This dissertation deals with the phenomenon of movement in relation to architecture. What does it entail to create an architecture of movement and what possibilities can this type of design offer us outside of 'conventional' architecture. The research behind the topic bases itself in the theory of Game Design as a tool to structure movement with meaning. Game Design proves itself to be a potent tool in encouraging interactivity, in turn handing over a certain level of control and design over to the players participating. The more control afforded to the players, the more they can affect the outcome and experience of a game. This means games are a medium with the possibility for high replay value as players go back to experience the multiple outcomes of a game. The theory of using Game Design as a tool in architecture is then tested through my own design located on the edge of the Grand Parade in Cape Town. The design aims to create an architecture of multiple outcomes including an 'Everyday' and 'Festival' narratives through the use of movement. Game Design is used as a tool of analysis in order to determine whether the movement is integral to the architecture and its intended function through spatializing the key principles identified in the theory of Game Design. The conclusion drawn is that Game Design is an effective tool in aiding design in architecture. Its value emerges through the fact that it challenges design decisions made within architecture based on how integral they are in relation to the social and programmatic rules and expectations the project attempts to deal with.

Abstract:
The Class Action Fairness Act of 2005 (CAFA) was signed into law on February 18, 2005. Prior to CAFA, plaintiffs found it easier for class action lawsuits to be tried in their preferred venue—state courts. Changes introduced by CAFA practically removed the majority of class action jurisdiction from state to federal courts. Since law and regulation might serve as an external corporate governance mechanism, an interesting question is whether CAFA has strengthened or weakened corporate governance. If CAFA improves corporate governance, associated marginal benefits would outweigh marginal costs. The opposite would be true if CAFA weakens corporate governance. This issue was hotly debated in the US Congress. The proponents argued that CAFA would reduce costs for the affected firms, while opponents argued the opposite. The main purpose of this paper is to examine which side of the debate is reflected in market reactions to various events that either enhanced or reduced the chances of the passage of CAFA. We identify the firms that are most likely to be affected by CAFA and find that the overall market reaction for these firms is positive when the likelihood of CAFA passage increases, while the reaction has been negative when the chance of its passage diminishes. We also hypothesize that firms that are more likely to be exposed to product liability litigation would experience a significantly higher (positive or negative) abnormal return than firms that are more likely to be involved in contract liability law suits. The results support this hypothesis. Includes Bibliographical references (leaves 83- 94).Forty-three male athymic nude mice were implanted with human vaginal mucosal cysts under general anaesthesia with Ketamine [25mg/kg] and Medetomidine [0.5mg/kg]. Cysts in 37 mice were recovered after 9 weeks of growth. twenty three cyst linings had retained the original structure of the vaginal epithelium. No marked deifference was present between the thickness of 9 week old linings and donor vaginal epithelium. The contaminants isolated from the skin of mice before implantation were mainly normal commercals of healthy experimental animals. There was no distinct difference in the number of cases with intact cyst formation between the terramycin/vitamin cocktaik group. The frequency of poor wound healing and/ or murine epidermis ingrowth was three times higher in animals stitched with silk sutures that in those cases where nylon sutures were used.

Abstract:
The growing problem of antibiotic resistance and the lack of promising prospective antibiotics have forced us to search for new classes of antibiotics. Among the candidates to develop into future antibacterials are antimicrobial peptides (AMPs). These potent, broad spectrum compounds are important components of innate immunity of organism from all kingdoms of life. One such family of mini-proteins from plants is called cyclotides, whose members are defines by cyclic backbone and a cystine knot (CCK), which confers to them extreme stability in the face of biological, chemical and physical insults.     Some cyclotides possess Gram-negative specific antibacterial activity; the purpose of this thesis was to characterize how these molecules kill bacteria, and how bacteria would respond to treatment with cyclotides. For this purpose, Salmonella enterica and Escherichia coli mutants resistant to the cyclotides cycloviolacin O2 and cycloviolacin O19, respectively, were selected. These mutants were characterized by whole genome sequencing, genetic reconstitution, fitness measurements, and cross-resistance studies. These studies identified a number of genetic pathways for resistance development to cyclotides. These mutants displayed variable fitness profiles in laboratory growth media and in mice competition experiments, with some mutants possessing a fitness advantage in mice. Cross-resistance studies resulted in the identification of several cases of cross-resistance and collateral sensitivity between cyclotides and other AMPs/antibiotics.      Antimicrobial effects of cyclotides were assayed in different conditions and in bacterial organisms with different surface characteristics. In addition, immunolocalization experiments were performed to explore the biological distribution of cyclotides in plants and to determine the mechanism of action of cyclotides in bacteria, respectively. Antibodies raised against cyO2 were used for this purpose. Immunohistochemical techniques applied to plant cells, tissues and organs provided the information that cyclotides were distributed in all plant organs, and were found in tissues vulnerable to pathogen attack, and that cyclotides were stored in the vacuoles of plant cells. Immunogold staining of cyclotide treated cells of S. typhimurium, showed effects of cyclotide treatment on the cell envelope components as well as cytoplasm. A higher number of cyclotide molecules was associated with the cell envelope, but a considerable fraction of them penetrated into the cytoplasm.

