Abstract:
Glucose homeostasis requires the proper expression and regulation of the catalytic subunit of glucose-6-phosphatase (G-6- Pase), which hydrolyzes glucose 6-phosphate to glucose in glucose-producing tissues. Glucose induces the expression of G-6- Pase at the transcriptional and posttranscriptional levels by unknown mechanisms. To better understand this metabolic regulation, we mapped the cis-regulatory elements conferring glucose responsiveness to the rat G-6- Pase gene promoter in glucose-responsive cell lines. The full-length (−4078/+64) promoter conferred a moderate glucose response to a reporter construct in HL1C rat hepatoma cells, which was dependent on coexpression of glucokinase. The same construct provided a robust glucose response in 832/13 INS-1 rat insulinoma cells, which are not glucogenic. Glucose also strongly increased endogenous G-6- Pase mRNA levels in 832/13 cells and in rat pancreatic islets, although the induced levels from islets were still markedly lower than in untreated primary hepatocytes. A distal promoter region was glucose responsive in 832/13 cells and contained a carbohydrate response element with two E-boxes separated by five base pairs. Carbohydrate response element-binding protein bound this region in a glucose-dependent manner in situ. A second, proximal promoter region was glucose responsive in both 832/13 and HL1C cells, with a hepatocyte nuclear factor 1 binding site and two cAMP response elements required for glucose responsiveness. Expression of dominant-negative versions of both cAMP response element-binding protein and CAAT/enhancer-binding protein blocked the glucose response of the proximal region in a dose-dependent manner. We conclude that multiple, distinct cis-regulatory promoter elements are involved in the glucose response of the rat G-6- Pase gene.

Abstract As models for β-cell metabolism, rat islets are, to some extent, a, heterogeneous cell population stressed by the islet isolation procedure, whereas rat-derived clonal β-cells exhibit a tumor-like phenotype. To describe to what extent either of these models reflect normal cellular metabolism, we compared metabolite profiles and gene expression in rat islets and the INS-1 832/13 line, a widely used clonal β-cell model. We found that insulin secretion and metabolic regulation provoked by glucose were qualitatively similar in these β-cell models. However, rat islets exhibited a more pronounced glucose-provoked increase of glutamate, glycerol-3-phosphate, succinate, and lactate levels, whereas INS-1 832/13 cells showed a higher glucose-elicited increase in glucose-6-phosphate, alanine, isocitrate, and α-ketoglutarate levels. Glucose induced a decrease in levels of γ-aminobutyrate (GABA) and aspartate in rat islets and INS-1 832/13 cells, respectively. Genes with cellular functions related to proliferation and the cell cycle were more highly expressed in the INS-1 832/13 cells. Most metabolic pathways that were differentially expressed included GABA metabolism, in line with altered glucose responsiveness of GABA. Also, lactate dehydrogenase A, which is normally expressed at low levels in mature β-cells, was more abundant in rat islets than in INS-1 832/13 cells, confirming the finding of elevated glucose-provoked lactate production in the rat islets. Overall, our results suggest that metabolism in rat islets and INS-1 832/13 cells is qualitatively similar, albeit with quantitative differences. Differences may be accounted for by cellular heterogeneity of islets and proliferation of the INS-1 832/13 cells.

1, 2, 9, 10-tetramethoxyaporphine phosphate (MDL-832) was once considered a potential human antitussive. MDL-832 was administered orally in the diets of Sprague-Dawley rats at dose levels of 0, 5, 10, 20, 40, 80 and 160 mg/kg/day for 3 and 6 months and in gelatin capsules to Beagle dogs at 0, 5, 10, 15, 30 and 60 mg/kg/day for 3, 6 and 12 months. Histopathologic examinations of hematoxylin and eosin-stained cerebellar sections revealed intracytoplasmic brown pigment accumulations in large fusiform neurons (presumably the motor type) of the pons. The pigment granules were found to be PAS-positive, non-acid fast, iron-free, Sudan B-positive and fuchsinophilic. Intraneuronal pigment accumulations were seen in rats after 3 months of treatment at 80 mg but not at 40 mg and after 6 months at 20 mg but not at 10 mg. For dogs the effect was observed after 3 months at 60 mg but not at 30 mg and after 12 months at 10 mg but not at 5 mg.

