Abstract:
Die quenching of an AA2024 aluminum alloy billet was carried out using a servo press with ram motion control of WC-20mass%Co dies directly after solution heat treatment (SHT). The cylindrical billets were heated in an electric furnace and transferred to the press, then subjected to uniaxial compression with a reduction in height (Δh/h0) of 2%, 5% or 10% after SHT at 773 K or 823 K. The die quenching was successfully carried out without the precipitation hardening only on the billet after SHT at 823 K by sandwiching for 8 s. The subsequent artificial aging behavior at 463 K in an oil bath was investigated. It is found that the precipitation kinetics is accelerated in the die-quenched billets after SHT at 823 K. The peak hardness of the billet processed with 5% after SHT at 823 K is as high as that of the water-quenched billet, HV 149.0.

e15556 Background: Chemotherapy based on DNA-damaging anti-cancer drugs remains the most predominant treatment for advanced gastric cancer. The predictors for their sensitivity are under development. Our data showed that sperm-associated antigen 5 (Spag5) contributed to cancer cell proliferation and decreased DNA-damaging drug sensitivity in vivo and vitro. Methods: Spag5 mRNA expression were compared in The Cancer Genome Atlas (TCGA) and GEO datasets (GSE56807, GSE63089, GSE65801), while protein level were detected in 52 patients diagnosed with gastric cancer in Nanfang Hospital by immunohistochemistry. BGC-823 and MGC-803 were transfected with spag5 and control siRNA and then subjected to MTT and EDU assay to verify proliferation rate. Cell viability and apoptosis were tested in spag5-silenced BGC-823 and MGC-803 treated with oxaliplatin, epirubicin and 5-fluorouracil independently. DNA-damaged were detected by comet assay and yH2ax level with western blot. Treatment of oxaliplatin was given intraperitoneally to nude mice carrying xerographs of Shspag5 or control BGC-823 cells every three days. Results: Spag5 mRNA level was significantly upregulated in cancer tissues compared with paired normal tissues in all of the datasets. Immunohistochemistry shows higher protein level of spag5 in paired cancer tissue. Knockdown of Spag5 BGC-823 and MGC-803 showed a significantly decreased proliferation in both MTT and EDU assay. Spag5 silencing led to increase vulnerability to oxaliplatin, epirubicin and 5-fluorouracil, thus less cell viability and more apoptosis. Spag5 knockdown BGC-823 and MGC-803 subjected to DNA-damaging drugs showed upregulated yH2ax level and increased DNA damage in comet assay. Tumor growth rate were significantly reduced in shspag5 group and treatment of oxaliplatin led to relatively higher inhibition of tumor growth. Conclusions: Spag5 serves as a candidate to predict proliferation and chemosensitivity to DNA-damaging chemotherapy.

Five new meroterpenoids, zizhines P-S and U (1−4,7), together with two known meroterpenoids (5 and 6) were isolated from Ganoderma sinensis. Their structures including absolute configurations were assigned by using spectroscopic, computational, and chemical methods. Racemics zizhines P and Q were purified by HPLC on chiral phase. Biological evaluation found that 4, 5 and 6 are cytotoxic toward human cancer cells (A549, BGC-823, Kyse30) with IC50 values in the range of 63.43–80.83 μM towards A549, 59.2 ± 2.73 μM and 64.25 ± 0.37 μM towards BGC-823, 76.28 ± 1.93 μM and 85.42 ± 2.82 μM towards Kyse30.

Abstract:
The chlorination reaction of Y2O3 with ZrCl4 in LiCl-KCl eutectic melt was investigated by X-ray absorption fine structure (XAFS) technique. The chlorination reaction was observed between 773 K and 823 K as the 1st peak shift of the Fourier transform magnitude function |FT(k3χ(k))|. The peak corresponding to the nearest Y3+-Cl− correlation was observed in the XAFS analysis at 823 K as the result of the chlorination. It was confirmed that the mixture melts after the reaction is almost equivalent to a molten 5% YCl3-(LiCl-KCl eutectic) mixture.

Gastric carcinoma is a frequently detected malignancy worldwide, while its mainstream drugs usually result in some adverse reactions, including immunosuppression. λ-carrageenan oligosaccharides (COS) have attracted increasing attention as potential anticancer agents due to their ability to enhance immune function. Our current work assessed the antitumor mechanism of λ-COS using BGC-823 cells. Our findings indicated that λ-COS alone did not have a significant impact on BGC-823 cells in vitro; however, it was effective in inhibiting tumor growth in vivo. When THP-1 cells were pre-incubated with λ-COS and used to condition the medium, BGC-823 cells in vitro displayed a concentration-dependent induction of cell apoptosis, nuclear damage, and the collapse of mitochondrial transmembrane potential. These findings suggested that the antineoplastic effect of λ-COS was primarily due to its immunoenhancement property. Treatment with λ-COS was found to significantly enhance the phagocytic capability of macrophages, increase the secretion of TNF-α and IFN-γ, and improve the indexes of spleen and thymus in BALB/c mice. In addition, λ-COS was found to inhibit the growth of BGC-823-derived tumors in vitro by activating the Par-4 signaling pathway, which may be stimulated by the combination of TNF-α and IFN-γ. When used in combination with 5-FU, λ-COS demonstrated enhanced anti-gastric carcinoma activity and improved the immunosuppression induced by 5-FU alone. These findings suggested that λ-COS could be used as an immune-modulating agent for chemotherapy.

