Full textPulchellin is a type 2 ribosome-inactivating protein (RIP) isolated from seeds of the Abrus pulchellus tenuiflorus plant. The DNA fiagment encoding Pulchellin A-chain (PAC) was cloned and inserted in pGEX-5X to express the recombinant pulchellin Achain (rPAC) as a fusion protein in Escherichin coli. The deduced amino acid sequence analyses of the rPAC presented a high sequential identity (> 86%) with the A-chain of abrin-c. The ability of the rPAC to depurinate rRNA in yeast ribosome was also demonstrated in vitro. Intending to validate the toxic activity we promoted the in vitro association of the rPAC with the recombinant pulchellin binding chain (rPBC). Both chains were incubated in the presence of a reducedloxidized system, yielding an active heterodimer (rPAB). The rPAB showed an apparent molecular mass of about 60 D a similar to the native pulchellin. The toxic activities of the rPAB and native pulchellin were compared by intraperitoneal injection in mice using different dilutions. The rPAB was able to kill 50% of the tested mice with doses of 45μg.kg-1. Our results indicated that the recombinant heterodimer presented toxic activity and a conformational pattern similar to pulchellin. Studies using tissue cultures were also performed to investigate the presence of the pulchellin in callus established from seed explants of A. pulchellus. Cotyledon segments of immature seeds were inoculated in basal medium MS supplemented with different concentrations of auxin, citokinin and sucrose in order to determine the best callus induction. The pulchellin expression was monitored in callus cultures by RT-PCR and biological activity. The calli obtained aRer 35 days were freeze dried, macerated and submitted to extraction of total RNA and proteins. A specific DNA fragment codifying the A-chain pulchellin was amplified from callus RNA suggesting the synthesis of the protein. This was confirmed in the calli crude extract that showed haemagglutinating activity against rabbit blood cells and a high intraperitoneal toxicity to mice. The crude extract was also submitted to affinity chromatography on a Sepharose-4B column. The retained protein, showed to be composed by two main bands in polyacrylamide gel electrophoresis, in denaturating conditions, with a similar pattern to the results obtained with seeds pulchellin.

Full textSchistosomiasis, caused by Schistosoma mansoni, is one of the most prevalent parasitic diseases in humans, its intermediate host is a freshwater mollusc from the species Biomphalaria glabrata. For the control of this disease, besides treatment of infected patients, it is also important that the mollusc populations are controlled. In order to find strategies for control of molluscs, this study aimed to investigate the activity of plant extracts as inhibitors of parasite development in mollusc. The main objective was to evaluate the biological activity of extracts from the leaves of two plants from Guinea-Bissau (Abrus precatorius and Khaya senegalensis) in Biomphalaria glabrata exposed to Schistosoma mansoni. The parameters evaluated were survival / mortality, oviposition, elimination of cercariae and its viability on the definitive host (mouse) as well as DNA damage in the molluscs through the technique of Comet Assay. Although the results are very preliminary, they seem to indicate that the extracts from leaves of Abrus precatorius and Khaya senegalensis don’t have molluscicidal activity, as the mortality rate was low in the groups of molluscs exposed to the extracts and mortality occurred later in the bioassay and not during the first 24 hours post-exposure. Through the technique of Comet assay we found that the DNA from molluscs exposed to extracts showed damage but that it was not significant. Regarding the group of molluscs infected with S.mansoni and exposed to the extracts, only those exposed to the extract of A. precatorius and DMSO showed significant DNA damage. Exposure to extracts induced an increase in the release of cercariae by the molluscs when compared to the group of molluscs infected only with S.mansoni, however these differences were not statistically significant. It was not detected an increase in the average number of eggs produced by B.glabrata exposed to the extracts. Although there were differences in the mean number of eggs produced between the various experimental groups, they were only significant between the group of molluscs infected and exposed to the extract from Khaya senegalensis and in the group infected but not exposed to any of the extracts (specifically in the control for the molluscs exposed to the extract of Khaya senegalensis). On the other hand the extracts appear to affect the viability of the infective forms in the definitive host (cercariae), and in the intermediate host (miracidia).