Abstract:
Using light as an energy source allows chemical reactions to be carried out with high precision in time and space. In time, because light can be switched on and off, in space, because light can be highly focussed. Further, different colours of light can be used to activate different reactants individually, even when they are present in the same reaction mixture. Herein, a system of two light-sensitive molecules was developed, which react completely sequence-independently with UV or visible light. The wavelength-dependence as well as the influence of chemical-physical parameters on such reactions were investigated in-depth.

Abstract:
Apoptosis, or programmed cell death, is an essential process in all multi-cellular organisms. It is indispensable to an organismâ s survival, preventing the malicious propagation of DNA damage and pathogenic alterations, through the clean disposal of afflicted cells. The BAD/tBID/BAK pathway is a portion of the apoptosis molecular pathway, albeit an important pathway since it is known to be deregulated and lead to pathological ailments such as cancer. Using chemical kinetics the BAD/tBID/BAK signaling pathway is modeled as a set of (nonlinear) ordinary differential equations. A first-cut numerical analysis reveals a mechanism where BAD sensitizes a switch from tBID activation to BAK activation. The phosphorylation of BAD is shown to inhibit this sensitizing effect. All behaviors are supported by experimental data, thereby validating the model of the BAD/tBID/BAK pathway. Moreover, modeling the phosphorylation of BAD as one of two modes, some conflicting experimental data about BADâ s phosphorylation can be disentangled. Parameter values (in this case the kinetic rate constants) are prone to error or missing altogether. Chemical reaction network theory, however, provides a theoretical approach to complement the initial numerical analysis without having to specify rate constant values. We extend the global asymptotic stability and robustness results in [92] to include any complex-balanced mass-action network. This enables us to study the BAD/tBID/BAK signaling network by breaking it into two sub-networks: one with BAD and tBID, and the other with tBID and BAK. The complex-balanced BAD/tBID sub-network is shown to possess a unique steady state which is globally asymptotically stable. This verifies the simple and dynamically well-behaved exchange of BAD for Bcl-2 proteins which guard against tBID activation. The second sub-network, tBID/BAK, is formulated as a complex-balanced network with a perturbation representing the reaction of tBID catalyzing the activation of BAK. Our theoretical results produce a non-conservative, though state-dependent, condition which can be used to prove global convergence to a neighborhood of the unperturbed steady state. We then illustrate the biological importance of the result for tBID/BAK sub-network with an example design for a drug delivery system.

Abstract:
This dissertation addresses the development of integrative modeling strategies capable of combining deterministic and stochastic, discrete and continuous, as well as multi-scale features. The first set of studies combines the purely deterministic modeling methodology of Biochemical Systems Theory (BST) with a hybrid approach, using Functional Petri Nets, which permits the account of discrete features or events, stochasticity, and different types of delays. The efficiency and significance of this combination is demonstrated with several examples, including generic biochemical networks with feedback controls, gene regulatory modules, and dopamine based neuronal signal transduction. A study expanding the use of stochasticity toward systems with small numbers of molecules proposes a rather general strategy for converting a deterministic process model into a corresponding stochastic model. The strategy characterizes the mathematical connection between a stochastic framework and the deterministic analog. The deterministic framework is assumed to be a generalized mass action system and the stochastic analogue is in the format of the chemical master equation. The analysis identifies situations where internal noise affecting the system needs to be taken into account for a valid conversion from a deterministic to a stochastic model. The conversion procedure is illustrated with several representative examples, including elemental reactions, Michaelis-Menten enzyme kinetics, a genetic regulatory motif, and stochastic focusing. The last study establishes two novel, particle-based methods to simulate biochemical diffusion-reaction systems within crowded environments. These simulation methods effectively simulate and quantify crowding effects, including reduced reaction volumes, reduced diffusion rates, and reduced accessibility between potentially reacting particles. The proposed methods account for fractal-like kinetics, where the reaction rate depends on the local concentrations of the molecules undergoing the reaction. Rooted in an agent based modeling framework, this aspect of the methods offers the capacity to address sophisticated intracellular spatial effects, such as macromolecular crowding, active transport along cytoskeleton structures, and reactions on heterogeneous surfaces, as well as in porous media. Taken together, the work in this dissertation successfully developed theories and simulation methods which extend the deterministic, continuous framework of Biochemical Systems Theory to allow the account of delays, stochasticity, discrete features or events, and spatial effects for the modeling of biological systems, which are hybrid and multiscale by nature.

