Abstract:
In 1935, Alberto Sartoris transformed an old mill into a private club for artists: le Cercle de l’Ermitage. The young architect conceptualized the space as a manifesto of rationalistic architecture. The resolutely modernist choice of the intervention contrasted with the rural and bucolic existing building. In 1971, the work disappeared under fake rustic decoration and everybody considered it lost, only remaining one of the most famous axonometries of the Italian master. Forty-five years later, the work reappears miraculously while the space is under transformation into a private residence. The demolition work has revealed an unexpected amount of original elements, which will allow a faithful and rigorous restoration of the Cercle de l’Ermitage.

Abstract Introduction. The two ETS transcription factors ETS1 and FLI1 are co-mapped in the 11q24.3 region recurrently gained in up to 25% of diffuse large B cell lymphomas (DLBCL) (Bonetti et al, 2013). The two proteins largely co-regulate a series of genes involved in B-cell signaling, differentiation and cell cycle (Priebe et al, 2020; Sartori et al, 2021). While FLI1 is expressed at a higher level in DLBCL of the germinal center B-cell (GCB) type than in the activated B-cell-like (ABC) DLBCL, ETS1 is more expressed in the latter subgroup. We and others have reported preclinical anti-tumor activity in lymphomas and other tumors with small molecules blocking the binding of ETS factors and RNA helicases (Erkizan et al, 2009; Spriano et al, 2019), and encouraging clinical data are coming from the first trials enrolling Ewing Sarcoma patients (Ludwig et al, 2020). Here, proteomic experiments were performed to investigate ETS1 interactome in DLBCL. Methods. To investigate interaction partners, protein precipitation was performed using streptavidin beads to pull down strep-tagged ETS1 in DLBCL ABC cell line, HBL-1, followed by Liquid Chromatography tandem Mass Spectrometry (LC-MS/MS). Shotgun proteomics analysis identified biologically relevant ETS1 protein interactors, with a spectral count above four, that were validated by normal and reverse co-immunoprecipitation experiments. Transcriptome analysis was done via RNASeq after DDX21 siRNAs silencing in four DLBCL cell lines, two derived from ABC (HBL1 and U2932) and two from GCB (OCI-Ly1 and VAL). Results. Identified ETS1 interactors in the ABC DLBCL HBL-1 included proteins related to RNA processing, more specifically in spliceosome (NOP56 and ALYREF) and in ribosome biogenesis (SF3B1 and DDX21). Among these, we focused on the novel interactor DDX21, an RNA helicase also regulated by FLI1 (Sartori et al, 2021). DDX21 was found differentially expressed between GCB and ABC DLBCL, with higher expression in the latter (P<0.001) (GSE98588: n= 117 cases, phs001444.v2.p1: n= 432 cases, GSE95013: n= 33 cases, and GSE10846, n= 350 cases). When we silenced DDX21 with siRNAs, toxicity was seen in both ABC and GCB cell lines. To better characterize the role of DDX21 as a transcriptional factor in DLBCL and to further investigate its link with ETS1 and FLI1 we obtained RNASeq transcriptome after DDX21 silencing. Transcriptional profiling and functional annotation of transcripts regulated by DDX21 identified genes coding proteins involved in cell cycle (FDR <0.001), ribosomes (FDR <0.001) and spliceosome (FDR <0.001).Conclusions. ETS1 interacts with proteins involved in spliceosome and in ribosome biogenesis, including DDX21. More expressed in ABC than GCB DLBCL, DDX21 appears essential for the survival of lymphoma cells by regulating cell cycle and RNA processing. Abstract:
