Abstract Background Due to severe immunosuppression, patients undergoing allogeneic hematopoietic stem cell transplantation (aSCT) are at increased risk of infection and especially bloodstream infections (BSI) remain a major cause of death. Knowledge of the specific epidemiology of pathogens and resistances is of utmost importance to optimize antimicrobial treatment strategies. Methods Based on the Cologne Cohort of Neutropenic Patients (CoCoNut) database, we conducted a retrospective analysis of blood cultures collected within 100 days following transplantation of patients undergoing aSCT between January 2009 and December 2018 at the University Hospital of Cologne, Germany. Contamination of coagulase-negative Staphylococci (CoNS) isolates (single positive isolate within 5 days) was considered within the analysis. Results In total, 843 aSCT patients were available for analysis (484/843 [57%] male). The median age was 53 (interquartile range [IQR] 43–62) years, predominant underlying diseases were acute myeloid leukemia (47%, 397/843), lymphoma (14%, 117/843), and acute lymphoblastic leukemia (11%, 89/843). Median inpatient stay was 39 (IQR 34–50) days, while 67/843 (8%) patients died. Antibacterial prophylaxis was administered in 289/843 (34%) and antifungal prophylaxis in 738/843 (88%) patients. BSI was diagnosed in 233/843 (28%) patients. In total, 5,489 pairs of blood cultures were taken (median 4 per patient, IQR 2–8), while a pathogen could only be detected in 922/5,489 (17%). Most frequent pathogens were CoNS (259/922, 28%), Enterococcus spp. (219/922, 24%), E. coli (132/922, 14%), Klebsiella spp. (44/922, 5%), P. aeruginosa (39/922, 4%), S. aureus (37/922, 4%), and Candida spp. (42/922, 5%). Polymicrobial infection was detected in 58/922 (6%) cases. Within Enterococci isolates, 24/219 (11%) were VRE. None of the Klebsiella, but 9/132 (7%) of E. coli isolates were ESBL positive. In 4/37 (11%) cases S. aureus isolates were MRSA. Conclusion Patients in the early phase after aSCT are at high risk of BSI with a predominantly gram-positive spectrum. Empirical antimicrobial treatment must consider pathogen epidemiology and resistance patterns while waiting for blood culture results. Disclosures All authors: No reported disclosures.

Abstract Background Osteoporosis is one of the leading causes of morbidity and mortality in older women and men. Men with osteoporosis have higher fracture-related morbidity and mortality rates compared to women, but screening rates in men remain much lower. Moreover, there is limited veteran-specific data despite the suggestion that veterans may be at higher risk for osteoporosis. (1) Objective: To assess current dual energy X-ray absorptiometry (DEXA) ordering practices within a veteran cohort. Design: A retrospective cohort study of male veterans aged71-75 years who were evaluated by a primary care provider (PCP) within the last 5 years (n = 13,174). Results DEXAs wereordered for ∼7% of the population (920/13,174), and 91.6% were completed (843/920). Over 60% of completed DEXAs were ordered due to the presence of risk factors (308/843, 36.5%), history of fractures (60/843, 7%), known osteopenia or osteoporosis (59/843, 7%), or osteopenia mentioned on another imaging modality (78/843, 9.3%), while only 33.5% (282/843) were ordered for screening purposes. Of those with DEXA results reported (838/843, 99.4%), 34.5% (289/838) were normal, 51% (427/838) were in the osteopenia range and 14.5% (122/838) were in the osteoporosis range. Regarding DEXAs obtained for screening purposes, 42% (118/282) were normal, 48% (136/282) were in the osteopenia range, and 10% (28/282) were in the osteoporosis range. Conclusions Despite the Endocrine Society and National Osteoporosis Foundation recommending screening men aged ≥70 years, less than 10% of male veterans eligible for osteoporosis screening based on age alone had a DEXA scan. Of those male veterans who received a DEXA for screening purposes, almost 60% were found to have an abnormal result. The lack of screening for osteoporosis in this population has the potential to worsen the disparity in bone health between men and women, and between veterans and non-veterans, making it difficult to determine the scope of a problem which has the potential to cause significant morbidity and mortality. <p>Two cultivars of faba bean (<em>Vicia faba</em> ‘Giza 843’ and ‘Nobaria 3’) that differ in aluminum (Al) tolerance were used to study cytogenetic and genomic alterations under the influence of Al Cl<sub>3</sub> (5, 15, and 25 mmol AlCl<sub>3</sub>) for different periods (6, 12 and 24 h). Under Al treatments, mitotic index in both cultivars decreased and total chromosomal abnormalities increased. The frequencies of micronuclei and chromosomal abnormalities (C-anaphase, metaphase-star chromosomes, breaks, sticky and disturbed chromosomes during metaphase or anaphase) in ‘Giza 843’ were lower than in ‘Nabaria 3’. Increase of the registered cytogenetic events under the influence of Al stress led to increase the detected polymorphism using RAPD and ISSR markers. Application of RAPD primers gave the same value of polymorphism in both faba bean cultivars under Al stress. Polymorphism average of nine ISSR primers of ’Giza 843’ (65.36 %) was lower than that of ‘Nobaria 3’ (71.59 %). Molecular markers, cytogenetic characteristics and seedling growth data indicate that Al tolerance of ‘Giza 843’ was higher than of ‘Nobaria 3’. This work shows that cytogenetic and ISSR techniques could be used efficiently to distinguish between the ability of two faba bean cultivars to tolerate toxic effects of Al.</p>