Full textSince the new penal code of 1994, misappropriation funds and breach of trust find one's way in a common repressive sphere of operations. These violations lead to a difficulty of differentiation and a confusion risk, which don't go with legal security principles and criminal legality. Indeed, risk materialize through the opportunity of a dual qualification suggested to judges. The doctrine, in its majority, is favourable to a reform in this context. Some of the authors aim at precise aspect of misappropriation funds as the prescription delay. Others consider that a content reform is necessary. Therefore, simple deletion of misappropriation funds violation would not be contemplate as a solution. Then, it's advisable to determinate in what extent breach of trust will supplant misappropriation funds. In this way, it is also necessary to find improvement for this incrimination in order to cover completely the repressive sphere of operations

Full textTaxpayers can abuse double tax treaties. The primary form of abuse is treaty shopping which defeats the bilateral nature of tax treaties. The concept of tax treaty abuse needs to receive a firm theoretical basis. A comparative study of its mechanism and its elements reveals that tax treaty abuse, form of evasion of law, can be defined as a situation where a taxpayer intentionally invokes a tax treaty in a manner that frustrates the spirit and purpose of the treaty (invoked rule) in order to avoid the application of the domestic rule which gives rise to tax liability (avoided rule). A conflict of norms can arise if the treaty benefit claimed by the taxpayer is disallowed by operation of a domestic anti-avoidance rule. However, the risk of incompatibility is limited due to the deficient nature of tax treaties, heavily relying on domestic law for their application, as well as to the duty to interpret treaties in good faith. The attitude of states with respect to tax treaty abuse is incoherent. On one hand, the number and the diversity of anti-abuse rules becomes ever greater while at the same time there are less business reasons to engage in treaty shopping because of the gradual erosion of source taxation. On the other hand, states do not fight treaty abuse with equal determination. In fact, the United States are the only country to engage in a coherent line of policy against treaty abuse through specific anti-abuse rules

Full textBecause of the extraordinary means to which the public sector has access, public competitors often occupy a stronghold on the market. This privileged increases the risk of anticompetitive practices being adopted by public actors and therefore obliges the public and juridical authorities in charge of market competition to keep a particularly keen watch on this sector's activities. In this context, the abuse of a dominant position, as defined in article 102 of the TFUE, is a central issue. Far from overlooking the specificity of the public sector, the European Law on the abuse of a dominant position constitutes a model, which deserves to be upheld. The scope of this model, which is know to be particularly broad, includes the case of public actors, which is explicitly addressed in article 106 of the TFUE. Public competitors easily meet the criteria defining those in a market power, and the rules on abuse of a dominant position enable the authorities to control these players' market activities extremely closely. The regulatory approach adopted in the European model for competition can be explained in terms of the ordo-liberal intellectual background. It is in the public sector that this model has been expressed most effectively. However, article 102 of the TFUE has not been spared by the wave of modernisation which has pervaded competition law. The theoretical concepts underlying these widely used regulatory tools, which have been criticized for being too strict and too rigid, are being completely revised. National authorities and the European Commission, inspired by the latest economic analyses, are now tending to judge the anti-competitive behaviour of the most powerful firms in terms of its concrete effects on the market. Consumer welfare is becoming the ultimate goal. However, the European Court of Justice still maintains its traditional approach to preventing the misuse of competitive power: it supports a legal system designed to protect competitive structures. This model is still highly relevant when it comes to examine the public sector conducts. Since its premises are sufficiently flexible to be able to support both public privileges and general welfare, this model seems to provide an efficient means of regulating competition in the public sector

Full textCompetition provisions seek to regulate the abuse of economic power in unequal commercial relationships. However, in the process, such provisions should not have the effect of adversely impacting the market or harming the proper functioning of other competition rules. When there exists a significant power disparity between parties, the stronger party can impose unequal conditions on the weaker party, particularly through what is called the "fear factor". During the duration of the contractial relationship, the weaker party will not be able to stand up to the stronger party in fear of the commercial relationship being terminated. This is why oppressive behaviors continue in reality, despite the presence of numerous regulations designed to prevent such behaviors. Law makers have a tendency to react by hastily enacting ad hoc regulations. The adaptability of powerful corporations to circumvent the rules is often overlooked. Ultimately, abuses of economic power can only be regulated on a sustainable basis through the creation of a more balanced and less concentrated market. It is in this context that a truly structural approach should be considered. This analysis compares the principal measures against abuses of economic power employed in France and Korea, which sometimes adopt different responses to similar circumstances

