Abstract:

PURPOSE: To determine the maximum-tolerated dose, pharmacokinetic interaction, and activity of PSC 833 compared with daunorubicin (DNR) and cytarabine in patients with poor-risk acute myeloid leukemia. PATIENTS AND METHODS: Patients received ara-C 3 g/m2/d on 5 consecutive days, followed by an IV loading dose of PSC 833 (1.5 mg/kg) and an 84-hour continuous infusion escalating from 6, 9, or 10 mg/kg/d. Daunorubicin was administered as a 72-hour continuous infusion at 34 or 45 mg/kg/d. Responding patients received consolidation chemotherapy with DNR pharmacokinetics performed without PSC-833 on day 1, and with PSC-833 on day 4. Response was correlated with expression of P-glycoprotein and lung resistance protein (LRP), and in vitro sensitization of leukemia progenitors to DNR cytotoxicity by PSC 833. RESULTS: All 43 patients are assessable for toxicity and response. Grade 3 or greater hyperbilirubinemia (70%) was the only dose-dependent toxicity. Four patients (9%) succumbed to treatment-related complications. Twenty-one patients (49%) achieved a complete remission or restored chronic phase, including 10 of 20 patients treated at the maximum-tolerated dose of 10 mg/kg/d of PSC-833 and 45 mg/m2 of DNR. The 95% confidence interval for complete response was 33.9% to 63.7%. Administration of PSC 833 did not alter the mean area under the curve for DNR, although clearance decreased approximately two-fold (P = .04). Daunorubicinol clearance decreased 3.3-fold (P = .016). Remission rates were not effected by mdr-1 expression, but LRP overexpression was associated with chemotherapy resistance. CONCLUSION: Combined treatment with infused PSC 833 and DNR is well tolerated and has activity in patients with poor risk acute myeloid leukemia. Administration of PSC 833 delays elimination of daunorubicinol, but yields variable changes in DNR systemic exposure.

Abstract:

Purpose. This study was aimed at developing co-encapsulated stealth nanoliposomes containing PSC 833, an efficient MDR modulator, and doxorubicin (DOX) in order to increase the effectiveness and decrease adverse effects of the anticancer drug. Methods. In attempt to increase the encapsulation efficiency of drugs, different methods for liposome preparation were tested and the effect of different parameters such as drug to lipid molar ratio, cholesterol mole percent and lipid compositions, were investigated. The final product with a lipid composition of EPC:DSPE-PEG2000:Chol (60:5:30 %mol) was prepared by thin layer film hydration method. After preparation of empty liposomes, DOX and PSC 833 were loaded using ammonium sulfate gradient and remote film loading methods, respectively. Physical characteristics of optimized liposomes (DOX/PSC-L) such as particle size, zeta potential, encapsulation efficiency, in-vitro drugs release and stability were evaluated. Furthermore, in vitro cytotoxicity study of various liposomal formulations as well as drugs, solutions against resistant human breast cancer cell line, T47D/TAMR-6, was evaluated using MTT assay. Results. The best formulation showed a narrow size distribution with average diameter of 91.3 ± 0.2 nm with zeta potential of -6 ± 1.2, the encapsulation efficiency for DOX and PSC 833 were more than 95% and 65.5%, respectively. In DOX-resistant T47D/TAMR-6 cells, dual-agent stealth liposomes showed significantly greater cytotoxicity (P < 0.05) than free DOX and liposomal DOX plus free PSC 833 treatments. Conclusions. Co-encapsulation of DOX and PSC 833 presents a promising anticancer formulation, capable of effective reversal of drug resistance, and should be explored further in therapeutic studies with animal tumor xenograft models. Abstract:

Abstract Older patients (pts) have a high incidence of P-glycoprotein (P-gp) positive AML blast cells. P-gp acts as drug efflux pump for a variety of agents including daunorubicin (DNR). The P-gp inhibitor PSC-833 can inhibit the function of P-gp in AML cells and restore sensitivity to DNR. Combining PSC-833 with DNR may improve response rate and survival in AML pts. However, previous intervention studies have produced conflicting results. From 1997 to 1999 we conducted a multi-center phase 3 study in pts of older age with previously untreated AML, in which we prospectively investigated the effect of PSC-833 combined with standard antileukemic treatment in relation to the flowcytometric P-gp status at diagnosis. Pts were randomized to receive two cycles of DNR (45 mg/m2, days 1-3) and Ara-C (200 mg/m2, 7 days) or the same regimen with a lower dose of DNR (35 mg/m2, days 1–3) plus PSC-833 (2 mg/kg loading dose followed by 10 mg/kg/24h for 72 hours). Pts who attained a complete remission (CR) were to receive a common consolidation cycle of Ara-C (1 g/m2, days 1–4), mitoxantrone (6 mg/m2, days 1–4) and etoposide (80 mg/m2, days 1–4). Results. 428 pts were randomized (214 in each arm) with 419 eligible and evaluable. Median age was 67 years (range 58–85). Baseline characteristics were comparable between the two arms. Cytogenetics was successfully performed in 293 pts; 5 had favorable and 66 had unfavorable abnormalities. P-gp expression was assessed in fresh, purified bone marrow samples of 309/419 pts (74%) by determining the Rhodamin123 efflux ratio with/without PSC-833 in vitro and by assessment of the UIC2 and MRK16 expression ratios. Upon P-gp assessment combining the 3 tests, pts were classified as P-gp negative (27%), low positive (29%), positive (28%) or highly positive (17%). Median follow up is 56 months. CR and 5-year survival rates with 95% confidence intervals are in the table below. Increased P-gp expression was associated with a lower CR rate (P=0.02), worse event-free survival (EFS) and overall survival (OS), P=0.01. There was also no significant effect of PSC-833 on the CR rate, OS, EFS or disease-free survival (DFS) when P-gp status was included in a multivariate analysis. In the planned subgroup analysis of pts with P-gp expression, who were expected to benefit most from PSC-833, CR rate was somewhat higher in the PSC arm (61% vs. 51%, P=0.07), however, OS, EFS and DFS were not different (P=0.8, 0.9 and 0.15). Analysis of frequently occurring polymorphisms of P-gp in exons 12, 21 and 26 revealed that pts who had wildtype genotype alleles in all 3 exons had a relatively poor survival as compared with those with heterozygous or homozygous mutant alleles in all 3 exons. Conclusions. Addition of PSC-833 to DNR and Ara-C does not improve CR rate nor long-term OS, EFS and DFS in elderly AML pts. P-gp expression remains an independent adverse prognostic factor in AML pts treated with P-gp reversal. Abstract:

