Abstract:
ABSTRACT Enterocin AS-48 is a cyclic peptide produced by Enterococcus faecalis S-48 whose genetic determinants have been identified in the conjugative plasmid pMB2. A region of 7.8 kb, carrying the minimum information required for production of and immunity against AS-48, had been previously cloned and sequenced in pAM401 (pAM401-52). In this region, the as-48A structural gene and as-48B, as-48C, as-48C1 , as-48D, and as-48D1 genes and open reading frame 6 (ORF6) and ORF7 had been identified. The sequence analysis carried out in this work in the BglII B fragment (6.6-kb) from pMB2 cloned downstream from the last ORF identified (ORF7) revealed the existence of two new ORFs, as-48G and as-48H, necessary for full AS-48 expression. Thus, JH2-2 transformants obtained with the pAM401-81 plasmid became producers and resistant at the wild-type level. Tn5 disruption experiments in the last genes, as-48EFGH, were not able to reproduce these expression levels, confirming that expression of these genes is necessary to get the phenotype conferred by the wild-type pMB2 plasmid. The as-48EFGH operon encodes a new ABC transporter that could be involved in producer self-protection. On the basis of the observed similarities, As-48G would be the ATP-binding domain, the deduced amino acid sequences of As-48E and As48-H could be assigned as transmembrane subunits, and As-48F, with an N-terminal transmembrane segment and a coiled-coil domain, strongly resembles the structure of some known ABC transporter accessory proteins whose localization in the cell is discussed. This cluster of genes is expressed by two polycistronic mRNAs, T2 and T3, in JH2-2(pAM401-81) in coordinate expression. Our results also suggest that expression of T3 could be regulated, because in JH2-2(pAM401EH) transformants, T3 was not detected, suggesting that these genes do not by themselves confer immunity, in accordance with the requirement for the as-48D1 gene for immunity against AS-48.

Abstract:
Purpose: To evaluate the effect of microRNA-489 (miR-489) on pituitary prolactinoma and its mechanisms of action. Methods: MMQ and GH3 cells were transfected with miR-489, cell viability assessed with cell counting kit-8 (CCK-8), and clone spots was evaluated by colony formation assay. Transwell assay was applied to measure cell migration and invasion while TargetScan was employed to the presumed targets of miR-489, followed by luciferase reporter assays. was MiR-489 and p21-activated kinase 3 (PAK3) gene expression were determined by quantitative reverse transcription-polymerase chain reaction (qRT-PCR. Protein levels of PAK3 were measured using western blots. Results: Transfection significantly increased miRNA-489 levels (p < 0.01). Cell viability, number of clone spots, as well as cell migration and invasion diminished in MMQ and GH3 cells following miR-489 transfection when compared to miR-NC mimic group (p < 0.01). The presumed binding site of miRNA- 489 was located in 3′-untranslated region (UTR) of PAK3, and miR-489 transfection repressed luciferase activity with the wild-type 3′-UTR (p < 0.05). In addition, miR-489 decreased PAK3 levels in MMQ and GH3 cells. Knockdown of PAK3 significantly suppressed cell viability, clone formation ability, as well as cell migration and invasion when compared to negative control (p < 0.01). Conclusion: MiR-489 overexpression suppresses pituitary prolactinoma by targeting PAK3, thus providing a potential therapeutic strategy for the management of pituitary prolactinoma.

