Full textThe objective of the present work was to evaluate the physiologic quality of carrot seeds, cilantro and parsley, associated with Alternaria alternata and A. dauci under two inoculation methods, conidial suspension and restriction hidric. The isolated of A. dauci and A. alternata were obtained, respectively, of carrot plants with symptom of the disease and of seeds of each one of the species, submitted to an initial test of sanity (Blotter Test). The inoculation treatments for suspension consisted in: testifies (distilled water and sterilized), conidial suspension of A. alternata, conidial suspension of A. dauci and conidial suspension of both species (A. alternata + A. dauci). Para restriction hidric: PDA + manitol (testifies); PDA + manitol + A. alternata; PDA + manitol + A. dauci; PDA + manitol + A.alternata + A.dauci. The physiologic quality of the seeds was evaluated by a group of tests accomplished in controlled conditions of laboratory and green house. The quality of carrot seeds, cilantro and parsley is influence by the association with it would Alternaria alternata and A. dauci. The use of the inoculation of seeds through the conidial suspension is efficient in the contamination of carrot seeds for A. alternata and A. dauci. The use of the manitol, as a restritor hidric, is effective for the obtaining of seeds infected by A. dauci in the culture of the parsley. Already for cilantro seeds, both methods demonstrated satisfactory levels of infection.
O objetivo do presente trabalho foi avaliar a qualidade fisiológica de sementes de cenoura, coentro e salsa, associadas com Alternaria alternata e A. dauci, sob dois métodos de inoculação, suspensão de conídios e restrição hídrica. Os isolados de A. dauci e A. alternata foram obtidos, respectivamente, de plantas de cenoura com sintomas da doença e de sementes de cada uma das espécies, submetidas a um teste inicial de sanidade (Blotter Test). Os tratamentos de inoculação para suspensão consistiram em: tratamento testemunha (água destilada e esterilizada), suspensão de conídios de A. alternata, suspensão de conídios de A. dauci e suspensão de conídios de ambas as espécies (A. alternata + A. dauci). Para restrição hídrica: BDA + manitol (tratamento testemunha); BDA + manitol + A. alternata; BDA + manitol + A. dauci; BDA + manitol + A.alternata + A.dauci. A qualidade fisiológica das sementes foi avaliada por um conjunto de testes realizados em condições controladas de laboratório e em casa de vegetação. A qualidade das sementes de cenoura, coentro e salsa é prejudicada pela associação com Alternaria alternata e A. dauci. A utilização da inoculação de sementes através da suspensão de conídios é eficiente na contaminação de sementes de cenoura por A. alternata e A. dauci. A utilização do manitol, como um restritor hídrico, é eficaz para a obtenção de sementes infectadas por A. dauci na cultura da salsa. Para sementes de coentro, ambos os métodos demonstram níveis satisfatórios de infecção.

Full textThe present study aimed to evaluate the effects of different gamma irradiation doses on the growth of Alternaria alternata and on production of mycotoxins Alternariol (AOH) and Alternariol Monomethyl Ether (AME) in cereal samples. The results showed a significant reduction in the number of CFU/g in rice grains and sunflower seeds, which were proportional to radiation dose used. However, in corn and wheat grains was observed an increase in the number of CFU/g with the increase of gamma irradiation. The radiation doses used resulted in a reduction of AOH levels. In rice and corn grains, the production of AOH was highest in the group irradiated with 5 kGy. Similar result was obtained with relation to AME. Scanning electron microscopy made it possible to visualize structural alterations on A. alternata induced by the different g-radiation doses used. Analysis of irradiated AOH and AME toxins standards didn´t show any alteration comparing to the control group.

The alternaria brown spot, caused by Alternaria alternata f sp. citri, causes large economic damages in tangor Murcott (Citrus sinensis Osbeck x Citrus reticulata [L.] Blanco). Its control is carried out through the spraying of agrochemicals, implying up to 15 pulverizations per year, which causes an increase in the production costs of the cultures and damages to the environment. As an alternative, the use of microorganisms, in particular Bacillus spp., has been used to diseases’ control. Therefore, the aim of this work was to evaluate the viability of Bacillus spp in in vitro and in vivo conditions. The methodologies were based on the interactions between biological control agents (Bacillus spp.) and the phytopathogen A. alternata, evaluated by the paired culture technique, by the production of volatile, thermostable and cell-free metabolites by different Bacillus spp. isolates. The molecular identification of the isolates tested and the efficacy of bacterial isolates were evaluated in leaves and plants under greenhouse conditions. The results showed that most of the isolates affected the development of phytopathogen and produced some types of metabolite, being antibiosis one of the probable mechanisms of action of the bacterium. The isolates ACB-01, ACB-07, ACB-08, ACB-18 and ACB-57 presented potential for disease control of A. alternata.
A mancha marrom de alternaria, causada por Alternaria alternata f sp. citri, causa grandes danos econômicos em tangor Murcott (Citrus sinensis L. Osbeck x Citrus reticulata [L.] Blanco). Seu controle é realizado através de pulverizações com agroquímicos, implicando em até 15 pulverizações por ano, o que acarreta em aumento no custo de produção da cultura e prejuízos ao meio ambiente. Como alternativa, o uso de microrganismos, em particular, as bactérias do gênero Bacillus spp., têm sido empregadas para o controle de doenças. Portanto, esse trabalho teve por objetivo avaliar em condições in vitro e in vivo a viabilidade de 47 isolados de Bacillus spp. para o controle da doença. As metodologias foram embasadas nas interações entre agentes de controle biológico (Bacillus spp.) e o fitopatógeno A. alternata avaliadas pela técnica de cultivo pareado, pela produção de metabólitos voláteis, termoestáveis e livre de células por diferentes isolados de Bacillus spp.. Realizou-se, ainda, a identificação molecular dos isolados testados e a eficácia dos isolados da bactéria em folhas destacadas e em plantas, sob condições de casa de vegetação. Os resultados obtidos mostraram que a maioria dos isolados afetou o desenvolvimento do fitopatógeno e produziram algum tipo de metabólito, sendo, a antibiose um dos prováveis mecanismos de ação da bactéria. Os isolados ACB-01, ACB-07, ACB-08, ACB-18 e ACB-57 apresentaram potencial para o biocontrole de A. alternata.

The results showed that: In in vitro test the ingredients active trifloxystrobin, iprodione, azoxystrobin, difenoconazole, pyraclostrobin, trifloxystrobin + propiconazole and trifloxystrobin + tebuconazole inhibited the micelial growth of the pathogen completely. In the field all fungicides treatments were effectives with productivity. Among the used products the treatment trifloxystrobin + propiconazole was profitable being compared cost and productivity.

A mancha marrom de alternaria, causada pela Alternaria alternata f. sp. citri, afeta o tangor murcote causando lesões em folhas, ramos e frutos, acarretando em desfolha e manchas que depreciam o fruto comercialmente. Os objetivos deste trabalho foram verificar o efeito do meio de cultura no isolamento, da temperatura e do fotoperíodo no crescimento micelial do patógeno, avaliar a sensibilidade do patógeno à fungicidas in vitro e estabelecer o melhor produto e dose para o controle da doença no campo. As avaliações laboratoriais constaram de avaliações de crescimento micelial, medição do tamanho de conídios, concentração de conídos/mL e avaliação de meios de culturas para isolamento do patógeno. Os resultados do teste in vitro mostraram que os ingredientes ativos trifloxystrobin, iprodione, azoxystrobin, difenoconazole, pyraclostrobin, trifloxystrobin + propiconazole e trifloxystrobin + tebuconazole inibiram completamente o crescimento micelial do patógeno. No ensaio de campo todos os tratamento foram superiores a testemunha quanto a produtividade. Concluindo que o controle da doença é recomendado independente do produto utilizado. Entre os produtos utilizados o tratamento com trifloxystrobin + propiconazole foi rentável comparando-se custo e produtividade.

Resumo: O patótipo tangerina do fungo Alternaria alternata produz a toxina hospedeiro específica ACT que afeta tangerinas e seus híbridos e causa a doença conhecida como mancha marrom de alternaria (MMA). A toxina ACT causa necrose em plantas susceptíveis, afetando ramos, folhas novas e frutos. Os sintomas incluem pontos marrons a negros circundados por um halo amarelado. Laranjas doces eram consideradas resistentes à doença uma vez que MMA nunca havia sido descrita no campo. Foi demonstrado que uma linhagem do fungo A. alternata produtor de toxina ACT é capaz de penetrar e colonizar de forma assintomática folhas de laranja 'Pera' sem perder a capacidade de causar necrose em genótipos susceptíveis. Microscopia óptica, inoculação e isolamento de A. alternata em plantas de laranja doce seguida pela inoculação cruzada em folhas susceptíveis de tangor 'Murcott', em adição a detecção por PCR do fungo em folhas assintomáticas de laranja doce no campo confirmam que laranja 'Pera' atua como hospedeiro assintomático do patógeno. A análise de genes (RT-qPCR) e proteínas (2DE) expressas durante a infecção com o fungo demonstrou que diferentes vias de sinalização foram ativadas em plantas de citros em resposta a A. alternata. Durante a resistência plantas responderam ativando as vias de sinalização mediadas por etileno e ácido jasmônico, as quais estão tipicamente envolvidas na defesa de plantas contra microrganismos necrotróficos. Aparentemente a produção de etileno também é requerida durante a susceptibilidade. Genes envolvidos na síntese de etileno aparecem induzidos durante a infecção e desenvolvimento de sintomas. Durante a infecção assintomática as plantas cítricas ativaram genes e proteínas associadas à síntese de fenilpropanóides, indicando o possível papel destes em plantas de laranja
Abstract: The tangerine pathotype of Alternaria alternata produces the host-specific ACT-toxin that affects mandarins and their hybrids and causes the disease known as Alternaria brown spot (ABS). ACT-toxin causes necrosis in susceptible plants, and the disease affects twigs, young leaves and fruits. The symptoms include brown to black necrotic spots that are surrounded by a yellow halo. Sweet oranges were considered resistant to the disease since ABS symptoms have never been reported in the field. It was demonstrated that an ACT-toxin-producing strain of A. alternata is able to penetrate and colonize Pera sweet orange leaves asymptomatically without losing its ability to cause necrosis on susceptible genotypes. Light microscopy, inoculation and recovery of A. alternata from sweet orange leaves followed by subsequent cross-inoculation in susceptible leaves of Murcott tangor and PCR detection of the fungus in sweet orange symptomless leaves collected in the field confirmed that Pera sweet orange may act as an asymptomatic host for the pathogen. The analysis of genes (RT-qPCR) and proteins (2DE) expressed during fungal infection demonstrated that different signaling pathways were activated in citrus plants in response to A. alternata. During resistance, plants respond activating ethylene and jasmonic acid signaling pathways which are typically involved in plant defense against necrotrophic pathogens. Apparently the production of ethylene is also required during susceptibility. Genes involved in ethylene biosynthesis appear to be induced during infection and symptoms development. During asymptomatic infection, citrus plants activate genes and proteins associated to phenylpropanoids biosynthesis, indicating their possible role in sweet orange plants response to the disease