Four new sesquiterpenoids, 1α-hydroxymethyl-3β-hydroxy-7,8-dihydro-ionol (1), 1α-hydroxymethyl- 3β-hydroxy-7,8-dihydro-ionol-9-O-β-D-glucopyranoside (2), (1α,5β,7β)-3,10(14)-guaiadien- 11,12-diol (3), and (6S)-13-O-β-D-glucopyranosyl-abscisic acid (4), together with 10 known sesquiterpenoids and 5 diterpenes were isolated from the branches and leaves of Chamaecyparis obtusa var. breviramea f. crippsii. Their structures were mainly determined on the basis of MS, IR, 1D and 2D NMR spectral evidence. Compound 13-epi-toruolsol (17) showed cytotoxicities against BGC-823 and Hela cancer cell lines with IC50 values of 23.0 and 49:9 μM, and compound 3-epitriptobenzene B (19) showed cytotoxicities against BGC-823, Hela and A549 cancer cell lines with IC50 values of 19.1, 30.3 and 24:5 μM, respectively

Abstract:
In the present paper, on the basis of molecular hybridization, a series of 4,6-dihydrazone pyrimidine derivatives containing the pyridine moiety were synthesized, structurally characterized, and evaluated in vitro for their antitumor activity. According to the results, all the tested compounds demonstrated broad-spectrum antitumor activity against selected tumor cell lines (MCF-7, BGC-823, A549, and BEL-7402) and no obvious toxicity toward normal cells HL-7702. In particular, compounds 10a and 10f were found to be the most promising antitumor agents among the tested compounds against BGC-823 cells (IC50 = 9.00 μM and 7.89 μM) and BEL-7402 cells (IC50 = 6.70 μM and 7.66 μM), respectively. Compounds 10a and 10f exhibited higher potency against BGC-823 and BEL-7402 than the positive control 5-FU (IC50 = 15.18 μM and 15.81 μM). Further mechanism investigations demonstrated that compounds 10a and 10f could significantly increase the level of cellular ROS and induce early apoptosis of BGC-823 cells in a dose-dependent manner. Moreover, the DNA binding results from UV/Vis, CD spectroscopy, and molecular docking studies indicated that 10a and 10f bind with DNA via groove binding and partial intercalation. These results demonstrated that 10a and 10f may serve as novel lead compounds for the discovery of more dihydrazone pyrimidine derivatives with improved antitumor potency and selectivity.

Commercial purity aluminum powder of irregular shape and ligamental morphology having average particle size of 75 µm was blended with medium carbon steel short fibers having average diameter of 100 µm and maximum length up to 1000 µm. The green compacts of pure aluminum and reinforced compositions were hot-pressed in 10−3 torr vacuum, at 723 K, 773 K, and 823 K for 5, 10, and 15 min durations under 50 MPa axial stress on Gleeble 3800 simulator. Microstructures of the sintered composites have been studied by scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDAX). The sintered compacts were characterized for densification behavior, hardness, and growth of FexAly reaction interface. Positive densification parameter was achieved for up to 10 wt.% reinforcement fraction. The maximum hardness of 51 Hv was achieved for 10 wt.% at 823 K for 15 min sintering time. The reaction interface was analysed by scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDAX). The reaction interface was found to be composed of Fe3Al, FeAl2, Fe2Al, Fe3Al, Fe4Al, Fe2Al5, and FeAl intermetallics. Growth of the reaction interface was diffusion-controlled which followed a nearly parabolic law with a rate constant of 1.41 × 10−12 m2 s−1 at 823 K.

Previous studies on the conversion of olefins to aromatics with bifunctional Ga- or Zn-ZSM-5 catalysts have concluded that benzene, toluene, and xylenes (BTX) yields are significantly higher than for ZSM-5 alone. These results were attributed to the higher aromatic dehydrogenation rate of Ga or Zn. In this study, a highly active, bifunctional PtZn/SiO<sub>2</sub> (1.3 wt% Pt, 2.6 wt% Zn) with H-ZSM-5 (Si/Al = 40) catalyst is investigated for propene aromatization at 723 K and 823 K. At low to moderate propene conversions, in addition to BTX, small alkanes and olefins are produced. Many of these may also be converted to aromatics at higher propene conversion while others are not, for example, methane, ethane and propane. When compared at equivalent space velocity or propene conversion, the bifunctional catalyst has a much higher selectivity to aromatics than ZSM-5; however, when compared at equivalent conversion of all reactive intermediates, the bifunctional catalyst exhibits very similar BTX selectivity. At 723 K, for ZSM-5 the primary non-reactive by-products are propane and butane; while, for the bifunctional catalyst the major non-reactive product is propane. At 823 K, both ZSM-5 and the bifunctional catalyst convert propane and butane to aromatics increasing the aromatic yields, and the by-products are methane and ethane.

Introduction: To investigate antitumor activity and mechanism of flavonoids from Alpinia officinarum Hance against gastric cancer. Methods: Transplanted mouse fore-stomach carcinoma (MFC) tumor mice were divided into six groups: control group, model group, low dose (20 mg/kg), middle dose (40 mg/kg), and high dose (80 mg/kg) groups of TFAO and 5-Fu group. Mice were treated with TFAO or 5-Fu for 14 days, except those of control and model group. Tumor inhibitory rate, spleen, and thymus index were calculated. Contents of proliferating cell nuclear antigen, MMP-9, vascular endothelial growth factor, IL-1β, IL-6, and IL-17 in serum were detected. Effect of galangin on BGC-823 cell growth was detected. Cell apoptosis and cell cycle distribution were measured. Enzyme activity of Caspase-3, Caspase-8, and Caspase-9 was detected. Western blot was used to detect STAT3, Bcl-2, Bax, Caspase-3, Caspase-8, Caspase-9, CyclinB1, and CyclinD1 protein expression in BGC-823 cell. Results: Compared with model group, tumor weight of mice decreased significantly ( p < .01) in 5-Fu group, low dose, middle dose, and high dose group of TFAO; thymus index of mice decreased significantly ( p < .05) in 5-Fu group; and spleen index decreased significantly ( p < .05) in low dose and middle dose groups of TFAO. Compared with model group, levels of PCNA, MMP-9, IL-1β, and IL-6 in serum of mice decreased obviously ( p < .01) in all administration groups; levels of VEGF in serum of mice decreased obviously ( p < .01) in low dose and high dose group of TFAO and 5-Fu group; and levels of IL-17 in serum of mice decreased significantly ( p < .01) in low-dose and middle-dose groups of TFAO and 5-Fu group. Galangin could inhibit BGC-823 cell growth; accelerate apoptosis; block cell cycle; increase cell Caspase-3, Caspase-8, and Caspase-9 enzyme activity; upregulate expression of Caspase-3, Caspase-8, Caspase-9, and Bax; and downregulate expression of STAT3, CyclinB1, CyclinD1, and Bcl-2 protein. Conclusion: Flavonoids from A. officinarum showed antitumor activity in gastric cancer. Mechanisms may be associated with inhibition of tumor angiogenesis, tumor cell proliferation, and cancer-associated inflammation.

