Abstract:
The pulmonary isozyme of angiotensin-converting enzyme (ACEP) is present in the body both as a cell-associated protein in endothelial, epithelial, and monocytic cells and as a soluble protein in various body fluids including serum. The mechanism by which soluble ACEP is produced in vivo is unknown. Using in vitro transfected cell culture systems, we previously demonstrated that the rabbit testicular isozyme of ACE (ACET), which shares extensive homology with ACEP, is first synthesized as a plasma membrane-anchored ectoprotein and then secreted to the culture medium by cleavage removal of its COOH-terminal membrane-anchored tail. Here, using in vitro cultures of arterial endothelial cells and acutely isolated renal epithelial cells, we demonstrate that ACEP is also cleavage secreted from their natural producer cells. Biochemical and immunological characterization of the in vitro secreted ACEP protein revealed that it is missing the COOH-terminal membrane-anchored region of the cell-associated ACEP. Similar analysis of ACEP proteins present in rabbit serum, lung, and kidney established that ACEP secretion in vivo is also caused by the cleavage removal of the COOH-terminal region of the cell-associated protein. To characterize the proteolytic enzyme responsible for ACEP secretion, we employed rabbit renal proximal tubular epithelial cells and demonstrated significant inhibition of secretion by compound 3, a hydroxamic acid-based inhibitor of specific metalloproteases. In contrast, the inhibitors of chymotrypsin, trypsin, serine, aspartate, and cysteine proteases were ineffective. These results indicate that soluble ACEP production by vascular endothelial and renal epithelial cells, both in vitro and in vivo, is achieved by cleavage removal of its membrane-anchoring COOH-terminal tail by a metalloprotease.

O objetivo do estágio foi analisar o trabalho da ONG na área da boa governação, em~particular o projeto “Sociedade Civil Pelo Desenvolvimento - Mais Transparência,~Melhor Governação” em São Tomé e Príncipe, executado pela ACEP e pela FONG-STP. Como tal, o trabalho proporciona uma contextualização em torno do debate que envolve o conceito de boa governação para melhor enquadrar a análise do projeto supramencionado.
The present study adopts the format of an Internship Report carried out within the Master of International Development and Cooperation at the Higher Institute of Economics and Management – University of Lisbon. This Report is the result of the completion of an internship at the Association of Cooperation Between Peoples (ACEP), based in Lisbon. The goal of the internship was to analyse the work implemented by the NGO in the field of good governance, particularly the project “Civil Society for Development – More Transparency, Better Governance” in São Tomé and Príncipe, carried out by ACEP and FONG-STP. For this purpose, it is provided a contextualization of the debate around the concept of good governance to better frame the analysis of the aforementioned project.

Depuis une vingtaine d'années, l'entreprise québécoise Hydro-Québec est engagée dans le projet de l'Accumulateur à Électrolyte Polymère ACEP. L'avenir de cette technologie apparait riche et prometteur, et ses performances sont proches des attentes les plus exigeantes. Les électrodes sont constituées par une feuille de lithium pour l'anode et- un mélange de polymère et d'oxyde de vanadium pour la cathode. L'électrolyte est, elle aussi, une feuille de polymère. Afin de fabriquer les cellules électrochimiques de cette batterie, Hydro-Québec et ses partenaires se sont intéressés à l'irradiation par faisceau d'électrons de la structure brute constituée de la feuille métallique et des polymères sous forme de solvants. La structure est alors figée par la polymérisation des solvants sous l'action du faisceau d'électrons à haute énergie. L'hétérogénéité de la structure empêche de pouvoir estimer directement la dose déposée dans les couches de polymères, et ainsi de prévoir les conditions d'irradiation optimales. Puisque les méthodes expérimentales ne peuvent fournir que des résultats très limités, le meilleur moyen disponible pour comprendre les phénomènes survenant dans le polymère réside dans la simulation, qui sera faite par la méthode de Monte-Carlo pour ce projet.

The dissertation seeks to understand the relationship of the visually impaired with the radio, and how broadcast content are crucial in everyday social relations and the study group. Part is the assertion that the radio airs daily various aspects of daily life. These aspects are perceived both in radio content, that aims to approximate the listener, in order to gain audience, but also the intimacy generated between the listener and the speaker (the speaker). The research was applied to the Association of the Blind of Piauí (ACEP), focusing on the observation of the regulars that environment, and the interference of the communication medium, radio, in daily relations of the members of the institution. Theoretically, the work is based on studies of radio which include: history, functions and features and Sociology of Everyday Life, with the main theoretical , the Frenchman Michel Meffesoli . The methodological approach is ethnography , divided into three stages of development: application of closed questionnaires ( 100 visually impaired ACEP , exploratory ) , in-depth interview ( conducted with some members of the association and arranged at work) and participant observation ( performed both in the ACEP , as the monitoring of two members for one day ) . The empirical analysis of object points that radio can be seen at various times of daily public studied, this being a decisive factor in socialities and daily make the blind.
A dissertação busca compreender a relação do deficiente visual com o rádio, e como os conteúdos veiculados são determinantes nas relações sociais e cotidianas do grupo em estudo. Parte-se da assertiva, que o rádio leva ao ar diariamente vários aspectos do cotidiano. Esses aspectos são percebidos tanto no conteúdo radiofônico, que objetiva a aproximação do ouvinte, com o intuito de ganhar audiência, como também da intimidade gerada entre quem escuta e quem fala (o locutor). A pesquisa foi aplicada na Associação do Cego do Piauí (ACEP), tendo como foco a observação dos frequentadores daquele ambiente, e a interferência do meio de comunicação, rádio, nas relações cotidianas dos membros da instituição. Teoricamente, o trabalho está fundamentado nos estudos sobre rádio, que englobam: histórico, funções e características e na Sociologia do Cotidiano, tendo como principal teórico, o francês Michel Maffesoli. O procedimento metodológico é a etnografia, dividido em três etapas de desenvolvimento: aplicação de questionários fechados (a 100 deficientes visuais da ACEP, de caráter exploratório), entrevista em profundidade (realizadas com alguns membros da associação e dispostas no trabalho) e observação participante (realizada tanto na ACEP, como o acompanhamento de dois membros durante um dia). A análise do objeto empírico aponta que o rádio pode ser percebido em vários momentos do cotidiano do público estudado, sendo este, um elemento determinante nas socialidades e no fazer diário do cego.