Este estudo teve como objetivo a síntese e caracterização de dietilaminoetilquitosana (QHdDEAE) e dodecil-dietilaminoetil-quitosana (QHdDEAEDOD) de baixa massa molar (Mw), para avaliar suas atividades antifúngicas contra os fungos Alternaria solani, Alternaria alternata e Penicilliun expansum, que provoca um grande impacto na produção e preservação de alimentos processados e minimamente processados. Os derivados de baixa massa molar (Mw) foram obtidos por reação de degradação de quitosana desacetilada com nitrito de sódio, seguido pela inserção de grupos DEAE e Dodecil na estrutura do polímero. O grau de desacetilação (DD) e de substituição (DS) por grupos dietilaminoetil (DEAE) e dodecil foram determinados usando ressonância magnética nuclear de hidrogênio (RMN-H) e caracterizados por infravermelho (IR). O grau de desacetilação e o grau de substituição por grupos DEAE foram de 94,8% 39,8%, respectivamente. Os derivados hidrofóbicos obtidos foram substituídos com 15% e 39,8% de grupos dodecil, conforme determinado por RMN-H. As massas molares foram determinadas usando Cromatografia de Permeação em Gel (GPC) e um Mw de 9,2 kDa foi obtido para quitosana desacetilada degradada. Ensaios in vitro contra os fungos Alternaria alternata, Peninicillium expansum e Alternaria solani foram realizados. Na concentração de 0,5g.L-1, QHdDEAEDOD e QHdDEAE foram mais eficazes que as quitosanas comercial e desacetilada e apresentaram inibições de 80% contra Alternaria alternata e Alternaria solani. A quitosana deacetilada degradada foi mais eficiente na inibição do crescimento fúngico de Penicillium expansum e para 0,5 g.L-1 atingiu cerca de 80% de inibição. Todos os derivados apresentaram viabilidade celular superior a 70% em células 3T3, demonstrando uma boa citocompatibilidade e potencial para aplicações na conservação de alimentos
This study aimed at the synthesis and characterization of Diethylaminoethyl-Chitosan (QHdDEAE) and Dodecyl-Diethylaminoethyl-Chitosan (QHdDEAEDOD) of low molar mass(Mw), to evaluate their antifungal activities against the fungi Alternaria solani, Alternaria alternata and Penicilliun expansum, which causes a great impact on the production and preservation of processed and minimally processed foods. The low Mw derivatives were obtained by degradation reaction with sodium nitrite, followed by the insertion of DEAE and Dodecyl groups on the polymer backbone. The degrees of deacetylation (DD) and substitution (DS) for DEAE and Dodecyl groups were determined using nuclear magnetic resonance of hydrogen (¹H-NMR) and characterized by FTIR. The degree of deacetylation and degree of substitution by DEAE gropus were 94,8% 39,8%, respectively. The obtained hydrophobicized derivatives were substituted with 15% and 39,8% of dodecyl groups as evaluated by ¹H-NMR. The molar masses were determined by using Gel Permeation Chromatography (GPC) and a Mw of 9,2kDa was obtained for deacetylated chitosan. In vitro assays against the fungi Alternaria alternata, Peninicillium expansum and Alternaria solani. At the concentration of 0.5 g.L-1 both, QHdDEAEDOD and QHdDEAE were more effective than commercial and deacetylated chitosans and exhibited inhibitions of 80% against Alternaria alternata and Alternaria solani. Degraded deacetylated chitosan was more efficient in inhibiting the fungal growth of Penicillium expansum and for 0.5 g.L-1 reached around 80% of inhibition. All the derivatives presented cell viabilities higher 70% on 3T3 demonstrated a good cytocompatibility and potential for applications in food preservation.

Full textThe southern states of Brazil are responsible for most of the national wheat production. However, is one of most important factor to produced wheat in southern Brazil, is the excess accumulation of rain and cloudy days, favoring the occurrence and increase diseases intensity. Necrotrophic fungi survive on seed and wheat straw, and among them, the fungus Alternaria alternata is considered one of the causal agents of wheat black point seeds. This disease is commonly found in wheat seeds in Brazil, but no reports of survival, seeds viability, transmission and control measures by seeds the treatment. The aim of this study was to evaluate the presence of A. alternata in wheat seed produced in southern Brazil, to quantify the viability of the fungus in stored seeds, during the inter-harvest period, and to evaluate the fungicides efficacy to Alternaria seed treatment indicated by the Wheat Research Committee to seed treatment to Alternaria control. In 2007 growing seasons, the presence of the fungus A. alternata was quantified in 75 wheat seeds samples at south Brazil, at three regions of Cropping and Use Value (CUV I, II and III). Before seeding them in potato-dextrose-agar media + antibiotic (PDA+A), two hundred seeds per sample were disinfested. The survival of A. alternata was established by the seeds from the 2008 harvest, stored inside propylene bags at a storehouse during 180 days- inter-harvest period being evaluated each 45 days. Three seed lots were analyzed from six wheat cultivars. The efficacy of fungicides to control the A. alternata was determinate by the treatment of the seeds sowed in PDA+A environment, with 400 seeds by treatment. The A. alternata was detected at all of the evaluated samples, with the average incidence of 39.6%, with the incidences of 48.0%, 38.8% and 35.9% respectively for CUV I, CUV II and CUV III regions. At CUV I region, the coolest and moistest region, had the biggest average incidence of the fungus, being significantly different than the others. The viability of A. alternata had average reduction of 49.5% in the mean of wheat cultivars during the inter-harvest, proving that infected seeds are primary inoculum source for the fungus. The fungicide iprodione+triticonazol was the most efficient to control the A. alternata in vitro
Os Estados da região sul do Brasil são responsáveis pela maior parte da produção tritícola nacional. Uma das dificuldades encontradas na produção trigo no sul do Brasil é o excesso de chuva e o acúmulo de dias encobertos, favoráveis à ocorrência e o aumento da intensidade de doenças. Fungos necrotróficos sobrevivem em sementes e na palha do trigo. Dentre eles, o fungo Alternaria alternata, é considerado um dos agentes causais de ponta preta em sementes de trigo. Comumente é detectado em sementes de trigo no Brasil. Porém, sem relatos de sua sobrevivência, viabilidade nas sementes, transmissão para planta e medidas de controle que envolva o tratamento de sementes. O objetivo deste trabalho foi detectar a presença de A. alternata em sementes de trigo produzidas no Sul do Brasil, quantificar a viabilidade do fungo em sementes armazenadas no período de entre safra e testar a eficácia dos fungicidas indicados pela Comissão de Pesquisa de Trigo para tratamento de sementes visando ao controle de A. alternata. A detecção do fungo foi quantificada em 75 amostras de sementes de trigo no Sul do Brasil, na safra agrícola de 2007, em três Regiões de Valor de Cultivo e Uso (VCU I, II e III). Duzentas sementes por amostra foram desinfestadas antes de serem plaqueadas em meio de batata-dextrose-ágar + antibiótico (BDA+A). Sobrevivência de Alternaria alternata foi determinada em sementes colhidas na safras 2008 e armazenadas em sacos de propileno em armazém de alvenaria durante 180 dias, período de entre safra, com avaliações em intervalos de 45 dias. Foram analisados três lotes de sementes de seis cultivares de trigo. A eficácia de fungicidas no controle de A. alternata foi determinada via tratamento de sementes, plaqueadas em meio de BDA+A, com 400 sementes por tratamento. Foi detectado A. alternata em todas as amostras analisadas, com incidência média de 39,6%, sendo 48,0%, 34,8% e 35,9%, respectivamente para Regiões de VCU1, VCU2 e VCU3. Na Região VCU1, considerada mais fria e úmida, houve maior incidência média do fungo, diferindo significativamente das demais. Houve redução média de 49,5% na viabilidade de A. alternata na média das cultivares de trigo durante o período de entre safra, comprovando que as sementes infectadas são fonte de inóculo primário para o fungo. O fungicida iprodiona+triticonazole foi o mais eficiente no controle de A. alternata in vitro

Full textTrichoderma spp. are beneficial fungi that after interacting with the roots improves plant health. Trichoderma spp. produce a variety of MAMPs (Associated Molecular Patterns Micro-organisms) that stimulate the induction of resistance (IR) in plants. The IR by Trichoderma spp. can increase resistance against pathogens mediated through salicylic acid (SA), jasmonic acid (JA), ethylene (ET) responses. These responses regulated are antagonistic between different defense signaling pathways related to JA/ET and AS, sensitizing the plant for enhanced resistance against pathogens. In this study we used a system of three components Solanum lycopersicum, Trichoderma harzianum and Alternaria alternata. The molecular signaling involved during disease suppression by T. harzianum was analyzed in tomato plants cv. Micro-Tom subsequent to infection by A. alternata. The signaling pathways of SA, JA and ET have been explored for IR T. harzianum. In the application of spores of T. harzianum in the roots of tomato plants increased resistance against A. alternata causing leaf spot of culture. The 15 day interval between application of T. harzianum and A. alternata was sufficient to suppress symptoms caused by A. alternata in tomato. The effect of IR elicited by T. harzianum was from his association with the roots of tomato for 15 days. Disease severity was estimated 96 hours after inoculation the challenge pathogen. DNA quantification by qPCR was used to determine differences in biomass of the pathogen. The result of the amount of DNA of A. alternata was 74 times lower in treated plants T. harzianum isolated Th1 compared with plants not treated with Th 1. Treatment with ethylene and methyl jasmonate indicated that the hormones JA and ET pathways are partially related to susceptibility to A. alternata in tomato plants. IR phenotype of tomato plants silenced for gene ET response factor was evaluated. The analysis of the development of symptoms demonstrated that IR by T. harzianum did not involve the response of ET. Finally, gene expression analysis indicates that IR by T. harzianum is controlled by increasing the biosynthesis of JA combined with the decrease in the response signaling SA and ET.