Abstract Wolfram syndrome is a rare genetic disorder characterized by juvenile-onset diabetes mellitus, optic nerve atrophy, hearing loss, diabetes insipidus, and progressive neurodegeneration. Pathogenic variants in the WFS1 gene are the main causes of Wolfram syndrome. WFS1 encodes a transmembrane protein localized to the endoplasmic reticulum (ER) and regulates the unfolded protein response (UPR). Loss of function of WFS1 leads to dysregulation of insulin production and secretion, ER calcium depletion, and cytosolic calpains activation, resulting in the activation of apoptotic cascades. Although the terminal UPR is well known to yield an inflammatory response called "sterilized inflammation" that accelerates β-cell dysfunction and death in diabetes, the contribution of β-cell inflammation to the development of diabetes in Wolfram syndrome has not been fully understood. Here we show that WFS1-deficiency enhances the gene expression of pro-inflammatory cytokines and chemokines, leading to cytokine-induced ER-stress and cell death in pancreatic β-cells. Under thechronic[BL1] high-glucose condition, CRISPR/Cas9-mediated Wfs1-knockout (KO) INS-1 832/13 cells, the pancreatic β-cell model of Wolfram syndrome, showed enhanced gene expression levels of ER stress markers (Chop, sXbp1, Bip, and Txnip). Moreover, the gene expression levels of pro-inflammatory cytokines (Il-1β, Il-6), chemokine (Ccl2), and vascular endothelial growth factor A (VegfA) were higher in Wfs1- KO INS-1 832/13 cells when compared to Wfs1-wild type INS-1 832/13 cells. [BL2] The high glucose-induced pro-inflammatory cytokine genes in Wfs1-KO INS-1 832/13 cells were mediated by the PKR-like ER-resident kinase (PERK) and inositol-requiring enzyme 1α (IRE1α)pathways. Furthermore, the IFN-γ and IL-1β treatment enhanced ER stress-mediated cell death and upregulated the pro-inflammatory cytokine gene expression in Wfs1-KO INS-1 832/13 cells. To confirm these findings, we investigated whether inflammation occurs in a mouse model of Wolfram syndrome. Compared to wild-type mice, 10-month-old whole body Wfs1-KO mouse islets showed a significantly higher density of Iba1, which is a marker for macrophage expression. The islet-infiltrated macrophages in these Wfs1-KO mice expressed the M1-macrophage marker (CD68). There was also strong fibrosis and a high density of endothelial cell marker (CD31) in the Wfs1-KO mouse islets. These results demonstrate that WFS1 loss-of-function enhances the inflammatory responses in pancreatic β-cells. The results in our study identified sterile inflammation as a new pathological mechanism in the development of diabetes in Wolfram syndrome. In Wolfram syndrome model mice, islets are highly infiltrated with macrophages, suggesting that the vicious cycle of ER stress and sterile inflammation accelerates the progression of diabetes in a cell-autonomous and cell-nonautonomous manner. Abstract:
Insulin secretion is coupled with changes in β-cell metabolism. To define this process, 195 putative metabolites, mitochondrial respiration, NADP+, NADPH and insulin secretion were measured within 15 min of stimulation of clonal INS-1 832/13 β-cells with glucose. Rapid responses in the major metabolic pathways of glucose occurred, involving several previously suggested metabolic coupling factors. The complexity of metabolite changes observed disagreed with the concept of one single metabolite controlling insulin secretion. The complex alterations in metabolite levels suggest that a coupling signal should reflect large parts of the β-cell metabolic response. This was fulfilled by the NADPH/NADP+ ratio, which was elevated (8-fold; P<0.01) at 6 min after glucose stimulation. The NADPH/NADP+ ratio paralleled an increase in ribose 5-phosphate (>2.5-fold; P<0.001). Inhibition of the pentose phosphate pathway by trans-dehydroepiandrosterone (DHEA) suppressed ribose 5-phosphate levels and production of reduced glutathione, as well as insulin secretion in INS-1 832/13 β-cells and rat islets without affecting ATP production. Metabolite profiling of rat islets confirmed the glucose-induced rise in ribose 5-phosphate, which was prevented by DHEA. These findings implicate the pentose phosphate pathway, and support a role for NADPH and glutathione, in β-cell stimulus-secretion coupling.

Abstract:
Neurotoxins such as rotenone, 1-methyl-4-phenylpyridinium (MPP+) and 6-hydroxydopamine (6-OHDA) are well known for their high toxicity on dopaminergic neurons and are associated with Parkinson’s disease (PD) in murine models and humans. In addition, PD patients often have glucose intolerance and may develop type 2 diabetes (T2D), whereas T2D patients have higher risk of PD compared to general population. Based on these premises, we evaluated the toxicity of these three toxins on pancreatic β-cell lines (INS-1 832/13 and MIN6) and we showed that rotenone is the most potent for reducing β-cells viability and altering mitochondrial structure and bioenergetics in the low nanomolar range, similar to that found in dopaminergic cell lines. MPP+ and 6-OHDA show similar effects but at higher concentration. Importantly, rotenone-induced toxicity was counteracted by α-tocopherol and partially by metformin, which are endowed with strong antioxidative and cytoprotective properties. These data show similarities between dopaminergic neurons and β-cells in terms of vulnerability to toxins and pharmacological agents capable to protect both cell types.