Abstract:
MicroRNAs (miRNAs) have been shown to be involved in bladder cancer progression. miR-489 (also known as miR-489-3p) was recently reported to be a tumor suppressor in several cancers. However, its exact role and mechanism in the progression of bladder cancer are largely unknown. In this study, we explore the role of miR-489 in the proliferation and invasion of human bladder cancer cells. The miR-489 expression levels were detected in bladder cancer and normal adjacent tissues, as well as in human normal bladder epithelial cells and bladder cancer cell lines. The results showed that miR-489 was sharply reduced in bladder cancer tissues and cell lines. Then the miR-489 mimic or oligo anta-miR-489 was transfected into T24 and UMUC3 bladder cancer cell lines. The results showed that the miR-489 mimic greatly increased the miR-489 level and significantly decreased the proliferation and invasion of T24 and UMUC3 cells. In contrast, the anta-miR-489 had a completely opposite effect on miR-489 expression, cell proliferation, and cell invasion. Moreover, bioinformatics and luciferase reporter gene assays confirmed that miR-489 targeted the mRNA 3′-untranslated region (3′-UTR) region of Jagged1 (JAG1), a Notch ligand. In conclusion, miR-489 suppressed proliferation and invasion of human bladder cancer cells.

Abstract:
MicroRNAs (miRs) are small conserved RNA that regulate gene expression. Bioinformatic analysis of miRNA profiles during mouse lung development indicated a role for multiple miRNA, including miRNA-489. miR-489 increased on completion of alveolar septation [postnatal day 42 (P42)], associated with decreases in its conserved target genes insulin-like growth factor-1 ( Igf1) and tenascin C ( Tnc). We hypothesized that dysregulation of miR-489 and its target genes Igf1 and Tnc contribute to hyperoxia-induced abnormal lung development. C57BL/6 mice were exposed to normoxia (21%) or hyperoxia (85% O2) from P4 to P14, in combination with intranasal locked nucleic acid against miR-489 to inhibit miR-489, cytomegalovirus promoter (pCMV)-miR-489 to overexpress miR-489, or empty vector. Hyperoxia reduced miR-489 and increased Igf1 and Tnc. Locked nucleic acid against miR-489 improved lung development during hyperoxia and did not alter it during normoxia, whereas miR-489 overexpression inhibited lung development during normoxia. The 3′ untranslated region in vitro reporter studies confirmed Igf1 and Tnc as targets of miR-489. While miR-489 was of epithelial origin and present in exosomes, its targets Igf1 and Tnc were produced by fibroblasts. Infants with bronchopulmonary dysplasia (BPD) had reduced lung miR-489 and increased Igf1 and Tnc compared with normal preterm or term infants. These results suggest increased miR-489 is an inhibitor of alveolar septation. During hyperoxia or BPD, reduced miR-489 and increased Igf1 and Tnc may be inadequate attempts at compensation. Further inhibition of miR-489 may permit alveolar septation to proceed. The use of specific miRNA antagonists or agonists may be a therapeutic strategy for inhibited alveolarization, such as in BPD.

Abstract:
Abstract Scholars continue to give different dates for Egypt’s second revolt against the Persians: Classicists generally date the revolt to 487–485 or 487/486–485/484 BC; Egyptologists and historians of the Achaemenid Empire generally date it to 486–485/484; while some scholars date it to 486/485–485/484. Such chronological differences may sound small, but they have important consequences for the way the rebellion is understood. The purpose of the present article is therefore twofold: first, it aims to clarify what we can and cannot know about the rebellion’s exact chronology. After a review of the relevant evidence, it will be argued that the best chronological framework for the rebellion remains the one provided by Herodotus’s Histories, which places the rebellion in ca. 487–484. Second, the article will show how this chronology influences our understanding of the geographical extent and social impact of the rebellion. The adoption of Herodotus’s chronological framework, for example, results in a larger number of Egyptian sources that can be connected to the period of revolt than was previously recognized. These sources, it will be argued, suggest that some people in the country remained loyal to the Persian regime while others were already fighting against it. Moreover, they indicate that the revolt reached Upper Egypt and that it may have affected the important city of Thebes.