Abstract The majority of metastatic colorectal cancers (mCRC) are mismatch repair (MMR) proficient (MMRp) and unresponsive to immunotherapy, while MMR deficient (MMRd) tumors often respond to immune checkpoint blockade (ICB). We previously reported that treatment of CRC preclinical models with temozolomide (TMZ) leads to MMR deficiency, increased tumor mutational burden (TMB) and, sensitization to immunotherapy. To clinically translate these findings, we designed the ARETHUSA clinical trial whereby O6-Methylguanine-DNA-methyltransferase (MGMT) deficient, MMR proficient and KRAS mutant mCRC patients receive priming therapy with TMZ. Analysis of solid tissue biopsies and circulating tumor DNA (ctDNA) obtained after TMZ treatment revealed the emergence of TMZ mutational signature, alterations in MMR genes and increased TMB in 14 out of 16 patients. Genetic mutations induced by TMZ were dose-dependent and multiple alterations in the nucleotide context favored by the TMZ signature emerged in MMR genes such as the MSH6 T1219I variant which was detected in ctDNA and tissue of 13/14 (93%) of the cases. A subset of the patients whose tumors after TMZ priming displayed the MSH6 mutation, the TMZ mutational signature and increased TMB, achieved disease stabilization upon pembrolizumab treatment. Overall, we provide proof-of-concept that treatment of MGMT deficient/MMR proficient KRAS mutant mCRCs with TMZ can be tracked by mutational signature analysis and lead to inactivation of the MMR pathway, emergence of the TMZ mutational signature, TMB increase, and, in some cases, to disease stabilization during ICB. Young America: The Daguerreotypes of Southworth & Hawes (2005) is a monumental exhibition catalogue showcasing the work of Albert Sands Southworth and Josiah Johnson Hawes. Together the partners established a renowned daguerreotype studio in mid-nineteenth-century Boston that catered to the city’s bourgeoisie. This paper seeks to unravel the mystery of dozens of daguerreotypes found in Young America, in which elite Boston women appear to be nearly nude. The unidentified women stand in stark contrast to the carefully concealed bodies of Southworth & Hawes’ other female subjects. Why would they expose themselves in such a manner before the camera’s lens? This paper attributes the women’s state of (un)dress to their deliberate emulation of two sculptures in the classical tradition: Clytie, a marble bust dating to antiquity, and Proserpine, a mid-nineteenth-century marble bust by American neoclassical sculptor Hiram Powers. This argument first reveals how a general “classical statue” aesthetic prevailed for women’s deportment in antebellum America, then demonstrates that the busts of Clytie and Proserpine had special significance as icons of white, elite female beauty in the period. Next, this paper makes the case that Southworth & Hawes devised a special style of photography deriving from their own daguerreotypes of the two statues, in which the women’s off-shoulder drapery was deliberately obscured allowing their female clientele to pose in the guise of these famous statues. The paper concludes by arguing that the women shown in these images could pose in this style without contravening societal norms, as these mythological figures were construed by women and men in the period to reflect the central precepts of the mid-nineteenth-century “Cult of True Womanhood.” Moreover, the busts offered sartorial models that reinforced standards of female dress as they related to class and privilege. By baring their flawless, white skin, however, the women positioned themselves at the crux of contentious beliefs about race in a deeply divided nation prior to the American Civil War.

Esta edición de Encrucijada Americana busca aportar a la discusión académica presentando una selección de artículos elaborados por profesores, investigadores y estudiantes de postgrado de Chile, Uruguay y México. Los trabajos han sido agrupados en tres categorías: I. Relaciones internacionales e integración regional; II. Democracia y sistema político; III. Gobiernos locales, comunidades y actores sociales. En la primera sección se incluyen tres artículos que analizan las relaciones internacionales en Latinoamérica, a nivel regional dos de ellos, y bilateral el restante. El primero, titulado UNASUR y la integración latinoamericana: propuesta de un nuevo modelo del regionalismo post-liberal, explora el nuevo contexto regional y expone algunas claves para entender y proyectar el proceso de integración actualmente en curso, poniendo el