Full textCurrently, the productivity of potato crop (Solanum tuberosum L.) may be reduced due to the occurrence of disease throughout the country, especially due to early blight, caused by Alternaria spp. Historically it was believed that A. solani (AS) was the main cause, most currently was founded others associated species, being predominant today the occurrence of the disease associated with the species A. grandis (AG). The objective of this study was to evaluate the in vitro susceptibility of three isolates of A. grandis and one isolate of A. solani for eleven fungicides widely used in the management of this disease in the field, was evaluated the effect of fungicides on mycelial growth using the fungicidal method that incorporate it in the medium culture. Dilutions of fungicides were performed in sterile distilled water according to the methodology described by Camargo (2013) and Ishizuka (2016). For the fungicidates with active ingredient mixture, was considered the sum of the both concentrations in the calculation.The concentrations of the culture medium were 100, 10, 1, 0.1, 0.01 and 0.001 mg L-1. The doses were adjusted according to the CI50 of each fungicide, was quantified also the inhibition of spore germination, and the fungicides were incorporated into the agar medium - water in discriminatory contractions of 0; 0.1; 1; 10 and 100 mg.L-1 . Then, 24 hours after preparation of the medium the plates were observed under an optical microscope, determining the spore germination observed at 300 units per plate. All tested isolates were highly sensitive to the active ingredients fluazinam, pyrimethanil, procymidone and pyraclostrobin. There was no sensitivity behavioral differences between species of A. solani and A. grandis which may characterize a loss of sensitivity due to the predominance of species of A. grandis associated with early blight in Brazil, was found evidences of insensitivity of isolates of A. grandis and A. solani to the active ingredients Chlorothalonil and Boscalida.

During its evolutionary process, plants have developed biosynthetic pathways by which they produce and accumulate a wide variety of secondary compounds that receive this name because they are not directly involved in the primary processes of plant growth and development. Many of these secondary compounds are responsible for chemical protection of plants against pathogens, herbivores, competing plants, ultraviolet radiation and other environmental stresses. The production of secondary metabolites can vary quantitatively and qualitatively between individuals of the same species, between plants of different species, between organs, between stages of plant development and are under influences of environmental factors. Studies of the biological effects of these compounds represent an important tool for the development of more specific and natural pesticides that are less harmful to the environment, as well as drugs and natural preservatives for food. Myrcia splendens (Sw.) DC. is a tree species found in the Brazilian cerrado and there are few published papers dealing with this species and its secondary metabolites to act as potential natural pesticides, antioxidant and antifungal. The methodology for the extraction of secondary metabolites can determine the composition of extracts and influence the activity of them. In this sense, in the first chapter of this work investigations were carried out on the phytotoxic potential of extracts of M. splendens young leaves from two different extraction methodologies. The 2C extract had the highest overall phytotoxic activity, showing promise in the search phytotoxic substances. In the second chapter, we investigate the phytotoxic potential of extracts and fractions of mature leaves of M. splendens from the two extraction methods carried out and was identified one of the secondary metabolites possibly associated with the phytotoxicity of the most active fractions. 2B fraction had the highest total inhibitory activity and so was fractionated, resulting in 14 fractions, which were also tested in wheat coleoptile. The FM8 and FM9 fractions, which had inhibitory activity and sufficient income, were subjected to HPLC for isolation of major compound. Identification by NMR spectra revealed that the major compound is miricitrin (myricetin-3-O-rhamnoside). In the last chapter, the extracts of mature leaves of M. splendens were tested for their antioxidant potential based on their ability to react with the free radical DPPH (2,2-diphenyl-1-picrylhidrazila) and antifungal potential using the plant pathogenic fungus Alternaria alternata as the target species. Most of the extracts showed a strong antioxidant activity, especially those iv! ! extracted with more polar solvents. On the other hand, only one of the extracts (2B) showed inhibitory activity on mycelial growth of the phytopathogenic fungus tested.
Ao longo do seu processo evolutivo, as plantas desenvolveram rotas biossintéticas pelas quais produzem e acumulam uma imensa variedade de compostos secundários, que recebem este nome por não estarem diretamente envolvidos nos processos primários de crescimento e desenvolvimento vegetal. Muitos desses compostos secundários são responsáveis pela defesa química das plantas contra patógenos, herbívoros, plantas competidoras, radiação ultravioleta e outros estresses ambientais. A produção de metabólitos secundários pode variar quantitativa e qualitativamente entre indivíduos de uma mesma espécie, entre plantas de espécies diferentes, entre órgãos, entre estágios do desenvolvimento vegetal e sobre influências de fatores ambientais. Os estudos realizados sobre os efeitos biológicos destes compostos representam uma importante ferramenta para o desenvolvimento de agroquímicos naturais mais específicos e menos prejudiciais ao ambiente, bem como de medicamentos e conservantes naturais para a indústria alimentícia. Myrcia splendens (Sw.) DC. é uma especie arbórea encontrada no cerrado brasileiro e existem poucos os trabalhos publicados que tratam de investigações do potencial desta espécie e de seus metabólitos secundários para atuarem como pesticidas naturais, antioxidantes e antifúngicos. A metodologia empregada para a extração de metabólitos secundários pode determinar a constituição e rendimento dos extratos e influenciar a atividade dos mesmos. Nesse sentido, no primeiro capítulo deste trabalho foram realizadas investigações sobre o potencial fitotóxico de extratos de folhas jovens de M. splendens oriundos de duas metodologias de extração diferentes. O extrato 2C teve a maior atividade fitotóxica geral, se mostrando promissor na busca de substâncias fitotóxicas. No segundo capítulo, foi investigado o potencial fitotóxico de extratos e frações de folhas maduras de M. splendens das duas metodologias de extração realizadas e foi identificado um dos metabólitos secundários possivelmente associado à fitotoxicidade das frações mais ativas. A fração 2B apresentou a maior atividade inibitória total e por isso foi fracionada, resultando em 14 frações, que também foram testadas em coleóptilos de trigo. As frações FM8 e FM9, que possuíam atividade inibitória e rendimento suficiente, foram submetidas a CLAE para isolamento do composto majoritário. A identificação foi feita por espectros de RMN e revelou que o composto majoritário é a miricitrina (miricetina-3-O-ramnosídeo). No último capítulo, os extratos de folhas maduras de M. splendens foram testadas quanto ao seu potencial antioxidante baseado habilidade de reagir com o radical livre DPPH (2,2-difenil-1-picrilhidrazila) e antifúngico usando como espécie-alvo o fungo fitopatogênico Alternaria ii! ! alternata. A maior parte dos extratos apresentaram atividade antioxidante muito forte, especialmente aqueles extraídos com solventes mais polares. Por outro lado, apenas um dos extratos (2B) apresentou atividade inibitória no crescimento micelial do fitopatógeno testado.

Alternaria brown spot (ABS) disease is caused by a fungus of Alternaria genus, that causes injury and fruit depreciation, leaves fall, drought of pointers and new shoots. The action of the fungus Alternaria alternata f. sp. citri is directly associated by the presence of toxin receptors on susceptible genotypes. The objectives of this study were: (1) to characterize the phenotype of 264 citrus hybrids obtained from crossing LPA Pera de Abril sweet orange (Citrus sinensis L. Osbeck) vs H163, a hybrid between Murcott tangor (C. reticulata x C. sinensis) x Pera sweet orange (C. sinensis), for response to A. alternata, via conidia suspension inoculation of detached young leaves (in vitro) and (2) construction of a linkage map using DArT_seq markers to identify possible QTL (Quantitative Trait Loci) associated to disease resistance. The fungus was isolated from lesions of Murcott tangor fruits with ABS symptoms and inoculated via conidia suspension on detached young leaves (in vitro). Two hundred and thirty five hybrids were evaluated and 70 (30%) showed different levels of disease symptoms after 72 hours of inoculation with the fungus on detached leaves and 165 (70%) were asymptomatic. The integrated linkage map, built with OneMap program, resulted in 726 DArT_seqTM and three SSR molecular markers that covered nine linkage groups, corresponding to the haploid number of chromosomes of the specie. Using the values of AUDPC, seven QTLs associated with resistance to ABS were localized in linkage groups of the integrated map. The integrated linkage map of Pera de Abril sweet orange (LPA) vs. H163 has a high degree of synteny with pseudo-chromosomes of sweet orange (C. sinensis).
A mancha marrom de alternaria (MMA) é uma doença causada por um fungo do gênero Alternaria, que causa lesões e depreciação em frutos de tangerinas, quedas de folhas, seca de ponteiros e de brotações novas. A ação do fungo Alternaria alternata f. sp. citri está diretamente associada à presença de receptores de toxinas em genótipos suscetíveis. Os objetivos do presente estudo foram: (1) caracterizar o fenótipo de 264 híbridos de citros obtidos do cruzamento de LPA (laranja Pera de Abril) (Citrus sinensis L. Osbeck) vs o H163, híbrido de tangor Murcott (C. reticulata x C. sinensis) x laranja Pera (C. sinensis), quanto à resposta à A. alternata e (2) construir um mapa de ligação integrado utilizando marcadores moleculares DArT_seq e localizar QTLs (Quantitative trait loci) associados à resistência à doença. A partir de lesões de tangor Murcott, com sintomas de MMA, foi obtido um isolado do fungo e foram avaliados 235 híbridos após inoculação de suspensão de conídios em folhas jovens destacadas (in vitro). Após 72 horas de inoculação, setenta híbridos (30%) apresentaram diferentes níveis de sintomas da doença e 165 (70%) dos indivíduos foram assintomáticos. O mapa de ligação integrado, construído com o programa OneMap, resultou em 726 marcadores DArT_seqTM e três SSR cobrindo 9 grupos de ligação, correspondendo ao número haplóide de cromossomos da espécie. Os sete QTLs (Quantitative trait Loci) localizados, associados à resistência a A. alternata, foram oriundos de utilização de valores de BLUP (Best Linear Unbiased Prediction), obtidos a partir da AACPD (Àrea Abaixo da Curva de Progresso da Doença). O mapa de ligação gênica integrado de LPA vs H163 apresenta alto grau de sintenia com os pseudo-cromossomos de laranja doce (C. sinensis).