Abstract:
Acute exacerbation of chronic obstructive pulmonary disease (AECOPD) is becoming a common respiratory disease, leading to increased morbidity and mortality worldwide. Tumor necrosis factor-alpha (TNF-α) is a powerful proinflammatory cytokine involved in the pathogenesis of AECOPD. Therefore, we proposed a close correlation between the TNF-α polymorphism [-308G/A (rs1800629), +489G/A (rs1800610)] and the disease progress of patients with AECOPD. Comparison of the TNF-α genotypes between the 198 AECOPD diagnosed patients groups and 195 healthy peoples suggested their significant differences of the three genotypes (AA, GA, GG) distribution for TNF-α -308 ( P < 0.05 ), but no differences of that for TNF-α +489. We found that patients with TNF-α -308 GA/AA genotypes showed smaller adjacent arterial diameter, thicker bronchial wall, higher bronchial artery ratio, higher bronchial wall grading, and higher frequency of acute exacerbations than those with TNF-α -308 GG genotype. Patients with TNF-α +489 GA/AA genotypes showed the same AECOPD properties as patients with TNF-α -308 except for the high frequency of acute exacerbations. Further experiment showed that the TNF-α -308 and+489 gene polymorphisms could affect the expression level of TNF-α in macrophages, suggesting the involvement of the macrophage population in disease regulation of AECOPD patients with TNF-α -308G/A and+489G/A genotype heterogeneity. In conclusion, the TNF-α -308 G/A genotype was related to AECOPD susceptibility and progress, while the TNF-α +489G/A genotype was related to AECOPD progress, but not AECOPD susceptibility.

Abstract:
Background: Hepatocellular carcinoma is one of the most common malignant tumors found all over the globe. Despite advances in surgery and chemotherapy, the five-year survival rate of patients with hepatocellular carcinoma is still low. It is known that the proliferation of hepatocellular carcinoma cells is closely related to the occurrence, development and prog- nosis of hepatocellular carcinoma. The present work investigates the expression of microRNA-489 (miR-489) in human hepatocellular carcinoma cells and its effect on the biological behavior of human hepatocellular carcinoma cells. Methods: The expression of miR-489 by fluorescence quantitative PCR detection in 30 patients with hepatoblastoma of liver cancer tissues and adjacent tissues was studied. Also, the determination of hepatoblastoma in four cell lines with differ- ent metastatic potential (HR8348, HCT116, HT29 and HEPG2) and the expression of miR-489 during miR-489 simulation process was studied. MTT assay, flow cytometry and Western blot analysis were performed to know the cell proliferation to detect the changes in cell cycle, apoptosis of cells, and SOX4 gene expression respectively. Results: RT-PCR results showed that the cells compared with pre-cancerous tissue, the expression level of miR-489 in hepatocellular carcinoma tissues than in adjacent tissue significantly decreased (P<0.05), and with liver cancer cell metastasis increased (P<0.05); analogue transfection constructed miR-489 overexpressing HEPG2 cell line by microRNA. MTT results showed that miR-489 can inhibit the proliferation of HEPG2 cells, the differences were statistically significant (P<0.05); flow cytometry results showed that miR-489 mimics was transfected into HEPG2 cells at 48 hours had no significant effect on cell cycle distribution (P > 0.05); but miR-489 expression could induce apoptosis, compared with the control group, the apoptosis of miR-489 mimics was significantly increased and the difference was statistically significant (P < 0.05). Conclusion: In conclusion, miR-489 can significantly inhibit the occurrence and development of hepatocellular carcinoma cells. The mechanism may be down regulated by the expression of SOX4 and inhibit cell proliferation. Further this study showed that the tumor cells SOX4 gene as a regulatory factor target the genes of miR-489 in hepatocellular carcinoma. This paper has an special kind of liability or receivable as its focus. When a debt is not paid to a Government body after the expiration of the time allowed for the payment, the amount is subject to be booked under legal procedures provided by law. This debt can be originated from unpaid taxes. Due to the relevance of this source this paper is focused on it. But all the unpaid debts are subject to be collected by judicial means: the only requirement is to be properly registered by the authority. In the Federal level of Government this power is granted to the Federal Treasure General Attorney. All the circumstances grant to this kind of debt a presumptuous liquidness and certainty. It requires some attention the process one translates a word, or an idea. To write this abstract avoiding a free translation to Dívida Ativa was desirable. Even in Portuguese this combination of two antagonistic words is criticized when taking into accounting purposes. One word means liability while the other could means asset. But all these questions related to translation issues tend to be relevant when two different legal systems are involved. And problems can arise if these differences are not take into consideration and a simple translation is adopted. This situation is what we can see in Brazil when the standards issued by Brazilian Authorities are simple copies of the international similar; the subject of this paper is the Brazilian equivalent to IAS 37. And Brazilian environment demands a special attention to its huge tax matters. So combining the main ideas about Dívida Ativa in light of what that standards prescribe, some Financial Statements were studied. Thus in some of them no evidence of the existence of the debt was given