Twelve corn (Zea mays L.) cultivars were evaluated under two environments in Central America and one in Mexico in order to determine the genetic progress achieved for resistance to stunting, after four selection cycles of the population 73. The trial included the synthetics from the cycles 0, 1,2,3 and 4, composites from the cycles 2, 3 and 4, three tolerant control s N8-6, Santa Rosa 8073 and the hybrid H-53, and a susceptible control (hybrid 8-833). The percent averages of stunted plants were relatively low (between 10 and 20 %) at the three localities. The cycle 4 synthetic yielded an average (4214 kg/ha) statistically alike to the hybrids 8-833 and H-53 (4288 and 4030 kg/ha, respectively). The cycle 4 synthetic showed the lowest stunting percentage (8.9%), the commercial tolerant variety NB-6 showed 16.2% and the susceptible hybrid (B-833) 22.8%. A lineal regression between yield averages and the selection cycles reveal an average gain per cycle of 149 kg/ha (4.3 % per cycle) and a reduction per cycle of 2% of stunted plants (11 % gain/cycle). It is concluded that the proceedings among S1 lines has been effective in improving the performance of population 73, increasing productivity of grain yield and the stunting resistance.

Abstract:

Genotypic differences in transpiration rate responses to high vapour pressure deficit (VPD) was earlier reported. Here we tested the hypothesis that this limitation could relate to different degrees of dependence on the apoplastic (spaces between cells), and symplastic water transport pathways (through cells via aquaporin-facilitated transport), which are known to have different hydraulic conductivities. The low transpiration rate (Tr) genotype PRLT 2/89/33 either restricted its transpiration under high VPD, or was more sensitive to VPD than H77/833-2, when grown hydroponically or in soil. The slope of the transpiration response to an ascending series of VPD was lower in whole plants than in de-rooted shoots. In addition, the transpiration response of detached leaves to moderately high VPD (2.67 kPa), normalised against leaves exposed to constant VPD (1.27 kPa), was similar in low and high Tr genotypes. This suggested that roots hydraulics were a substantial limitation to water flow in pearl millet, especially under high VPD. The dependence on the apoplastic and symplastic water transport pathways was investigated by assessing the transpiration response of plants treated with inhibitors specific to the AQP-mediated symplastic pathway (AgNO3 and H2O2) and to the apoplastic pathway (precipitates of Cu(Fe(CN)6) or Cu(CuFe(CN)6)). When CuSO4 alone was used, Cu ions caused an inhibition of transpiration in both genotypes and more so in H77/833-2. The transpiration of high Tr H77/833-2 was decreased more by AQP inhibitors under low VPD (1.8 kPa) than in PRLT 2/89/33, whereas under high VPD (4.2 kPa), the transpiration of PRLT 2/89/33 was decreased more by AQP inhibitors than in H77/833-2. The transpiration rate of detached leaves from H77/833-2 when treated with AgNO3 decreased more than in PRLT 2/89/33. Although the root hydraulic conductivity of both genotypes was similar, it decreased more upon the application of a symplastic inhibitor in H77/833-2. The transpiration of low Tr PRLT 2/89/33 was decreased more by apoplastic inhibitors under both low and high VPD. Then the hydraulic conductivity decreased more upon the application of an apoplastic inhibitor in PRLT 2/89/33. In conclusion, both pathways contributed to water transport, and their contribution varied with environmental conditions and genotypes. Roots were a main source of hydraulic limitation in these genotypes of pearl millet, although a leaf limitation was not excluded. The similarity between genotypes in root hydraulic conductivity under normal conditions also suggests changes in this conductivity upon changes in the evaporative demand. The low Tr genotype depended more on the apoplastic pathway for water transport, whereas the high Tr genotype depended on both pathway, may be by ‘tuning-up’ the symplastic pathway under high transpiration demand, very likely via the involvement of aquaporins.

В возрасте 6 месяцев средний вес представителей симментальской породы составлял 175 кг, голштинской - 163 кг, симментальская × голштинская - 171 кг, абердин-ангусская × голштинская - 178 кг, Шароле × голштинская - 176 кг и зебу × симментальская - 192 кг. Целью исследования стало определение генотипа с оптимальным результатом роста и развития. В результате вскармливания привес от рождения до 6 месяцев составил: симментальская - 761 г, голштинская - 716 г, симментальская × голштинская - 750 г, абердин-ангусская × голштинская - 833 г, Шароле × голштинская - 804 г и зебу × симментальская - 844 г. В результате был сделан вывод, что сбалансированное питание положительно влияет на динамику роста и развитие всех исследуемых пород. The research was carried out at the training center for animal husbandry and horse breeding of the Azerbaijan State Agrarian University. Several breeds of cows have been studied. At the age of 6 months, the average weight of the representatives of the Simmental breed was 175 kg, Holstein - 163 kg, Simmental × Holstein - 171 kg, Aberdeen Angus × Holstein - 178 kg, Charolais × Holstein - 176 kg and Zebu × Simmental - 192 kg. The aim of the study was to determine the genotype with the optimal result of growth and development. As a result of feeding, the weight gain from birth to 6 months was: Simmental - 761 g, Holstein - 716 g, Simmental × Holstein - 750 g, Aberdeen-Angus × Holstein - 833 g, Charolais × Holstein - 804 g and Zebu × Simmental - 844 g. As a result, it was concluded that a balanced diet has a positive effect on the dynamics of growth and development of all the studied breeds.