In this paper, the precipitation of carbide and wear loss of high-carbon 8 mass% Cr tool steel at two tempering conditions (i.e., 773–803 K and 823–853 K) were studied by INCA Steel, EPMA-1720H, XRD, and ML-10 tester. The results show that the particles of test steels include the carbides (Cr7C3 and Cr23C6) and carbides nucleated on Al2O3. When carbides are of the same size, the number of carbides in test steel at a tempering temperature of 773–803 K is greater than that at a tempering temperature of 823–853 K, especially when the size of carbides is less than 5 μm. Compared with the test steel tempered at 823–853 K, the distance between adjacent actual particles reduced by 80.6 μm and the maximum amount of reduction was 9.4% for single wear loss at the tempering temperature of 773–803 K. It can be concluded from thermodynamics results that Al2O3 inclusions began to precipitate in liquid, and the precipitation of carbides was at the solid–liquid region. Al2O3 can be used as the nucleation interface of carbide, thus promoting the formation of carbides. During the cooling of molten steel, a lower temperature can increase the difference of actual solubility product bigger than equilibrium solubility product, thus promoting the carbide formation.

Chalcones1~8and 5-deoxyflavonoids9~22were synthesized in good yields by aldol condensation, Algar-Flynn-Oyamada reaction, glycosidation, and deacetylation reaction, respectively, starting from 2-acetyl phenols substituted by methoxy or methoxymethoxy group and appropriately benzaldehydes substituted by methoxy, methoxymethoxy group, or chlorine. Among them,13and17~22are new compounds. The cytotoxicity bioassays of these chalcones and 5-deoxyflavonoids were screened using the sulforhodamine B (SRB) protein staining method, and the results showed that compounds2, 4, 5, 6, 10, 15, and19exhibited moderate cytotoxicity against the cancer cell line of MDA-MB-231, U251, BGC-823, and B16 in comparison with control drugs (HCPT, Vincristine, and Taxol).

The herb medicine formula “Chong Lou Fu Fang” (CLFF) has efficacy in inhibiting the proliferation of human gastric cancerin vitroandin vivo. To explore the potentially useful combination of CLFF with chemotherapeutic agents commonly used in gastric cancer therapy, we assess the interaction between CLFF and these chemotherapeutic agents in both SGC-7901 cell lines and BGC-823 cell lines using a median effect analysis and apoptosis analysis, and we also investigate the influence of CLFF on chemotherapeutic agent-associated gene expression. The synergistic analysis indicated that CLFF had a synergistic effect on the cytotoxicity of 5-fluorouracil (5-FU) in a relative broad dose inhibition range (20–95% fraction affected in SGC-7901cell lines and 5–65% fraction affected in BGC-823 cell lines), while the synergistic interaction between CLFF and oxaliplatin or docetaxel only existed in a low dose inhibition range (≤50% fraction affected in both cell lines). Combination of CLFF and chemotherapeutic agents could also induce apoptosis in a synergistic manner. After 24 h, CLFF alone or CLFF combination with chemotherapeutic agents could significantly suppress the levels of expression of chemotherapeutic agent resistance related genes in gastric cancer cells. Our findings indicate that there are useful synergistic interactions between CLFF and chemotherapeutic agents in gastric cancer cells, and the possible mechanisms might be partially due to the down-regulation of chemotherapeutic agent resistance related genes and the synergistic apoptotic effect.

Indonesian society is a communal society, where there is a lack of awareness in protecting traditional culture. The emergence of various problems related to the Protection of Intellectual Property in Traditional Knowledge and Cultural Expression has made the public begin to realize the importance of Intellectual Property Law itself. In this study, the researcher uses a normative juridical law research method, namely reviewing and studying literature, legislation related to Traditional Knowledge and Cultural Expression by conducting a Law and Literature approach by analyzing secondary data through literature such as books, journals, articles and laws and regulations related to Traditional Knowledge. As a result, Decision No. 823/K/Pid.Sus/2009 showed that judges lacked understanding of traditional knowledge, which at that time had not been specifically regulated and only through the Patent Law, Trademark Law, and Copyright Law. Public awareness of Intellectual Property Rights is also low, leading to legal uncertainty. Law No. 5 of 2017 provides protection for traditional cultural knowledge and expressions, such as arts and customs, with the state obliged to carry out inventory, maintenance, and preservation. This law also emphasizes the importance of public legal awareness in its enforcement.

