Full textThis dissertation deals with the phenomenon of movement in relation to architecture. What does it entail to create an architecture of movement and what possibilities can this type of design offer us outside of 'conventional' architecture. The research behind the topic bases itself in the theory of Game Design as a tool to structure movement with meaning. Game Design proves itself to be a potent tool in encouraging interactivity, in turn handing over a certain level of control and design over to the players participating. The more control afforded to the players, the more they can affect the outcome and experience of a game. This means games are a medium with the possibility for high replay value as players go back to experience the multiple outcomes of a game. The theory of using Game Design as a tool in architecture is then tested through my own design located on the edge of the Grand Parade in Cape Town. The design aims to create an architecture of multiple outcomes including an 'Everyday' and 'Festival' narratives through the use of movement. Game Design is used as a tool of analysis in order to determine whether the movement is integral to the architecture and its intended function through spatializing the key principles identified in the theory of Game Design. The conclusion drawn is that Game Design is an effective tool in aiding design in architecture. Its value emerges through the fact that it challenges design decisions made within architecture based on how integral they are in relation to the social and programmatic rules and expectations the project attempts to deal with.

Full textIncludes Bibliographical references (leaves 83- 94).Forty-three male athymic nude mice were implanted with human vaginal mucosal cysts under general anaesthesia with Ketamine [25mg/kg] and Medetomidine [0.5mg/kg]. Cysts in 37 mice were recovered after 9 weeks of growth. twenty three cyst linings had retained the original structure of the vaginal epithelium. No marked deifference was present between the thickness of 9 week old linings and donor vaginal epithelium. The contaminants isolated from the skin of mice before implantation were mainly normal commercals of healthy experimental animals. There was no distinct difference in the number of cases with intact cyst formation between the terramycin/vitamin cocktaik group. The frequency of poor wound healing and/ or murine epidermis ingrowth was three times higher in animals stitched with silk sutures that in those cases where nylon sutures were used.

Full textCatalysts play an essential role in nearly every chemical production process. Platinum supported on high surface area carbon substrates (Pt/C) is one of the promising candidates as an electrocatalyst in low temperature polymer electrolyte fuel cells. Developing the activity of the Pt/C catalyst with narrow Pt particle size distribution and good dispersion has been a main concern in current research.
In this study, the main objective was the development and characterization of inexpensive and effective nanophase Pt/C electrocatalysts. A set of modified Pt/C electrocatalysts with high electrochemical activity and low loading of noble metal was prepared by the impregnation-reduction method in this research. The four home-made catalysts synthesized by different treatments conditions were characterized by several techniques such as EDS, TEM, XRD, AAS, TGA, BET and CV.
Pt electrocatalysts supported on acid treatment Vulcan XC-72 electrocatalysts were produced successfully. The results showed that Pt particle sizes of Pt/C (PrOH)x catalysts between 2.45 and 2.81nm were obtained with homogeneous dispersion, which were more uniform than the commercial Pt/C (JM) catalyst. In the electrochemical activity tests, ORR was confirmed as a structure-sensitive reaction. The Pt/C (PrOH/pH2.5) showed promising results during chemically-active surface area investigation, which compared well with that of the commercial standard Johnson Matthey Pt/C catalyst. The active surface area of Pt/C (PrOH/pH2.5) at 17.98m2/g, was higher than that of the commercial catalyst (17.22 m2/g ) under the conditions applied. In a CV electrochemical activity test of Pt/C catalysts using a Fe2+/Fe3+ mediator system study, Pt/C (PrOH/pH2.5) (67mA/cm2) also showed promise as a catalyst as the current density is comparable to that of the commercial Pt/C (JM) (62mA/cm2).
A remarkable achievement was attained in this study: the electrocatalyst Pt supported on CNTs was synthesized effectively. This method resulted in the smallest Pt particle size 2.15nm. In the electrochemically-active surface area study, the Pt/CNT exhibited a significantly greater active surface area (27.03 m2/g) and higher current density (100 mA/cm2) in the Fe2+/Fe3+ electrochemical mediator system than the other home-made Pt/C catalysts, as well as being significantly higher than the commercial Pt/C (JM) catalysts. Pt/CNT catalyst produced the best electrochemical activities in both H2SO4 and K4[Fe(CN)6] electrolytes. As a result of the characteristics of Pt/CNTï¼it can be deduced that the Pt/CNT is the best electrocatalyst prepared in this study and has great potential for use in fuel cell applications.