Abstract:
In February 2013, the OECD introduced its Base Erosion and Profit Shifting Project. This study investigates whether there is a market reaction on key event dates throughout the development of the BEPS project. The OECD has no regulatory power and analysts have doubted whether it will change the global corporate taxation structure or even lead to an increase in corporate taxation for multinational corporations. However, major accounting firms have published advice on how to comply with changes required by the BEPS Action Plan. I examine the three-day cumulative abnormal return for different categories of firms that may be disproportionately affected by changes brought about by the BEPS Project. I find the expected negative market reaction on several of the key event dates, indicating investors believe the OECD's plan will be implemented.

Abstract:

Catalysts play an essential role in nearly every chemical production process. Platinum supported on high surface area carbon substrates (Pt/C) is one of the promising candidates as an electrocatalyst in low temperature polymer electrolyte fuel cells. Developing the activity of the Pt/C catalyst with narrow Pt particle size distribution and good dispersion has been a main concern in current research.


In this study, the main objective was the development and characterization of inexpensive and effective nanophase Pt/C electrocatalysts. A set of modified Pt/C electrocatalysts with high electrochemical activity and low loading of noble metal was prepared by the impregnation-reduction method in this research. The four home-made catalysts synthesized by different treatments conditions were characterized by several techniques such as EDS, TEM, XRD, AAS, TGA, BET and CV.


Pt electrocatalysts supported on acid treatment Vulcan XC-72 electrocatalysts were produced successfully. The results showed that Pt particle sizes of Pt/C (PrOH)x catalysts between 2.45 and 2.81nm were obtained with homogeneous dispersion, which were more uniform than the commercial Pt/C (JM) catalyst. In the electrochemical activity tests, ORR was confirmed as a structure-sensitive reaction. The Pt/C (PrOH/pH2.5) showed promising results during chemically-active surface area investigation, which compared well with that of the commercial standard Johnson Matthey Pt/C catalyst. The active surface area of Pt/C (PrOH/pH2.5) at 17.98m2/g, was higher than that of the commercial catalyst (17.22 m2/g ) under the conditions applied. In a CV electrochemical activity test of Pt/C catalysts using a Fe2+/Fe3+ mediator system study, Pt/C (PrOH/pH2.5) (67mA/cm2) also showed promise as a catalyst as the current density is comparable to that of the commercial Pt/C (JM) (62mA/cm2).


A remarkable achievement was attained in this study: the electrocatalyst Pt supported on CNTs was synthesized effectively. This method resulted in the smallest Pt particle size 2.15nm. In the electrochemically-active surface area study, the Pt/CNT exhibited a significantly greater active surface area (27.03 m2/g) and higher current density (100 mA/cm2) in the Fe2+/Fe3+ electrochemical mediator system than the other home-made Pt/C catalysts, as well as being significantly higher than the commercial Pt/C (JM) catalysts. Pt/CNT catalyst produced the best electrochemical activities in both H2SO4 and K4[Fe(CN)6] electrolytes. As a result of the characteristics of Pt/CNT,it can be deduced that the Pt/CNT is the best electrocatalyst prepared in this study and has great potential for use in fuel cell applications.

Initially, a pilot study was carried out in order to reconstruct the major paternal and maternal lineages of the Muslim population living in the Cape metropolitan area. The Study has shown the ability of molecular genetic tools to give insight into the origins and history of local communities. The study was also used as a point of reference for the Strand Muslim Community project. Genetic variations of the Y-chromosome and mitochondrial DNA for the pilot study were analyzed using the RFLP technique. The SNaPshot mini-sequencing technique was used to genotype single nucleotide polymorphisms (SNP) on the Y-chromosome and mitochondrial DNA in 115 males from the Strand Muslim community.