Abstract Background: The Breast Cancer Index (BCI) is a validated gene expression assay that provides a quantitative individualized risk of late recurrence and predicts the likelihood of benefit from extended endocrine therapy (EET) in HR+ early-stage breast cancer. The objective of this analysis was to assess the impact of BCI on clinical decision-making regarding EET in the BCI Registry Study. Methods: The BCI Registry Study is an ongoing, prospective, large-scale study to investigate the long-term clinical outcome, decision impact, and medication adherence in HR+ early-stage breast cancer patients receiving BCI testing as part of routine clinical care. Per protocol, physicians and patients completed pre- and post-BCI test questionnaires to assess the following: physician decision-making regarding EET; physician confidence with decision-making; patient preferences for EET; patient concerns about the cost, side effects, drug safety and benefit of EET; and patient satisfaction regarding treatment recommendation. Pre- and post-BCI responses were compared using McNemar’s test and Wilcoxon signed rank test. Results: Pre- and post-BCI test physician and patient questionnaires were completed for 843 and 823 patients, respectively. For the patients included in this analysis, mean age at enrollment was 65y and 88.4% of patients were postmenopausal. 74.7% of tumors were T1, 53.4% were G2, 75.3% N0, and 13.8% HER2-positive. Following BCI testing, physicians changed treatment recommendations for EET in 40.1% (338/843) of patients (p&lt; 0.0001). In cases in which physicians recommended EET prior to BCI testing, 44.7% (214/479) changed their recommendation not to extend endocrine therapy. Of these cases, 98.1% (210/214) were BCI Prognostic low risk and/or BCI (H/I) Predictive low likelihood of benefit from EET. In cases in which physicians did not recommend EET prior to BCI testing, 34.6% (124/358) changed their recommendation in favor of EET. In such cases, 87.1% (108/124) of cases were BCI Prognostic high risk and/or BCI (H/I) Predictive high likelihood of benefit from EET. Further, following BCI testing, 38.8% (327/843) of physicians felt more confident in their recommendation (p&lt; 0.0001). The percentage of physicians having high confidence levels (confident or strongly confident) increased from 58.1% (490/843) before BCI testing to 80.5% (679/843) after BCI testing. The percentage of physicians having low confidence levels (not at all confident, not confident, or ambivalent) decreased from 39.1% (330/843) before BCI testing to 18.7% (158/843) after BCI testing. In addition, 40.5% (341/843) of patients felt more comfortable with their EET decision (p&lt; 0.0001) following BCI testing. Notably, changes in patient preferences for EET correlated with BCI test results. In BCI (H/I)-Low patients, 46.9% (241/514) showed a decreased preference for EET (p&lt; 0.0001) while in BCI (H/I)-High patients, 28.2% (87/309) showed an increased preference for EET (p&lt; 0.0001). Compared with baseline, after BCI testing, significantly more patients were less concerned about cost (20.9%, p&lt; 0.0001), drug safety (25.4%, p=0.0014), and the benefits of EET (29.3%, p=0.0002). No significant change in concern regarding side-effects was observed (p=0.1486). Conclusions: This first analysis in a large patient cohort of the BCI Registry confirms previous findings on the significant clinical decision-making impact of BCI for extending adjuvant endocrine therapy. Incorporating BCI into clinical practice resulted in changes in physician recommendations for EET in over 40% of cases, while at the same time increasing physician confidence in their recommendations. Knowledge of the BCI result also improved patient satisfaction and reduced patient concerns regarding cost, drug safety and benefit of EET. Citation Format: Tara B. ObjectivesAlthough family planning in China has changed gradually since 1994, there are few studies about family planning and women’s reproductive rights. The main objective of this study was to examine awareness of sexual and reproductive healthcare rights (SRHCRs), and learn how factors related to family planning influence awareness of SRHCRs among married women of reproductive age in China.Methods and participantsInner Mongolia, Chongqing, Guangdong and Henan were selected for the study, and a total of 2504 married women of reproductive age were recruited. A self-administered anonymous questionnaire was used to collect information on participants’ awareness of SRHCRs.ResultsThere were a total of 10 843 (≤6×2504) responses, with a response rate of 72.17% (10 843/15 024) on average among participants regarding SRHCRs (a multiple response set). The highest response rate was for choice (Right 3) (90.64%, 95% CI 89.47%, 91.81%), followed by privacy (Right 5) (86.11%, 95% CI 84.72%, 87.50%) and information (Right 1) (84.47%, 95% CI 83.02%, 85.93%). Only 43.39% (95% CI 41.40%, 45.38%) of participants gave responses to safety (Right 4). Participants without children showed more interest in Right 1, in access (Right 2) and in Right 4. Those who utilised tests for fetal sex determination paid more attention to Rights 2 and 4. Women who accepted informed choice were more likely to be aware of all six rights except for Right 3 and dignity (Right 6). Those individuals who were satisfied or very satisfied with comprehensive sexual and reproductive health counselling services were more likely to show interest in all six rights.ConclusionsAwareness of SRHCRs among reproductive aged women in China is still inadequate. Family planning service providers might strengthen the service awareness of sex and reproductive health rights according to the different needs of women.