Full textThe search for new active components of microorganisms is one of the areas of greatest investment in developed countries. Among these microorganisms, fungi are distinguished by their ability to produce diverse metabolites for pharmacological purposes. Endophytes are all microorganisms, cultivated or not, that live inside plant tissue, without causing harm to the host and without developing external structures, excluding nodule bacteria and mycorrhiza. Considering the importance of coffee production in the state of Minas Gerais, studies of endophytic fungi in coffee allow for greater understanding of the diversity of these fungi. In this study, a total of 22 endophytic fungi isolated from coffee (Coffea arabica L.) were cultivated in vitro. The screening was conducted using the agar diffusion method against Gram-positive and Gram-negative bacteria and yeast. The most effective isolated fungus among all of the tested microorganisms was the lineage Alternaria alternata, and subsequently, its extract was assayed. The total phenolic content was 3.44 μgGAE/mg of the crude extract. For the antimicrobial activity assays, the minimum inhibitory concentration (MIC) and minimum bactericidal and fungicidal concentration (MBFC) against Staphylococcus aureus, Escherichia coli and Candida albicans were determined. The ranges of MIC values were 50-100 μg/mL for S. aureus and 800-1000 μg/mL for E. coli. The crude extract did not show activity in the tested concentrations for C. albicans. Through these tests, the antioxidant activity of the fungus was assayed through its ability to scavenge DPPH radicals and through β-carotene/linoleic acid system oxidation, both of which were not significant. Antitumour activity was studied by the MTT assay. At a dilution of 400 μg/mL, the extract displayed a cytotoxic activity of approximately 50% towards HeLa cells in vitro. These results indicate that endophytic fungi could be a promising source of antimicrobial compounds and are worthy of further study.

Full textMorphological characteristics, sporulation and pathogenicity of A. solani and A. alternate were estudied. Isolates were obtained from tomato, sweet-pepper, eggplant, potato,"jiló"(Solanum gilo Raddi) and tobacco fungicidal sensibility of these isolates to various fungicidas was also evaluated. For morphological characterization, length and width of the conidium body and length of the conidium beak were measured. The results obtained confirmed the morphological variation between the two species of Altenaria,/i> studied. Influence of NUV, light and darkness, mycelium age and the addition of supplementary water to the medium was studied in relation to sporulation of both species of Alternaria. The best sporulation index for A. solani was obtained with mycelium discs removed from the colony margins, incubation under NUV light conditions and without the addition of supplementary water. For A. alternate only NUV light was enough to induce optimum sporulation. Cross inoculations tests with the various isolates of A. solani and A. alternate which were tested against tomato, pepper and eggplant, showed that all isolates were pathogenic to all hosts, with the exception of the A. solani isolate obtained from eggplant which was not pathogenic to pepper. Seed inoculations 'of tomato with A. solani was positive only for the tomato isolate, during the period of post-emergence of the seedlings. Isolates of A. alternate were not pathogenic to seedlings in pre and post-emergence. However, isolates of A. alternate,/i> from tomato, potato, eggplant and sweet-pepper were most pathogenic, whereas the"jiló"and tobacco isolates were the less agressive ones, when inoculated in semi-matured tomato fruits. With respect to the fungitoxicity of 10 fungicides, at different concentrations, against the isolates of A. solani and A. alternate from tomato, it was found that 100 ppm captafol, propiconazole and phenyltin acetate totally inhibited the mycelial growth of A. alternate, whereas for A. solani only 100 ppm phenyltin acetate was effective.

Alternaria Brown Spot (ABS), caused by Alernaria alternata a fungus which produces a host specific toxin (HST), is a disease of huge importance in mandarin orchards in many producing regions in the world. The selection of varieties resistant to disease is of great economic importance. In this study different hybrids of citrus were evaluated. These hybrids were obtained by directed crosses and preselected by agronomic characteristics that make them potential new varieties of oranges and mandarins, for industry or fresh fruit. The evaluation of these new genotypes is an essential step for the release of new varieties. Seventy-nine hybrids and their parents were subjected to inoculation with an isolated from A. alternaria in detached leaves and plant assays. The severity of disease was measured by the scale described in the literature. The evaluation of Mendelian segregation in hybrids of Murcott tangor and Pera sweet orange, evaluated in greater numbers, allows inference about the genetics of resistance heritability of ABS and to map the genomic region associated with the fungus using an integrated genetic map previously obtained. Different responses were observed after the fungus inoculation. Five hybrids of Willowleaf mandarin like, six hybrids of mandarin like, three hybrids of Murcott like and 21 hybrids of orange like, resistant to ABS, or with few symptoms of the disease were selected. High values of heritability and genetic variability measured indicate that the selection of resistant plants in the progeny can be successful. A resistance gene and molecular markers associated with it were located in the same linkage group 3 of integrated map of Murcott and Pera. The selected genotypes constitute potential material for commercial planting and for the control of disease in the field.
A mancha marrom de alternaria (MMA), causada pelo fungo Alternaria alternata, que produz uma toxina específica ao hospedeiro (Host Specific Toxin, HST) é uma doença de grande importância nos pomares de tangerinas em muitas regiões produtoras no mundo. A seleção de variedades resistentes à doença é de grande importância econômica. Neste trabalho foram avaliados diferentes híbridos de citros, obtidos por cruzamentos dirigidos e pré-selecionados, por apresentarem características agronômicas que os tornam potenciais novas variedades de laranjas e tangerinas, tanto para a citricultura industrial, quanto de mesa. A avaliação destes novos genótipos é uma etapa imprescindível para a liberação de novas variedades. Setenta e nove híbridos juntamente com seus genitores foram submetidos à inoculação com um isolado de A. alternaria em folhas destacadas e em plantas. A severidade da doença foi mensurada através de escala descrita na literatura. A avaliação da segregação mendeliana nos híbridos de tangor Murcott e laranja Pera, avaliados em maior número, permitiu inferir acerca da herdabilidade genética da resistência a MMA e mapear a região genômica associada à resposta das plantas ao fungo em um mapa genético integrado dos genitores previamente obtido. Foram observadas diferentes respostas à inoculação controlada do fungo. Foram selecionados cinco híbridos tipo mexerica, seis híbridos tipo tangerina, três híbridos tipo tangor Murcott e 21 híbridos tipo laranja resistentes a MMA, ou que apresentaram poucos sintomas da doença. Os valores elevados de herdabilidade e variabilidade genotípica mensurados indicam que a seleção de plantas resistentes na progênie pode ser realizada com sucesso. Marcadores ligados à região associada à resposta à infecção com A. alternata foram localizados no grupo de ligação 3 do mapa integrado de tangor Murcott e laranja Pera. Os genótipos selecionados se constituem em materiais potenciais para plantio comercial, visando ao controle da doença no campo.

