Abstract:
Abstract Post-transcriptional modification of tRNA wobble adenosine into inosine is crucial for decoding multiple mRNA codons by a single tRNA. The eukaryotic wobble adenosine-to-inosine modification is catalysed by the ADAT (ADAT2/ADAT3) complex that modifies up to eight tRNAs, requiring a full tRNA for activity. Yet, ADAT catalytic mechanism and its implication in neurodevelopmental disorders remain poorly understood. Here, we have characterized mouse ADAT and provide the molecular basis for tRNAs deamination by ADAT2 as well as ADAT3 inactivation by loss of catalytic and tRNA-binding determinants. We show that tRNA binding and deamination can vary depending on the cognate tRNA but absolutely rely on the eukaryote-specific ADAT3 N-terminal domain. This domain can rotate with respect to the ADAT catalytic domain to present and position the tRNA anticodon-stem-loop correctly in ADAT2 active site. A founder mutation in the ADAT3 N-terminal domain, which causes intellectual disability, does not affect tRNA binding despite the structural changes it induces but most likely hinders optimal presentation of the tRNA anticodon-stem-loop to ADAT2.

Abstract This article describes the customs and manners of the Malay heritage of Kelantan. They can be dividedinto still-practiced and abandoned. Good manners described include those of respecting for elders,knowledge seeking and friendship. The customs described are of neighborhood, visiting, marriage, birthand death. Exposure to these customs and manners for future generations should be maintained to ensurethe sustainability of the Malay heritage of Kelantan.

Pemerintah Indonesia mengakui keberadaan masyarakat hukum adat yang tersebar di berbagai wilayah Indonesia. Bahkan memberikan pengakuan akan keberadaan masyarakat hukum adat tersebut salah satunya terdapat di dalam Undang-undang Nomor 5 Tahun 1960 tentang Peraturan-Peraturan Dasar Pokok-Pokok Agraria. Penelitian ini merupakan jenis penelitian doktrinal dengan menggunakan pendekatan konsep dan perundang-undangan. Permasalahan yang diangkat dalam penelitian ini adalah mengenai bagaimana keberadaan sanksi adat sebagai bagian dari penerapan hukum pidana adat di lingkungan masyarakat adat. Hasil penelitian menunjukkan bahwa dari berbagai lingkungan masyarakat adat di Kalimantan Timur dan Kalimantan Utara ternyata sanksi terkait hukum pidana adat seharusnya hanya diberlakukan terhadap beberapa perbuatan tertentu saja dan tidak diberlakukan bagi semua orang. Akan tetapi pada kenyataannya masih belum ada kepastian hukum terkait pemberian sanksi pidana adat tersebut. Selain itu ternyata sulit untuk menerapkan aturan hukum pidana adat di tengah masyarakat adat saat ini. Hal tersebut dikarenakan masyarakat sudah banyak yang memahami mengenai keberadaan hukum nasional Indonesia serta cukup beratnya sanksi adat yang dijatuhkan terkait tindak pidana adat.Keyword: hukum, pidana, adat, sanksi

Masyarakat ilalang embayya adalah masyarakat yangmasih menjalankan adat secara penuh sehingga sangat selektif terhadap kemajuan zaman, namunmasyarakat di ipantarang embayyasudah lebih terbuka. Dengan keberadaan sarana pendidikanformal di desa mereka, menyebabkan masyarakat Adata Ammatoa pun harus bernegosiasi untukmemastikan agar adat dan tradisi mereka tetap lestari. Tujuan penelitian ini adalah untuk mengetahui tentang proses Upacara Adat Mupunjung dan nilai-nilai kearifan lokal Upacara Adat Mupunjung. Metode penelitian yang digunakan adalah metode penelitian Sejarah. Hasil penelitian menunjukan pelaksanaan upacara adat ini dilaksanakan setiap tahun sekali pada bulan Maulid yang sebulan sebelumnya telah di persiapkan untuk dana, sesaji, pusaka, panitia, dan situs. Prosesi pelaksanaan upacara adat dilaksanakan pada pagi hari dengan acara puncaknya yaitu tawasulan, membaca ikrar bubuka, wawacan Nabi Adam serta diakhiri dengan berdoa bersama. Nilai-nilai kearifan yang terkandung dalam Upacara Adat Mupunjung adalah nilai agama, nilai sosial, nilai bahasa, nilai seni, nilai sejarah, nilai budaya, nilai ekonomi, nilai pendidikan, nilai etika, nilai estetis.