It has been documented in isolation but is seen in association with congenital heart disease in 0.3% of cases and in association with ventricular septal defect in 19% of cases. PURPOSE: The aim of this study was to investigate whether the hind limbs or intestinal tract is the most important initiator of the inflammatory response secondary aortic clamping and hind limb ischemia/reperfusion injury. METHODS: Blood samples of Wistar rats obtained from posterior cava vein, portal vein, and heart cavity during either laparotomy (control group, n=8) or laparotomy + 2 h of aortic clamping and bilateral hind limb ischemia (ischemia group, n=8), or 2 h after ischemia and 2 h of reperfusion (ischemia-reperfusion group, n=8) were assayed for interleukin 6 (IL-6) and C-reactive protein (CRP). RESULTS: Serum IL-6 at the heart (223.6±197.9 [10-832] pg/mL) was higher (p<0.001) than at both portal (133.08±108.52 [4-372] pg/mL) and posterior cava veins (127.58±109.15 [8-388] pg/mL). CRP was not significant different among groups. CONCLUSION: The splanchnic region is also a source of inflammatory response secondary to ischemia and reperfusion of the hind limbs.

Introduction. Available evidence for routine terminal ileoscopy during colonoscopy is equivocal. We investigated the place of routine terminal ileoscopy and biopsy during colonoscopy, in a tropical setting.Materials and Methods. All consenting adults undergoing colonoscopy had routine TI and biopsy. Patients with right iliac fossa (RIF) pain, diarrhoea, anaemia, suspected inflammatory bowel disease (IBD), and raised inflammatory markers were defined as Group A and all others undergoing colonoscopy as Group B.Results. Caecal intubation and TI were achieved in 988/1096 (90.15%) and 832/1096 (75.9%) cases, respectively. 764/832(91.8%) patients were included in final analysis. 81/764 (10.6%) patients had either macroscopic (34/81) or microscopic (47/81) abnormalities of terminal ileum; 20/81 had both. These were CD (28/47), tuberculosis (TB) (6/47), ileitis due to resolving infection (8/47), and drug-induced ileitis (5/47). 27/81 with macroscopically normal ileum had CD (18/27), ileitis due to resolving infection (5/27) and drug-induced ileitis (4/27) on histology. 12/764 (1.57%) patients with macroscopically normal colon had ileal CD (8/12), drug-induced ileitis (2/12), and resolving ileal infection (2/12) on histology. 47/764 (6.15%) patients had ileal pathology that influenced subsequent management. These were significantly higher in Group A (43/555 (8%)) than in Group B (4/209 (1.9%)) (P=0.0048,χ2=7.968).Conclusion. TI and biopsy improve diagnostic yield of colonoscopy in patients with RIF pain, diarrhoea, anaemia, suspected IBD, and raised inflammatory markers.

Objective. We estimated time to initiation, outpatient resource use, and costs of outpatient care during the 6 months prior to ART initiation for HIV-infected pediatric patients in Zambia.Methods. We enrolled 1,102 children who initiated ART at <15 years of age between 2006 and 2011 at 5 study sites. Of these, 832 initiated ART ≤6 months after first presenting to care at the study sites. Data on time in care and resources utilized during the 6 months prior to ART initiation were extracted from patient medical records. Costs were estimated from the provider’s perspective and are reported in 2011 USD.Results. For the patients who initiated ART ≤6 months after presenting to care, median age at presentation to care was 3.9 years; median CD4 percentage was 13%. Median time to ART initiation was 26 days. Patients made, on average, 2.38 clinic visits prior to ART initiation and received 0.81 CD4 tests, 0.74 full blood count tests, and 0.49 blood chemistry tests. The mean cost of pre-ART care was $20 per patient.Conclusions. Zambian pediatric patients initiating ART ≤6 months after presenting to care do so quickly, utilize fewer resources than mandated by national guidelines, and accrue low costs.

Background & Aims: Respiratory complications represent an important adverse event of endoscopic procedures. We screened for respiratory complications after endoscopic procedures using a questionnaire and followed-up patients suggestive of respiratory infection.Method: In this prospective observational, multicenter study performed in Outpatient practices of gastroenterology we investigated 15,690 patients by questionnaires administered 24 hours after the endoscopic procedure.Results: 832 of the 15,690 patients stated at least one respiratory symptom after the endoscopic procedure: 829 patients reported coughing (5.28%), 23 fever (0.15%) and 116 shortness of breath (SOB, 0.74%); 130 of the 832 patients showed at least two concomitant respiratory symptoms (107 coughing + SOB, 17 coughing + fever, 6 coughing + coexisting fever + SOB) and 126 patients were followed-up to assess their respiratory complaints. Twenty-nine patients (follow-up: 22.31%, whole sample: 0.18%) reported signs of clinically evident respiratory infection and 15 patients (follow-up: 11.54%; whole sample: 0.1%) received therefore antibiotic treatment. Coughing or vomiting during the endoscopic procedure resulted in a 156.12-fold increased risk of respiratory complications (95% CI: 67.44 - 361.40) and 520.87-fold increased risk of requiring antibiotic treatment (95% CI: 178.01 - 1524.05). All patients of the follow-up sample who coughed or vomited during endoscopy developed clinically evident signs of respiratory infection and required antibiotic treatment while this occurred in a significantly lower proportion of patients without these symptoms (17.1% and 5.1%, respectively).Conclusions: We demonstrated that respiratory complications following endoscopic sedation are of comparably high incidence and we identified major predictors of aspiration pneumonia which could influence future surveillance strategies after endoscopic procedures.