Abstract Introduction : Since the first description of hematopoietic stem and progenitor cell (HSPC) mobilization over forty years ago, it has become the standard of care for both autologous and allogeneic transplantation. A five-day course of G-CSF represents the most commonly used mobilization regimen today. The CXCR4 inhibitor, plerixafor, is a more rapid but weak mobilizer when used as a single agent, thus emphasizing the need for faster acting agents with more predictable mobilization responses and fewer side effects. Methods : Given the critical role of VLA4/VCAM1 signaling for migration and retention of HSPC, we were seeking to identify small molecule antagonists of VLA4 with improved potency and bioavailability. Relative to previously described comparators Bio5192 (▢4▢1-specific) and firategrast (▢4▢1 and ▢4▢7 dual-specific), our lead candidate, CWHM-823, exhibited increased aqueous solubility and ~10-100 fold better activity in blocking VLA4 and mobilizing HSPC in mice. CWHM-823 pharmacokinetics and mobilization were assessed in BALB/c and DBA/2 mice at different doses (3 to 15 mg/kg) and time points (15 to 240 min) when administered alone or in combination with the truncated isoform of the CXCR2 agonist Gro-beta (tGro-β, 2.5 mg/kg, generously provided by GlaxoSmithKline). HSPC mobilization was monitored using flow cytometry and clonogenic in vitro assays. "True" stem cells were measured in a serial competitive transplantation assay. The combination of tGro-β and VLA4 antagonist was further tested in diabetic mice in comparison to G-CSF (9 x 100μg/kg, q12h). RNA profiling of flow-sorted HSPC was performed via microarray analysis. Results : The combination of tGro-β with each VLA4 antagonist resulted in a dramatic synergistic increase in circulating HSPC numbers when compared to steady state (50-70-fold) or treatment with single agents (3-10 fold) including tGroβ. Mobilization with tGro-β + CWHM-823 was rapid, peaking at 15-30 minutes after injection. In a model of streptozotocin-induced diabetes, the mobilopathy (reduction in stem cell mobilization compared to wild type mice) was considerably less pronounced with the combination tGro-β + CWHM-823 (~1.5-fold lower CFU mobilization in diabetic mice) versus the 5-day course of G-CSF (~3-fold reduction). Despite the superior progenitor cell mobilization achieved with G-CSF (~2-fold more CFU and LSK/ml), the concentration of serially repopulating units (RU) was equally high in the tGro-β + CWHM-823 and G-CSF mobilized grafts suggesting a higher HSC frequency (1 RU out of 200 vs. 1 RU out of 400 LSK/CFU) in the tGro-β + CWHM-823 mobilized grafts (Figure 1). RNA profiling demonstrated close similarity between the expression profile of tGro-β + CWHM-823 mobilized, BM resident, and G-CSF mobilized LSK, with less than 0.5% of genes found to be significantly up- or downregulated. CXCR2 chemokine receptor stimulation was critical for the observed synergistic response, as pretreatment ("priming") or simultaneous treatment with tGro-β resulted in subsequent enhanced mobilization using VLA4 inhibitors, whereas reversed administration (VLA4 antagonist followed by tGro-β) had no effect on potency of either agent. Lack of surface CXCR2 expression on HSPC suggested that a rapidly acting effector molecule released from tGro-β-stimulated mature myeloid cells may subsequently influence VLA4-mediated HSPC adhesion/retention. Consistent with this theory, we observed increased protease MMP-9 in plasma within minutes after treatment with tGro-β + CWHM-823. Conclusions: We describe a novel strategy for rapid, reliable, and potent mobilization of HSPC in mice using a combination of VLA4 blockade (via novel and potent ▢4▢1 inhibitors) and CXCR2 activation (via tGro-β). The combination of tGro-β + VLA4 inhibitors or tGro-β followed by VLA4 inhibitors results in synergistic and rapid HSPC mobilization with quantity and quality of repopulating units similar to optimal mobilization with G-CSF. These data suggest further development of tGro-β + VLA4 inhibitor combinations for clinical testing is warranted. Figure 1. Abstract Apart from the importance of accurate meaning transference, the key to English translation of Traditional Chinese Medicine (TCM) terms lies in the proper translation of term forms, in particular, of long term structures and length. This article reports on an empirical study of the English translation of long TCM terms by the following procedures: (1) collecting 1220 TCM terms and their English translations from dictionaries, journals and official websites related to TCM terminology translation; (2) segmenting and POS-tagging with ICTCLAS to obtain 823 long TCM terms with 3 or more Chinese words; (3) selecting 150 out of the 823 long TCM terms through random sampling; (4) POS-tagging the 150 English translations with CLAWS5; (5) basing on the parallel corpus and systematically discussing the structures, term length change, translation techniques and translation regularities generalized from the English translation of long TCM terms. The result shows nominalization, shift of some pre-modifiers into post-modifiers, and amplification of a predicate in the 9 kinds of structural features, and some translation techniques like literal translation, paraphrase, adaptation, amplification and simplification employed in the English translation of long TCM terms.

Then-hexane cracking performance over the as-prepared MeAPOs was estimated under different reaction conditions. MeAPO-5 molecular sieves exhibited higher activity for the cracking conversion in the temperature range of 723~823 K than MeAPO-11. The conversion level ofn-hexane over MeAPOs with the same topology had the following order: MgAPOs > ZnAPOs > CuAPOs. The cracking results showed that the conversion level ofn-hexane increased with increasing reaction temperatures and contact time, and that the first order is determined in then-hexane concentration.

