Abstract:
The prime objective of this study was to research the possibilities of synchronizing information such as episodic progression and tracking as well as subscriptions in regards to audio podcasting channels , and to do this without the need of consideration to the underlying platforms. An application providing podcatching capabilities is developed and its features determined through the process of comparing similar existing solutions. Based on this comparison a model is shaped and used as a tool of measuring podcatching capabilities of any media playing software, including the very solution resulting from this study. Supporting questions such as how to find and subscribe to podcast channels will be answered through the analysis of syndication feeds , their structure and how its contents may not only be read but also stored to best accomodate requirements deemed as necessary.

Saunders, Janet Catherine. "An in vivo platform for identifying protein aggregation inhibitors." Thesis, University of Leeds, 2014. http://etheses.whiterose.ac.uk/7622/.

Full text
Abstract:
Protein aggregation is the basis of a vast array of diseases and one of the most expensive problems to overcome during production of biopharmaceuticals. The tendency of proteins to aggregate ensures that demanding purification techniques are a pre-requisite to in vitro analysis of aggregation mechanisms and screening for aggregation inhibitors. In this thesis, a powerful new system was developed to identify aggregation-prone sequences in vivo, and to screen for inhibitors of aggregation. The screen is based on a β-lactamase-tripartite fusion system, where the minimal inhibitory concentration of antibiotic, conferred by the β-lactamase enzyme, is used to evaluate the level of test protein aggregation. Using this in vivo system, the aggregation propensity of the two disease-related proteins human islet amyloid polypeptide (hIAPP) and amyloid beta peptide (Aβ) was found to be significantly higher than non-aggregating controls. Importantly, this system provides a new approach to assess aggregation-propensity without the need for purified protein. The system was used to screen small molecules for their aggregation-inhibiting properties against hIAPP. It was found that many results correlated well with the published literature on these molecules, but notably, a number did not. In vitro analysis of hIAPP aggregation in the presence of these molecules validated the results from the in vivo assay, refuting a number of published studies and confirming the power of the tripartite system for identifying aggregation inhibitors. Finally, the system was used to differentiate between an aggregating and non-aggregating human VH antibody domain, to demonstrate the application of the screen to biopharmaceuticals. The in vivo system successfully identified the aggregating test protein, wherein the addition of excipients prevented its aggregation in vivo in a titratable manner. Overall the work presented herein describes a novel, and experimentally simple, in vivo system that provides rapid and accurate analysis of protein aggregation and its inhibition.

Olurin, Olumuyiwa. "Policy Merger System for P3P in a Cloud Aggregation Platform." Thèse, Université d'Ottawa / University of Ottawa, 2013. http://hdl.handle.net/10393/23624.

Full text
Abstract:
The need for aggregating privacy policies is present in a variety of application areas today. In traditional client/server models, websites host services along with their policies in different private domains. However, in a cloud-computing platform where aggregators can merge multiple services, users often face complex decisions in terms of choosing the right services from service providers. In this computing paradigm, the ability to aggregate policies as well as services will be useful and more effective for users that are privacy conscious regarding their sensitive or personal information. This thesis studies the problems associated with the Platform for Privacy Preference (P3P) language, and the present issues with communicating and understanding the P3P language. Furthermore, it discusses some efficient strategies and algorithms for the matching and the merging processes, and then elaborates on some privacy policy conflicts that may occur after merging policies. Lastly, the thesis presents a tool for matching and merging P3P policies. If successful, the merge produces an aggregate policy that is consistent with the policies of all participating service providers.

Abstract:
The global cyber-infrastructure comprehends a growing number of resources, spanning over several abstraction layers. These resources, which can include wireless sensor devices or mobile networks, share common requirements such as richer inter-connection capabilities and increasing data consumption demands. Additionally, the service model is now widely spread, supporting the development and execution of distributed applications. In this context, new challenges are emerging around the “big data” topic. These challenges include service access optimizations, such as data-access context sharing, more efficient data filtering/ aggregation mechanisms, and adaptable service access models that can respond to context changes. The service access characteristics can be aggregated to capture specific interaction models. Moreover, ubiquitous service access is a growing requirement, particularly regarding mobile clients such as tablets and smartphones. The Session concept aggregates the service access characteristics, creating specific interaction models, which can then be re-used in similar contexts. Existing Session abstraction implementations also allow dynamic reconfigurations of these interaction models, so that the model can adapt to context changes, based on service, client or underlying communication medium variables. Cloud computing on the other hand, provides ubiquitous access, along with large data persistence and processing services. This thesis proposes a Session abstraction implementation, deployed on a Cloud platform, in the form of a middleware. This middleware captures rich/dynamic interaction models between users with similar interests, and provides a generic mechanism for interacting with datasources based on multiple protocols. Such an abstraction contextualizes service/users interactions, can be reused by other users in similar contexts. This Session implementation also permits data persistence by saving all data in transit in a Cloud-based repository, The aforementioned middleware delivers richer datasource-access interaction models, dynamic reconfigurations, and allows the integration of heterogenous datasources. The solution also provides ubiquitous access, allowing client connections from standard Web browsers or Android based mobile devices.