The aims of this research are to analyze the differences of local wisdom and food security household in an indigenous village and non-indigenous village and to analyze the correlation between local wisdom and food security. Research respondents are housewives or housewives who also act as head of household. The research method used a quantitative approach with questionnaire instruments supported by qualitative data with in-depth interview. The results show that there are the difference of food security in Ciptagelar Village and Tonjong II Village which the level of household local wisdom and food security in Kampung Ciptagelar is higher than in Kampung Tonjong II. In addition, from the three dimensions of local wisdom tested, there are two dimensions that have a positive and quite strong relationship with food security, that is the dimensions of local knowledge and community solidarity and compliance with rules and leaders.Keywords: Food Security, Indigenous Village, Local Wisdom, Non-Indigenouse VillageABSTRAKPenelitian ini bertujuan untuk menganalisis perbedaan kearifan lokal dan ketahanan pangan rumah tangga di Kampung adat dan non-kampung adat serta menganalisis hubungan antara kearifan lokal dengan ketahanan pangan. Responden penelitian merupakan ibu rumah tangga atau ibu rumah tangga yang juga berperan sebagai kepala rumah tangga. Metode penelitian menggunakan pendekatan kuantitatif dengan instrumen kuesioner yang didukung dengan data kualitatif dengan metode wawancara mendalam. Hasil penelitian menunjukkan bahwa terdapat perbedaan tingkat ketahanan pangan di Kampung Ciptagelar dan Kampung Tonjong II yang mana tingkat kearifan lokal dan tingkat ketahanan pangan rumah tangga di Kampung Ciptagelar lebih tinggi dibandingkan dengan Kampung Tonjong II. Selain itu, dari tiga dimensi kearifan lokal yang diuji, terdapat dua dimensi yang memiliki hubungan positif dan cukup kuat dengan ketahanan pangan yakni dimensi pengetahuan lokal dan solidaritas masyarakat dan kepatuhan pada aturan serta pemimpin. Pada satu sisi, kedatangan Islam di tanah Lampung memperkaya budaya masyarakat Lampung; Sementara pada sisi lain, kultur atau budaya masyarakat Lampung berpengaruh pada pengamalan ajaran Islam di masyarakat. Inkulturasi Islam sebagai ajaran baru ke dalam konteks kebudayaan lokal Lampung berjalan secara akomodatif atau adaptif sehingga Islam mewarnai budaya lokal tanpa kehilangan identitasnya.

Abstract:
This study examines the development of the informal micro-business sector and how it is influenced by adat in the traditional village markets of Gianyar Regency on the island of Bali, Indonesia. Informal micro-business is an important element of the Indonesia domestic economy. The adat, commonly defined as customary ancestral laws and cultural traditions, refers specifically to customary laws, both oral and written, and is ingrained in the everyday lives of the Balinese, especially at the local village level. These "laws" act as codes of conduct that the citizens of the community must maintain and abide by. The traditional village is the place where the adat is firmly entrenched and inextricably linked to economic activities. The traditional village market is also the place where informal micro-businesses gather and perform daily economic transactions. Using ethnography and mental map research methods, this research seeks to further the knowledge and understanding of the informal micro-business sector in and near the traditional village markets of Gianyar Regency and how their development is influenced by the adat. The district markets of Payangan, Ubud and Sukawati, along with several random village markets, are selected as the research sites to demonstrate the commonalities, differences and tendencies in the traditional markets despite the presence of various economic forces. This research finds that the adat significantly influences the informal microbusiness sector and provides an opportunity for entrepreneurs to satisfy village customary and cultural needs. Further, the research finds that the traditional village, known as the Desa Adat, is the primary source of the adat's influence in the traditional market.

Abstract:
This thesis investigates the history and phenomenon of legal pluralism in Indonesia. The need to explore this topic has been urged by the revival there of Islamic law and adat law, the two greatest non-state normative orderings, in the last two decades. At the same time the ideal of modernity in Indonesia has been characterized by a state-driven effort in the post-colonial era to make the institution of law an inseparable part of national development. The result has been a conception of law as a homogenous system in which the ideology of legal positivism represents the basic tool for lawmaking. This, however, has led to an impasse, seeing that pluralism and multiculturalism are in fact self-evident phenomena in the society. The state has been obliged, therefore, to accommodate these non-state normative orderings.
The discussion of Indonesian legal pluralism in this thesis focuses on understanding the state's attitude and behavior towards the three largest legal traditions currently operative in the society, i.e., adat law, Islamic law and civil law. Socio-political factors are shown to have much influenced the relations between state and non-state laws. The state's strategy of accommodation of legal pluralism has in fact largely depended on the extent to which those legal traditions have been able to conform to national ideology. Certain "national legal postulates" have functioned as a yardstick by which the country's legislative and judicial institutions have measured the extent of their accommodation of legal pluralism, although they have had little choice but to do so.
Influenced by Masaji Chiba's theory of "three levels of law" (i.e., official law, unofficial law and legal postulates), this thesis analyzes two aspects of legal pluralism in Indonesia: the political and "conflictual" domains of legal pluralism. The analysis is thus generally based on the state policy of legal pluralism reflected in the legal and political strategies confronting the issue of unofficial laws as well as the conflicts arising from such situations. The first aspect is addressed by looking at a number of statutes and regulations promulgated specifically to deal with Islamic law and adat law, while the second is analyzed in terms of actual cases of private interpersonal law arising from conflict between state and non-state legal traditions, as reflected in legislation and court decisions. From a discussion of these two aspects, the thesis concludes that, although the form of the relations between official and unofficial laws may have changed in conjunction with the socio-political situation of the country, the logic behind legal pluralism has in fact never altered, i.e., to use law as a tool of state modernism. Thus conflicts arising from the encounter between different legal traditions will usually be resolved by means of "national legal postulates," making the unofficial laws more susceptible to the state's domination of legal interpretation and resolution.