Abstract:
The publication presents results of the reproductive biology study of the Lissotriton lantzi (Wolterstorff, 1914) from the Abrau Peninsula (Krasnodar Territory, Russia). Ten females and ten males of this species were caught before breeding began. Animals were kept in plastic containers with water at a temperature of 11.5–23.0°C, one pair of newts in each container. Every day the reproductive behavior was recorded and laid eggs were selected. Males showed reproductive behavior (mating dance and position of spermatophore) within 1–36 days after the start of the research. Position of first eggs was observed in different females from 6 to 16 of April at a water temperature of 13.0–16.5°C. All ten females bred in April and May, only five females laid eggs in June and three females in July. The total duration of the oviposition period in different females was 41–99 days. Last eggs were found from May 26 to July 18 at a temperature of 17.5 to 23.0°C. Fertility of females in the breeding period was 67–832 eggs. The eggs of newts had a length of 3.1–4.0 mm and width of 1.9–2.8 mm. The duration of egg incubation at a temperature 14.5–20.5°C was from 13 to 24 days. Pre-larvae length after the hatching was equal to 6.5–9.8 mm. The larvae started to eat during 0–5 days after hatching. The length of larvae at the beginning of exogenous feeding was 8.2–11.3 mm. It is noted that most of the studied females (9 out of 10) had higher fertility than previously noted for this species (283–832 against 70–220 eggs).

Intracellular thioguanine nucleotides (6-TGN) are the major cytotoxic metabolites of mercaptopurine (6-MP). Red blood cell (RBC) 6-TGN concentrations were measured in a group of 120 consecutive children with lymphoblastic leukemia (ALL) to assess interpatient variability and its clinical importance. Assays were performed after at least 2 months 6-MP maintanance chemotherapy and a minimum 7 days unattenuated protocol dose of 75 mg/m2. Observed 6-TGN concentrations ranged from 126 to 832 pmol/8 x 10(8) RBCs (median, 275). There was a correlation between 6-TGN and neutropenia 14 days postassay (rs = .51; P less than .0005), and an inverse correlation between 6-TGN and the length of time uninterrupted full protocol dose was tolerated without neutropenia (rs = -.3; P less than .01). After a median follow-up of 49 months, 19 children had relapsed, of whom 17 (89%) had 6-TGN concentrations below the group median (log-rank chi 2 = 11.9; P less than .001). Multivariate analysis using Cox's proportional hazards regression showed the 6-TGN effect on disease control to be independent of diagnostic WBC count, sex, age, immunological cell type, French-American-British (FAB) type, variation in other antineoplastic therapy, and duration of remission at the time of 6-TGN assay. Children with ALL taking the same dose of 6-MP show great variability in its measurable cytotoxic effect, and this variability is apparently important in predicting treatment outcome.

Untuk mendapatkan hasil yang maksimal dalam pengelolaan tanaman, apotek hidup agar dapat tumbuh dan berkembang dengan baik, maka sebaiknya tanaman apotek hidup dapat tumbuh dengan tidak diganggu oleh tumbuhan gulma yang sering tumbuh di dekat tanaman apotek hidup. Maka tujuan penelitian adalah untuk mengidentifikasi dan mencari tahu jenis gulma apa saja yang tumbuh di lahan tanaman apotek hidup di SMP Negeri 6 Kota Ternate. Metode yang digunakan yaitu metode observasi langsung. Dari hasil penelitian yang telah dilakukan di dapatkan sebanyak 27 spesies gulma yang terdapat di lahan tanaman apotek hidup yang terdiri dari 832 indivifu dan terbagi dalam 3 kelompok gulma. Kelompok gulam berdaun lebar ditemukan dengan jumlah 691 individu (83%), sedangkan gulma rerumputan atau rumput-rumputan yaitu sebanyak 97 tumbuhan (12%) dan gulma jenis teki dengan jumlah 44 individu (5%). Secara umum bahwa gulma daun lebar merupakan gulma dominan yang tumbuh di lahan apotek hidup.

Patients with either HIV, diabetes or hypertension had the same waiting areas, the same pharmacy, were seen by the same clinical staff, had similar provision of adherence counselling and tracking if they failed to attend appointments.Primary outcome measuresRetention in care, plasma viral load.FindingsBetween 5 August 2018 and 21 May 2019, 2640 patients were screened of whom 2273 (86%) were enrolled into integrated care (832 with HIV infection, 313 with diabetes, 546 with hypertension and 582 with multiple conditions). They were followed up to 30 January 2020. Overall, 1615 (71.1%)/2273 were female and 1689 (74.5%)/2266 had been in care for 6 months or more. The proportions of people retained in care were 686/832 (82.5%, 95% CI: 79.9% to 85.1%) among those with HIV infection, 266/313 (85.0%, 95% CI: 81.1% to 89.0%) among those with diabetes, 430/546 (78.8%, 95% CI: 75.4% to 82.3%) among those with hypertension and 529/582 (90.9%, 95% CI: 88.6 to 93.3) among those with multimorbidity. Among those with HIV infection, the proportion with plasma viral load <100 copies/mL was 423(88.5%)/478.ConclusionIntegrated management of chronic diseases is a feasible strategy for the control of HIV, diabetes and hypertension in Africa and needs evaluation in a comparative study.