With the advent of the new Code of Civil Procedure (Law no. 13.105/2015), it emerged the obligation of analytical reasoning in judgments, laid down in article 489, paragraph 1, of the CPC, which sets forth the hypothesis in which will be not considered, for all legal purposes, reasoned the judgments. It turns out that part of the doctrine and the jurisprudence opposed barriers to the application of the referred legal dispositive to the procedural labor law, for they understand that it would not be fulfilled the requirements set out in article 769 of the CLT. In other words, under the arguments that there is no gap in the labor process ordinance and that the analytical reasoning would be incompatible with the procedural labor law. On the other hand, the other part of the doctrine and jurisprudence holds that the analytical reasoning is an important legal mechanism in combating judicial activism present in the labor courts, supporting the completion of the requirements set out in the law for implementation of common standards in labor process. Therefore, the doctrinal and jurisprudential debate, it abides to this work to address scientifically the question inherent in the requirement of analytical reasoning of judicial decisions in the labor process, mainly aiming to submit a proposal of a solution to this problem. After long research stabilized in doctrine and jurisprudence, it reaches two main conclusions. The first is that the article 489 of the CPC is not incompatible with the systematic and principiology of the procedural labor law, which is, therefore, perfectly applicable supplementarily to the labor process, before the existence of partial regulatory gap in CLT. Finally, the second conclusion is that it is essential that they are committed to efforts for the creation of a Code of Labor Procedure that contains sufficient mechanisms for the solution of the problems that plague labor relations in the present days, because, only that way, the labor process will return to have its vanguard status of a rapid, simple and effective process model, pursued by other procedural systems.

489, §1º, do CPC/15. Palavras-chave: Decisão judicial; dever de motivação; fundamentação adequada; Estado Democrático de Direito; devido processo legal; CPC/15; art. 489,§1º.

Abstract:
The present study has as object of study the duty to justify the judicial decisions established in art. 489 of the Brazilian Code of Civil Procedure, which, in accordance with the Federal Constitution, established the grounds as an essential element of the judicial pronouncement, but innovated in the conception of §1º which deals with the inherent contours of the obligation to state reasons, and §2º which provides for the justification of a consolidated judicial decision in the weighting of legal rules. Seen in this way is that once consolidated in the judicial decision the preponderance of the implementation of legal norms, the legitimation of the exercise of the judicial activity and thus the guarantee of correction of the legal discourse by means of its consistency is in the duty of justification of the judicial decision an important tool to control the interpretation and application of legal norms. Thus, in view of the dimension of the thematic, several theories were developed in order to better clarify the phenomenon described and with that to trace parameters of its verification, among these the present work took as object of investigation the doctrine undertaken by the German Robert Alexy, which widely received in Brazil has as theoretical framework the development of an argumentative legal theory, besides being responsible for the introduction of the weighting technique as a form of resolution of the normative conflict between principles. For this reason, the purpose of this study is to analyze the duty to state reasons for the judicial decision, set forth in art. 489, §1º and §2º in the light of the discursive theory developed by Alexy, in order to draw the parallel between the theoretical framework chosen and the regulation of the thematic. Such an approach presupposes the necessary verification of the compatibility of that with the current procedural system, so that its scope and limitations are established.