Viruses of phytophatogens are present in the most diverse groups of hosts, infecting from Spiroplasma to nematode and playing an important role in the ecology of these hosts. Because of the potential as biological control agents, the discoveries about these viruses have increased which encourages the ecological management of plant diseases. In this context, this study aimed: isolate and characterize viruses that infect plant pathogenic fungi (1) and Ralstonia spp. in the Americas (2) and investigate antiviral defense mechanisms in Ralstonia spp. (3). In this thesis, three viruses were characterized at biological and molecular level. Two viruses infect bacteria, Ralstonia spp. and the third infect a fungus, Alternaria alternata. The bacterial virus displayed two propagation modes: pseudolysogenic for the RsIBR1 inovirus (Inoviridae) and lytic for the phiAP1 phikmvvirus (Podoviridae). RsIBR1 has been shown to dramatically change the phenotype of the host, while not causing cell lysis, converting the phytopathogenic R. pseudosolanacearum into a commensal bacterium. PhiAP1 was characterized as a new member of the genus Phikmvvirus, and the presence of exolisinas, in addition to the bactericidal properties and EPS degradation make this virus attractive for future proposals of the bacterial wilt management. In A. alternata, it was characterized Alternaria alternata parititivirus 1 (AtPV1), a new divergent mycovirus related to genus Gammapartitivirus. Its persistent lifestyle prevented to dissect completely the interaction, because in the absence of isogenic lineage, it is unclear whether the viral infection led to a conversion of a pathogen in a epiphytic or still if is responsible for controlling phenotypic plasticity expressed by the host. The presence of canonical CRISPR loci in Ralstonia spp. isolates suggested that, in these hosts, this system could compose the antiviral defense arsenal. However, here was demonstrated that in R. solanacearum, the CRISPR-Cas system is unable to acquire new spacers or interfere with foreign DNA, even in a state of priming. The expression of the Cas genes was not detected; indicating that Cas operon apparently is a suppressed transcriptional unit. Two genes h-ns, expressed in R. solanacearum CFBP2957 are candidates to the putative transcriptional repression of CRISPR. The BIMs (Bacteriophages Insensitive Mutants) who escaped of infection by an alternative strategy to the CRISPR, were less competent, but still permissive to viral adsorption, and were able to survive under high inoculum pressure, which excludes the presence of abortive systems. Viral replication was also not detected and sequencing of two BIMs genome revealed the presence of particular mutations in genes encoding components of the secretory and motility pathways (such as Type II and Type IV), potentially involved in the antiviral resistance. Additional analyzes are being conducted to characterize in detail this defense strategy. In conclusion, the discovery and characterization of viruses infecting phytopathogens in Brazil expand the information on this diversity, little known in America, exposes the biotechnological potential of these natural enemies and reveals the important role of these viruses in the modulation and evolutionary course of their hosts
Vírus que infectam fitopatógenos são encontrados nos mais diversificados grupos de hospedeiros, infectando desde spiroplasmas a nematóides e desempenhando papel relevante na ecologia desses hospedeiros. Graças ao potencial como agentes de controle biológico, as descobertas sobre esses vírus tem aumentado o que amplia as perspectivas do manejo ecológico de doenças de plantas. Neste contexto, este trabalho objetivou: Isolar e caracterizar vírus que infectam fungos fitopatogênicos (1) e os que infectam Ralstonia spp. no continente americano (2) e Investigar mecanismos de defesa antiviral in Ralstonia spp (3). Nesse trabalho, três vírus foram caracterizados a nível biológico e molecular. Dois deles infectam bactérias do complexo Ralstonia spp. e o terceiro infecta um fungo, Alternaria alternata. Os virus de bacterias isolados possuem dois modos de propagação distintos: pseudolisogênico, o inovírus RsIBR1 (Inoviridae) e lítico, o phikmvvirus phiAP1 (Podoviridae). RsIBR1 demonstrou ser capaz de alterar drasticamente o fenótipo do hospedeiro, convertendo a bactéria fitopatogênica R. pseudosolanacearum em uma comensal. PhiAP1 foi caracterizado como um novo membro do gênero Phikmvvirus, e a presença de exolisinas em adição as propriedades bactericida e de degradação de EPS o tornam atrativo para futuras propostas de manejo da murcha-bacteriana. Em A. alternata, foi caracterizado o Alternaria alternata parititivirus 1 (AtPV1), um novo micovírus divergente, relacionado ao gênero Gammapartitivirus. Seu estilo de vida persistente impediu dissecar por completo a interação, pois na ausência da linhagem isogênica, não foi possível confirmar se a infecção viral conduziu a uma conversão de um patógeno em um epifítico ou ainda se é responsável pelo controle da plasticidade fenotípica manifestada pelo hospedeiro. A presença de loci CRISPR canônicos entre isolados de Ralstonia spp. sugeriu que nesses hospedeiros esse sistema pudesse compor o arsenal de defesa antiviral. No entanto, aqui foi demonstrado que em R. solanacearum, o sistema CRISPR é incapaz de adquirir novos spacers ou interferir com DNA invasor, mesmo em estado de priming. A expressão dos genes Cas não foi detectada, indicando que o operon Cas aparentemente é uma unidade transcricional reprimida. Dois genes h-ns, expressos em R. solanacearum CFBP2957 são candidatos na execução da provável repressão do CRISPR. Foram obtidos BIMs (Bacteriophages Insensitive Mutants) que escaparam da infecção por uma estratégia alternativa ao CRISPR. Os BIMs foram menos competentes, mas ainda permissivos a adsorção viral, e foram hábeis para sobreviver sob alta pressão de inóculo, o que exclui a presença de sistemas abortivos. A replicação viral também não foi detectada e o sequenciamento do genoma de dois BIMs revelou a presença de algumas mutações em genes que codificam proteínas componentes das maquinarias de secreção e motilidade bacteriana (Tipo II e Tipo IV), potencialmente envolvidas com o fenótipo da resistência. Análises adicionais estão sendo conduzidas para caracterizar em detalhes essa(s) estratégia(s) de defesa. Em conclusão, a descoberta e caracterização de vírus que infectam fitopatógenos no Brasil, além de ampliar as informações sobre essa diversidade, pouco conhecida na América, expõe o potencial biotecnológico desses inimigos naturais e revela o papel relevante desses vírus na modulação e curso evolutivo dos seus hospedeiros.

Full textThe development and implementation of integrated disease-management system on sunflower (Helianthus annuus) require precise and accurate information on the damage caused by Alternaria leaf spot (Alternaria helianthi). The objectives of this thesis were: to develop and to validate a diagrammatic scale for Alternaria leaf spot; to determine the influence of temperature (15-32.5 °C) and leaf wetness duration (2-24 h) on the monocycle of the disease; and to compare the relationship between variables related to Alternaria leaf spot or to leaf area and yield, in field experiments. The diagrammatic scale with levels representing: 0.03; 0.2; 0.6; 3; 7; 12; 25; 40; and 66% of diseased leaf area was validated by three experienced raters. The scale permitted assessments to be accurate and precise, when drawn or detached leaves were used. The absolute error in estimating disease severity was lower than 13% and the reproduc ibility of assessments was high. In controlled environment studies, relative lesion density and severity were influenced by temperature and leaf wetness duration. The disease was more severe in the temperature of 25 °C. The minimum temperature for disease development was 13.0 °C and the maximum was 35.8 °C. The disease increased with increasing periods of leaf wetness. The estimated minimum temperature for mycelial growth rate was 5.5 °C and the maximum was 32.9 °C. The estimated minimum temperature for conidia germination was 7.9 °C and the maximum was 40.0 °C. Three field experiments were carried out in 1997/1998, 1998/1999 and 1999/2000 to investigate the relationship between severity of Alternaria leaf spot, area under disease progress curve (AUDPC), healthy leaf area index at any given day (HLAI), healthy leaf area duration (HAD), radiation intercepted by healthy leaf area at any given day (HRI), total healthy leaf area absorption (HAA), and yield components of sunflower, sowed in four dates. AUDPC showed relationship with yield in two years, following the negative exponential model. The yield x HAD relationship was linear for each of three trials. Among the integral variables analyzed, the yield x HAA relationship was the best one, fitted by the exponential model. The slope of the yield-severity relationship was stable from R1 growth stage. The yield-HLAI relationship was stable between R1 and R6. The yield-HRI relationship was variable and not stable. The yield-severity relationship in R3 growth stage proved that plants with severity higher than 10% had yield lower than 500 kg/ha, despite the sowing date, which can be used as a damage threshold. Severity can be used as an independent variable to a system of sunflower Alternaria leaf spot management.

Este trabalho teve por objetivos: (i) selecionar genótipos de tangerinas resistentes a infecção por Alternaria alternata, e estabelecer relações entre níveis de severidade da doença entre folhas e frutos, (ii) avaliar a contribuição de metabólitos na resistência de folhas maduras de tangerinas a A. alternata, (iii) selecionar genótipos de tangerina resistentes a infecção por Elsinoe fawcettii. Para tais experimentos foram utilizados 22 genótipos de tangerina pertencentes ao banco de germoplasma da Estação experimental de Citricultura de Bebedouro, Bebedouro-SP. (i) Dentre os materiais genéticos resistentes, foram constatados quatro genótipos de clementinas (Citrus clementina), seis mandarinas, sendo duas pertencentes a C. reticulata, duas a C. tangerina, uma a C. deliciosa e uma a C. nobilis; um tangelo (C. tangerina x C. paradisi); dois híbridos de mandarinas, sendo um resultante do cruzamento entre C. nobilis x C. deliciosa, e o outro de C. clementina x C. reticulata; um híbrido de tangor (C. clementina x C. sinensis) e dois híbridos de satsumas (C. unshiu x C. deliciosa.). Também foi constatado indícios de relação entre severidade de doença em folhas jovens destacadas e incidência natural de doença em frutos. (ii) Ao final dos experimentos, não foi observado influência desses metabólitos na defesa das folhas maduras de tangerina a MMA. (iii) Dente os genótipos avaliados, foram observados 10 materiais genéticos resistentes a verrugose. Dentre estes materiais, observou-se que os genótipos Commune, Caffin, Bruno, Burgess, Peau Lisse, Zanzibar, Beuty of Glen Retreat, Rode King e Encore também se apresentaram resistentes a MMA. Estes materiais são promissores para o cultivo de tangerinas em áreas de ocorrência destas doenças, e poderão contribuir com futuros trabalhos de melhoramento genético para resistência a MMA e verrugose.
The aims of this work were: (i) select tangerine genotypes resistant to Alternaria alternata infection and classify relative degrees of disease severity among leaves and fruits, in order to ultimately control the pathogen; (ii) evaluate metabolites contribuition on mature leaves of tangerines to A. alternata; (iii) select tangerine genotypes resistant to Elsinoe fawcettii. For such experiments 22 tangerines genotypes from germoplasm bank at Estação experimental de Citricultura de Bebedouro, Bebedouro-SP were used. (i) Among the resistant genetic materials, four clementines genotypes (Citrus clementina), six mandarines, being two of C. reticulata, two C. tangerina, one C. deliciosa and one C. nobilis; one tangelo (C. tangerina x C. paradisi); two mandarines hybrids, being one resulted between C. nobilis x C. deliciosa crossing, and other at C. clementina x C. reticulata; one tangor hybrid (C. clementina x C. sinensis) and two satsumas hybrids (C. unshiu x C. deliciosa) were found. A consistent relationship was noted among level of susceptibility in leaves (as evaluated following either natural infection or artificial inoculation), severity and susceptibility of fruit, suggesting a new methodology for diagnosis, identification and selection of Citrus spp. varieties resistant to A. alternata. (ii) At the end of experiments, it was not found any relationship between the presence of these metabolites and resistance to MMA on tangerines mature leaves. (iii) Among the genotypes evaluated, 10 genetic materials resistant to scab were observed. Finally, Commune, Caffin, Bruno, Burgess, Peau Lisse, Zanzibar, Beuty of Glen Retreat, Rode King and Encore cultivars, being resistant to A. alternata and also to E. fawcettii, exhibited good agronomic characteristics and, consequently, show a promising economic exploration.