Eurycoma longifolia (Tongkat Ali
TA) is a Malaysian shrub used to treat various illnesses including male infertility. Considering that TA is also used to improve male fertility and no report 
regarding its safety has been published, this study investigated the effects of a patented, aqueous TA extract on various sperm and testicular functions. Materials and Methods This study 
encompasses two parts (part 1: on spermatozoa
part 2: on TM3-Leydig and TM4-Sertoli cells). Part 1: Semen samples of 27 patients and 13 fertile donors were divided into two groups, 
washed and swim-up prepared spermatozoa, and incubated with different concentrations of TA (1, 10, 20, 100, 2000 &mu
g/ml) for 1 hour at 37°
C. A sample without addition of TA served as control. After incubation with TA, 
the following parameters were evaluated: viability (Eosin-Nigrosin test), total and progressive motility (CASA), acrosome reaction (triple stain technique), sperm production of reactive oxygen

dihydroethidium test
DHE), sperm DNA fragmentation (TUNEL assay) and mitochondrial membrane potential (&Delta

Les deux methodes spectroscopiques utilisees dans cette etude, d'une part l'infrarouge appliquee a la caracterisation des defauts chimiques d'irradiation et a l'etude de l'hydratation et d'autre part la rmn 1 3c haute resolution en phase solide appliquee a la caracterisation des defauts d'irradiation et au suivi de la mobilite locale des groupements de la resine, se sont revelees tres precieuses. L'action des rayonnements ionisants sur les resines etudiees conduit d'une part a la formation de produits d'oxydation que sont les amides, acides carboxyliques, cetones et esters et d'autre part a des ruptures de chaine avec formation de phenol. Lors d'une irradiation en milieu humide, la proportion d'esters formes est plus faible alors que les ruptures des chaines sont plus importantes. Par ailleurs, le suivi de la mobilite locale, par rmn haute resolution en phase solide, montre une diminution des mouvements par irradiation avec des rayons , attribuee a la rigidification des resines concernees. Apres irradiation, la capacite d'absorption d'eau des resines augmente. Il n'existe pratiquement pas de difference d'absorption entre les resines dgeba/ddm irradiees en milieu humide et celles irradiees en milieu sec, a des doses d'irradiation similaires. Avant irradiation, la fixation de molecules h 2o sur d'autres molecules h 2o se fait dans l'environnement des alcools. Par contre, apres irradiation, de telles associations s'etablissent pres des c = o.

More than 20 million infants in the world (15.5 % of all births) are born with low birth weight. Ninety-five % of them are in developing countries. Oral colonization of Gramnegative anaerobes has 
been implicated as a risk factor for preterm delivery of low birth weight (PLBW) infants. The objective of this study was to investigate the association between periodontal pathogens and pre-term 
delivery of low birth weight (PLBW) infants. The study sample included 200 randomly selected women admitted to the department of obstetrics-gynecology of the teaching hospital of Butare in 
Rwanda. Mothers were asked to complete a questionnaire in order to identify factors which might pose a health risk to them and their infants. Gingival crevicular fluid (GCF) was collected from each quadrant of the mother&rsquo
s month (using paper points) within 24 hours of delivery. Ten ml of foetal cord serum samples were collected at delivery and 10 ml of maternal serum samples were 
collected within 48 of delivery. GCF was examined by PCR for the presence of 5 periodontopathogens and ELISA was used for the evaluation of cytokines (IL-6 and IL-10) and immunoglobulins (IgM, IgG) in foetal cord and maternal blood against the periodontopathogens. P. intermedia showed significant associations either on its own or in combinations with most indicators of 
periodontal disease used in this study, while Aa and members of the red complex were significantly associated with gum bleeding and reduced frequency of tooth brushing. A strong association 
between PLBW and maternal and foetal cord serum sample levels of IL-10 was observed. Also, a good association was observed between PLBW and FCB sample levels of IL-6. Significant 
associations were observed between PLBW and maternal IgG against the different peridontopathogens. The findings of this study may suggest that the levels of maternal IgG and foetal IgM 
against the different periodontopathogens are associated with dissemination of maternal periodontopathogens to the foetus thereby illiciting an inflammatory response which contributes to 
PLBW.