Abstract:

We have identified the glucose-6-phosphate dehydrogenase mutations responsible for enzyme deficiency in nine individuals with chronic nonspherocytic hemolytic anemia. We found the variants Tokyo, Iowa, Shinshu, and Guadalajara in British subjects and Kobe in an Italian. In addition we have determined the variant Corum has the mutation 820 G-->A and have found in British subjects the mis-sense mutations 224 T-->C, 488 G-->A and 833 C-->T which have not been described before. Some, but not all, of the mutations involve amino acids located near putative substrate binding sites.

Phys. 6 (1994) 833–853] and where we determined the leading behaviour of the spectral shift function and the scattering amplitude as the separation of the wells tends to infinity. In this article we determine the explicit form of the subleading contributions, which in particular show strong oscillatory behaviour. Also we apply our methods to the critical and subcritical double well problems.

Aneurysm of interventricular septum (IVSA) is a rare congenital anomaly. It has been documented in isolation but is seen in association with congenital heart disease in 0.3% of cases and in association with ventricular septal defect in 19% of cases. Etiologies are considered to be idiopathic formation, trauma, infection or spontaneous closure of pre-existing ventricular septal defect. Clinical manifestations are variable, some patients are asymptomatic whereas others can present with hemodynamic compromise. Abstract:

P-glycoprotein (pgp), which is the product of the MDR1(multidrug resistance-1) gene, has an established role as a mediator of cytotoxic drug resistance in acute myeloid leukemia (AML). To study the role of pgp in mediating apoptosis resistance in AML cells deprived of serum and growth factors, apoptosis was quantified by flow cytometry using uptake of the dye 7-amino-actinomycin D (7-AAD) alongside low forward scatter. In pgp+ve primary AML samples, there was a significant increase in apoptosis in the presence of the pgp-specific antibody UIC2 (mean increase: 58%; range: 11%-95%; P < .05). Likewise, apoptosis in growth factor–deprived TF1 cells cultured for 30 hours increased 2.5-fold in the presence of 25 μg/mL UIC2. The pgp reversal agent PSC-833 (1 μmol/L) augmented in vitro apoptosis by a median of 52% in pgp+ve patient samples and to a comparable degree in 6 pgp−ve samples. To determine whether the sphingomyelin-ceramide (SM-ceramide) pathway of apoptosis occurs in AML blasts in response to cytotoxic drugs, cells were incubated with daunorubicin at the patient-specific IC30 (the concentration of daunorubicin that caused apoptotic cell death in 30% of cells) in the presence of the ceramide synthase inhibitor fumonisin B1, which inhibited apoptosis by 18%-81% (median: 40%). Exogenous SM failed to augment apoptosis induced by growth factor withdrawal in pgp+ve TF1 cells and was significantly more effective at augmenting apoptosis in pgp−ve patient blasts (median increase in cell death: 33%; range: 19%-88%) than in pgp+ve samples (median: 7%; range: 0%-27%;P = .028). Cellular accumulation of exogenous SM was associated with apoptosis and also occurred in nonapoptotic patient cells treated with PSC-833. However, this effect was not seen following treatment with the UIC2 antibody. These results indicate that pgp is able to exert a protective effect on AML cell viability and that this is associated with a reduced effect of exogenous SM on apoptosis. The pgp reversal agent PSC-833 acts, at least in part, by a pgp independent mechanism to alter SM distribution and to augment apoptosis induced in AML cells by serum and growth factor withdrawal.

Abstract:

Abstract. The impact of acid deposition and tree harvesting on three lakes and their representative sub-catchments in the Muskoka-Haliburton region of south-central Ontario was assessed using a critical loads approach. As nitrogen dynamics in forest soils are complex and poorly understood, for simplicity and to allow comparison among lakes and their catchments, CLs (A) for both lakes and forest soils were calculated assuming that nitrate leaching from catchments will not change over time (i.e. a best case scenario). In addition, because soils in the region are shallow, base cation weathering rates for the representative sub-catchments were calculated for the entire soil profile and these estimates were also used to calculate critical loads for the lakes. These results were compared with critical loads obtained by the Steady State Water Chemistry (SSWC) model. Using the SSWC model, critical loads for lakes were between 7 and 19 meq m-2yr-1 higher than those obtained from soil measurements. Lakes and forests are much more sensitive to acid deposition if forests are harvested, but two acid-sensitive lakes had much lower critical loads than their respective forested sub-catchments implying that acceptable acid deposition levels should be dictated by the most acid-sensitive lakes in the region. Under conditions that assume harvesting, the CL (A) is exceeded at two of the three lakes and five of the six sub-catchments assessed in this study. However, sulphate export from catchments greatly exceeds input in bulk deposition and, to prevent lakes from falling below the critical chemical limit, sulphate inputs to lakes must be reduced by between 37% and 92% if forests are harvested. Similarly, sulphate leaching from forested catchments that are harvested must be reduced by between 16 and 79% to prevent the ANC of water draining the rooting zone from falling below 0 μeq l-1. These calculations assume that extremely low calcium leaching losses (9–27 μeq l-1) from forest soils can be maintained without any decrease in forest productivity. Calcium concentrations in the three lakes have decreased by between ∼10 and 25% over the past 20 years and calculations assume that calcium concentrations in lakes can fall to around 30% of their current values without any harmful effects on biota. Components of the pig innate immune system, such as neutrophils, have not been fully described, and epigenetic mechanisms controlling gene expression remain unclear in pigs. We performed RNA- and assay for transposase-accessible chromatin (ATAC)-sequencing on two porcine neutrophil samples to characterize epigenetic regulation of gene expression in these differentiated cells. A deep neutrophil transcriptome detected expression of 10,974 genes. In comparison to previously reported RNA-seq data on eight sorted immune cell types from the same animals, we predicted 833 enriched (significantly higher and 2-fold higher than average across all other cell types) genes in neutrophils. The 833 gene list was significantly enriched for biological processes related to typical neutrophil functions using Gene Ontology and in comparison to human neutrophil RNA-seq data. ATAC-seq detected open chromatin (OP) regions from ~227 million total reads that identified on average 151,789 peaks located in intergenic (67%), intron (24%), promoter-TSS (6%), exon (3%) or TTS (1%) regions. OP regions whose signal is correlated with, and nearby to, genes with enriched neutrophil expression are being analyzed for sites predicted to be bound by transcription factors that could explain neutrophil-enriched expression patterns. The data reported here is the first integrative study of the epigenetics of porcine neutrophil gene expression and provide a valuable resource to elucidate neutrophil transcriptional regulatory networks.