O tomate industrial destina-se à produção de derivados, como extratos, polpas, molhos e conservas de tomate sem pele. A indústria de processamento de tomates no Brasil apresentou um grande crescimento a partir da década de 1970. O Brasil figura entre os dez maiores produtores de tomate industrial do mundo. A cultura do tomate é afetada por um grande número de doenças causadas por fungos e bactérias. Entre os fungos produtores de toxinas que acometem os tomates industriais destacam-se os do gênero Alternaria. O objetivo do presente trabalho foi avaliar a qualidade físico-química, microbiológica e a ocorrência de micotoxinas de Alternaria alternata em derivados de tomate e em tomates frescos. Derivados de tomate de três marcas (codificadas como A, B, C) e três tipos (extrato, polpa, catchup) foram adquiridos no comércio local do Distrito Federal. Frutos de tomate foram obtidos em uma indústria de processamento do Estado de Goiás e levados para o Laboratório de Fitopatologia da Embrapa Hortaliças para realizar o levantamento da ocorrência de fungos. Os derivados de tomate foram avaliados quanto às características físicas e químicas: teor de água, sólidos totais, sólidos solúveis, pH e acidez titulável. Avaliou-se também a ocorrência de matéria estranha, sujidades, larvas e parasitos, bem como a qualidade da matéria-prima por meio da contagem de fungos filamentosos (Howard) e a adequação, quanto aos parâmetros microbiológicos, à legislação vigente. Os tomates com ocorrência natural de A. alternata e os derivados de tomate foram avaliados quanto à ocorrência das micotoxinas alternariol (AOH) e alternariol monometil éter (AME) por meio de Cromatografia Líquida de Alta Eficiência. Quanto aos parâmetros físico-químicos, a polpa da marca A apresentou teores de sólidos solúveis inferiores a 6% (mínimo estabelecido pela legislação brasileira). Houve diferença significativa (p<0,001) entre todas as marcas de catchup e polpa de tomate avaliadas em relação a teor de água, sólidos totais, sólidos solúveis, pH e acidez titulável. Amostras de catchup da marca C apresentaram os menores teores de sólidos totais (24,28 ± 0,48 g/100g) e sólidos solúveis (27,32 ± 0,12 ºBrix), enquanto a marca A apresentou os maiores (sólidos totais: 30,96 ± 0,30 g/100g e sólidos solúveis: 32,47 ± 0,10 ºBrix). Em relação ao extrato de tomate, não foram observadas diferenças significativas quanto ao teor de água, sólidos totais e acidez titulável. Todos os extratos apresentaram teor de sólidos totais inferior ao mínimo (18%) estabelecido pela legislação vigente. Dos produtos avaliados apenas a marca B de extrato de tomate apresentou ausência de sujidades, larvas e parasitos. Nas demais amostras foram encontrados fragmentos de inseto, pelos de rato e ácaros. Pelos de ratos foram encontrados em amostras de polpa de tomate e extrato de tomate, já fragmentos de insetos foram identificados em todos os produtos de todas as marcas, exceto no extrato da marca B. Apenas o catchup da marca C e a polpa da marca A apresentaram ácaros. A legislação brasileira estabelece, para produtos de tomate, a ausência de sujidades, parasitos e larvas e a ausência de pelos de roedores com base na RDC nº 175 de 2003. Em relação à contagem de filamentos micelianos pelo método Howard todos os produtos avaliados se mostraram adequados, porém o extrato de tomate da marca B apresentou contagem de 12,88 ± 2,04%, o que pode indicar a presença de 1,2% de matéria-prima deteriorada (tomates com mofo). Quanto as análises microbiológicas, todas as amostras avaliadas apresentaram ausência de Salmonella sp. por 25 g, contagem de coliformes a 45 ºC inferior a 3 NMP/g e estafilococos coagulase positiva inferior a 10 UFC/g. Nas amostras de polpa e extrato de tomate de todas as marcas não foram identificados níveis detectáveis das micotoxinas. Nas amostras de catchup da marca A foram encontrados teores de AOH variando de 1,22 a 8,45 μg/g e em amostras de tomates naturalmente contaminadas com A. alternata os teores de AOH variaram de 15,98 a 18,18 μg/g. A micotoxina AME foi identificada apenas em uma amostra de 8 catchup em sache da marca C. Concluiu-se que a maioria dos concentrados de tomate está em desacordo com a legislação vigente quanto ao teor de sólidos totais, sólidos solúveis e apresentam fragmentos de inseto e pelos de ratos. Porém, estão livres dos principais microrganismos patogênicos. Micotoxinas de A. alternata foram identificadas apenas no produto catchup e em duas amostras de frutos tomate naturalmente contaminados com esse fungo. ________________________________________________________________________________ ABSTRACT
Processing tomatoes are intended for the production of tomato derivatives such as pulps, sauces and canned peeled tomatoes. The tomato processing industry had a great increase from the 1970s in Brazil. The country is among the ten largest processing tomato producers of in the world. Many diseases caused by fungi and bacteria are reported affecting this crop. Species of the genus Alternaria are among the most important toxin-producing fungi attacking processing tomatoes. The objective of the present work was to evaluate the physicochemical and microbiological quality as well as the occurrence of toxins produced by Alternaria alternata in tomato products and in fresh tomato fruits. Tomato products from three distinct brands (codified as A, B, and C) and three processing tomato items (extract, pulp, and ketchup) were obtained in the Federal District of Brasilia local market. Fruits were obtained from a processing industry in the State of Goiás and taken to the Plant Pathology Laboratory at Embrapa Vegetable Crops to confirm the presence of fungal infection. Tomato products were evaluated for the following physical and chemical characteristics: water content, total solids, soluble solids, pH, and titratable acidity. Strange materials, dirtiness, larvae and parasites were also evaluated, as well as the raw material quality by counting filamentous fungi (Howard). The adequacy for microbiological parameters, according to legislation was also assessed. Tomato fruits with natural occurrence of A. alternata and tomato derivatives were evaluated via High Performance Liquid Chromatography for the occurrence of two toxins: alternariol (AOH) and alternariol monomethyl ether (AME). The pulp of brand “A” presented soluble solids content lower than 6% (the minimum established by the Brazilian legislation). Significant differences (p<0,001) were detected among all ketchup and pulp samples evaluated for water content, total solids, soluble solids, pH and titratable acidity. Ketchup samples of the brand “C” had the lowest contents of total solids (24.28 ± 0.48 g/100g) and soluble solids (27.32 ± 0.12 ºBrix), whereas the brand “A” displayed the highest values for total solids (30.96 ± 0.30 g/100g) and soluble solids (32.47 ± 0.10 ºBrix). No significant differences were observed in relation to water content, total solids and titratable acidity in all evaluated tomato extracts. However, all tomato extracts had total solids content lower than the minimum (18%) established by the current legislation. From all products analyzed, brand “B” was the only extract that did not present dirtiness, larvae, and parasites. In the other samples, fragments of insects, rodent’s hair, and mites were found. Rodent hairs were found in samples of pulp and in extract samples, but insect fragments were identified in all products of all brands, except in brand B extract. Mites were detected only in the brand “C” Ketchup and in the brand “A” pulp. The Brazilian legislation establishes the absence of dirt, parasites and larvae and the absence of rodent hairs in tomatoes products, according to the RDC#175 (from 2003). Regarding the counting of mycelium filaments by the Howard method, all products evaluated are adequate; however the B brand extract presented counting of 12,88 ± 2,04%, which may indicate the presence of up to 1.2% of deteriorated raw material (moldy tomatoes). Salmonella (25 g samples) coliforms (counting at 45oC lower than 3 NMP/g) and positive coagulase staphylococcus (lower than 10 UFC/g) were not found is all evaluated processing items. Detectable levels of mycotoxins were not identified in the samples of pulp and extract of all brands. Contents of AOH in the samples of brand “A” ketchup ranged from 1.22 to 8.45 μg/g. In tomato fruit samples naturally contaminated with A. alternata the AOH contents ranged from 15.98 to 18.8 μg/g. AME toxin was identified only in one ketchup (sachet) sample of the brand “C”. Based on our results, the majority of the tomato concentrates were not in accordance with the current Brazilian legislation regarding their total solids content and soluble solids as well as in relation to the presence of insect fragments and rodent hairs. Alternaria alternaria toxins were identified only in ketchup and in two samples of tomato fruits naturally contaminated by this fungus