The aim of this study was to investigate the genetic diversity of the SLC22A1 gene and to deduce its possible pharmacogenetic implications within the Cape Coloured population of South 
Africa
a uniquely admixed population of immigrant Europeans, Asians and the indigenous populations. Recent studies have reported an abundance of polymorphic variants within this solute 
carrier transporter gene encoding for the organic cation transporter 1, as well as evidence linking these variants to an effect on metformin uptake. This study included establishing baseline 
frequency distribution of previously reported alleles for 20 SNP variants within the SLC22A1 gene, as well as the development of SNaPshot®
and Multiplex AS-PCR genotyping assays, and 
also exploring the possibility of using High-resolution melt (HRM) analysis as a costeffective alternative for SNP genotyping. Ethics clearance was obtained from the Ethics Committee of the 
University of the Western Cape. Biological samples in the form of buccal (oral) swabs were collected from 132 unrelated voluntary donors from the Cape Coloured population residing in the 
Cape Metropolitan area. Two SNaPshot®
Multiplex Systems were specifically designed for the study,successfully optimized and used for genotyping. Hundred genetic profiles were then generated for a total of 20 SNP variants on SLC22A1 gene, using this primer extension-based genotyping method that enables multiplexing up 10 SNPs. Population genetics data obtained for 
the investigated SNPs were analysed using various statistical analysis software. Important population genetic parameters were calculated, and possible pharmacogenetics implications were then discussed. Among others, allelic and genotypic frequencies, as well as linkage disequilibrium were determined and compared with world populations. Minor deviation from Hardy- Weinberg equilibrium was observed in the Cape Coloured population. No significantLinkage Disequilibrium between the investigated SNPs was observed in this population. A Multiplex allele specific &ndash
PCR (MAS-PCR) genotyping 
system was successfully designed and optimized for the genotyping of 10 SNPs from the SLC22A1. This system, also developed specifically for this study, was made of 2 multiplexes each covering 5 SNPs. It is an inexpensive genotyping assay that allows for efficient discrimination of SNP polymorphisms in one reaction tube with standard PCR conditions. A pilot study was 
conducted to explore the possibility of using High-resolution melt (HRM) analysis as a cost-effective alternative for SNP genotyping. In addition to genotyping, HRM analysis can be used to scan 
large numbers of samples for novel genetic variations.

Two strategies were considered for the synthesis of 2,3-unsaturated O-glucosyl-1,2,3-triazoles. The reaction between tri-O-acetyl-D-glucal and triazole alcohols gave no stereoselectivity. In fact, β-isomer formation was increased. A second strategy furnished 2,3-unsaturated O-glucosides from tri-O-acetyl-D-glucal and alkynols via Ferrier rearrangement; this methodology employing montmorillonite K-10 doped with FeCl3∙6H2O affords new glycosides in good to excellent yields, short time and high α-stereoselectivity in dichloromethane. Subsequently, the glucosides were coupled with 2-azido-1,4-naphthoquinone to give a new series of 1,2,3-1H-triazolyl O-glucoside derivatives based on click reaction.
Duas estratégias foram consideradas para a síntese de 1,2,3-triazóis O-glicosídeos2,3-insaturados. A reação entre o tri-O-acetil-D-glucal e os alcoóis triazólicos não mostrou seletividade. De fato, a formação do isômero β foi aumentada. Uma segunda estratégia forneceu O-glicosídeos 2,3-insaturados a partir do tri-O-acetil-D-glucal e alquinóis via rearranjo de Ferrier; esta metodologia formou novos glicosídeos, empregando montmorilonite K-10 dopado com FeCl3∙6H2O em diclorometano, em rendimentos de bons à excelentes, baixo tempo reacional e alta α-estereosseletividade. Subsequentemente, os glicosídeos foram acoplados com 2-azido-1,4-naftoquinona formando uma nova série de derivados 1,2,3-1H-triazolil O-glicosídeos através da reação Click.

Cette thèse rapporte les résultats de deux études cherchant à approfondir nos connaissances sur l’efficacité et les mécanismes d’action du disulfirame dans le traitement de l’alcoolodépendance. Dans la première étude, en cross-over et randomisée, 33 sujets alcoolodépendants ont reçu des messages avec menace d’effet aversif ou neutres pendant 2 séances d’exposition au stimulus alcool. La condition menace a consisté à annoncer aux patients qu’ils avaient pris du disulfirame et la condition neutre qu’ils avaient pris un placebo. En réalité, tous avaient reçu un placebo. Dans la condition menace comparée à la condition neutre, la tension artérielle diastolique a baissé (p = 0. 04). Les réponses subjectives sont restées inchangées. Des effets bénéfiques alternatifs qui découlent d’autres processus cognitifs sont discutés. La deuxième étude rapporte les résultats préliminaires d’une méta-analyse de l’efficacité du disulfirame dans l’alcoolodépendance. Des 16 études randomisées et contrôlées confondues, notre méta-analyse a montré une efficacité supérieure du disulfirame à la condition contrôle OR=2. 35 (95%CI=1. 48-3. 78). Dans les études en ouvert comparées à celles en aveugle, le disulfirame avait une efficacité supérieure. Nous avons également pu confirmer que le disulfirame est efficace quand son administration est supervisée. En conclusion, nous avons pu montrer que le disulfirame est un traitement efficace si les études sont ouvertes et supervisées. Son mécanisme d’action, étudié en laboratoire, passe par un effet de la menace d’une réaction aversive, effet qui n’a pas semblé mesurable au niveau subjectif, mais qui s’est révélé au niveau neurovégétatif

Abstract:

In many forensic cases, such as mass disasters reconstruction cases, the recovered DNA is highly degraded. In such incidences, typing of STR loci has become one of the most powerful tools for retrieving information from the degraded DNA. However, as DNA degradation proceeds, three phenomena occur consecutively: loci imbalance, allele dropout and no amplification. To solve the problem of degraded DNA, redesigned primer sets have been developed in which the primers were positioned as close as possible to the STR repeat region. These reduced primer sets were called Miniplexes. Unfortunately, a few of the CODIS STR loci cannot be made into smaller amplicons. For this reason non-CODIS miniSTRs have been developed. The present study was undertaken for the population genetic analysis of microsatellite variation in four South African populations
Afrikaner, Xhosa, Mixed Ancestry and Asian Indian using eight non-CODIS miniSTR loci. These miniSTRs loci were characterized within the populations by estimating the levels of diversity of the markers, estimating the population genetic parameters, and studying the inter-population relationships. All of the miniSTRs were amplified successfully and the genetic variability parameters across all loci in Afrikaner, Mixed Ancestry, Asian Indian and Xhosa were estimated to be in the range of 3 (D4S2364) to 12 (D9S2157) alleles, the total number of alleles over all loci ranged from 100 to 204, the allelic richness ranged from 3.612 to 10.307 and the heterozygosity ranged from 0.4360 to 0.8073. Genetic distance was least between Afrikaner and Asian Indian and highest between Xhosa and Mixed Ancestry. Deviations from Hardy-Weinberg equilibrium were not observed for most of the loci. The low mean FIS (-0.027) and FIT (-0.010) and FST (0.017) values across the populations indicated low level of inbreeding within (FIS) and among (FST) the populations. The Asian Indian population showed higher levels of the inbreeding coefficient, indicating less gene exchange between it and other populations. These 8 markers can be used for genetic investigations and assessing population structure. The study contributed to the knowledge and genetic characterization of four South African populations. In addition, these MiniSTRs prove to be useful in cases where more genetic information is needed.

L’objectif de la thèse consiste à dresser les apports théoriques récents sur les actions et réactions stratégiques autour des écosystèmes d’affaires et sous-tendent en partie notre problématique de la recherche puisque nous nous attachons à un nouveau champ organisationnel porté par les plateformes numériques. La particularité de ces plateformes est caractérisée par un modèle d’organisation reposant sur un système coopératif ou sur un système capitaliste traditionnel, conduit par la recherche du profit. Cette recherche s’inscrit sur le champs de la stratégie et consiste à étudier les jeux d’acteurs, en s’appuyant plus particulièrement sur les écosystèmes d’affaires (en abrégé ESA), concept apparu dans les travaux de Moore (1993, p.76) et définit comme un ensemble d’« acteurs entretenant des relations de coopétition » (Koenig, 2012, p. 210). L’ESA désigne une communauté économique regroupant des organisations qui interagissent entre elles afin de produire des biens et/ou des services susceptibles de créer de la valeur pour les clients et pour elles-mêmes. L’ensemble des ESA que nous étudions se regroupent autour des plateformes appelées mobil banking (nouvel entrant) et des établissements bancaires (firmes installées). La problématique qui a conduit notre travail de thèse est de comprendre comment les plateformes se sont imposées, par quelles actions stratégiques elles sont parvenues à se faire une place sur le marché et éventuellement le dominer. Notre terrain de recherche est le Tchad et notre méthodologie repose sur une analyse qualitative basée sur une étude de cas. Les données collectées nous permettront de retracer l’historique des évènements et d’étudier les discours des acteurs impliqués dans le processus à l’œuvre au moyen d’une analyse manuelle des corpus. The objective of the thesis is to draw up recent theoretical contributions on strategic actions and reactions around business ecosystems and partly underpin our research problematic since we are focusing on a new organizational field carried by digital platforms. The particularity of these platforms is characterized by an organizational model based on a cooperative system or on a traditional capitalist system, driven by the search for profit. This research is part of the field of strategy and consists of studying the games of actors, relying more specifically on business ecosystems (abbreviated to ESA), a concept that appeared in the work of Moore (1993, p.76) and is defined as a set of "actors maintaining relationships of coopetition" (Koenig, 2012, p. 210). ESA refers to an economic community of organizations that interact with each other to produce goods and/or services that can create value for customers and for themselves. The ESAs we are studying are grouped around platforms called mobile banking (new entrants) and banking institutions (established firms). The problem that led to our thesis is to understand how the platforms have imposed themselves, by what strategic actions they have managed to make a place for themselves on the market and eventually dominate it. Our research field is Chad and our methodology is based on a qualitative analysis of a case study. The data collected will allow us to retrace the history of events and to study the discourses of the actors involved in the process at work by means of a manual analysis of the corpus. The proposals produced can shed new lighting on the strategic actions of the actors