Objective 1) Report the incidence and outcome of delayed facial nerve palsy following middle ear surgery in our department, and 2) review the medical literature. Methods This is a retrospective chart review study of 833 patients who were operated for chronic otitis media with cholesteatoma (572 patients), otosclerosis (192 patients), and profound hearing loss who received a cochlear implant (68 patients) since 1993. Results Delayed facial nerve palsy was observed in 10 patients (1.2%). In all cases, immediate postoperative facial nerve function was normal and the palsy occurred 5 to 8 days postoperatively. Facial nerve function recovered in all patients within 6 months. Assessment and management issues are discussed. Conclusions Delayed facial nerve palsy may rarely occur following middle ear surgery and has an excellent recovery rate.

Abstract P-glycoprotein (Pgp) is a 170-kDa membrane transporter that mediates drug efflux and is an effector of multidrug resistance. SDZ PSC 833 (PSC), a nonimmunosuppressive cyclosporine that potently modulates Pgp, is currently under clinical evaluation in patients with cancer. We have developed a reversed-phase HPLC assay for determining PSC blood concentrations that utilizes a step gradient with linear segments to resolve PSC into two distinct peaks (likely to be keto and enol isomers). To clinically validate the assay, PSC concentrations were obtained by HPLC from nine patients receiving oral doses of 5 mg/kg every 6 h. Values ranged from 0.91 to 5.4 mg/L during the dosing period, comparable with concentrations of PSC that modulate Pgp in vitro. In addition, we investigated the immunoreactivity of the Abbott TDx cyclosporin A (CsA) monoclonal whole-blood assay for PSC. The TDx CsA assay cross-reacts ∼17% with PSC as determined by adding known amounts of PSC to whole blood. When PSC concentrations obtained by the TDx CsA assay were divided by 0.17, we found agreement between the TDx CsA assay and the HPLC PSC assay for samples from nine patients.

Abstract Background The eighth edition of the American Joint Committee on Cancer TNM staging for esophageal cancer will be implemented at the beginning of 2018. The nodal staging process in the eighth edition remains unchanged from that in the seventh edition in that it was based on the number of lymph nodes (LNs) involved, but the regional lymph node map has been revised. The aortopulmonary (station 5), anterior mediastinal (station 6), and tracheobronchial (station 10) nodes have been omitted from the regional lymph node map for the new TNM staging. However, the role and prognostic significance of these LN stations are not clear. The purpose of this study was to investigate whether the revised nodal staging used in the eighth edition staging system is appropriate, and to verify the role, prognostic significance, and therapeutic value of these LNs in esophageal cancer. Methods The records of patients who underwent esophagectomy for cancer in our department between January 2007 and January 2013 were retrospectively analyzed. The rate of metastases and the index of estimated benefit from lymph node dissection (IEBLD) were calculated for stations 5, 6, and 10 LNs. LN metastasis and patient survival were analyzed and the efficacy of the eighth edition TNM staging system was verified. Results A total of 1637 patients (1350 men, 287 women) were included. The frequencies of dissection of stations 5, 6, and 10 LNs were 34.3% (562/1637), 15.9% (260/1637), and 50.9% (833/1637), respectively. The calculated rate of metastasis to these stations was 3.2% (18/562), 2.3% (6/260), and 4.9% (41/833), respectively. No difference was found in the N stage determined by the seventh and eighth edition N staging systems. The survival curves differed significantly between N stages calculated using the eighth edition TNM system (P < 0.001). The IEBLD values of stations 5, 6, and 10 LNs were 0.57, 0, and 0.97, respectively. Station 5 or 10 LN(+ ) patients had worse median survival time and 5-year overall survival rate compared with LN(–) patients (P < 0.01). Univariate analysis showed that differentiation, T stage, N stage (both seventh and eighth edition calculations), and metastasis to stations 5 and 10 LNs were associated with long-term survival. Conclusion Metastasis to stations 5, 6, or 10 LNs was infrequent. If stations 5, 6, and 10 LNs were omitted in the eighth edition calculation to determine the N stage based on the number of metastatic LNs, this did not influence the accuracy and survival-predicting efficacy of the eighth edition TNM staging. The therapeutic value of lymphadenectomy of stations 5, 6, and 10 was limited. Metastasis to stations 5, 6, and 10 LNs indicated more advanced N stage, which was associated with poor survival. However, no survival difference was found between station 6 LN(+ ) and LN(–) subgroups, possibly because of the limited numbers of cases. Considerando o excesso de peso em escolares, objetivamos analisar a incidência de sobrepeso e obesidade nas turmas do 6° ano do Ensino Fundamental de um Colégio da Rede Pública Estadual em Ponta Grossa – Paraná, entre os anos de 2015 a 2019. Para tanto, utilizamos os recursos metodológicos da pesquisa exploratória. Ao todo, foram analisados os índices de massa corporal de 833 alunos. Desse modo, identificamos que 26,94% dos alunos do sexo masculino e 33,75% das alunas do sexo feminino apresentaram excesso de peso, o que permite concluir que tais comorbidades são predominantes nas alunas e é necessário intervenções para a mudança neste quadro dentro da escola ao longo dos anos finais do ensino fundamental para que essa predominância diminua e não haja complicações na idade adulta. Também observamos que os dados da escola de Ponta Grossa são superiores aos nacionais, porém inferiores ou muito parecidos com outros dados da região sul.

