Full textThe work carried out focuses on the characterisation of a resistance mechanism described in E. faecium. A single nucleotide mutation in the eat(A) gene leads to the substitution of an amino acid (T450I) coding for the variant form of the Eat(A)v protein. This mutation leads to cross-resistance to different antibiotics (Lincosamides, Streptogramins A and Pleuromutilins) which all target the peptide transfer centre of the ribosome. Eat(A) is one of the ABC-F proteins, some of which are Ribosome Protection Proteins, while the role of the wild type remains unknown. The aim of this thesis is to characterise the biological activity of the two forms of the Eat(A) protein. The mutation causing the resistance phenotype is located 3 residues upstream of one of the two ATP hydrolysissites. A difference in ATPase activity between the native and the variant form of the protein may be at the origin of the resistance mechanism. To address these questions, we developed a purification method to obtain sufficient soluble protein for in vitro assays. We studied the impact of the T450I mutation on the ATPase activity of the proteins, revealing the importance of isoleucine in the biological activity. We also developed an in vitro translation system using E. faecium S30 extracts to study the impact of the two Eat(A) proteins on translation by monitoring GFP synthesis, with or without antibiotics. Finally, we undertook the characterisation of the interactions between the two forms of Eat(A) and the E. faecium ribosome using cryo-EM technology

Full textProteins belonging to the ABC-F family are found in prokaryotes as well as eukaryotes. They interact with the ribosome and are considered as translation factors. The Escherichia coli strain (MG1655) has 4 ABC-F paralogs named: EttA, YheS, YbiT and Uup. Here I present a study of the physiological and functional role of E. coli ABC-F proteins and the Streptococcus pneumoniae MsrD protein. My work has shown that the ABC-F proteins studied are important for bacteria adaptation to stress conditions. Bacteria deleted from the ettA gene are more sensitive to salt stress, bacteria deleted from the uup gene are more sensitive to acid stress while the overexpression of the protein YheS or MsrD in bacteria provide resistance to macrolides. I also demonstrated that the ettA gene is important for the growth of E. coli under conditions of saline stress at low phosphate concentration when the amino acids, malate or fumarate are used as the only carbon source. The deletion of the ettA gene makes bacteria more sensitive to salt stress and causes dysregulation of certain genes linked to the malate / glyoxylate metabolism and genes involved in ribosomal activity. The deleted strain of the ettA gene under-expresses the malate synthase aceB gene by post-transcriptional regulation. The under- expression of malate synthase in bacteria promotes their growth in the glyoxylate medium. The deletion of the ettA gene also induced overexpression of ribosome modulation factor (RMF) in LB medium, which caused an increase in the formation of 100S ribosomes

Full textBtuCD-F is a binding protein-dependent ABC transporter system that uses the power of ATP hydrolysis to pump vitamin B₁₂ into the cytoplasm of E. coli. The crystal structure of BtuF, the protein that binds vitamin B₁₂ and delivers it to the transporter, BtuCD, has been solved by x-ray crystallography. BtuF is a bi-lobed protein and B₁₂ is bound in a deep cleft formed at the interface between the two lobes. A stable complex between BtuF and BtuCD is demonstrated to form in vitro and was modeled using the individual crystal structures. Two conserved surface glutamates from BtuF may interact with conserved arginine residues on the periplasmic surface of the BtuCD transporter, playing a role in docking and the transmission of conformational changes.
BtuCD has also been reconstituted in vitro into proteoliposomes. In the presence of ATP, BtuCD proteoliposomes can mediate uptake of vitamin B₁₂ in a BtuF dependent fashion. In the absence of ATP, B₁₂ appears to become sequestered between BtuF and BtuCD. The ATPase activity of BtuCD was examined in proteoliposomes as well as in detergent solution. BtuCD has a significant basal rate of hydrolysis under all conditions tested, and B₁₂-bound and apo-BtuF can stimulate that rate. Interestingly, the rate of ATP hydrolysis, as well as the effect of BtuF, vitamin B₁₂ and sodium ortho-vanadate on that rate, is different in each detergent and lipid environment. These results indicate that ABC transporters are highly sensitive to their environment and underline the importance of detergent or lipid choice in functional reconstitution and membrane protein crystallization experiments. Our results lead us to propose a revised model of the ABC transport cycle.

Zr-based AB2 hydrogen storage alloys prepared by inert gas atomization only have 73% of the discharge capacity of the arc-melting alloys. High solidification rates in atomization process lead the formation of an amorphous phase and dense oxide layers on the surfaces of atomized powders. The outside layers of atomized powders retarded the initial activation rate and decreased the discharge capacities of electrodes. The hydrogenation ability of atomized powders was reduced because hydrogen could not pass through the outside amorphous and oxide layers to form hydrides in the alloy powders. Although F-treatment did not show much improvement on the discharge capacity of arc-melting alloy powders, the atomized powders treated by HF immersion possessed better activation properties and discharge capacities. This was because of the removal of oxide layers on the surfaces by the F-treatment. Larger atomized powders showed a profoundly dendritic structure on the surface as compared to that on smaller powders. They also have a relatively high capacity (305mA/g) after 60min. F-treatment. The F-ions preferably dissolved the oxide layer on the dendrites, and form highways for hydrogen passage. The channels created by F-treatment were able to connect the surfaces and Ni-rich layers inside of atomized powders. F-treatment also enhanced the rate capability of alloy electrodes and lowered the oxygen content of surface layers. F-treated atomized powders after 1-month exposure in air have a high discharge capacity and a good initial activation rate. But after 6 months storage in air, the discharge capacity decayed to that of untreated atomized powders. After immersion in water for 1 month, not only the untreated powders but also the F-treated powders were deteriorated, as caused by the formation of oxide on the surface. According to AES surface analysis of alloy powders, the deterioration of the discharge capacity and activation was due to the fact that the oxide reformed on the new surface created by F-treatment, and retards the hydrogen passage into the atomized powders.