Du, Qianzhou. "Extracting the Wisdom of Crowds From Crowdsourcing Platforms." Diss., Virginia Tech, 2019. http://hdl.handle.net/10919/102032.

Full text
Abstract:
Enabled by the wave of online crowdsourcing activities, extracting the Wisdom of Crowds (WoC) has become an emerging research area, one that is used to aggregate judgments, opinions, or predictions from a large group of individuals for improved decision making. However, existing literature mostly focuses on eliciting the wisdom of crowds in an offline context—without tapping into the vast amount of data available on online crowdsourcing platforms. To extract WoC from participants on online platforms, there exist at least three challenges, including social influence, suboptimal aggregation strategies, and data sparsity. This dissertation aims to answer the research question of how to effectively extract WoC from crowdsourcing platforms for the purpose of making better decisions. In the first study, I designed a new opinions aggregation method, Social Crowd IQ (SCIQ), using a time-based decay function to eliminate the impact of social influence on crowd performance. In the second study, I proposed a statistical learning method, CrowdBoosting, instead of a heuristic-based method, to improve the quality of crowd wisdom. In the third study, I designed a new method, Collective Persuasibility, to solve the challenge of data sparsity in a crowdfunding platform by inferring the backers' preferences and persuasibility. My work shows that people can obtain business benefits from crowd wisdom, and it provides several effective methods to extract wisdom from online crowdsourcing platforms, such as StockTwits, Good Judgment Open, and Kickstarter.

Digital Health-Data is being collected at unprecedented rates today as biometric micro sensors continue to diffuse into our lives in the form of smart devices, wearables, and even clothing. From this data, we hope to learn more about preventative health so that we can spend less money on the doctor. To help users aggregate this perpetual growth of biometric "big" data, Apple HealthKit, Google Fit, and Samsung SAMI were each created with the hope of becoming the dominant design platform for Digital Health-Data. The research for this paper consists of citings from technology strategy literature and relevant journalism articles regarding recent and past developments that pertain to the wearables market and the digitization movement of electronic health records (EHR) and protected health information (PHI) along with their rules and regulations. The culmination of these citations will contribute to my hypothesis where the analysis will attempt to support my recommendations for Apple, Google, and Samsung. The ending chapters will encompass discussions around network effects and costs associated with multi-homing user data across multiple platforms and finally ending with my conclusion based on my hypothesis.

Abstract:
This essay explores the early experiences of Bangladeshi blue-collar service workers in digitalising their livelihoods. It is a qualitative study that surveys and interviews service-providers in Dhaka who use the business aggregator platform Sheba.xyz, an online service marketplace, and seeks to understand what brought these self-employed micro-entrepreneurs, previously outside the digital economy, to adopt an ICT-enabled solution. The study is guided by Diffusion of Innovations (DOI) theory, one of the historically dominant paradigms in the field of communication for development (C4D). The overarching research question is, “Why did blue-collar service-providers in Dhaka adopt and use a digital business aggregator platform?” The aim is to explore what motivated/discouraged and enabled/hindered innovation adoption among a group of users previously marginalised from digital and financial inclusion. The findings suggest that adoption of Sheba.xyz among service-providers was not driven so much by a desire to digitalise one’s business per se, and as a means of mitigating a previous inability to do so. Rather, the factors that emerge from the qualitative data are other perceived relative advantages of the solution – of increase in customers, income, and opportunity. Survey respondents and interviewees also displayed strong affiliation with, and trust in, the platform provider; an alertness for fair treatment; and a drive to prosper, suggesting that they embraced a comprehensive concept and altered life situation where belonging, respect, and self-fulfilment matters, rather than narrowly adopted a new mobile application.