O presente trabalho apresenta os resultados de uma pesquisa de campo realizada com crianças Tikuna, educadores e pais da comunidade do Umariaçú II. O objetivo deste estudo é investigar as ações e reações das crianças indígenas no processo de ensino e aprendizagem na educação infantil, na Escola Municipal AEGATU DECATUCU. Os objetivos específicos visam: a) observar as relações de convivência entre criança-criança e criança-educador frente às atividades escolares diante da prática pedagógica do professor em sala de aula; b) descrever os modos de como a família (pais/ responsáveis) participa como verdadeiro núcleo, sendo parceira da educação e da aprendizagem da vida escolar do filho; c) registrar por meio da observação e convivência como a criança indígena reage ao aprendizado formal na sala de aula, mediante as atividades propostas pelos educadores. No que se refere ao comportamento, a criança Tikuna tem seu momento de diversão, e em relação à aprendizagem, é atenciosa quando os educadores estão a explicar. Percebe-se que o estímulo e a motivação da criança devem estar contemplados na proposta pedagógica dos educadores Tikuna. Os achados da pesquisa revelam que os pais procuram ser colaboradores e parceiros da educação dos seus filhos. Declaram que o ensinamento e a aprendizagem ainda são heranças marcadas e enraizadas pelas gerações. Para os educadores, a escola é a instituição que tende a viabilizar muitas atividades para motivar os pais a estarem presentes no cotidiano escolar de seus filhos. A educação indígena para crianças pequenas enfrenta desafios para adequar o currículo à demanda do ensino e da aprendizagem. Percebe-se a falta de materiais e de recursos pedagógicos específicos para atender a demanda escolar da criança indígena. O entorno social imposto pela sociedade urbanizada se infiltra na cultura e tradição local gerando um processo de aculturação e descaracterização da criança indígena. Diante disto a criança indígena, como ser em desenvolvimento e marcada por sua cultura, não tem o poder de mudar o atual contexto sociocultural, mas, pode ressignificá-lo por meio dos modos de vida e sobrevivência que a educação e o educador podem lhe oferecer.
This paper presents the results of field research carried out with the children, educators and parents of the Umariaçú II community. The goal of this study is to investigate the actions and reactions of the indigenous children in the teaching and learning process in early childhood education, in the AEGATU DECATUCU Municipal School. The specific goals laid out in this study were: Observe within the daily life of the school the communal interaction relations among the children and between children and educator in regard to the school activities connected to the pedagogical practice of the teacher in the classroom. Describe the ways in which the family (parents/those/responsible), in relation to the school, participates as true nuclei, being these partners in the education and learning process within the school life of their children. Register, through observation and interaction, how the indigenous children react to formal learning in the class room, confronted with the proposed activities of the educators. With regard to behavior, the children, as all children, have their moments of fun, and with regard to the learning process, these children are very attentive when the educators are explaining things. One perceives that stimulus and motivation must always walk together as a proposal of the educators. Besides this, the research revealed that the parents are not such great collaborators and partners in the [formal] education of their children, because they still have the image that teaching and learning are legacies marked and rooted by family generations. For the educators the school is the institution which tends to make possible many activities to have the presence of the parents in its daily life, even though they are confronted with the challenges which indigenous education faces in its curricula, resources, community, acculturation and social life which is being daily infiltrated by a more urbanized society. Faced with this, the presence of the child remains as a being in development, with dreams, fulfillments, many times being marked by its culture, which the child itself does not have the power to change, but, re-signifies through their ways of life and survival.

Differents facteurs tels les rayonnements uv ou ionisants, les agents oxydants, les radicaux libres, peuvent engendrer des dommages au niveau des bases de l'adn. Ces modifications peuvent avoir une influence notable sur la structure et les proprietes du biopolymere. Afin de mieux comprendre la nature de ces lesions, leur mode de formation et les effets qu'elles peuvent avoir sur l'integrite de l'adn, l'etude de certains degradations de la thymine a ete realisee au moyen des outils de la chimie quantique. La premiere partie de ce travail est consacree a l'etude comparative des proprietes conformationnelles et electroniques de produits d'oxydation du groupement methyle de la thymidine par l'intermediaire d'une methode locale de la dft. Le chapitre suivant traite des intermediaires radicalaires formes par l'addition d'un atome d'hydrogene ou du radical hydroxyle sur la double liaison 5,6 de la thymine. Un protocole general d'etude de la structure et des constantes de couplage hyperfin isotrope rpe, incluant une methode hybride de la dft (b3lyp), les effets du solvant, des vibrations et de la temperature a ete mis en place. Ce protocole a ete applique a l'analyse des produits radicalaires formes par addition de l'atome d'hydrogene. D'autre part, les parametres thermodynamiques et cinetiques des differentes reactions d'addition ont ete determines. Un modele de reactivite en solution est finalement propose pour la comprehension des mecanismes d'addition du radical hydroxyle. La derniere partie concerne l'etude des produits finaux non radicalaires formes par l'addition initiale du radical hydroxyle sur la double liaison 5,6 de la thymine. La comparaison des conformations et des proprietes electroniques des deux isomeres cis d'hydroxyhydroperoxydes de thymine et des deux isomeres cis de la 5,6-dihydrothymidine a ete effectuee.

