resultados obtidos apontam que: existe uma relação sinérgica entre os herbicidas glifosato e saflufenacil; a associação de saflufenacil e glifosato é eficiente no controle de buva resistente ao herbicida glifosato e; a adição de glifosato ao herbicida saflufenacil, além de melhorar o controle, previne a ocorrência de rebrote de plantas de buva. No capítulo III, foram realizadas amostragens quinzenais de plantas de buva em Boa Vista do Incra e São Vicente do Sul afim de se avaliar a artropodofauna associada à esta planta daninha. Os resultados mostram que a buva assume importante função como hospedeiro alternativo de pragas importantes da cultura da soja, tais como espécies de percevejos e lagartas, assim como de inimigos naturais.

Abstract:
In many parts of the country principal candidates are being licensed through alternative pathways. Some view this movement as a plausible solution to the shortage of principals and inadequacy of principal preparation programs (Hess, 2003; Southern Regional Education Board, 2006; Thomas B. Fordham Institute, 2003). Others are insulted and threatened by the prospect of a person from a non-traditional background leading a school (Fenwick & Pierce, 2001). The debate as to whether or not these candidates possess the prerequisite skills and knowledge to effectively lead a school continues. But, will superintendents, as gatekeepers to school districts, afford these individuals the opportunity to prove their worth as principals? In this study, superintendents’ attitudes toward alternative licensure of school principals; past behaviors related to hiring alternatively licensed school personnel; attitudes toward specific alternatively licensed personnel; perceptions of the conditions in their school districts; anticipated concerns about hiring alternatively licensed principals; and the presence or absence of a clearly articulated induction program for new principals are used as predictors of superintendents' willingness to recommend hiring alternatively licensed principals to their school boards. The composite model of attitude-behavior consistency and data from a qualitative study of 18 superintendents were the bases for the development of a theory. To test the theory, an on-line questionnaire, using Likert and Thurstone scaled items, was administered to 1200 randomly selected superintendents who were members of the American Association of School Administrators (AASA) in 2005. Multiple regression analysis was used for the analysis of the quantitative data. Five of the 19 predictor variables were significant predictors of superintendents’ willingness to recommend hiring alternatively licensed principals. The strongest relationship existed between superintendents’ willingness to recommend hiring alternatively licensed principals and their perceptions of the instructional leadership ability of alternatively licensed principals. Other significant predictors were superintendents’ past experiences hiring alternatively licensed principals, perceptions of the community acceptance of alternatively licensed principals, general attitude toward alternative licensure, and willingness to hire under the given definition. Superintendents displayed a low neutral (more unfavorable) score on the Thurstone scale, which means they view the employment of alternatively licensed principals slightly unfavorably.

La présente thèse a pour objet d'élaborer une étude d'ensemble de la modalité alternative de l'obligation. Sont analysées les principales caractéristiques de l'obligation alternative, notamment la faculté d'option offerte à l'une des parties entre plusieurs modalités d'exécution. La confrontation de l'obligation alternative avec les autres modalités de l'obligation et les mécanismes optionnels voisins permet de clarifier la notion. Un recensement des principales applications contractuelles et légales de la modalité alternative telles que certaines valeurs mobilières composées, les warrants financiers et les obligations pluri-monétaires, est effectué aux fins d'observer l'évolution de la pratique contractuelle la concernant depuis le code napoléon. L'étude du régime juridique de l'obligation alternative précise l'ensemble des règles applicables à cette modalité notamment les règles relatives aux rapports entre les parties antérieurement à l'option (le régime des risques des biens compris dans l'obligation) et postérieurement à la date de l'option (les conséquences de l'option) ainsi que les effets juridiques du refus d'effectuer la faculté de choix.