Full textInitially, a pilot study was carried out in order to reconstruct the major paternal and maternal lineages of the Muslim population living in the Cape metropolitan area. The Study has shown the ability of molecular genetic tools to give insight into the origins and history of local communities. The study was also used as a point of reference for the Strand Muslim Community project. Genetic variations of the Y-chromosome and mitochondrial DNA for the pilot study were analyzed using the RFLP technique. The SNaPshot mini-sequencing technique was used to genotype single nucleotide polymorphisms (SNP) on the Y-chromosome and mitochondrial DNA in 115 males from the Strand Muslim community.

TA) is a Malaysian shrub used to treat various illnesses including male infertility. Considering that TA is also used to improve male fertility and no report 
regarding its safety has been published, this study investigated the effects of a patented, aqueous TA extract on various sperm and testicular functions. Materials and Methods This study 
encompasses two parts (part 1: on spermatozoa
part 2: on TM3-Leydig and TM4-Sertoli cells). Part 1: Semen samples of 27 patients and 13 fertile donors were divided into two groups, 
washed and swim-up prepared spermatozoa, and incubated with different concentrations of TA (1, 10, 20, 100, 2000 &mu
g/ml) for 1 hour at 37°
C. A sample without addition of TA served as control. After incubation with TA,

Full textMore than 20 million infants in the world (15.5 % of all births) are born with low birth weight. Ninety-five % of them are in developing countries. Oral colonization of Gramnegative anaerobes has 
been implicated as a risk factor for preterm delivery of low birth weight (PLBW) infants. The objective of this study was to investigate the association between periodontal pathogens and pre-term 
delivery of low birth weight (PLBW) infants. The study sample included 200 randomly selected women admitted to the department of obstetrics-gynecology of the teaching hospital of Butare in 
Rwanda. Mothers were asked to complete a questionnaire in order to identify factors which might pose a health risk to them and their infants. Gingival crevicular fluid (GCF) was collected from each quadrant of the mother&rsquo
s month (using paper points) within 24 hours of delivery. Ten ml of foetal cord serum samples were collected at delivery and 10 ml of maternal serum samples were 
collected within 48 of delivery. GCF was examined by PCR for the presence of 5 periodontopathogens and ELISA was used for the evaluation of cytokines (IL-6 and IL-10) and immunoglobulins (IgM, IgG) in foetal cord and maternal blood against the periodontopathogens. P. intermedia showed significant associations either on its own or in combinations with most indicators of 
periodontal disease used in this study, while Aa and members of the red complex were significantly associated with gum bleeding and reduced frequency of tooth brushing. A strong association 
between PLBW and maternal and foetal cord serum sample levels of IL-10 was observed. Also, a good association was observed between PLBW and FCB sample levels of IL-6. Significant 
associations were observed between PLBW and maternal IgG against the different peridontopathogens. The findings of this study may suggest that the levels of maternal IgG and foetal IgM 
against the different periodontopathogens are associated with dissemination of maternal periodontopathogens to the foetus thereby illiciting an inflammatory response which contributes to 
PLBW.