We used three-dimensional reconstruction 1) to determine the effect of treating rats with dexamethasone, from age 4 to 13 days, on alveolar volume (v) and number (Na) and 2) to determine if v, Na, or both change between age 14 and 60 days. At age 14 days, v, Na, and gas exchange surface area (Sa) were (2.7 +/- 0.3) x 10(4) microns3, (20.2 +/- 2.1) x 10(6), and 832 +/- 29 cm2, respectively, in diluent-treated rats; in dexamethasone-treated 14-day-old rats the same parameters were (7.5 +/- 1.1) x 10(4) microns3, (9.9 +/- 1.6) x 10(6), and 733 +/- 16 cm2, respectively. At age 60 days, v, Na, and Sa were (7.1 +/- 0.4) x 10(4) microns3, (60.8 +/- 4.1) x 10(6), and 4,495 +/- 187 cm2 in diluent-treated rats and in rats treated with dexamethasone from age 4 to 13 days, v, Na, and Sa at age 60 days were (15.9 +/- 2.4) x 10(4) microns3, (25.8 +/- 3.8) x 10(6), and 3,424 +/- 203 cm2. We conclude that treatment with dexamethasone from age 4 to 13 days (the period of normal septation) resulted in larger alveoli at age 14 and 60 days, and in diluent- and dexamethasone-treated rats the increase in Sa between age 14 and 60 days is due, at least in part, to the formation of new alveoli.

Soursop (Annona muricata L.) is climacteric fruit with a short ripening period and postharvest shelf life, leading to a rapid softening. In this study, transcriptome analysis of soursop fruits was performed to identify key gene families involved in ripening under postharvest storage conditions (Day 0, Day 3 stored at 28 ± 2 °C, Day 6 at 28 ± 2 °C, Day 3 at 15 ± 2 °C, Day 6 at 15 ± 2 °C, Day 9 at 15 ± 2 °C). The transcriptome analysis showed 224,074 transcripts assembled clustering into 95, 832 unigenes, of which 21, 494 had ORF. RNA-seq analysis showed the highest number of differentially expressed genes on Day 9 at 15 ± 2 °C with 9291 genes (4772 up-regulated and 4519 down-regulated), recording the highest logarithmic fold change in pectin-related genes. Enrichment analysis presented significantly represented GO terms and KEGG pathways associated with molecular function, metabolic process, catalytic activity, biological process terms, as well as biosynthesis of secondary metabolites, plant hormone signal, starch, and sucrose metabolism, plant–pathogen interaction, plant–hormone signal transduction, and MAPK-signaling pathways, among others. Network analysis revealed that pectinesterase genes directly regulate the loss of firmness in fruits stored at 15 ± 2 °C.

Interobserver variations between three radiologists with different backgrounds (duration of practice, and departments) were studied in connection with the diagnosing of urolithiasis. Films from 152 consecutive patients (examined with conventional abdominal films, urography/tomography) were analyzed independently. With respect to the number of patients with or without calculi there was agreement between the three observers in 129 of the 152 cases, which was taken to represent very good agreement beyond chance. The total number of stones was interpreted as 832, 460 and 570 respectively, and the median stone size as 6, 9 and 11 mm2. There was a fair interobserver agreement rate regarding whether stones were localized to the renal parenchyma or calyces, and whether stenosis/stricture or hydronephrosis was present. Tomography appeared to reduce the interobserver variability significantly. Adequate bowel cleaning, oblique projections, and lack of clinical information apparently did not influence the agreement rate. It was concluded that such interobserver variations are factors of importance in the reliability of the radiologic evaluation of urolithiasis.