Abstract Background Recent studies suggest a measles-induced immune amnesia that could have long-term immunosuppressive effects via preferential depletion of memory B and T CD150+ lymphocytes. Methods We examined the association between past measles and tetanus antibody levels among children participating in the 2013–2014 Democratic Republic of Congo (DRC) Demographic and Health Survey (DHS). Our sample consisted of 833 children aged 6–59 months whose mothers were selected for interview. Mothers reported (via recall) history of measles within the lifetime of the child. Classification of children who previously had measles was completed using maternal recall and measles immunoglobulin G (IgG) serostatus obtained via dried blood spot (DBS) analysis. A multiplex chemiluminescent immunoassay platform was used to obtain serologic results and Assay Score (AS) was calculated as a ratio to a positive control included in each run. Tetanus serostatus was categorized as being above or below the sample median serology AS value. Tetanus vaccination status was obtained via dated vaccination card and limited to children receiving the complete 3-dose vaccination series. Results The median AS for tetanus serology among the entire sample of 833 children was 0.085, while children with history of measles had a median AS of 0.053 (N = 41) and children with no history of measles had a median AS of 0.088 (N = 792), chi-square P-value < 0.05. A random intercept logistic regression model was used to examine the association between previous measles disease and odds of having below median levels of tetanus antibody. Controlling for potential confounding variables, the odds of a child with past history of measles having less than the median level of tetanus antibody was 3.86 (95% CI: 1.70, 8.78) among children fully vaccinated for tetanus. Conclusion The results suggest that, among children 6–59 months in DRC, measles may have a long-term impact on levels of pre-existing, vaccine-induced immunity to tetanus. These findings suggest the need for laboratory studies examining measles’ impact on pre-existing, vaccine-induced immunity and underscore the need for continued evaluation and improvement of DRC’s measles vaccination program. SUMMARYThere are an estimated 277 000 cases of campylobacteriosis in Australia each year, most of which are thought to be sporadically acquired. To explore causes for these infections, we conducted a multi-centre case-control study of patients and community controls across five Australian States during 2001–2002. A total of 881 campylobacter cases and 833 controls aged ⩾5 years were recruited into the study. Crude logistic analyses were conducted within various food and non-food exposure groups. A final most parsimonious multivariable logistic regression model was developed and adjusted odds ratios (aOR), 95% confidence intervals (95% CI) were derived together with adjusted population attributable risks (PAR). Consumption of undercooked chicken (aOR 4·7, 95% CI 2·6–8·4) and offal (aOR 2·0, 95% CI 1·0–4·0), ownership of domestic chickens aged <6 months (aOR 12·4, 95% CI 2·6–59·3) and domestic dogs aged <6 months (aOR 2·1, 95% CI 1·1–4·2) were found to be independent risk factors for illness in the final model. The PAR proportions indicate that eating chicken meat, either cooked or undercooked may account for approximately 30% of campylobacter cases that occur each year in Australia. These results justify the continued need for education of consumers and foodhandlers about the risks associated with the handling of raw chicken and the potential for cross-contamination.

Abstract:

Trehalose-6-phosphate synthase (TPS) plays an important role in the synthesis of trehalose. In the current study, a TPS gene was obtained from Diaphorina citri, and named as DcTPS1 which encoded a protein of 833 amino acid residues. Real-time quantitative PCR (qPCR) analysis revealed that DcTPS1 had the highest expression level in the midgut and fifth-instar nymph stage. Knockdown of DcTPS1 by RNA interference (RNAi) induced an abnormal phenotype and increased mortality and malformation rate with a decreased molting rate. In addition, silencing of DcTPS1 significantly inhibited D. citri chitin metabolism and fatty acid metabolism, while the expression levels of fatty acid decomposition-related genes were downregulated. Furthermore, comparative transcriptomics analysis revealed that 791 differentially expressed genes (DEGs) were upregulated and 678 DEGs were downregulated when comparing dsDcTPS1 groups with dsGFP groups. Bioinformatics analysis showed that upregulated DEGs were mainly involved in oxidative phosphorylation, whereas downregulated DEGs were mainly attributed to the lysosome and ribosome. These results indicated that DcTPS1 played an important role in the growth and development of D. citri.

During childhood and pre-adolescence, the family environment is key to initiating and consolidating healthy styles in children through a balanced diet and basic hygiene habits. This study analyses hygiene, nutrition and health practices in Spanish families with children between 6 and 14 years of age according to the type of family (nuclear, single-parent or reconstituted) and the quantity, age and gender of the children. A representative Spanish national sample of 1103 Spanish parents, 270 fathers and 833 mothers, with children aged 6 to 14, is analysed. The study is descriptive, using statistical techniques with classic indicators (means, percentages). The results show that nuclear families manifest healthier habits, in general, and consider the consumption of pastries, ultra-processed food and excessive amounts of salt to be harmful. Furthermore, this family typology develops healthy and hygienic habits, such as brushing teeth daily, sleeping at least 8 h a day, drinking a glass of milk a day, eating fish more than once a week and eating fast food sporadically. They are also concerned about their children’s annual medical check-ups (paediatrician and dentist). It is concluded that the family type is related to the hygiene and feeding habits of the children.

e16720 Background: Pancreatic cancer (PC) is the third most lethal form of cancer in the United States, and is most often diagnosed having already metastasized, making its 5-year survival rate one of the lowest. Metastatic PC (mPC) is most common at sites such as liver, lymph nodes, and lung while, in contrast, brain metastases from PC are rare. Brain metastases present a major challenge for future oncological research, as they are difficult to treat. If PC patients most likely to develop brain metastases can be identified earlier, there is an opportunity for rapid therapeutic intervention. There is limited research and understanding of the relationship between pancreatic cancer and brain metastases. Methods: We performed a retrospective, non-interventional study using deidentified data. Inteliquet’s data was aggregated from its consortium of 175 US healthcare locations, which includes a variety of source systems at each site. These sources include electronic medical record systems, laboratory information management systems, and other sources. A subset of the adult mPC data originally treated at HonorHealth was identified as an exploratory cohort. Results: 833 patient records were identified with drug treated mPC, 33 (4%) of which had metastasized to the brain. The PC dataset had a median diagnosis age of 65 and was 46% female. The site of the primary tumor within the pancreas was acquired from ICD10 code: 35% were in the head, 14% in the tail, 15% in the body, 5% in overlapping sites, 6% in other parts and 26% where location was unspecified. Metastatic locations came from secondary malignancy ICD10 codes: 63% had liver and 25% had lung metastases, which aligns with prior studies. For patients with brain metastases, the gender distribution was 42% female, while the distribution of primary PC site was: 33% head, 9% tail, 6% body, 6% in overlapping locations, 12% in other parts and 33% with unspecified location. The median age of diagnosis in the brain metastasis group was 67 years. Conclusions: This is the largest study of PC patients with brain metastases that we could find. 33 patients out of 833 had brain metastases compared to a recent review which had 25 cases. Our data suggests that specification of the primary pancreatic site is more difficult, aside from the head of the pancreas. Analyses are underway to explore correlations between other clinical factors and brain metastases, and to calculate the time in between the initial pancreatic cancer diagnosis and detection of the brain metastasis. This hypothesis-generating cohort will be tested in patient data from the rest of the consortium.