Background: The anemic children tend to have average or below average academic performance due to effect of anemia on cognitive and behavioral aspect of the child. The objective of the study is to study prevalence and types of anemia among children at a tertiary care hospital.Methods: A total of 6000 patients attended the outpatient department of the SVS hospital, Mahabubnagar during the study period. Out of them 1237 patients were pediatric patients who attended the pediatric outpatient department. Among these 1237 children, 823 children were found anemic. Among these 823 anemic children, 100 were randomly included in the present study for further investigations. Thus, final sample size of the present study was randomly selected 100 anemic children.Results: The incidence of anemia was found to be very high i.e. 66.5%. It was found that the prevalence of anemia was 60.9% among females compared to only 39.1% among male children. The prevalence of anemia was significantly higher in females compared to males. In both males and females, the iron deficiency anemia was the most common type of anemia found in the present study. Maximum prevalence of iron deficiency anemia was seen in 1-5 years of age in females. Maximum prevalence of iron deficiency anemia was seen in 1-5 years of age in males. The most common grade of anemia was moderate grade followed by mild.Conclusions: Iron deficiency anemia is more prevalent in female children. In males also, it is not found to be very low. Its prevalence in males is also noticeable.

The electrochemical properties of as-cast Zr56Cu19Ni11Al9Nb5 metallic glass and samples annealed at different temperatures were investigated using potentiodynamic polarization tests and electrochemical impedance spectroscopy (EIS) in phosphate buffer saline (PBS) solution. It was shown that passivation occurred for the as-cast sample and the samples annealed at 623–823 K, indicating good corrosion resistance. At higher annealing temperature, the corrosion resistance first increased, and then decreased. The sample annealed at 823 K exhibited the best corrosion resistance, with high spontaneous corrosion potential Ecorr at −0.045 VSCE, small corrosion current density icorr at 1.549 × 10−5 A·cm−2, high pitting potential Epit at 0.165 VSCE, the largest arc radius, and the largest sum of Rf and Rct at 5909 Ω·cm2. For the sample annealed at 923 K, passivation did not occur, with low Ecorr at −0.075 VSCE, large icorr at 1.879 × 10−5 A·cm−2, the smallest arc radius, and the smallest sum of Rf and Rct at 2173 Ω·cm2, which suggested the worst corrosion resistance. Proper annealing temperature led to improved corrosion resistance due to structural relaxation and better stability of the passivation film, however, if the annealing temperature was too high, the corrosion resistance deteriorated due to the chemical inhomogeneity between the crystals and the amorphous matrix. Optical microscopy and scanning electron microscopy (SEM) examinations indicated that localized corrosion occurred. Results of energy dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS) illustrated that the main corrosion products were ZrO2, CuO, Cu2O, Ni(OH)2, Al2O3, and Nb2O5.

The objective of this study is to explore the effects of microRNA-33a-5p (miR-33a-5p)-ras-related protein Rap-2a (RAP2A) on biological functions of gastric cancer (GC) and to find the potential functional mechanism. Methods. We measured the miR-33a-5p expression in 30 GC tissues and cellular level and 30 adjacent normal tissues as control. Besides, the expression of miR-33a-5p was checked at cell level as well. To screen the possible targets of miR-33a-5p, prediction software was used and gene RAP2A attracted our attention. Through a series of experiments including real-time polymerase chain reaction (qRT-PCR), luciferase assay, and western blotting (WB), we verified RAP2A as a potential target of miR-33a-5p. The impacts of miR-33a-5p and RAP2A on biological functions of GC cell lines (BGC-823 and MGC-803) were analyzed by subsequent experiments. Cell invasion was tested by invasion assays. Cell proliferation was measured by cell counting kit-8 (CCK-8) assay. Cell clone was measured by clone formation assays. Finally, the expression of RAP2A protein was analyzed by WB assay. Results. We found miR-33a-5p was expressed lowly in GC tissues and cells. Overexpression of miR-33a-5p in BGC-823 and MGC-803 cells greatly inhibited the cell invasion and colony number. Furthermore, compared to sh-control (shControl), RAP2A knockdown (sh-RAP2A/shRAP2A) raised the sensitivity of GC cells to 5-FU significantly, characterized as reducing cell apoptosis. Conclusions. The expression of miR-33a-5p was lower in GC cell lines and tissues obviously, indicating that miR-33a-5p served as the antitumor gene in GC. The expression of RAP2A regulated negatively the sensitivity of GC cells to 5-FU. According to our in vitro experiments, miR-33a-5p/RAP2A was likely to become a new therapeutic target for GC.

Comparative studies of the effect of hydrogenation on creep regularities of the titanium alloy of the Ti–Al–V system in the fine-grained (dav ≈ 7 μm) and ultrafine-grained (dav ≈ 0.3 μm) states are performed for the creep rates in the interval 10-7–10–5 s-1 at temperatures in the range 723–823 K. Hydrogenation of the alloy to 0.23 % is found to lead to a decrease in the steady creep rate and increase in the time to failure with simultaneous decrease in the value of deformation to failure. Possible reasons for the violation of the creep power law for the hydrogenated alloy are discussed.