Abstract Background and Purpose. Limited evidence exists regarding the characteristics of people who benefit most from constraint-induced movement therapy (CIMT). This study’s purpose was to investigate 6 potential descriptors in predicting CIMT outcomes. Subjects. The participants were a convenience sample (N=55) of people who were more than 6 months poststroke. Methods. The Wolf Motor Function Test (WMFT) and the Motor Activity Log amount scale (MALa) were used to assess outcomes for CIMT. The potential predictors (side of stroke, time since stroke, hand dominance, age, sex, and ambulatory status) were entered into a linear regression model using stepwise entry, with simultaneous entry of the dependent variables’ pretest scores as the covariate. Results. Age was the only significant predictor of the 6 potential predictors in the model and was predictive only of MALa scores. None of the independent variables showed a predictive relationship with the WMFT. Discussion and Conclusion. Although age was the only significant predictor, an equally strong finding in this study was that side of stroke, chronicity, hand dominance, sex, and ambulatory status were not found to be predictors at the follow-up session. Abstract:
The concentrations of polychlorinated biphenyls (PCBs), polychlorinated naphthalenes (PCNs), and polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) were determined in soil samples collected around an industrial park in Northwest China, to investigate the potential impacts of park emissions on the surrounding environment. The total concentration ranges of PCBs, PCNs, and PCDD/Fs in the soil samples were in 13.2–1240, 141–832, and 3.60–156 pg/g, respectively. The spatial distribution and congener patterns of PCBs, PCNs, and PCCD/Fs indicated that there might be multiple contamination sources in the study area, so source apportionments of PCBs, PCNs, and PCCD/Fs were performed by a positive matrix factorization model based on the concentrations of all target congeners together. The results revealed that these highly chlorinated congeners (CB-209, CN-75, and OCDF) might be derived from phthalocyanine pigments, the legacy of Halowax 1051 and 2,4-D products, which together contributed nearly half of the total concentration of target compounds (44.5%). In addition to highly chlorinated congeners, the local industrial thermal processes were mainly responsible for the contamination of PCBs, PCNs, and PCDD/Fs in the surrounding soil. The total carcinogenic risk of PCBs, PCNs, and PCDD/Fs in a few soil samples (0.22 × 10−6, 0.32 × 10−6, and 0.40 × 10−6) approached the threshold of potential carcinogenic risk (1.0 × 10−6). Since these pollutants can continuously accumulate in the soil, the contamination of PCBs, PCNs, and PCDD/Fs in surrounding soil deserves continuous attention.

Abstract The effects of storage for 6 months or 2 years at -20 degrees C were studied in urine samples from Type II diabetic patients by assaying albumin by immunoturbidity, N-acetylglucosaminidase (EC 3.2.1.30) by methoxynitrovinylphenol release, and creatinine by the Jaffé method. There were significant decreases (P &lt; 0.001) in albumin/creatinine ratios from 1.14 (0.63-2.98) to 0.83 (0.32-2.12) g/mol (median + interquartile ranges) after 6 months (n = 97), and from 1.64 (0.74-5.72) to 1.00 (0.37-4.54) g/mol after 2 years (n = 89). The percentage of samples with results below the detection limit of the albumin assay (2 mg/L) increased from 5% to 21% after 6 months and from 0% to 34% after 2 years. N-Acetylglucosaminidase/creatinine ratios decreased (P &lt; 0.001) from 520 (358-832) to 380 (263-695) U/mol after 6 months and from 520 (330-865) to 258 (82-462) U/mol after 2 years. The effect of storage was greater in samples with concentrations in the normal range (&lt; 2.5 g/mol for albumin/creatinine, &lt; 500 U/mol for N-acetylglucosaminidase/creatinine). Samples with albumin concentrations more than twice the normal range were still detected as abnormal after storage at -20 degrees C; e.g., 18% were &gt; 5 g/mol (albumin/creatinine) initially, with 17% &gt; 5 g/mol after 6 months of storage. We therefore recommend storage of urine samples at 4 degrees C for no longer than 7 days before assay.

Abstract In 832, the Frankish emperor Lothar I promulgated a capitulary in the royal palace of Pavia („Hlotharii capitulare Papiense“) confirming selected chapters of the capitularies of his predecessors Charlemagne and Louis the Pious. These chapters are mostly cited verbatim, but some (chs. 3, 12, 13) seem to have no equivalent in earlier capitularies and are therefore usually considered to be new provisions by Lothar himself. The article focuses particularly on ch. 13 containing provisions for the office of notary. It includes a clause that has been interpreted as a prohibition against notaries in the service of counts writing charters outside their own county. This article argues for an alternative interpretation: that the chapter aims only to ensure that transfers of property certified by notaries are always certified in the region in which the property is located. Moreover, the chapter very likely refers to a decree by Louis the Pious of 818/819 („Capitula legibus addenda“, ch. 6). It thus emerges that ch. 13 of the „Capitulare Papiense“ does in fact take up a provision by Lothar’s predecessors, and the same can be suggested for ch. 12. According to the latter chapter, unfree people are ineligible to attain their freedom based on the 30-year term of Roman law if one of their parents was unfree. This coincides largely with a known provision ascribed to Louis the Pious in the „Liber Papiensis“, which has not hitherto been connected with Lothar’s capitulary. It can thus be considered the model, adopted and slightly modified by Lothar, for ch. 12. These new findings make it highly probable that ch. 3, for which no source has yet been identified, also repeats an older provision of Charlemagne or Louis the Pious, now lost.