Full textThe aim of this study was to investigate the genetic diversity of the SLC22A1 gene and to deduce its possible pharmacogenetic implications within the Cape Coloured population of South 
Africa
a uniquely admixed population of immigrant Europeans, Asians and the indigenous populations. Recent studies have reported an abundance of polymorphic variants within this solute 
carrier transporter gene encoding for the organic cation transporter 1, as well as evidence linking these variants to an effect on metformin uptake. This study included establishing baseline 
frequency distribution of previously reported alleles for 20 SNP variants within the SLC22A1 gene, as well as the development of SNaPshot®
and Multiplex AS-PCR genotyping assays, and 
also exploring the possibility of using High-resolution melt (HRM) analysis as a costeffective alternative for SNP genotyping. Ethics clearance was obtained from the Ethics Committee of the 
University of the Western Cape. Biological samples in the form of buccal (oral) swabs were collected from 132 unrelated voluntary donors from the Cape Coloured population residing in the 
Cape Metropolitan area. Two SNaPshot®
Multiplex Systems were specifically designed for the study,successfully optimized and used for genotyping. Hundred genetic profiles were then generated for a total of 20 SNP variants on SLC22A1 gene, using this primer extension-based genotyping method that enables multiplexing up 10 SNPs. Population genetics data obtained for 
the investigated SNPs were analysed using various statistical analysis software. Important population genetic parameters were calculated, and possible pharmacogenetics implications were then discussed. Among others, allelic and genotypic frequencies, as well as linkage disequilibrium were determined and compared with world populations. Minor deviation from Hardy- Weinberg equilibrium was observed in the Cape Coloured population. No significantLinkage Disequilibrium between the investigated SNPs was observed in this population. A Multiplex allele specific &ndash
PCR (MAS-PCR) genotyping 
system was successfully designed and optimized for the genotyping of 10 SNPs from the SLC22A1. This system, also developed specifically for this study, was made of 2 multiplexes each covering 5 SNPs. It is an inexpensive genotyping assay that allows for efficient discrimination of SNP polymorphisms in one reaction tube with standard PCR conditions. A pilot study was 
conducted to explore the possibility of using High-resolution melt (HRM) analysis as a cost-effective alternative for SNP genotyping. In addition to genotyping, HRM analysis can be used to scan 
large numbers of samples for novel genetic variations.

Two strategies were considered for the synthesis of 2,3-unsaturated O-glucosyl-1,2,3-triazoles. The reaction between tri-O-acetyl-D-glucal and triazole alcohols gave no stereoselectivity. In fact, β-isomer formation was increased. A second strategy furnished 2,3-unsaturated O-glucosides from tri-O-acetyl-D-glucal and alkynols via Ferrier rearrangement; this methodology employing montmorillonite K-10 doped with FeCl3∙6H2O affords new glycosides in good to excellent yields, short time and high α-stereoselectivity in dichloromethane. Subsequently, the glucosides were coupled with 2-azido-1,4-naphthoquinone to give a new series of 1,2,3-1H-triazolyl O-glucoside derivatives based on click reaction.
Duas estratégias foram consideradas para a síntese de 1,2,3-triazóis O-glicosídeos2,3-insaturados. A reação entre o tri-O-acetil-D-glucal e os alcoóis triazólicos não mostrou seletividade. De fato, a formação do isômero β foi aumentada. Uma segunda estratégia forneceu O-glicosídeos 2,3-insaturados a partir do tri-O-acetil-D-glucal e alquinóis via rearranjo de Ferrier; esta metodologia formou novos glicosídeos, empregando montmorilonite K-10 dopado com FeCl3∙6H2O em diclorometano, em rendimentos de bons à excelentes, baixo tempo reacional e alta α-estereosseletividade. Subsequentemente, os glicosídeos foram acoplados com 2-azido-1,4-naftoquinona formando uma nova série de derivados 1,2,3-1H-triazolil O-glicosídeos através da reação Click.

Full textIn many forensic cases, such as mass disasters reconstruction cases, the recovered DNA is highly degraded. In such incidences, typing of STR loci has become one of the most powerful tools for retrieving information from the degraded DNA. However, as DNA degradation proceeds, three phenomena occur consecutively: loci imbalance, allele dropout and no amplification. To solve the problem of degraded DNA, redesigned primer sets have been developed in which the primers were positioned as close as possible to the STR repeat region. These reduced primer sets were called Miniplexes. Unfortunately, a few of the CODIS STR loci cannot be made into smaller amplicons. For this reason non-CODIS miniSTRs have been developed. The present study was undertaken for the population genetic analysis of microsatellite variation in four South African populations
Afrikaner, Xhosa, Mixed Ancestry and Asian Indian using eight non-CODIS miniSTR loci. These miniSTRs loci were characterized within the populations by estimating the levels of diversity of the markers, estimating the population genetic parameters, and studying the inter-population relationships. All of the miniSTRs were amplified successfully and the genetic variability parameters across all loci in Afrikaner, Mixed Ancestry, Asian Indian and Xhosa were estimated to be in the range of 3 (D4S2364) to 12 (D9S2157) alleles, the total number of alleles over all loci ranged from 100 to 204, the allelic richness ranged from 3.612 to 10.307 and the heterozygosity ranged from 0.4360 to 0.8073. Genetic distance was least between Afrikaner and Asian Indian and highest between Xhosa and Mixed Ancestry. Deviations from Hardy-Weinberg equilibrium were not observed for most of the loci. The low mean FIS (-0.027) and FIT (-0.010) and FST (0.017) values across the populations indicated low level of inbreeding within (FIS) and among (FST) the populations. The Asian Indian population showed higher levels of the inbreeding coefficient, indicating less gene exchange between it and other populations. These 8 markers can be used for genetic investigations and assessing population structure. The study contributed to the knowledge and genetic characterization of four South African populations. In addition, these MiniSTRs prove to be useful in cases where more genetic information is needed.