Background: Social media is like a magnanimous tree and us being leaves connected through twigs of imagination Today 4.33 billion people are connected to the internet, and almost all do by using their mobiles. In Pakistan, there were 71 million broadband subscribers and 69 million 3G/4G users in June 2019. Aims & Objectives: The current study aimed to find the relationship between social support, social media usage, and psychological well-being among undergraduates aged 18-25 years. Methods: A sample of eight hundred and forty-three male and female students was included in this research. A stratified random sampling technique was applied in the research. The sample was taken from departments of various institutions in Lahore. The duration of the study was 6 months. The study was a cross-sectional study and the data were collected using questionnaires based on Ryff's scale of psychological well-being, Media and Technology Usage and Attitudes Scale, and Inventory of Social Supportive Behavior (ISSB) scale. Results: The data were analyzed on SPSS 25 and the results indicated that social support received from close ties was positively correlated with psychological well-being [r(841) = .523, n = 843, p< .001]. At the same time, social support received from online sources had no significant impact on psychological well-being while social media usage was negatively correlated with psychological well-being [r(841) = -.215, n = 843, p< .001]. Using the regression analysis technique, the three variables in combination explained 34.6% of the variance in psychological well-being [R2 = 0.346, F(6, 836) = 75.37, p< 0.01]. Age and gender had no significant effect. Conclusion: These findings implicate that healthy interpersonal relationships between close ties and undergraduate students should be encouraged and controlled usage of social networking sites should be practiced.

Abstract. The mesoscale snow distribution over the Namco lake area of the Tibetan Plateau on October 2005 has been investigated in this paper. The control and sensitive experiments were conducted using Weather Research Model (WRF) with three nested grids that included a 1 km finest grid centered on the Namco station. Our simulation ran from 6 October through 10 October 2005, which was concurrent with long term meteorological observations. Evaluation against boundary layer meteorological tower measurements and flux observations showed that the model captured the observed 2 m temperature and 10 m winds reasonably well in the sensitive experiment. The results suggested that output snow depth maximum amounts from two simulated experiments were centered downwind shore of Namco lake. Modified surface parameters for example surface skin temperature on the lake help to increase simulated credibility.

Abstract A precise colorimetric method is described for the estimation of phenyltoloxamine dihydrogen citrate, diphenhydramine hydrochloride, and chlorpheniramine maleate. The method is based on the interaction of these drugs as n-electron donors with 7,7,8,8-tetracyanoquinodimethane as π-acceptor to give a highly colored radical anion. This is corroborated by electron spin resonance spectroscopy. The colored product is quantitated spectrophotometrically at 843 nm. Beer's law is obeyed in the concentration range 2-6 µg/mL for each drug. Optimization of the experimental conditions is described. The proposed method was applied to the analysis of these drugs in tablet form, and results were in good agreement with those obtained by the official method

180 Background: The Institute of Medicine (IOM) 2013 report recommends supportive oncology care from diagnosis through survivorship, to end of life. The Coleman Supportive Oncology Collaborative (CSOC) developed a city-wide plan to improve supportive oncology. Metrics derived from the Commission on Cancer (CoC), ASCO Quality Oncology Practice Initiative (ASCO-QOPI) and National Quality Forum (NQF) were used to assess the CSOC impact. Methods: Medical records of consecutive cancer patients from 6 practice improvement cancer centers in Chicago (3 academic, 2 safety-net, 1 public) were reviewed for 2 periods: 2014 (n = 843) and Q1 of 2015 (n = 313). Descriptive statistics assessed differences in quality metrics. Results: Significant improvement was achieved in 6 of 8 core supportive oncology metrics (see table). Conclusions: Consolidated metrics are feasible to assess supportive oncology quality. Early data indicate improvement and effectiveness of the collaborative approach. [Table: see text]