Sàng lọc kháng thể bất thường là xét nghiệm cần thiết đảm bảo trong an toàn truyền máu và giảm thiểu phản ứng chậm sau truyền máu.Trong nghiên cứu sàng lọc kháng thể bất thường ở bệnh nhân trước phẫu thuật tại Trung tâm tim mạch – Bệnh viện E trung ương từ tháng 7 năm 2013 đến tháng 8 năm 2014 , tổng số 1680 trường hợp (832 nam, 848 nữ), 11 trường hợp có kết quả dương tính với kháng thể bất thường chiếm tỷ lệ 0,65%. Tỷ lệ dương tính ở bệnh nhân nữ 0,83% cao hơn so với bệnh nhân nam đáng kể (0,48%). Có 176 bệnh nhân từ 0 đến 6 tháng tuổi tỷ lệ dương tính là 1,7%; sau là nhóm tuổi từ 19 đến 40 tuổi tỷ lệ dương tính là 0,97%. Tỷ lệ kháng kháng thể bất thường có liên quan đến tuổi, giới, và chẩn đoán lâm sàng. Nghiên cứu này có vai trò quan trọng của sàng lọc kháng thể bất thường cho bệnh nhân trước phẫu thuật.

This study intends to find a relation between children´s perceived restoration and their environmental orientations. In order to do that, a designed perceived restoration scale adapted from the PRCS-C II was used as well as an adapted version of the Children´s Environmental Perceptions Scale (CEPS). A total of 832 children aged between 6 and 13 participated in the study. The results show that there is a relation between the perceived restoration due to the nature present in the school playground and the environmental orientations children show. AbstractObjectiveTo assess published reports of oncological surgical success rates in patients who underwent transoral laser supraglottic surgery and robotic surgery for supraglottic cancer.MethodsA systematic review of the literature was conducted and a meta-analysis of published data was performed. PubMed, Sage, Medline and Cochrane data sources were investigated. Overall survival rates, disease-specific survival rates, additional treatments and recurrence rates were investigated to determine the success of the surgical procedures.ResultsThe meta-analysis included 24 studies; 1617 studies were excluded. There were no statistically significant differences between the transoral laser supraglottic surgery and transoral robotic supraglottic surgery groups in terms of overall survival (77.0 per cent and 82.4 per cent respectively) and disease-specific survival (75.8 per cent and 87.0 per cent respectively). There was recurrence in 164 out of 832 patients (19.7 per cent) in the transoral laser supraglottic surgery group and in only 6 out of 66 patients (9 per cent) in the transoral robotic supraglottic surgery group.ConclusionTransoral laser surgery and robotic surgery appear to have comparable and acceptable oncological success rates.

Evaluation of success rate of manual DSEK in terms of central corneal thickness and BSCVA in patients of PBK. : Prospective, non-comparative, interventional study of 18 patients of PBK, who underwent manual DSEK between June 2021 to May 2022 with minimum six months to maximum 15 months post-operative follow up. DSEK lenticule was prepared by manual dissection. Push in technique with 26 G needle was used for graft insertion. Preoperative and post-operative CCT and BSCVA were measured and compared. : Preoperative central pachymetry in patients (n=18) was (832±162) μm which significantly reduced in the post operative period. The mean thickness of lenticule on day 1 post op was 203±93.5μm which became 156±76 μm (p =.0436) at 6 months and remained almost same till last follow up. Pre operative BSCVA ranged from HM to 2/60 in these 18 patients which was improved in 16/18 eyes post DSEK. Eighty eight percent (15/17) patients had regained BSCVA of 6/60 to 6/12. In early post operative period, complications noted were, partial detachment of lenticule and pupillary block glaucoma which were managed accordingly.: Manual DSEK is feasible, low-cost widely accessible alternative to DSAEK/DMEK for patients of PBK with good visual outcome in resource limited settings.: Manual DSEK has short learning curve and it’s training should be imparted to corneal surgeons for better outcome in PBK and other causes of endothelial decompensation.

Purpose Fenretinide induces apoptosis in malignant gliomas in vitro. This two-stage phase II trial was conducted to determine the efficacy of fenretinide in adults with recurrent malignant gliomas. Patients and Methods Twenty-two patients with anaplastic gliomas (AG) and 23 patients with glioblastoma (GBM) whose tumors had recurred after radiotherapy and no more than two chemotherapy regimens were enrolled. Fenretinide was given orally on days 1 to 7 and 22 to 28 in 6-week cycles in doses of 600 or 900 mg/m2 bid. Results Six of 21 (29%) patients in the AG arm and two of 23 (9%) patients in the GBM arm had stable disease at 6 months. One patient with AG treated at 900 mg/m2 bid dosage had a partial radiologic response. Median progression-free survival (PFS) was 6 weeks for the AG arm and 6 weeks for the GBM arm. PFS at 6 months was 10% for the AG arm and 0% for the GBM arm. Grade 1 or 2 fatigue, dryness of skin, anemia, and hypoalbuminemia were the most frequent toxicities reported. The trial was closed after the first stage because of the inadequate activity at the fenretinide doses used. The first-administration mean plasma Cmax for fenretinide was 832 ± 360 ng/mL at the 600 mg/m2 bid dosage and 1,213 ± 261 ng/mL at the 900 mg/m2 bid dosage. Conclusion Fenretinide was inactive against recurrent malignant gliomas at the dosage used in this trial. However, additional studies using higher doses of the agent are warranted based on the tolerability of the agent and the potential for activity of a higher fenretinide dosage, as suggested in this trial.