The study of phytopathogenic fungi Alternaria alternata has improved the chemical of this micro-organism. The fungus was grown up in large scale incubated in rice medium for 26 days. After grown period was interrupted an extract of the microorganism was made and fractionated using several chromatographic techniques, obtaining 11 different classes of metabolites which are: alternariol monomethyl éter, alternariol, altertoxin, altenuen, altenusin, uridine, uracila, 5 -inosil, ergosterol, peróxido de ergosterol e triglycerides. Also, a study was made via LC-MS in order to identify some toxins produced by A. alternata like a pathogen of citrus, and it was possible to identify four of these. Other research via LC-MS was the study of co-cultivation of the fungus A. alternata, the causal agent of alternaria brown spot, and Guignardia citricarpa, the causal agent of citrus black spot. In this study was possible to observe the variation in concentration of alternariol and thenuazonic acid, mycotoxins produced by A. alternata when this fungus is in contact with G. citricarpa. The study identification of the volatiles produced by A. Alternaria in different ways contributed significantly to improve the knowledge of their chemistry. Once, the A. alternata is a pathogenic fungus of 'Murcott' a study was performed via scanning electron microscopy (ESEM). Thus it was possible to observe the penetration of this fungus by stomatal cavity of young leaves of citrus. In order to search for natural inhibitors to control the disease, were tested several classes of natural products against the A. alternata, and some alkaloids, a xxvi coumarin, a xanthyletin, and a flavonoid substances showed Higher inhibition of mycelial growth, spore germination and appressorium formation. With the aim to obtain information on natural inhibitors were tested metabolites produced by A. alternata opposite and G. citricarpa, the results indicated that alternariol monomethyl ether and alternariol were substances that had a potential fungistatic against the causal agent of citrus black spot.
O estudo do fungo fitopatógeno Alternaria alternata contribuiu com a química desse micro-organismo. O fungo foi cultivado em larga escala utilizando como meio de cultura arroz por um período de 26 dias. Após esse período o crescimento foi interrompido e foi feito um extrato do microorganismo o qual foi fracionado utilizando várias técnicas cromatográficas obtendo 11 metabólitos de distintas classes sendo eles: alternariol monometil éter, alternariol, altertoxina, altenueno, altenusina, uridina, uracila, 5 -inosil, ergosterol, peróxido de ergosterol e triglicerídeo. Além disso, foi feito um estudo via LC-MS buscando identificar algumas toxinas de citros produzidas por A. alternata, sendo que foi possível a identificação de quatro dessas. Outra pesquisa realizada via LC-MS foi o estudo de co-cultivo entre os fungos A. alternata, o agente causal da mancha marrom de alternaria, e Guignardia citricarpa, o agente causal da mancha preta de citros. Nesse estudo foi possível observar a variação de alternariol e ácido tenuazônico, micotoxinas produzidas por A. alternata, quando este fungo está em contato com outro fitopatógeno. O estudo de identificação dos voláteis produzidos por A. alternaria em diferentes meios contribuiu muito para um maior conhecimento de sua química. Sabendo que A. alternata é um fitopatógeno de tangor Murcott foi realizado um estudo via microscopia eletrônica de varredura (MEV) a qual foi possível observar a penetração desse fungo pela cavidade estomatal de folhas jovens desse citros. Visando a busca de inibidores naturais para o controle da doença, foram ensaiadas diversas classes de produtos naturais frente a A. alternata, sendo que alcalóides e uma cumarina, a xantiletina, foram as substâncias que apresentaram uma maior inibição no crescimento micelial, germinação de esporos e formação de apressórios. Pensando em inibidores naturais, foram ensaiados metabólitos produzidos pelo A. alternata frente ao G. citricarpa, e os resultados obtidos revelaram que alternariol monometil éter e alternariol foram as substâncias que apresentaram um potencial fungistático frente ao agente causal da mancha preta de citros.

Full text[ES] RESUMEN La mancha marrón causada por un patotipo del hongo Alternaria alternata (Fr.) Keissl. es un factor limitante para algunas variedades de mandarina como 'Fortune', 'Nova' y otros híbridos. La información existente sobre la epidemiologia de la enfermedad procede de las regiones húmedas de Florida, y no puede extrapolarse a las zonas semi-áridas. Por ello se inició un estudio para caracterizar la epidemiología de la mancha marrón en las condiciones de España durante dos años consecutivos. Se determinó la presencia de inóculo en las hojas, brotes y frutos de los árboles afectados, en la hojarasca del suelo y las malas hierbas. Para ello se siguió la dinámica de las conidias de Alternaria en el aire con un capturador y se realizaron exposiciones de placas con medio semi-selectivo ARSA. Se estudió también el efecto de diversos factores ambientales en la incidencia de la enfermedad a través de exposición de plantas trampa. Las hojas y brotes afectados constituyeron la principal fuente de inoculo, con 15-76% de aislados patógenos. Se detectaron también aislados patógenos en la hojarasca y en malas hierbas del género Sonchus. El patógeno sobrevivió en hojas inmaduras caídas al suelo hasta los 76 días. Se detectaron conidias de Alternaria en el aíre de forma continua. Se identificaron colonias patógenas en todas las exposiciones de medio semi-selectivo. Las infecciones de las plantas trampa se observaron principalmente en primavera y otoño, en semanas con precipitaciones >2.5 mm y temperatura media >12.5°C. Se estudió la eficiencia infectiva del patógeno mediante la inoculación de plantas de 'Fortune' y 'Nova' bajo diferentes combinaciones de temperatura (8-35°C) y duraciones de humectación foliar (8-48 horas). Se estudió el efecto de los períodos interrumpidos de humectación foliar sobre la infección y también la influencia de la temperatura en la duración del período de incubación. La incidencia de la enfermedad se incrementó con humectaciones foliares superiores a 16 horas, descendió de con temperaturas inferiores a 12ºC y 27ºC, apenas se detectaron infecciones a 35ºC. La incidencia se detuvo con un período seco de una hora en 'Fortune' y dos horas en 'Nova'. La temperatura tuvo un efecto significativo en la duración del periodo de incubación. Se evaluaron los modelos Alter-Rater y un modelo empírico simple (SRS) basado en temperatura y pluviometría con datos obtenidos en exposiciones de plantas trampa durante los años 2011 a 2013. La evaluación de los modelos se realizó mediante análisis de curvas ROC. Ninguno de los modelos evaluados se consideró adecuado para predecir las infecciones en 'Fortune'. En 'Nova', el modelo SRS permitió predecir los periodos semanales de infección con una sensibilidad de 0,93 y una especificidad de 0,84. Este modelo se ha implementado en la web 'gipcitricos' del IVIA para la gestión integrada de la mancha marrón en cítricos. Algunos estudios indican que el riego por inundación podría incrementar la duración de la humectación foliar y el riesgo de infección, pero no existe ninguna referencia específica en cítricos. Se realizó un estudio durante los años 2012 y 2013 en tres parcelas de mandarina con riego por inundación y otras tres con riego localizado. En cada parcela se registraron los valores del contenido de humedad del suelo, duración de humectación foliar, humedad relativa, temperatura y pluviometría. El promedio de la duración de la humectación foliar diaria fue de 4-8 horas y la máxima de 20-23 horas. La dinámica de la duración de la humectación foliar se analizó mediante un modelo de series temporales ARIMA (1,1,1). En los análisis gráficos de los residuos no se detectó ninguna asociación entre los riegos por inundación y la duración de humectación foliar.

En les anàlisis gràfics dels residus no es va detectar cap associació entre els regs per inundació i la duració d'humectació foliar. Els augments en la duració de la humectació foliar es van relacionar únicament amb alguns esdeveniments de pluja.

Full textThe present study aimed to evaluated the mycoflora and occurrence of mycotoxins by high performance liquid chromatography coupled to tandem mass in freshly harvested and stored wheat samples. The results showed the predominance of genus Alternaria in all samples, followed by Epicoccum (12.6%), Fusarium (8.3%) and over 16 other genera of filamentous fungi. The species of Fusarium and Alternaria were identified using molecular methods, and F. graminearum and A. alternata were the most frequent species. Of 70 samples, 69 (98.6%) were contaminated with toxin DON (210-2910 ppb), and 3 (4.3%) samples contaminated with the toxin ZEA (20-30.1 ppb). The level of DON contamination is below of the limits recently established by Brazilian legislation (3000 ppb), but exceed the limits of the European Community (1750 ppb). It is prudent to perform a continuous monitoring of wheat to assess consumer exposure to contaminants and establish guidelines for food safety.

Este trabalho teve como objetivos determinar o efeito da idade da lesão de mancha marrom de Alternária (MMA) na produção conidial em folhas, ramos e frutos, a produção conidial sob diferentes condições de umidade relativa, a influência da aplicação de fungicidas na supressão da produção de conídios, o efeito da idade de maturação de folhas na suscetibilidade ao patógeno e a influência do tamanho de frutos na suscetibilidade à Alternaria alternata. A reação de espécies cítricas à MMA também foi avaliada. A esporulação em folhas teve início cerca de 10 dias depois do aparecimento de lesões de mancha marrom de Alternária, sendo que a maior produção de conídios ocorreu a partir de 20 dias, estendendo-se até 40 dias de idade. A produção de conídios foi superior em folhas do que em ramos ou frutos. A esporulação por unidade de área de lesão em folhas foi maior nos tangelos Minneola e Orlando , em relação ao tangor Murcott . A produção conidial em lesões de folhas foi mais abundante a partir de 85, 92,5, 96 e 100% de umidade relativa. A aplicação de fungicidas do grupo das estrobilurinas e dos cúpricos suprimiu a esporulação em lesões nas folhas por cerca de 14 a 21 dias após a aplicação. Folhas de tangelo Minneola apresentaram maior quantidade e maiores tamanhos de lesões até 20 dias de idade. O híbrido Nova foi o mais resistente entre os materiais genéticos estudados, sendo que folhas a partir de 16 dias de idade...

Application of pyraclostrobin, trifloxyztrobin, azoxystrobin and copper fungicides suppressed the sporulation on leaf lesions for about 14 to 21 days after application. Full textAbstract: The aims of this work were: (i) select tangerine genotypes resistant to Alternaria alternata infection and classify relative degrees of disease severity among leaves and fruits, in order to ultimately control the pathogen; (ii) evaluate metabolites contribuition on mature leaves of tangerines to A. alternata; (iii) select tangerine genotypes resistant to Elsinoe fawcettii. For such experiments 22 tangerines genotypes from germoplasm bank at Estação experimental de Citricultura de Bebedouro, Bebedouro-SP were used. (i) Among the resistant genetic materials, four clementines genotypes (Citrus clementina), six mandarines, being two of C. reticulata, two C. tangerina, one C. deliciosa and one C. nobilis; one tangelo (C. tangerina x C. paradisi); two mandarines hybrids, being one resulted between C. nobilis x C. deliciosa crossing, and other at C. clementina x C. reticulata; one tangor hybrid (C. clementina x C. sinensis) and two satsumas hybrids (C. unshiu x C. deliciosa) were found. A consistent relationship was noted among level of susceptibility in leaves (as evaluated following either natural infection or artificial inoculation), severity and susceptibility of fruit, suggesting a new methodology for diagnosis, identification and selection of Citrus spp. varieties resistant to A. alternata. (ii) At the end of experiments, it was not found any relationship between the presence of these metabolites and resistance to MMA on tangerines mature leaves. (iii) Among the genotypes evaluated, 10 genetic materials resistant to scab were observed. Finally, Commune, Caffin, Bruno, Burgess, Peau Lisse, Zanzibar, Beuty of Glen Retreat, Rode King and Encore cultivars, being resistant to A. alternata and also to E. fawcettii, exhibited good agronomic characteristics and, consequently, show a promising economic exploration.