96 Background: The IOM 2013 report recommends comprehensive supportive oncology care from diagnosis through survivorship to end of life. The Coleman Foundation supported a baseline assessment of supportive oncology care using key COC, ASCO-QOPI and NQF measures. Methods: Data extracted from medical records at 6 Chicago cancer centers in 2014 (n = 843 patients), including stage I-III (n = 391) and stage IV solid tumor or refractory hematologic malignancy (n = 452). Results: Fifty-four percent of patients had discussions on understanding of illness, 24% were informed of prognosis timeframe, and 5% had a documented healthcare power of attorney. Only 6% of stage IV patients received distress screening within 2 weeks of diagnosis and 15% of Stage IV patients had referrals to palliative care. Among patients with Stage I-III cancer, 38% had a supportive oncology screening, and 2% received treatment summaries and a SCPs. Conclusions: There are still large gaps in important supportive care processes based on IOM 2013 recommendations, COC, and ASCO QOPI measures. Future steps include implementing supportive care processes that address gaps across the 6 cancer centers. [Table: see text]

Operational national-scale hydrological forecasting systems are widely used in many countries for flood early warning systems and water management. However, this kind of system has never been implemented in Russia. OpenForecast v2—the first national-scale operational runoff forecasting system in Russia—has been developed and deployed to fill this gap. OpenForecast v2 delivers 7 day-ahead streamflow forecasts for 843 gauges across Russia. The verification study has been carried out using 244 gauges for which operational streamflow data were openly available and quality-controlled for the entire verification period (14 March–6 July 2020). The results showed that the developed system provides reliable and skillful runoff forecasts for up to one week. The benchmark testing against climatology and persistence forecasts showed that the system provides skillful predictions for most analyzed basins. OpenForecast v2 is in operational use and is openly available on the Internet.

The manuscript containing the service of St. Lazar Hrebeljanovic was presented to the Russian Tsar in 1550 by the abbot of the Hilandar monastery Paisius. We currently know only two manuscripts containing hymns to the Saint Lazar: RNB, Kir.-Bel. 586/843 (eighth decade of the 16th century) and RBB, f. 379 ? 66 (mid- 17th century). The older source is more complete. It includes stichera of Small and Great Vespers (on Lord I have cried and aposticha), stichera at the Litya, stichera at the end of Psalm 50, aposticha of Matins, as well as exapostilarion and theotokion. In addition, there are indications that two canons have been performed (a total of 31 hymns). In the younger source the chanting repertoire is much smaller - there are only doxastika and , stichera at the end of Psalm 50 (a total of 6 songs). The doxastika presented in these manuscripts have different musical variants.

The objective of this study was to optimize the epoxidation of fatty acid methyl esters (FAME) derived from Jatropha oil. This experiment was carried out with peracetic acid generated in situ by using hydrogen peroxide and acetic acid. A response surface methodology (RSM) central composite design (CCD) was used, with one response variable, the percentage (%) of epoxidation yield. The factors evaluated were temperature (50-80°C), the molar ratio of hydrogen peroxide (HP) to unsaturation (1.1-2 mol), the molar ratio of acetic acid (AA) to unsaturation (0.5-0.8 mol), and time (2-7 hours). The optimum percentage of epoxidation yield (90.98%) was at the condition of 65°C reaction temperature, HP to unsaturation molar ratio of 2.19, AA to unsaturation molar ratio of 0.65 for 6 hours. The formation of epoxides product (oxirane) was confirmed using Fourier transform infrared spectroscopy oxirane peaks (doublet) at 825 and 843 cm-1. The result showed good agreement with the predicted values from the RSM model.

The primary structure of a novel envelope protein from shrimp white spot syndrome virus (WSSV) was characterized using a combination of SDS–PAGE and mass spectrometry. The resulting amino acid sequence matched an open reading frame (ORF), ORF1050, of the WSSV genome ORF database. ORF1050 contained 843 nt, encoding 281 aa, and was termed the vp281 gene. Computer-assisted analysis showed that both the vp281 gene and its product shared no significant homology with other known viruses. However, they shared striking identity/similarity with another WSSV structural protein, VP292, at both the nucleotide and amino acid sequence level, suggesting that vp281 and vp292 might have evolved by gene duplication from a common ancestral gene. WSSV VP281 cDNA was cloned into a pET32a(+) expression vector containing a T7 RNA polymerase promoter to produce (His)6-tagged fusion proteins in Escherichia coli strain BL21. Specific mouse antibodies were raised using the purified fusion protein (His)6-VP281. Western blot analysis showed that the mouse anti-(His)6-VP281 antibodies bound specifically to VP281 of WSSV, without cross-reactivity with VP292. The transmission electron microscope immunogold-labelling method was used to localize VP281 in the WSSV virion as an envelope protein. The cell attachment ‘Arg–Gly–Asp’ motif in VP281 indicated that this protein might play an important role in mediating WSSV infectivity.