Purpose:To investigate whether anthropometric profiles and fitness measures vary according to birth-date distribution in the German national youth soccer teams and to analyze whether there is a difference in the chance of becoming a professional soccer player depending on birth quarter (BQ).Methods:First, 554 players were divided into 6 age groups (U16–U21), each subdivided into 4 BQs. Every player performed at least one 30-m sprint, a countermovement jump, and an incremental test to determine individual anaerobic threshold. For players performing more than 1 test within a team, the best 1 was included. Since some players were part of several different teams, a total of 832 data sets from 495 individual soccer players, all born from 1987 to 1995, divided into 6 age categories (U16–U21) were included.Results:Overall, more players were born in BQ1 than in all other BQs (P < .05). No significant difference between BQs could be observed in any anthropometric or performance characteristics (P > .18). Players born in BQ4 were more likely to become professional than those born in BQ1 (odds ratio 3.04, confidence limits 1.53–6.06).Conclusion:A relative age effect exists in elite German youth soccer, but it is not explained by an advantage in anthropometric or performance-related parameters. Younger players selected into national teams have a greater chance to become professionals later in their career.

Parkinson's disease (PD) patients may experience fluctuations in executive performance after oral levodopa (LD). Their relationship with the pharmacokinetic profile of LD and with distinct cognitive processes associated with frontal-basal ganglia circuits is not well understood. In this randomized, double-blind, crossover study we plotted acute cognitive changes in 14 PD patients challenged with faster (immediate-release, IR) versus slower (controlled-release, CR) increases in LD plasma concentrations. We monitored motor status, LD plasma levels, and performance on four tasks of executive function (Wisconsin Card Sorting Test—WCST, Sternberg test, Stroop and Tower of Hanoi), 1 hr before and over +6 hr after IR and CR-LD dose. Analysis of variance demonstrated significant but divergent changes in the Sternberg (6-digit but not 2- and 4-digit) test: improvement after CR-LD and worsening after IR-LD. Marginal improvement (p = .085) was observed with CR-LD in the WCST, while no significant differences were seen for the Stroop or Tower of Hanoi tests. Executive-related performance after LD challenge may differ depending on the LD time-to-peak plasma concentration and specific task demands. A slower rise in LD levels appears to have a more favorable impact on more difficult working memory tests. These results require replication to determine their generalization. Abstract Deletions of chromosome 6q have been reported in a variety of hematological malignancies. Regarding their biological and prognostic impact, however, the data available are not conclusive. Therefore the current study focused on 60 pediatric and adolescent patients all diagnosed with T-cell lymphoblastic lymphoma (T-LBL), a clearly defined histological sub-entity of NHL, and all treated uniformly according to the protocol NHL-BFM 95. Deletions of chromosome 6 were analyzed by loss of heterozygosity (LOH) analysis of 25 microsatellite markers located on chromosome 6q14-q24 and the prognostic impact of LOH on 6q was evaluated after combing the results with clinical data of the patients. Selection criteria for this study were 1) treatment according to multicentre protocol NHL-BFM 95, 2) T-cell immunophenotype of LBL, 3) germ line control and tumor DNA available and 4) three or more informative markers for the analysis of LOH. High-molecular weight germ line DNA was extracted from blood and bone marrow smears or frozen cells. Tumor DNA was isolated from tumor touch imprints or cytospin preparations of malignant effusions or frozen tumor cells. Paired normal and tumor DNA samples from each patient were amplified by PCR, after which fragment length analysis was performed on an ABI PRISM 3100 Genetic Analyzer. From 04/95 until 03/03 a total of 186 patients were diagnosed with T-LBL and treated according to the protocol NHL-BFM 95. The probability of event free survival (pEFS) for these 186 patients was 79±3% (median follow-up of 4.5 (0.5–9.1) years). 60 of the 186 T-LBL patients were evaluable for LOH analysis, including 19 out of the total 25 of T-LBL patients who suffered from tumor failure. Fragment length analysis comparing germ line DNA and tumor DNA was performed for all 25 microsatellite markers per patient resulting in a total of 832 successfully analyzed markers for the whole group of 60 patients. The status of these 832 results was LOH in 10%, retention of heterozygosity in 55%, homozygosity in germ line control and tumor DNA in 25% and microsatellite instability in 9% of results respectively. In 19 out of the 60 evaluable patients (32%) LOH of at least one microsatellite marker was detected. LOH was observed in 13 out of the 19 evaluable patients with tumor failure compared to 6 out of the 41 patients without tumor failure. The probability of disease free survival (pDFS) (5y) for the 19 patients with LOH was 24±11% compared to 85±6% for the 41 patients without LOH (p &lt;0.0001). A critical region of deletion could be assigned to microsatellite markers D6S1284 and D6S1716. pDFS (5y) was 0% for patients with LOH involving these markers (n=7) compared to 82±7% for patients without LOH at these markers (n=31) (p &lt;0.0001). We conclude that LOH on chromosome 6q14-q24, especially of markers D6S1284 and D6S1716, confers poor outcome on children and adolescents with T-LBL.