The particularity of these platforms is characterized by an organizational model based on a cooperative system or on a traditional capitalist system, driven by the search for profit. This research is part of the field of strategy and consists of studying the games of actors, relying more specifically on business ecosystems (abbreviated to ESA), a concept that appeared in the work of Moore (1993, p.76) and is defined as a set of "actors maintaining relationships of coopetition" (Koenig, 2012, p. 210). ESA refers to an economic community of organizations that interact with each other to produce goods and/or services that can create value for customers and for themselves. The ESAs we are studying are grouped around platforms called mobile banking (new entrants) and banking institutions (established firms). The problem that led to our thesis is to understand how the platforms have imposed themselves, by what strategic actions they have managed to make a place for themselves on the market and eventually dominate it. Our research field is Chad and our methodology is based on a qualitative analysis of a case study. The data collected will allow us to retrace the history of events and to study the discourses of the actors involved in the process at work by means of a manual analysis of the corpus. The proposals produced can shed new lighting on the strategic actions of the actors

Full textThis paper presents the results of field research carried out with the children, educators and parents of the Umariaçú II community. The goal of this study is to investigate the actions and reactions of the indigenous children in the teaching and learning process in early childhood education, in the AEGATU DECATUCU Municipal School. The specific goals laid out in this study were: Observe within the daily life of the school the communal interaction relations among the children and between children and educator in regard to the school activities connected to the pedagogical practice of the teacher in the classroom. Describe the ways in which the family (parents/those/responsible), in relation to the school, participates as true nuclei, being these partners in the education and learning process within the school life of their children. Register, through observation and interaction, how the indigenous children react to formal learning in the class room, confronted with the proposed activities of the educators. With regard to behavior, the children, as all children, have their moments of fun, and with regard to the learning process, these children are very attentive when the educators are explaining things. One perceives that stimulus and motivation must always walk together as a proposal of the educators. Besides this, the research revealed that the parents are not such great collaborators and partners in the [formal] education of their children, because they still have the image that teaching and learning are legacies marked and rooted by family generations. For the educators the school is the institution which tends to make possible many activities to have the presence of the parents in its daily life, even though they are confronted with the challenges which indigenous education faces in its curricula, resources, community, acculturation and social life which is being daily infiltrated by a more urbanized society. Faced with this, the presence of the child remains as a being in development, with dreams, fulfillments, many times being marked by its culture, which the child itself does not have the power to change, but, re-signifies through their ways of life and survival.

Full textAn atomic-level description of the presumed catalytic action of β-galactosidase is described. This large enzyme, from E. coli , carries out two reactions which allow the bacterium to live on the disaccharide lactose. First, it breaks down lactose to the two monosaccharides galactose and glucose. Second, it converts lactose into another disaccharide, allolactose, which is the inducer for the lac operon, and thus is the signal to the bacterium to produce more β-galactosidase. The work is based on high resolution x-ray crystallography and enzyme kinetics. A crystal form of β-galactosidase was isolated that permits data collection up to 1.5 à resolution. Using this crystal form, the structures of several ligands bound to the enzyme were determined. These ligands were chosen to mimic various points in the reaction: binding of substrate, covalent intermediates, transition states, and products. Together these complexes suggest a reaction coordinate for β-galactosidase which clarifies and enhances previous ideas about the reaction mechanism. The reaction includes a conformational change triggered by the progression of the substrate towards the transition state. Additional investigation suggests that this conformational change is involved in determining whether the enzyme carries out its hydrolysis or isomerization reaction. Considerations of the structure in the context of other related enzymes suggest an evolutionary path for β-galactosidase. It is suggested that a progenitor enzyme which catalyzed the hydrolysis of long polysaccharide substrates recruited additional domains which permit β-galactosidase to act on smaller substrates and produce the inducer, allolactose. This dissertation includes both my previously published and co-authored material.