1. Introduction Ti has superior properties, such as high specific strength, corrosion resistance, biocompatibility, and is used for many fields such as airplane, chemical plant, consumer products. However, its high manufacturing cost and poor processability have hindered its further spread. Thus, a number of smelting processes and processing methods have been investigated to reduce the cost and spread the use of Ti. Among them, plating Ti metal on inexpensive substrates is one of the methods that can utilize the surface properties of Ti. Electrodeposition is a promising plating method from the viewpoints of the cost and the flexibility in shape of substrate. Therefore, electrodeposition of Ti metal in high-temperature molten salts has been investigated for a long time [1–3]. Our group has reported the electrodeposition of Ti films with compact and smooth surfaces in molten KF–KCl and LiF–LiCl containing Ti(III) ions at 923 K [4–7]. We also reported that the Ti films obtained in LiF–LiCl at 823 K had smoother surfaces [8]. In the present study, we investigated the effect of temperature on the grain growth and smoothness of Ti films electrodeposited in LiF–LiCl eutectic melt at 823–973 K. 2. Experimental The experiments were carried out in eutectic LiF–LiCl melt (30:70 mol%, melting point 774 K) at 823–973 K. The melt was placed in a nickel crucible and the experiments were conducted in an airtight Kanthal container in an argon glove box. As Ti(III) ion sources, Li2TiF6 (2.00 mol%) and Ti sponge (1.33 mol%) were added to the melt. The added amounts of Ti sponge were twice the amounts necessary to generate Ti(III) ions by comproportionation reaction between Ti(IV) ions and Ti(0) according to Eq. 1. Ti(0) + 3Ti(IV) → 4Ti(III) (1) Ni plates were used as working electrodes, and Ti rods was used as counter and reference electrodes. The samples prepared by galvanostatic electrolysis were analyzed by XRD and SEM/EDX after washing with distilled water to remove adhered salts. 3. Results and Discussion Fig. 1 shows the optical and surface SEM images of the samples obtained by galvanostatic electrolysis at 823, 873, 923, and 973 K. The cathodic current density was 50 mA cm−2 and electrolysis time was 20 min. All the samples have a metallic luster and are confirmed to be Ti metal by XRD analysis. However, the brightness differs between samples, with the Ti film at 823 K showing the highest brightness. From the surface SEM images, the crystal grain size of Ti increases as the temperature rises. This tendency is reasonably explained by previous work on the temperature dependence on grain growth of Ti [9]. Also, the value of surface roughness (Sa) measured by SEM increases as the temperature increases. The smoothest Ti film with Sa = 2.05 ± 0.22 μm was obtained at 823 K. These results show that Ti film with smoother surface can be electrodeposited by suppressing the grain growth of Ti at lower temperatures. To obtain thicker films, electrolysis was conducted for longer time at 823 K, where grain growth is suppressed. Fig. 2 shows the optical and SEM images of the samples obtained by galvanostatic electrolysis at a cathodic current density of 50 mA cm−2 and charge densities of 60, 150, and 450 C cm−2. As shown in the SEM images, the thickness of the Ti film increases with the increase in charge density. Although some deposition onto the edge was observed at 450 C cm−2, the thickness of the Ti film reaches 100 μm. In the literature, there are data about its mechanical characteristics for tensile tests in a range of temperatures of 20 – 750 °С and the limited characteristics of rupture strength. Data on its creep velocity is absent. Laws of creep of steel EP-823 were investigated on metal of three heats with weight of 3 kg. The compression tests at air were applied for cylindrical samples of 5 ÷ 6 mmat temperatures of 600 – 760 °С and stresses of 70 – 310 MPa. The base of compression tests did not exceed 11 hours. The structure after quenching and tempering consisted of tempered martensite and 6 – 12 vol. % of delta-ferrite, the grain size was less 20 μm. It is shown, that the description of creep tests results in double logarithmic (log (σ) – log (έ)) coordinates provides the best concurrence of results of approximation and experiment, than in half-logarithmic (σ – log (έ)). The analysis of parametrical dependences on Hollomon’s PS = (T/1000)[CS – log (έ) and to Larsen-Miller’s PE = (T/1000)[СE – log (σ)] has allowed to find the equations for creep velocity for the set pressure level of 100 – 220 MPa in the form of log (έ) = –19,355 + 9,17 (T/1000) log (σ) and ultimate strength of creep under the set admissions for creep velocity of 0,01 – 1 %/hour in the form of log (σ) = 4,304 – – 0,109 (T/1000) [20 – log (έ)]. Calculations of ultimate strength of creep and creep velocity on pair models and models of Hollomon (Larsen-Miller) give close results, but the preference should be given the second ones as these models consider all three varied factors. Data of control tests under the scheme of a tensile in the same conditions are cited. It is shown, that between results of tests on compression and on tensile at definition of durability characteristics, there is the linear dependence expressed by the equation σ0.2 at compression = 1.3σ0.2 at tensile. At the analysis of creep it has been established, that creep velocity for steel of one grade (09Cr12W3NbB) and for one heat at different type of loading (tensile or compression) have similar values while creep velocities for steel of one grade (EP-823), but of different heats even at one type of loading – compression, can differ substantially.