Abstract:
Studi ini mengambil lokasi di permukiman Kampung Adat Hewokloang, Kabupaten Sikka dengan tujuan untuk mengidentifikasi dan mengkaji wujud bangunan rumah adat dan perubahannya dalam upaya revitalisasi kampung secara menyeluruh. Kesadaran masyarakat akan perlindungan kawasan wisata budaya harus dibangun agar pelestarian situs warisan budaya dapat dilakukan dengan baik, seperti halnya warisan artefak Budaya Hewokloang berupa kondisi fisik bangunan (lepo) di Hewokloang yang telah mengalami perubahan sehingga perlu dilakukan revitalisasi. Penelitian ini mengkaji bentuk asli bangunan tradisional suku Hewokloang berupa wujud dan tipologi bangunan. Metode deskriptif kualitatif dengan paradigma naturalistik digunakan dalam dalam mengkaji dan didukung oleh hasil wawancara dengan tokoh adat, pengamatan langsung dan rekaman sejarah berupa dokumen tertulis. Hasil penelitian menunjukan ada tujuh suku dengan 21 tipe rumah adat (lepo) yang terdapat pada permukiman kampung adat Hewokloang dilihat dari wujud bangunan dan gaya arsitektur. Selain itu bentuk arsitektural murni Hewokloang sudah jarang ditemui karena beberapa sudah ditinggalkan pemiliknya atau diubah dengan bentuk dan style yang lain. Tujuan kajian ini adalah untuk mengidentifikasi bentuk tipologi fasad dari bangunan adat di Kabupaten Paser agar dapat menjadi acuan desain gedung baru, selain itu karakter bangunan akan mencadi citra yang menjadi ekspresi arsitektur. Untuk mengkaji tipologi arsitektur adat Paser melalui pendekatan tipologi arsitektur menurut Raphael Moneo, khususnya bentuk citra atau fasade bangunan adat. Sasaran kajian ini ditujukan pada bangunan tradisional adat Paser yaitu Museum Sadurengas terkait pada bentuk atap, dinding, pintu, jendela, warna dan ukiran sebagai fasad depan bangunan. Museum Sadurengas sebelumnya memliki fungsi istana raja. Kajian ini menggunakan pendekatan tipologi dengan metode penuelusuran secatra kajian literatur pustaka maupun lapangan yang kemudian dideskriptif secara kualitatif. Penelitian ini menyajikan hasil analisis tipologi arsitektur yang menunjukan bahwa Museum Sadurangas sebagian besar pengaruh kerajaan melayu dan Langgam Moorish pada bukaan pintu dan jendela. Perkembangan zaman menyebabkan saat ini kelestarian rumah adat terancam dan berangsur-angsur berkurang. Minangkabau sebagai salah satu etnis terbesar di Indonesia yang menjadi bagian penting dari identitas sosial, dengan Rumah Gadang sebagai bangunan adatnya mulai berkurang dalam penggunaannya. Sedangkan Rumah Gadang memiliki arsitektur yang unik, yang dapat terus dikembangkan tanpa menghilangkan identitasnya. Tujuan dari penelitian ini adalah untuk mengumpulkan data yang relevan tentang bangunan tradisional, khususnya Rumah Gadang sebagai warisan budaya di Sumatra Barat. Pengumpulan data meliputi pembentuk elemen fasade bangunan dan pembentuk elemen ruang dalam sebagai karakter arsitektural rumah adat Minangkabau. Sementara karakter yang lain dapat ditentukan dari denah dasar dan bentuk bangunan. Indikator dari variabel-variabel tersebut yang memperkuat karakter arsitektural pada Rumah Adat Minangkabau sebagai kekayaan budaya serta identitas etnis Mianngkabau Sumatra Barat.