Visando entender os mecanismos envolvidos na resistência de quatro cultivares de batata à pinta preta, esse trabalho teve por objetivos: 1) quantificar os componentes epidemiológicos da resistência dos cultivares Aracy, Delta, Desirée e Bintje que apresentam diferentes níveis de resistência à pinta preta, e estudar o efeito da idade dos tecidos nesses componentes; 2) realizar a análise histológica do processo infeccioso nesses cultivares, visando identificar possíveis mecanismos estruturais e relacionados com os níveis de resistência dos cultivares, e com a resistência de tecidos jovens; 3) verificar se a expressão diferencial de genes homólogos aos genes que codificam as proteínas PR gluB (glucanase básica de tabaco) e chiB (quitinase básica de feijão) respectivamente, se correlaciona com os níveis de resistência dos cultivares analisadas, e com a maior resistência dos tecidos jovens. Os componentes epidemiológicos da resistência, período de incubação (PI), número de lesões (NL), a severidade, taxa de expansão das lesões (TEL), período latente (PL) e o número de esporos por área foliar afetada (NEAA) foram quantificados nos terços inferior, médio e superior de plantas dos cultivares Aracy (resistente), Delta (resistência intermediária), Desirée(suscetível) e Bintje (suscetível) inoculadas com Alternaria solani numa concentração de 10 3 conídios.mL -1 . No cultivar Aracy constatou-se maior PI, redução do NL, da TEL e conseqüentemente da severidade em todos os terços avaliados. Já o menor NL, TEL, e severidade no terço médio explicam a resistência moderada do cultivar Delta. O cultivar Desirée, classificado como suscetível, apresentou, no terço superior, NL e severidade similar ao cultivar Aracy. O PL e o NEAA não foram adequados para a avaliação da resistência em condições de casa de vegetação e, provavelmente, foram influenciados pela falta de molhamento foliar. Todos os componentes foram influenciados pela idade dos tecidos. Maior PI e menores valores de NL, TEL, severidade e AACPD foram observados no terço superior, comprovando a menor suscetibilidade desses tecidos. Foram efetuados estudos histológicos do processo infeccioso de A. solani nas diferentes cultivares e terços usando a técnica de diafanização de tecidos. Foi quantificado o número de conídios germinados, número de apressórios, número de penetrações diretas e número de sítios de penetração exibindo células com reação de hipersensibilidade (HR). Não se constatou mecanismos de resistência que pudessem ser associados com os níveis de resistência dos cultivares estudados, nem com a maior resistência dos tecidos jovens. Constatou-se uma relação positiva entre o número de sítios de penetração mostrando HR e a resistência do cultivar Aracy. Houve associação desse componente com a resistência dos tecidos jovens. A expressão de genes homólogos à glucanase básica de tabaco (gluB) e à chitinase básica de feijão (chiB) como resposta a infecção de A. solani, foi analisada nos terços inferior e superior dos cultivares Bintje e Aracy, mediante a técnica de "Northern Blotting". Um aumento na expressão de transcritos a gluB e chiB foi constatado no terço inferior do cultivar Aracy, mas não em Bintje. Esse padrão de expressão sugere um possível envolvimento de isoformas destes genes com a resistência de Aracy a A. solani. Essa expressão diferenciada, não foi observada em folhas do terço superior desse cultivar sugerindo um padrão de regulação gênica dependente da idade dos tecidos, e o envolvimento de outros mecanismos bioquímicos na resistência dos tecidos jovens.
The resistance components of four potato cultivars to Alternaria solani were studied in three different levels: 1) quantification of epidemiological components related to resistance in cultivars Aracy, Delta, Desirée, and Bintje, and the effect of age-related resistance in these components; 2) histological analysis of the infectious process in these cultivars, aiming at identifying possible structural mechanisms related to resistance; 3) differential expression of genes homologous to gluB (basic glucanase of tobacco) and chiB (basic chitinase of bean) that encode pathogenesis-related proteins (PR) and the correlation between gene expression levels and cultivar/tissue age resistance levels. The epidemiological components related to resistance, incubation period (PI), number of lesions (NL), early blight severity, lesion expansion rate (TEL), latent period (PL), and the number of spores produced by lesion area (NEAA) were quantified in the lower, middle, and upper parts of cultivars Aracy (resistant), Delta (intermediate resistance), Desirée (susceptible), and Bintje (susceptible) inoculated with a suspension of 103 conidia of A. solani.mL-1. Longer PI, reduced NL, low TEL, and low severity were recorded in Aracy in all parts of the plant. For 'Delta', the reduced NL, low TEL and severity on the middle part of the plant corroborates its intermediate resistance level. Desirée is a susceptible cultivar, but NL and severity on the upper part did not differ from Aracy. The PL and NEAA were not informative when assessing resistance under greenhouse conditions. Probably, PL and NEAA were influenced by the shortage or lack of leaf wetness. All components were influenced by tissue age. Longer PI and smaller values of NL, TEL, and severity were observed in the upper leaves, indicating higher resistance of these tissues. Histological analyses of the infectious process of A. solani in different cultivars were carried out by tissue discoloration. The number of germinated conidia, number of apressoria, number of direct penetrations, and number of penetration sites with cells exhibiting hypersensitive reaction (HR) were quantified. No mechanisms of resistance were associated with cultivar resistance levels or with tissue age. For cultivar Aracy there was a direct relationship between number of penetration sites showing HR and cultivar resistance. The number of sites with HR was also associated with resistance of young tissues. The expression of gluB and chiB in response to A. solani infection was analyzed in the lower and upper parts of 'Bintje' and 'Aracy' plants by Northern blotting analysis. An increase in the level of transcripts related to gluB and chiB was recorded in the lower part of Aracy, but not in Bintje. Differential expression suggests the involvement of isoforms of these genes in Aracy resistance. Differential expression was not observed in leaves of the upper part of Aracy suggesting an age-dependent gene regulation process and/or the involvement of other biochemical mechanisms associated with young tissue high resistance level.

Full textResumo: O presente trabalho visou verificar a ação "in vitro" e "in vivo" de alguns fungicidas e do indutor de resistência acibenzolar-S-methyl (BTH) no controle da pinta preta do tomateiro e seus reflexos na produção. A ação "in vitro" de fungicidas, nas concentrações de 0, 1, 10 e 100 μgmL-1 do ingrediente ativo, foi verificada com base no crescimento micelial, germinação de conídios e efeito anti-esporulante sobre A. solani. Em condições de casa de vegetação foram estudadas as ações: preventiva, residual e curativa de fungicidas em cultivares com diferentes níveis de resistência a pinta preta. Em experimentos de campo, realizados nos municípios paulistas de Sumaré (1999), Socorro (2000) e Bragança Paulista (2000 e 2001), foi avaliada a ação de diferentes grupos fungicidas e acibenzolar-S-methyl (isolado, em mistura com fungicidas e em programas de aplicação) sobre: a severidade da doença (folíolos e hastes); redução de área foliar; incidência de frutos sadios; doentes e danificados pelo sol; massa fresca de frutos sadios, produção em kilos por 10 plantas e porcentagem de frutos grandes, médios e pequenos (Bragança Paulista, 2001). Todos os fungicidas apresentaram ação positiva e diferenciada para todos os parâmetros avaliados, em função das características inerentes de cada princípio ativo. Abstract: The aim of this study was to verify the "in-vitro" and in-vivo effectiveness of various fungicides and plant activator acibenzolar-S-methyl (BTH) in the control of tomato early blight and its effects on yield. The in-vitro effect of the fungicides, in the concentrations 0, 1, 10 and 100 μgmL-1 of the active ingredient, was assessed based on mycelial growth, conidial germination, and the antisporulating effect on A. solani. The various groups of fungicides were studied to compare their preventative, residual and curative efficacy, in greenhouse environment. Field tests were conducted in the state of São Paulo, Brazil, in the countries of Sumaré (1999), Socorro (2000) and Bragança Paulista (2000 and 2001), to compare the effect of various fungicide groups and acibenzolar-S-methyl (alone, mixed with the fungicides, or applied in conjunction with fungicidal treatments) on various parameters: the severity of the disease (leaflets and stems); percentage of leaf drop; relative numbers of healthy fruit as compared to diseased or sun-damaged fruit; yield in kg per 10 plants; and the relative percentages of large, medium and small-sized fruits (Bragança Paulista, 2001). All the fungicides presented a positive effect on all of the parameters evaluated, consistent with the active ingredients of each. The greatest efficacies of control were shown by systemic, more-specific fungicides. In regard to mycelial growth and conidial germination, the greatest inhibition percentages were obtained with metconazole, tebuconazole, difenoconazole, iprodione, cyprodinil, procymidone, prochloraz, fluazinam and pyrimethanil. The fungicides kresoxim methyl, azoxystrobin, pyraclostrobin+methiram, fenamidone+chlorothalonil and famoxadone+mancozeb presented an intermediate degree of inhibition on the mycelial growth and complete inhibition of conidial germination at concentrations of 1 μgmL-1 and above...