In 1998, a seminal study on adverse childhood experiences (ACEs) and subsequent health risks catapulted ACEs and the study questionnaire into the zeitgeist. However, its childhood sexual abuse (CSA) item is problematic as it requires the perpetrator have been 5-years or older than the victim. To assess whether some survivors’ CSA is not identified by the current item, whether their exclusion prevents access to services requiring a four-threshold ACE score, and how their health outcomes compared to other CSA groups and controls, an international sample of 974 women completed an online survey assessing their current health and CSA history using the original item and an experimental item without the 5-year modifier. Results indicated many CSA survivors are not identified by a 5-year modifier, exclusion has service implications for some, and on most variables, they had increased adverse health outcomes compared to controls. Means of assessing CSA must be thoughtfully revised.

Isolated hybrid wind microgrids operate within three distinct modes, depending on the wind resources and the consumer grid demand: diesel-only (DO), wind-diesel (WD) and wind-only (WO). Few successful systems have been shown to consistently and smoothly transition between wind-diesel and wind-only modes. The University of Alaska – Fairbanks Alaska Center for Energy and Power (ACEP) has constructed a full scale test bed of such a system in order to evaluate technologies that facilitate this transition. The test bed is similar in design to the NREL Power Systems Integration Laboratory (PSIL) and sized to represent a typical off-grid community. The objective of the present work is to model the ACEP test bed in DO and WD modes using MATLAB™ SIMULINK© and then validate the model with actual full-scale laboratory measurements. As will be shown, the frequency responses are grouped into three classifications based on their behavior. The model is shown to be successful in describing the frequency response of relatively small (0.15 per unit) steps in load. Modifications to the excitation system model are discussed which could improve the accuracy for larger steps in load. The ACEP test bed and associated SIMULINK© model are to be used in future work to support investigating WO operation.

The mixing of fluids using AC Electrokinetic is presented in this paper. Both AC electrothermal (ACET) and AC electroosmosis (ACEO) techniques are investigated for mixing operation. AC electrokinetic mixing utilizes the characteristics of short diffusion distance and large specific interface area, and the characteristics of laminar flow and multiphase flow in a microchannel. The proposed mixer will have advantages of easy implementation and compatibility with microchip fabrication. Furthermore low and high conductive fluid has been experimented for mixing operation. In this research, the ACET and ACEO mixing will be optimized by surface modification using a biocompatible hydrophobic nanocomposite monolayer. This coating will modify the mixer surface to a hydrophobic surface and improve the friction losses at the interface, and eventually increase the mixing rate. Both ACEO and ACET flow is a promising technique in microfluidic mixing toward laboratory automation applications, such as clinical diagnostics and high-throughput drug screening. But the mixing efficiency and type of AC electrokinetic usage depends on the conductivity range of the fluids. These mixers can be integrated with the lab-on-a-chip and can provide inexpensive disposable devices.

Abstract AC electrokinetics is one of the widely used methods as an actuating mechanism in the lab-on-a-chip devices because of the absence of moving mechanical parts. In this paper, an analysis is done by using two perpendicular electrodes which are placed in V-shape to each other while maintaining an angle of 45 degrees with the third horizontal electrode. A semiconductive fluid was used to observe the microfluidic behavior and characteristics under the application of a DC biased AC signal. Applying the AC signal produced two different mechanisms of electrokinetics such as DC-biased AC electrothermal (ACET) and DC-biased AC electroosmosis (ACEO). In the ACEO process, a time-varying voltage was applied to the electrodes to create the double layer capacitance and zeta potential. This ACEO mechanism required lower voltage. On the other hand, the AC Electrothermal (ACET) flow produced a non-uniform electric field that generated spatially varying heat sources which in turn created a non-uniform temperature distribution. Two surface characteristics were also analyzed experimentally; one of these was by using the hydrophobic surface and the other used glass-surface only. At the microscale, mechanical microdevice encounter very high flow resistance and put stringent requirements on the strength of fluid channels, chambers and the interconnects. While many types of microfluidic manipulations can be effectively done by AC electrokinetics techniques, current research work focused on the observation of varying frequencies and voltages and their effects on microfluidic manipulations.

Abstract:
The recent discovery of angiotensin converting enzyme 2 (ACE2) as a functional peptidase within the renin-angiotensin system (RAS) has added a new layer of complexity to the enzymatic cascade of this hormonal system. ACE2 is highly expressed in the proximal tubules of the kidney, an important tissue site involved in blood pressure regulation. Therefore, we derived a model for the processing of Ang I which is the immediate precursor to the biologically active peptides Ang II and Ang-(1-7) based on metabolism data in isolated proximal tubules of the sheep kidney (1). Given the individual experimental velocities for several peptidases expressed in the proximal tubules including ACE, ACE2 and neprilysin, rate constants were calculated to describe the conservation equations for the processing of Ang I, Ang II and Ang-(1-7) We modeled the system with Ang I as the initial substrate and peptide concentrations for the downstream products were calculated using Euler’s method.