Salacia is one of the medicinally valuable genus, distributed throughout tropical areas which include India, Sri Lanka, Southern China and other Southern Asian Countries. The genus Salacia is represented by 21 species in India, among them eight species are recorded from the state of Karnataka in the Southern part of India. Despite its pharmaceutical importance, very little information exists about the genetic diversity of Salacia at molecular level. Hence the present study was carried out to evaluate the genetic among six species of Salacia namely S. chinensis, S. malabarica, S. oblonga, S. macrosperma, S. reticulata and S. gambleana with the help of ISSR marker analysis. Dendrogram and genetic distance were generated adopting Unweighted Paired Group Method with Arithmetic mean (UPGMA) in the NTSYS-pc software. Basic genetic parameters were calculated by analysing the genetic data with Pop gene 1.32 and GenAlEx 6.2 software. The overall polymorphism across the ten primers screened revealed 26 % polymorphism. A 60% polymorphism was scored for the primer UBC 841, whereas, no polymorphism was observed for primer UBC 840 and ISSR 6. The average observed heterozygosity was more than expected heterozygosity. Observed heterozygosity (Ho) ranged from 0.15 (UBC 841) to 0.38 (ISSR 6) with an average of 0.25, whereas expected heterozygosity (He) ranged from 0.10 (UBC 843) to 0.35 (ISSR 6) with an average of 0.23 for Salacia species. The higher heterozygosity pointed towards increased genetic diversity amongst the species. ISSR marker analysis showed high level of inter and intra population genetic differentiation.

Abstract Abstract 1706 TET2 (tet oncogene family member 2) on chromosome 4q24 was identified as a candidate tumor suppressor gene. Recurring submicroscopic deletions and copy-neutral loss of heterozygosity (CN-LOH) involving 4q in MDS patients detected by SNP microarray analyses suggested TET2 as an interesting candidate gene. Subsequent sequencing studies revealed TET2 mutations in 10–25% of patients with AML, MDS, and MPNs, while a mutation frequency of up to 42% was reported in CMML. Only a subset of studies evaluated both TET2 mutation status and TET2 copy number status, although this might be of pathophysiological and even of prognostic relevance if TET2 functions as a classical tumor suppressor gene. In the majority of studies copy number status was determined by SNP array analysis, although being expensive and time consuming. Here, in order to investigate TET2 deletions in a large cohort of patients an easy to perform FISH assay was developed applying BACs covering the TET2 gene (RP11-351K6 and RP11-16G16; BlueGnome, Cambridge, UK). This assay was validated on samples with TET2 deletions proven by SNP array analysis. With these FISH probes we analyzed 843 cases with myeloid malignancies (404 AML (323 de novo AML, 68 s-AML, 13 t-AML), 166 MDS, 201 CMML, and 72 MPN). Overall 50 (5.9%) cases with TET2 deletion were identified. These included 22 AML (5.0%), in detail 14 de novo AML (4.3%), 6 s-AML (8.8%), and 2 t-AML (15.4%) as well as 15 CMML (7.5%), 9 MDS (5.4%) and 4 MPN (5.6%). Patients with TET2 deletions showed the following karyotypes: normal: n=15, cytogenetically balanced rearrangements involving 4q24: n=3, 4q deletion as the sole abnormality: n=2, complex: n=25, other abnormalities: n=5. In 25/50 (50%) cases the TET2 deletion was cytogenetically cryptic. In patients with complex aberrant karyotype loss of 4q material was due to interstitial deletion in 7/25 cases and due to unbalanced rearrangements in 16/25 cases, while in 2/25 cases chromosomes 4 were normal in chromosome banding analysis. Furthermore, in patients with TET2 deletions mutation analyses was performed for mutations in TET2 (n=37 investigated), RUNX1 (n=13), NPM1 (n=18), JAK2V617F (n=18), CBL (n=36), NRAS (n=17), KRAS (n=36), FLT3-ITD (n=26), FLT3-TKD (n=8), IDH1 (n=9) and MLL-PTD (n=24). Mutations in TET2 were detected in 19/37 cases (51%), in RUNX1 in 1/13 (8%), in JAK2V617F in 6/18 (33.3%), in CBL in 2/36 (5.6%), in NRAS in 1/17 (6%), in KRAS in 1/36 (2.8%) and in NPM1 in 1/18 (6%) cases, whereas no mutations within the other genes analyzed were found. In the cohort of cases with TET2 deletion and concomitantly TET2 mutation (n=19) 10 had a normal karyotype (52.6%), 5 a complex karyotype (26.3%) and 4 had other abnormalities (21.1%). Importantly, in the cohort of CMML, in 10 of 14 cases (71.4%) both a TET2 deletion and TET2 mutation was detected. Overall, TET2 mutations were significantly more frequent in patients with cytogenetically cryptic TET2 deletion as compared to cytogenetically visible 4q deletions (68.2% vs. 26.7%, p=0.020). In addition FISH screening identified 2 cases (one CMML, one t-MDS) which showed a translocation involving the TET2 locus not leading to a deletion of the BAC signals but a separation suggesting a fusion with yet unidentified partner genes. In conclusion, FISH analyses identified TET2 deletions in 6% of myeloid malignancies. 50% of these deletions were submicroscopic and therefore not detectable by chromosome banding analysis. TET2 deletions were accompanied by TET2 mutations in 51% of respective cases. FISH is a reliable and efficient method to determine the copy number state of TET2. Still, the prognostic impact of TET2 deletions with and without additional TET2 mutations in the various myeloid malignancies has to be evaluated in future investigations. Tres especies resultaron ser las más abundantes y más frecuentes, Aleochara (Aleochara) curtula (Goeze, 1777), Aleochara (Aleochara) lata Gravenhorst, 1802 y Aleochara (Xenochara) haematoptera. To improve the quality and digestibility of legumes, and to examine effects of varying germination periods (6, 12 and 18 hrs) on the protein quantity, digestibility, ash, moisture, fibre and fat content of locally available legumes namely green gram, chickpea, lentil and moth bean the present study was undertaken. It was found that the protein content and protein digestibility of legumes increased by 4.87 to 42.79 and 3.48 to 19.77 per cent, respectively depending on the type of legume, and length of germination. The highest per cent increase in protein content was found in chickpea (42.79%) germinated for 18 hrs. Maximum increase in in-vitro protein digestibility (19.77 %) was noticed in moth bean, ranging from 5.31 to 19.77 per cent at 6, 12 and 18 hrs of germination. Highest enhancement in protein value was observed in legumes which were germinated for 18 hrs. In comparison to raw, a significant reduction was noted in the ash content of all types of germinated legumes and in terms of per cent, the decrease varied from 2.98 to 8.46. Moisture and crude fibre content increased significantly where fat content was decreased in all legumes. Cervi cornu extracts have been used in traditional medicine for the treatment of various disorders, including osteoporosis. However, since it is not easy to separate the active ingredients, limited research has been conducted on their functional properties. In this study, we extracted the low-molecular-weight (843 Da) collagen NP-2007 from cervi cornu by enzyme hydrolyzation to enhance absorption and evaluated the therapeutic effect in monosodium iodoacetate-induced rat osteoarthritis (OA) model. NP-2007 was orally administered at 50, 100, and 200 mg/kg for 21 days. We showed that the production of matrix metalloproteinase-2, -3, and -9, decreased after NP-2007 treatment. The levels of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, and prostaglandin E2 were also reduced after treatment of NP-2007. Furthermore, the administration of NP-2007 resulted in effective preservation of both the synovial membrane and knee cartilage and significantly decreased the transformation of fibrous tissue. We verified that the treatment of NP-2007 significantly reduced the production of nitric oxide and pro-inflammatory cytokines including TNF-α, IL-1β, and IL-6 in lipopolysaccharides-stimulated RAW 264.7 cells by regulation of the NF-kB and MAPK signaling pathways. This study indicates that NP-2007 can alleviate symptoms of osteoarthritis and can be applied as a novel treatment for OA treatment.