Molten salts have a number of potential applications including metals’ electrowinning and electrorefining, pyrochemical spent nuclear fuels’ reprocessing. Considerable efforts were devoted over decades to investigation of various electrochemical processes in molten salts, especially in fused chlorides. Silver chloride electrode is one of convenient types of reference electrodes often employed in potentiometry measurements. This electrode has several advantages (especially compactness) compare to the standard chlorine reference electrode, potential of which in chloride melts is taken as zero. Surprisingly little attention was paid so far to the systematic study (covering the effect of temperature and concentration) of silver electrode potentials in molten chlorides, with majority of the works dating back to 1950-s and 60-s. Molten LiCl–KCl eutectic was, on obvious reasons, the best studied, and several other melts (NaCl, NaCl–KCl, LiCl–KCl–CsCl, NaCl–CsCl) were also looked at. The present study was directed at the systematic study of silver electrochemistry in fused alkali chlorides and determining silver electrode potentials. Here we present the results of the experiments performed in the melts based on the ternary NaCl–KCl–CsCl (30–24.5–45.5 mol. %) eutectic mixture at 823–1073 K. Electrode potentials of silver were determined using the electromotive force measurements method and the following galvanic cell: Ag|(NaCl–KCl–CsCl)+AgCl||(NaCl–KC1–CsCl)|Cl2 (C) Concentration of silver in the electrolyte varied from 0.1 to 5 wt. %. Background Almost 50% of the adults in the United States have hypertension. Although clinical trials indicate that home blood pressure monitoring can be effective in managing hypertension, the reported results might not materialize in practice because of patient adherence problems. Objective The aims of this study are to characterize the adherence of Medicaid patients with hypertension to daily telemonitoring, identify the impacts of adherence reminder calls, and investigate associations with blood pressure control. Methods This study targeted Medicaid patients with hypertension from the state of Texas. A total of 180 days of blood pressure and pulse data in 2016-2018 from a telemonitoring company were analyzed for mean transmission rate and mean blood pressure change. The first 30 days of data were excluded because of startup effects. The protocols required the patients to transmit readings by a specified time daily. Patients not transmitting their readings received an adherence reminder call to troubleshoot problems and encourage transmission. The patients were classified into adherent and nonadherent cohorts; adherent patients were those who transmitted data on at least 80% of the days. Results The mean patient age was 73.2 (SD 11.7) years. Of the 823 patients, 536 (65.1%) were women, and 660 (80.2%) were urban residents. The adherent cohort (475/823, 57.7%) had mean transmission rates of 74.9% before the adherence reminder call and 91.3% after the call, whereas the nonadherent cohort (348/823, 42.3%) had mean transmission rates of 39% and 58% before and after the call, respectively. From month 1 to month 5, the transmission rates dropped by 1.9% and 10.2% for the adherent and nonadherent cohorts, respectively. The systolic and diastolic blood pressure values improved by an average of 2.2 and 0.7 mm Hg (P<.001 and P=.004), respectively, for the adherent cohort during the study period, whereas only the systolic blood pressure value improved by an average of 1.6 mm Hg (P=.02) for the nonadherent cohort. Conclusions Although we found that patients can achieve high levels of adherence, many experience adherence problems. Although adherence reminder calls help, they may not be sufficient. Telemonitoring lowered blood pressure, as has been observed in clinical trials. Furthermore, blood pressure control was positively associated with adherence.

Underweight adults have higher rates of respiratory death than the normal weight but it is unclear whether this association is causal or reflects illness-induced weight loss (reverse causality). Evidence from a 45-year follow-up of underweight participants for respiratory mortality in the Whitehall study (N=18 823; 2139 respiratory deaths) suggests that excess risk among the underweight is attributable to reverse causality. The age-adjusted and smoking-adjusted risk was 1.55-fold (95% CI 1.32 to 1.83) higher among underweight compared with normal weight participants, but attenuated in a stepwise manner to 1.14 (95% CI 0.76 to 1.71) after serial exclusions of deaths during the first 5–35 years of follow-up (Ptrend<0.001).

1. The isolated, intact heart of the marine arachnid Limulus polyphemus continues to beat in vitro for many hours. Application of γaminobutyric acid (GABA) decreased the heart beat frequency with a threshold of 3xlO−7 moll−1 and an EC50 of 2.0±0.6xlO−5 moll−1 (mean±s.D., N = 8). At lO−4moll−1 and above the heart beat was completely and reversibly inhibited. 2. The agonist potency profile of the Limulus heart chronotropic GABA receptor was very similar to that of the vertebrate GABAA receptor: muscimol &gt; ZAPA&gt;GABA=⋍TACA&gt;isoguvacine&gt;THIP&gt;3-aminopropane sulphonic acid&gt; imidazole-4-acetic acid ⋍ß-guanidino proprionic acid ⋍5-aminovalerate. In contrast, the antagonist profile differed dramatically: bicuculline, pitrazepin and SR 95103, as well as the channel blocker picrotoxin, were without effect at concentrations up to 10−4moll−1. 3. The benzodiazepines clorazepate, flunitrazepam, flurazepam and diazepam, as well as the barbiturate sodium pentobarbital, were without effect on the GABA response, suggesting that the Limulus heart GABA receptor is not complexed with the benzodiazepine and barbiturate modulatory subunits that characterize vertebrate GABAA receptor. 4. The GABAB ligands baclofen, phaclophen and kojic amine were inactive on the heart. However, 3-aminopropyl-phosphonous acid (CGA147 823), a potent and highly selective GABAb agonist, was the most active of the compounds tested. It inhibited the heart beat with a threshold of about SnmolP1, an EC50 of 4.0±2.7×10−7 mol1−1, and produced total inhibition of the heart at 10−5moll−1. CGA 147 823 was inactive on the locust thoracic somal GABA receptors. 5. cis-4-aminocrotonic acid (CACA), the ligand defining a proposed GAB Actype receptor, was inactive on the heart. 6. The GABA-induced inhibition of the heart beat was enhanced by pretreatment with the GABA uptake inhibitor nipecotic acid but not with sodium valproate or ß-alanine. 7. The Limulus heart chronotropic GABA receptor appears to be of a hitherto undescribed type that differs in pharmacology from the vertebrate GABAA and GABAB receptors as well as from the well-defined GABA receptors on the somata of locust neurones and the muscle fibres of insects and the nematode Ascaris.

Penelitian ini merupakan jenis penelitian kuantitatif, menggunakan metode eksperimen yang terdiri dari 4 perlakuan yaitu A (kontrol tanpa menggunakan hormon), B (pemberian Oocyte Developer 0,1 cc/kg betina), C (Oodev 0,3 cc/kg), dan D (Oodev 0,5 cc/kg betina). Analisis data menggunakan analisis varians (ANOVA). Parameter uji dalam penelitian ini meliputi fekunditas dan daya tetas. Hasil penelitian menunjukkan rerata fekunditas terbaik diberikan pada indukan 0,5 cc/kg dengan total fekunditas 823 ± 125 butir telur dengan daya tetas terbaik juga pada dosis 0,5 cc/kg nila betina dengan hasil rata-rata 49 ± 5 bumbu. Penggunaan Oocyte Developer berpengaruh nyata terhadap fekunditas dan daya tetas ikan nila (Oreochromis niloticus).