Abstract:
We introduce a new point process, the dynamic contagion process, by generalising the Hawkes process and Cox process with shot noise intensity. Our process includes both self-excited and externally excited jumps, which could be used to model the dynamics of contagion impact from endogenous and exogenous factors of the underlying system. We systematically analyse the theoretical distributional properties of this new process, based on the piecewise-deterministic Markov process theory developed in Davis (1984), and the extension of the martingale methodology used in Dassios and Embrechts (1989). The analytic expressions of the Laplace transform of the intensity process and probability generating function of the point process are derived. A simulation algorithm is provided for further industrial implementation and statistical analysis. Some extensions of this process and comparison with other similar processes are also investigated. The major object of this study is to produce a general mathematical framework for modelling the dependence structure of arriving events with dynamic contagion, which has the potential to be applicable to a variety of problems in economics, finance and insurance. We apply our research to the default probability of credit risk and ruin probability of risk theory.

Abstract:
In this thesis I explore aspects of the tradition of radical Romanticism in a pedagogical context, with the focus on the teaching of English in secondary (11-18 age range) schools in England. I draw on my personal professional history as English school-teacher and teacher educator in this project, alongside more theoretical considerations, aiming towards a creative synthesis between Romantic and critical paradigms. In so doing I look first at the general contexts of such relationships, introducing some of the principal figures in the debate, in Section One. I go on to look more closely, in Section Two, at the nature of experience in education through the lens of slippery but crucial ideas of immersion, criticality and wonder, culminating in some illustrative descriptions of English classroom activity. For Section Three this leads into an exploration of the nature of creativity in education – a concept much used currently, but in need of more careful, critically purposeful and painstaking theorising. Following this discussion, I look in Section Four at aspects of the Secondary English curriculum in more detail, both theoretically and practically, before making a specific study in Section Five of interdisciplinary possibilities – a key area, by definition suggesting the forging of connections in theory and practice and thus tending to bring to the fore both unifying possibilities and challenging disjunctions. In the sense that my project here is to do with developing pedagogy, I explore next, in Section Six, some of the issues in the context of initial teacher education (ITE), currently my own sphere of professional activity. Section Seven represents something of a poetic interlude, if (I hope) an illuminating one. Section Eight is essentially an attempt to tie together the various strands of my study and look towards future possibilities, including the tentative conclusions presented in Section Nine.

Joubert, Pieter. "Trichotomous therapy : a proposed pastoral paradigm / Pieter Joubert." Thesis, North-West University, 2005. http://hdl.handle.net/10394/489.

Abstract:
The relationship between Psychology and Theology has become an issue of increasing concern in recent years for the Christian community. With the growing complexities of cultural differences, the moral decline of society and its drift away from a Christian worldview, the pastoral ministry seems to be as challenging as ever. The problem about the relationship between theology and psychology is that there is growing confusion in today's Christian community about the best way to help people overcome their personal problems of living. In this regard some scholars believe that Christians should only submit to Biblical counselling, while others support psychotherapy, as long as it is integrated with Biblical principles and has a firm Scriptural foundation. In researching the literature on the subject, it was found that various theoretical models have been defined and described as attempts to scientifically justify the union of the above mentioned two points of view. In this thesis the viewpoints of four prominent groups of scholars about the subject are described and analysed in order to determine to what extent will it be possible to identify common denominators amongst their different models and to eventually establish if it is possible to formulate a synthesis about the commonalities so defined. The integration models of the following four prominent groups of authors about the subject were analysed: (1) Larry Crabb (1977); (2) William T. Kirwan (1993), (3) John D. 2 Carter & Bruce Narramore (1979) and (4) Eric L. Johnson & Stanton L. Jones, eds., (2000). These scholar's contributions were chosen for analysis because, from the research about the subject, it was found that these 4 groups of authors made some of the most significant scientific contributions about the integration of theology with psychology. Before attempting to formulate a pastoral paradigm out of a synthesis of the contributions of the aforementioned scholars, it was fundamentally important to distinguish between the 3 prominent views of man. Some scholars argue that man is a monistic being, while others defend a dichotomous view of man as consisting of a body and a soul. A third significant view describes man as consisting of three significant parts, namely a body, a soul and a spirit. In this thesis it was argued that the trichotomous view of man was the most correct way of defining the fundamental composition of man. According to the researcher an understanding of the trichotomous nature of man is important as it serves as a theological basis for pastoral counselling in order to clearly communicate the precise truths of the believer's union with Christ. In the last part of the thesis a practical pastoral paradigm was proposed based on the trichotomous view of man in which psychology and theology can be harmoniously integrated by means of a meta-systemic approach. The first phase of the paradigm focused on a model within which Psychotherapy can be applied while the second phase described Spirituotherapy as a model within which pastoral counselling can be applied.