At the present moment, corruption crimes committed by civil service employees not only interfere with the work of public officials endowed with authority, but they also undermine the credibility of state power, the trust and respect of people for state institutions and, primarily, for equitable justice. The article describes the experience of counteracting corruption in the practice of courts and judicial bodies, including the High Qualification Board of Judges, the Councils of Judges of the Russian Federation and of its regions. Adhering to the requirements of international law and taking into consideration the national law systems, the authors based their research on the inter-disciplinary systemic approach, which is necessary for the effective prevention of corruption in the court system. The theoretical results of the research were reflected in determining both the general regularities of combating corruption in the court system and the specific characteristics that take into consideration the structural and functional features of court power and the legal status of public officials. The practical results include suggestions on optimizing the organization of corruption prevention in the court system, the cooperation of courts and judicial bodies with other state bodies and public institutions on counteracting corruption while preserving the independence of the judicial power. The practical conclusions could be used for optimizing the work of courts and judicial bodies connected with the organization and implementation of anti-corruption measures.

Abstract:

The use of lactic acid bacteria (LAB) in the dairy industry has undergone an enormous evaluation in technologically advanced countries. The exploitation of this kind is however very limited in some other countries such as Algeria. This limitation concerns the diversity of products present on the market but also the number of strains introduced in dairy products, this is related to the constraints posed by LAB. To isolate new probiotic strains, samples of cow's milk were subjected to spontaneous fermentation to enrich its LAB. 21 lactic acid isolates active against four pathogenic and food spoilage bacteria were selected from sixty-one isolated LAB, and only ten strains were selected showing considerable adhesive capacities. Indeed, they have moderate to high levels of hydrophobicity. They can all auto-aggregate with percentages ≥85.86 ± 0.59%. The power to co-aggregate with 4 pathogenic strains differs from strain to strain. Thus, strains C2, H9, V5, and L7 showed different percentage levels of 56.06 ± 0.36%, 32.33 ± 0.63%, 22.60 ± 079%, and 6.55 ± 0.20% against B. cereus, S. aureus, P. aeruginosa, and E. coli respectively. They were all resistant to the acidity of the stomach environment and have a good tolerance to 0.3% and 0.5% bile salts. All isolates were able to grow at 0.4% phenols after exposure for 24 h. Only our lactic acid bacteria V1 and V8 showed resistance after exposure to pepsin at pH =2 and all strains at pH =3. The results obtained are interesting which makes our strains presumed probiotic, they can play an important role in the gastrointestinal tract (GIT).

Abstract:

Transforming growth factor-alpha (TGF-alpha) and epidermal growth factor (EGF) are similar in structure and biological activity. In the present study, the distributions of TGF-alpha mRNA, TGF-alpha immunoreactivity, and TGF-alpha-EGF receptor mRNA were examined in epithelial and nonepithelial compartments of the jejunum, and the effect of TGF-alpha on growth of a jejunal crypt cell line (IEC-6) was determined. Epithelial cells eluted from the rat jejunal cryptvillus axis expressed TGF-alpha mRNA at twofold higher levels in the villus tip than in the crypt and EGF receptor mRNA at sevenfold higher levels in the villus tip. Expression of these two mRNA transcripts in the subepithelium was low. Immunohistochemical staining showed TGF-alpha immunoreactivity predominantly in the epithelium and muscularis. Immunostaining of villus cells was uniform, whereas crypt cells did not stain. IEC-6 cells bound 125I-EGF to a single class of high-affinity (dissociation constant = 833 pM) receptors. EGF and TGF-alpha (10 ng/ml) only modestly stimulated IEC-6 cell growth in the presence of 5% serum but increased expression of the protooncogenes c-jun and c-myc threefold over control cells. These findings suggest that, among the potential physiological roles for TGF-alpha produced by the jejunal epithelium, promotion of cell migration and modulation of fluid and electrolyte transport may be as relatively important as stimulation of cell proliferation.

Abstract Objectives When bloodstream infections are caused by resistant bacteria, rapid antimicrobial susceptibility testing (RAST) is important for adjustment of therapy. The EUCAST RAST method, directly from positive blood cultures, was validated in a multi-laboratory study in Europe. Methods RAST was performed in 40 laboratories in northern Europe (NE) and 15 in southern Europe (SE) from clinical blood cultures positive for Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Staphylococcus aureus or Streptococcus pneumoniae. Categorical results at 4, 6 and 8 h of incubation were compared with results for EUCAST standard 16–20 h disc diffusion. The method, preliminary breakpoints and the performance of the laboratories were evaluated. Results The total number of isolates was 833/318 in NE/SE. The number of zone diameters that could be read (88%, 96% and 99%) and interpreted (70%, 81% and 85%) increased with incubation time (4, 6 and 8 h). The categorical agreement was acceptable, with total error rates in NE/SE of 2.4%/4.9% at 4 h, 1.1%/3.5% at 6 h and 1.1%/3.3% at 8 h. False susceptibility at 4, 6 and 8 h of incubation was below 0.3% and 1.1% in NE and SE, respectively, and the corresponding percentages for false resistance were below 1.9% and 2.8%. After fine-tuning breakpoints, more zones could be interpreted (73%, 89% and 93%), with only marginally affected error rates. Conclusions The EUCAST RAST method can be implemented in routine laboratories without major investments. It provides reliable antimicrobial susceptibility testing results for relevant bloodstream infection pathogens after 4–6 h of incubation.