The master’s thesis deals with the design of steam back-pressure turbine with integrated gearbox for compressor drive according to given parameters. The first part of the thesis is literature review which comprises a general division of steam turbines, especially a description of the types of blading. Furthermore, it describes the conversion of energy in the steam turbine stage in detail. The second part of the thesis contains calculations and includes the design of the flow parts of the steam turbine, the design of the outlet and the integrated gearbox. Attached to this work are drawings of a simplified section of the steam turbine with a gearbox and the layout drawing of the machine.

An atomic-level description of the presumed catalytic action of β-galactosidase is described. This large enzyme, from E. coli , carries out two reactions which allow the bacterium to live on the disaccharide lactose. First, it breaks down lactose to the two monosaccharides galactose and glucose. Second, it converts lactose into another disaccharide, allolactose, which is the inducer for the lac operon, and thus is the signal to the bacterium to produce more β-galactosidase. The work is based on high resolution x-ray crystallography and enzyme kinetics. A crystal form of β-galactosidase was isolated that permits data collection up to 1.5 à resolution. Using this crystal form, the structures of several ligands bound to the enzyme were determined. These ligands were chosen to mimic various points in the reaction: binding of substrate, covalent intermediates, transition states, and products. Together these complexes suggest a reaction coordinate for β-galactosidase which clarifies and enhances previous ideas about the reaction mechanism. The reaction includes a conformational change triggered by the progression of the substrate towards the transition state. Additional investigation suggests that this conformational change is involved in determining whether the enzyme carries out its hydrolysis or isomerization reaction. Considerations of the structure in the context of other related enzymes suggest an evolutionary path for β-galactosidase. It is suggested that a progenitor enzyme which catalyzed the hydrolysis of long polysaccharide substrates recruited additional domains which permit β-galactosidase to act on smaller substrates and produce the inducer, allolactose. This dissertation includes both my previously published and co-authored material.

Abstract This thesis presents a novel control architecture, Samba, for reactive task execution. Reactive task execution implies goal-oriented and reactive properties from a robot and the ability to execute several tasks at the same time, also in a dynamic environment. These requirements are fullfilled in Samba by the rrepresentation of goals, intermediate results, and robots actions. The key idea in Samba is to produce continously reactions for all the important objects in the environment. These reactions are represented as action maps, which are a novel representation for robot actions. An action map specifies for each possible action how preferable the action is from the perspective of the producer of the map. the preferences are shown by assigning a weight to each action. Tasks are executed by modifying and combining action maps. The tasks can be either reasoned by a higher layer or triggered by sensor data. Action maps, and the methods for modifying and combining them, enable executing tasks inparallel and considering the dynamics of the environment. further, as the action maps are produced continously from sensor data, the robot actions are based on the current state of the environment. Markers describe goals and intermediate results. They facilitate managing the complexity of the system. Markers describing intermediate results decompose the system vertically, into producers and consumers of data. Markers describing goals decompose the control system horizontally, into a Samba layer and a higher layer of reasoning tasks. Tasks flow via markers from the higher layer to the Samba layer. Markers are tested on a real robot equipment with stereo gaze platform. Further, the samba architecture is applied to playing soccer. Experiments were carried out in the 1997 and 1998 RoboCup competitions. These experiments show that the Samba architecture is a potential alternative for controlling a mobile robot in a dynamic environment.

Abstract:
I investigate whether investors have statistically significant negative reactions to the local clients of sanctioned auditors at the time of the PCAOB’s announcement of an enforcement action against that auditor. I also investigate whether or not the significance of the reactions has uniformly varied over time to account for the possibility of changing comprehensiveness in PCAOB enforcement actions. I measure abnormal returns using CAPM over three different measurement periods. I also disaggregate the data by year of enforcement action. 6 of the 27 data sets produced a statistically significant negative abnormal return suggesting that clients of sanctioned auditors do experience statistically significant abnormal negative returns at the time of the PCAOB’s announcement of the enforcement action. The significance of the abnormal returns did not vary overtime.