Abstract:
The traditional electricity market, holding centralized authority over consumers, is no longer adequate seeing a shift towards a more electrified, decentralized, and digitalized society. Increased energy prices, raising concerns about climate change, and tightening governmental regulations have resulted in that an extensive diffusion of renewable energy sources has evolved. This development is expected to change the structure of the sector, despite that an appropriate market design that can deal with these remains to be identified. The purpose of this study was to investigate how a business model of a digital platform, managing energy trading within a local community could be designed. This study contributes to a new dimension of energy transitions within a Multi-Level Perspective by studying a particular field of the transition in terms of flexibility market platforms. The rising need for flexible solutions, making the consumer a prosumer, and enabling shared energy through a digital platform involves uncertainty and challenges, where a suitable business model linking new technology to the emerging market needs to be defined. Despite the novelty of the research field of local energy markets, the aim of investigating business model designs for a local energy market platform has been reached through an exploratory case study and integration of theories from several fields. This study makes an analytical contribution of investigating five pioneering projects, all developing digital platforms enabling integration of flexibility into the electricity market. This further contributes to the design-implementation gap of theories when developing a local energy market, by suggesting the most vital parameters to take into account. Based on the findings, a suggestion on a suitable business model design and a corresponding market design was developed. The main objective of the proposed market design is to serve as a basis to bring forward flexibility available from prosumers and their controllable demand and supply arrangement, including renewable energy technology generation and storage devices. The intention is to maintain a balanced and transparent distribution network at the lowest possible costs, while, at the same time functioning as reserve storage towards the main grid, reducing the risk of capacity shortage. Additional insights were raised that can be helpful in the evaluation of utilizing flexibility energy assets before making grid investments, following the recently presented recommendation of the EU's Clean Energy package.

Modern people are familiar to use search engines to solve their daily problems. Google provides well search services; however, search results are optimized for general purpose not for user’s requirements about specific domain problems. Also, Google search results present relevant web links, users frequently spend long time to navigate possible links for seeking adequate information. Such problems also happen in the IT domain. People having IT troubles always wait for IT staffs to solve problems. When IT staffs cannot solve hard problems, they are hard to find solutions from Web search engines on time. To improve IT services, we develop the itQA social platform that integrates social services for general users and IT staffs. Based on the platform, IT questions are aggregated into the same pool. Also, itQA integrates Web Crawler and Search Engine techniques that aggregate IT information into the platform by crawling and indexing QA contents from popular IT websites. In this way, all IT QA contents can be aggregated into the same platform so that accumulated user logs and feedbacks can be analyzed to gradually improve the quality of the itQA platform.

Alzheimer Disease is the most prevalent dementia. Abnormal folding of Tau leads to generation of aggregated Tau species like oligomers and further NFTs. Toxic Tau species were suggested to spread trough human brain and damage the neuronal circuit. To halt disease progression a noteworthy development in therapies based on phosphorylation modulation, Microtubule stabilization and enchantment of Tau clearance have been done. To develop therapies against Tau aggregation and aggregates clearance, the build up of models that recapitulates Tau pathology are required. In the following work the “seeding” strategy was used to achieve aggregation of hTauP301L. We worked with two in-vitro seeding models – cellular and primary neuronal- where phosphorylated insoluble hTauP301L is present. GSK3β was shown to increase ser202/thr205 phosphorylation in soluble and insoluble hTauP301L expressed in QBI but was not sufficient to induce aggregation alone. In primary neuronal seeding model tau aggregation and phosphorylation was increased over-time. Hsp90 inhibition was found to reduce total and AT8 immuno-reactive hTauP301L levels, emerging as a potential drug for Tau clearance. With this work we provide two models to study the mechanisms behind tau nucleation, aggregation, and trans-synaptic spreading. These models are valuable tools for the development of drugs for AD and Tauopathies.

Parkinson’s disease (PD) is the second most common neurodegenerative disorder affecting people in their middle and old age. It is characterized by resting tremor, rigidity, akinesia and postural instability and associated with selected loss of dopaminergic (DA) neuron in the substantianigra pars compacta. Combinations of environmental and genetic factors are thought to cause PD. Among the genetic factors contributing to the pathogenesis of PD, mutations in the α-synuclein gene such as A30P, A53T and E46K increased aggregate formation and mutations in Parkin, DJ-1 and PINK1 impaired mitochondrial function. In addition, environmental factors as well as aging are also thought to contribute the development of the disease. No effective treatment for the disease currently urges the development of new agents that may halt the degeneration of PD. In this study, human neuroblastoma SH-SY5Y cells were differentiated toward the DA phenotype in the presence of retinoic acid (RA) follow by 12-O-tetradecanoyl-phorbol-13-acetate (TPA). Greatly up-regulated tyrosine hydroxylase (TH, DA neuronal marker) in SH-SY5Y cells was observed following 7 days of differentiation. These TH-positive cells exhibited a mature morphology with long, branched cell processes and large cell bodies. Addition of MPP+ (1-methyl-4-phenylpyridinium), a potent complex I inhibitor in DA neurons, to the above SH-SY5Y cells caused significant cell death compared with untreated cells. This MPP+-induced cell death was used to screen herbal extracts provided by Industrial Technology Research Institute (ITRI). Cell viability assay indicated that pretreatment of herbal extracts NTNU-043, -057, -059, -125, -293, -313, -331, -385, -450, and -514 protected undifferentiated SH-SY5Y cells against MPP+-induced toxicity, whereas pretreatment of herbal extracts NTNU-092 and -313 protected differentiated SH-SY5Y cells against MPP+-induced toxicity.