Abstract:
AbstractAppreciating beauty is part of everyday life, when we contemplate fine arts, architecture, music, and natural scenes. Aesthetic appreciation, like any ordinary phenomenon of human life, triggers affective and cognitive processes that can provide the subject with sensations of hedonic pleasure and cognitive self-reward (Leder H, Belke B, Oeberst A, Augustin D. Br J Psychol 95(4):489–508, 2004). Although humans share several neuropsychological processes, the experience of aesthetic appreciation is undeniably idiosyncratic, and sometimes it is not that simple to find beauty where we were supposed to find it, and more often the same object can elicit different reactions amongst observers.

Abstract:
A large, 530 MW, Compressed Air Storage with Humidification (CASH) cycle was configured as an energy-storage plant utilizing as the low pressure expander the turbine of the Siemens-Westinghouse W 501F2 with a firing temperature of about 2400°F (1315C) and a 1600°F (870C) high pressure Dresser turbine. A selling price for electricity was developed for this CASH plant and compared with that for power produced by simple and combined cycle power plants of similar size. At 90°F ambient, where cycling power is most necessary, the specific capital cost of the CASH plant was calculated to be 405 $/kW while the simple and combined cycle were 403 and 489 $/kW respectively. The CASH plant has a much lower incremental cost than does the simple cycle. These factors result in the CASH plant having lower overall costs than the simple cycle for operation of over about 400 hr/yr. and lower than the combined cycle for operation of less than about 4000 hr/yr.

Abstract:
"Indiscriminability action spectra" of the red/green opponent channel can be measured by using a foveally presented 500 ms raised cosine without spatial edges. These spectra represent lights that are rendered indiscriminable at a cone-antagonistic locus. I found that mid-waveand long-wave low-frequency stimuli falling on the same contour, when presented simultaneously on an intense 578 nm adapting field, cancel the "green" and "red" sensations, respectively, elicited by each. That is, the isocancellation contours of the red/green channel are identical to the indiscriminability spectra. Also, using the low-frequency stimulus, I isolated a short-wave mechanism that codes a single "violet" both at and above threshold. Lights between 400 and 489 nm, which fall on the same indiscriminability contour up to 2 log units suprathreshold, are indiscriminable. This single "violet" is distinct from a single "blue" elicited by a second short-wave mechanism whose sensitivity is suppressed by a 578 nm adapting field. This mechanism is manifestly opponent since the color it codes is cancelled by an appropriate yellow low-frequency light.

Abstract:
Naturally occurring retirement communities (NORCs) are apartment, condo, co-op and social housing buildings that while not purpose-built for older adults, have become home to a high number of them. In Toronto, there are 489 residential buildings that are NORCs. Of these, 256 are located in neighbourhoods with the highest cumulative incidence of SARS-CoV-2, and are home to 40,955 older adults 65 years of age and above, including 18,144 older adults 80 years of age and above. Prioritizing COVID-19 vaccination by both age and neighbourhood of residence is an effective strategy to minimize deaths, morbidity, and hospitalization. Targeting people living in NORCs in high-risk neighbourhoods for early vaccination is a practical application of that strategy, which will also address barriers to vaccination in this population.