foco en la experiencia de la Unión de Naciones Suramericanas (UNASUR). Fue elaborado por un grupo de estudiantes y académicos de la carrera de Ciencia Política y Relaciones Internacionales de la Universidad Alberto Hurtado, coordinados por Isabel Rodríguez y Cristóbal Bywaters. El tercero, La relación bilateral Chile-Perú: un caso de visiones geopolíticas opuestas, analiza, desde la geopolítica crítica, los divergentes puntos de vista con que Chile y Perú afrontan discursivamente la relación bilateral, caracterizada en buena cuenta por el alto nivel de competencia entre las partes. El autor, Lester Cabrera, es Profesor de Ciencia Política y Relaciones Internacionales de la Universidad San Sebastián, y Coordinador Académico del Programa de Magíster en Seguridad y Defensa de la ANEPE-UdeC. La segunda sección contiene dos artículos, ambos relacionados con el funcionamiento de la democracia y el sistema político, en Uruguay y en Chile. El primero de ellos, Estado de Derecho, democracia y gobernabilidad en Uruguay, analiza el funcionamiento de la democracia y las características del Estado de Derecho en aquel país, poniendo especial atención al período de gobierno del Frente Amplio, entre el 2005 y el 2010. El autor, Carlos Moreira, es politólogo uruguayo, y miembro de la Red de Estudios de Calidad de la Democracia en América Latina. El segundo, Nominando candidatos con “encuestas en mano”: el uso de las encuestas como mecanismo de selección de candidatos, examina el escenario político-electoral chileno entre el 2005 y el 2009, indagando en aquella particular fórmula, empleada por los principales conglomerados políticos del país (Concertación y Alianza por Chile) para elegir a los candidatos a las elecciones parlamentarias, presidenciales y municipales realizadas en aquel período. Tal mecanismo “nuevo” es contrastado con lo que los autores denominan “métodos tradicionales de selección de candidatos”. El artículo fue co-elaborado por Andrés Dockendorff, Alejandro Salinas y Pedro Figueroa, todos ellos investigadores externos del Instituto de Asuntos Públicos de la Universidad de Chile. Finalmente, en la tercera sección se incluyen tres artículos, relacionados entre sí por la preeminencia que los autores confieren a los gobiernos locales, municipios y actores sociales en el análisis de las problemáticas que estudian. El primero de ellos, Gobierno local: una lectura de municipios y comunidades desde la teoría democrática, explora – en palabras del propio autor- “los conceptos de municipio y comunidad, como elementos integrantes de la idea de gobierno local”, empleando como base interpretativa los postulados de Robert Dahl y Giovanni Sartori. El escrito fue elaborado por Roberto Lagos, alumno del Magíster en Sociología de la Universidad Alberto Hurtado. El segundo, El Municipio en Chiapas en la coyuntura zapatista: un actor inesperado, analiza el rol que desempeñó y desempeña el municipio –como institución- en el contexto que siguió a la confrontación armada, después de 1994, en Chiapas. La autora, Araceli Burguete, es mexicana, e integrante del Centro de Investigaciones y Estudios Superiores en Antropología Social (CIESAS), Unidad Sureste (Chiapas, México). Por último, en el tercer artículo, La participación de las Comunidades Eclesiales de Base (CEBs) en la regeneración del tejido social popular brasileño durante la dictadura militar 1964-1985, se examina el papel que le correspondió a la Iglesia Católica brasileña, y en particular a las Comunidades Eclesiales de Base, en la reconstrucción del tejido social popular, especialmente en el complejo período de 1964 a 1985, marcado por la violencia institucionalizada practicada por los gobiernos militares. El autor del artículo, Fabián Bustamante, es estudiante del Magíster en Historia de Chile, impartido por la Universidad de Santiago. Esperamos que esta edición de Encrucijada Americana contribuya a enriquecer el debate académico en torno a los temas estudiados, y que tal debate posibilite e impulse transformaciones que, en definitiva, vayan en beneficio de los pueblos de nuestra América.