The role and potential mechanism of TIMP1 in resistance to 5-Fluorouracil (5-Fu) and stem properties in gastric cancer (GC) were investigated. The expressions of HIF-1α and TIMP1, as well as the chemosensitivity of the 5-Fu in GC cell lines (GCCL) (e.g., MGC-803, BGC-823, SGC-7901, HGC-27 and AGS) upon normoxia or hypoxia were analyzed by means of RT-PCR and CCK-8 assay, respectively. Meanwhile, the population of stem cells was determined by using sphere formation assay, while stem cell markers (SCM) (Oct4 and CD44) were detected by western blot to evaluate stem properties. Hypoxia led to upregulated expression of HIF-1α and TIMP1, and enhanced resistance to 5-Fu, sphere formation capability, and expression of SCM in GC cells (GCCs). Indeed, the expressions of TIMP1 and HIF-1α were positively related to each other. The protein levels of both HIF-1α and TIMP1 were increased and decreased by overexpressing and silencing TIMP1, respectively. Under hypoxia conditions, overexpression of TIMP1 conferred 5-Fu-resistance and stem properties to MGC-803 and AGS cells, as revealed by increased IC50 value of 5-Fu, enhanced sphere formation, and up-regulation of Oct4 and CD44; silencing TIMP1 caused the contrary results. TIMP1 is an effective regulator of HIF-1 and plays a critical role in resistance to 5-Fu and stem properties in GCCs upon hypoxia.

e15031 Background: Theetiology andreliable biomarkers for the risk of bilateral TGCT are still under investigation. Long-term relapse risks of up to 20 years after the therapy in contralateral testes demand a specific model of surveillance. Due to the rarity of the tumor type, reports of national experiences of bilateral TGCT are of a crucial significance for a future global database. Methods: The study was conducted during the treatment of 1 823 patients with TGCT in a single center from 1987-2011. Their mean age was 24 years at the time of diagnosis. Results: Out of 1 823 patients diagnosed with TGCT, 33 developed the bilateral disease (1.81%), 11 of them had synchronous tumors (33%). In the rest of patients the second testicular tumor was diagnosed 1 to 17 years after the initial diagnosis. The mean age of patients with bilateral TGCT was 28. According to the histological type: 11 were seminoma, 14 non-seminoma and 8 mixed seminoma and non-seminoma. Seven patients with bilateral tumor had a seminoma component at initial diagnosis. Compared to 5 patients out of 11 with synchronous tumor, the yolk sac component was detected only in 2 out of 22 metachronous patients. Conclusions: The presented findings of bilateral TGCT prevalence (1.8%) are in concordance with similar previous studies. As this type of tumor may be effectively treated, the risk assessment of metachronous tumor development should in future be facilitated by new biomarkers based on omics techniques. Thus, the investigation of karyotyping, c-KIT and transcriptome specificities will contribute to elucidation which pathways contribute to germ cell neoplasms. The stem cell markers for pluripotency maintenance may in future become potential diagnostic markers used as predictors for metastatic germ cell tumors. The development of a global database on these rare tumors will be of a crucial importance.

The present study aimed to develop a scale for measuring Qataris’ attitudes toward coaching and their intentions towards this profession. The survey target population included Qataris who were 18 years or older and who lived in residential housing units in Qatar. To assure representation of population a proportionate stratified sampling method was used. Overall, 823 Qatari nationals participated in the study. To develop the scale for measuring Qataris’ attitudes toward coaching (ATC), standard scale development procedures were followed. These procedures resulted in a 5-factors measure; Image, Personal development, Job nature, Rewards and Limitations. Exploratory and confirmatory factor analyses provided initial evidence regarding the factorial, convergent and discriminant validity of the scale. The reliability of all factors were also satisfactory. The five aforementioned factors predicted 22% of the variability in job intentions. Theoretical and practical implications of the study are discussed.

Amid the COVID-19 pandemic, a large number of patients with joint lesions resistant to nonsteroidal anti-inflammatory drugs (NSAIDs) was encountered in our clinical practice. The article summarizes the positive experience of outpatient therapy of 916 patients (mean age 47.1±11.3 years), including 774 women, with arthritis of various localization, which occurred on average 45.0±7.1 days after a previously transmitted infection caused by SARS-CoV-2. 709 (77.4%) patients used NSAIDs before going to the clinic, the effect of which was absent or insufficient. In this regard, intramuscular injections of betamethasone prescribed to 823 patients were used as the initial therapy (considering the comorbid background and allergy history). During the drug intake, most of the patients had relief of inflammation, pain reduction, improvement of the disease segment function already on the 7th day. In 69.3% of cases, a single-dose administration was sufficient. The obtained results allow drawing preliminary conclusions about the feasibility of using this technique in the complex treatment of reactive arthritis after the SARS-CoV-2 infection. In vitro experimental plants obtained by clonal micropropagation of 9 grape somaclones of 5 original forms were the material for cytogenetic research. A biological microscope XSP-146TP was used for cytogenetic analysis. 823 cases of deviation from diploidy were observed in total. Significant tissue ploidy was observed in the meristematic tissue of in vitro plants of grape somaclones obtained by colchicine treatment of proembryogenic cells of various varieties. The significant direct correlation was found between the frequency of polyploidy in meristem tissues of in vitro plants and the number of chloroplasts in the stomata of grape somaclones. The reverse correlation was found between the frequency of polyploidy and the number of stomata on the leaf area. Somaclone No. 72, obtained as a result of regeneration from colchicinated proembryogenic cells of the Ruta grape variety and identified as a tetraploid (2n = 4x = 76), is recommended for use in the polyploid creation program.