Цель исследования - улучшение исходов лечения пациентов старших возрастных групп с переломами проксимального отдела бедренной кости (ППОБК) путем разработки алгоритма оказания медицинской помощи с учетом характера перелома и коморбидного фона пациента. В статье изложены подходы к выбору тактики и техники помощи пострадавшим старших возрастных групп с ППОБК. В исследование вошли 4 491 пациент с ППОБК, проходивший лечение в Госпитале для ветеранов войн с 2010 по 2019 г. Все больные были разделены на две группы в зависимости от тактики применяемого лечения: 1-я - 2 082 больных, пролеченных в 2010-2014 гг. с применением традиционных подходов; 2-я - 2 409 человек, лечение которых осуществляли по разработанному алгоритму, учитывающему характер перелома и тяжесть коморбидной патологии. Результаты были оценены по шкале Лекена, а качество жизни - по шкале ВОЗ. В 1-й группе хорошие исходы были достигнуты через 1 год у 126 (10,5 %) пациентов, через 3 года - у 111 (9,1 %); во 2-й группе через 1 год были достигнуты отличные исходы у 103 (6 %), хорошие - у 960 (55,7 %), а через 3 года отличные исходы - у 87 (5,4 %), хорошие - у 843 (52,3 %). Таким образом, использование предложенного алгоритма оказания помощи пациентам с ППОБК позволяет достоверно (p<0,001) улучшить исходы лечения этой категории пострадавших. The objective of the study was to improve the treatment outcome of patients of older age groups with fractures of the proximal femur by developing an algorithm for providing medical care while taking into account the nature of a fracture and the comorbid background of a patient. The article describes approaches to the choice of tactics and techniques of medical assistance to patients of older age groups with fractures of the proximal femur. 4 491 patients with fractures of the proximal femur who had been treated at the Hospital for War Veterans in Saint Petersburg from 2010 to 2019 were included for analysis. All the patients were divided into two groups depending on the tactics of the treatment used. Group 1 (2 082 patients) were treated in 2010-2014 using traditional methods. Group 2 (2 409 patients) were treated according to the developed algorithm while taking into account the nature of a fracture and the severity of comorbid pathology. The results were evaluated by the Leken’s functional index, and the quality of life was assessed by WHO scale. In patients of the 1st group, good outcomes were achieved after a year in 126 (10,5 %), after 3 years in 111 (9,1 %), while in the second group after a year, excellent outcomes were achieved in 103 (6 %) cases, good in 960 (55,7 %), and after 3 years excellent in 87 (5,4 %), good in 843 (52,3 %). Thus, the use of the proposed algorithm for providing care to patients with fractures of the proximal femur significantly (p<0,001) improves the treatment outcomes for this category of patients.