Abstract:

A genetic predisposition to central nervous system (CNS) toxicity induced by antimicrobial drugs (antibiotics, antivirals, antifungals, and antiparasitic drugs) has been suspected. Whole genome sequencing of 66 cases and 833 controls was performed to investigate whether antimicrobial drug-induced CNS toxicity was associated with genetic variation. The primary objective was to test whether antimicrobial-induced CNS toxicity was associated with seventeen efflux transporters at the blood-brain barrier. In this study, variants or structural elements in efflux transporters were not significantly associated with CNS toxicity. Secondary objectives were to test whether antimicrobial-induced CNS toxicity was associated with genes over the whole genome, with HLA, or with structural genetic variation. Uncommon variants in and close to three genes were significantly associated with CNS toxicity according to a sequence kernel association test combined with an optimal unified test (SKAT-O). These genes were LCP1 (q = 0.013), RETSAT (q = 0.013) and SFMBT2 (q = 0.035). Two variants were driving the LCP1 association: rs6561297 (p = 1.15x10-6, OR: 4.60 [95% CI: 2.51–8.46]) and the regulatory variant rs10492451 (p = 1.15x10-6, OR: 4.60 [95% CI: 2.51–8.46]). No common genetic variant, HLA-type or structural variation was associated with CNS toxicity. In conclusion, CNS toxicity due to antimicrobial drugs was associated with uncommon variants in LCP1, RETSAT and SFMBT2.

Abstract:

The effect of the transesterification reaction time of 0.6% mass % NaOH catalyst in the formation of methyl ester (biodiesel) made from used waste cooking oil as raw material 6 times usage and methanol 1: 5 mass has been studied at 60°C. The reaction times used were 30, 40, and 45 minutes. All samples resulting from the transesterification reaction have been studied for their chemical and physical properties. Parameters of chemical properties included in the Indonesian National Standard (SNI) for all samples were % FFA, pH, while the water content was above the SNI standard. The physical property parameters for all samples that meet SNI are only flash point, color and 90% distillation. While the density of all test samples was lower than the SNI which was set at 850-890 Kg/m3, the results obtained for reaction times of 30, 40 and 45 minutes were 828, 833 and 836 Kg/m3 respectively. The highest yield was owned by a reaction time of 40 minutes, namely 80%. As for the reaction time of 35 and 45 minutes respectively 79 and 65%.

This study was an interpersonal communication study on the politeness methods used by the main characters in the film Joy in the setting mostly of a family cum workplace communication. The purpose of this study was to describe the major characters' politeness tactics. The data for this descriptive qualitative study, the dialogues uttered by all of the movie's principal characters, were scrutinized using Brown and Levinson's (1987) paradigm. The investigation studied how the characters used politeness strategies such as positive politeness, Bald-on -Record, negative politeness and off-record techniques in their utterances. The data came from the script of Joy, and it was identified that 833 politeness strategies were employed. Bald on Record (8%), Positive Politeness (51%), Negative Politeness (35%), and Off Record (6%). The findings revealed that positive politeness strategies were the most frequently used in this film. The results indicated that the main characters had a tendency to use positive politeness to show their respect and also to maintain a harmonious relationship in the family and workplace.

FMS patients demonstrate an altered profile of chemokines relative to healthy controls (HC). Eotaxin-2 is a potent chemoattractant distributed in a variety of tissues. The aim of the study was to compare serum levels of eotaxin-2 between FMS patients and HC and to examine a potential correlation between eotaxin-2 levels and clinical parameters of FMS. Methods. 50 patients with FMS and 15 HC were recruited. Data on the severity of FMS symptoms and depression were collected. Serum levels of eotaxin-2 (ELISA) were determined in all participants. High-sensitive CRP (hs-CRP) was measured in the FMS group. Results. The FMS cohort included predominantly females (84%), mean age of 49, and mean disease duration of 6 years. FMS patients exhibited significantly higher eotaxin-2 levels (pg/ml) versus HC: 833 (±384) versus 622 (±149), p=0.04. Mean hs-CRP level among FMS patients was 4.8 ± 6 mg/l, a value not indicative of acute inflammation. No correlation was found between eotaxin-2 and hs-CRP levels. No correlation was found between eotaxin-2 and severity measures of FMS or depression. Conclusion. Eotaxin-2 does not appear to be a candidate for a disease activity biomarker in FMS. Further research is warranted into the role of this chemokine in the pathophysiology of the FMS.

Functional and morphological alterations were generated by p.o. (per os) administration of a single oral dose of carbon tetrachloride (CCl4; 0.125 mL/kg b.w., equivalent to 293 mg/kg) to adult male Wistar rats. CCl4 significantly increased (p < 0.05) the serum activities of alanine aminotransferase (ALT; 7478 ± 1044%) and aspartate aminotransferase (AST; 6964 ± 833%), compared to control rats; CCl4 also significantly decreased serum concentration of albumin (23 ± 5.5%) and increased the concentration of malondialhdeyde (MDA) in liver (300 ± 33%). Furthermore, CCl 4 down-regulated the mRNA steady-state level of tumor necrosis factor a(TNF-a). CCl4 produced necrosis in the central lobe area, extended to the periphery, nuclear alterations (pycnosis, karyolysis and karyorrhexis), and cytoplasmic acidophilia. The pretreatment with 4 mg/kg (p.o.) of Ginkgo biloba extract (GbE), for 5 days, prevented most of the damage caused by CCl4: significantly decreased the serum activities of ALT and AST (54 and 65%, respectively), compared to CCl4-treated rats; GbE partially prevented the increase of liver MDA (55 ± 14%) and the decrease of albumin concentration to 12 ± 0.2%. This pretreatment prevented the down-regulation of TNF-a and up-regulated the interleukine 6 (IL-6) mRNA steady-state level. Moreover, the GbE reduced the amount of necrotic areas in the central lobe area, compared to CCl4-treated rats.