Abstract:
Abstract Introduction: Mismatch repair (MMR) protein expression in colorectal cancer (CRC) is usually homogeneously retained or lost. However, rare CRC lesions may show a heterogeneous pattern of MMR protein expression. We aimed to evaluate the molecular effect of a heterogeneous expression of the MMR proteins in a complex context of concomitant loss of one of the other effectors of the DNA repair mechanism. Experimental procedures: We evaluated MMR protein expression (by immunohistochemistry -IHC- for MLH1, MSH2, MSH6 and PMS2) in 200 CRCs, identifying 3 groups with proficient (MMRp), deficient (MMRd) and heterogeneous (MMRh) MMR. MMRh tumors were micro-dissected based on the expression of the heterogeneous marker, DNA was extracted and subjected to targeted sequencing (TSO500 panel, Illumina). RNA was purified from bulk tumors of all MMRh cases and in a control series of 10 MMRp and 10 MMRd CRCs and analyzed using the IO360 panel (NanoString). The degree of tumor infiltrating lymphocytes (TILs) was calculated on H&E slides and on IHC sections stained with antibodies raised against CD8. Results: Twenty-nine out of 200 cases were MMRd (14.5%). Nine cases (4.5%) showed a heterogeneous pattern of MMR expression, with 6 tumors harboring concomitant loss of one of the other MMR proteins, resulting in a microsatellite instable (MSI) phenotype. Five out of the 9 cases were suitable for separate mutational analysis of IHC-positive and IHC-(double)negative components of the tumor. Both components of all the lesions exhibited a high tumor mutation burden (mean TMB: 59 mut/Mb) in line with the MSI status, nevertheless a significant increase in TMB in the double-negative components was observed (mean TMB: neg. 67 mut/Mb vs pos. 53 mut/Mb), due to a higher number of sub-clonal, non-synonymous variants compared to the positive component. Comparative gene expression analyses between MMRd, MMRp and MMRh CRCs highlighted differential gene expression patterns, with MMRh tumors displaying a strong activation of angiogenesis, MAPK and PI3K-AKT axes. Moreover, these tumors showed the highest number of TILs, characterized by a substantial population of exhausted CD8+ lymphocytes. Conclusions: We describe a unique subgroup of CRCs showing heterogeneous expression of MMR proteins, in a background of concomitant loss of one of the other markers. This heterogeneity is associated with i) an increase of TMB and of unstable loci in the negative component, with additional markedly sub-clonal variants, ii) a high number of TILs, mostly exhausted CD8+ lymphocytes, and iii) a differential activation of signaling pathways. Taken together these data provide indirect evidence that heterogeneous loss of MMR proteins may impact on the biology of CRCs. Whether this pattern may influence response to immune checkpoint inhibition remains to be determined. La arquitectura moderna de Brasil va incorporando a partir del final de los años treinta varios conceptos del Movimiento Moderno europeo, pero, como escribe Alberto Sartoris, sin hacerlo de manera directa: sino reinterpretándolos, traduciéndolos y adaptándolos a las especificas condiciones climáticas y paisajísticas del País (SARTORIS, 2005) El texto describe la apropiación de las nuevas teorías a partir del análisis de algunos proyectos modélicos en los cuales aparece contundente la concatenación entre espacialidad, paisaje natural y paisajismo. Las obras, realizadas por equipos multidisciplinarios guiados por el maestro Niemeyer, demuestran como la nueva espacialidad brasileña surge de la fusión entre elementos arquitectónicos, artísticos, naturales y estructurales, con el uso de reglas morfológicas de tipo similar en los diferentes aspectos de los proyectos, como acontece por ejemplo en la iglesia Pampulha, donde arquitecto, artista, ingeniero y paisajista -Oscar Niemeyer, Candido Portinari, Joaquim Cardoso y Roberto Burle Marx- operan en comunión aplicando criterios estéticos de naturaleza similar. La revolución brasileña es la asimilación de un método de trabajo que se fundamenta en el dialogo entre maestros de diferentes saberes, en el cual el arquitecto tiene el importante papel de “orquestador” (LE CORBUSIER, 1995) y como objetivo la construcción holística de un nuevo paisaje moderno, tropical e integrado.

La arquitectura, también la de la vanguardia, se debate entre la referencia a episodios pasados y la invención de estrategias. Esta investigación es el resultado de un viaje alrededor de las obras de la primera etapa de Frank Gehry en California. En el mismo, se suceden múltiples encuentros, entre otros, con la arquitectura de un maestro moderno, Frank Lloyd Wright, y de un crítico revisor, Charles Moore, de los que se derivan inesperadas coincidencias. La evolución progresiva del lenguaje arquitectónico de Gehry hacia la fragmentación está precedida por distorsiones volumétricas e investigaciones geométricas que, previamente a las vanguardias contemporáneas, cuestionan la unidad y coherencia del proyecto en favor de mecanismos como la agregación simbólica, la dislocación volumétrica o la banalización y vulgarización del vocabulario moderno. En el núcleo de la propia tradición moderna, de la mano de Alberto Sartoris, se anticipan investigaciones sobre la desestabilización del concepto de jerarquía, lo que confirma la fragilidad de las categorizaciones. En el trazado de estos acuerdos insospechados con búsquedas pretéritas no se pretende verificar ni la repetición ni la copia. Pero sí contribuir a diluir, de una vez por todas, la especulación sobre su invención.