Introduction. The authors arise the question of increasing the efficiency of the municipal machine. Moreover, the influence of the clamping force of the brush body on the surface and on the brush deterioration and, as a consequence, on the quality of the roadway cleaning is highlighted in the article.Methods and materials. The results of the changing process of the brushing tool vertical coordinate in the municipal machine were presented, depending on the displacement of the hydraulic cylinder rod. The analysis of the existing types of the brushing working body suspension was carried out. As a result of such analysis, the authors established the most frequent usage of the three-point and parallelogram suspensions, the schemes of which were also presented in the article.Results. The equations of geometrical connections of the municipal machine elements, which could be used in mathematical model, are presented. In addition, the kinematic diagram of the municipal machine with the brush working body suspension is drawn up. Therefore, the presented model takes into account such parameters as the length of the levers’ arms, the linear movements of the hinges, the rod stroke, the levers’ angles and the distances between the hinges. The methods and the course of the experiment for determining the displacement dependence of the brush body from the displacement of the hydraulic cylinder rod are described. The authors also present the photographs of the experimental research fragments.Discussion and conclusions. On the basis of the obtained data, the authors construct the experimental and theoretical dependences’ graph of the brush body displacement and the actuator hydraulic cylinder rod displacement. The analysis of such graphs confirms the adequacy of the geometric constraint equations and also proves that these equations could be used in the mathematical model of the position controlling process of the brush operating body and also for determination the optimum value of the brush working element clamping force to the surface.

CD22, a B lymphocyte membrane glycoprotein, contains immunoreceptor tyrosine-based inhibition motifs (ITIMs) in the cytoplasmic region and recruits Src homology 2-containing protein-tyrosine phosphatase-1 (SHP-1) to the phosphorylated ITIMs upon ligation of B lymphocyte antigen receptor (BCR), thereby negatively regulating BCR signaling. Among the three previously identified ITIMs, both ITIMs containing tyrosine residues at position 843 (Tyr843) and 863 (Tyr863), respectively, are shown to be required for CD22 to recruit SHP-1 and regulate BCR signaling upon BCR ligation by anti-Ig antibody (Ab), indicating that CD22 has the SHP-1-binding domain at the region containing Tyr843 and Tyr863. Here we address the requirement of CD22 for SHP-1 recruitment and BCR regulation upon BCR ligation by antigen, which induces much stronger CD22 phosphorylation than anti-Ig Ab does. We demonstrate that the CD22 mutant in which both Tyr843 and Tyr863 are replaced by phenylalanine (CD22F5/6) recruits SHP-1 and regulates BCR signaling upon stimulation with antigen but not anti-Ig Ab. This result strongly suggests that CD22 contains another SHP-1 binding domain that is specifically activated upon stimulation with antigen. Both of the flanking sequences of Tyr783 and Tyr817 fit the consensus sequence of ITIM, and the CD22F5/6 mutant requires these tyrosine residues for SHP-1 binding and BCR regulation. Thus, these ITIMs constitute a novel conditional SHP-1-binding site of CD22 that is activated upon BCR ligation by antigen but not by anti-Ig Ab.

Abstract Our research group has focused on how Ca salts of soybean oil (CSSO) supplementation, a source of omega-6 fatty acids (FA), impacts reproductive performance of beef cows. In Nelore (Bos indicus) grazing cows assigned to fixed-time artificial insemination (AI), supplementing 100 g/d (as-fed basis) of CSSO for 21 d after AI increased (P &lt; 0.01) pregnancy rates from 38.1% (623/1635 as pregnant/total non-supplemented cows) to 49.0% (843/1720 as pregnant/total CSSO-supplemented cows). These outcomes were later associated with enhanced early embryonic development and pregnancy establishment from supplemental omega-6 FA. More specifically, supplementing 100 g/d of CSSO to Nelore cows increased (P ≤ 0.05) incorporation of omega-6 FA in circulation and in the conceptus, increased (P ≤ 0.04) conceptus length (2.2-fold increase), and upregulated mRNA expression of interferon-tau (4.1-fold increase) on d 15 of gestation. Similar outcomes were noted in B. taurus beef cows receiving 100 g/d of CSSO for 21 d after AI. Supplementing CSSO to Angus-influenced cows increased (P = 0.01) pregnancy rates from 51.7% (193/388 as pregnant/total non-supplemented cows) to 60.2% (226/383 as pregnant/total CSSO-supplemented cows), increased (P &lt; 0.01) plasma concentrations of omega-6 FA, and upregulated (P = 0.05) mRNA expression of interferon-tau in d 15 conceptuses (1.8-fold increase). Across these research efforts, CSSO was mixed with granular feed ingredients and hand-fed to cows, which demands labor and may discourage the use of CSSO supplementation by commercial cow-calf producers. Accordingly, our research group also demonstrated that CSSO inclusion into a self-fed low-moisture block resulted in similar incorporation (P ≥ 0.35) of omega-6 FA in the circulation of Angus-influenced cows compared with CSSO inclusion into a hand-fed granular concentrate. Collectively, CSSO improves reproductive performance of B. indicus and B. taurus beef cows, and may be supplemented via self-fed low-moisture blocks to minimize labor needs.