Abstract:
The culture of human induced pluripotent stem cells (hiPSCs) at large-scale becomes feasible with the aid of scalable suspension setups in continuously stirred tank reactors (CSTRs). Suspension cul- tures of hiPSCs are characterized by the self-aggregation of single cells into macroscopic cell aggre- gates that increase in size over time. The development of these free-floating aggregates is dependent on the culture vessel and thus represents a novel process parameter that is of particular interest for hiPSC suspension culture scaling. Further, aggregates surpassing a critical size are prone to spon- taneous differentiation or cell viability loss. In this regard, and, for the first time, a hiPSC-specific suspension culture unit was developed that utilizes in situ microscope imaging to monitor and to characterize hiPSC aggregation in one specific CSTR setup to a statistically significant degree while omitting the need for error-prone and time-intensive sampling. For this purpose, a small-scale CSTR system was designed and fabricated by fused deposition modeling (FDM) using an in-house 3D- printer. To provide a suitable cell culture environment for the CSTR system and in situ microscope, a custom-built incubator was constructed to accommodate all culture vessels and process control devices. Prior to manufacture, the CSTR design was characterized in silico for standard engineering parameters such as the specific power input, mixing time, and shear stress using computational fluid dynamics (CFD) simulations. The established computational model was successfully validated by comparing CFD-derived mixing time data to manual measurements. Proof for system functionality was provided in the context of long-term expansion (4 passages) of hiPSCs. Thereby, hiPSC aggregate size development was successfully tracked by in situ imaging of CSTR suspensions and subsequent automated image processing. Further, the suitability of the developed hiPSC culture unit was proven by demonstrating the preservation of CSTR-cultured hiPSC pluripotency on RNA level by qRT-PCR and PluriTest, and on protein level by flow cytometry

All potential OTT initiatives from different stakeholders that impact the industry are examined. The incumbent technology, traditional TV, is challenged by innovative business models that provide sports content over-the-top via the Internet. Pure OTT channels such as DAZN and digital giants such as Amazon entered the sports rights market forcing old leaders to rethink their business model and to adapt to the new technology. Besides of changing consumer requirements, different market dynamics such as the digitization of sports rights, the trend towards D2C (direct-to-consumer), niche targeting and features around digital fan experiences push OTT in becoming the leading sports consumption technology. The fast-paced and vibrant market movements in the sports media industry make estimations difficult. The findings from qualitative and quantitative findings identified the consolidation of the market to a concentrated number of content aggregators operating in the premium sports right segment as most the likely scenario for the industry in 5-10 years. This trend goes along with a starting content endgame initiated by top rightsholders who implement D2C business models for secondary markets. The major change in the market will be the end of market fragmentation to a more consolidated sports rights market led by major D2C and OTT platforms.

Abstract:
We are currently witnessing an expansion of football in Portugal, with several key players thriving in the best European clubs and being transferred by massive compensations. Some of the existing sports press, both in paper and electronic format, are not open to increase football knowledge among the youth players because they do not have a great visibility with the public, as does senior football and nowadays, even women's football. With the identification of this problem among youth teams and the analysis made on several websites, both institutional and journalistic/informative, the need to aggregate dispersed information about youth football players in Portugal was detected. Taking into account that social networks are very present in our daily lives with football clubs, players and supporters using them frequently, the information contained proved to be essential for the performance of the players. In this context, a platform has been developed which will provide more content and information regarding youth football players, through an approach that allows the use of the platform in different devices of different sizes. With the tests carried out on the platform, it was observed that the aggregation of the information obtained is useful and applicable to youth football players, guaranteeing almost instant access to data taken from various sources, which contributes to the improvement of football and its clubs in Portugal, more specifically, amateur clubs that do not have the financial resources to invest in advanced technology for the sourcing of information and statistical analysis.