Abstract:

Studies have suggested that maternal PUFA status during pregnancy may influence early childhood allergic diseases, although findings are inconsistent. We examined the relationship between maternal PUFA status and risk of allergic diseases in early childhood in an Asian cohort. Maternal plasma samples from the Growing Up in Singapore Towards Healthy Outcomes mother–offspring cohort were assayed at 26–28 weeks of gestation for relative abundance of PUFA. Offspring (n 960) were followed up from 3 weeks to 18 months of age, and clinical outcomes of potential allergic diseases (rhinitis, eczema and wheezing) were assessed by repeated questionnaires. Skin prick testing (SPT) was also performed at the age of 18 months. Any allergic disease with positive SPT was defined as having any one of the clinical outcomes plus a positive SPT. The prevalence of a positive SPT, rhinitis, eczema, wheezing and any allergic disease with positive SPT was 14·1 % (103/728), 26·5 % (214/808), 17·6 % (147/833), 10·9 % (94/859) and 9·4 % (62/657), respectively. After adjustment for confounders, maternal total n-3, n-6 PUFA status and the n-6:n-3 PUFA ratio were not significantly associated with offspring rhinitis, eczema, wheezing, a positive SPT and having any allergic disease with positive SPT in the offspring (P>0·01 for all). A weak trend of higher maternal n-3 PUFA being associated with higher risk of allergic diseases with positive SPT in offspring was observed. These findings do not support the hypothesis that the risk of early childhood allergic diseases is modified by variation in maternal n-3 and n-6 PUFA status during pregnancy in an Asian population.

Дано описание макета нового компактного лазерного локатора, предназначенного для дистанционной регистрации характеристик морского волнения с борта судна. Прибор регистрирует с помощью ПЗС-линейки отраженное от морской поверхности лазерное излучение, имеющее форму «веера». В макете использован лазерный диод Oclaro HL63193, работающий в непрерывном режиме в области длин волн 634-637 нм с выходной мощностью до 500 мВт. Освещаемый лазером участок на морской поверхности представляет собой полосу шириной 4-5 мм и длиной 2-3 м (при размещении макета на судне на высоте 6-9 м над уровнем моря). Приемный блок макета выполнен на базе кинопроекционного объектива ОП-55АР и ПЗС-линейки Sony ILX554A. Выполнена проверка работоспособности макета в лабораторных и морских условиях. Показано, что скорость сканирования морской поверхности может быть значительно выше (до 833 Гц), чем у ранее созданных макетов с узким пучком, когда используется механическое сканирование зеркала (7-15 Гц). Это позволяет увеличивать скорость накопления данных и тем самым уменьшать дисперсию измеряемых за заданное время величин, что в свою очередь делает возможным регистрацию более слабых воздействий гидродинамических процессов на морскую поверхность. При этом масса макета уменьшилась до 5 кг вместо 35 кг (у макета с узким пучком и механическим сканером).

Este trabalho teve como objetivo caracterizar a composição florística e fitossociológica de um fragmento de caatinga, localizado no município de Jardim de Piranhas, Rio Grande do Norte. Realizou-se uma amostragem inteiramente aleatória, a partir de 15 unidades amostrais de 20 x 20 (400 m²). Foi inventariado para cada unidade amostral, todos os indivíduos florestais arbustivo-arbóreos vivos com circunferência da altura do peito (CAP ≥ 6 cm) mensurados a 1,30 m do solo, registrando-se a circunferência, altura total e nome comum por indivíduo. Avaliou-se a composição florística, diversidade e as estruturas vertical e horizontal. Foram amostrados 833 indivíduos distribuídos em onze espécies e seis famílias, sendo a família Fabaceae a mais representativa, com o total de cinco espécies, enquanto as demais famílias apresentaram apenas uma espécie cada. Já as espécies com maior valor de importância foram a Mimosa tenuiflora (Willd.) Poir. e Aspidosperma pyrifolium Mart & Zucc., responsáveis por 67,11% dos indivíduos amostrados. Assim, a diversidade de espécie encontrada no fragmento através dos índices estudados foi considerada fora do padrão normal em áreas de Caatinga, apresentando baixa riqueza e diversidade, com apenas nove espécies, grande heterogeneidade ambiental e baixa dominância ecológica.

Objectives. Infusion of chemotherapy at home provides an alternative to hospitalization for children with cancer. Few programs of pediatric home chemotherapy have been described or evaluated. The purpose of this work was to compare prospectively chemotherapy in the hospital to chemotherapy at home with respect to billed medical charges, out-of-pocket expenses, and quality of life. Methods. Eligibility criteria for home therapy were defined. Parents and nurses were trained. Billed charges, loss of wages, out-of-pocket expenses, medical outcome, and quality of life of 14 patients for one course of chemotherapy in the hospital were compared with those for an identical course at home. Results. Daily charges for chemotherapy were $2329 ± 627 in the hospital and $1865 ± 833 at home; out-of-pocket costs, $68 ± 31 and $11 ± 6, respectively; and loss of income, $265 ± 233 and $67 ± 107, respectively. Patients' independence, well-being, appetite, mood, and school work were significantly better at home, and parental time at work and with the family was greater. Conclusion. Administration of selected chemotherapy at home results in lower billed charges, reduced expenses, reduced loss of income for parents, and a more satisfying lifestyle for patients and parents.

ABSTRACT Aim The aim of this in vitro study was to evaluate the apical seal obtained with four root canal sealers AH 26, Sealapex, Endoflas FS and AH Plus, with lateral condensation. Materials and methods Sixty root canals were prepared using the step-back technique. The specimens were divided into four experimental groups of 12 teeth and two control groups of 12 teeth. The experimental groups were obturated by laterally condensed gutta-percha with one of the tested sealers and control groups were obturated without any sealer. Methylene blue dye penetration with centrifuging method was used to evaluate the apical sealing ability. The quantitative apical leakage of each specimen was measured after 2 weeks. Results The results showed no significant differences between all groups except between AH Plus and Endoflas FS (<0.05). AH Plus showed significantly less leakage than Endoflas FS.