We assessed the impact of pre-exposure prophylaxis (PrEP) in the context of a community-based HIV program among female sex workers (FSWs) in Kolkata, India. This was an open-label, uncontrolled demonstration trial. HIV seronegative FSWs over 18 years were eligible. Participants were administered daily tenofovir/emtricitabine (TDF-FTC) with follow-up visits at months 1, 3, 6, 9, 12, and 15. Drug adherence was monitored by self-report, and a random subset of participants underwent plasma TDF testing. 843 women were screened and 678 enrolled and started on PrEP. Seventy-nine women (11%) did not complete all scheduled visits: four women died of reasons unrelated to PrEP and 75 withdrew, for a 15-month retention rate of 89%. Self-reported daily adherence was over 70%. Among those tested for TDF, the percentage of women whose level reached ≥40 ng/mL was 65% by their final visit. There were no HIV seroconversions, and no evidence of significant changes in sexual behavior. This study demonstrated the feasibility and effectiveness of PrEP for FSWs in Kolkata, with very high levels of adherence to PrEP and no HIV seroconversions. The integration of PrEP into an existing community-based HIV prevention program ensured community support and facilitated adherence.

AbstractThe development of the radio remnant of SN 1987A has been followed using the Australia Telescope Compact Array since its first detection in 1990 August. The remnant has been observed at four frequencies, 1.4, 2.4, 4.8, and 8.6 GHz, at intervals of 4–6 weeks since the first detection. These data are combined with the 843 MHz data set of Ball et al. (2001) obtained at Molonglo Observatory to study the spectral and temporal variations of the emission. These observations show that the remnant continues to increase in brightness, with a larger rate of increase at recent times. They also show that the radio spectrum is becoming flatter, with the spectral index changing from −0.97 to −0.88 over the 11 years. In addition, at roughly yearly intervals since 1992, the remnant has been imaged at 9 GHz using super-resolution techniques to obtain an effective synthesised beamwidth of about 0″.5. The imaging observations confirm the shell morphology of the radio remnant and show that it continues to expand at ˜3000 km s−1. The bright regions of radio emission seen on the limb of the shell do not appear to be related to the optical hot spots which have subsequently appeared in surrounding circumstellar material.

3-Aminobutanoic acid is an unnatural amino acid and by analogy with other amino acids, is represented in its zwitterionic form by R-CH(COO¯)NH­3+ (where side group R = aryl, alkyl or any other group) in solid phase. Experimental IR and Raman modes near 3029, 2871, 1577, 1288 cm-1 measured for 3ABA are strongly in favor of the zwitterionic dimer structure formed of –N-H∙∙∙O–C bonding between –NH­3+ and anion – CO2¯ groups. Zwitterionic monomer and dimer structures at B3LYP/6-311++G(d,p) level are computed. It has been found that the stable zwitterionic monomer and dimer structures are possible only in a water solvent medium computed using SCRF SMD solvation model. The zwitterionic dimer vibrational structure is in good agreement with the experiment and both zwitterionic monomer and dimer models have identically predicted the frequencies of the modes that are localized and unperturbed except those that characterize the H-bonding due to the –N-H∙∙∙O bonding. This is supported by the appearance of two characteristic medium strong Raman modes at 900 and 843 cm-1 that suggest unperturbed C-C skeletal structure and localized modes due to CH, CH2 and CH3 groups bound to it.

Abstract An attempt has been made to determine the organization of variable-heavy chain gene locus in cattle, based on 91% assembled Hereford cow genome sequence, where both functional VH and pseudogenes are identified on chromosome 7 and 21. Of a total of 13 VH genes identified unambiguously, 6 VH genes are located on chromosome 21 and 7 VH genes are present on chromosome 7. Interestingly, the functional VH genes are distributed on both chromosome 7 and 21 raising questions about VDJ recombination occurring on two different chromosomes (Hosseini, A. et al. 2004. Int. Immunol.16:843) or via as yet unidentified mechanism that possibly permitted allelic exclusion. The presence of 10 DH-genes in the Hereford genome is confirmed as earlier reported by our laboratory (Koti, M. et al. 2010. Mol Immunol. 47:2119). Come cryptic recombination signal sequences (RSS) seem to exist within the VH encoding region requiring elucidation of their role in VDJ recombination. Nevertheless, V-gene expression in bovine B cells is influenced by the composition of RSSs. These findings, especially the presence of variable heavy chain locus on two chromosomes, raise fundamental questions about the phenomenon and mechanisms of allelic exclusion in bovine B cells.
