Abstract:
A novel cold-adapted siphovirus, vB_PagS_AAS21 (AAS21), was isolated in Lithuania using Pantoea agglomerans as the host for phage propagation. AAS21 has an isometric head (~85 nm in diameter) and a non-contractile flexible tail (~174 × 10 nm). With a genome size of 116,649 bp, bacteriophage AAS21 is the largest Pantoea-infecting siphovirus sequenced to date. The genome of AAS21 has a G+C content of 39.0% and contains 213 putative protein-encoding genes and 29 genes for tRNAs. A comparative sequence analysis revealed that 89 AAS21 open reading frames (ORFs) code for unique proteins that have no reliable identity to database entries. In total, 63 AAS21 ORFs were functionally annotated, including those coding for the proteins responsible for virion morphogenesis, phage-host interactions, and DNA metabolism. Proteomic analysis led to the experimental identification of 19 virion proteins, including 11 that were predicted by bioinformatics approaches. Based on comparative phylogenetic analysis, AAS21 cannot be assigned to any genus currently recognized by ICTV and may represents a new branch of viruses within the family Siphoviridae.

Abstract:
A cold-adapted siphovirus, vB_PagS_AAS23 (AAS23) was isolated in Lithuania using the Pantoea agglomerans strain AUR for the phage propagation. The double-stranded DNA genome of AAS23 (51,170 bp) contains 92 probable protein encoding genes, and no genes for tRNA. A comparative sequence analysis revealed that 25 of all AAS23 open reading frames (ORFs) code for unique proteins that have no reliable identity to database entries. Based on the phylogenetic analysis, AAS23 has no close relationship to other viruses publicly available to date and represents a single species of the genus Sauletekiovirus within the family Drexlerviridae. The phage is able to form plaques in bacterial lawns even at 4 °C and demonstrates a depolymerase activity. Thus, the data presented in this study not only provides the information on Pantoea-infecting bacteriophages, but also offers novel insights into the diversity of cold-adapted viruses and their potential to be used as biocontrol agents.

Abstract:
Vip3 proteins are increasingly used in insect control in transgenic crops. To shed light on the structure of these proteins, we used the approach of the trypsin fragmentation of mutants altering the conformation of the Vip3Af protein. From an alanine scanning of Vip3Af, we selected mutants with an altered proteolytic pattern. Based on protease digestion patterns, their effect on oligomer formation, and theoretical cleavage sites, we generated a map of the Vip3Af protein with five domains which match some of the domains proposed independently by two in silico models. Domain I ranges amino acids (aa) 12–198, domain II aa199–313, domain III aa314–526, domain IV aa527–668, and domain V aa669–788. The effect of some mutations on the ability to form a tetrameric molecule revealed that domains I–II are required for tetramerization, while domain V is not. The involvement of domain IV in the tetramer formation is not clear. Some mutations distributed from near the end of domain I up to the end of domain II affect the stability of the first three domains of the protein and destroy the tetrameric form upon trypsin treatment. Because of the high sequence similarity among Vip3 proteins, we propose that our domain map can be extended to the Vip3 family of proteins.

Abstract:
The experiment was carried out at the experimental farm of Bangabandhu Sheikh Mujibur Rahman Agricultural University (BSMRAU), Salna, Gazipur during 2015 using twenty promising genotypes (AA21 to AA40) of green amaranth in Randomized Complete Block Design (RCBD) with three replications to study the variability, genetic parameters, association of yield related morphological traits to foliage yield and to find out the direct and indirect effects of components characters towards foliage yield. The significant analysis of variance revealed wide range of variability among the twenty genotypes of green amaranth. Except for stem diameter, foliage yield and all the yield contributing morphological traits had high heritability including high genetic advance in percent of mean, high to moderate genotypic and phenotypic variances and coefficient of variations with minimum differences of PCV and GCA indicated that the traits were governed by additive gene actions and had a great scope of selection for the improvement of green amaranth. Correlation study revealed that total biomass and shoot weight showed significant association with foliage yield per m2. Hence, selection based on total biomass and shoot weight could remarkably improve the foliage yield of green amaranth. However, the path coefficient analysis revealed that shoot weight had the highest positive direct effect (1.19) with significant genotypic correlation on foliage yield. It indicated that direct selection based on shoot weight would be effective for the improvement of foliage yield of green amaranth. Mean performance study revealed that the genotypes, such as AA21, AA22, AA24, AA25, AA27, AA28, AA31, AA36 and AA39 had high foliage yields including most of the yield related morphological traits over the grand mean. These genotype could be selected as high yielding varieties or parental lines for future breeding program.