Abstract:
La plupart des gènes humains codants génèrent des transcrits alternatifs (isoformes) par épissage alternatif (alternative splicing, AS) et polyadénylation alternative (APA) en général dans la région codante et la région 3’ non traduite (3’UTR), respectivement. Le rôle de l’AS et la 3’UTR-APA est de plus en plus reconnu dans l’oncogenèse. En particulier, des réseaux d’AS connectant des facteurs d’épissage et des variants d’épissage ont récemment été identifiés. L’AS est aussi largement régulé par les agents anticancéreux génotoxiques, tel que la doxorubicine et le cisplatine (induisant des différents types de lésions sur l’ADN), qui sont régulièrementt utilisés dans les traitements du cancer du sein et du poumon non-à-petites-cellules (non-small-cell lung cancer, NSCLC), respectivement. Étant donné l’apparition fréquente de résistances aux chimiothérapies, comprendre les mécanismes sous-jacents est crucial pour surmonter ce problème clinique. Il existe des exemples d’évènements d’AS associés à la résistance aux agents anticancéreux, mais l’implication des facteurs d’épissage et des réseaux d’AS est très peu connue. De plus, une étude précédente a démontré que la doxorubicine réprime un grand groupe d’exon terminaux alternatifs (alternative last exons, ALE), qui correspondent à l’utilisation de sites de polyadénylation introniques (intronic polyadenylation, IPA). Les ALEs ont un rôle émergent dans le cancer, mais on ne sait encore que très peu sur leur régulation par d’autres agents anticancéreux, tel que le cisplatine. Afin de mieux comprendre le rôle des régulations d’AS et d’APA dans la réponse et la résistance cellulaire à la chimiothérapie, mon projet de thèse avait deux objectifs principaux : 1) déterminer l’étendue, les réseaux régulateurs, et les fonctions des régulations d’AS dans la résistance à la doxorubicine des cellules de cancer du sein, et 2) déterminer l’étendue, les mécanismes, et l’impact des régulations d’ALE en réponse au cisplatine dans des cellules de NSCLC. Dans la première partie, j’ai identifié par RNA-seq des milliers d’évènements d’AS et des dizaines de facteurs d’épissage régulés dans un modèle cellulaire de cancer du sein ER+ résistant à la doxorubicine. Par un miniscreen siARN, j’ai identifié deux facteurs, ZRANB2 et SYF2, impliqués dans la résistance à la doxorubicine. D’autres analyses RNA-seq ont révélé les évènements d’AS régulés par ces deux facteurs peu étudiés, ainsi que leur convergence vers l’exon 5 alternatif de l’oncogène ECT2. La déplétion de ZRANB2, SYF2, et du variant ECT2-ex5 réduit l’arrêt en phase S induit par la doxorubicine et la résistance des cellules. De plus, un niveau élevé d’inclusion de l’exon 5 d’ECT2 corrèle avec une mauvaise survie spécifiquement de patientes ER+ traitées par chimiothérapie. Dans la deuxième partie, j’ai identifié par 3’-seq que le traitement cisplatine (mais pas oxaliplatine) induit des ALEs/IPAs dans des milliers de gènes enrichis en gènes de cycle et de mort cellulaire. Cet effet est lié à une inhibition de la processivité de l’élongation dans les longs gènes. Une analyse 3’-seq sur polysomes m’a permis de montrer que ces régulations d’ALEs impactent le traductome, et a révélé un groupe d’isoformes particulièrement courtes peu efficacement traduites, dont un transcrit connu avec une fonction non-codante. En conclusion, j’ai pu identifier durant ma thèse un nouveau réseau d’AS impliqué dans la résistance à la doxorubicine des cancers du sein ER+, et une importante régulation d’ALEs impactant le traductome en réponse au cisplatine dans des cellules NSCLC. Ces travaux améliorent notre compréhension du rôle de l’AS et des ALE/IPA dans la réponse et la résistance cellulaire à la chimiothérapie anticancéreuse. Au plus long terme, les transcrits alternatifs et les régulateurs identifiés constituent des biomarqueurs candidats de chimiorésistance
Most human coding genes generate alternative transcripts (isoforms) through alternative splicing (AS) and alternative polyadenylation (APA), most often within the coding region and the 3’ untranslated region (3’UTR), respectively. Both AS and 3’UTR-APA regulations have been increasingly involved in oncogenesis. In particular, AS networks connecting oncogenic splicing factors and oncogenic splicing variants have been recently identified. AS is also widely regulated by genotoxic anticancer drugs, like doxorubicin and cisplatin that induce different types of DNA lesions and are widely used in breast cancer and non-small-cell lung cancer (NSCLC) therapy, respectively. Given the frequent occurrence of resistance to chemotherapy, understanding the underlying mechanisms is crucial to overcome this major issue. There are examples of AS events associated with anticancer drug resistance, but very little is known about the splicing factors and therefore the AS networks involved. In addition, a previous study showed that doxorubicin represses a large set of alternative last exons (ALE) corresponding to the use of intronic polyadenylation (IPA) sites. ALEs have an emerging role in cancer, but little is known about its regulation by other anticancer drugs, like cisplatin. In order to better understand the role of AS and APA regulation in cell response and resistance to chemotherapy, my PhD project had two main aims: 1) determine the extent, regulatory networks and function of AS regulation in breast cancer cell resistance to doxorubicin, and 2) determine the extent, mechanism and impact of ALE regulation in response to cisplatin in NSCLC cells. In the first part, I identified by RNA-seq thousands of AS events and dozens of splicing factors regulated in a cell model of acquired resistance to doxorubicin in ER+ breast cancer. Through an siRNA miniscreen, I found two splicing factors, ZRANB2 and SYF2, involved in doxorubicin resistance. Further RNA-seq analyses revealed the AS events regulated by depletion of these poorly characterized splicing factors, and their convergence on the alternative exon 5 of the oncogene ECT2. Depletion of ZRANB2, SYF2 and the ECT2-Ex5 variant reduces doxorubicin-induced S phase arrest and doxorubicin resistance. In addition, high inclusion levels of ECT2-Ex5 correlate with poor survival specifically in ER+ breast cancer treated with chemotherapy. In the second part, I found by 3’-seq that in NSCLC cell treatment with cisplatin (but not oxaliplatin) induces ALE/IPA in thousands of genes enriched in cell cycle and cell death. This effect is linked to an inhibition of transcription elongation processivity in long genes. 3’-seq analysis on polysomes showed that this ALE regulation impacts the translatome, and revealed a set of particularly short isoforms that were inefficiently translated, including a transcript with a non-coding function. In conclusion, during my thesis, I could identify a novel AS network involved in doxorubicin resistance in ER+ breast cancer, and widespread ALE regulation impacting the translatome in lung cancer cisplatin response. This work increases our understanding of AS and IPA role in cell response and resistance to anti-cancer chemotherapy. In the longer term, the identified alternative transcripts and regulators constitute candidate biomarkers of chemoresistance