Full textOver the last decade, the growing popularity of Online Social Networks has attracted a pervasive presence of social spammers. While this presence has started with spam advertising and common scams, the recent years have seen this escalate to the far more concerning mass manipulation attempts. This targeted and largely automated abuse of social platforms is risking the credibility and usefulness of the information disseminated on these platforms. The social spam detection problem has been traditionally modeled as a supervised problem where the goal is to classify individual social accounts. This common choice is problematic for two reasons. First, the dynamic and adversarial nature of social spam makes the performance achieved by features-based supervised systems hard to maintain. Second, features-based modeling of individual social accounts discards the collusive context in which social attacks are increasingly undertaken. Acting synchronously allows spammers to gain greater exposure and efficiently disseminate their content. Thus, even when spammers change their characteristics, they continue to act collusively, inevitably creating links between collusive spammingaccounts. This constitutes an unsupervised signal that is relatively easy to maintain and hard to evade. It is therefore beneficial to find a suitable similarity measure that captures this collusive behavior. Accordingly, we propose in this work to cast the social spam detection problem in probabilistic terms using the undirected graphical models framework. Instead of the individual detection paradigm that is commonly used in the literature, we aim to model the classi_cation task as one of joint inference. In this context, accounts are represented as random variables and the dependency between these variables is encoded in a graphical structure. This probabilistic setting allows to model theuncertainty that is inherent to classification systems while simultaneously leveraging the dependency that _ows from the similarity induced by the spammers collusive behavior. We propose two graphical models: the Markov Random Field with inference performed via Loopy Belief Propagation, and the Conditional Random Field with a setting that is more adapted to the classification problem, namely by adopting the Tree Reweighted message passing algorithm for inference and a loss that minimizes theempirical risk. Both models, evaluated on Twitter, demonstrate an increase in classification performance compared to state-of-the-art supervised classifiers. Compared to the Markov Random Field, the proposed Conditional Random Field framework offers a better classification performance and a higher robustness to changes in spammers input distribution

Full textThe qualified behaviours of abuse by the criminal law present criteria which establish the existence of a general concept of abuse in this branch of law. Thus any repressive text which aims at an abuse requires that it be the factor of a person holding a legal power expressly determined by the penal norm. This prerogative, granted or recognized by the law and making it possible for its holder to impose his will on others in an interest at least partially distinct from his, is conceived in an extensive way by the criminal law which admits that it can result from a situation of right or fact. Prerequisite to the abuse, necessary to its characterization, the power comes to delimit the field of it and to found repression of it. But the abuse in criminal law necessarily, leading its author to the illegality, if it is caused by the power, cannot be defined as while being the exercise. The definition of abuse forces to resort to the concept of authority connected the power, characterized by the aptitude for being believed or being obeyed recognized by the victim to the author of the abuse because the confidence which causes the legal power that he holds on her. The protean nature of the authority and the system which it forms with the power from which it remains distinct give all its coherence to the abuse, which the consists of an excess of power by the diversion of the authority which is connected there. This unit definition makes it possible to identify among the accused acts those which can or not be qualified abuse. Beyond, the concept of abuse has some utility for the criminal law, since it influences the nature of the incrimination and the methods of the repression of the abusive behaviours

The 1st part of this thesis analyses the figure of liminal man which has emerged through the discourse and actions of local decision makers in response to the increased presence of homeless in the public arena. The 2nd part deals with the various economic, institutional and emotional ties by which homeless are bound, provides biographies and recollections, and examines the importance for them of both place and networking to relieve a sense of rejection and insecurity which can destroy their persona. The 3rd part analyses how, between mutual rejection and mutual recognition, and between the institutional, collective and public spheres, viable relationships between the homeless of the “Maison de Nanterre” and the inhabitants of the neighbourhood have progressively evolved

Full textIn Canada, the strawberry producers are facing several major challenges. The production period is short, the competition is strong and the production has to deal with important telluric pathogens. The soilless production could help overcome these problems. To maximize productivity, experimental device was set up in 2013 and 2014 to Onésime Pouliot farm on the island of Orleans in the province of Quebec. This project seeks to demonstrate that in soilless production, the umbrella like rain shelter can produce more marketable fruit and decrease the risk of foliar disease. The project also aimed to evaluate some methods of culture that could help to extend the production period (greenhouse forcing) or optimize the use of water and fertilizers (capillary mat, alternate substrate). Five treatments were tested, the first four under umbrella like rain shelters (T1-T4) and the last one without coverage (C). The treatments T1 was grown on peat (PE) and T2 on a mixture of sawdust and peat (PS25). For T3, plants forced greenhouse were tested to extend the harvest. T4 treatment was cultivated on PE substrate and deposited on a capillary mat. When compared with the control, umbrella covered treatments led to a significantly lower incidence of late blight [Sphaerotheca macularis (Wall. Ex Fries)] and a higher yield of marketable fruit. In protected growing conditions, the plants forced in greenhouses achieved peak production at the most profitable period for fresh strawberries in Quebec for the summer of 2013. The capillary mat (T4) proved interesting since for a similar yield fruit, it had to use less water and fertilizer. Our results put lights on the potential of using umbrella production above ground strawberries in Quebec and offers appropriate recommendations as to the most cost-effective and sustainable production methods for local growers.
